Jerdonitin is a P-II class snake venom metalloproteinase comprising metalloproteinase and disintegrin domains. In this study, we established a high-level expression system in Pichia pastoris and developed a purification strategy for the recombinant Jerdonitin. This recombinant Jerdonitin degraded fibrinogen at a level of activity comparable with its wild type. The effects of recombinant Jerdonitin on inhibiting ADP-induced human platelet aggregation were in a dose-dependent manner with an IC50 of 248 nM. In addition, we reported here that Jerdonitin can significantly inhibit the growth of several cell lines, including human liver cancer cells (Bel7402), human leukemia cells (K562) and human gastric carcinoma cells (BGC823). This study offers recombinant Jerdonitin that will be valuable for further functional and structural studies of Jerdonitin. (C) 2009 Elsevier Ltd. All rights reserved.

The contra-rotating open rotor is, once again, being considered as an alternative to the advanced turbofan to address the growing pressure to cut aviation fuel consumption and carbon dioxide emissions. One of the key challenges is meeting community noise targets at takeoff. Previous open rotor designs are subject to poor efficiency at takeoff due to the presence of large regions of separated flow on the blades as a result of the high incidence needed to achieve the required thrust. This is a consequence of the fixed rotor rotational speed constraint typical of variable pitch propellers. Within the study described in this paper, an improved operation is proposed to improve performance and reduce rotorrotor interaction noise at takeoff. Three-dimensional computational fluid dynamics (CFD) calculations have been performed on an open rotor rig at a range of takeoff operating conditions. These have been complemented by analytical tone noise predictions to quantify the noise benefits of the approach. The results presented show that for a given thrust, a combination of reduced rotor pitch and increased rotor rotational speed can be used to reduce the incidence onto the front rotor blades. This is shown to eliminate regions of flow separation, reduce the front rotor tip loss and reduce the downstream stream tube contraction. The wakes from the front rotor are also made wider with lower velocity defect, which is found to lead to reduced interaction tone noise. Unfortunately, the necessary increase in blade speed leads to higher relative Mach numbers, which can increase rotor alone noise. In summary, the combined CFD and aero-acoustic analysis in this paper shows how careful operation of an open rotor at takeoff, with moderate levels of re-pitch and speed increase, can lead to improved front rotor efficiency as well as appreciably lower overall noise across all directivities. The protection of the environment against pollutants produced by aviation is of great concern in the 21st century. Among the multiplicity of proposed solutions, modifying flight profiles for existing aircraft is a promising approach. The aim is to deliver and understand the trade-off between environmental impact and operating costs. This work will illustrate the optimisation process of aircraft trajectories by minimising fuel consumption and flight time for the climb phase of an aircraft that belongs to A320 category. To achieve this purpose a new variant of a multi-objective Tabu Search optimiser was evolved and integrated within a computational framework, called GATAC, that simulates flight profiles based on altitude and speed. © 2013 Springer-Verlag.

The aerodynamic design of turbomachinery presents the design optimisation community with a number of exquisite challenges. Chief among these are the size of the design space and the extent of discontinuity therein. This discontinuity can serve to limit the full exploitation of high-fidelity computational fluid dynamics (CFD): such codes require detailed geometric information often available only sometime after the basic configuration of the machine has been set by other means. The premise of this paper is that it should be possible to produce higher performing designs in less time by exploiting multi-fidelity techniques to effectively harness CFD earlier in the design process, specifically by facilitating its participation in configuration selection. The adopted strategy of local multi-fidelity correction, generated on demand, combined with a global search algorithm via an adaptive trust region is first tested on a modest, smooth external aerodynamic problem. Speed-up of an order of magnitude is demonstrated, comparable to established techniques applied to smooth problems. A number of enhancements aimed principally at effectively evaluating a wide range of configurations quickly is then applied to the basic strategy, and the emerging technique is tested on a generic aeroengine core compression system. A similar order of magnitude speed-up is achieved on this relatively large and highly discontinuous problem. A five-fold increase in the number of configurations assessed with CFD is observed. As the technique places constraints neither on the underlying physical modelling of the constituent analysis codes nor on first-order agreement between those codes, it has potential applicability to a range of multidisciplinary design challenges. © 2012 by Jerome Jarrett and Tiziano Ghisu.

An increasin g interest in biofuel applications in modern engines requires a better understanding of biodiesel combustion behaviour. Many numerical studies have been carried out on unsteady combustion of biodiesel in situations similar to diesel engines, but very few studies have been done on the steady combustion of biodiesel in situations similar to a gas turbine combustor environment. The study of biodiesel spray combustion in gas turbine applications is of special interest due to the possible use of biodiesel in the power generation and aviation industries. In modelling spray combustion, an accurate representation of the physical properties of the fuel is a first important step, since spray formation is largely influenced by fuel properties such as viscosity, density, surface tension and vapour pressure. In the present work, a calculated biodiesel properties database based on the measured composition of Fatty Acid Methyl Esters (FAME) has been implemented in a multi-dimensional Computational Fluid Dynamics (CFD) spray simulation code. Simulations of non-reacting and reacting atmospheric-pressure sprays of both diesel and biodiesel have been carried out using a spray burner configuration for which experimental data is available. A pre-defined droplet size probability density function (pdf) has been implemented together with droplet dynamics based on phase Doppler anemometry (PDA) measurements in the near-nozzle region. The gas phase boundary condition for the reacting spray cases is similar to that of the experiment which employs a plain air-blast atomiser and a straight-vane axial swirler for flame stabilisation. A reaction mechanism for heptane has been used to represent the chemistry for both diesel and biodiesel. Simulated flame heights, spray characteristics and gas phase velocities have been found to compare well with the experimental results. In the reacting spray cases, biodiesel shows a smaller mean droplet size compared to that of diesel at a constant fuel mass flow rate. A lack of sensitivity towards different fuel properties has been observed based on the non-reacting spray simulations, which indicates a need for improved models of secondary breakup. By comparing the results of the non-reacting and reacting spray simulations, an improvement in the complexity of the physical modelling is achieved which is necessary in the understanding of the complex physical processes involved in spray combustion simulation. Growing concerns regarding fluctuating fuel costs and pollution targets for gas emissions, have led the aviation industry to seek alternative technologies to reduce its dependency on crude oil, and its net emissions. Recently blends of bio-fuel with kerosine, have become an alternative solution as they offer "greener" aircraft and reduce demand on crude oil. Interestingly, this technique is able to be implemented in current aircraft as it does not require any modification to the engine. Therefore, the present study investigates the effect of blends of bio-synthetic paraffinic kerosine with Jet-A in a civil aircraft engine, focusing on its performance and exhaust emissions. Two bio-fuels are considered: Jatropha Bio-synthetic Paraffinic Kerosine (JSPK) and Camelina Bio-synthetic Paraffinic Kerosine (CSPK); there are evaluated as pure fuels, and as 10% and 50% blend with Jet-A. Results obtained show improvement in thrust, fuel flow and SFC as composition of bio-fuel in the blend increases. At design point condition, results on engine emissions show reduction in NO x, and CO, but increases of CO is observed at fixed fuel condition, as the composition of bio-fuel in the mixture increases. Against a background of increasing energy demand and rising fuel prices, hybrid-electric propulsion systems (HEPS) have the potential to significantly reduce fuel consumption in the aviation industry, particularly in the lighter sectors. By taking advantage of both Electric Motor (EM) and Internal Combustion Engine (ICE), HEPS provide not only a benefit in fuel saving but also a reduction in take-off noise and the emission levels. This research considers the design and sizing process of a hybrid-electric propulsion system for a single-seat demonstrator aircraft, the experimental derivation of the ICE map and the EM parameters. In addition to the experimental data, a novel modeling approach including several linked desktop PC software packages is presented to analyze and optimize hybrid-electric technology for aircraft. Further to the analysis of a parallel hybrid-electric, mid-scale aircraft, this paper also presents a scaling approach for a 20 kg UAV and a 50 tonne inter-city airliner. At the smaller scale, two different mission profiles are analyzed: an ISR mission profile, where the simulation routine optimizes the component size of the hybrid-electric propulsion system with respect to fuel saving, and a maximum duration profile; where the flight endurance is determined as a function of payload weight. At the larger scale, the performance of a 50 tonne inter-city airliner is modeled, based on a hybrid-electric gas-turbine, assuming a range of electric boost powers and battery masses.

A recent study has shown that nonanoic acid (NA) is one of the strongest allelochemicals to a cyanobacterium Microcystis aeruginosa, but the physiological responses of M. aeruginosa to NA stress remain unknown. In this study, physiological characters such as the growth rate, photosynthetic processes, phosphorus and nitrogen uptake kinetics, and the contents of intracellular microcystin of M. aeruginosa PCC7806 were studied under the NA stress. The results showed that the growth rates of M. aeruginosa PCC 7806 were significantly inhibited in all NA stress treatments during first 3 days after exposure, and the growth rate was recovered after 5-day exposure. After 2-day exposure, the contents of both phycocyanin and allophycocyanin per cell decreased at NA concentration of 4 mg L-1, and oxygen evolution was inhibited even at the concentration of 0.5 mg L-1, but carotenoid content per cell was slightly boosted in NA stress. Physiological recovery of M. aeruginosa PCC7806 was observed after 7-day exposure to NA. It was shown that NA stress had no effect on uptake of nitrogen, but could stimulate the uptake of phosphorus. Polyfluorinated and perfluorinated compounds (PFCs) are used in numerous commercial products and have been ubiquitously detected in the environment as well as in the blood of humans and wildlife. To assess the combined effects caused by PFCs in mixtures, gene expression profiles were generated using a custom cDNA microarray to detect changes in primary cultured hepatocytes of rare minnows exposed to six individual PFCs (perfluorooctanoic acid, perfluorononanoic acid, perfluorodecanoic acid, perfluorododecanoic acid, perfluorooctane sulfonate, and 8:2 fluorotelomer alcohol) and four formulations of the PFCs mixtures. Mixtures as well as individual compounds consistently regulated a particular gene set, which suggests that these conserved genes may play a central role in the toxicity mediated by PFCs. Specifically, a number of genes regulated by the mixtures were identified in this study, which were not affected by exposure to any single component. These genes are implicated in multiple biological functions and processes, including fatty acid metabolism and transport, xenobiotic metabolism, immune responses, and oxidative stress. More than 80% of the altered genes in the PFOA- and PFOS-dominant mixture groups were of the same gene set, while the gene expression profiles from single PFOA and PFOS exposures were not as similar. This work contributes to the development of toxicogenomic approaches in combined toxicity assessment and allows for comprehensive insights into the combined action of PFCs mixtures in multiple environmental matrices. A liquid chromatography electrospray mass spectrometry (LC/ESI/MS) method working in multiple reactions monitoring mode for the determination of trace amounts of microcystin variants (MC-LR and [Dha(7)] MC-LR) in waters was developed. The limit of quantification was 0.05 mu g/L and the limit of detection was 0.015 mu g/L for MC-LR and [Dha(7)] MC-LR, respectively. Recoveries for MCs were in the range of 68%-81%. MC-LR and [Dha(7)] MC-LR were chemically stable with similar physiochemical behavior.

With the widespread exposure of people to nicotine through recreational use of tobacco products, research into nicotine has attracted increasing attention. Tobacco smoking is by far the most important cause of lung cancer. As the world's largest producer and consumer of tobacco products, China bears a large proportion of the global burden of smoking-related disease; therefore, information on nicotine publications should be collected to formulate future research policy. In the present study, we investigated nicotine-related research articles published by Chinese authors that were indexed in the Science Citation Index (SCI) from 1991 to 2007. An indicator "citations per publication" (CPP) was used in the study to evaluate the impact of journals, articles, and institutes. The quantity of publications has increased at a quicker pace than the worldwide trend. Article visibility, measured as the frequency of being cited, also increased during the period. However, the overall quality of articles, based on the impact factor of journals publishing those articles, dropped behind the worldwide average level. There has been an increase in international collaboration, mainly with researchers in the USA. The average CPP of international co-authorship articles was higher than that of single country publications. Besides the USA, nicotine research in China will benefit from more collaboration with Taiwan, England, and Germany. Some 110 of 264 articles were published by a single institute, and the top six institutes were compared from various angles. Seventy-two subject categories were covered, and trends (in terms of both quantity and quality) of nicotine research in China were compared with worldwide trends. In addition, analysis of keywords in both nicotine and lung cancer research fields was applied to indicate research interests. Mutual cooperation among multiple disciplines needs further strengthening.

The effects of organic-rich sediment and sulfide exposure on Hydrilla verticillata were investigated. The organic richness of sediment was simulated by adding sucrose into sediments, and sulfide exposure was conducted by adding sodium sulfide to plant roots. The length, biomass and density of shoot reduced in the sucrose-amended sediments, and the largest reduction occurred in the highest 1.0% addition treatment by 84.2%, 56.7% and 92.4%, respectively. However, the 0.1% addition treatment stimulated the growth of root. The effects of below-ground sulfide exposure on the physiological activities of H. verticillata were determined by adding sulfide to the below-ground tissue. Significantly inhibitory effects of sulfide were observed on plant photosynthesis, root carbohydrate and nitrogen synthetic reserves. The net photosynthetic rates, soluble carbohydrate and soluble protein contents in root were reduced by 104%, 71.8% and 49.8%, respectively, in the 0.6 mM sulfide treatment.

A 115 days feeding trial was conducted to evaluate the effect of dietary cyanobacteria on growth, microcystins (MCs) accumulation in hybrid tilapia (Oreochromis niloticus x O. aureus) and the recovery when the fish were free of cyanobacteria. Three experimental diets were formulated: the control (cyanobacteria free diet); one test diet with cyanobacteria from Lake Taihu (AMt 80.0 mu g MCs g(-1) diet) and one with cyanobacteria from Lake Dianchi (AMd, 410.0 rho g MCs g(-1) diet). Each diet was fed to fish for 60 days and then all fish were free of cyanobacteria for another 55 days. A significant increase in feeding rate (FR) was observed in fish fed AMd diet after a first 30-day exposure (1(st) EP), and in fish fed both AMt diet and AMd diet after a second 30-day exposure (2(nd) EP). Specific growth rates (SGR) of fish fed AMt diet and AMd diet were both obviously affected after the first 30-day exposure, but SGR was only significantly affected in fish fed AMt diet after the second 30-day exposure. After a 55-day recovery, there were no significant differences among diets in the indices mentioned above. Much higher concentrations of MCs were accumulated in tissues of all fish exposed to cyanobacteria. After the 55-day recovery, MC concentrations in fish tissues were significantly lower than those on day 60. (C) 2009 Elsevier Ltd. All rights reserved.

Fluorotelomer alcohols (FTOHs) have shown estrogenic activity in vitro and in vivo, but the mechanism of this activity is not known. In this study, 18-week-old zebrafish (Danio rerio) were exposed to 0, 0.03, 0.3 and 3.0 mg/l 1H, 1H, 2H, 2H-perfluorooctan-1-ol (6:2 ETCH) for 7 days, and the effects on plasma sex hormone levels were measured followed by use of real-time PCR to examine selected gene expression in hypothalamic-pituitary-gonadal (HPG) axis and liver. Exposure to 6:2 FTOH significantly increased plasma estradiol (E2) and testosterone (T) levels in both males and females. Furthermore, the ratio of T/E2 was reduced in females while increased in males. In females, the increase of E2 was accompanied by up-regulated hepatic estrogenic receptor alpha (ER alpha) and vitellogenin (VTG1 and VTG3) expression. In males, the elevation of the T level is consistent with the up-regulation of cytochrome P450 c17 alpha-hydroxylase, 17, 20-lase (CYP17) and the down-regulation of cytochrome P450 aromatase A (CYP19A). The present study demonstrated that waterborne exposure to 6:2 FTOH alter plasma sex hormone levels and the ratio of T/E2, as well as the transcriptional profiles of some genes in the HPG axis and liver. The results suggested that FTOHs may disturb fish reproduction through endocrine disrupted activity. (C) 2009 Elsevier B.V. All rights reserved.

Perfluorooctane sulfonate (PFOS) is widely distributed and persistent in the environment and in wildlife, and it has the potential for developmental toxicity. However, the molecular mechanisms that lead to these toxic effects are not well known. In the present study, proteomic analysis has been performed to investigate the proteins that are differentially expressed in zebrafish embryos exposed to 0.5 mg/l PFOS until 192 h postfertilization. Two-dimensional electrophoresis coupled with mass spectrometry was employed to detect and identify the protein profiles. The analysis revealed that 69 proteins showed altered expression in the treatment group compared to the control group with either increase or decrease in expression levels (more than twofold difference). Of the 69 spots corresponding to the proteins with altered expression, 38 were selected and subjected to matrix-assisted laser desorption/ionization tandem time-of-flight mass spectrometry (TOF/TOF) analysis; 18 proteins were identified in this analysis. These proteins can be categorized into diverse functional classes such as detoxification, energy metabolism, lipid transport/steroid metabolic process, cell structure, signal transduction, and apoptosis. Overall, proteomic analysis using zebrafish embryos serves as an in vivo model in environmental risk assessment and provides insight into the molecular events in PFOS-induced developmental toxicity.
