5-HT-moduline is an endogenous tetrapeptide [Leu-Ser-Ala-Leu (LSAL)] that was first isolated from bovine brain tissue. To understand the physiological role of this tetrapeptide, we studied the localization of 5-HT-moduline binding sites in rat and mouse brains. Quantitative data obtained with a gaseous detector of β-particles (β-imager) indicated that [3H]-5-HT-moduline bound specifically to rat brain sections with high affinity (Kd = 0.77 nM and Bmax = 0.26 dpm/mm2). Using film autoradiography in parallel, we found that 5-HT-moduline binding sites were expressed in a variety of rat and mouse brain structures. In 5-HT1B receptor knock-out mice, the specific binding of [3H]-5-HT-moduline was not different from background labeling, indicating that 5-HT-moduline targets are exclusively located on the 5-HT1B receptors. Although the distribution of 5-HT-moduline binding sites was similar to that of 5-HT1B receptors, they did not overlap totally. Differences in distribution patterns were found in regions containing either high levels of 5-HT1B receptors such as globus pallidus and subiculum that were poorly labeled or in other regions such as dentate gyrus of hippocampus and cortex where the relative density of 5-HT-moduline binding sites was higher than that of 5-HT1B receptors. In conclusion, our data, based on autoradiographic localization, indicate that 5-HT-moduline targets are located on 5-HT1B receptors present both on 5-HT afferents and postsynaptic neurons. By interacting specifically with 5-HT1B receptors, this tetrapeptide may play a pivotal role in pathological states such as stress that involves the dysfunction of 5-HT neurotransmission.

We are conducting a genome scan at an average resolution of 10 centimorgans (cM) for type 2 diabetes susceptibility genes in 716 affected sib pairs from 477 Finnish families. To date, our best evidence for linkage is on chromosome 20 with potentially separable peaks located on both the long and short arms. The unweighted multipoint maximum logarithm of odds score (MLS) was 3.08 on 20p (location, x̂ = 19.5 cM) under an additive model, whereas the weighted MLS was 2.06 on 20q (x̂ = 57 cM, recurrence risk, λ̂s = 1.25, P = 0.009). Weighted logarithm of odds scores of 2.00 (x̂ = 69.5 cM, P = 0.010) and 1.92 (x̂ = 18.5 cM, P = 0.013) were also observed. Ordered subset analyses based on sibships with extreme mean values of diabetes-related quantitative traits yielded sets of families who contributed disproportionately to the peaks. Two-hour glucose levels in offspring of diabetic individuals gave a MLS of 2.12 (P = 0.0018) at 9.5 cM. Evidence from this and other studies suggests at least two diabetes-susceptibility genes on chromosome 20. We have also screened the gene for maturity-onset diabetes of the young 1, hepatic nuclear factor 4-a (HNF-4α) in 64 affected sibships with evidence for high chromosomal sharing at its location on chromosome 20q. We found no evidence that sequence changes in this gene accounted for the linkage results we observed.

Hypoxanthine (H), the deamination product of adenine, has been implicated in the high frequency of A to G transitions observed in retroviral and other RNA genomes. Although H·C base pairs are thermodynamically more stable than other H·N pairs, polymerase selection may be determined in part by kinetic factors. Therefore, the hypoxanthine induced substitution pattern resulting from replication by viral polymerases may be more complex than that predicted from thermodynamics. We have examined the steady-state kinetics of formation of base pairs opposite template H in RNA by HIV-RT, and for the incorporation of dITP during first- and second-strand synthesis. Hypoxanthine in an RNA template enhances the k2app for pairing with standard dNTPs by factors of 10–1000 relative to adenine at the same sequence position. The order of base pairing preferences for H in RNA was observed to be H·C >> H·T > H·A > H·G. Steady-state kinetics of insertion for all possible mispairs formed with dITP were examined on RNA and DNA templates of identical sequence. Insertion of dITP opposite all bases occurs 2–20 times more frequently on RNA templates. This bias for higher insertion frequencies on RNA relative to DNA templates is also observed for formation of mispairs at template A. This kinetic advantage afforded by RNA templates for mismatches and pairing involving H suggests a higher induction of mutations at adenines during first-strand synthesis by HIV-RT.

O objetivo do presente trabalho é a investigação e o desenvolvimento de estratégias de otimização contínua e discreta para problemas de Fluxo de Potência Ótimo (FPO), onde existe a necessidade de se considerar as variáveis de controle associadas aos taps de transformadores em-fase e chaveamentos de bancos de capacitores e reatores shunt como variáveis discretas e existe a necessidade da limitação, e/ou até mesmo a minimização do número de ações de controle. Neste trabalho, o problema de FPO será abordado por meio de três estratégias. Na primeira proposta, o problema de FPO é modelado como um problema de Programação Não Linear com Variáveis Contínuas e Discretas (PNLCD) para a minimização de perdas ativas na transmissão; são propostas três abordagens utilizando funções de discretização para o tratamento das variáveis discretas. Na segunda proposta, considera-se que o problema de FPO, com os taps de transformadores discretos e bancos de capacitores e reatores shunts fixos, possui uma limitação no número de ações de controles; variáveis binárias associadas ao número de ações de controles são tratadas por uma função quadrática. Na terceira proposta, o problema de FPO é modelado como um problema de Otimização Multiobjetivo. O método da soma ponderada e o método ε-restrito são utilizados para modificar os problemas multiobjetivos propostos em problemas mono-objetivos. As variáveis binárias associadas às ações de controles são tratadas por duas funções, uma sigmoidal e uma polinomial. Para verificar a eficácia e a robustez dos modelos e algoritmos desenvolvidos serão realizados testes com os sistemas elétricos IEEE de 14, 30, 57, 118 e 300 barras. Todos os algoritmos e modelos foram implementados em General Algebraic Modeling System (GAMS) e os solvers CONOPT, IPOPT, KNITRO e DICOPT foram utilizados na resolução dos problemas. Os resultados obtidos confirmam que as estratégias de discretização são eficientes e as propostas de modelagem para variáveis binárias permitem encontrar soluções factíveis para os problemas envolvendo as ações de controles enquanto os solvers DICOPT e KNITRO utilizados para modelar variáveis binárias não encontram soluções.
