We examined the efficiency of digestion of hemoglobin from four mammalian species, human, cow, sheep, and horse by acidic extracts of mixed sex adults of Schistosoma japonicum and S. mansoni. Activity ascribable to aspartic protease(s) from S. japonicum and S. mansoni cleaved human hemoglobin. In addition, aspartic protease activities from S. japonicum cleaved hemoglobin from bovine, sheep, and horse blood more efficiently than did the activity from extracts of S. mansoni. These findings support the hypothesis that substrate specificity of hemoglobin-degrading proteases employed by blood feeding helminth parasites influences parasite host species range; differences in amino acid sequences in key sites of the parasite proteases interact less or more efficiently with the hemoglobins of permissive or non-permissive hosts.

OBJECTIVES: The inoculum effect (IE) is an increase in the minimum inhibitory concentration (MIC) at high bacterial densities. The effect of three inoculum sizes on the selection of resistance to vancomycin, daptomycin, and linezolid was investigated in methicillin-resistant Staphylococcus aureus (MRSA). METHODS: Low (10(4) CFU/ml), medium (10(6) CFU/ml), and high (10(8) CFU/ml) inocula of MRSA were exposed to twofold increasing concentrations of either drug during 15 days of cycling. MICs for low (MICL), medium (MICM), and high (MICH) inocula were determined daily. Conventional MICs were measured at days 1, 5, 10, and 15. Experiments were performed in triplicate. RESULTS: At the beginning of the experiment a small IE was observed for vancomycin (MICL=1 μg/ml, MICM=1-2 μg/ml, and MICH=2 μg/ml) and a significant IE for daptomycin (MICL=0.25 μg/ml, MICM=0.25-0.5 μg/ml, and MICH=2 μg/ml). Linezolid exhibited no IE at low and medium inocula (MICL=1 μg/ml and MICM=1-2 μg/ml), but with the high inoculum, concentrations up to 2,048 μg/ml did not fully inhibit visual growth. During cycling, increase of MIC was observed for all antibiotics. At day 15, MICL, MICM, and MICH of vancomycin were 2-4, 4-8, and 4-16 μg/ml and of daptomycin were 0.5-2, 8-128, and 64-256 μg/ml, respectively. MICL and MICM of linezolid were 1 and 2-4 μg/ml, respectively. Conventional MICs showed vancomycin and daptomycin selection of resistance since day 5 depending on the inocula. No selection of linezolid resistance was observed. CONCLUSIONS: Our results showed the importance of the inoculum size in the development of resistance. Measures aimed at lowering the inoculum at the site of infection should be used whenever possible in parallel to antimicrobial therapy.

This research project looked at the economic benefits and costs associated with alternative strategies for abandoning low volume rural highways and bridges. Three test counties in Iowa were studied, each 100 square miles in size: Hamilton County having a high agricultural tax base and a high percentage of paved roads and few bridges; Shelby County having a relatively low agricultural tax base, hilly terrain and a low percentage of paved road and many bridges; and Linn County having a high agricultural tax base, a high percentage of paved roads and a large number of non-farm households. A questionnaire survey was undertaken to develop estimates of farm and household travel patterns. Benefits and costs associated with the abandonment of various segments of rural highway and bridge mileages in each county were calculated. "Benefits" calculated were reduced future reconstruction and maintenance costs, whereas "costs" were the added cost of travel resulting from the reduced mileage. Some of the findings suggest limited cost savings from abandonment of county roads with no property access in areas with large non-farm rural population; relatively high cost savings from the abandonment of roads with no property access in areas with small rural population; and the largest savings from the conversion of public dead-end gravel roads with property or residence accesses to private drives.

In 2004 Walnut Creek was placed on the 303d list of impaired water bodies for lack of aquatic life with biological causes. Sediment from farmland as well as the stream banks was listed as the most likely stressor. In response to this listing a preliminary watershed assessment was completed by the six counties which have land in the Walnut Creek watershed. Walnut Creek flows through portions of Shelby, Pottawattamie, Montgomery, Mills, Page, and Fremont Counties before reaching its confluence with the West Nishnabotna River. The preliminary study assessed resource concerns and evaluated anticipated landowner participation levels for the six Huc 12 sub-watersheds which divide the Walnut Creek basin. These preliminary assessments revealed a priority sub-watershed which lies between US Hwy 6 and Hwy 34. A development grant was then funded by the Division of Soil Conservation to conduct a detailed assessment of this area. The detailed assessment involved an assessment of the uplands as well as the stream itself. A better understanding of the resource concerns was gained through the assessment, allowing for a comprehensive watershed plan to be developed. A variety of best management practices will be necessary for our project to be a success, many of which will be funded by other sources besides the WIRB. This grant is the first request for funding submitted by the East Pottawattamie and Montgomery SWCDs’. This grant will serve as the first critical step in building what is destined to be a true watershed success story.

Manteno Lake is located 8 miles northwest of Earling, IA in Shelby County. The lake has been impaired with an overload of silt and nutrients. In October of 2007, land directly north of the lake will be coming out of CRP and back into agricultural production. The Shelby Soil and Water Conservation District, Shelby County Board of Supervisors and Shelby County Conservation Board feel strongly that treatment of this area is necessary prior to the CRP contract expiring. The placement and construction of three structures will reduce the amount of silt and nutrients affecting Manteno Lake. This project calls for the construction of three structures on crucial tributaries to the lake.
