Com o objetivo de avaliar a contaminação do solo, sedimentos, água e plantas por metais pesados, decorrente do uso de agroquímicos, na microbacia de Caetés em Paty do Alferes, RJ, amostras de solo foram analisadas, em 1996, para obter os teores totais de Cd, Co, Cu, Mn, Ni, Pb e Zn de duas áreas com diferentes declividades (25% e 45%) tendo, ambas, como cobertura vegetal, capoeira, pasto e olericultura. Os resultados referentes aos solos sob pastagem (antiga área de olericultura) mostraram maiores teores de metais pesados do que a área de capoeira. Entretanto, esses valores não atingiram níveis críticos no solo, e esses elementos estavam presentes em formas químicas pouco disponíveis para absorção pelas plantas, como foi constatado na análise de metais pesados em tomate (Lycopersicum esculentum L.), pimentão (Capsicum annuum L.), repolho (Brassica oleracea L.) e pepino (Cucumis sativus L.), que apresentaram níveis baixíssimos desses elementos. Nas amostras de sedimentos, o comportamento dos metais pesados foi semelhante ao obtido nos solos, estando esses elementos distribuídos em maior porcentagem nas frações residual, ligadas a óxidos de Mn e Fe e ligadas à matéria orgânica. As amostras de água do córrego e do açude que cortam a microbacia apresentaram valores acima dos padrões internacionais com relação a Cd, Pb e Mn.

To identify common variants influencing body mass index (BMI), we analyzed genome-wide association data from 16,876 individuals of European descent. After previously reported variants in FTO, the strongest association signal (rs17782313, P = 2.9 x 10(-6)) mapped 188 kb downstream of MC4R (melanocortin-4 receptor), mutations of which are the leading cause of monogenic severe childhood-onset obesity. We confirmed the BMI association in 60,352 adults (per-allele effect = 0.05 Z-score units; P = 2.8 x 10(-15)) and 5,988 children aged 7-11 (0.13 Z-score units; P = 1.5 x 10(-8)). In case-control analyses (n = 10,583), the odds for severe childhood obesity reached 1.30 (P = 8.0 x 10(-11)). Furthermore, we observed overtransmission of the risk allele to obese offspring in 660 families (P (pedigree disequilibrium test average; PDT-avg) = 2.4 x 10(-4)). The SNP location and patterns of phenotypic associations are consistent with effects mediated through altered MC4R function. Our findings establish that common variants near MC4R influence fat mass, weight and obesity risk at the population level and reinforce the need for large-scale data integration to identify variants influencing continuous biomedical traits.

Whole-grain foods are touted for multiple health benefits, including enhancing insulin sensitivity and reducing type 2 diabetes risk. Recent genome-wide association studies (GWAS) have identified several single nucleotide polymorphisms (SNPs) associated with fasting glucose and insulin concentrations in individuals free of diabetes. We tested the hypothesis that whole-grain food intake and genetic variation interact to influence concentrations of fasting glucose and insulin. Via meta-analysis of data from 14 cohorts comprising ∼ 48,000 participants of European descent, we studied interactions of whole-grain intake with loci previously associated in GWAS with fasting glucose (16 loci) and/or insulin (2 loci) concentrations. For tests of interaction, we considered a P value <0.0028 (0.05 of 18 tests) as statistically significant. Greater whole-grain food intake was associated with lower fasting glucose and insulin concentrations independent of demographics, other dietary and lifestyle factors, and BMI (β [95% CI] per 1-serving-greater whole-grain intake: -0.009 mmol/l glucose [-0.013 to -0.005], P < 0.0001 and -0.011 pmol/l [ln] insulin [-0.015 to -0.007], P = 0.0003). No interactions met our multiple testing-adjusted statistical significance threshold. The strongest SNP interaction with whole-grain intake was rs780094 (GCKR) for fasting insulin (P = 0.006), where greater whole-grain intake was associated with a smaller reduction in fasting insulin concentrations in those with the insulin-raising allele. Our results support the favorable association of whole-grain intake with fasting glucose and insulin and suggest a potential interaction between variation in GCKR and whole-grain intake in influencing fasting insulin concentrations.

O objetivo deste trabalho foi avaliar o efeito residual dos herbicidas imazaquin, imazethapyr e diclosulam aplicados na cultura da soja sobre o girassol em sucessão. Nas parcelas, foi semeado o girassol aos 60 e 90 dias após a aplicação (DAA) do imazaquin e do diclosulam e 45 e 75 DAA do imazethapyr. As subparcelas foram compostas pelos herbicidas imazaquin (150 g/ha), imazethapyr (70 g/ha) e diclosulam (33,6 g/ha) aplicados na cultura da soja, além da testemunha (sem aplicação). O girassol, semeado aos 90 e aos 75 dias após a aplicação do imazaquin e do imazethapyr na cultura da soja, respectivamente, não apresenta sintomas de fitotoxicidade. O diclosulam causa redução total do estande de girassol nas duas épocas de semeadura. A lesão causada pelos herbicidas ao girassol, observada na produtividade da cultura, é maior em ordem decrescente de fitotoxicidade: diclosulam > imazaquin > imazethapyr.

Algodoeiros de fibras coloridas apresentam qualidade inferior em relação aos de fibra branca, além de menor potencial de rendimento, porque, comparados aos brancos, os coloridos foram pouco explorados em termos de melhoramento genético. Este trabalho teve como objetivo avaliar o efeito da seleção massal para porcentagem de fibra alta, média e baixa, dentro de uma população da cultivar BRS Verde, lançada em 2003. Quinhentas plantas da cultivar BRS Verde foram colhidas ao acaso e estratificadas em quatro classes quanto à porcentagem de fibra. As sementes de cada classe foram misturadas e participaram de um ensaio em blocos ao acaso com quatro tratamentos, mais a população original e quatro repetições. Ficou evidenciada a eficiência da seleção massal em alterar a média das populações selecionadas e o ganho de seleção foi de 11% quanto à porcentagem de fibra.

The work presented here is part of a larger study to identify novel technologies and biomarkers for early Alzheimer disease (AD) detection and it focuses on evaluating the suitability of a new approach for early AD diagnosis by non-invasive methods. The purpose is to examine in a pilot study the potential of applying intelligent algorithms to speech features obtained from suspected patients in order to contribute to the improvement of diagnosis of AD and its degree of severity. In this sense, Artificial Neural Networks (ANN) have been used for the automatic classification of the two classes (AD and control subjects). Two human issues have been analyzed for feature selection: Spontaneous Speech and Emotional Response. Not only linear features but also non-linear ones, such as Fractal Dimension, have been explored. The approach is non invasive, low cost and without any side effects. Obtained experimental results were very satisfactory and promising for early diagnosis and classification of AD patients.

Alzheimer's disease is the most prevalent form of progressive degenerative dementia; it has a high socio-economic impact in Western countries. Therefore it is one of the most active research areas today. Alzheimer's is sometimes diagnosed by excluding other dementias, and definitive confirmation is only obtained through a post-mortem study of the brain tissue of the patient. The work presented here is part of a larger study that aims to identify novel technologies and biomarkers for early Alzheimer's disease detection, and it focuses on evaluating the suitability of a new approach for early diagnosis of Alzheimer’s disease by non-invasive methods. The methods selected in this case are Automatic Spontaneous Speech Analysis (ASSA) and Emotional Temperature (ET), that have the great advantage of being non invasive, low cost and without any side effects.

O objetivo deste trabalho foi avaliar a produção e o desenvolvimento radicular e da parte aérea de plantas de café 'Conilon', provenientes de sementes e de estacas, no Município de Vila Valério, ES. Foi utilizado o delineamento experimental em blocos ao acaso com dois tratamentos (mudas provenientes de sementes e de estacas) e 12 repetições com cinco plantas úteis por parcela. As avaliações da parte aérea foram realizadas a partir de sete meses do plantio. Foram realizadas seis desbrotas e cinco colheitas. A avaliação do sistema radicular foi realizada aos 52 meses depois do plantio em quatro profundidades (0-10, 10-20, 20-40 e 40-60 cm). Foram quantificados o diâmetro, o comprimento e a área superficial de raízes. As plantas propagadas por estaca emitiram maior número de ramos plagiotrópicos, durante os sete primeiros meses de idade, e menor número de brotos ortotrópicos durante todo o experimento. Não houve diferenças no comprimento e na área superficial de raízes finas, entre plantas propagadas por sementes e por estacas. Houve maior concentração de raízes finas nas camadas superficiais do solo. A produtividade das plantas propagadas por estacas é maior do que a das plantas propagadas por sementes.

Numerous genetic loci have been associated with systolic blood pressure (SBP) and diastolic blood pressure (DBP) in Europeans. We now report genome-wide association studies of pulse pressure (PP) and mean arterial pressure (MAP). In discovery (N = 74,064) and follow-up studies (N = 48,607), we identified at genome-wide significance (P = 2.7 × 10(-8) to P = 2.3 × 10(-13)) four new PP loci (at 4q12 near CHIC2, 7q22.3 near PIK3CG, 8q24.12 in NOV and 11q24.3 near ADAMTS8), two new MAP loci (3p21.31 in MAP4 and 10q25.3 near ADRB1) and one locus associated with both of these traits (2q24.3 near FIGN) that has also recently been associated with SBP in east Asians. For three of the new PP loci, the estimated effect for SBP was opposite of that for DBP, in contrast to the majority of common SBP- and DBP-associated variants, which show concordant effects on both traits. These findings suggest new genetic pathways underlying blood pressure variation, some of which may differentially influence SBP and DBP.

BACKGROUND: Chronic endoplasmic reticulum (ER) stress contributes to the apoptotic cell death in the myocardium, thereby playing a critical role in the development of cardiomyopathy. ER stress has been reported to be induced after high-fat diet feeding in mice and also after saturated fatty acid treatment in vitro. Therefore, since several studies have shown that peroxisome proliferator-activated receptor (PPAR)β/δ inhibits ER stress, the main goal of this study consisted in investigating whether activation of this nuclear receptor was able to prevent lipid-induced ER stress in cardiac cells. METHODS AND RESULTS: Wild-type and transgenic mice with reduced PPARβ/δ expression were fed a standard diet or a high-fat diet for two months. For in vitro studies, a cardiomyocyte cell line of human origin, AC16, was treated with palmitate and the PPARβ/δ agonist GW501516. Our results demonstrate that palmitate induced ER stress in AC16 cells, a fact which was prevented after PPARβ/δ activation with GW501516. Interestingly, the effect of GW501516 on ER stress occurred in an AMPK-independent manner. The most striking result of this study is that GW501516 treatment also upregulated the protein levels of beclin 1 and LC3II, two well-known markers of autophagy. In accordance with this, feeding on a high-fat diet or suppression of PPARβ/δ in knockout mice induced ER stress in the heart. Moreover, PPARβ/δ knockout mice also displayed a reduction in autophagic markers. CONCLUSION: Our data indicate that PPARβ/δ activation might be useful to prevent the harmful effects of ER stress induced by saturated fatty acids in the heart by inducing autophagy.

O objetivo deste trabalho foi avaliar o acúmulo e a liberação de nutrientes (N, P, K, Ca, Mg e S) de resíduos culturais de plantas de cobertura na entressafra, em condições de Cerrado. O experimento foi conduzido em um Latossolo Vermelho distroférrico com textura argilosa. As plantas de cobertura avaliadas foram: amaranto (Amaranthus cruentus L.), milheto (Pennisetum glaucum L.) e capim-pé-de-galinha (Eleusine coracana (L.) Gaertn.). O delineamento experimental utilizado foi o de blocos ao acaso, no esquema de parcelas subdivididas, com quatro repetições. Na fase de florescimento das espécies, foi avaliada a produção de matéria seca e o acúmulo de nutrientes. A fim de avaliar a liberação de nutrientes dos resíduos culturais, o material vegetal de cada espécie foi acondicionado em sacolas de náilon, as quais foram dispostas sobre o solo e seu conteúdo analisado em intervalos de 30 dias, até 240 dias após sua instalação. As maiores quantidades de nutrientes acumulados na fitomassa das plantas de cobertura foram observadas no milheto e no capim-pé-de-galinha. O potássio foi o nutriente acumulado em maior quantidade, chegando a atingir 416,9 kg ha-1 no milheto. As maiores taxas de liberação de nutrientes foram observadas nos resíduos culturais do amaranto.

Gene-lifestyle interactions have been suggested to contribute to the development of type 2 diabetes. Glucose levels 2 h after a standard 75-g glucose challenge are used to diagnose diabetes and are associated with both genetic and lifestyle factors. However, whether these factors interact to determine 2-h glucose levels is unknown. We meta-analyzed single nucleotide polymorphism (SNP) × BMI and SNP × physical activity (PA) interaction regression models for five SNPs previously associated with 2-h glucose levels from up to 22 studies comprising 54,884 individuals without diabetes. PA levels were dichotomized, with individuals below the first quintile classified as inactive (20%) and the remainder as active (80%). BMI was considered a continuous trait. Inactive individuals had higher 2-h glucose levels than active individuals (β = 0.22 mmol/L [95% CI 0.13-0.31], P = 1.63 × 10(-6)). All SNPs were associated with 2-h glucose (β = 0.06-0.12 mmol/allele, P ≤ 1.53 × 10(-7)), but no significant interactions were found with PA (P > 0.18) or BMI (P ≥ 0.04). In this large study of gene-lifestyle interaction, we observed no interactions between genetic and lifestyle factors, both of which were associated with 2-h glucose. It is perhaps unlikely that top loci from genome-wide association studies will exhibit strong subgroup-specific effects, and may not, therefore, make the best candidates for the study of interactions.

O objetivo deste trabalho foi avaliar a eficiência de solubilização de FePO4 por Aspergillus niger, em meio de cultura com diferentes fontes de carbono (C) e nitrogênio (N) e com concentrações crescentes de fosfato. A quantidade de fosfato solúvel, acidez e pH final foram determinados após o crescimento do fungo, em cultura estacionária a 30ºC. A eficiência de solubilização aumentou conforme o crescimento do fungo, atingiu o máximo no 11º dia (68%) e depois regrediu. Das fontes de C e de N testadas, as maiores eficiências de solubilização foram obtidas com manitol (21%) e ácido glutâmico (17%). Com o aumento da concentração de fosfato (0 a 1.330 µg mL-1), a máxima eficiência de solubilização (70%) foi obtida com 330 µg mL-1 de PO4(3-) e decresceu até 47% com 1.330 µg mL-1. A produção de ácidos foi o principal mecanismo de solubilização do FePO4, com base na correlação positiva e significativa entre a produção de fosfato e a acidez.

The objective of this work was to determine the inheritance of cotton blue disease resistance by cotton plants. Populations derived from the CD 401 and Delta Opal resistant varieties were evaluated, through a greenhouse test with artificial inoculation by viruliferous aphids. Cotton blue disease resistance is conditioned by one dominant gene, both in CD 401 and Delta Opal varieties.
