The cultivation of hybrid rice is a technology that allows for an increase in grain yield of 30% relative to the grain yield of conventional cultivars. However, the main challenge for this technology is related to seed production, which has high production costs and low seed yields. Therefore, agronomic techniques that could enhance flowering synchrony of parental lines in the field are essential for an efficient production system of hybrid rice seeds. The objective of this work was to study the effects of sowing depth, plant density and fertilization with nitrogen or phosphorus as potential techniques to increase the pollen availability in the field and, consequently, the flowering synchrony between parental lines in the production of hybrid rice seeds. The experiments were conducted during two growing seasons in the Central Region of Brazil. All of the experiments were conducted as a randomized complete block in a split plot scheme; however, the experiment with P fertilization had a factorial design. Our research allow inferring that nitrogen fertilization technique applied to the soil or foliar at the time of panicle differentiation does not affect the time of onset of flowering of rice varieties INTA Puitá CL and L106R, which are potential R lines for the production of hybrid rice. Agronomic techniques of variation in sowing depth, seeding rate and the phosphate fertilization affect the time of onset of flowering from 10 to 19 degree-days, which could represent two days in the crop cycle, for the line L106R. Such techniques constitute potential alternatives for use in hybrid rice seed production systems and could be applied in alternated blocks of R lines in the field to obtain longer periods of pollen availability in the field.

La stesura del seguente elaborato di tesi è avvenuta simultaneamente all’inizio di un progetto Lean presso il plant produttivo, situato a Crevalcore (Bo), di Fonderie di Montorso Spa, azienda produttrice di getti in ghisa. Lo scopo del seguente elaborato è di descrivere l’implementazione di un progetto Lean in uno dei settori più antichi esistenti. Negli ultimi decenni, si è sentito molto parlare di Lean production in settori come quello dell’automotive ma, raramente, si sente parlare di lean thinking o kaizen in settori metalmeccanici. I processi produttivi tipici di una fonderia, infatti, sono difficilmente prevedibili e controllabili come può essere un processo di assemblaggio di componenti. Trasferire, dunque, un sistema di gestione della produzione, il Toyota Production System, nato in una linea di assemblaggio, in un settore con una grande quantità di vincoli di progettazione e produzione, è una sfida, una sfida necessaria per rispondere ad una situazione di estrema variabilità della domanda, in cui il mercato richiede i prodotti in maniera rapida e puntuale. Per rimanere, dunque, competitivi è necessario un cambiamento radicale, una rivisitazione di tutti i processi aziendali partendo dall’arrivo dell’ordine, alla spedizione dello stesso. Alla luce di ciò, nel corso della trattazione, sono state svolte analisi al fine di ridurre il Lead time di partenza da 18 a 4 settimane, ponendo molta attenzione al coinvolgimento delle persone, il vero motore del cambiamento. Le parole cardine di questo elaborato sono: standardizzazione dei processi, eliminazione dei colli di bottiglia, aumento della comunicazione tra le funzioni aziendali, prioritizzazione dei clienti e per ultimo, ma non per importanza, la valorizzazione del gemba.

Agriculture market instability impedes achieving the global goal of sustainable and resilient food systems. Currently, the support to producers reaches the mammoth USD 540 billion a year and is projected to reach USD 1.8 trillion by 2030. This gigantic increase requires a repurposing agricultural support strategy (RASS), considering the market country-specific circumstances. These circumstances may vary with geographic locations, marketing structures, and product value chains. The fruit production system is crucial for health-conscious consumers and profit-oriented producers for food and nutritional security. Export is one of the main driving forces behind the expansion of the fruit sector, and during the year 2010-2018, trade significantly outpaced production increases. The previous literature states that irregular and unpredictable behaviour — Chaos — can arise from entirely rational economic decision-making within markets. Different markets' direct/indirect linkages through trade create trade hubs, and uncertainty may function as an avenue to transmit adverse shocks and increase vulnerability rather than contribute to resilience. Therefore, distinguishing Chaos into an endogenous and exogenous pattern of behaviour is cradled to formulate an effective RASS for resilient food systems and to understand global food crises. The present research is aimed at studying the market dynamics of three regional trade hubs, i.e., Brazil (South America), Italy (Europe), and Pakistan (Asia), each representing advanced to traditional value chains to control uncertainty (risks). The present research encompasses 1) a systematic review to highlight the research dynamism and identify grey-areas of research. Based on the findings, we have investigated the 2) nonlinear impacts of climate-induced price responsiveness in monopsony markets. Once we highlighted the importance of marketing structures/arrangements, 3) we developed a risk transmission framework to address the co-evolving impacts in complex dynamic interactions.

The Internet of Vehicles (IoV) paradigm has emerged in recent times, where with the support of technologies like the Internet of Things and V2X , Vehicular Users (VUs) can access different services through internet connectivity. With the support of 6G technology, the IoV paradigm will evolve further and converge into a fully connected and intelligent vehicular system. However, this brings new challenges over dynamic and resource-constrained vehicular systems, and advanced solutions are demanded. This dissertation analyzes the future 6G enabled IoV systems demands, corresponding challenges, and provides various solutions to address them. The vehicular services and application requests demands proper data processing solutions with the support of distributed computing environments such as Vehicular Edge Computing (VEC). While analyzing the performance of VEC systems it is important to take into account the limited resources, coverage, and vehicular mobility into account. Recently, Non terrestrial Networks (NTN) have gained huge popularity for boosting the coverage and capacity of terrestrial wireless networks. Integrating such NTN facilities into the terrestrial VEC system can address the above mentioned challenges. Additionally, such integrated Terrestrial and Non-terrestrial networks (T-NTN) can also be considered to provide advanced intelligent solutions with the support of the edge intelligence paradigm. In this dissertation, we proposed an edge computing-enabled joint T-NTN-based vehicular system architecture to serve VUs. Next, we analyze the terrestrial VEC systems performance for VUs data processing problems and propose solutions to improve the performance in terms of latency and energy costs. Next, we extend the scenario toward the joint T-NTN system and address the problem of distributed data processing through ML-based solutions. We also proposed advanced distributed learning frameworks with the support of a joint T-NTN framework with edge computing facilities. In the end, proper conclusive remarks and several future directions are provided for the proposed solutions.

L'elaborato proposto illustra l'implementazione della metodologia di Lean Manufacturing nella progettazione di una linea di assemblaggio destinata alla realizzazione di bruciatori residenziali. Vengono messi in evidenza i principali strumenti avanzati dal Toyota Production System e viene condotta un'analisi sull'influenza di tale approccio sulle maggiori aziende del settore Automotive degli ultimi decenni. Il caso di studio mette a confronto la linea di produzione in oggetto a monte e a valle dell'implementazione Lean presso l'azienda Riello S.p.a. di Legnago (VR). Si evidenziano i benefici apportati in termini di output produttivo giornaliero e riduzione dei costi di manodopera attraverso il miglioramento continuo del processo, l'adozione di nuovi utensili e l'applicazione del metodo Yamazumi nel ribilanciamento delle operazioni manuali.

Les défis conjoints du changement climatique d'origine anthropique et la diminution des réserves de combustibles fossiles sont le moteur de recherche intense pour des sources d'énergie alternatives. Une avenue attrayante est d'utiliser un processus biologique pour produire un biocarburant. Parmi les différentes options en matière de biocarburants, le bio-hydrogène gazeux est un futur vecteur énergétique attrayant en raison de son efficacité potentiellement plus élevé de conversion de puissance utilisable, il est faible en génération inexistante de polluants et de haute densité d'énergie. Cependant, les faibles rendements et taux de production ont été les principaux obstacles à l'application pratique des technologies de bio-hydrogène. Des recherches intensives sur bio-hydrogène sont en cours, et dans les dernières années, plusieurs nouvelles approches ont été proposées et étudiées pour dépasser ces inconvénients. À cette fin, l'objectif principal de cette thèse était d'améliorer le rendement en hydrogène moléculaire avec un accent particulier sur l'ingénierie métabolique et l’utilisation de bioprocédés à variables indépendantes. Une de nos hypothèses était que la production d’hydrogène pourrait être améliorée et rendue plus économiquement viable par ingénierie métabolique de souches d’Escherichia coli producteurs d’hydrogène en utilisant le glucose ainsi que diverses autres sources de carbone, y compris les pentoses. Les effets du pH, de la température et de sources de carbone ont été étudiés. La production maximale d'hydrogène a été obtenue à partir de glucose, à un pH initial de 6.5 et une température de 35°C. Les études de cinétiques de croissance ont montré que la μmax était 0.0495 h-1 avec un Ks de 0.0274 g L-1 lorsque le glucose est la seule source de carbone en milieu minimal M9. .Parmi les nombreux sucres et les dérivés de sucres testés, les rendements les plus élevés d'hydrogène sont avec du fructose, sorbitol et D-glucose; 1.27, 1.46 et 1.51 mol H2 mol-1 de substrat, respectivement. En outre, pour obtenir les interactions entre les variables importantes et pour atteindre une production maximale d'hydrogène, un design 3K factoriel complet Box-Behnken et la méthodologie de réponse de surface (RSM) ont été employées pour la conception expérimentale et l'analyse de la souche d'Escherichia coli DJT135. Le rendement en hydrogène molaire maximale de 1.69 mol H2 mol-1 de glucose a été obtenu dans les conditions optimales de 75 mM de glucose, à 35°C et un pH de 6.5. Ainsi, la RSM avec un design Box-Behken était un outil statistique utile pour atteindre des rendements plus élevés d'hydrogène molaires par des organismes modifiés génétiquement. Ensuite, l'expression hétérologue de l’hydrogénases soluble [Ni-Fe] de Ralstonia eutropha H16 (l'hydrogénase SH) a tenté de démontrer que la mise en place d'une voie capable de dériver l'hydrogène à partir de NADH pourrait surpasser le rendement stoechiométrique en hydrogène.. L’expression a été démontrée par des tests in vitro de l'activité enzymatique. Par ailleurs, l'expression de SH a restaurée la croissance en anaérobie de souches mutantes pour adhE, normalement inhibées en raison de l'incapacité de réoxyder le NADH. La mesure de la production d'hydrogène in vivo a montré que plusieurs souches modifiées métaboliquement sont capables d'utiliser l'hydrogénase SH pour dériver deux moles d’hydrogène par mole de glucose consommé, proche du maximum théorique. Une autre stratégie a montré que le glycérol brut pourrait être converti en hydrogène par photofermentation utilisant Rhodopseudomonas palustris par photofermentation. Les effets de la source d'azote et de différentes concentrations de glycérol brut sur ce processus ont été évalués. À 20 mM de glycérol, 4 mM glutamate, 6.1 mol hydrogène / mole de glycérol brut ont été obtenus dans des conditions optimales, un rendement de 87% de la théorie, et significativement plus élevés que ce qui a été réalisé auparavant. En prolongement de cette étude, l'optimisation des paramètres a également été utilisée. Dans des conditions optimales, une intensité lumineuse de 175 W/m2, 30 mM glycérol et 4.5 mM de glutamate, 6.69 mol hydrogène / mole de glycérol brut ont été obtenus, soit un rendement de 96% de la valeur théorique. La détermination de l'activité de la nitrogénase et ses niveaux d'expression ont montré qu'il y avait relativement peu de variation de la quantité de nitrogénase avec le changement des variables alors que l'activité de la nitrogénase variait considérablement, avec une activité maximale (228 nmol de C2H4/ml/min) au point central optimal. Dans la dernière section, la production d'hydrogène à partir du glucose via la photofermentation en une seule étape a été examinée avec la bactérie photosynthétique Rhodobacter capsulatus JP91 (hup-). La méthodologie de surface de réponse avec Box-Behnken a été utilisée pour optimiser les variables expérimentales de façon indépendante, soit la concentration de glucose, la concentration du glutamate et l'intensité lumineuse, ainsi que d'examiner leurs effets interactifs pour la maximisation du rendement en hydrogène moléculaire. Dans des conditions optimales, avec une intensité lumineuse de 175 W/m2, 35 mM de glucose, et 4.5 mM de glutamate,, un rendement maximal d'hydrogène de 5.5 (± 0.15) mol hydrogène /mol glucose, et un maximum d'activité de la nitrogénase de 246 (± 3.5) nmol C2H4/ml/min ont été obtenus. L'analyse densitométrique de l'expression de la protéine-Fe nitrogenase dans les différentes conditions a montré une variation significative de l'expression protéique avec un maximum au point central optimisé. Même dans des conditions optimales pour la production d'hydrogène, une fraction significative de la protéine Fe a été trouvée dans l'état ADP-ribosylée, suggérant que d'autres améliorations des rendements pourraient être possibles. À cette fin, un mutant amtB dérivé de Rhodobacter capsulatus JP91 (hup-) a été créé en utilisant le vecteur de suicide pSUP202. Les résultats expérimentaux préliminaires montrent que la souche nouvellement conçue métaboliquement, R. capsulatus DG9, produit 8.2 (± 0.06) mol hydrogène / mole de glucose dans des conditions optimales de cultures discontinues (intensité lumineuse, 175 W/m2, 35 mM de glucose et 4.5 mM glutamate). Le statut d'ADP-ribosylation de la nitrogénase-protéine Fe a été obtenu par Western Blot pour la souche R. capsulatus DG9. En bref, la production d'hydrogène est limitée par une barrière métabolique. La principale barrière métabolique est due au manque d'outils moléculaires possibles pour atteindre ou dépasser le rendement stochiométrique en bio-hydrogène depuis les dernières décennies en utilisant les microbes. À cette fin, une nouvelle approche d’ingénierie métabolique semble très prometteuse pour surmonter cette contrainte vers l'industrialisation et s'assurer de la faisabilité de la technologie de la production d'hydrogène. Dans la présente étude, il a été démontré que l’ingénierie métabolique de bactéries anaérobiques facultatives (Escherichia coli) et de bactéries anaérobiques photosynthétiques (Rhodobacter capsulatus et Rhodopseudomonas palustris) peuvent produire de l'hydrogène en tant que produit majeur à travers le mode de fermentation par redirection métabolique vers la production d'énergie potentielle. D'autre part, la méthodologie de surface de réponse utilisée dans cette étude représente un outil potentiel pour optimiser la production d'hydrogène en générant des informations appropriées concernant la corrélation entre les variables et des producteurs de bio-de hydrogène modifiés par ingénierie métabolique. Ainsi, un outil d'optimisation des paramètres représente une nouvelle avenue pour faire un pont entre le laboratoire et la production d'hydrogène à l'échelle industrielle en fournissant un modèle mathématique potentiel pour intensifier la production de bio-hydrogène. Par conséquent, il a été clairement mis en évidence dans ce projet que l'effort combiné de l'ingénierie métabolique et la méthodologie de surface de réponse peut rendre la technologie de production de bio-hydrogène potentiellement possible vers sa commercialisation dans un avenir rapproché.

We present an outlook on the climate system thermodynamics. First, we construct an equivalent Carnot engine with efficiency and frame the Lorenz energy cycle in a macroscale thermodynamic context. Then, by exploiting the second law, we prove that the lower bound to the entropy production is times the integrated absolute value of the internal entropy fluctuations. An exergetic interpretation is also proposed. Finally, the controversial maximum entropy production principle is reinterpreted as requiring the joint optimization of heat transport and mechanical work production. These results provide tools for climate change analysis and for climate models’ validation.

The HACCP system is being increasingly used to ensure food safety. This study investigated the validation of the control measures technique in order to establish performance indicators of this HACCP system in the manufacturing process of Lasagna Bolognese (meat lasagna). Samples were collected along the manufacturing process as a whole, before and after the CCPs. The following microorganism s indicator (MIs) was assessed: total mesophile and faecal coliform counts. The same MIs were analyzed in the final product, as well as, the microbiological standards required by the current legislation. A significant reduction in the total mesophile count was observed after cooking (p < 0.001). After storage, there was a numerical, however non-significant change in the MI count. Faecal coliform counts were also significantly reduced (p < 0.001) after cooking. We were able to demonstrate that the HACCP system allowed us to meet the standards set by both, the company and the Brazilian regulations, proved by the reduction in the established indicators

Biofuels are both a promising solution to global warming mitigation and a potential contributor to the problem. Several life cycle assessments of bioethanol have been conducted to address these questions. We performed a synthesis of the available data on Brazilian ethanol production focusing on greenhouse gas (GHG) emissions and carbon (C) sinks in the agricultural and industrial phases. Emissions of carbon dioxide (CO(2)) from fossil fuels, methane (CH(4)) and nitrous oxide (N(2)O) from sources commonly included in C footprints, such as fossil fuel usage, biomass burning, nitrogen fertilizer application, liming and litter decomposition were accounted for. In addition, black carbon (BC) emissions from burning biomass and soil C sequestration were included in the balance. Most of the annual emissions per hectare are in the agricultural phase, both in the burned system (2209 out of a total of 2398 kg C(eq)), and in the unburned system (559 out of 748 kg C(eq)). Although nitrogen fertilizer emissions are large, 111 kg C(eq) ha-1 yr-1, the largest single source of emissions is biomass burning in the manual harvest system, with a large amount of both GHG (196 kg C(eq) ha-1 yr-1). and BC (1536 kg C(eq) ha-1 yr-1). Besides avoiding emissions from biomass burning, harvesting sugarcane mechanically without burning tends to increase soil C stocks, providing a C sink of 1500 kg C ha-1 yr-1 in the 30 cm layer. The data show a C output: input ratio of 1.4 for ethanol produced under the conventionally burned and manual harvest compared with 6.5 for the mechanized harvest without burning, signifying the importance of conservation agricultural systems in bioethanol feedstock production.

Sugar and ethanol production are key components of Brazil`s rural development and energy strategies, yet in recent years sugar production has been widely criticized for its environmental and labor practices. This study examines the relationship between rural development and sugarcane, ethanol, and cattle production in the state of Sao Paulo. Our results suggest that the value added components of sugarcane production, which include sugar refining and ethanol production, may have a strong positive affect on local human development in comparison to primary agricultural production activities and other land uses. These results imply that sugar production, when accompanied by a local processing industry can stimulate rural development. However, this paper also highlights the significant environmental and social harms generated by the sugar industry at large, which may undermine its development benefits if not addressed. The success in the adoption of peach integrated production (IP) was evaluated in small orchards of the Parana State. The importance of specific technical accompaniment; points of strangulating in adoption of technology and the classification of the areas to IP conformity were evaluated. The seasons 2005/2006 (without IP orientation) and 2006/2007 (with IP orientation) were compared considering 20 producers who were oriented monthly to attend the minimum requisites. The incidence of peach rust (Tranzschelia discolor) and of brown rot (Monilinia fructicola) in full bloom was evaluated in 2006/2007 and 2007/2008 seasons, as biological parameters to accompany the efficiency of system adoption. After the technical accompaniment in 2007/2008 season, the software APOIA-Novo Rural-PI (APOIA-PI) was applied to measure the conformity to IP in peach orchards. The conformity index of each orchard was compared to the minimum requisite to classify as IP (0.7). The major difficulties in register of field book were: pests monitoring; collect of climate data and the harvest evaluation. The technical accompaniment increased in 60% the conformity in use of field book. In 2007/2008 season, the brown rot incidence increased in some areas, caused by not following IP recommendations. The inadequate management caused the increment in pathogen inoculum, promoting the disease development in peach orchards. The APOIA-PI classified two orchards as good agricultural practices (GAP) (0.7 <= conformity index >= 0.4), two as integrated production (IP) (>= 0.7) and the other orchards had conformity index lower than 0.4.

Fed-batch culture can offer significant improvement in recombinant protein production compared to batch culture in the baculovirus expression vector system (BEVS), as shown by Nguyen et al. (1993) and Bedard et al. (1994) among others. However, a thorough analysis of fed-batch culture to determine its limits in improving recombinant protein production over batch culture has yet to be performed. In this work, this issue is addressed by the optimisation of single-addition fed-batch culture. This type of fed-batch culture involves the manual addition of a multi-component nutrient feed to batch culture before infection with the baculovirus. The nutrient feed consists of yeastolate ultrafiltrate, lipids, amino acids, vitamins, trace elements, and glucose, which were added to batch cultures of Spodoptera frugiperda (Sf9) cells before infection with a recombinant Autographa californica nuclear polyhedrosis virus (Ac-NPV) expressing beta-galactosidase (beta-Gal). The fed-batch production of beta-Gal was optimised using response surface methods (RSM). The optimisation was performed in two stages, starting with a screening procedure to determine the most important variables and ending with a central-composite experiment to obtain a response surface model of volumetric beta-Gal production. The predicted optimum volumetric yield of beta-Gal in fed-batch culture was 2.4-fold that of the best yields in batch culture. This result was confirmed by a statistical analysis of the best fed-batch and batch data (with average beta-Gal yields of 1.2 and 0.5 g/L, respectively) obtained from this laboratory. The response surface model generated can be used to design a more economical fed-batch operation, in which nutrient feed volumes are minimised while maintaining acceptable improvements in beta-Gal yield. We cloned the complete complementary DNA of an isolate of the hepatitis C virus, HCV-S1, into a tetra cycline-inducible expression vector and stably transfected it into two human hepatoma cell lines, Huh7 and HepG2. Twenty-six Huh7 and two HepG2-positive clones were obtained after preliminary screening. Two Huh7 (SH-7 and -9) and one HepG2 (G-19) clones were chosen for further characterisation. Expression of HCV proteins in these cells accumulated from 6 In to 4 days posttreatment. Full-length viral plus-strand RNA was detected by Northern analyses. Using RT-PCR and ribonuclease protection assay, we also detected the synthesis of minus-strand HCV RNA. Plus- and minus-strand viral RNA was still detected after treatment with actinomycin D. Indirect immunofluorescence staining with anti-E2, NS4B, and NS5A revealed that these proteins were mostly localised to the endoplasmic reticulum (ER). Culture media from tet-induced SH-9 cells was separated on sucrose density gradients and analysed for the presence of HCV RNA. Viral RNA levels peaked at two separate ranges, one with a buoyant density of 1.08 g/ml and another from 1.17 to 1.39 g/ml. Electron microscopy demonstrated the presence of subviral-like particles (approximately 20-25 nm in diameter) in the cytoplasm of SH-9 and G-19 cells, which were positively labelled by anti-HCV core antibodies. Anti-E2 antibodies strongly labelled cytoplasmic vesicular structures and some viral-like particles. Complete viral particles of about 50 nm which reacted with anti-E2 antibodies were observed in the culture media of tet-induced SH-9 cells following negative staining. Supernatant from tet-treated SH-9 cells was found to infect naive Huh7 and stable Huh7-human CD81 cells. (C) 2002 Elsevier Science (USA).
