Avaliou-se a contaminação da casca e do conteúdo interno de ovos inoculados com Salmonella enterica sorovar enteritidis fagotipo 4, lavados com água de torneira (AT) ou solução de amônia quaternária (AQ) e armazenados a 8ºC e 25ºC. Duzentos e cinqüenta e dois ovos foram divididos em três grupos. Os tratamentos de cada grupo consistiram de imersão em AT, AQ a 25ºC e a 43ºC. Após a secagem natural, todos os grupos foram contaminados com solução de S. enteritidis. Seguindo-se a contaminação, cada grupo tratado foi estocado a 8ºC ou 25ºC, e a presença de S. enteritidis na casca e no conteúdo interno foi avaliada após zero, 24, 96 e 168 horas. A sanitização com AQ mostrou-se eficiente na redução de S. enteritidis nas cascas dos ovos. O armazenamento dos ovos a 8ºC demonstrou ser preponderante na redução e na ausência de S. enteritidis na casca. Nos ovos lavados com AT, o armazenamento a 25ºC permitiu a permanência da bactéria nas cascas até 168 horas. Não se detectou S. enteritidis no conteúdo interno dos ovos em nenhum dos grupos.

Societal concerns about environmental sustainability has lead to the development of ecologically-friendly alternatives to chemical insecticides for crop protection. One such alternative is biological pest control. In particular, baculoviruses are well suited as insect biopesticides due to their narrow host specificity and relative ease of propagation. In Brazil, the baculovirus Anticarsia gemmatalis nucleopolyhedrovirus (AgMNPV) is the main biological control agent employed for the soybean pest, Anticarsia gemmatalis. This baculovirus biopesticide is currently produced using caterpillars, but increasing market demand for the product has encouraged the development of an in vitro manufacturing process, which can be scaled up to much higher virus productivities. In this study, three wild-type AgMNPV isolates (AgMNPV-2D, AgMNPV-MP2 and AgMNPV-MP5) and a recombinant form (vAgEGT-LacZ) were characterised in terms of occlusion body (OB) production and infection kinetics, to enable future optimisation of the in vitro production process. These viruses were propagated using a Spodoptera frugiperda (IPLB-SF21) insect cell line grown in shaker-flask batch cultures. Among the virus isolates tested, AgMNPV-MP5 was found to be the best producer, yielding (5.3±0.85)x108 OB/mL after 8 days post infection. The characterisation of vAgEGT-LacZ propagation in suspension cell cultures has not been previously reported in the literature; hence it became the main focus for this thesis. In particular, it was carried out a study on the effect of the multiplicity of infection (MOI) on OB production. Five successive batches were performed getting a final production (8.9±1.42)x1014 occlusion bodies, considering that production is related for a bioreactor with final volume of 10m3. A low MOI associated with a fed-batch process for vAgEGT-LacZ production was found to support a 3-fold higher OB yield when compared to the default batch process (1.8x107 and 5.3x107 OB/mL, respectively). This yield is competitive with regards to the production process.

The robustness and performance of the Variable Structure Adaptive Pole Placement Controller are evaluated in this work, where this controller is applied to control a synchronous generator connected to an infinite bus. The evaluation of the robustness of this controller will be accomplished through simulations, where the control algorithm was subjected to adverse conditions, such as: disturbances, parametric variations and unmodeled dynamic. It was also made a comparison of this control strategy with another one, using classic controllers. In the simulations, it is used a coupled model of the synchronous generator which variables have a high degree of coupling, in other words, if there is a change in the input variables of the generator, it will change all outputs simultaneously. The simulation results show which control strategy performs better and is more robust to disturbances, parametric variations and unmodeled dynamics for the control of Synchronous Generator

Survival models deals with the modeling of time to event data. However in some situations part of the population may be no longer subject to the event. Models that take this fact into account are called cure rate models. There are few studies about hypothesis tests in cure rate models. Recently a new test statistic, the gradient statistic, has been proposed. It shares the same asymptotic properties with the classic large sample tests, the likelihood ratio, score and Wald tests. Some simulation studies have been carried out to explore the behavior of the gradient statistic in fi nite samples and compare it with the classic statistics in diff erent models. The main objective of this work is to study and compare the performance of gradient test and likelihood ratio test in cure rate models. We first describe the models and present the main asymptotic properties of the tests. We perform a simulation study based on the promotion time model with Weibull distribution to assess the performance of the tests in finite samples. Logic courses represent a pedagogical challenge and the recorded number of cases of failures and of discontinuity in them is often high. Amont other difficulties, students face a cognitive overload to understand logical concepts in a relevant way. On that track, computational tools for learning are resources that help both in alleviating the cognitive overload scenarios and in allowing for the practical experimenting with theoretical concepts. The present study proposes an interactive tutorial, namely the TryLogic, aimed at teaching to solve logical conjectures either by proofs or refutations. The tool was developed from the architecture of the tool TryOcaml, through support of the communication of the web interface ProofWeb in accessing the proof assistant Coq. The goals of TryLogic are: (1) presenting a set of lessons for applying heuristic strategies in solving problems set in Propositional Logic; (2) stepwise organizing the exposition of concepts related to Natural Deduction and to Propositional Semantics in sequential steps; (3) providing interactive tasks to the students. The present study also aims at: presenting our implementation of a formal system for refutation; describing the integration of our infrastructure with the Virtual Learning Environment Moodle through the IMS Learning Tools Interoperability specification; presenting the Conjecture Generator that works for the tasks involving proving and refuting; and, finally to evaluate the learning experience of Logic students through the application of the conjecture solving task associated to the use of the TryLogic

The general aim of this work is to verify the occurence of variant forms of negation in spoken English with the purpose of making a comparative study between the English and the Portuguese languages. As for Portuguese, we used as a matter of reference a study already made on negation. As for English, we analized a corpus of the North American English variant organized by a university in the United States. This study is based on the North American Functional Linguistics theoretical perspective, which considers relevant the study of language used in real situations of communicative interaction. The data analisys proved that there is at least one form of negative variant in spoken English which is not allowed by prescriptive grammar. This phenomenon turns out to be similar to Portuguese, which includes three variant strategies. According to the data obtained, it was possible to verify that the variant strategy used in English, from a contrastive point of view, corresponds to a negative strategy ruled by Portuguese prescriptive grammar. Finally, we discussed about the different conceptions of language, grammar and teaching, giving suggestions to colaborate to a productive and reflexive teaching of first or second language

Este artigo é parte de uma pesquisa qualitativa mais abrangente que utilizou como referencial teórico-metodológico a Grounded Theory e o Interacionismo Simbólico, resultando no modelo teórico denominado Entre o sofrimento e a esperança: a reabilitação da incontinência urinária como componente interveniente. Com a intenção de comunicar todo o conhecimento produzido, apresenta-se parte desse modelo, referente ao processo de enfrentamento da incontinência urinária por mulheres sem perspectivas de acesso ao tratamento cirúrgico, após falha dos procedimentos conservadores. Ao inter-relacionar os componentes (categorias e subcategorias) relativos à experiência dessas mulheres, buscando compará-los e analisá-los para compreender a interação entre eles, notou-se vulnerabilidade moral e psicossocial no movimento da experiência do grupo, suscetibilizando-o a riscos à saúde e ao comprometimento da qualidade de vida. Pesquisas são necessárias para aprofundar a compreensão de experiências em que haja barreira ao tratamento cirúrgico por descrédito do profissional médico sobre sua efetividade.

This study aimed to evaluate the use of hydrogel in the survival of cuttings of Eucalyptus urograndis produced with different types of substrate and water management. The experiment was carried out using cuttings of the same clone, produced up to 50 days after the cutting, in two different nurseries ("C" and "E"). Then, all the cuttings were allocated to the same nursery for 80 days, where they received irrigation at different frequencies. After this period, the cuttings were transplanted into polyethylene pots, in clay soil, with and without hydrogel. The results obtained were subjected to the variance technique for totally randomized experiments. Data analysis revealed that the plants with hydrogel displayed delayed symptoms of water deficit. Regardless of the cutting's origin ("C" or "E"), the hydrogel influenced survival, guaranteeing 37 days without additional irrigation. The nursery water management only influenced the plants from nursery "E", and the plants adapted to the lack of water (water management with one daily irrigation) took more time to present symptoms of water deficit.

A dispersão de diásporos é um evento de elevada importância para as espécies vegetais, que dispõem de diversas estratégias para este fim. O presente estudo teve por objetivos conhecer e caracterizar o espectro das diferentes estratégias de dispersão do componente arbóreo em uma floresta estacional de Santa Maria, Rio Grande do Sul, Brasil (próximo às coordenadas 53º54'W e 29º38'S) e, a partir desses dados, investigar sua relação com os estratos verticais da floresta. Foram amostrados todos os indivíduos com perímetro à altura do peito > 15 cm, em 100 unidades amostrais de 10×10 m. A caracterização das estratégias de dispersão foi realizada por meio de observações a campo de frutos e potenciais dispersores, e consulta à bibliografia especializada. Foram amostradas 58 espécies pertencentes a 26 famílias botânicas. Dessas espécies, 74% apresentaram estratégia de dispersão zoocórica; 24%, estratégia de dispersão anemocórica e apenas Gymnanthes concolor Spreng. apresentou a estratégia autocórica. Analisando-se a proporção das estratégias de dispersão por meio da densidade relativa, verificou-se que 80% dos indivíduos são zoocóricos, 12% anemocóricos e 8% autocóricos. Para a floresta analisada foram encontradas diferenças significativas nos padrões de distribuição vertical das espécies agrupadas de acordo com a estratégia de dispersão. A zoocoria ocorreu em espécies de todos os estratos, enquanto a autocoria ficou limitada ao sub-bosque. Já a anemocoria foi mais importante entre as espécies com indivíduos emergentes.

Neste artigo discute-se o trabalho com valores em Educação Ambiental, o que exige uma fundamentação e posicionamento adequados. Diante do impasse entre posições relativistas e universalistas para a educação em valores, aponta-se para a necessidade de explicitar nossa posição e ação em favor dos valores ambientalmente desejáveis, considerando que as questões envolvidas com o meio ambiente dizem respeito à vida e à sobrevivência de todos os seres do planeta. Estes valores podem ser identificados junto aos princípios presentes no Tratado de educação global para sociedades sustentáveis e responsabilidade global, apresentado pela sociedade civil na ECO-92. Por fim, ressalta-se a necessidade de se desenvolverem estratégias educativas que envolvam as dimensões aqui denominadas de cognição, afetividade e ação, resultando em um trabalho abrangente, que amplie as possibilidades de o indivíduo apreender, de maneira mais efetiva, um dado valor, tendo, então, melhores condições de construí-lo em sua vida.
