Professional cleaning is a basic service occupation with a wide variety of tasks carried out in all kind of different sectors and workplaces by a large workforce. One important risk for cleaning workers is the exposure to chemical substances that are present in cleaning products.Monoethanolamine was found to be often present in cleaning products such as general purpose cleaners, bathroom cleaners, floor cleaners and kitchen cleaners. Monoethanolamine can injure the skin, and exposure to monoethanolamine was associated to asthma even when the air concentrations were low. It is a strong irritant and known to be involved in sensitizing mechanisms. It is very likely that the use of cleaning products containing monoethanolamine gives rise to respiratory and dermal exposures. Therefore there is a need to further investigate the exposures to monoethanolamine for both, respiratory and dermal exposure.The determination of monoethanolamine has traditionally been difficult and analytical methods available are little adapted for occupational exposure assessments. For monoethanolamine air concentrations, a sampling and analytical method was already available and could be used. However, a method to analyses samples for skin exposure assessments as well as samples of skin permeation experiments was missing. Therefore one main objective of this master thesis was to search an already developed and described analytical method for the measurement of monoethanolamine in water solutions, and to set it up in the laboratory. Monoethanolamine was analyzed after a derivatisation reaction with o-pthtaldialdehyde. The derivated fluorescing monoethanolamine was then separated with high performance liquid chromatography and detection took place with a fluorescent detector. The method was found to be suitable for qualitative and quantitative analysis of monoethanolamine. An exposure assessment was conducted in the cleaning sector to measure the respiratory and dermal exposures to monoethanolamine during floor cleaning. Stationary air samples (n=36) were collected in 8 companies and samples for dermal exposures (n=12) were collected in two companies. Air concentrations (Mean = 0.18 mg/m3, Standard Deviation = 0.23 mg/m3, geometric Mean = 0.09 mg/m3, Geometric Standard Deviation = 3.50) detected were mostly below 1/10 of the Swiss 8h time weighted average occupational exposure limit. Factors that influenced the measured monoethanolamine air concentrations were room size, ventilation system and the concentration of monoethanolamine in the cleaning product and amount of monoethanolamine used. Measured skin exposures ranged from 0.6 to 128.4 mg/sample. Some cleaning workers that participated in the skin exposure assessment did not use gloves and had direct contact with the solutions containing the cleaning product and monoethanolamine. During the entire sampling campaign, cleaning workers mostly did not use gloves. Cleaning workers are at risk to be regularly exposed to low air concentrations of monoethanolamine. This exposure may be problematic if a worker suffers from allergic reactions (e.g. Asthma). In that case a substitution of the cleaning product may be a good prevention measure as several different cleaning products are available for similar cleaning tasks. Currently there are no occupational exposure limits to compare the skin exposures that were found. To prevent skin exposures, adaptations of the cleaning techniques and the use of gloves should be considered. The simultaneous skin and airborne exposures might accelerate adverse health effects. Overall the risks caused by exposures to monoethanolamine are considered as low to moderate when the cleaning products are used correctly. Whenever possible, skin exposures should be avoided. Further research should consider especially the dermal exposure routes, as very high exposures might occur by skin contact with cleaning products. Dermatitis but also sensitization might be caused by skin exposures. In addition, new biomedical insights are needed to better understand the risks of the dermal exposure. Therefore skin permeability experiments should be considered.

BACKGROUND: Environmental conditions play a crucial role in mite growth, and optimal environmental control is key in the prevention of airway inflammation in chronic allergic rhinoconjunctivitis or asthma. OBJECTIVE: To evaluate the relationship between building energy performance and indoor mite allergen concentration in a cross-sectional study. METHODS: Major allergen concentration (Der f 1, Der p 1, mite group 2, Fel d 1 and Bla g 2) was determined by quantitative dot blot analysis from mattress and carpet dust samples in five buildings designed for low energy use (LEB) and in six control buildings (CB). Inhabitants had received 4 weeks prior to mite measurement a personal validated questionnaire related to the perceived state of health and comfort of living. RESULTS: Cumulative mite allergen concentration (with Der f 1 as the major contributor) was significantly lower in LEB as compared with CB both in mattresses and in carpets. In contrast, the two categories of buildings did not differ in Bla g 2 and Fel d 1 concentration, in the amount of dust and airborne mould collected. Whereas temperature was higher in LEB, relative humidity was significantly lower than in CB. Perceived overall comfort was better in LEB. CONCLUSIONS: Major mite allergen Der f 1 preferentially accumulates in buildings not specifically designed for low energy use, reaching levels at risk for sensitization. We hypothesize that controlled mechanical ventilation present in all audited LEB may favour lower air humidity and hence lower mite growth and allergen concentration, while preserving optimal perceived comfort.

Airborne transmission of Pneumocystis sp. from host to host has been demonstrated in rodent models and several observations suggest that interindividual transmission occurs in humans. Moreover, it is accepted that the Pneumocystis organisms infecting each mammalian species are host specific and that the hypothesis of an animal reservoir for Pneumocystis jirovecii (P. jirovecii), the human-specific Pneumocystis species, can be excluded. An exosaprophytic form of the fungus cannot be strictly ruled out. However, these data point toward the potential for the specific host to serve as its own reservoir and for Pneumocystis infection in humans as an anthroponosis with humans as a reservoir for P. jirovecii. This review highlights the main data on host-to-host transmission of Pneumocystis in rodent models and in humans by the airborne route and provides a rationale for considering the occurrence of nosocomial infections and measures for their prevention

Airborne particles can come from a variety of sources and contain variable chemical constituents. Some particles are formed by natural processes, such as volcanoes, erosion, sea spray, and forest fires, while other are formed by anthropogenic processes, such as industrial- and motor vehicle-related combustion, road-related wear, and mining. In general, larger particles (those greater than 2.5 μm) are formed by mechanical processes, while those less than 2.5 μm are formed by combustion processes. The chemical composition of particles is highly influenced by the source: for combustion-related particles, factors such as temperature of combustion, fuel type, and presence of oxygen or other gases can also have a large impact on PM composition. These differences can often be observed at a regional level, such as the greater sulphate-composition of PM in regions that burn coal for electricity production (which contains sulphur) versus regions that do not. Most countries maintain air monitoring networks, and studies based on the resulting data are the most common basis for epidemiology studies on the health effects of PM. Data from these monitoring stations can be used to evaluate the relationship between community-level exposure to ambient particles and health outcomes (i.e., morbidity or mortality from various causes). Respiratory and cardiovascular outcomes are the most commonly assessed, although studies have also considered other related specific outcomes such as diabetes and congenital heart disease. The data on particle characteristics is usually not very detailed and most often includes some combination of PM2.5, PM10, sulphate, and NO2. Other descriptors that are less commonly found include particle number (ultrafine particles), metal components of PM, local traffic intensity, and EC/OC. Measures of association are usually reported per 10 μg/m3 or interquartile range increase in pollutant concentration. As the exposure data are taken from regional monitoring stations, the measurements are not representative of an individual's exposure. Particle size is an important descriptor for understanding where in the human respiratory system the particles will deposit: as a general rule, smaller particles penetrate to deeper regions of the lungs. Initial studies on the health effects of particulate matter focused on mass of the particles, including either all particles (often termed total suspended particulate or TSP) or PM10 (all particles with an aerodynamic diameter less than 10 μm). More recently, studies have considered both PM10 and PM2.5, with the latter corresponding more directly to combustion-related processes. UFPs are a dominant source of particles in terms of PNC, yet are negligible in terms of mass. Very few epidemiology studies have measured the effect of UFPs on health; however, the numbers of studies on this topic are increasing. In addition to size, chemical composition is of importance when understanding the toxicity of particles. Some studies consider the composition of particles in addition to mass; however this is not common, in part due the cost and labour involved in such analyses.

The presence of hydrogen in polysilicon films obtained at low temperatures by hot-wire CVD and the post-deposition oxidation by air-exposure of the films are studied in this paper. The experimental results from several characterization techniques (infrared spectroscopy, X-ray photoelectron spectroscopy, secondary ion mass spectrometry and wavelength dispersive spectroscopy) showed that hydrogen and oxygen are homogeneously distributed at grain boundaries throughout the depth of the films. Hydrogen is introduced during the growth process and its concentration is higher in samples deposited at lower temperatures. Oxygen diffuses along the grain boundaries and binds to silicon atoms, mainly in Si 2O groups.

The main objective of this study was todo a statistical analysis of ecological type from optical satellite data, using Tipping's sparse Bayesian algorithm. This thesis uses "the Relevence Vector Machine" algorithm in ecological classification betweenforestland and wetland. Further this bi-classification technique was used to do classification of many other different species of trees and produces hierarchical classification of entire subclasses given as a target class. Also, we carried out an attempt to use airborne image of same forest area. Combining it with image analysis, using different image processing operation, we tried to extract good features and later used them to perform classification of forestland and wetland.

Uimaveden klooridesinfioinnissa syntyy sivutuotteena haihtuvia ja haitallisia halogeeniyhdisteitä, kuten trihalometaaneja ja triklooriamiinia, jotka voivat heikentää allas-tilan sisäilman laatua merkittävästi. Tämän tutkimuksen tavoitteena oli kartoittaa näiden desinfioinnin sivutuotteiden pitoisuuksia suomalaisissa uimahalleissa sekä selvittää epäpuhtauksien kulkeutumista allastiloissa. Lisäksi pyrittiin löytämään merkittävimmät veden laatu- ja käsittelyparametrit sekä ilmanvaihtotekniset tekijät, jotka vaikuttavat vedestä haihtuvien epäpuhtauksien pitoisuuksiin hallitiloissa. Mittaukset tehtiin kymmenessä eri puolilla Suomea sijaitsevassa uimahallissa. Mittausten perusteella havaittiin, että allastilojen kloroformipitoisuudet vaihtelivat välillä 8,9-84,0 ¿g/m3. Terapia-allasostoilta mitatut pitoisuudet olivat pääallastiloista mitattuja pitoisuuksia suurempia ja aamulla mitattu pitoisuus alhaisempi kuin illalla mitattu. Lisäksi ilmastoiduista valvomoista mitatut pitoisuudet olivat merkittävästi allastilojen pitoisuuksia pienempiä. Triklooriamiininäytteistä suurin osa oli alle määritysrajan. Sisäilman kloroformipitoisuuden havaittiin korreloivan veden lämpötilan sekä ilman kosteuden kanssa. Triklooriamiinille tilastollista analyysiä ei voitu tehdä mm. määritys-rajan alle jääneiden näytteiden suuren osuuden vuoksi. Teknisten kyselyiden ja ilmanvaihtomittausten perusteella todettiin, että uimahallien ilmanvaihto toimii lämmitystarpeen vuoksi sekoittavana ja epäpuhtauksien leviämistä allastilassa ei voida käytännössä katsoen estää. Ilman virtausnopeudet pääaltaiden reunoilla olivat pieniä ja ilman virtauskenttien ei todettu vaikuttavan epäpuhtauksien kulkeutumiseen allastiloissa. Mittauskohteiden vähyydestä ja vedenkäsittelyn hallikohtaisista erityispiirteistä johtuen luotettavia johtopäätöksiä vedenkäsittelymenetelmien vaikutuksesta allasveden ja allastilan ilman laatuun ei pystytty tekemään.

Puhdastilojen suunnittelussa pyritään saamaan hallittu ja valvottu ilmanpuhtaus luokiteltuun tilaan.Luokittelu tapahtuu puhdastilastandardeilla, lisäksi lääkevalmisteita valmistettavassa tilassa GMP -säädösten mukaisin luokituksin. Puhdastilastandardi ISO 14644 käsittää seitsemän osaa, jossa on käsitelty puhdastilaa koskevia määräyksiä suunnittelusta käyttöön ja testaukseen. GMP-säädökset sisältävät yhdeksän kappaletta, joista kappale 3: 'Tilat ja laitteet' on keskeinen osa lääkeainevalmistuksen puhdastilasuunnittelua. Puhtaan ilman aikaansaamiseksi puhdastilaan merkittävimmät roolit ovat ilmanvaihdolla, puhdastilarakenteilla ja rakennusautomaatiolla. Ilma voidaan tuoda tilaan kolmella eri periaatteella. Ilmaa tuodaan tilaan yhdensuuntaisesti, turbulenttisesti tai sekavirtauksena HEPA -suodattimien kautta, joilla varmistetaan epäpuhtauksien korkea suodatusaste. Ilmapoistetaan rei'itettyjen, korotettujen lattioiden kautta tai tilan alaosassa olevien poistoilmasäleikköjen kautta, josta se johdetaan noin 75-90%:sti kierrätettynä takaisin tilaan. Lääketeollisuudessa rei'itettyjä, korotettuja lattioita eivoida käyttää kontaminaatiovaaran, vuoksi. Tilaan suunniteltuja olosuhteita ylläpidetään rakennusautomaation avulla ja monitorointijärjestelmällä valvotaan tilassa olevan ilman laatua. Kaikki GMP-luokituksen mukaiset puhdastilat tulee validoida. Validointiin kuuluu teknisten järjestelmien kvalifiointi ja koko prosessin validointi. Teknisten järjestel-mien kvalifiointi käsittää suunnitelmien tarkastuksen (DQ), asennus - ja käyttöönotto tarkastukset (IQ), toiminnan testauksen (OQ) ja suorituksen testauksen (PQ). Kvali-fiointi kuuluu yhtenä osa-alueena validointiin. Prosessin validointi on osa yrityksen laadunvarmistusta. Validoinnilla hankitaan dokumentoidut todisteet siitä, että tila tai prosessi todella täyttää annetut vaatimukset. Tässä työssä laadittiin esimerkinomainen kvalifiointisuunnitelma puhdastilan tekni-sille järjestelmille. Suunnitelma sisältää asennus- ja käyttöönoton mukaiset tarkastukset (IQ)ja toiminnan aikaiset testaukset (OQ).

Steep mountain catchments typically experience large sediment pulses from hillslopes which are stored in headwater channels and remobilized by debris-flows or bedload transport. Event-based sediment budget monitoring in the active Manival debris-flow torrent in the French Alps during a two-year period gave insights into the catchment-scale sediment routing during moderate rainfall intensities which occur several times each year. The monitoring was based on intensive topographic resurveys of low- and high-order channels using different techniques (cross-section surveys with total station and high-resolution channel surveys with terrestrial and airborne laser scanning). Data on sediment output volumes from the main channel were obtained by a sediment trap. Two debris-flows were observed, as well as several bedload transport flow events. Sediment budget analysis of the two debris-flows revealed that most of the debris-flow volumes were supplied by channel scouring (more than 92%). Bedload transport during autumn contributed to the sediment recharge of high-order channels by the deposition of large gravel wedges. This process is recognized as being fundamental for debris-flow occurrence during the subsequent spring and summer. A time shift of scour-and-fill sequences was observed between low- and high-order channels, revealing the discontinuous sediment transfer in the catchment during common flow events. A conceptual model of sediment routing for different event magnitude is proposed.

Diplomityön tavoitteena oli tutkia selluradan leijutusta sekä hallintaa kapealla ja täysleveällä selluradalla ilmakuivaimessa. Työn sisältö on jaettu neljään osaan teoriaan, numeeriseen virtauslaskentaan, kokeelliseen osuuteen ja johtopäätöksiin. Työn alussa kiinnitettiin huomiota ilmakuivaimen ja selluradan rakenteeseen, selluradan lujuusominaisuuksiin ja niiden testaamiseen laboratorio-oloissa. Teoria osuudessa tarkasteltiin selluradan leijutusteoriaa ja siihen liittyvää virtaustekniikkaa. Leijutusta kuvattiin mekaniikasta tutun vapaakappalekuvan muodossa. Vapaakappalekuva auttaa ymmärtämään sellurataan vaikuttavia voimia ja niiden aiheuttajia. Numeerisessa virtauslaskennassa rakennettiin Fluent -virtauslaskentamalli. Malli kuvaa leijutussuuttimen alusta 0,2 metrin pituista pätkää. Laskennassa simuloitiin selluradan lei-jutusta 2, 5 ja 7 mm etäisyyksillä. Tuloksia tarkasteltiin paine- ja nopeusjakaumien muodossa. Kokeellisessa osuudessa tehtiin mittauksia yksittäisellä ylä- ja alasuuttimella sekä suutinpaketilla. Yksittäisistä suuttimista mitattiin ilmasuihkujen ulostulokulmaa, suuttimensisäistä painejakaumaa ja selluradan pintaan vaikuttavaa staattista painetta suuttimen pituuden funktiona. Suutinpaketilla tutkittiin ilmasuihkujen ja tasomaisen levyn välistä vuorovaikutusta. Ylä- ja alasuuttimen välissä leijutettiin levyä, josta mitattiin ilmasuihkujen aiheuttama z-suuntainen voima ja leijutuskorkeus. Työn lopussa arvioitiin teorian, numeerisen virtauslaskennan ja kokeellisen osuuden tuloksia. Näiden pohjalta tehtiin teknisiä parannusehdotuksia ilmakuivaimen toiminnan tehostamiseksi ja selluradan hallinnan parantamiseksi kapealla ja täysleveällä selluradalla.

Prosessiteollisuudessa syntyviä ilmapäästöjä seurataan jatkuvatoimisilla mittausjärjestelmillä. Jatkuvatoimisen päästöjen seurantajärjestelmän tulee tuottaa luotettavaa tietoa prosessin aiheuttamista päästöistä. Päästöjen seurannasta saataviin tuloksiin syntyy aina epävarmuutta, joka koostuu mittausolosuhteiden, mittausjärjestelmän tai mittauksen suorittamisen aiheuttamista epävarmuuksista. Työn alussa on kuvattu Stora Enso Oyj:n Imatran tehtaitten typen oksidien jatkuvatoiminen päästöjen seurantamenetelmä ja selvitetty seurantajärjestelmän suorittama päästöjen laskentamenetelmä. Lisäksi on tuotu esille päästömittaustuloksen epävarmuuteen vaikuttavia tekijöitä kyseisessä järjestelmässä. Selvityksessä merkittävimmäksi järjestelmän luotettavuuteen vaikuttavaksi tekijäksi osoittautui virheellinen mittaustieto, jota voidaan hallita järjestelmässä asettamalla raja-arvo mittaustiedolle ja tehostamalla käytönaikaista valvontaa. Lisäksi korvaamalla häiriöaikainen virheellinen mittaustieto laskennallisella pitoisuuden odotusarvolla päästölaskentaa varten, tuottaa seurantajärjestelmä luotettavampia typen oksidien päästötietoja raportointia varten.

Engineered nanomaterials (ENMs) exhibit special physicochemical properties and thus are finding their way into an increasing number of industries, enabling products with improved properties. Their increased use brings a greater likelihood of exposure to the nanoparticles (NPs) that could be released during the life cycle of nano-abled products. The field of nanotoxicology has emerged as a consequence of the development of these novel materials, and it has gained ever more attention due to the urgent need to gather information on exposure to them and to understand the potential hazards they engender. However, current studies on nanotoxicity tend to focus on pristine ENMs, and they use these toxicity results to generalize risk assessments on human exposure to NPs. ENMs released into the environment can interact with their surroundings, change characteristics and exhibit toxicity effects distinct from those of pristine ENMs. Furthermore, NPs' large surface areas provide extra-large potential interfaces, thus promoting more significant interactions between NPs and other co-existing species. In such processes, other species can attach to a NP's surface and modify its surface functionality, in addition to the toxicity in normally exhibits. One particular occupational health scenario involves NPs and low-volatile organic compounds (LVOC), a common type of pollutant existing around many potential sources of NPs. LVOC can coat a NP's surface and then dominate its toxicity. One important mechanism in nanotoxicology is the creation of reactive oxygen species (ROS) on a NP's surface; LVOC can modify the production of these ROS. In summary, nanotoxicity research should not be limited to the toxicity of pristine NPs, nor use their toxicity to evaluate the health effects of exposure to environmental NPs. Instead, the interactions which NPs have with other environmental species should also be considered and researched. The potential health effects of exposure to NPs should be derived from these real world NPs with characteristics modified by the environment and their distinct toxicity. Failure to suitably address toxicity results could lead to an inappropriate treatment of nano- release, affect the environment and public health and put a blemish on the development of sustainable nanotechnologies as a whole. The main objective of this thesis is to demonstrate a process for coating NP surfaces with LVOC using a well-controlled laboratory design and, with regard to these NPs' capacity to generate ROS, explore the consequences of changing particle toxicity. The dynamic coating system developed yielded stable and replicable coating performance, simulating an important realistic scenario. Clear changes in the size distribution of airborne NPs were observed using a scanning mobility particle sizer, were confirmed using both liquid nanotracking analyses and transmission electron microscopy (TEM) imaging, and were verified thanks to the LVOC coating. Coating thicknesses corresponded to the amount of coating material used and were controlled using the parameters of the LVOC generator. The capacity of pristine silver NPs (Ag NPs) to generate ROS was reduced when they were given a passive coating of inert paraffin: this coating blocked the reactive zones on the particle surfaces. In contrast, a coating of active reduced-anthraquinone contributed to redox reactions and generated ROS itself, despite the fact that ROS generation due to oxidation by Ag NPs themselves was quenched. Further objectives of this thesis included development of ROS methodology and the analysis of ROS case studies. Since the capacity of NPs to create ROS is an important effect in nanotoxicity, we attempted to refine and standardize the use of 2'7-dichlorodihydrofluorescin (DCFH) as a chemical tailored for the characterization of NPs' capacity for ROS generation. Previous studies had reported a wide variety of results, which were due to a number of insufficiently well controlled factors. We therefore cross-compared chemicals and concentrations, explored ways of dispersing NP samples in liquid solutions, identified sources of contradictions in the literature and investigated ways of reducing artificial results. The most robust results were obtained by sonicating an optimal sample of NPs in a DCFH-HRP solution made of 5,M DCFH and 0.5 unit/ml horseradish peroxidase (HRP). Our findings explained how the major reasons for previously conflicting results were the different experimental approaches used and the potential artifacts appearing when using high sample concentrations. Applying our advanced DCFH protocol with other physicochemical characterizations and biological analyses, we conducted several case studies, characterizing aerosols and NP samples. Exposure to aged brake wear dust engenders a risk of potential deleterious health effects in occupational scenarios. We performed microscopy and elemental analyses, as well as ROS measurements, with acellular and cellular DCFH assays. TEM images revealed samples to be heterogeneous mixtures with few particles in the nano-scale. Metallic and non-metallic elements were identified, primarily iron, carbon and oxygen. Moderate amounts of ROS were detected in the cell-free fluorescent tests; however, exposed cells were not dramatically activated. In addition to their highly aged state due to oxidation, the reason aged brake wear samples caused less oxidative stress than fresh brake wear samples may be because of their larger size and thus smaller relative reactive surface area. Other case studies involving welding fumes and differently charged NPs confirmed the performance of our DCFH assay and found ROS generation linked to varying characteristics, especially the surface functionality of the samples. Les nanomatériaux manufacturés (ENM) présentent des propriétés physico-chimiques particulières et ont donc trouvés des applications dans un nombre croissant de secteurs, permettant de réaliser des produits ayant des propriétés améliorées. Leur utilisation accrue engendre un plus grand risque pour les êtres humains d'être exposés à des nanoparticules (NP) qui sont libérées au long de leur cycle de vie. En conséquence, la nanotoxicologie a émergé et gagné de plus en plus d'attention dû à la nécessité de recueillir les renseignements nécessaires sur l'exposition et les risques associés à ces nouveaux matériaux. Cependant, les études actuelles sur la nanotoxicité ont tendance à se concentrer sur les ENM et utiliser ces résultats toxicologiques pour généraliser l'évaluation des risques sur l'exposition humaine aux NP. Les ENM libérés dans l'environnement peuvent interagir avec l'environnement, changeant leurs caractéristiques, et montrer des effets de toxicité distincts par rapport aux ENM originaux. Par ailleurs, la grande surface des NP fournit une grande interface avec l'extérieur, favorisant les interactions entre les NP et les autres espèces présentes. Dans ce processus, d'autres espèces peuvent s'attacher à la surface des NP et modifier leur fonctionnalité de surface ainsi que leur toxicité. Un scénario d'exposition professionnel particulier implique à la fois des NP et des composés organiques peu volatils (LVOC), un type commun de polluant associé à de nombreuses sources de NP. Les LVOC peuvent se déposer sur la surface des NP et donc dominer la toxicité globale de la particule. Un mécanisme important en nanotoxicologie est la création d'espèces réactives d'oxygène (ROS) sur la surface des particules, et les LVOC peuvent modifier cette production de ROS. En résumé, la recherche en nanotoxicité ne devrait pas être limitée à la toxicité des ENM originaux, ni utiliser leur toxicité pour évaluer les effets sur la santé de l'exposition aux NP de l'environnement; mais les interactions que les NP ont avec d'autres espèces environnementales doivent être envisagées et étudiées. Les effets possibles sur la santé de l'exposition aux NP devraient être dérivés de ces NP aux caractéristiques modifiées et à la toxicité distincte. L'utilisation de résultats de toxicité inappropriés peut conduire à une mauvaise prise en charge de l'exposition aux NP, de détériorer l'environnement et la santé publique et d'entraver le développement durable des industries de la nanotechnologie dans leur ensemble. L'objectif principal de cette thèse est de démontrer le processus de déposition des LVOC sur la surface des NP en utilisant un environnement de laboratoire bien contrôlé et d'explorer les conséquences du changement de toxicité des particules sur leur capacité à générer des ROS. Le système de déposition dynamique développé a abouti à des performances de revêtement stables et reproductibles, en simulant des scénarios réalistes importants. Des changements clairs dans la distribution de taille des NP en suspension ont été observés par spectrométrie de mobilité électrique des particules, confirmé à la fois par la méthode dite liquid nanotracking analysis et par microscopie électronique à transmission (MET), et a été vérifié comme provenant du revêtement par LVOC. La correspondance entre l'épaisseur de revêtement et la quantité de matériau de revêtement disponible a été démontré et a pu être contrôlé par les paramètres du générateur de LVOC. La génération de ROS dû aux NP d'argent (Ag NP) a été diminuée par un revêtement passif de paraffine inerte bloquant les zones réactives à la surface des particules. Au contraire, le revêtement actif d'anthraquinone réduit a contribué aux réactions redox et a généré des ROS, même lorsque la production de ROS par oxydation des Ag NP avec l'oxygène a été désactivé. Les objectifs associés comprennent le développement de la méthodologie et des études de cas spécifique aux ROS. Etant donné que la capacité des NP à générer des ROS contribue grandement à la nanotoxicité, nous avons tenté de définir un standard pour l'utilisation de 27- dichlorodihydrofluorescine (DCFH) adapté pour caractériser la génération de ROS par les NP. Des etudes antérieures ont rapporté une grande variété de résultats différents, ce qui était dû à un contrôle insuffisant des plusieurs facteurs. Nous avons donc comparé les produits chimiques et les concentrations utilisés, exploré les moyens de dispersion des échantillons HP en solution liquide, investigué les sources de conflits identifiées dans les littératures et étudié les moyens de réduire les résultats artificiels. De très bon résultats ont été obtenus par sonication d'une quantité optimale d'échantillons de NP en solution dans du DCFH-HRP, fait de 5 nM de DCFH et de 0,5 unité/ml de Peroxydase de raifort (HRP). Notre étude a démontré que les principales raisons causant les conflits entre les études précédemment conduites dans la littérature étaient dues aux différentes approches expérimentales et à des artefacts potentiels dus à des concentrations élevées de NP dans les échantillons. Utilisant notre protocole DCFH avancé avec d'autres caractérisations physico-chimiques et analyses biologiques, nous avons mené plusieurs études de cas, caractérisant les échantillons d'aérosols et les NP. La vielle poussière de frein en particulier présente un risque élevé d'exposition dans les scénarios professionnels, avec des effets potentiels néfastes sur la santé. Nous avons effectué des analyses d'éléments et de microscopie ainsi que la mesure de ROS avec DCFH cellulaire et acellulaire. Les résultats de MET ont révélé que les échantillons se présentent sous la forme de mélanges de particules hétérogènes, desquels une faible proportion se trouve dans l'échelle nano. Des éléments métalliques et non métalliques ont été identifiés, principalement du fer, du carbone et de l'oxygène. Une quantité modérée de ROS a été détectée dans le test fluorescent acellulaire; cependant les cellules exposées n'ont pas été très fortement activées. La raison pour laquelle les échantillons de vielle poussière de frein causent un stress oxydatif inférieur par rapport à la poussière de frein nouvelle peut-être à cause de leur plus grande taille engendrant une surface réactive proportionnellement plus petite, ainsi que leur état d'oxydation avancé diminuant la réactivité. D'autres études de cas sur les fumées de soudage et sur des NP différemment chargées ont confirmé la performance de notre test DCFH et ont trouvé que la génération de ROS est liée à certaines caractéristiques, notamment la fonctionnalité de surface des échantillons.

Background: Some countries have recently extended smoke-free policies to particular outdoor settings; however, there is controversy regarding whether this is scientifically and ethically justifiable. Objectives: The objective of the present study was to review research on secondhand smoke (SHS) exposure in outdoor settings. Data sources: We conducted different searches in PubMed for the period prior to September 2012. We checked the references of the identified papers, and conducted a similar search in Google Scholar. Study selection: Our search terms included combinations of"secondhand smoke,""environmental tobacco smoke,""passive smoking" OR"tobacco smoke pollution" AND"outdoors" AND"PM" (particulate matter),"PM2.5" (PM with diameter ≤ 2.5 µm),"respirable suspended particles,""particulate matter,""nicotine,""CO" (carbon monoxide),"cotinine,""marker,""biomarker" OR"airborne marker." In total, 18 articles and reports met the inclusion criteria. Results: Almost all studies used PM2.5 concentration as an SHS marker. Mean PM2.5 concentrations reported for outdoor smoking areas when smokers were present ranged from 8.32 to 124 µg/m3 at hospitality venues, and 4.60 to 17.80 µg/m3 at other locations. Mean PM2.5 concentrations in smoke-free indoor settings near outdoor smoking areas ranged from 4 to 120.51 µg/m3. SHS levels increased when smokers were present, and outdoor and indoor SHS levels were related. Most studies reported a positive association between SHS measures and smoker density, enclosure of outdoor locations, wind conditions, and proximity to smokers. Conclusions: The available evidence indicates high SHS levels at some outdoor smoking areas and at adjacent smoke-free indoor areas. Further research and standardization of methodology is needed to determine whether smoke-free legislation should be extended to outdoor settings.

Occupational exposures to fungi are very frequent and are known to cause chronic or acute symptoms. To better assess health risks related to fungal exposure, it is crucial to characterize precisely the airborne fungal community in terms of quantity and composition. The objective of this chapter is to synthesize existing knowledge of airborne fungal contamination in various occupational settings. We analyzed 134 papers published between 2000 and 2014 focusing on five different work sectors considered as highly contaminated (i.e., more than 1000 fungal particles/m3): animal confinement buildings, sawmills, waste handling, the food industry, and grain/plant handling. Results show that harvesting grain, washing cheese, and handling salami seem to be the occupational situations with the worst potential for exposure. Moreover, a lack of standardized sampling and analysis methods among countries and even within the same country is highlighted. Occupational exposure limit values do not exist. Recommendations and guidelines based on culture-dependent methods, which are now recognized to underestimate true concentrations, are proposed. Those recommendations are frequently exceeded and protective measures are not always easy to implement.

The layered double hydroxides, known as anionic clays and represented by the general formula [M2+1-x M3+x (OH) 2]x+ Am-x/m·nH 2O, are a group of materials which are of much interest currently. They present a variety of potential applications as adsorbents, catalysts and catalyst support, ion-exchangers, antacids and as a polymer stabilizer. It is possible to obtain a broad variety of layered double hydroxides (LDHs), depending on the identity and ratio of the cations M2+ and M3+, as well as the interlamelar anion. The aim of this review is to give out some information about this class of materials, concerning to the synthesis, characterization, properties and applications.
