The pathogenicity and immunogenicity of six recently isolated Paracoccidioides brasiliensis samples derived from patients presenting distinct and well defined clinical forms of paracoccidioidomycosis (PCM) were compared as to their virulence, tropism to different organs and ability to induce specific cellular and humoral immune response in susceptible (B10.A) inbred mice. Isolates Pb44 and Pb47 were obtained from acute cases, Pb50 from a chronic severe form, Pb45 from a chronic moderate case and both Pb56 and Pb57 from chronic mild forms of PCM. Pathogenicity and tropism of each fungal sample were evaluated by LD50% estimation, examination of gross lesions on various organs at 2, 4, 12 and 16 weeks post-infection, and by colony-forming unit (CFU) counts in the lungs at week 16 post-infection of mice. Fungal tropism in human PCM and in B10.A mice was always dissociated. A well defined relationship between virulence of the fungal sample and the clinical findings of the correspondent patient was not evident, although a tendency to higher LD50% and less intense paracoccidioidic lesions was observed in mice infected with Pb56 and Pb57. The specific DTH response patterns varied according to the infectant sample, but positive DTH reactions at the beginning of the infection and a tendency to anergy or low DTH responses at week 12 and/or week 16 post-infection were always observed. A correspondence between the DTH response in humans and in mice was noticeable only when the isolates from the most benign cases (Pb56 and Pb57) were considered. The specific antibody patterns in mice and in the correspondent patients were also not analogous. Collectively, these results indicate that an association between the fungal pathogenicity and immunogenicity in the human disease and in susceptible mice was discernible only when isolates obtained from very mild cases (Pb56 and Pb57) were considered.

A joint experimental and theoretical study has been carried out to rationalize the results of visible photoluminescence measurements at room temperature on Sr1-xTiO3-x (ST) perovskite thin films. From the experimental side, ST thin films, x = 0 to 0.9, have been synthesized following soft chemical processing, and the corresponding photoluminescence properties have been measured. First principles quantum mechanical techniques, based on density functional theory at the B3LYP level, have been employed to study the electronic structure of a crystalline, stoichiometric (x = 0) ST-s model and a nonstoichiometric (SrO-deficient, x not equal 0) and structurally disordered ST-d model. The relevance of the present theoretical and experimental results of the photoluminescence behavior of ST is discussed. The optical spectra and the calculations indicate that the symmetry-breaking process on going from ST-s to ST-d creates electronic levels in the valence band. Moreover, an analysis of the Mulliken charge distribution reveals a charge gradient in the structure. These combined effects seem to be responsible for the photoluminescence behavior of deficient Sr1-xTiO3-x.

Strong photoluminescent emission has been measured at room temperature for noncrystalline BaT'O-3 (BT) perovskite powders. A joint experimental and theoretical study has been carried out to rationalize this phenomenon. From the experimental side, BT powder samples have been synthesized following a soft chemical processing, their crystal structure has been confirmed by x-ray data and the corresponding photoluminescence (PL) properties have been measured. Only the structurally disordered samples present PL at room temperature. From the theoretical side, first-principles quantum-mechanical techniques, based on density-functional theory at the B3LYP level, have been employed to study the electronic structure of crystalline (BT-c) and asymmetric (BT-a) models. Theoretical and experimental results are found to be consistent and their confrontation leads to an interpretation of the PL apparition at room temperature in the structurally disordered powders.

Intense and broad photoluminescence (PL) emission at room temperature was observed on structurally disordered Ba[Zr0.25Ti0.75]O-3 (BZT) powders synthesized by the polymeric precursor method. BZT powders were annealed at 573 K for different times and at 973 K for 2 h in oxygen atmosphere. The single-phase cubic perovskite structure of the powder annealed at 973 K for 2 It was identified by X-ray diffraction and Fourier transform Raman techniques. PL emission increased with the increase of annealing time, which reached its maximum value in the powder annealed at 573 K for 192 h. First principles quantum mechanical calculations based on density functional theory (B3LYP level) were employed to study the electronic structure of ordered and disordered models. The theoretical calculations and experimental measurements of Ultraviolet-visible absorption spectroscopy indicate that the presence of intermediary energy levels in the band gap is favorable for the intense and broad PL emission at room temperature in disordered BZT powders. The PL behavior is probably due the existence of a charge gradient on the disordered structure, denoted by means of a charge transfer process from [TiO5]-[ZrO6] or [TiO6]-[ZrO5] clusters to [TiO6]-[ZrO6] clusters. The behavioral and hematological effects of treatment with Chamomilla 6cH in mice subjected to experimental stress are described. Swiss mice were randomly divided into pairs, one animal was inoculated with Ehrlich's tumor, the other was treated daily with Chamomilla 6cH or control or received no treatment. After 7 days, the animals were observed in an open-field arena and blood samples taken. Mice who cohabitated with a sick cage-mate showed a decrease in their general activity, but those treated with Chamomilla 6cH were less severely affected (p = 0.0426). No hematologicall changes were observed.In a second experiment, the forced swimming test was applied to mice pre-treated with Chamomilla 6cH, controls were: water, 10% ethanol or amitriptyline. Only the amitriptyline and ethanol treated groups showed significant excitatory behavior (p = 0.0020), Chamomilla 6cH treated animals' scores intermediate between water control and ethanol or amitriptyline. A decrease in the leukocyte count was observed in the amitriptyline and Chamomilla 6cH treated groups (p = 0.039). These data suggest that treatment with Chamomilla 6cH is related to the recovery of basal behavioral conditions in mice subjected to stressful conditions.

Venous rethrombosis following thrombectomy is a common event. The aim of the present study was to verify the action of heparin, heparin plus acetyl salicylic acid (ASA) and dipyridamole, and of an arteriovenous fistula (AVF) in the prevention of this complication. Thrombosis was induced in 48 male rabbits by the injection of thrombin in a segment of the left jugular vein, in which the blood flow was arrested for 10 minutes. After 48 hours, the animals were randomly allocated into one of 4 groups of treatment: (1) control, (2) subcutaneous heparin (600 S.I. Units/kg - 8/8 hours), (3) heparin, in the same dose, plus ASA (10 mg/kg/once a day), and dipyridamole (0.5 mg/kg thrice a day), (4) an AVF was surgically constructed between the left carotid after and the left maxillar vein. After 30 minutes, thrombectomy was performed. The venous blood flow, the hematocrit, activated partial thromboplastin time and thrombin time tests were performed before, right after the thrombectomy and 48 hours after thrombectomy. Venography was performed after thrombectomy and at the end of the experiment. The animals were killed 48 hours after thrombectomy and the veins were examined macroscopically. Venous rethrombosis was significantly prevented only in the AVF group (9/12), when compared to control group (0/12), heparin group (1/12) and heparin plus antiaggregating agents group (2/12). These results validate further clinical and experimental investigations with the use of AVF to prevent rethrombosis after venous thrombectomy, when a reduction of venous flow is present.

In this study we optimized an enzyme-linked immunosorbent assay (ELISA) to evaluate bothropic venom levels in biological samples. These samples were obtained by two distinct protocols. In the first one, Swiss mice were injected with 1 LD 50 of Bothrops jararaca (B. jararaca) venom and 15 minutes later, animals were treated with ovine antibothropic serum. Blood and spleen homogenate samples were obtained 6 hours after antiserum therapy. Ovine antibothropic serum significantly neutralized venom levels in serum and spleen. In the second protocol, BALB/c mice were injected with 1 LD 50 of bothropic venom by either intraperitoneal (IP) or intradermal (ID) route and venom levels were evaluated 1, 3 and 6 hours after, in blood, spleen homogenates and urine. Serum and splenic venom levels were significantly higher in animals envenomed by IP route comparing with animals envenomed by ID route. Higher venom levels were also detected in urine samples from animals envenomed by IP route. However, these differences were not statistically significant. These results demonstrated that the optimized ELISA was adequate to quantify venom levels in different biological samples. This assay could, therefore, substitute the in vivo neutralizing assay and also be useful to evaluate the severity of human and experimental envenomations.

This paper presents a comparative analysis between the experimental characterization and the numerical simulation results for a three-dimensional FCC photonic crystal (PhC) based on a self-assembly synthesis of monodispersive latex spheres. Specifically, experimental optical characterization, by means of reflectance measurements under variable angles over the lattice plane family [1,1, 1], are compared to theoretical calculations based on the Finite Di®erence Time Domain (FDTD) method, in order to investigate the correlation between theoretical predictions and experimental data. The goal is to highlight the influence of crystal defects on the achieved performance.

A Análise do Comportamento oferece diversas explicações para o fenômeno denominado depressão, uma das quais envolve a referência ao modelo do desamparo aprendido. O desamparo aprendido é definido como a dificuldade de aprendizagem resultante da exposição a estímulos aversivos incontroláveis. Como produtos desta exposição, surgiriam padrões comportamentais comuns àqueles observados em indivíduos depressivos, a exemplo de inatividade. Em razão do paralelo entre os efeitos da experiência com incontrolabilidade sobre o repertório comportamental em humanos e não-humanos, o desamparo aprendido tem sido apontado como um modelo animal de depressão. Frequentemente, menções à experiência com incontrolabilidade são encontradas na literatura em associação com o desamparo aprendido, cuja ocorrência é estritamente vinculada àquela condição. A incontrolabilidade também parece relevante para a instalação de respostas identificadas com a depressão. No presente trabalho, descrevem-se as definições de incontrolabilidade referidas por publicações da área comportamental experimental e clínica, discutindo-se a pertinência deste conceito em explicações funcionais da depressão na Análise do Comportamento, bem como suas possíveis contribuições para um modelo de intervenção clínica da depressão à luz desta abordagem. A relação entre incontrolabilidade e depressão é tratada a partir de cinco categorias de análise: 1) Variabilidade de fenômenos investigados, de resultados produzidos e de definições oferecidas; 2) Efeitos diferenciais da incontrolabilidade frente a estímulos aversivos e apetitivos; 3) Enfoques transversais das variáveis relevantes: instaladoras x mantenedoras, históricas x atuais, exclusivas x sobrepostas a outros fenômenos; 4) Incontrolabilidade em Humanos: suposições numerosas, evidências empíricas escassas e Contingências Verbais; 5) Tratamento da depressão: pontos de contato e de distanciamento frente à investigação empírica. Diferentes usos do conceito de incontrolabilidade são assinalados, indicando-se como a mesma topografia verbal é emitida por diversos autores sob controle de eventos distintos. Variáveis relevantes para a generalidade do desamparo aprendido – enquanto modelo experimental e equivalente animal da depressão – são discutidas, justificando-se a necessidade de maior investigação de aspectos como: correspondência entre o conceito de incontrolabilidade e a condição experimentalmente estabelecida no laboratório; efeitos de diferentes tipos de estimulação incontrolável; produção de desamparo aprendido em humanos e participação de processos verbais; diferentes efeitos da sinalização pré-aversiva de estímulos incontroláveis. Assinala-se que, em geral, o tratamento analítico-comportamental da depressão é constituído por procedimentos com foco sobre a aprendizagem de que responder controla o ambiente e pode disponibilizar reforçadores. É examinado o papel da incontrolabilidade na instalação da depressão, constatando-se, por fim, que se apresenta como condição suficiente, porém não necessária para a ocorrência e/ou manutenção do fenômeno.

O enriquecimento ambiental e os efeitos do envelhecimento sobre as alterações morfológicas das micróglias e no comportamento foram investigados em modelo murino de encefalite sub-letal por arbovírus. Para medir possíveis influências da idade e do ambiente sobre a progressão da encefalite, camundongos suíços albinos fêmeas de 2 meses de idade foram mantidos em Ambiente Padrão (AP) ou em Ambiente Enriquecido (AE), durante: 6 meses (Adulto - A) e 16 meses (Senil –S). Após os testes comportamentais, os camundongos A e S foram inoculados intranasalmente com igual volume de homogenado de cérebro de camundongo infectado pelo vírus Piry (Py) ou homogenado de cérebro de camundongo normal. Oito dias após a inoculação (8DPI), quando os primeiros testes comportamentais revelaram as alterações relacionadas à doença, os cérebros foram seccionados e inumomarcados seletivamente para IBA-1 e antígenos virais. Aos 20 ou 40DPI, os animais restantes foram testados comportamentalmente e processados para os mesmos marcadores e nenhum sinal neuropatológico foi detectado. Em camundongos adultos infectados o ambiente padrão (APPyA), a atividade de burrowing diminuiu e se recuperou rapidamente (8-10DPI), a atividade de campo aberto (20-40DPI), mas manteve-se inalterado em animais da mesma idade e de ambiente enriquecido (AEPyA). Em contraste animais senis tanto de ambiente enriquecido (AEPyS) quanto de ambiente empobrecido (APPyS) reduzem significativamente a atividade de burrowing em todas janelas. A encefalite causada pelo virus Piry, induziu perdas olfativas transitórias em animais APPyA e AEPyA, mas permanents em APPyS e AEPyS. A imunomarcação para os antígenos viral do Piry atingiram seu pico no parênquima do SNC aos 5 e 6DPI e desapareceu aos 8DPI. Todas as reconstruções tridimensionais das micróglias, foram realizadas aos 8DPI. Mudanças Microgliais foram significativamente mais graves em camundongos adultos do que em camundongos senis, mas os animais AE parecem recuperar a morfologia microglia homeostática mais cedo do que os animais de AP. Os efeitos benéficos do AE foram menores em camundongos envelhecidos.

MgTiO3 (MTO) thin films were prepared by the polymeric precursor method with posterior spin-coating deposition. The films were deposited on Pt(111)/Ti/SiO2/Si(100) substrates and heat treated at 350 °C for 2 h and then heat treated at 400, 450, 500, 550, 600, 650 and 700 °C for 2 h. The degree of structural order−disorder, optical properties, and morphology of the MTO thin films were investigated by X-ray diffraction (XRD), micro-Raman spectroscopy (MR), ultraviolet− visible (UV−vis) absorption spectroscopy, photoluminescence (PL) measurements, and field-emission gun scanning electron microscopy (FEG-SEM) to investigate the morphology. XRD revealed that an increase in the annealing temperature resulted in a structural organization of MTO thin films. First-principles quantum mechanical calculations based on density functional theory (B3LYP level) were employed to study the electronic structure of ordered and disordered asymmetric models. The electronic properties were analyzed, and the relevance of the present theoretical and experimental results was discussed in the light of PL behavior. The presence of localized electronic levels and a charge gradient in the band gap due to a break in the symmetry are responsible for the PL in disordered MTO lattice.

In this paper, we report a detailed structural and electronic characterization of PbMoO4 crystals by using a conventional hydrothermal (CH) method. The samples were characterized by X-ray diffraction (XRD), Fourier transform Raman (FT-Raman), field-emission gun scanning electron microscopy (FEG-SEM) and photoluminescence (PL) measurements. In addition, first-principles quantum mechanical calculations based on the density functional theory were employed in order to understand the band structure and density of states for the PbMoO4. Analysis of both theoretical and experimental results allows to rationalize the role of order-disorder effects in the observed green PL emissions in these ordered powders.
