Resolution and Absolute Configuration Assignment of a Natural Racemic Chromane from Peperomia obtusifolia (Piperaceae)

The resolution of the natural racemic chromane 3,4-dihydro-5-hydroxy-2,7-dimethyl-8-(3 ''-methyl-2 ''-butenyl)-2-(4'-methyl-1',3'-pentadienyl)-2H-1-benzopyran-6-carboxylic acid (1) isolated from the leaves of Peperomia obtusifolia has been accomplished using stereoselective HPLC. The absolute coil figuration of the resolved enantiomers was determined by the analysis of optical rotations and CD spectra. The finding of a racemic mixture instead of an enantiomerically pure metabolite raises questions about the final steps in the biosynthesis of this class of natural products, suggesting that the intramolecular chromane ring formation step may not be enzymatically controlled at all in P. obtusifolia. Effect of tranquilization and anesthesia on high-resolution electrocardiographic indices of dogs in the chronic chagasic myocardiopathy

Avaliou-se o efeito da tranquilização e da anestesia sobre os índices da eletrocardiografia de alta resolução (ECGAR) em cães portadores de doença-de-chagas na fase crônica indeterminada. Foram utilizados oito cães, adultos, sem raça definida, fêmeas, submetidas a seis protocolos (grupos). No grupo 1, os animais estavam sem efeito de tranquilização ou anestesia; no grupo 2, foram tranquilizados com acepromazina; no 3, foram tranquilizados com a associação acepromazina e buprenorfina; no 4, estavam sob anestesia geral inalatória com isofluorano; no 5, sob anestesia geral inalatória com sevofluorano; e no 6, sob anestesia com propofol. Os animais foram submetidos a todos os protocolos, com um período de 15 dias entre cada avaliação. Não se verificou alteração significativa na duração do complexo QRS e do LAS40 entre os grupos, e o RMS40 permaneceu sem alteração significativa. O nível de ruído foi significativamente menor nos grupos 4, 5 e 6 em relação ao grupo 1. A anestesia facilitou o registro da ECGAR sem alterar os índices eletrocardiográficos .

High resolution spectroscopy of (CH3OH)-C-13 around the 10R(20) CO2 laser emission: New FIR laser lines, frequency measurements and assignments

We investigated the IR absorption spectrum of (CH3OH)-C-13 around the frequency of the 10R(20) CO2 laser line. We found two absorption lines which can be excited by 10R(20) and studied the FIR laser emissions excited by this pump line using a waveguide CO2 laser of 300 MHz tunability: We report two new FIR laser lines of large offset, not previously observed due to their weakness and closeness to other stronger lines. We measured the frequencies of five FIR laser lines for the first time by an accurate heterodyne technique and present the complete assignments of the IR-FIR laser systems relative to this pump line. Furthermore we present new frequency values for two FIR laser lines whose frequencies had been previously wrongly measured. Copyright (C) 1997 Elsevier B.V. Ltd.

Structure of human uropepsin at 2.45 angstrom resolution

The molecular structure of human uropepsin, an aspartic proteinase from the urine produced in the form of pepsinogen A in the gastric mucosa, has been determined by molecular replacement using human pepsin as the search model. Crystals belong to space group P2(1)2(1)2(1), with unit-cell parameters a = 50.99, b = 75.56, c = 89.90 Angstrom. Crystallographic refinement led to an R factor of 0.161 at 2.45 Angstrom resolution. The positions of 2437 non-H protein atoms in 326 residues have been determined and the model contains 143 water molecules. The structure is bilobal, consisting of two predominantly beta -sheet lobes which, as observed in other aspartic proteinases, are related by a pseudo-twofold axis. A model of the uropepsin-pepstatin complex has been constructed based on the high-resolution crystal structure of pepsin complexed with pepstatin.

OFF-LINE SUPERCRITICAL-FLUID EXTRACTION - HIGH-RESOLUTION GAS-CHROMATOGRAPHY APPLIED TO THE STUDY OF MORACEAE SPECIES

Supercritical fluid extraction with CO2, Performed in a home-made system, of rhizomes of Dorstenia bryoniifolia Mart. ex Miq. (Moraceae) and of bark roots of Brosimum gaudichaudii Trecul (Moraceae) afforded crude extracts that were analysed by high resolution gas chromatography (HRGC). The D. bryoniifolia extract contained, besides the previously reported pimpinelin and isobergapten, the furocoumarins psoralen, bergapten, isopimpinelin and the triterpenes alpha- and beta-amyrin and the acetate of the latter. The B. gaudichaudii extract contained a number of terpenoids as well as the previously reported psoralen and bergapten. Supercritical fluid extraction gave extracts qualitatively similar to those obtained by Soxhlet extraction with hexane and, together with off-line HRGC, was shown to be a fast and accurate technique to be used in rapid phytochemical examination.
