Pain transmission at the spinal cord is modulated by descending actions that arise from supraspinal areas which collectively form the endogenous pain control system. Two key areas involved of the endogenous pain control system have a circunventricular location, namely the periaqueductal grey (PAG) and the locus coeruleus (LC). The PAG plays a crucial role in descending pain modulation as it conveys the input from higher brain centers to the spinal cord. As to the LC, it is involved in descending pain inhibition by direct noradrenergic projections to the spinal cord. In the context of neurological defects, several diseases may affect the structure and function of the brain. Hydrocephalus is a congenital or acquired disease characterized by an enlargement of the ventricles which leads to a distortion of the adjacent tissues, including the PAG and LC. Usually, patients suffering from hydrocephalus present dysfunctions in learning and memory and also motor deficits. It remains to be evaluated if lesions of the periventricular brain areas involved in pain control during hydrocephalus may affect descending pain control and, herein, affect pain responses. The studies included in the present thesis used an experimental model of hydrocephalus (the rat injected in the cisterna magna with kaolin) to study descending modulation of pain, focusing on the two circumventricular regions referred above (the PAG and the LC). In order to evaluate the effects of kaolin injection into the cisterna magna, we measured the degree of ventricular dilatation in sections encompassing the PAG by standard cytoarquitectonic stanings (thionin staining). For the LC, immunodetection of the noradrenaline-synthetizing enzyme tyrosine hydroxylase (TH) was performed, due to the noradrenergic nature of the LC neurons. In general, rats with kaolin-induced hydrocephalus presented a higher dilatation of the 4th ventricle, along with a tendency to a higher area of the PAG. Due to the validated role of detection the c-fos protooncogene as a marker of neuronal activation, we also studied neuronal activation in the several subnuclei which compose the PAG, namely the dorsomedial, dorsolateral, lateral and ventrolateral (VLPAG) parts. A decrease in the numbers of neurons immunoreactive for Fos protein (the product of activation of the c-fos protooncogene) was detected in rats injected with kaolin, whereas the remaining PAG subnuclei did not present changes in Fos-immunoreactive nuclei. Increases in the levels of TH in the LC, namely at the rostral parts of the nucleus, were detected in hydrocephalic animals. The following pain-related parameters were measured, namely 1) pain behavioural responses in a validated pain inflammatory test (the formalin test) and 2) the nociceptive activation of spinal cord neurons. A decrease in behavioral responses was detected in rats with kaolin-induced hydrocephalus was detected, namely in the second phase of the test (inflammatory phase). This is the phase of the formalin test in which the motor behaviour is less important, which is important since a semi-quantitative analysis of the motor performance of rats injected with kaolin indicates that these animals may present some motor impairments. Collectively, the results of the behavioral studies indicate that rats with kaolin-induced hydrocephalus exhibit hypoalgesia. A decrease in Fos expression was detected at the superficial dorsal layers of the spinal cord in rats with kaolin-induced hydrocephalus, further indicating that hydrocephalus decreases nociceptive responses. It remains to be ascertained if this is due to alterations in the PAG and LC in the rats with kaolin-induced hydrocephalus, which may affect descending pain modulation. It remains to be evaluated what are the mechanisms underlying the increased pain inhibition at the spinal dorsal horn in the hydrocephalus rats. Regarding the VLPAG, the decrease in neuronal activity may impair descending modulation. Since the LC has higher levels of TH in rats with kaolininduced hydrocephalus, which also appears to increase the noradrenergic innervation in the spinal dorsal horn, it is possible that an increase in the release of noradrenaline at the spinal cord accounts for pain inhibition. Our studies also determine the need to study in detail patients with hydrocephalus namely in what concerns their thresholds to pain and to perform imaging studies focused on the structure and function of pain control areas in the brain.

Uma nova área tecnológica está em crescente desenvolvimento. Esta área, denominada de internet das coisas, surge na necessidade de interligar vários objetos para uma melhoria a nível de serviços ou necessidades por parte dos utilizadores. Esta dissertação concentra-se numa área específica da tecnologia internet das coisas que é a sensorização. Esta rede de sensorização é implementada pelo projeto europeu denominado de Future Cities [1] onde se cria uma infraestrutura de investigação e validação de projetos e serviços inteligentes na cidade do Porto. O trabalho realizado nesta dissertação insere-se numa das plataformas existentes nessa rede de sensorização: a plataforma de sensores ambientais intitulada de UrbanSense. Estes sensores ambientais que estão incorporados em Data Collect Unit (DCU), também denominados por nós, medem variáveis ambientais tais como a temperatura, humidade, ozono e monóxido de carbono. No entanto, os nós têm recursos limitados em termos de energia, processamento e memória. Apesar das grandes evoluções a nível de armazenamento e de processamento, a nível energético, nomeadamente nas baterias, não existe ainda uma evolução tão notável, limitando a sua operacionalidade [2]. Esta tese foca-se, essencialmente, na melhoria do desempenho energético da rede de sensores UrbanSense. A principal contribuição é uma adaptação do protocolo de redes Ad Hoc OLSR (Optimized Link State Routing Protocol) para ser usado por nós alimentados a energia renovável, de forma a aumentar a vida útil dos nós da rede de sensorização. Com esta contribuição é possível obter um maior número de dados durante períodos de tempo mais longos, aproximadamente 10 horas relativamente às 7 horas anteriores, resultando numa maior recolha e envio dos mesmos com uma taxa superior, cerca de 500 KB/s. Existindo deste modo uma aproximação analítica dos vários parâmetros existentes na rede de sensorização. Contudo, o aumento do tempo de vida útil dos nós sensores com recurso à energia renovável, nomeadamente, energia solar, incrementa o seu peso e tamanho que limita a sua mobilidade. Com o referido acréscimo a determinar e a limitar a sua mobilidade exigindo, por isso, um planeamento prévio da sua localização. Numa primeira fase do trabalho analisou-se o consumo da DCU, visto serem estes a base na infraestrutura e comunicando entre si por WiFi ou 3G. Após uma análise dos protocolos de routing com iv suporte para parametrização energética, a escolha recaiu sobre o protocolo OLSR devido à maturidade e compatibilidade com o sistema atual da DCU, pois apesar de existirem outros protocolos, a implementação dos mesmos, não se encontram disponível como software aberto. Para a validação do trabalho realizado na presente dissertação, é realizado um ensaio prévio sem a energia renovável, para permitir caracterização de limitações do sistema. Com este ensaio, tornou-se possível verificar a compatibilidade entre os vários materiais e ajustamento de estratégias. Num segundo teste de validação é concretizado um ensaio real do sistema com 4 nós a comunicar, usando o protocolo com eficiência energética. O protocolo é avaliado em termos de aumento do tempo de vida útil do nó e da taxa de transferência. O desenvolvimento da análise e da adaptação do protocolo de rede Ad Hoc oferece uma maior longevidade em termos de tempo de vida útil, comparando ao que existe durante o processamento de envio de dados. Apesar do tempo de longevidade ser inferior, quando o parâmetro energético se encontra por omissão com o fator 3, a realização da adaptação do sistema conforme a energia, oferece uma taxa de transferência maior num período mais longo. Este é um fator favorável para a abertura de novos serviços de envio de dados em tempo real ou envio de ficheiros com um tamanho mais elevado.

RESUMO: Promover a qualidade de vida no envelhecimento implica responder às necessidades de cuidados dos mais velhos. Actualmente, muitos idosos permanecem no seu domicílio, o que exige a prestação adequada de cuidados informais e/ou formais nesse contexto. O presente estudo teve como objectivo identificar e comparar as necessidades de cuidados de utentes de um centro de dia e de um serviço de apoio domiciliário numa Instituição Particular de Solidariedade Social. A amostra foi constituída por 54 utentes idosos e pelos seus cuidadores informais. Os utentes foram avaliados com recurso a: Camberwell Assessment of Need for the Elderly, Geriatric Depression Scale, Mini Mental State Examination, Índice de Barthel e Escala de Lawton e Brody para as actividades de vida diária. Os respectivos cuidadores informais foram avaliados quanto a necessidades de cuidados e a sobrecarga familiar. As necessidades de cuidados mais frequentes na amostra estavam relacionadas com alimentação, companhia, saúde física e actividades diárias. Parte das necessidades estavam cobertas. Porém, muitas necessidades psicológicas e sociais, nomeadamente actividades diárias, companhia e memória, não estavam cobertas, em particular nos utentes de apoio domiciliário. Tal como esperado, foi encontrado um maior número de necessidades de cuidados em situações de dependência e de doenças neuropsiquiátricas. Comparativamente com os utentes de centro de dia, os utentes de apoio domiciliário apresentaram números superiores de necessidades de cuidados, necessidades não cobertas. Uma melhor identificação das necessidades de cuidados e dos factores associados a estas poderá ajudar a delinear intervenções adequadas em centros de dia e em serviços de apoio domiciliário.------------ABSTRACT: To promote quality of life in aging means responding to the health and social needs of older people. Today, elderly people tend to stay at home until later stages of disease, which requires the provision of adequate informal care, formal care or both. This study aimed to identify and compare the needs for care of users of a day centre and a domiciliary care service, in a nonprofit organization in Portugal. The sample consisted of 54 elderly users and of their informal caregivers. The users were assessed using the Camberwell Assessment of Need for the Elderly (CANE), the Geriatric Depression Scale, the Mini Mental State Examination, the Barthel Index and the Lawton and Brody Scale for activities of daily living. Informal caregivers were assessed using the CANE and the Zarit Burden Interview. The more frequent care needs were related to food, company, physical health and daytime activities. A proportion of needs were met. However, many psychological and social needs were unmet, namely daytime activities, company and memory, and this was so in particular concerning domiciliary care users.As expected, a higher number of needs was related to dependency and the presence of neuropsychiatric conditions. The domiciliary care users had more total needs and more unmet needs when compared with day centre users. The identification of needs for care and their associated factors can help in the planning of appropriate interventions in day centres and domiciliary care services.

The amorphous silicon photo-sensor studied in this thesis, is a double pin structure (p(a-SiC:H)-i’(a-SiC:H)-n(a-SiC:H)-p(a-SiC:H)-i(a-Si:H)-n(a-Si:H)) sandwiched between two transparent contacts deposited over transparent glass thus with the possibility of illumination on both sides, responding to wave-lengths from the ultra-violet, visible to the near infrared range. The frontal il-lumination surface, glass side, is used for light signal inputs. Both surfaces are used for optical bias, which changes the dynamic characteristics of the photo-sensor resulting in different outputs for the same input. Experimental studies were made with the photo-sensor to evaluate its applicability in multiplexing and demultiplexing several data communication channels. The digital light sig-nal was defined to implement simple logical operations like the NOT, AND, OR, and complex like the XOR, MAJ, full-adder and memory effect. A pro-grammable pattern emission system was built and also those for the validation and recovery of the obtained signals. This photo-sensor has applications in op-tical communications with several wavelengths, as a wavelength detector and to execute directly logical operations over digital light input signals.

O presente estudo tem como objetivo compreender a valoração de uma mensagem de correio eletrónico em ambiente corporativo, e respetiva integração no arquivo empresarial. Objetivamos, nos primeiros capítulos, descrever o modelo metodológico aplicado. Seguidamente, apresentamos uma revisão bibliográfica assente em literatura inerente à temática dos circuitos informacionais na empresa, e documentação eletrónica resultante dos mesmos, com ênfase nas mensagens de correio eletrónico. A investigação empírica teve por base a aplicação de um inquérito por questionário, tendo por amostra um grupo profissional pré-selecionado, com acesso regular a um sistema de correio eletrónico. Os resultados obtidos através da análise do material empírico permitem-nos observar uma utilização preferencial do correio eletrónico face a outros suportes comunicacionais, constituindo-se as mensagens eletrónicas no documento produzido em maior escala, em ambiente profissional. Foram igualmente verificados casos de ausência de regulamentação para criação, gestão ou arquivo de mensagens, ou mesmo de um sistema de gestão documental que permita integração das mesmas. Com base na análise empírica, e em associação com a revisão bibliográfica explanada, concluímos que estas, e outras questões enunciadas ao longo do estudo, poderão constituir-se num sério obstáculo à gestão, preservação, recuperação e integridade da informação registada por meio de mensagens de correio eletrónico, caso não sejam criados e aplicados procedimentos e sistemas de gestão orientados para a documentação eletrónica, de forma a garantir o acesso a informação de fulcral importância para empresa, enquanto evidência e memória dos processos e atividades organizacionais.

Since its introduction 16 years ago, the astrocyte-neuron lactate shuttle (ANLS) model has profoundly modified our understanding of neuroenergetics by bringing a cellular and molecular resolution. Praised or disputed, the concept has never ceased to attract attention, leading to critical advances and unexpected insights. Here, we summarize recent experimental evidence further supporting the main tenets of the model. Thus, evidence for distinct metabolic phenotypes between neurons (mainly oxidative) and astrocytes (mainly glycolytic) have been provided by genomics and classical metabolic approaches. Moreover, it has become clear that astrocytes act as a syncytium to distribute energy substrates such as lactate to active neurones. Glycogen, the main energy reserve located in astrocytes, is used as a lactate source to sustain glutamatergic neurotransmission and synaptic plasticity. Lactate is also emerging as a neuroprotective agent as well as a key signal to regulate blood flow. Characterization of monocarboxylate transporter regulation indicates a possible involvement in synaptic plasticity and memory. Finally, several modeling studies captured the implications of such findings for many brain functions. The ANLS model now represents a useful, experimentally based framework to better understand the coupling between neuronal activity and energetics as it relates to neuronal plasticity, neurodegeneration, and functional brain imaging.

The influence of proximal olfactory cues on place learning and memory was tested in two different spatial tasks. Rats were trained to find a hole leading to their home cage or a single food source in an array of petri dishes. The two apparatuses differed both by the type of reinforcement (return to the home cage or food reward) and the local characteristics of the goal (masked holes or salient dishes). In both cases, the goal was in a fixed location relative to distant visual landmarks and could be marked by a local olfactory cue. Thus, the position of the goal was defined by two sets of redundant cues, each of which was sufficient to allow the discrimination of the goal location. These experiments were conducted with two strains of hooded rats (Long-Evans and PVG), which show different speeds of acquisition in place learning tasks. They revealed that the presence of an olfactory cue marking the goal facilitated learning of its location and that the facilitation persisted after the removal of the cue. Thus, the proximal olfactory cue appeared to potentiate learning and memory of the goal location relative to distant environmental cues. This facilitating effect was only detected when the expression of spatial memory was not already optimal, i.e., during the early phase of acquisition. It was not limited to a particular strain.

The purpose of this study was to assess the efficacy of the Process Specific Approach to cognitive rehabilitation for a client with schizophrenia who has attentional deficits. The study was a single case experimental design which followed a variation of the multiple baseline approach. Prior to training of the attentional deficit, multiple baseline assessments were completed. These included an ov:erview of the sUbject's information processing ability, random measures of attention and a genera.l level of functioning in living, learning and working environments. During the re-training, attention tests were administered at the completion of each attention component. A general functional evaluation through interviews and a measure of information processing ability were. completed after the re-training was concluded. The results of the study demonstrate a significant i'mprovement in attention and memory measures. Qualitative data indicate si·gni.ficant others observed improvements in performance in r livi'ng, learning and working environments. The results suggest this approach to cognitive rehabilitation was effective with this subject and further research to establish generalizability is recommended.

The present thesis investigates the responses to reflection in both the crayfish Procambarus clarkii and the fruit fly Drosophila melanogaster. Responses to reflection in crayfish depend on social status and the current work suggests that learning and memory consolidation are required for these responses to be altered. Crayfish were treated to either massed or spaced training fights prior to reflection testing. The results show that subordinate crayfish treated to spaced training display a response typical of subordinate crayfish but subordinate crayfish treated to massed training exhibit a response typical of dominant crayfish. Fruit flies are shown to be attracted to reflection and responses to reflection are described here for the first time. Responses in fruit flies are shown to be dependent on social status. The frequency of behaviours were altered in isolated flies but not socialized flies. The addition of pheromones cVA and 7,11-HD were used to investigate how the addition of chemical cues altered responses to reflection in fruit flies. Socialized fruit flies treated with cVA exhibited an increase in the frequency of behaviours on both mirrored and clear glass walls, while isolated flies exhibited a decrease. Socialized flies treated with 7,11-HD spent more time on mirrored walls compared to glass walls, whereas the frequency of all behaviours were decreased in isolated flies treated with 7,11-HD.

La mémoire et l’apprentissage sont des phénomènes complexes dont on ne comprend pas encore bien l’origine au niveau cellulaire et moléculaire. Cependant, il est largement admis que des changements plus simples au niveau synaptique, tels que la potentialisation à long-terme (long-term potentiation ou LTP) pourraient constituer la base cellulaire de la formation des nouveaux souvenirs. Ces mécanismes sont couramment étudiés au niveau de l’hippocampe, une région du lobe temporal reconnue comme étant nécessaire à la formation de la mémoire explicite chez les mammifères. La LTP est classiquement définie comme un renforcement durable de l’efficacité de connexions synaptiques ayant été stimulées de façon répétée et soutenue. De plus, on peut distinguer deux formes de LTP: une LTP précoce, qui repose sur la modification de protéines déjà formées, et une LTP tardive, qui requiert, elle, la synthèse de nouvelles protéines. Cependant, bien que de nombreuses études se soient intéressées au rôle de la traduction pour la maintenance de la LTP, les mécanismes couplant l’activité synaptique à la machinerie de synthèse protéique, de même que l’identité des protéines requises sont encore peu connus. Dans cette optique, cette thèse de doctorat s’est intéressée aux interactions entre l’activité synaptique et la régulation de la traduction. Il est par ailleurs reconnu que la régulation de la traduction des ARNm eukaryotiques se fait principalement au niveau de l’initiation. Nous avons donc étudié la modulation de deux voies majeures pour la régulation de la traduction au cours de la LTP : la voie GCN2/eIF2α et la voie mTOR. Ainsi, nos travaux ont tout d’abord démontré que la régulation de la voie GCN2/eIF2α et de la formation du complexe ternaire sont nécessaires à la maintenance de la plasticité synaptique et de la mémoire à long-terme. En effet, l’activité synaptique régule la phosphorylation de GCN2 et d’eIF2α, ce qui permet de moduler les niveaux du facteur de transcription ATF4. Celui-ci régule à son tour la transcription CREB-dépendante et permet ainsi de contrôler les niveaux d’expression génique et la synthèse de protéines nécessaires pour la stabilisation à long-terme des modifications synaptiques. De plus, la régulation de la voie mTOR et de la traduction spécifique des ARNm 5’TOP semble également jouer un rôle important pour la plasticité synaptique à long-terme. La modulation de cette cascade par l’activité synaptique augmente en effet spécifiquement la capacité de traduction des synapses activées, ce qui leur permet de traduire et d’incorporer les protéines nécessaires au renforcement durable des synapses. De telles recherches permettront sans doute de mieux comprendre la régulation des mécanismes traductionnels par l’activité synaptique, ainsi que leur importance pour la maintenance de la potentialisation à long-terme et de la mémoire à long-terme.

Peu importe la discipline qui l’appréhende, de la médecine humorale à la psychanalyse aujourd’hui, de l’histoire de l’art à la philosophie, la mélancolie se définit par un manque. S’il ne succombe pas à l’apathie, le mélancolique s’efforcera de pallier cette insuffisance par ses activités intellectuelles et artistiques : la mélancolie est carence et génie. La mélancolie travaille : elle compose avec l’absence. De quel ordre est ce manque ? Dans les écrits savants et les œuvres visuelles, la mélancolie a l’image en défaut : un souvenir ou une représentation juste, idéale. La mélancolie ne donne rien à voir sinon ce rapport à l’image, ce travail de mise en ordre et de mise en œuvre que l’on résume sous les noms « intellection » et « création ». La mélancolie est formaliste : elle cherche un modèle, une représentation, un nom, la forme d’une narration. Peu d’œuvres se prêtent à l’étude du génie de la mélancolie comme celle de Roland Barthes (1915-1980). Critique, ce corpus questionne la mélancolie de la forme et du sens. Écrite, cette œuvre donne à lire une figure de la mélancolie qui diffère selon ce qui lui manque. Toujours, la mélancolie compose avec l’absence de l’image. Cache de l’écriture, la photographie a été utilisée comme image du réel et du souvenir. L’image photographique participe d’une quête théorique en même temps qu’elle donne forme à la mélancolie de l’écriture. Avec la photographie, la mélancolie apparaît à la ville (L’empire des signes), au miroir (Roland Barthes par Roland Barthes), en amour (Fragments d’un discours amoureux) et au tombeau (La chambre claire). En figurant ce qui échappe à la littérature, la photographie ordonne autour d’elle une narration mélancolique. Ainsi la fragmentation, la collection, la spécularisation, l’investigation et la formalisation, sont autant d’opérations qui caractérisent la poétique narrative mise en place dans l’œuvre de Roland Barthes. Dans ces opérations, nous voyons également un modèle de la mélancolie du processus de création.

Un certain nombre de théories pédagogiques ont été établies depuis plus de 20 ans. Elles font appel aux réactions de l’apprenant en situation d’apprentissage, mais aucune théorie pédagogique n’a pu décrire complètement un processus d’enseignement en tenant compte de toutes les réactions émotionnelles de l’apprenant. Nous souhaitons intégrer les émotions de l’apprenant dans ces processus d’apprentissage, car elles sont importantes dans les mécanismes d’acquisition de connaissances et dans la mémorisation. Récemment on a vu que le facteur émotionnel est considéré jouer un rôle très important dans les processus cognitifs. Modéliser les réactions émotionnelles d’un apprenant en cours du processus d’apprentissage est une nouveauté pour un Système Tutoriel Intelligent. Pour réaliser notre recherche, nous examinerons les théories pédagogiques qui n’ont pas considéré les émotions de l’apprenant. Jusqu’à maintenant, aucun Système Tutoriel Intelligent destiné à l’enseignement n’a incorporé la notion de facteur émotionnel pour un apprenant humain. Notre premier objectif est d’analyser quelques stratégies pédagogiques et de détecter les composantes émotionnelles qui peuvent y être ou non. Nous cherchons à déterminer dans cette analyse quel type de méthode didactique est utilisé, autrement dit, que fait le tuteur pour prévoir et aider l’apprenant à accomplir sa tâche d’apprentissage dans des conditions optimales. Le deuxième objectif est de proposer l’amélioration de ces méthodes en ajoutant les facteurs émotionnels. On les nommera des « méthodes émotionnelles ». Le dernier objectif vise à expérimenter le modèle d’une théorie pédagogique améliorée en ajoutant les facteurs émotionnels. Dans le cadre de cette recherche nous analyserons un certain nombre de théories pédagogiques, parmi lesquelles les théories de Robert Gagné, Jerome Bruner, Herbert J. Klausmeier et David Merrill, pour chercher à identifier les composantes émotionnelles. Aucune théorie pédagogique n’a mis l’accent sur les émotions au cours du processus d’apprentissage. Ces théories pédagogiques sont développées en tenant compte de plusieurs facteurs externes qui peuvent influencer le processus d’apprentissage. Nous proposons une approche basée sur la prédiction d’émotions qui est liée à de potentielles causes déclenchées par différents facteurs déterminants au cours du processus d’apprentissage. Nous voulons développer une technique qui permette au tuteur de traiter la réaction émotionnelle de l’apprenant à un moment donné au cours de son processus d’apprentissage et de l’inclure dans une méthode pédagogique. Pour atteindre le deuxième objectif de notre recherche, nous utiliserons un module tuteur apprenant basé sur le principe de l’éducation des émotions de l’apprenant, modèle qui vise premièrement sa personnalité et deuxièmement ses connaissances. Si on défini l’apprenant, on peut prédire ses réactions émotionnelles (positives ou négatives) et on peut s’assurer de la bonne disposition de l’apprenant, de sa coopération, sa communication et l’optimisme nécessaires à régler les problèmes émotionnels. Pour atteindre le troisième objectif, nous proposons une technique qui permet au tuteur de résoudre un problème de réaction émotionnelle de l’apprenant à un moment donné du processus d’apprentissage. Nous appliquerons cette technique à une théorie pédagogique. Pour cette première théorie, nous étudierons l’effet produit par certaines stratégies pédagogiques d’un tuteur virtuel au sujet de l’état émotionnel de l’apprenant, et pour ce faire, nous développerons une structure de données en ligne qu’un agent tuteur virtuel peut induire à l’apprenant des émotions positives. Nous analyserons les résultats expérimentaux en utilisant la première théorie et nous les comparerons ensuite avec trois autres théories que nous avons proposées d’étudier. En procédant de la sorte, nous atteindrons le troisième objectif de notre recherche, celui d’expérimenter un modèle d’une théorie pédagogique et de le comparer ensuite avec d’autres théories dans le but de développer ou d’améliorer les méthodes émotionnelles. Nous analyserons les avantages, mais aussi les insuffisances de ces théories par rapport au comportement émotionnel de l’apprenant. En guise de conclusion de cette recherche, nous retiendrons de meilleures théories pédagogiques ou bien nous suggérerons un moyen de les améliorer.

L’importance respective des lymphocytes T régulateurs naturels générés dans le thymus ou induits en périphérie dans la régulation immunitaire et la résolution de l’inflammation est désormais bien établie. Nous avons contribué à mettre en évidence une nouvelle voie d’induction de lymphocytes T régulateurs périphériques à partir de cellules T humaines CD4+CD25- naïves et mémoires. Nous avons montré que l’engagement de la molécule ubiquitaire transmembranaire CD47 sur la cellule T par un anticorps monoclonal ou par le peptide 4N1K (peptide dérivé du domaine carboxy-terminal de la thrombospondine-1 et spécifique du site de liaison à CD47) induisait des lymphocytes T CD4+ régulateurs exerçant une fonction suppressive sur les lymphocytes T effecteurs. Les propriétés suppressives induites par la thrombospondine-1 confortent les fonctions anti-inflammatoires de cette protéine de la matrice extracellulaire. L’inhibition exercée par les lymphocytes T régulateurs induits dépend du contact intercellulaire entre les cellules T régulatrices et leurs cibles, et est indépendante du TGF-. Nos résultats démontrent également le rôle de CD47 sur le lymphocyte T CD4+ dans la réponse immunitaire spécifique de l’antigène in vivo. En effet, les souris BALB/c déficientes pour CD47 présentent un biais de la sécrétion d’anticorps et de cytokines de type Th1, alors que les souris BALB/c sont décrites comme exprimant un profil de production de cytokines de type Th2. Nos travaux mettent en évidence le rôle de CD47 dans l’inhibition du développement d’une réponse cellulaire et humorale de type Th1 in vivo, confirmant de précédentes études in vitro réalisées avec des cellules T CD4+ humaines. Nous présentons également le rôle inhibiteur de l’engagement de CD28 in vitro sur la différenciation en cellules Th17 des lymphocytes T CD4+ naïfs isolés de souris BALB/c. Le mécanisme proposé est dépendant de la production de l’IL-2 et de l’IFN- et indépendant de la présence de lymphocytes T régulateurs. Notre étude du rôle de deux molécules transmembranaires CD47 et CD28 exprimées sur la cellule T CD4+, contribue à une meilleure connaissance des mécanismes impliqués dans la tolérance immunologique, la résolution de l’inflammation et la différenciation des cellules T "helper" CD4+.
