Introduction: Prior repeated-sprints (6) has become an interesting method to resolve the debate surrounding the principal factors that limits the oxygen uptake (V'O2) kinetics at the onset of exercise [i.e., muscle O2 delivery (5) or metabolic inertia (3)]. The aim of this study was to compare the effects of two repeated-sprints sets of 6x6s separated by different recovery duration between the sprints on V'O2 and muscular de-oxygenation [HHb] kinetics during a subsequent heavy-intensity exercise. Methods: 10 male subjects performed a 6-min constant-load cycling test (T50) at intensity corresponding to half of the difference between V'O2max and the ventilatory threshold. Then, they performed two repeated-sprints sets of 6x6s all-out separated by different recovery duration between the sprints (S1:30s and S2:3min) followed, after 7-min-recovery, by the T50 (S1T50 and S2T50, respectively). V'O2, [HHb] of the vastus lateralis (VL) and surface electromyography activity [i.e., root-mean-square (RMS) and the median frequency of the power density spectrum (MDF)] from VL and vastus medialis (VM) were recorded throughout T50. Models using a bi-exponential function for the overall T50 and a mono-exponential for the first 90s of T50 were used to define V'O2 and [HHb] kinetics respectively. Results: V'O2 mean value was higher in S1 (2.9±0.3l.min-1) than in S2 (1.2±0.3l.min-1); (p<0.001). The peripheral blood flow was increased after sprints as attested by a higher basal heart rate (HRbaseline) (S1T50: +22%; S2T50: +17%; p≤0.008). Time delay [HHb] was shorter for S1T50 and S2T50 than for T50 (-22% for both; p≤0.007) whereas the mean response time of V'O2 was accelerated only after S1 (S1T50: 32.3±2.5s; S2T50: 34.4±2.6s; T50: 35.7±5.4s; p=0.031). There were no significant differences in RMS between the three conditions (p>0.05). MDF of VM was higher during the first 3-min in S1T50 than in T50 (+6%; p≤0.05). Conclusion: The study show that V'O2 kinetics was speeded by prior repeated-sprints with a short (30s) but not a long (3min) inter-sprints-recovery even though the [HHb] kinetics was accelerated and the peripheral blood flow was enhanced after both sprints. S1, inducing a greater PCr depletion (1) and change in the pattern of the fibres recruitment (increase in MDF) compared with S2, may decrease metabolic inertia (2), stimulate the oxidative phosphorylation activation (4) and accelerate V'O2 kinetics at the beginning of the subsequent high-intensity exercise.

Purpose: To compare the efficacy and safety of a slowly resorbable glaucoma implant, Healaflow (HF) and Healon 5 (H5) as spacers injected into failing filtration blebs at the time of bleb needlings, augmented with MMC Methods: Eighty-four glaucoma patients with filtration bleb failure following glaucoma surgery (deep sclerectomy/ trabeculectomy) underwent bleb needling using either HF (n=44) or H5 (n=40) as an intrableb viscoelastic spacer. All needlings were augmented by MMC and performed in the operating room, by a single surgeon. The choice between HF and H5 was randomised. Postoperative data for IOP, number of glaucoma medications (GMs), antimetabolite injections (AMIs), bleb morphology and all complications were recorded at day 1 (D1), weeks 1 (W1), 4 (W4), month 6 (M6) and last visit. Success was defined as IOP≤21mmHg and 20% reduction in IOP from baseline Results: Age was comparable between groups (HF vs H5; 66 vs 72 years; p=0.13*). There were no differences in mean postoperative IOP or GMs at any time point between groups (mean IOP = HF vs H5; prior to needling=19.8mmHg vs 18.7mmHg, p=0.48*; D1=10.4mmHg vs 10.2mmHg, p=0.85*; W1=12.3mmHg vs 13.8mmHg, p=0.33*; W4=15.7mmHg vs 15.1mmHg, p=0.69*; M6=14.3mmHg vs 13.8mmHg, p=0.69*. Mean GMs= HF vs H5; prior to needling=1.7 vs 1.7, p=0.96*; M6=0.9 vs 1.1, p=0.66*). Success rates were comparable between HF and H5 groups (74% vs 71%). Requirements for AMIs were similar between groups; post filtration surgery and prior to needling (mean AMIs 1.54 vs 1.65 p=0.82*) and within the first six months following needling ( 0.77 vs 0.65 p=0.70*). Complication rates were infrequent in both groups (9%, HF; 3%, H5) * two-sample t-test Conclusions: The early to mid-term success rates of needlings using intrableb spacers were high. However there were no differences observed between the use of crosslinked and high molecular weight sodium hyaluronate

Abstract In humans, the skin is the largest organ of the body, covering up to 2m2 and weighing up to 4kg in an average adult. Its function is to preserve the body from external insults and also to retain water inside. This barrier function termed epidermal permeability barrier (EPB) is localized in the functional part of the skin: the epidermis. For this, evolution has built a complex structure of cells and lipids sealing the surface, the stratum corneum. The formation of this structure is finely tuned since it is not only formed once at birth, but renewed all life long. This active process gives a high plasticity and reactivity to skin, but also leads to various pathologies. ENaC is a sodium channel extensively studied in organs like kidney and lung due to its importance in regulating sodium homeostasis and fluid volume. It is composed of three subunits α, ß and r which are forming sodium selective channel through the cell membrane. Its presence in the skin has been demonstrated, but little is known about its physiological role. Previous work has shown that αENaC knockout mice displayed an abnormal epidermis, suggesting a role in differentiation processes that might be implicated in the EPB. The principal aim of this thesis has been to study the consequences for EPB function in mice deficient for αENaC by molecular and physiological means and to investigate the underlying molecular mechanisms. Here, the barrier function of αENaC knockout pups is impaired. Apparently not immediately after birth (permeability test) but 24h later, when evident water loss differences appeared compared to wildtypes. Neither the structural proteins of the epithelium nor the tights junctions showed any obvious alterations. In contrary, stratum corneum lipid disorders are most likely responsible for the barrier defect, accompanied by an impairment of skin surface acidification. To analyze in details this EPB defect, several hypotheses have been proposed: reduced sensibility to calcium which is the key activator far epidermal formation, or modification of ENaC-mediated ion fluxes/currents inside the epidermis. The cellular localization of ENaC and the action in the skin of CAPl, a positive regulator of ENaC, have been also studied in details. In summary, this study clearly demonstrates that ENaC is a key player in the EPB maintenance, because αENaC knockout pups are not able to adapt to the new environment (ex utero) as efficiently as the wildtypes, most likely due to impaired of sodium handling inside the epidermis. Cette fonction nommée barrière épithéliale est localisée dans la partie fonctionnelle de la peau : l'épiderme. A cette fin, l'évolution s'est dotée d'une structure complexe composée de cellules et de lipides recouvrant la surface, la couche cornée. Sa formation est finement régulée, car elle n'est pas seulement produite à la naissance mais constamment renouvelée tout au long de la vie, ce qui lui confère une grande plasticité mais ce qui est également la cause de nombreuses pathologies. ENaC est un canal sodique très étudié dans le rein et le poumon pour son importance dans la régulation de l'homéostasie sodique et la régulation du volume du milieu intérieur. Il est composé de 3 sous unités, α, ß et y qui forment un pore sélectif pour le sodium dans les membranes. Ce canal est présent dans la peau mais sa fonction n'y est pas connue. Des travaux précédents ont pu montrer que les souris dont le gène codant pour αENaC a été invalidé présentent un épiderme pathologique, suggérant un rôle dans la différentiation et pourrait même être impliqué dans la barrière épithéliale. Le but de cette thèse fut l'étude de la barrière dans ces souris knockouts avec des méthodes moléculaires et physiologiques et la caractérisation des mécanismes moléculaire impliqués. Dans ce travail, il a été montré que les souris mutantes présentaient un défaut de la barrière. Ce défaut n'est pas visible immédiatement à la naissance (test de perméabilité), mais 24h plus tard, lorsque les tests de perte d'eau transépithéliale montrent une différence évidente avec les animaux contrôles. Ni les protéines de structures ni les jonctions serrées de l'épiderme ne présentaient d'imperfections majeures. A l'inverse, les lipides de la couche cornée présentaient un problème de maturation (expliquant le phénotype de la barrière), certainement consécutif au défaut d'acidification à la surface de la peau que nous avons observé. D'autres mécanismes ont été explorées afin d'investiguer cette anomalie de la barrière, comme la réduction de sensibilité au calcium qui est le principal activateur de la formation de l'épiderme, ou la modification des flux d'ions entre les couches de l'épiderme. La localisation cellulaire d'ENaC, et l'action de son activateur CAPl ont également été étudiés en détails. En résumé, cette étude démontre clairement qu'ENaC est un acteur important dans la formation de la barrière épithéliale, car la peau des knockouts ne s'adapte pas aussi bien que celle des sauvages au nouvel environnement ex utero à cause de la fonction d'ENaC dans les mouvements de sodium au sein même de l'épiderme. Dans ce travail, il a été montré que les souris mutantes présentaient une peau plus perméable que celle des animaux contrôles, grâce à une machine mesurant la perte d'eau à travers la peau. Ce défaut n'est visible que 24h après la naissance, mais nous avons pu montrer que les animaux mutants perdaient quasiment 2 fois plus d'eau que les contrôles. Au niveau moléculaire, nous avons pu montrer que ce défaut provenait d'un problème de maturation des lipides qui composent la barrière de la peau. Cette maturation est incomplète vraisemblablement à cause d'un défaut de mouvement des ions dans les couches les plus superficielles de l'épiderme, et cela à cause de l'absence du canal ENaC. En résumé, cette étude démontre clairement qu'ENaC est un acteur important dans la formation de la barrière épithéliale, car la peau des mutants ne s'adapte pas aussi bien que celle des sauvages au nouvel environnement ex utero à cause de la fonction d'ENaC dans les mouvements de sodium au sein même de l'épiderme.

This investigation was conducted to study the performance characteristics of low cost roadway surfaces of soil-aggregate-sodium chloride mixtures. Many roads have been successfully stabilized with sodium chloride. However, little information is available on either the properties of the road materials or the effects of sodium chloride on the materials. The performance of some of the sodium chloride stabilized roads in Franklin County, Iowa, and the performance of some near-by nonchemically treated roads has been studied. The study of sodium chloride stabilized roads was restricted to the roads in which the binder soil used in construction came from the same source. The effects of sodium chloride on some of the engineering properties of the soil and soil-aggregate mixtures used were studied in the laboratory.

The amiloride-sensitive epithelial sodium channel is the limiting step in salt absorption. In mice, this channel is composed of three subunits (alpha, beta, and gamma), which are encoded by different genes (Scnn1a, Scnn1b, and Scnn1c, respectively). The functions of these genes were recently investigated in transgenic (knockout) experiments, and the absence of any subunit led to perinatal lethality. More defined phenotypes have been obtained by introducing specific mutations or using transgenic rescue experiments. In this report, these approaches are summarized and a current gene-targeting strategy to obtain conditional inactivation of the channel is illustrated. This latter approach will be indispensable for the investigation of channel function in a wide variety of organ systems.

Phosphate (Pi) acquisition of crops via arbuscular mycorrhizal (AM) symbiosis acquires increasing importance due to the limited rock Pi reserves and the demand for environmentally sustainable agriculture. However, the symbiotic Pi uptake machinery has not been characterized in any monocotyledonous plant species. Among these, rice is the primary staple food for more than half of the human population and thus central for future food security. However, the relevance of the AM symbiosis for rice Pi nutrition is presently unclear. Here, we show that 70% of the overall Pi acquired by rice is delivered via the symbiotic route. To better understand this pathway we combined genetic, molecular and physiological approaches to determine the specific functions of the two rice Pi transporters, PT11 and PT13, which are expressed only during AM symbiosis. The PT11 lineage of proteins is present in mono- and dicotyledons whereas PT13, while found across the Poaceae, is absent from dicotyledons. Surprisingly, mutations in either PT11 or PT13 affected fungal colonization and arbuscule formation demonstrating that both genes are essential for AM symbiosis between rice and Glomus intra.rad.ices. Importantly, for symbiotic Pi uptake, only PT11 is necessary and sufficient. We found that mycorrhizal rice, remarkably, received almost all Pi via the symbiotic route. Such dominating mycorrhizal Pi uptake was found in plants grown under controlled conditions as well as in field soils, suggesting that the AM symbiosis is relevant for the Pi nutrition of field grown rice. Development of smaller arbuscules in PT11 mutants suggested that symbiotic Pi signaling is required for fungal nourishment by the plant. However, co-culture of mutant with wild type nurse plants did not restore normal arbuscule size in mutant roots, indicating that other factors than malnutrition accounted for the altered arbuscule phenotype. Surprisingly, the loss of PT13 did not affect symbiotic Pi uptake although it impacted arbuscule morphology, suggesting that PT13 is involved in signaling during arbuscule development. However, induction of PT13 was not only monitored in arbusculated cells but also in inner cortex cells of non-inoculated roots of plants grown under high Pi fertilization conditions. According to preliminary observations, PT13 localized at the tonoplast in arbusculated and non-arbusculated cells, suggesting that it might be involved in transporting Pi into the vacuole, possibly for maintaining cellular Pi homeostasis. The further investigation showed that fungal colonization level was significantly affected in the crown roots of two ptlS mutant alleles, but not in large lateral roots, implying the possible role of PT13 for maintaining Pi homeostasis in the crown roots. - L'acquisition de phosphate (Pi) par les plantes cultivées s'effectue grâce à une symbiose mycorhizienne arbasculaire (AM). L'étude de cette symbiose devient fondamentale puisque d'une part, les réserves en phosphate minéral sont limitées, et, d'autre part, la demande pour une agriculture écologiquement soutenable se renforce. La machinerie d'absorption symbiotique du phosphate n'est cependant pas encore élucidée chez les plantes monocotylédones. Parmi celles-ci, le riz occupe une place primordiale. Aliment de base pour plus de la moitié de la population mondiale, il revêt de ce fait une dimension essentielle en termes de sécurité alimentaire. Pourtant, l'importance de la symbiose AM chez le riz dans le processus d'acquisition du phosphate n'est, encore de nos jours, que peu comprise. Dans cette étude, nous montrons que 70% du phosphate acquis par le riz est mis à disposition de la plante grâce à la symbiose AM. Afin de mieux comprendre ce mécanisme, nous avons employé des approches physiologiques et génétiques nous permettant de déterminer les fonctions spécifiques de deux transporteurs de Pi, PT11 et PT13, présents chez le riz et exprimés uniquement durant la symbiose AM. La famille de gènes à laquelle appartient PT11 est présente chez les monocotylédones ainsi que chez les dicotylédones tandis que PT13, bien que retrouvé au sein des Poaceae, est absent chez les dicotylédones. Etonnamment, des versions mutées de PT11 ou de PT13 affectent la colonisation par le champignon endo-mycorhizien ainsi que la formation d'arbuscules, démontrant l'importance de ces deux gènes dans la symbiose AM entre le riz et Glomus intraradices. Il est à noter que seul PT11 se révèle nécessaire et suffisant pour l'apport de Pi grâce à la symbiose. Nous avons observé que la presque totalité du phosphate dont dispose le riz lors d'une symbiose AM provient du champignon. De telles proportions ont été observées tant chez des plantes cultivées en conditions contrôlées que chez des plantes cultivées dans les champs. Cela suggère l'importance de la symbiose AM dans le processus d'acquisition du Pi chez le riz cultivé à l'extérieur. Le développement d'arbuscules plus petits chez le mutant PT11 tend à montrer qu'une voie signalétique impliquant le Pi symbiotique est nécessaire pour l'entretien du champignon par la plante. Toutefois, une co-culture du mutant avec des plantes sauvages ne permet pas de restaurer des arbuscules de taille normale dans les racines du mutant. Ce résultat indique le rôle de facteurs autres que la malnutrition aboutissant à la formation d'arbuscules altérés. Si la perte de PT13 n'affecte pas l'acquisition de phosphate symbiotique, la morphologie de l'arbuscule est, quant à elle, modifiée. Ceci suggère un rôle de PT13 durant le développement de l'arbuscule. Or, l'induction de PT13 est non seulement détectée dans des cellules contenant des arbuscules mais également dans des cellules du cortex, ceci chez des plantes cultivées sans champignon mais dans des conditions de fortes concentrations en engrais phosphaté. En accord avec des observations précédentes, PT13 est localisé au niveau du tonoplaste des cellules contenant ou non des arbuscules. Ceci suggère que PT13 pourrait être impliqué dans le transport du Pi vers la vacuole, éventuellement pour maintenir une certaine homéostasie du phosphate. Dans cette étude, nous démontrons également que le niveau de colonisation par le champignon est affecté de manière significative dans les racines principales des deux allèles du mutants ptl3, mais pas dans les grosses racines latérales. Cela impliquerait un rôle possible de PT13 dans le maintien de l'homéostasie du phosphate dans les racines principales. RESUME POUR UN LARGE PUBLIC Le phosphate (Pi), l'un des éléments minéraux essentiel au développement des plantes, se trouve généralement en faible quantité dans le sol, limitant ainsi la croissance des plantes. Le rendement de la production agricole dépend dès lors de l'addition d'engrais contenant du phosphate inorganique (Pi), obtenu à partir de ressources minières riches en phosphate. Or, ces ressources devraient être épuisées d'ici la fin du siècle. Les racines des plantes possèdent des transporteurs de phosphate efficaces leur permettant d'acquérir rapidement le Pi présent dans le sol. Comme le Pi s'avère immobile dans le sol, l'absorption rapide par les racines crée des zones pauvres en Pi autour des systèmes racinaires. Pour surmonter cet obstacle, les plantes ont développé une symbiose avec des champignons endomycorhiziens, la symbiose mycorhizienne arbusculaire (AM). Cette association leur donne accès à d'autres ressources en phosphate puisque le mycélium de ces champignons se développe sur une surface 100 fois supérieure à celle des racines. Cela augmente considérablement la surface de nutrition, dépassant ainsi la zone appauvrie en Pi. Le phosphate, transporté grâce au champignon jusqu'à l'intérieur des racines, est fourni à la plante par le biais de structures établies à l'intérieur des cellules végétales, appelées arbuscules. De leur côté, les plantes possèdent des transporteurs spécifiques afin de recevoir le Pi fourni par les champignons. A l'heure actuelle, la machinerie nécessaire à cette absorption a été uniquement décrite chez des plantes dicotylédones. Or, comprendre l'apport de phosphate par les champignons mycorhiziens s'avère particulièrement pertinent dans le cas des espèces monocotylédones cultivées telles que les céréales. Ces dernières constituent en effet la majeure partie de l'alimentation humaine. Parmi les céréales, le riz demeure l'aliment de base de la population mondiale, d'où son importance en terme de sécurité alimentaire. Durant mon travail de thèse, j'ai identifié et caractérisé le transporteur du riz impliqué dans l'apport de phosphate par ce type de symbiose AM. J'ai également démontré que le riz, lorsqu'il vit en symbiose, bénéficie de la presque totalité du Pi transporté par le champignon. Environ 40% de la production globale de riz est cultivée dans des conditions permettant la symbiose avec des mycorhizes arbusculaires. Les variétés de riz adaptées à ces conditions aérobiques deviennent des alternatives favorables aux cultivars actuels nécessitant une forte irrigation. Elles se révèlent en effet plus tolérantes aux pénuries d'eau et permettent l'utilisation de pratiques agricoles moins intensives. Les données présentées dans cette étude enrichissent nos connaissances concernant l'absorption du phosphate chez le riz grâce à la symbiose AM. Ces connaissances peuvent s'avérer décisives pour le développement de cultivars du riz plus adaptés à une agriculture écologiquement soutenable.

The coupling between synaptic activity and glucose utilization (neurometabolic coupling) is a central physiologic principle of brain function that has provided the basis for 2-deoxyglucose-based functional imaging with positron emission tomography. Approximately 10 y ago we provided experimental evidence that indicated a central role of glutamate signaling on astrocytes in neurometabolic coupling. The basic mechanism in neurometabolic coupling is the glutamate-stimulated aerobic glycolysis in astrocytes, such that the sodium-coupled reuptake of glutamate by astrocytes and the ensuing activation of the Na(+)-K(+) ATPase triggers glucose uptake and its glycolytic processing, which results in the release of lactate from astrocytes. Lactate can then contribute to the activity-dependent fueling of the neuronal energy demands associated with synaptic transmission. Analyses of this coupling have been extended in vivo and have defined the methods of coupling for inhibitory neurotransmission as well as its spatial extent in relation to the propagation of metabolic signals within the astrocytic syncytium. On the basis of a large body of experimental evidence, we proposed an operational model, "the astrocyte-neuron lactate shuttle." A series of results obtained by independent laboratories have provided further support for this model. This body of evidence provides a molecular and cellular basis for interpreting data that are obtained with functional brain imaging studies.

Background and objectives: Interleukin-18 (IL-18) is a pleiotropic cytokine involved in rheumatoid arthritis (RA) pathogenesis. This studywas carried out to evaluate the efficicacy of interleukin-18 binding protein (IL-18BP) gene therapy in the rat adjuvant-induced arthritis (AIA) model and to decipher the mechanisms by which IL-18BP delivery lessens bone destruction. Materials and methods: Arthritis was induced in female Lewis rat by Mycobacterium butyricum and the mRNA expression of IL-18 and IL-18BP was determined in the joints. In a preventative study, rats were divided into an adenovirus producing IL-18BP-Fc (AdmIL-18BP-Fc) group (n=8) and an adenovirus producing green fluorescent protein (AdGFP) group (n=7). On day 8 after AIA induction, adenoviruses were injected. Clinical parameters were assessed. At day 18, during maximal arthritis, the rats were euthanized, ankles were collected, and X-rays were performed. mRNA and protein were extracted from joints for analyses by qRT-PCR, multiplex, Western blot, and zymography. Results: We observed a decrease in the [IL-18BP/IL-18] ratio from day 7 to day 45. Administration of AdmIL-18BPd-Fc decreased clinical parameters and prevented bone and joint destruction compared to AdGFP administration. IL-18BP delivery reduced the metalloproteinase 9 (MMP-9) levels by 33% (at protein level (Fig. 1B) and functional level (Fig. 1C) and the tartrate-resistant acid phosphatase (TRAP) level by 44% (Fig. 1D) in the joint homogenates from AdmIL-18BPd-Fc compared to AdGFP treated rats.However, no variationwas observed forMMP-2 at the protein level (Fig.1A) and functional level (Fig. 1C). Conclusions: In rat AIA, a decrease in the [IL-18BP/ IL-18] ratio was observed. IL-18BP delivery prevented joint and bone destruction by downregulating MMP-9 and TRAP, suggesting a potential benefit of a similar therapy in RA.

Introduction: Prior repeated-sprints (6) has become an interesting method to resolve the debate surrounding the principal factors that limits the oxygen uptake (V'O2) kinetics at the onset of exercise [i.e., muscle O2 delivery (5) or metabolic inertia (3)]. The aim of this study was to compare the effects of two repeated-sprints sets of 6x6s separated by different recovery duration between the sprints on V'O2 and muscular de-oxygenation [HHb] kinetics during a subsequent heavy-intensity exercise. Methods: 10 male subjects performed a 6-min constant-load cycling test (T50) at intensity corresponding to half of the difference between V'O2max and the ventilatory threshold. Then, they performed two repeated-sprints sets of 6x6s all-out separated by different recovery duration between the sprints (S1:30s and S2:3min) followed, after 7-min-recovery, by the T50 (S1T50 and S2T50, respectively). V'O2, [HHb] of the vastus lateralis (VL) and surface electromyography activity [i.e., root-mean-square (RMS) and the median frequency of the power density spectrum (MDF)] from VL and vastus medialis (VM) were recorded throughout T50. Models using a bi-exponential function for the overall T50 and a mono-exponential for the first 90s of T50 were used to define V'O2 and [HHb] kinetics respectively. Results: V'O2 mean value was higher in S1 (2.9±0.3l.min-1) than in S2 (1.2±0.3l.min-1); (p<0.001). The peripheral blood flow was increased after sprints as attested by a higher basal heart rate (HRbaseline) (S1T50: +22%; S2T50: +17%; p≤0.008). Time delay [HHb] was shorter for S1T50 and S2T50 than for T50 (-22% for both; p≤0.007) whereas the mean response time of V'O2 was accelerated only after S1 (S1T50: 32.3±2.5s; S2T50: 34.4±2.6s; T50: 35.7±5.4s; p=0.031). There were no significant differences in RMS between the three conditions (p>0.05). MDF of VM was higher during the first 3-min in S1T50 than in T50 (+6%; p≤0.05). Conclusion: The study show that V'O2 kinetics was speeded by prior repeated-sprints with a short (30s) but not a long (3min) inter-sprints-recovery even though the [HHb] kinetics was accelerated and the peripheral blood flow was enhanced after both sprints. S1, inducing a greater PCr depletion (1) and change in the pattern of the fibres recruitment (increase in MDF) compared with S2, may decrease metabolic inertia (2), stimulate the oxidative phosphorylation activation (4) and accelerate V'O2 kinetics at the beginning of the subsequent high-intensity exercise.

Sixteen patients with refractory hypertension were submitted to vigorous sodium depletion while cardiovascular homeostasis was monitored with measurements of hormonal and hemodynamic parameters and repeat saralasin tests. This regimen resulted in a negative sodium balance by an average of 300 mEq. The loss of sodium closely correlated to the decrease of body weight (r = 0.70, p less than 0.005). Blood pressure (BP) decreased from 176/166 +/- 8/3 to 155/109 +/-6/3 mm Hg. There was a significant correlation between percent increments in plasma renin activity (PRA) and the rise in plasma norepinephrine (r = 0.68, p less than 0.05) and a close negative correlation between percent increase in PRA and the ratio of fall in mean blood pressure (MAP) per unit of weight loss (r = -0.73, p less than 0.005). Thus, patients with the least percent increase in PRA demonstrated the greatest fall in BP per unit of weight loss, indicating that relative rather than absolute elevation of renin may be the factor limiting antihypertensive efficacy of sodium depletion. Sodium depletion induced increase in peripheral resistance and decrease in cardiac output, both mostly attributable to relative hyperreninemia. Indeed, the adverse hemodynamic changes were reversed by angiotensin inhibition, during which BP normalized. It is concluded that vigorous sodium depletion complemented by angiotensin blockade or suppression with sympatholytic agents improves management of otherwise refractory hypertension.

The remaining phosphorus (Prem) has been used for estimating the phosphorus buffer capacity (PBC) of soils of some Brazilian regions. Furthermore, the remaining phosphorus can also be used for estimating P, S and Zn soil critical levels determined with PBC-sensible extractants and for defining P and S levels to be used not only in P and S adsorption studies but also for the establishment of P and S response curves. The objective of this work was to evaluate the effects of soil clay content and clay mineralogy on Prem and its relationship with pH values measured in saturated NaF solution (pH NaF). Ammonium-oxalate-extractable aluminum exerts the major impacts on both Prem and pH NaF, which, in turn, are less dependent on soil clay content. Although Prem and pH NaF have consistent correlation, the former has a soil-PBC discriminatory capacity much greater than pH NaF.
