This work quantifies two important epidemiological features of the bean (Phaseolus vulgaris)/Phaeoisariopsis griseola pathosystem. The first is the effect of the number of nights of leaf wetness on infection efficiency. Infection efficiency was below 10% when inoculated leaflets were exposed to less than two nights of leaf wetness. Optimum infection efficiencies were obtained after three to four nights of leaf wetness, at about 50%. Further nights of leaf wetness did not increase the infection efficiency. The second feature quantified is the relative rate of leaflet defoliation for varying levels of angular leaf spot severity. It increased with disease severity according to a logarithm-like curve, and a relative rate of 0.23 day-1 was estimated for a severity of 18%. The implications of these results on the disease epidemiology are discussed.

As primary objective, this thesis examines Finnair Technical Procurement’s service quality with its underlying process. As an internal unit, Technical Procurement serves as a link between external suppliers and internal customers. It is argued that external service quality requires a certain quality level within an organization. At the same time, aircraft maintenance business is subject to economic restraints. Therefore, a methodology was developed with a modified House of Quality that assists management in analyzing and evaluating Technical Procurement’s service level and connected process steps. It could be shown that qualitative and quantitative objectives do not exclude each other per se.

A doença de Chagas é causada pelo Trypanosoma cruzi (T. cruzi) e a detecção de anticorpos anti-T. cruzi no soro é um método para diagnosticar a doença. Neste trabalho, ELISA indireto foi otimizado para a detecção de anticorpos no soro de pacientes com doença de Chagas empregando a proteína Tc85-11 (Ag) da superfície do parasita tripomastigota. Na otimização das condições experimentais foi aplicado planejamento fatorial completo para os parâmetros tempo de incubação, diluições do Ag (0,08 mg mL-1), anticorpo primário (Ac) e anti-IgG conjugada com a peroxidase (Ac*). Os melhores resultados foram obtidos nas seguintes condições: 0,14 mg Ag/poço, 60 min para o tempo de incubação, diluições de 1:35 e 1:1000 para Ac e Ac*, respectivamente. O valor do limiar de reatividade ("cut off") foi A450nm = 0,371. O ELISA indireto foi aplicado em amostras de soros de pacientes infectados com doença de Chagas e pacientes com diferentes doenças sistêmicas. A proteína Tc85-11, a qual está envolvida com a adesão do parasita na célula hospedeira, é também apropriada para o diagnóstico sorológico da doença de Chagas.

Flow injection (FI) methodology, using diffuse reflectance in the visible region of the spectrum, for the analysis of total sulfur in the form of sulfate, precipitated in the form of barium sulfate, is presented. The method was applied to biodiesel, to plant leaves and to natural waters analysis. The analytical signal (S) correlates linearly with sulfate concentration (C) between 20 and 120 ppm, through the equation S=-1.138+0.0934 C (r = 0.9993). The experimentally observed limit of detection is about 10 ppm. The mean R.S.D. is about 3.0 %. Real samples containing sulfate were analyzed and the results obtained by the FI and by the reference batch turbidimetric method using the statistical Student's t-test and F-test were compared.

A qualitative spot-test and tandem quantitative analysis of dipyrone in the bulk drug and in pharmaceutical preparations is proposed. The formation of a reddish-violet color indicates a positive result. In sequence a quantitative procedure can be performed in the same flask. The quantitative results obtained were statistically compared with those obtained with the method indicated by the Brazilian Pharmacopoeia, using the Student's t and the F tests. Considering the concentration in a 100 µL aliquot, the qualitative visual limit of detection is about 5×10-6 g; instrumental LOD ≅ 1.4×10-4 mol L-1 ; LOQ ≅ 4.5×10-4 mol L-1.

Two Lettuce mosaic virus isolates capable of overcoming the resistance afforded by the resistance gene mo1² in lettuce, LMV-AF199 from Brazil, and LMV-E, an European isolate, were evaluated for the rapidity and severity of symptoms induced on the lettuce variety Salinas 88 (mo1²). The mosaic symptoms on Salinas 88 plants inoculated with LMV-AF199 appeared 7 days post-inoculation (dpi) and 15 dpi for LMV-E. The symptoms induced by LMV-AF199 in this cultivar were also more severe than those induced by LMV-E. In order to identify the region of the viral genome responsible for this phenotype, recombinant viruses were constructed between these isolates and the phenotype of each recombinant was analysed. The region encoding proteins P1 and HcPro from LMV-AF199 was associated with the increased virulence in Salinas 88.

This research focuses on the link between quantitative sustainability disclosure and information asymmetry. It builds upon previous research which links information asymmetry with voluntary disclosure. Stakeholders from the financial services sector claim that sustainability disclosure needs to be more numerical and comparable between companies. This research covers 111 firms from Denmark, Finland, the Netherlands and Sweden from non-service industries and studies how quantitative their sustainability disclosure is, and whether or not there is a negative relation with information asymmetry. The results support the hypotheses, where two out of three information asymmetry proxies have a significant negative relation with quantitative disclosure. Size is supported as a moderating factor. Quantitativity also proves to have a significant link with third party sustainability ratings. The direct link between quantitativity and cost of capital is not however supported.

Since its introduction, fuzzy set theory has become a useful tool in the mathematical modelling of problems in Operations Research and many other fields. The number of applications is growing continuously. In this thesis we investigate a special type of fuzzy set, namely fuzzy numbers. Fuzzy numbers (which will be considered in the thesis as possibility distributions) have been widely used in quantitative analysis in recent decades. In this work two measures of interactivity are defined for fuzzy numbers, the possibilistic correlation and correlation ratio. We focus on both the theoretical and practical applications of these new indices. The approach is based on the level-sets of the fuzzy numbers and on the concept of the joint distribution of marginal possibility distributions. The measures possess similar properties to the corresponding probabilistic correlation and correlation ratio. The connections to real life decision making problems are emphasized focusing on the financial applications. We extend the definitions of possibilistic mean value, variance, covariance and correlation to quasi fuzzy numbers and prove necessary and sufficient conditions for the finiteness of possibilistic mean value and variance. The connection between the concepts of probabilistic and possibilistic correlation is investigated using an exponential distribution. The use of fuzzy numbers in practical applications is demonstrated by the Fuzzy Pay-Off method. This model for real option valuation is based on findings from earlier real option valuation models. We illustrate the use of number of different types of fuzzy numbers and mean value concepts with the method and provide a real life application.

The present research was carried out at the DER/FCAV, São Paulo State University - Brazil, with the purpose of analyzing the quantity and quality of biogas produced by six types of substrates. The substrates used in the anaerobic digestion were characterized as: 1- Litter of broiler (LB); 2- Grinded broiler litter (GBL); 3- Broiler litter with wood shavings (BLWS); 4- Grinded broiler litter with wood shavings (GBLWS); 5- Broiler litter with peanut hulls (BLPH); 6- Grinded broiler litter with peanut hull (GBLPH). It was concluded from the collected data that: the (GBL) substrate showed a superior biogas accumulated production in relation to the other substrates, while the BLWS presented an inferior accumulated production; the grinded substrates showed higher quantities of accumulated biogas in relation to the non-grinded substrates, except for GBLPH, with 20.9 m³ inferior than BLPH; the period of maximum biogas production started from 45 to 60 days, declining after 120 days; at 57 days after filling up the digesters the biogas produced had levels of CH4 superior than 53%, and from 99 days all of them produced biogas with levels of CH4 superior than 70%; LB and GBL presented higher concentrations of CH4 in the biogas.

OBJETIVO: Investigar a expressão imunoistoquímica dos marcadores p53, Ki-67, CK20 em neoplasias uroteliais papilíferas superficiais da bexiga e correlacionar com o grau histológico, progressão tumoral e recidiva. MÉTODOS: Foram selecionadas amostras de 43 pacientes portadores de carcinoma de células transicionais superficiais da bexiga. Elas foram distribuídas em dois grupos, um denominado recorrente, de 18 indivíduos e outro não recorrente, com 25 casos. Foram confeccionados blocos multiamostrais. A técnica imunoistoquímica empregada foi de imunoperoxidase e os anticorpos foram: p53 (clone DO7), o Ki-67 (clone SP6) e CK20. RESULTADOS: A expressão do p53 foi observada em 11 casos, todos tumores de alto grau (p=0,0001). A progressão histológica ocorreu em seis indivíduos (p=0,0076). Dos 18 casos recorrentes, seis apresentaram imunorreação para o p53 e 12 foram negativos para este anticorpo (p=0,1715). O Ki-67 foi positivo em 17 dos 18 casos do grupo recorrente (p=0,0001) e dos 20 tumores de alto grau, 18 apresentaram reação para este anticorpo (p=0,0001). Dos 18 indivíduos que tiveram recorrência, 13 apresentaram expressão anômala para CK20 (p=0,0166). Nos carcinomas de alto grau, dos 20 casos, 16 apresentaram expressão anômala para este anticorpo, enquanto que 18 dos 23 indivíduos com tumores de baixo grau mostraram expressão habitual para a CK20 (p=0,0002). CONCLUSÃO: O p53 mostrou boa correlação com a progressão histológica e grau histológico. O Ki-67 apresentou forte associação com a recidiva e grau histológico, e a CK20 também associou-se com estas variáveis.

The purpose of this thesis is twofold. The first and major part is devoted to sensitivity analysis of various discrete optimization problems while the second part addresses methods applied for calculating measures of solution stability and solving multicriteria discrete optimization problems. Despite numerous approaches to stability analysis of discrete optimization problems two major directions can be single out: quantitative and qualitative. Qualitative sensitivity analysis is conducted for multicriteria discrete optimization problems with minisum, minimax and minimin partial criteria. The main results obtained here are necessary and sufficient conditions for different stability types of optimal solutions (or a set of optimal solutions) of the considered problems. Within the framework of quantitative direction various measures of solution stability are investigated. A formula for a quantitative characteristic called stability radius is obtained for the generalized equilibrium situation invariant to changes of game parameters in the case of the H¨older metric. Quality of the problem solution can also be described in terms of robustness analysis. In this work the concepts of accuracy and robustness tolerances are presented for a strategic game with a finite number of players where initial coefficients (costs) of linear payoff functions are subject to perturbations. Investigation of stability radius also aims to devise methods for its calculation. A new metaheuristic approach is derived for calculation of stability radius of an optimal solution to the shortest path problem. The main advantage of the developed method is that it can be potentially applicable for calculating stability radii of NP-hard problems. The last chapter of the thesis focuses on deriving innovative methods based on interactive optimization approach for solving multicriteria combinatorial optimization problems. The key idea of the proposed approach is to utilize a parameterized achievement scalarizing function for solution calculation and to direct interactive procedure by changing weighting coefficients of this function. In order to illustrate the introduced ideas a decision making process is simulated for three objective median location problem. The concepts, models, and ideas collected and analyzed in this thesis create a good and relevant grounds for developing more complicated and integrated models of postoptimal analysis and solving the most computationally challenging problems related to it.

An indirect enzyme linked immunoassay (ELISA-I) was developed and standardized for the serological diagnosis of classical swine fever (CSF). For the comparison, nine hundred and thirty-seven swine serum samples were tested by serum neutralization followed by immunoperoxidase staining (NPLA), considered as the standard. Of these, 223 were positive and 714 negative for neutralizing antibodies to classical swine fever virus (CSFV). In relation to the NPLA, the ELISA-I presented a 98.2% sensitivity; 92.86% specificity, 81.11% positive predictive value, 99.4% negative predictive value and a 94.1% precision. Statistical analysis showed a very strong correlation (r=0,94) between both tests. When compared to a commercially available ELISA kit, the performance of both, in relation to the NPLA, was similar. It was concluded that the ELISA-I is suitable for large scale screening of antibodies to classical swine fever virus, although it does not distinguish antibodies to classical swine fever virus from those induced by other pestiviruses.

Um ensaio imunoenzimático do tipo ELISA de bloqueio com anticorpo monoclonal (ELISA-M) foi desenvolvido e padronizado para a detecção de anticorpos contra o vírus da Rinotraqueíte Infecciosa Bovina (Herpesvírus Bovino tipo 1; BHV-1). Foram utilizadas nesta avaliação 266 amostras de soros bovinos, sendo 148 negativos e 118 positivos em testes de soroneutralização (SN). Em comparação com este último, o ELISA-M demonstrou uma sensibilidade de 92,37%, especificidade de 92,56%, valor preditivo positivo de 90,83%, valor preditivo negativo de 93,83% e precisão de 92,48%. O índice de concordância (k) entre os testes foi de 0,85. O ELISA-M apresentou como vantagens a rapidez e a praticidade de execução. Com base nestes resultados, o ELISA-M foi considerado uma alternativa apropriada para o diagnóstico sorológico de infecções pelo BHV-1. Entretanto, o teste não foi capaz de diferenciar anticorpos induzidos por BHV-1 ou BHV-5.
