Phosphatases have long been regarded as tumor suppressors, however there is emerging evidence for a tumor initiating role for some phosphatases in several forms of cancer. Low Molecular Weight Protein Tyrosine Phosphatase (LMWPTP; acid phosphatase 1 [ACP1]) is an 18 kDa enzyme that influences the phosphorylation of signaling pathway mediators involved in cancer and is thus postulated to be a tumor-promoting enzyme, but neither unequivocal clinical evidence nor convincing mechanistic actions for a role of LMWPTP have been identified. In the present study, we show that LMWPTP expression is not only significantly increased in colorectal cancer (CRC), but also follows a step-wise increase in different levels of dysplasia. Chemical inhibition of LMWPTP significantly reduces CRC growth. Furthermore, downregulation of LMWPTP in CRC leads to a reduced migration ability in both 2D- and 3D-migration assays, and sensitizes tumor cells to the chemotherapeutic agent 5-FU. In conclusion, this study shows that LMWPTP is not only overexpressed in colorectal cancer, but it is correlated with the malignant potential of this cancer, suggesting that this phosphatase may act as a predictive biomaker of CRC stage and represents a rational novel target in the treatment of this disease.

Plants that deploy a phosphorus (P)-mobilising strategy based on the release of carboxylates tend to have high leaf manganese concentrations ([Mn]). This occurs because the carboxylates mobilise not only soil inorganic and organic P, but also a range of micronutrients, including Mn. Concentrations of most other micronutrients increase to a small extent, but Mn accumulates to significant levels, even when plants grow in soil with low concentrations of exchangeable Mn availability. Here, we propose that leaf [Mn] can be used to select for genotypes that are more efficient at acquiring P when soil P availability is low. Likewise, leaf [Mn] can be used to screen for belowground functional traits related to nutrient-acquisition strategies among species in low-P habitats.

The aim of this research was to optimize osmotic dehydration of pineapple, according to two criteria: maximize water loss and minimize solid gain. The process was made as an application to Combined Methods Technology, in which three preservation factors were combined: water activity, pH and chemical preservatives, all being applied at low levels, in order to get a product resembling non-processed fruit. The experiment was divided into three treatments, being: non-coated pineapple pieces (A), pieces coated with alginate (B) and coated with low-methoxyl pectin (C). Process involved the following main steps: enzymatic inactivation of fruit pieces; in treatments B and C, incorporation of their respective coatings; and osmotic dehydration, in sucrose syrup containing potassium sorbate and citric acid. Optimum conditions, determined from Response Surface Methodology, were the following: dehydration of fruit pieces coated by alginate, at 42-47° C, in sucrose syrup at 66-69° Brix, for 220 to 270 minutes. Results indicated that both coatings significantly affected the mass transfers of the process, reducing solid incorporation and increasing water loss; therefore, increasing weight loss and performance ratio (water loss: solid incorporation) took place. Water activity was not significantly affected by the coatings. The product obtained under optimum conditions was submitted to sensorial evaluation, and presented a good general acceptance. Moulds and yeasts countings indicated good microbiological stability of the product for at least 60 days at 30ºC.

The recombinant human growth hormone (rhGH) can improve weight gain, physical growth, clinical and lung in cystic fibrosis (CF). However, the routine use, although promising, is not established in the literature. The objective of this study was to assess the benefits of rhGH in children and adolescents with CF. We conducted a systematic review in the database PubMed, Lilacs, SciELO and Cochrane, in the period 2000-2010, using the keywords: cystic fibrosis, growth hormone, children and adolescents. We found 77 articles and included 11 randomized controlled trials, with 290 children and adolescents with CF. The short-term use (1-24 months) of rhGH improved the height, weight, growth rate, bone mineral content and components of pulmonary function. Adverse effects, like diabetes, were not observed in the studies. The short-term use of rhGH improved growth and body composition in patients with CF.

OBJECTIVE: Removable partial dentures (RPD) require different hygiene care, and association of brushing and chemical cleansing is the most recommended to control biofilm formation. However, the effect of cleansers has not been evaluated in RPD metallic components. The aim of this study was to evaluate in vitro the effect of different denture cleansers on the weight and ion release of RPD. MATERIAL AND METHODS: Five specimens (12x3 mm metallic disc positioned in a 38x18x4 mm mould filled with resin), 7 cleanser agents [Periogard (PE), Cepacol (CE), Corega Tabs (CT), Medical Interporous (MI), Polident (PO), 0.05% sodium hypochlorite (NaOCl), and distilled water (DW) (control)] and 2 cobalt-chromium alloys [DeguDent (DD), and VeraPDI (VPDI)] were used for each experimental situation. One hundred and eighty immersions were performed and the weight was analyzed with a high precision analytic balance. Data were recorded before and after the immersions. The ion release was analyzed using mass spectrometry with inductively coupled plasma. Data were analyzed by two-way ANOVA and Tukey HSD post hoc test at 5% significance level. RESULTS: Statistical analysis showed that CT and MI had higher values of weight loss with higher change in VPDI alloy compared to DD. The solutions that caused more ion release were NaOCl and MI. CONCLUSIONS: It may be concluded that 0.05% NaOCl and Medical Interporous tablets are not suitable as auxiliary chemical solutions for RPD care.

Breast weight has great economic importance in poultry industry, and may be associated with other variables. This work aimed to estimate phenotypic correlations between performance (live body weight at 7 and 28 days, and at slaughter, and depth of the breast muscle measured by ultrasonography), carcass (eviscerated body weight and leg weight) and body composition (heart, liver and abdominal fat weight) traits in a broiler line, and quantify the direct and indirect influence of these traits on breast weight. Path analysis was used by expanding the matrix of partial correlation in coefficients which give the direct influence of one trait on another, regardless the effect of the other traits. The simultaneous maintenance of live body weight at slaughter and eviscerated body weight in the matrix of correlations might be harmful for statistical analysis involving systems of normal equations, like path analysis, due to the observed multicollinearity. The live body weight at slaughter and the depth of the breast muscle as measured by ultrasonography directly affected breast weight and were identified as the most responsible factors for the magnitude of the correlation coefficients obtained between the studied traits and breast weight. Individual pre-selection for these traits could favor an increased breast weight in the future reproducer candidates of this line if the broilers' environmental conditions and housing are maintained, since the live body weight at slaughter and the depth of breast muscle measured by ultrasonography were directly related to breast weight.

Um experimento foi conduzido com o objetivo de avaliar os efeitos da inclusão de ácido benzoico (Vevovitall®) na dieta sobre o desempenho e a morfologia intestinal do duodeno de leitões de 28 a 70 dias de idade. Foram utilizados 160 leitões Dalland × Penarlan desmamados aos 21 dias de idade, separados por sexo. O experimento foi dividido em fases pré-inicial 1 (28 a 35 dias de idade), pré-inicial 2 (36 a 46 dias de idade) e fase inicial (47 a 70 dias idade). Utilizou-se o delineamento experimental tipo blocos ao acaso com 5 tratamentos, cada um com 8 repetições de 4 animais, totalizando 40 unidades experimentais. Foram utilizados quatro níveis de ácido benzoico (0; 0,25; 0,50 e 0,75%) e um tratamento testemunha com ácido fumárico. Abateram-se, aos 70 dias de idade, dois animais por repetição para coleta de amostras do duodeno e estudo da morfologia intestinal e um animal por repetição para estudo de lesões gástricas. Foram avaliadas as características de desempenho (ganho de peso, consumo de ração e conversão alimentar) para cada fase e para o período total. Como características morfológicas, foram avaliadas a altura das vilosidades, a profundidade de criptas e a relação altura de vilosidades/profundidade de criptas. Avaliou-se ainda a incidência de diarreia. No período de 28 a 70 dias, o uso de ácido benzoico influencia o desempenho de suínos, que neste estudo foi melhor os níveis de 0,50 e 0,75%, e reduz a incidência de diarreia. Animais alimentados com dietas contendo ácido benzoico apresentam maior altura de vilosidade e profundidade de criptas.

Foram conduzidos dois experimentos com o objetivo de avaliar a utilização de duas fontes de metionina (em pó ou líquida) em rações formuladas com base em aminoácidos digestíveis ou totais para frangas de reposição leves e semipesadas, nas fases inicial (de 1 a 6 semanas de idade) e de crescimento (de 11 a 16 semanas de idade). Em cada experimento, foram utilizadas 384 aves distribuídas em delineamento inteiramente ao acaso, em esquema fatorial 2 × 2 × 2, composto de duas linhagens (Hy-Line W36 e Hy-Line Brown), duas fontes de metionina (HMTBA - 88% e DL-Metionina - 99%) e dois critérios de formulação da dieta (aminoácidos totais e digestíveis), totalizando oito tratamentos, cada um com seis repetições de oito aves. Foram avaliados os consumos de ração, proteína bruta e energia metabolizável, o peso e ganho de peso corporal e a conversão alimentar das aves. Não houve interação entre os fatores para nenhuma das variáveis estudadas. As aves da linhagem Brown apresentaram maior consumo de ração, peso corporal e ganho de peso. As fontes de metionina e os critérios de formulação de ração não influenciaram as características de desempenho. As aves das linhagens W36 e Brown da linhagem Hy-Line apresentam diferentes perfis de desenvolvimento corporal. A metionina em pó (DL-metionina - 99%) ou a líquida (HMTBA - 88%), assim como os critérios de formulação de ração (aminoácidos totais ou digestíveis) proporcionam desempenho semelhante em aves de reposição.

Avaliaram-se os efeitos do fornecimento da dieta pré-inicial (DPI) e do peso ao alojamento sobre o desempenho e as características de carcaça de frangos de corte. Utilizaram-se 960 frangos machos, linhagem comercial Cobb-500, alojados em 32 boxes, em um delineamento inteiramente casualizado em esquema fatorial 2 x 4, composto de dois pesos ao alojamento (42 e 46 g) e quatro períodos de fornecimento da dieta pré-inicial (0, 1 a 7, 1 a 10 e 1 a 14 dias de idade), durante 49 dias. Os programas alimentares compreenderam três ou quatro fases (pré-inicial e/ou inicial; engorda; final) e as dietas fornecidas eram à base de milho e farelo de soja. De modo geral, aves mais pesadas ao alojamento apresentaram, ao longo de todo o período experimental, maior consumo de ração (CR) e ganho de peso (GP). Não foram observadas diferenças significativas na conversão alimentar (CA) dos animais. Considerando todo o período experimental, as aves alimentadas com a dieta pré-inicial durante sete dias apresentaram maior consumo de ração e ganho de peso, porém, menor rendimento de carcaça. O peso ao alojamento determina o desempenho e o oferecimento da dieta pré-inicial durante sete dias beneficia o desempenho das aves.

Foi conduzido um experimento para avaliar a utilização de subprodutos do arroz em dietas formuladas com base nos conceitos de proteína bruta e ideal para frangos de corte de 1 a 42 dias de idade. Foram utilizados 720 pintos machos de 1 dia de idade da linhagem "Hybro", em delineamento inteiramente ao acaso, em esquema fatorial 3 × 2, composto de três dietas (sem subprodutos, farelo de arroz integral e quirera de arroz) e dois conceitos de formulação de rações (proteína bruta e ideal), totalizando seis tratamentos e quatro repetições de 30 aves. O ganho de peso, o consumo de ração e a conversão alimentar foram avaliados aos 21 e 42 dias e as características de carcaça aos 42 dias de idade. As aves alimentadas com dietas formuladas pelo conceito tradicional (baseado na proteína bruta) apresentaram melhor conversão alimentar e menor taxa de deposição de gordura abdominal.
