Eight bufadienolides were successfully isolated and purified from ChanSu by high-speed counter-current chromatography (HSCCC) combined with preparative HPLC (prep-HPLC). First, a stepwise elution mode of HSCCC with the solvent system composed of petroleum ether - ethyl acetate - methanol - water (4:6:4:6, 4:6:5:5, v/v) was employed and four bufadienolides, two partially purified fractions were obtained from 200 mg of crude extract. The partially purified fractions III and VI were then further separated by prep-HPLC, respectively, and another four bufadienolides were recovered. Their structures were confirmed by ESI-MS and ¹H-NMR spectra.

In the present study, a reversed-phase high-performance liquid chromatographic (RP-HPLC) procedure was developed and validated for the simultaneous determination of seven water-soluble vitamins (thiamine, riboflavin, niacin, cyanocobalamin, ascorbic acid, folic acid, and p-aminobenzoic acid) and four fat-soluble vitamins (retinol acetate, cholecalciferol, α-tocopherol, and phytonadione) in multivitamin tablets. The linearity of the method was excellent (R² > 0.999) over the concentration range of 10 - 500 ng mL-1. The statistical evaluation of the method was carried out by performing the intra- and inter-day precision. The accuracy of the method was tested by measuring the average recovery; values ranged between 87.4% and 98.5% and were acceptable quantitative results that corresponded with the label claims.

An effective method for the rapid separation and purification of three stilbenes from the radix of Polygonum cillinerve (Nakai) Ohwl by macroporous resin column chromatography combined with high-speed counter-current chromatography (HSCCC) was successfully established. In the present study, a two-phase solvent system composed of chloroform-n-butanol-methanol-water (4:1:4:2, v/v/v/v) was used for HSCCC separation. A one-step separation in 4 h from 150 mg of crude extract produced 26.3 mg of trans-resveratrol-3-O-glucoside, 42.0 mg of pieceid-2"-O-gallate, and 17.9 mg of trans-resveratrol with purities of 99.1%, 97.8%, and 99.4%, respectively, as determined by high-performance liquid chromatography (HPLC). The chemical structures of these compounds were identified by nuclear magnetic resonance (NMR) spectroscopy.

This work describes a method to determine Cu at wide range concentrations in a single run without need of further dilutions employing high-resolution continuum source flame atomic absorption spectrometry. Different atomic lines for Cu at 324.754 nm, 327.396 nm, 222.570 nm, 249.215 nm and 224.426 nm were evaluated and main figures of merit established. Absorbance measurements at 324.754 nm, 249.215 nm and 224.426 nm allows the determination of Cu in the 0.07 - 5.0 mg L-1, 5.0 - 100 mg L-1 and 100 - 800 mg L-1 concentration intervals respectively with linear correlation coefficients better than 0.998. Limits of detection were 21 µg L-1, 310 µg L-1 and 1400 µg L-1 for 324.754 nm, 249.215 nm and 224.426 nm, respectively and relative standard deviations (n = 12) were £ 2.7%. The proposed method was applied to water samples spiked with Cu and the results were in agreement at a 95% of confidence level (paired t-test) with those obtained by line-source flame atomic absorption spectrometry.

Työn tuloksien perusteella energiankäytön kannalta paras pyörimisnopeus Proto 10 sekoittimelle on 1500 min-1. Nostamalla kierrosnopeutta yli tämän ei saavutettu merkittävää parannusta dispersion laadussa varsinkaan suuremmilla virtaamilla. Virtaamalla todettiin olevan suuri merkitys sekoitustulokseen. Laboratoriosekoittimella tehtyjen kokeiden todettiin vastaavan parhaiten teollisen mittakaavan tuloksia pyörimisnopeudella 1200 min-1.

The deficient environmental preservation on agricultural properties has reflected in great soil losses for erosion and the high rate of sedimentation. Therefore, this work brings the application of software to demonstrate that planters can get benefits by the conservationist practices. The benefits will be obtained from the environment aspect (erosion and sedimentation control), as well as economical aspect (financial incentives). A proposal has been created by the Technical Chamber of Rural Water Use and Conservation (CT-Rural) to estimate the reduction in raw water use charge for the rural water users of the PCJ Watershed - this reduction is being possible since January 2007, at the PCJ Committee. The software develops a simple relationship, so that if the rural conservationist action goes more efficient, such as if the conserved rural area goes larger, then the reduction value will bring better benefits for the rural water users. Within the purposes of this study, there is the idealization of mechanisms that may support specific management matters related to the water users of federal rivers from the rural sector of PCJ Watershed.

The increasing demand for water resources accentuates the need to reduce water waste through a more appropriate irrigation management. In the particular case of irrigated coffee planting, which in recent years presented growth with the predominance of drip irrigation, the improvement of drip irrigation management techniques is a necessity. The proper management of drip irrigation depends on the knowledge of the spatial pattern of soil moisture distribution inside the wetted strip formed under the irrigation lines. In this study, grids of 24 tensiometers were used to determine the water storage within the wetted strip formed under drippers, with a 3.78 L h-1 discharge, evenly spaced by 0.4 m, subjected to two different management criteria (fixed irrigation interval and 60 kPa tension). Estimates of storage based on a one-dimensional analysis, that only considers depth variations, were compared with two-dimensional estimates. The results indicate that for high-frequency irrigation the one-dimensional analysis is not appropriate. However, under less frequent irrigation, the two-dimensional analysis is dispensable, being the one-dimensional sufficient for calculating the water volume stored in the wetted strip.

The objective this study has been the selection of lipase productor microorganism, for removal of oils and grease, in the pre-treatment of biodiesel wastewater washing. For this, analyses of the physicist-chemistries characteristics had been made with the wastewater of the biodiesel washing, and then it had been isolated and chosen, by means of determinations of the lipase activity. Following, it was made a test of fat biodegradation, in the conditions: pH (5.95), temperature (35 ºC), rotation (180 rpm) and ammonium sulfate as nitrogen source (3 g L-1) and establishing as variable the two microorganism preselected and the time (24; 48; 72; 96 and 120 h). The biodiesel purification wastewater had presented high potential of environmental impact, presenting a concentration of O of 6.76 g L-1. From the six isolated microbiological cultures, two microorganisms (A and B) had been selected, with enzymatic index of 0.56 and 0.57, respectively. The treatment of the wastewater using the isolated microorganism (Klebsiella oxytoca) had 80% of the fatty removal in 48 h.

This study was carried out to determine some physiological and phenological responses of the bean under high [CO2] and drought stress. The experiment was conducted from April to July 2009 in Viçosa, Brazil. The open-top chambers were used to enrich the air with CO2, whereas the drought stress was applied between the flowering and the ripening. The randomized block design was used, with four replicates in the subplots. The following plots were [CO2] at 700ppm (F1) and [CO2] environmental (F2) and the subplots were well watering (S1) and drought stress (S2). The results were subjected to Anova and the Tukey test (P < 0.05). For the treatments F1S1 and F1S2 the photosynthetic rate showed increments of 59% and the transpiration reduction of 12%. The yield, leaf temperature and stomatal conductance were not significant different to high [CO2], different from the dry matter, who showed increment of 20% (F1S1) and the water use efficiency who showed increase of 90% for high [CO2]. The osmotic potential was lower in plants under drought stress (F2S2 and F1S2), followed by plants under high [CO2] (F1S1). Despite the increment in photosynthesis, high [CO2] does not guarantee higher yield.

The aim of this study was to identify the relation between the evapotranspirometer demand and the supply of water from local rainfall, evaluating the possibility of using water excess for irrigation of Green Roofs in the State of Mato Grosso, in Brazil. The study was done using a series of historical data provided by the National Institute of Meteorology (INMET - Instituto Nacional de Meteorologia) which has official climatological stations in 12 cities and regions of the State. The evapotranspiration values were obtained by the Penman-Monteith method and by the Climatic Water Balance (CWB) by the Thornthwaite and Mather method using Available Water Capacity (AWC) of 12mm. With the CWB the excess and deficit were calculated, which were used for the estimative of the volume and area of a reservoir as a function of a collector area of a roof of 100m² and the volume of supplementary water for irrigation. With the obtained results, it was found that in most investigated regions of the State the use of green roofs is not compromised by the water deficiency. On the other hand, the use of a reservoir to accumulate the rain water excess may be impractical, because it requires a considerable area for installation and also because of the high cost of the land.

The determination of volumetric water content of soils is an important factor in irrigation management. Among the indirect methods for estimating, the time-domain reflectometry (TDR) technique has received a significant attention. Like any other technique, it has advantages and disadvantages, but its greatest disadvantage is the need of calibration and high cost of acquisition. The main goal of this study was to establish a calibration model for the TDR equipment, Trase System Model 6050X1, to estimate the volumetric water content in a Distroferric Red Latosol. The calibration was carried out in a laboratory with disturbed soil samples under study, packed in PVC columns of a volume of 0.0078m³. The TDR probes were handcrafted with three rods and 0.20m long. They were vertically installed in soil columns, with a total of five probes per column and sixteen columns. The weightings were carried out in a digital scale, while daily readings of dielectric constant were obtained in TDR equipment. The linear model θν = 0.0103 Ka + 0.1900 to estimate the studied volumetric water content showed an excellent coefficient of determination (0.93), enabling the use of probes in indirect estimation of soil moisture.

Frequent traffic of tractors in agricultural soils, promotes soil compaction and affects the flow and water availability. The aim of this research was to study the effect of compaction induced by tractor traffic on water availability in the soil under different traffic intensities on the same path (0, 1, 3, 6 and 10 passages), to two tractors with 3.3 and 2.6 ton of weights, over three different surface conditions. The study was conducted in an Andisol, representative soil of the study area. It was determined the behavior of the water retention curve, obtaining the gravitational water, available water and hygroscopic water, to 10 and 30 cm of depth. The hygroscopic water is the most prevalent with values of up to 80% of the total water present in the soil. The water retention curves showed increases in the values of field capacity and wilting point and behaviors "flattened" indicating a high sensitivity to the applied treatments, representative of compacted soils, which give the surfaces studied characteristics do not suitable for normal crop development.

The consideration of the streamflow seasonality has a high potential to improve the water use. In order to give subsidies to the optimization of water use, it was evaluated the impact of the change of reference annual streamflow by the monthly streamflows in the potential water use throughout the hydrography of Paracatu sub-Basin. It was evaluated the impact on Q7,10 (lowest average streamflow during a 7-day period with an average recurrence of 10 years) and on Q95 (permanent flow present 95% of the time). The use of monthly streamflow to substitute the annual streamflow had a high potential of improvement of water resources use in the sub-Basin studied. The use of monthly Q 7,10 in substitution of annual Q 7,10 increases the potential water use that vary from about 10% in the months of lower water availability to values exceeding 200% in the months with higher availability of surface water resources. The use of monthly Q95 in substitution of the annual Q95 implies in changes oscillating from reduction of 37% in months of higher water restriction to values exceeding 100% in the months of higher availability, so the use of monthly Q95 instead of the annual Q95 enables the more rational and safe use of water resources.
