Mining and metallurgical industries are the most responsible for heavy metal contamination. These contaminants are often associated to poor management strategies and the lack of suitable containment areas. This is the case of one of the largest metal alloy producers in the São Francisco river basin, in Minas Gerais state, Brazil. During operation, the tailings of this activity have been accumulated in the surrounding area, leading to heavy metal contamination (Mn, Cu, Zn, Ni, Pb, Cd, Cr and As), much above the critical levels, which accumulate in the sediments of the nearby waterways, in the soils of the drainage area, and in the water column. In this work we are going to discuss both the contamination level and its extent in the Consciência river, a small tributary of the São Francisco river, the longest river entirely Brazilian, by analyzing the geochemistry and mineralogy of the accumulated sediments. It will be also discuss the reclamation strategies of this site and the impact on the environment.

The market for table grapes is moving into mass production of specialty seed-less grapes in covered areas, aiming at obtaining premium prices with early or late production of high quality products. Production of quality seedless grapes is not straightforward since it is requires the correct combination of various independent characteristics, such as color, sugars, size and quantity at the right moment for successful harvesting and marketing. The present study was carried out at the two largest Portuguese producers located in Alentejo, and has the objective of studying the effect of irrigation management strategies and two different soils on the various relevant parameters for successful production and marketing. The management strategies were the application of ten day stress at the end of the cycle, in order to promote early maturing of the grapes. Three different timings of the stress were applied. Soil moisture, sap flow, bark thickness, as well as leaf water potential, stomatal conductance and chlorophyll content were measured regularly during the production season. The results indicate that the roots explore a rather large soil volume and the plants can successfully withstand reasonable periods of drought without significant changes to the plant physiology. Additionally late rains can mask the effect of any farmer applied drought and invalidate any farmer induced stress to the plants. Water-logged soils tend to cause early onset of maturity, but cause the ripening stage to extend over a longer period of time, and thus, in effect result in a delay in the harvest date. Topography also has some effect on the ripening, since hot air tends to accumulate under the plastic at the higher areas of the field. A linha de investigação deste estudo é a ‘articulação da Investigação em Didáctica das Ciências e Práticas de Ensino dos Professores de Ciências’. O seu enquadramento teórico e metodológico inicial assentou nos estudos das áreas ‘Relações entre Investigação e as Práticas’ e ‘Avaliação da Formação Pós-Graduada – vertente impacte/articulação’. Inicialmente, fez-se uma análise histórico-epistemológica da Didáctica desde a sua génese até aos dias de hoje, para se compreender quer as raízes do gap entre académicos e práticos em geral, quer a crescente necessidade de articulação. Posteriormente, avançou-se para a primeira área, partindo da polémica despoletada por Hargreaves, ao defender que o ensino deveria ser uma profissão baseada na investigação. Em seguida, partiu-se de uma análise mais geral sobre a Investigação e as Práticas no contexto educacional em vários países antes se centrar especificamente no contexto da Didáctica das Ciências (impacte da IDC nas Práticas e constrangimentos na articulação). Analisou-se também brevemente as relações entre a IDC e Práticas no contexto da Formação de Professores, e não a área da Formação de Professores, para mantermos sempre o foco nas questões inerentes à articulação. Culminou-se na análise das culturas e epistemologias da acção e da investigação, com especial destaque para o conceito de professorinvestigador na actualidade e para a descrição das barreiras epistemológicas e ontológicas. Concluiu-se que as comunidades da investigação e da acção utilizavam o conceito ‘articulação’ indiscriminadamente como sinónimo de interacções, contacto, aproximação, impacte, etc., o que acabou esvaziando-o do seu verdadeiro significado. No que diz respeito à segunda área, a sua análise partiu da descrição da evolução de dez anos (1997-2007) de estudos sobre a Avaliação do Impacte dos CM nas práticas por ser considerada uma forma eficaz de articular as dimensões da Didáctica em direcção a um novo olhar sobre o conceito ‘articulação’. Além disso, apontou a dificuldade de se avaliar o impacte ao nível macro, por um lado, por não se tratar de uma prática investigativa institucionalizada no estatuto da carreira dos professores dos EB e ES e, por outro, por ainda colidir com diferentes concepções da natureza das investigações realizadas por Professores encontradas em ambas as comunidades, entendida ora como processo cognitivo (para o desenvolvimento profissional dos professores), ora como prática social (para construção de conhecimento no campo da Didáctica). Foram compiladas ainda as sugestões para se potenciar o impacte da IDC nas Práticas e/ou a articulação entre a IDC-Práticas em contexto formativo de diversos estudos avaliativos. Finalizou-se a análise chamando a atenção para cinco aspectos que ainda carecem de maior aprofundamento nesta área. Este longo enquadramento evidenciou a complexidade da problemática ‘articulação’ decorrente da interdependência das várias dimensões (epistemológica, política, ontológica, psicológica, ética, entre outras). Por exemplo, a ausência de consenso sobre critérios para a avaliação da qualidade da investigação produzida por professores (dimensões política e epistemológica) acaba, por vezes, por não conferir legitimidade às mesmas e por influenciar a legitimação pela comunidade académica, o que resulta na necessidade de diferenciação dos contributos e no maior afastamento entre as comunidades (dimensão ontológica), entre outros. Assim, optou-se por iniciar a análise do fenómeno ‘articulação entre IDCPráticas’ através dos primeiros modelos de articulação investigação-ensino, os quais visavam, contudo, fundamentalmente o impacte da IDC nas Práticas de Ensino das Ciências. Posteriormente, foram apresentadas as potencialidades da Avaliação ↔ Feedback, TIC e Colaboração (estratégias/métodos) para potenciar a articulação entre Investigação- Práticas. No que diz respeito à investigação empírica, realizou-se um estudo de caso descritivo e explorativo de natureza mista. O caso único, crítico e instrumental foi o fenómeno “articulação entre a IDC-Práticas na Formação Didáctica Pós- Graduada” no contexto da unidade curricular ‘Metodologia do Ensino da Física’ (MEF) do Curso de Mestrado em Ensino de Física. A técnica de análise geral utilizada foi a “descrição do caso” pelo facto de não se ter um referencial teórico especificamente sobre o caso. O caso contemplou três unidades de análise, a saber: Caracterização dos Professores-Formandos; Funcionamento da Unidade Curricular e Dinâmica dos currículos dos módulos articuladores. Estas unidades de análises permitiram evidenciar de que forma as características e/ou alterações implementadas na disciplina MEF contribuíram (ou podem contribuir) para a articulação da IDC-Práticas e descrever as dinâmicas do currículo (intencional – negociado – acção), evidenciando em que medida promoveram (ou inibiram) a articulação IDC – práticas. O estudo de caso aqui descrito revelou, ainda, a existência de dois níveis de articulação entre a Investigação e as Práticas no contexto formativo. O primeiro nível foi a articulação entre a Investigação sobre o Ensino Superior/Formação de Professores de Ciências (patente nas estratégias/métodos utilizados na disciplina) e a prática formativa dos IF no contexto da disciplina. O segundo nível centrou-se na articulação entre a Investigação sobre o Ensino não-Superior/Didáctica das Ciências e as práticas de Ensino das Ciências, base orientadora do currículo da disciplina aqui analisada, concretizado nos dois módulos articuladores descritos. Destacam-se algumas dimensões de análise descritas na presente investigação empírica, a saber: Utilização das TIC; Avaliação do Ensino baseada no feedback dos alunos; Avaliação Formativa das Aprendizagens e feedback; Trabalho de grupo realizado nos módulos articuladores; Currículo centrado na IDC; Currículo centrado na articulação da IDC-Práticas de Ensino das Ciências; Currículo centrado nas Práticas de Ensino das Ciências; Currículo centrado na articulação da Investigação-Práticas formativas e Currículo centrado nas Políticas Educativas. Relativamente a dinâmica dos currículos (intencional - negociado - acção) dos dois módulos articuladores, foram definidos quatro construtos (objectos de ensino, objectos de aprendizagem, objectivos de ensino e objectivos de aprendizagem) que culminaram na discussão de vários aspectos a serem considerados nos próximos cursos como, por exemplo: 1) Importância de o contrato didáctico prever a inclusão de objectos de aprendizagem; 2) Incompatibilidade do objectivo de aprendizagem ‘compreender a importância da IDC e a sua relevância para as práticas lectivas em contextos específicos’ num quadro formativo articulador; e 3) Importância de os cursos de formação de professores explicitarem quais ferramentas investigativas são necessárias à produção autónoma de conhecimento no contexto escolar e académico (mesmo que não sejam mobilizadas), de forma a que os professores possam delinear previamente planos individuais de formação/investigação. O estudo termina com a apropriação do modelo de articulação entre a Investigação Educacional e Práticas de McIntyre (2005) ao contexto da Didáctica das Ciências evidenciando uma relação dialógica com a investigação empírica. Apesar de este modelo priorizar a dimensão epistemológica (que aceita o gap pela impossibilidade epistemológica do seu total desaparecimento), na sua apropriação foi considerada a influência das outras dimensões. Esta apropriação assentou, portanto, numa visão moderada de articulação e na complexidade inerente à interdependência das dimensões. Foram propostos três caminhos epistemológicos complementares para a articulação entre a IDC-Práticas: 1º) Interacções entre Didáctica Investigativa – Didáctica Profissional; 2º) Utilização de estratégias na IDC especialmente desenhadas para informar as práticas de ensino; e 3º) Realização de IDC pela escola. Em cada um destes caminhos procurou-se enquadrar algumas sugestões e iniciativas já levadas a cabo para potenciar o impacte e/ou articulação e que se encontravam referenciadas na literatura em geral e no contexto português em particular. O primeiro caminho (composto por cinco etapas) evidenciou-se como aquele que leva a maior vantagem pelas inúmeras intervenções possíveis. A investigação empírica aqui apresentada enquadrou-se inclusivamente neste primeiro caminho pelo facto de ter sido uma iniciativa com a intencionalidade explícita de articular a Didáctica Investigativa e Profissional e por ter sido realizada no contexto da Formação Pós-Graduada (cenário considerado privilegiado para a promoção de interacções). Esta iniciativa foi realizada exclusivamente no âmbito curricular da Formação Pós-Graduada (Didáctica Curricular) e procurou articular as dimensões epistemológicas da Didáctica através da utilização de ‘mecanismos potencialmente articuladores’ (Avaliação - feedback, TIC e Colaboração). Foram descritas as quatro etapas deste primeiro caminho percorridas empiricamente com variações no grau de concretização, com excepção da quinta etapa ‘Investigação sobre a prática de ensino com generalização situada’ porque a vertente dissertativa do respectivo curso não fez parte do corpus. Assim, a articulação ocorreu fundamentalmente no nível epistemológico (currículo da disciplina). No que diz respeito ao 2º caminho, é aquele em que a comunidade académica mais tem investido, quer pelas críticas voltadas especificamente para a investigação, quer pelo sucesso na potenciação do impacte nas propostas até agora implementadas. Deve ser utilizado de forma complementar ao 1º, envolvendo, de preferência, os Professores que percorrem frequentemente o 1º caminho na sua prática diária. Esta condição justifica-se pela necessidade de se integrar legitimamente os professores nas equipas de investigação, aumentando concomitantemente a contribuição das Práticas para a construção de conhecimento no campo educacional. Finalmente, o 3º caminho é aquele que ainda não pode ser concretizado porque, para as Escolas serem diferentes das actuais na dimensão epistemológica (tornando-se produtoras de conhecimento didáctico), seriam necessárias medidas estruturais e articuladas nas várias dimensões anteriormente referidas. Entretanto, foram apontadas algumas soluções como, por exemplo, a utilização de investigações de generalização situada nas Escolas e a ligação das Escolas em redes. Estas investigações locais não substituiriam, mas mobilizariam a IDC produzida nas Universidades (centradas na construção do campo Didáctica das Ciências). Este caminho visionário culmina por um lado, com uma análise prospectiva assente na relação de complementaridade entre as evidências científicas e experienciais porque uma prática sem suporte investigativo é imprudente e uma investigação sem suporte experiencial é imatura. Por outro com uma constatação tardia (deveras reconfortante) que os estudos centrados na relação entre a Investigação e Práticas são estudos voltados para a Formação de Investigadores-Seniores por exigirem uma meta-reflexão da prática investigativa e do processo investigativo. As implicações do estudo são: (i) futuras iniciativas de articulação entre IDCPráticas; (ii) implementar e avaliar as sugestões advindas em novos contextos formativos; e (iii) na Educação a distância na área da Didáctica e Formação Didáctica de Professores. Assume-se a limitação estrutural da investigação resultante da alteração do projecto inicial que o restringiu a uma única etapa. Faz-se ainda uma reflexão do processo formativo-investigativo mediante a descrição dos constrangimentos de natureza interna e externa. Explicitam-se as limitações de carácter geral e específico e algumas tentativas de minimização dos respectivos efeitos no estudo. Finaliza-se o estudo com algumas sugestões de trabalhos futuros, a saber: (i) Continuidade dos estudos centrados na articulação entre IDC-Práticas; (ii) Continuidade dos estudos de Avaliação da Formação Pós-Graduada em termos de eficiência, eficácia, impacte e articulação; (iii) Análise da Epistemologia da Prática Docente em comunidades de práticas escolares; (iv) Articulação entre a Investigação sobre a Formação de Professores e as práticas dos formadores e futuros-formadores; e (v) Constituição de “Scholarship of teaching” na Formação de Professores.

Vitis vinifera L., the most widely cultivated fruit crop in the world, was the starting point for the development of this PhD thesis. This subject was exploited following on two actual trends: i) the development of rapid, simple, and high sensitive methodologies with minimal sample handling; and ii) the valuation of natural products as a source of compounds with potential health benefits. The target group of compounds under study were the volatile terpenoids (mono and sesquiterpenoids) and C13 norisoprenoids, since they may present biological impact, either from the sensorial point of view, as regards to the wine aroma, or by the beneficial properties for the human health. Two novel methodologies for quantification of C13 norisoprenoids in wines were developed. The first methodology, a rapid method, was based on the headspace solid-phase microextraction combined with gas chromatography-quadrupole mass spectrometry operating at selected ion monitoring mode (HS-SPME/GC-qMS-SIM), using GC conditions that allowed obtaining a C13 norisoprenoid volatile signature. It does not require any pre-treatment of the sample, and the C13 norisoprenoid composition of the wine was evaluated based on the chromatographic profile and specific m/z fragments, without complete chromatographic separation of its components. The second methodology, used as reference method, was based on the HS-SPME/GC-qMS-SIM, allowing the GC conditions for an adequate chromatographic resolution of wine components. For quantification purposes, external calibration curves were constructed with β-ionone, with regression coefficient (r2) of 0.9968 (RSD 12.51 %) and 0.9940 (RSD of 1.08 %) for the rapid method and for the reference method, respectively. Low detection limits (1.57 and 1.10 μg L-1) were observed. These methodologies were applied to seventeen white and red table wines. Two vitispirane isomers (158-1529 L-1) and 1,1,6-trimethyl-1,2-dihydronaphthalene (TDN) (6.42-39.45 μg L-1) were quantified. The data obtained for vitispirane isomers and TDN using the two methods were highly correlated (r2 of 0.9756 and 0.9630, respectively). A rapid methodology for the establishment of the varietal volatile profile of Vitis vinifera L. cv. 'Fernão-Pires' (FP) white wines by headspace solid-phase microextraction combined with comprehensive two-dimensional gas chromatography with time-of-flight mass spectrometry (HS-SPME/GCxGC-TOFMS) was developed. Monovarietal wines from different harvests, Appellations, and producers were analysed. The study was focused on the volatiles that seem to be significant to the varietal character, such as mono and sesquiterpenic compounds, and C13 norisoprenoids. Two-dimensional chromatographic spaces containing the varietal compounds using the m/z fragments 93, 121, 161, 175 and 204 were established as follows: 1tR = 255-575 s, 2tR = 0,424-1,840 s, for monoterpenoids, 1tR = 555-685 s, 2tR = 0,528-0,856 s, for C13 norisoprenoids, and 1tR = 695-950 s, 2tR = 0,520-0,960 s, for sesquiterpenic compounds. For the three chemical groups under study, from a total of 170 compounds, 45 were determined in all wines, allowing defining the "varietal volatile profile" of FP wine. Among these compounds, 15 were detected for the first time in FP wines. This study proposes a HS-SPME/GCxGC-TOFMS based methodology combined with classification-reference sample to be used for rapid assessment of varietal volatile profile of wines. This approach is very useful to eliminate the majority of the non-terpenic and non-C13 norisoprenic compounds, allowing the definition of a two-dimensional chromatographic space containing these compounds, simplifying the data compared to the original data, and reducing the time of analysis. The presence of sesquiterpenic compounds in Vitis vinifera L. related products, to which are assigned several biological properties, prompted us to investigate the antioxidant, antiproliferative and hepatoprotective activities of some sesquiterpenic compounds. Firstly, the antiradical capacity of trans,trans-farnesol, cis-nerolidol, α-humulene and guaiazulene was evaluated using chemical (DPPH• and hydroxyl radicals) and biological (Caco-2 cells) models. Guaiazulene (IC50= 0.73 mM) was the sesquiterpene with higher scavenger capacity against DPPH•, while trans,trans-farnesol (IC50= 1.81 mM) and cis-nerolidol (IC50= 1.48 mM) were more active towards hydroxyl radicals. All compounds, with the exception of α-humulene, at non-cytotoxic levels (≤ 1 mM), were able to protect Caco-2 cells from oxidative stress induced by tert-butyl hydroperoxide. The activity of the compounds under study was also evaluated as antiproliferative agents. Guaiazulene and cis-nerolidol were able to more effectively arrest the cell cycle in the S-phase than trans,trans-farnesol and α-humulene, being the last almost inactive. The relative hepatoprotection effect of fifteen sesquiterpenic compounds, presenting different chemical structures and commonly found in plants and plant-derived foods and beverages, was assessed. Endogenous lipid peroxidation and induced lipid peroxidation with tert-butyl hydroperoxide were evaluated in liver homogenates from Wistar rats. With the exception of α-humulene, all the sesquiterpenic compounds under study (1 mM) were effective in reducing the malonaldehyde levels in both endogenous and induced lipid peroxidation up to 35% and 70%, respectively. The developed 3D-QSAR models, relating the hepatoprotection activity with molecular properties, showed good fit (R2LOO > 0.819) with good prediction power (Q2 > 0.950 and SDEP < 2%) for both models. A network of effects associated with structural and chemical features of sesquiterpenic compounds such as shape, branching, symmetry, and presence of electronegative fragments, can modulate the hepatoprotective activity observed for these compounds. In conclusion, this study allowed the development of rapid and in-depth methods for the assessment of varietal volatile compounds that might have a positive impact on sensorial and health attributes related to Vitis vinifera L. These approaches can be extended to the analysis of other related food matrices, including grapes and musts, among others. In addition, the results of in vitro assays open a perspective for the promising use of the sesquiterpenic compounds, with similar chemical structures such as those studied in the present work, as antioxidants, hepatoprotective and antiproliferative agents, which meets the current challenges related to diseases of modern civilization.

Os incêndios florestais, associados ao abandono do espaço rural, a pequena propriedade florestal e o desinteresse e o absentismo dos proprietários florestais têm sido apontados como fatores que têm afetado a sustentabilidade das florestas em Portugal. Apesar da formulação de políticas e de instrumentos de planeamento e de gestão florestal para lidar com estes constrangimentos, são ainda escassos os progressos para uma Gestão Florestal Sustentável. A nível internacional e europeu, a participação dos agentes já representa um aspeto-chave no processo de definição e de implementação de estratégias que promovam a multifuncionalidade da floresta, mas também se adeqúem às necessidades e aos interesses dos agentes locais. A temática da tese esteve focada nesta discussão, argumentando que existe uma escassa participação dos agentes nos processos de tomada de decisão relativos ao setor florestal. O principal objetivo da investigação foi o desenvolvimento de uma metodologia participativa para a discussão e negociação de estratégias locais para a Gestão Florestal Sustentável, que maximizem o potencial produtivo e o papel sócio-ambiental das florestas, diminuam o risco de incêndio e promovam o crescente interesse e participação dos agentes locais na gestão florestal. A tese está estruturada em três partes. A primeira parte apresenta uma avaliação do sector florestal nacional, com base numa revisão bibliográfica e numa comparação de indicadores, políticas e instrumentos de planeamento e gestão florestal (Capítulo 2) e com base num estudo de perceção social desenvolvido numa área de estudo localizada na região Centro de Portugal. Este estudo analisa as perceções técnicas (decisores políticos e técnicos) e sociais (proprietários florestais e outros membros da comunidade local) sobre as florestas, a gestão florestal e os incêndios florestais (Capítulo 4). As ‘Zonas de Intervenção Florestal’, enquanto ferramenta recente para a cooperação e organização dos proprietários e produtores florestais, foram também analisadas (Capítulo 3). A segunda parte da tese é dedicada à análise de processos de participação pública, com base numa revisão bibliográfica sobre os benefícios, níveis, abordagens e métodos de participação (Capítulo 5) e numa avaliação de processos de participação pública desenvolvidos em Portugal (Capítulo 6). A terceira parte da tese foca-se no desenho e no teste da metodologia participativa proposta no âmbito desta tese (Capítulo 7) e na formulação de algumas orientações para melhoria dos processos participativos na gestão florestal (Capítulo 8). Os resultados confirmaram a centralidade dos incêndios florestais e dos fatores associados ao contexto socioeconómico (e.g. despovoamento e envelhecimento populacional, absentismo, falta de gestão florestal, estrutura fundiária) como os principais problemas que afetam a floresta na região Centro de Portugal. A organização e cooperação dos proprietários florestais emergiu como solução possível para lidar com estas ameaças e promover a multifuncionalidade da floresta, sendo essencial aumentar o conhecimento e a participação dos agentes nas decisões associadas à floresta. É proposta uma ferramenta para esta participação, centrada no contexto local e facilmente utilizável por todos os agentes. A implementação da metodologia participativa revelou o seu potencial no desenvolvimento de uma participação equitativa e inclusiva dos múltiplos agentes.

O Design Social assume-se cada vez mais como ferramenta de fortalecimento social e como uma arma na luta contra a fealdade do mundo, o inútil, o disfuncional e outros aspectos da desumanização. Mas não basta só produzir produtos para as pessoas, é preciso produzir com as pessoas. Após uma intervenção institucional da Universidade de Aveiro, em parceria com a Vista Alegre, sobre responsabilidade social corporativa e uma análise do caso paradigmático da Fiskers Village, desenhámos um modelo orientador para o Design Social e definimos directivas para intervir junto da comunidade local da Serra dʼArga, em Portugal: (1) o design como método de acção (2) do local para o global, (3) que identifica, partilha e intervém (4) com a comunidade, (5) em modo sistémico e com base no ciclo de vida, (6) visando construir para a integração máxima nos ciclos da natureza. Estas directivas serviram de pano de fundo à nossa intervenção e deram origem a uma oficina que pretendeu associar design, craft e natureza. Aí foram criados produtos e serviços sustentáveis que ligam produtores e consumidores ao território, sugerem uma outra forma de ocupação do tempo, e, finalmente, ajudam a comunidade a repensar a sua vida social e a sua relação com a floresta.

The sustainable viticulture of a region passes, among other aspects, for maximizing the varieties potential minimizing subsequent interventions during winemaking, which should contribute to the production of quality wines maintaining their typicity and rationalizing costs. The detailed knowledge of each Appellation specificities, namely vineyard parcel (soil type and topographical peculiarities) and harvest climatic conditions is crucial for sustainability in this sector. Thus, in line with this current trend, the starting point for the development of this PhD thesis was to evaluate the oenological potential of different varieties cultivated throughout Bairrada Appellation (Portugal). During maturation several changes in grape varieties occur, namely berries become sweeter, less acidic, and they develop flavour, aroma and colour properties. The development of these characteristics is essential to define grapes oenological potential, i.e. to estimate the possibility of their usage to produce specific wines. A three years sampling plan was designed to evaluate the effect of harvest year and parcel characteristics on V. vinifera cv. Arinto, Bical, Sauvignon Blanc, Baga, Castelão, Touriga Nacional, and Sousão grapes composition. For each variety, 3 parcels with different characteristics were selected. Several physicochemical parameters were evaluated, during maturation: berry weight, pH, titratable acidity, sugar and phenolic contents, antiradical activity, and volatile composition (free fraction). Special attention was devoted to grapes at technologic maturity, since, besides these parameters, glycosidically-linked fraction was also considered. By using the results obtained at technologic maturity, a comprehensive approach was applied to identify the significance of harvest and parcel characteristics effects on each variety composition. Considering all the parameters under study, it may be highlighted some significant differences. According to the obtained results determined during maturation, it was possible to conclude that Arinto, Bical and Sauvignon Blanc grapes from parcels with clay-sandy and clay-calcareous soils have higher phenolic content and antiradical activity. Otherwise, Sauvignon Blanc presented similar volatile composition for grapes cultivated in the 3 parcels, while Arinto and Bical exhibited higher volatile content in grapes from claysandy and clay-calcareous soils. For Baga, Castelão and Touriga Nacional red varieties, grapes with higher phenolic content, antiradical activity, and volatile content were obtained from clayey and clay-calcareous soils. Furthermore, for Touriga Nacional, parcels altitude seems also to modulate grapes composition. Beyond parcel effect, harvest year conditions also influence grapes composition: 2011 harvest was related with lower phenolic and volatile contents, as well as lower antiradical activity.For grapes collected at technologic maturity, analysis of variance-simultaneous component analysis (ASCA) was applied combining all the parameters under study, in order to assess the influence of harvest and parcel characteristics on each variety oenological potential. The results obtained using this comprehensive approach is closely related with those observed during maturation and revealed that harvest was the main factor that influenced grapes composition (53% to 68% of the total data set variance) followed by parcel characteristics, explaining ca. 15-19% of the total data set variance. The oenological potential of each variety may be different from one parcel to another, i.e., clay-sandy and clay-calcareous related-environments seem to favour Arinto and Bical white grapes composition, but for the red varieties, grapes composition was favoured by clayey and clay-calcareous soils. Besides, also higher altitude seems to favour Touriga Nacional grapes composition. Sauvignon Blanc seems to be a variety well adapted to the different parcel characteristics. In order to go forward in the valuation of these varieties, the aroma properties of 6 monovarietal wines were studied based on an aroma network-approach, linking molecular data related to volatile composition and aroma data about the key odor active molecules. This approach allowed to identify different wine aroma properties and to infer about the consumer’s sensory perception. It was found that aroma properties differ from one wine variety to another: while Arinto and Sauvignon Blanc wine exhibited higher tree fruity, sweety and flowery aromas, related essentially with ester compounds and C13 norisoprenoids, the opposite was obtained for Bical wine, corroborating the aroma sensory perceptions of the trained panel. Sauvignon Blanc also exhibited higher toasted aromas (related with thiols, mainly with 2-methyl-3-furanthiol). Touriga Nacional red wine exhibited higher tree, tropical, and berry fruits notes (sensory described as sweet fruits), toasted and flowery aromas, while these are similar for the other red wines under study. Besides Portuguese Bairrada wines, this aroma network approach is a tool that can be used to explain the aroma properties of wines worldwide. The grape and wine data generated under the present PhD thesis, in the context of Bairrada Appellation, shows the unique character of each variety, and may be used by growers and wine producers as a support for decisionmaking based on objective criteria, increasing the sustainability in this sector. For instance, it is possible to take advantage of the natural resources and produce products with different characteristics obtained from the same variety, minimizing costs during the winemaking process.

Last-resort antibiotics are the final line of action for treating serious infections caused by multiresistant strains. Over the years the prevalence of resistant bacteria has been increasing. Natural environments are reservoirs of antibiotic resistance, highly influenced by human-driven activities. The importance of aquatic systems on the evolution of antibiotic resistance is highlighted from the assumption that clinically-relevant resistance genes have originated in strains ubiquitous in these environments. We hypothesize that: a) rivers are reservoirs and disseminators of antibiotic resistance; b) anthropogenic activities potentiate dissemination of resistance to last-resort antibiotics. Hence, the main goal of the work is to compare the last-resort antibiotics resistome, in polluted and unpolluted water. Rivers from the Vouga basin, exposed to different anthropogenic impacts, were sampled. Water quality parameters were determined to classify rivers as unpolluted or polluted. Two bacterial collections were established enclosing bacteria resistant to cefotaxime (3rd generation cephalosporin) and to imipenem (carbapenem). Each collection was characterized regarding: phylogenetic diversity, antibiotic susceptibility, resistance mechanisms and mobile genetic elements. The prevalence of cefotaxime- and imipenem-resistant bacteria was higher in polluted water. Results suggested an important role in the dissemination of antibiotic resistance for Enterobacteriaceae, Pseudomonas and Aeromonas. The occurrence of bacteria resistant to non-beta-lactams was higher among isolates from polluted water as also the number of multiresistant strains. Among strains resistant to cefotaxime, extended-spectrum beta-lactamase (ESBL) genes were detected (predominantly blaCTX-M-like) associated to mobile genetic elements previously described in clinical strains. ESBL-producers were often multiresistant as a result of co-selection mechanisms. Culture-independent methods showed clear differences between blaCTX-M-like sequences found in unpolluted water (similar to ancestral genes) and polluted water (sequences identical to those reported in clinical settings). Carbapenem resistance was mostly related to the presence of intrinsically resistant bacteria. Yet, relevant carbapenemase genes were detected as blaOXA-48-like in Shewanella spp. (the putative origin of these genes), and blaVIM-2 in Pseudomonas spp. isolated from polluted rivers. Culture-independent methods showed an higher than the previously reported diversity of blaOXA-48-like genes in rivers. Overall, clear differences between polluted and unpolluted systems were observed, regarding prevalence, phylogenetic diversity and susceptibility profiles of resistant bacteria and occurrence of clinically relevant antibiotic resistance genes, thus validating our hypotheses. In this way, rivers act as disseminators of resistance genes, and anthropogenic activities potentiate horizontal gene transfer and promote the constitution of genetic platforms that combine several resistance determinants, leading to multiresistance phenotypes that may persist even in the absence of antibiotics.

During the late twentieth century the supply chains for gold were considered by the majority of consumers (when they were considered at all) to be driven by simple commercial imperatives. That notion was shattered during the first decade of the twenty-first century by the appearance of ethical campaigns, led by advocates determined to present major players in the gold industry as morally reprehensible. The ‘No Dirty Gold’ campaign sought to shift the purchasing of gold onto a moral register, in order to challenge the activities of large mining corporations. It was followed by the Fairtrade Foundation’s ‘Fairtrade Gold’ initiative, which had aspirations to support subsistence mining communities at the expense of big business. By directly targeting a luxury material and playing on its inherent social ambiguities, campaigners hoped to thoroughly moralise the purchasing of gold objects. Dr Oakley’s presentation will examine the forces behind this developing social phenomenon, describe the trajectories of a selection of major campaigns, and consider the extent to which these have impacted on public attitudes, gold miners and the actions of consumers, producers and retailers of luxury goods.

The undesirable enrichment of water by nutrients may be a problem, especially in areas with restricted exchange with the sea. The tidal regime flushes the system and contributes for the removal of phytoplankton, favouring phytobenthos as the target of enhanced nutrients. Water samples were collected during the years of 2006 and 2007-08 for nutrients, chlorophyll a and dissolved oxygen. Sediment sample s were also collected for pore water nutrients and benthic chlorophyll a. From comparison with previous work, a decrease in the nitrogen concentration in the water column can be pointed out, which may indicate an improvement of the water quality. Pore water DAIN represents approximately 75% of the total DAIN of the whole lagoon. Benthic chlorophyll a concentrations were much larger than in the water column, representing around 99% of the total chlorophyll existent in the lagoon. Benthic microalgae play a relevant role in this system and therefore standard monitoring programs of the WFD, which do not consider this component, may fail to track nutrient-driven changes in primary producers. Dissolved oxygen concentration could be near critical levels during the summer (early in the morning), especially in the inner channels.

Cyanobacteria are a recognised public health hazard, because the majority of species is able to produce toxins. The monitoring is usually restricted to freshwater environments, like lakes or dams used for water supply. Cyanobacteria blooms have been regularly reported in the Guadiana River and high estuary and most of the observed cyanobacteria were toxin producers.

Infectious diseases often hamper the production of aquatic organisms in aquaculture systems, causing economical losses, environmental problems and consumer safety issues. The conventional way aquaculture producers had to control pathogens was by means of synthetic antibiotics and chemicals. This procedure had consequences in the emergence of more resilient pathogens, drug contamination of seafood products and local ecosystems. To avoid the repercussions of antibiotic use, vaccination has greatly replaced human drugs in western fish farms. However there is still massive unregulated antibiotic use in third world fish farms, so less expensive therapeutic alternatives for drugs are desperately needed. An alternative way to achieve disease control in aquaculture is by using natural bioactive organic compounds with antibiotic, antioxidant and/or immunostimulant properties. Such diverse biomolecules occur in bacteria, algae, fungi, higher plants and other organisms. Fatty acids, nucleotides, monosaccharides, polysaccharides, peptides, polyphenols and terpenoids, are examples of these substances. One promising source of bioactive compounds are salt tolerant plants. Halophytes have more molecular resources and defence mechanisms, when compared with other tracheophytes, to deal with the oxidative stresses of their habitat. Many halophytes have been used as a traditional food and medical supply, especially by African and Asian cultures. This scientific work evaluated the antibiotic, antioxidant, immunostimulant and metal chelating properties of Atriplex halimus L., Arthrocnemum macrostachyum Moric., Carpobrotus edulis L., Juncus acutus L. and Plantago coronopus L., from the Algarve coast. The antibiotic properties were tested against Listonella anguillarum, Photobacterium damselae piscicida and Vibrio fischeri. The immunostimulant properties were tested with cytochrome c and Griess assays on Sparus aurata head-kidney phagocytes. J. acutus ether extract inhibited the growth of P. damselae piscicida. A. macrostachyum, A. halimus, C. edulis, Juncus acutus and P. coronopus displayed antioxidant, copper chelating and iron chelating properties. These plants show potential as sources of bioactive compounds with application in aquaculture and in other fields.

Every can of tuna purchased by the consumer has taken a long journey before reaching the supermarket shelves. For each can bought there is a lengthy process from sea to shelf. A large proportion of the tuna cans purchased in the European Union come all the way from West Africa; a developing region with a high dependency on fisheries. Amidst an ever-increasing demand for tuna products the global tuna fisheries are set to continue expanding, apparently one of the last natural resource based industries fit to do so in West Africa. Tuna is the biggest fisheries export and dominates the fisheries sector in Ghana, a country situated in West Africa. This thesis aims to understand how this globally important industrial fisheries functions in terms of procedures, practices, Governance and finance. Socioeconomic influences, in the setting of a developing country, were also examined. For these purposes a Value Chain Analysis was employed. A Value Chain Analysis is a tool commonly used to understand how different companies and organizations participate in a domestic policy environment, which directs conclusion in the global economy. This analysis has the potential to allow researchers to fully understand a commodity chain and hence identify realistic opportunities for consequential improvements. Interviews and questionnaires were employed in-field Ghana along with secondary data collection techniques. It was found that the fisheries functions at the production level under influences from large multinational companies and tends to operate with a certain degree of lawlessness. Governance over the value chain is well defined, however implementation is poor or non-existent. The processors, whom are also dominated by multinationals, exert some control over the producers and their sales, however the high value links which are highlighted occur at the retail stage. Socioeconomic dynamics acting in the chain included the lack of communication between the public and private sector, power imbalances amongst players at production, the role of local businesswomen as actors in the chain and the general characteristics of the workers in the industry. Value addition and upgrading are needed the most in Governance over the chain, especially within Monitoring, Control and Surveillance. The results of the study provide a wealth of material about the components of a cost-heavy fishing industry in a developing country; an industry on which many eyes have recently turned due to illegal fishing activities. It highlights clearly where funding and future focus are needed. This value chain can be used as a guide for those that need to comprehend the financial complexities and real life dynamics of the Ghanaian tuna fishing industry today.

This article aims to approach the Wine Route in Algarve, while fueling the Enotourism in the Region. In methodological therms, secondary sources were consulted, including literature considered relevant, as well the analysis of the case Tenerife, as a destination where Enotourism was successfully implemented. The primary sources used for the case study were based on an analysis of interviews to the producers associates to the Algarve Wine Route (AWR) and an interview with the head of AWR, which led to the view and vision of the leaders in the implementation of this project. In order to give greater depth to the theme addressed the issue of external promotion of wine tourism, verifying which markets to consider in attracting tourists for using such statistics and information listed on the Portuguese Tourism Website. The main results point to the need for continuity the consolidation and reinforcing the AWR, increasing the collaboration between partners and among economic actors. In the opinion of the respondents, the external markets are the major destination for both the flow of wine production, as for attracting new customers and as such should be treated as strategic markets.
