Essential and Molecular Dynamics (ED/MD) have been used to model the conformational changes of a protein implicated in a conformational disease-cataract, the largest cause of blindness in the world-after non-enzymic post-translational modification. Cyanate modification did not significantly alter flexibility, while the Schiff's base adduct produced a more flexible N-terminal domain, and intra-secondary structure regions, than either the cyanate adduct or the native structure. Glycation also increased linker flexibility and disrupted the charge network. A number of post-translational adducts showed structural disruption around Cys15 and increased linker flexibility; this may be important in subsequent protein aggregation. Our modelling results are in accord with experimental evidence, and show that ED/MD is a useful tool in modelling conformational changes in proteins implicated in disease processes. Prebiotics are non-digestible (by the host) food ingredients that have a beneficial effect through their selective metabolism in the intestinal tract. Key to this is the specificity of microbial changes. The present paper reviews the concept in terms of three criteria: (a) resistance to gastric acidity, hydrolysis by mammalian enzymes and gastrointestinal absorption; (b) fermentation by intestinal microflora; (c) selective stimulation of the growth and/or activity of intestinal bacteria associated with health and wellbeing. The conclusion is that prebiotics that currently fulfil these three criteria are fructo-oligosaccharides, galacto-oligosaccharides and lactulose, although promise does exist with several other dietary carbohydrates. Given the range of food vehicles that may be fortified by prebiotics, their ability to confer positive microflora changes and the health aspects that may accrue, it is important that robust technologies to assay functionality are used. This would include a molecular-based approach to determine flora changes. The future use of prebiotics may allow species-level changes in the microbiota, an extrapolation into genera other than the bifidobacteria and lactobacilli, and allow preferential use in disease-prone areas of the body.

Current intakes of very long-chain omega-3 fatty acids, eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), are low in most individuals living in Western countries. A good natural source of these fatty acids is seafood, especially oily fish. Fish oil capsules contain these fatty acids also. Very long-chain omega-3 fatty acids are readily incorporated from capsules into transport (blood lipids), functional (cell and tissue), and storage (adipose) pools. This incorporation is dose-dependent and follows a kinetic pattern that is characteristic for each pool. At sufficient levels of incorporation, EPA and DHA influence the physical nature of cell membranes and membrane protein-mediated responses, lipid-mediator generation, cell signaling, and gene expression in many different cell types. Through these mechanisms, EPA and DHA influence cell and tissue physiology and the way cells and tissues respond to external signals. In most cases the effects seen are compatible with improvements in disease biomarker profiles or health-related outcomes. As a result, very long-chain omega-3 fatty acids play a role in achieving optimal health and in protection against disease. Long-chain omega-3 fatty acids not only protect against cardiovascular morbidity but also against mortality. In some conditions, for example rheumatoid arthritis, they may be beneficial as therapeutic agents. On the basis of the recognized health improvements brought about by long-chain omega-3 fatty acids, recommendations have been made to increase their intake. The plant omega-3 fatty acid, alpha-linolenic acid (ALA), can be converted to EPA, but conversion to DHA appears to be poor in humans. Effects of ALA on human health-related outcomes appear to be due to conversion to EPA, and since this is limited, moderately increased consumption of ALA may be of little benefit in improving health outcomes compared with increased intake of preformed EPA + DHA.

Obesity has become a major global health problem. Recently, attention has focused on the benefits of fermentable carbohydrates on modulating metabolism. Here, we take a system approach to investigate the physiological effects of supplementation with oligofructose-enriched inulin (In). We hypothesize that supplementation with this fermentable carbohydrate will not only lead to changes in body weight and composition, but also to modulation in neuronal activation in the hypothalamus. Male C57BL/6 mice were maintained on a normal chow diet (control) or a high fat (HF) diet supplemented with either oligofructose-enriched In or corn starch (Cs) for 9 weeks. Compared to HF+Cs diet, In supplementation led to significant reduction in average daily weight gain (mean ± s.e.m.: 0.19 ± 0.01 g vs. 0.26 ± 0.02 g, P < 0.01), total body adiposity (24.9 ± 1.2% vs. 30.7 ± 1.4%, P < 0.01), and lowered liver fat content (11.7 ± 1.7% vs. 23.8 ± 3.4%, P < 0.01). Significant changes were also observed in fecal bacterial distribution, with increases in both Bifidobacteria and Lactobacillius and a significant increase in short chain fatty acids (SCFA). Using manganese-enhanced MRI (MEMRI), we observed a significant increase in neuronal activation within the arcuate nucleus (ARC) of animals that received In supplementation compared to those fed HF+Cs diet. In conclusion, we have demonstrated for the first time, in the same animal, a wide range of beneficial metabolic effects following supplementation of a HF diet with oligofructose-enriched In, as well as significant changes in hypothalamic neuronal activity.

The global population is becoming increasingly older presenting medical and economic challenges to society. One factor associated with the aging process is immunosenescence, which may be defined as the decline in immunity with age, and represents a potential causative factor for many age related illnesses. The profile of the gut microbiota is also known to alter with aging and these changes have been linked the declines in the immunity observed in immunosenescence. For example, above the age of 60 years populations of bifidobacteria have been observed to decrease markedly, leading to a reduction in the inhibition of the growth of some pathogens and potentially an increase in the susceptibility to infections. As such, an interest exists in attempting to reverse their decline in elderly individuals, through the use of both probiotics and prebiotics. Both approaches have shown to be encouraging in altering microbiota profiles beneficially and in reducing immunosenescence by reducing the colonisation potential of pathogens and counteracting chronic inflammation. The current review will give an overview of the process of immunosenescence and its role in disease, detail how the microbiota are involved in its progression and highlight data suggesting that pre- and probiotics may counteract these age-related events.

Cleaner fishes are usually classified as obligate or facultative cleaners according to their diet and the extent to which their nutritional requirements in the different ontogenetic stages are gained from cleaning. While obligate cleaners clean throughout their lives and ingest mainly food taken from the clients` body surface, facultative cleaners clean only as juveniles and have a broader diet. In addition, some facultative cleaners may experience a relatively higher predation risk, and thus rarely interact with piscivorous fishes. Despite these acknowledged differences, there are very few studies that compare cleaning activity of obligate and facultative cleaners within the same area. Cleaning activity of the obligate cleaner goby Elacatinus cf. randalli and the facultative cleaner wrasse Thalassoma noronhanum were comparatively examined at Fernando de Noronha Archipelago, tropical West Atlantic. The client assemblage attended by the two cleaners differed, as the goby attended a slightly greater diversity of species (22), mostly piscivores and zoobenthivores, and the wrasse attended fewer species (19), mostly planktivores. Chromis multilineata was the most common client species of both cleaners, although body size (which is expected to be positively correlated to clients` ectoparasite load) of C. multilineata individuals attended by the goby was larger than that of the individuals attended by the wrasse. Despite such differences, T. noronhanum showed a surprisingly species-rich client assemblage when compared with other cleaners of the genus Thalassoma. In addition, the frequency and time spent on cleaning interactions, as well as the number of client species attended per 10-min period, was similar for both cleaner species, which indicate that they have important yet complimentary ecological roles in the reef community at Fernando de Noronha Archipelago.

Aim The microbial profile of localized aggressive periodontitis (LAgP) has not yet been determined. Therefore, the aim of this study was to evaluate the subgingival microbial composition of LAgP. Material and Methods One hundred and twenty subjects with LAgP (n=15), generalized aggressive periodontitis (GAgP, n=25), chronic periodontitis (ChP, n=30) or periodontal health (PH, n=50) underwent clinical and microbiological assessment. Nine subgingival plaque samples were collected from each subject and analysed for their content of 38 bacterial species using checkerboard DNA-DNA hybridization. Results Red complex and some orange complex species are the most numerous and prevalent periodontal pathogens in LAgP. The proportions of Aggregatibacter actinomycetemcomitans were elevated in shallow and intermediate pockets of LAgP subjects in comparison with those with GAgP or ChP, but not in deep sites. This species also showed a negative correlation with age and with the proportions of red complex pathogens. The host-compatible Actinomyces species were reduced in LAgP. Conclusion A. actinomycetemcomitans seems to be associated with the onset of LAgP, and Porphyromonas gingivalis, Tannerella forsythia, Treponema denticola, Campylobacter gracilis, Eubacterium nodatum and Prevotella intermedia play an important role in disease progression. Successful treatment of LAgP would involve a reduction in these pathogens and an increase in the Actinomyces species.

O presente estudo buscou analisar a viabilidade econômica, agronômica e ambiental de um sistema que está em execução em uma propriedade no norte do estado de Goiás. Esse sistema, denominado Sistema Triângulo, vai de encontro com a Integração Lavoura-Pecuária e busca formas alternativas para o uso dos restos culturais (fardos) de soja fornecendo esse material como volumoso para o gado no sistema de confinamento. Para o estudo da viabilidade, foram usadas as informações da própria fazenda num estudo de caso. Para tal, realizou-se uma revisão quanto à adoção de sistema de produção na área agrícola, quanto às interações dos macronutrientes que seriam retirados das áreas por intermédio dos fardos, o papel da matéria orgânica quanto à sustentabilidade do sistema, o cenário atual dos confinamentos no Brasil e os custos que compõem e determinam a lucratividade dessa atividade. Por ser um estudo de caráter incremental, a viabilidade econômica ficou focada à redução dos custos alimentares. Foram utilizadas informações produzidas pela Fazenda Triângulo que é produtora de soja desde 2002. Sendo assim, foi possível acompanhar a evolução dos teores de macronutrientes (nitrogênio, fósforo, potássio, cálcio, magnésio e enxofre) e do carbono em uma área total de 351 ha. Os resultados mostraram uma estabilidade nos níveis dos macroelementos após o início da retirada dos restos culturais mas, apresentou-se inconclusivo no caso do carbono. Foi estimada também a exportação dos macronutrientes por intermédio da retirada dos fardos das áreas de soja. Levantada a composição química dos fardos, foram estabelecidos seis cenários de reposição via adubação específica e esses custos foram acrescidos de custos de processamento dos fardos e o custo dos outros componentes da dieta no confinamento. Calculado o valor presente líquido para cada um dos cenários e comparando com a alternativa tradicional de dieta de confinamento, onde se usa a silagem de milho como 100% do volumoso, mostrou-se uma redução de até 5,02% nos custos alimentares. Com as práticas agronômicas da produção de soja vigentes hoje, recomenda-se a adoção do Sistema Triângulo com a ressalva de uma necessidade de maiores estudos quando seu impacto nos teores de carbono orgânico no solo.

Enterocutaneous fistulas are associated with prolonged hospital stay, high morbidity/mortality, and increase in hospital costs. This study aims to describe the use of a vacuum system and normal oral diet in dealing with this problem. Methods: Seventy-four consecutive patients with recent and defined external postoperative fistulas were analyzed. Abdominal imaging was used to exclude abscess and distal obstruction. The fistula tract was sealed with Foley catheter, connected to a negative pressure flask, changed daily for 5, 10 or 15 days, as necessary. Normal oral diet was permitted. Results: No patient died. Serum albumin and transferrin showed significantly higher levels at the end of treatment than at the beginning. The moderate and low-output fistulas had the best results (97% closed). Forty-eight (65%) fistulas closed after five days, 16(22%) after 10 days and 4(5%) after 15 days. Treatment failed in 6(8%) patients, who subsequently underwent surgery. Only one patient with low-output did not close her fistula. The cost of the treatment was US$ 41.75/day and it was considered cost effective. Conclusions: The vacuum system demonstrated good results in the treatment of fistulas. It included simplicity, low cost, short hospital stay, absence of skin breakdown, normal eating, good nutrition and activity patterns

O objetivo neste trabalho foi avaliar o desempenho de pacus (Piaractus mesopotamicus) criados em tanques-rede e alimentados com dietas contendo níveis de proteína bruta (PB) e energia digestível (ED). Foram utilizados 3.960 peixes com 293,38 ± 5,67 g de peso inicial, distribuídos em 18 tanques-rede de 5 m³, com 220 peixes por unidade experimental (44 peixes/m³), em esquema fatorial 3 × 2, composto de três níveis de proteína bruta (25, 30 e 35%) e dois de energia digestível (3.250 e 3.500 kcal/kg). O arraçoamento foi realizado quatro vezes ao dia (às 9 h, 11h30min, 14 h e 17 h) até a saciedade aparente dos animais. Não foram observadas diferenças no ganho de peso, na taxa de sobrevivência, na conversão alimentar aparente nem na taxa de crescimento específico. No entanto, houve diferença na deposição de gordura visceral, que foi maior nos animais alimentados com as rações de maior nível energético. Também não foi observada influência dos níveis de proteína e energia da dieta nos teores de umidade, proteína bruta, matéria mineral e lipídio dos filés. Rações contendo 25% de proteína bruta e 3.250 kcal/kg de energia digestível promovem melhores resultados de desempenho.

Um isolado de Fusarium graminearum estudado na UNESP/Campus de Jaboticabal, onde foi comprovada sua eficácia no controle de Egeria densa e E. najas, macrófitas aquáticas submersas. Para estudar o efeito de diferentes concentrações do inóculo na severidade de doença, foram conduzidos experimentos em incubadoras para BOD, com concentrações variando em um décimo, de 0,1 até 1,4 g/l de arroz moído colonizado com F. graminearum. Para verificar os possíveis efeitos da idade da planta sobre a severidade de doença, plantas com, no mínimo 35 cm, de comprimento foram excisadas em segmentos correspondentes às idades de crescimento. Os tratamentos com concentrações de inóculo a partir de 0,5 g/l apresentaram sintomas. Todos os tratamentos inoculados com o fungo, nas concentrações a partir de 0,5 g/l, apresentaram drástica redução na produção de biomassa fresca. Todos os segmentos utilizados como plantas-teste (0 a 32 cm de comprimento) apresentaram suscetibilidade ao fungo. Os ponteiros de plantas de ambas as espécies apresentaram maior severidade de sintomas no sexto e oitavo dias após a inoculação, contudo, os segmentos correspondentes às idades 2 e 3 apresentaram maior redução de biomassa fresca quando inoculadas, apesar de não apresentarem sintomas tão severos quanto a idade 1. Com relação ao ganho de biomassa fresca, as testemunhas apresentaram sempre maior crescimento do que os respectivos tratamentos inoculados, contudo os segmentos correspondentes à idade 4 apresentaram menor ganho de biomassa fresca do que as demais idades.

A citricultura é um mercado em expansão, principalmente no Estado de São Paulo, cuja importância na balança comercial já é reconhecida. Como em qualquer espécie cultivada, o crescimento das áreas de cultivo favorecem também o crescimento de problemas fitossanitários. Desta forma, as espécies de citros são afetadas por diversas doenças destacando-se entre elas a melanose, causada por Diaporthe citri (Wolf.), à qual a grande maioria das variedades comerciais são suscetíveis. O conhecimento da diversidade intra-específica é de grande importância, já que esta poderá auxiliar na seleção de variedades com resistência. O objetivo deste trabalho foi avaliar a variabilidade genética em isolados de Diaporthe citri, originários de diferentes locais, variedades e partes da planta, utilizando marcadores moleculares. Marcadores do tipo AFLP (Amplified Fragment Length Polymorphism) foram utilizados para caracterização de dez isolados do patógeno. Os DNAs genômicos extraídos da massa micelial foram utilizados nas reações de amplificação. A técnica fluorescent AFLP permitiu a distinção dos isolados estudados, tendo sido classificados em quatro grupos distintos. Contudo, estes grupos não foram formados em razão da região geográfica, parte da planta ou variedade.
