Histone RNA 3' end formation occurs through a specific cleavage reaction that requires, among other things, base-pairing interactions between a conserved spacer element in the pre-mRNA and the minor U7 snRNA present as U7 snRNP. An oligonucleotide complementary to the first 16 nucleotides of U7 RNA can be used to characterize U7 snRNPs from nuclear extracts by native gel electrophoresis. Using similar native gel techniques, we present direct biochemical evidence for a stable association between histone pre-mRNA and U7 snRNPs. Other complexes formed in the nuclear extract are dependent on the 5' cap structure and on the conserved hairpin element of histone pre-mRNA, respectively. However, in contrast to the U7-specific complex, their formation is not required for processing. Comparison of several authentic and mutant histone pre-mRNAs with different spacer sequences demonstrates that the formation and stability of the U7-specific complex closely follows the predicted stability of the potential RNA-RNA hybrid. However, this does not exclude a stabilization of the complex by U7 snRNP structural proteins.

Abstract Information-centric networking (ICN) offers new perspectives on mobile ad-hoc communication because routing is based on names but not on endpoint identifiers. Since every content object has a unique name and is signed, authentic content can be stored and cached by any node. If connectivity to a content source breaks, it is not necessarily required to build a new path to the same source but content can also be retrieved from a closer node that provides the same content copy. For example, in case of collisions, retransmissions do not need to be performed over the entire path but due to caching only over the link where the collision occurred. Furthermore, multiple requests can be aggregated to improve scalability of wireless multi-hop communication. In this work, we base our investigations on Content-Centric Networking (CCN), which is a popular {ICN} architecture. While related works in wireless {CCN} communication are based on broadcast communication exclusively, we show that this is not needed for efficient mobile ad-hoc communication. With Dynamic Unicast requesters can build unicast paths to content sources after they have been identified via broadcast. We have implemented Dynamic Unicast in CCNx, which provides a reference implementation of the {CCN} concepts, and performed extensive evaluations in diverse mobile scenarios using NS3-DCE, the direct code execution framework for the {NS3} network simulator. Our evaluations show that Dynamic Unicast can result in more efficient communication than broadcast communication, but still supports all {CCN} advantages such as caching, scalability and implicit content discovery.

Background. Providing an worksite fitness center for employees is an effective setting to increase an adult’s levels of physical activity, demonstrably reducing health care costs and elevating employee productivity. Increasing compliance and adherence among employees to consistently utilize an onsite facility remains a challenge.^ Purpose. To illustrate if two electronic behavioral journalism newsletters can increase employee participation in a free worksite wellness center.^ Design. A randomized control trial was implemented. ^ Setting. A large petroleum-producing corporation located in Houston, Texas Subjects.70 full or part time employees that had checked into visited the wellness center during September, October, and November 2009. ^ Intervention. Two behavioral journalism newsletters were created based on authentic community member role model interviews. Newsletters were distributed to intervention group participants via company email. Intervention and control group participant’s visits to the wellness center were monitored via a key card check in system at the facility for two weeks following the newsletter intervention.^ Analysis. Count variable statistics were used to identify rate differences between the intervention and control group wellness center visits. A binomial test for equality of proportions was used to identify differences between participants that had at least one visit to the wellness center compared with those with none.^ Results. Both intervention and control groups had a baseline of zero wellness center visits. The control group had a mean visit rate 0.441 and the intervention group had 0.857. A 0.416, almost one half more visits were found among the intervention group.^ Conclusion. This study indicates that behavioral journalism newsletters featuring a role model from within the worksite population can be a cost effective communication method to improve participation in an onsite wellness center. ^

This is an ethnographic study about the worldview of community-based initiatives in Houston, Texas, and the people who work in them. People who participated in this study recognize that their direct constructive action is at the heart of authentic social change in their minority communities. Through qualitative data analysis, a constellation of relationships and process patterns were found to constitute themselves into the system of the community-based initiative. The predominant patterns identified from the findings in this study are: the pervasiveness of place, the importance of people, unique initiatory patterns, the concrete local sustainability, the ever-present action orientation, the resourceful use of networks and inter-relationships, the significance of church influence, the core sense of spirituality and the essence of hope. These patterns emerged out of the local knowledge, which is acutely sensitive to the elements of history and lived experience, embedded in the distinctive moral and visionary patterns of meaning and expression. Findings from the research reveal that these community-based initiatives are not programs--they are people--people who keep hope alive in their communities and who, by their daily practice, liberate others. ^

Introduction: Both a systems approach to change and a focus on multi-sector interventions ensures obesity prevention programming within the community is equitable, sustainable, and cost-effective. An authentic community engagement approach is required to implement interventions guided by best-evidence research and practice. Although there are examples illustrating the benefits of community engagement, there is no standardized method to implement it. The San Antonio Sports Foundation (SA Sports), a non-profit community-based organization, implements a variety of free events and programs promoting active life styles. One such program is the Fit Family Challenge which is a summer-long program implemented at the school level targeted at families. ^ Aims: This thesis was a culmination of the experience from the student collaborating with SA Sports as part of a practicum opportunity. Using secondary data collected by the Fit Family Challenge during the 2011 year, the goals of this thesis were: to assess individual changes; evaluate short-term impact; and describe the community engagement process. ^ Methods: SA Sports collected quantitative and qualitative data during the implementation and evaluation of the FFC program. SA Sports allowed the used of de-identified data to be analyzed to study the aims of this thesis. ^ Results: The program was able to provide families with the knowledge, information, and opportunity to exercise as a family and cook healthier meals. School district coordinators were generally satisfied and illustrated the benefits of a community partnership. An authentic community engagement was present highlighting the importance of communication, collaboration and the sustainability of such partnerships in the community. ^ Conclusion: The success of an obesity program should focus on triggers that initiate behavioral change rather than physiological changes. The evaluation was guided by a community engagement approach, which illustrated the development of new partnerships and the strengthening of other collaborations. Ultimately, the engagement approach empowered the community to identify their own problems and build collaboration, rather than tackling obesity prevention alone. ^

This research focused on the topic of end-of-life planning and decision-making for adults affected by mental retardation. Adults with mental retardation have unique challenges in this regard, including difficulty communicating their wishes without assistance and diminished decision-making skills. The primary research objective was to identify factors that can affect opportunities for adults with mental retardation in community-based services settings (and their advocates) to be involved in planning and deciding about their own end-of-life experience. ^ A descriptive qualitative inquiry was designed to explore issues related to death and dying, and the notion of end-of-life planning, from the perspective of adults with mental retardation who receive publicly-funded community services ("clients") and family members of individuals who receive such services. Study participants were recruited from a single mental retardation service provider in a large urban setting (the "Agency"). Sixteen clients and 14 families of Agency clients took part. Client data collection was accomplished through face-to-face interviews, focus group meetings, and record reviews; family members were involved in a face-to-face interview only. ^ An initial coding scheme was developed based upon literature and policy reviews, and themes related to the research questions. Analysis involved extracting data from transcripts and records and placing it into appropriate thematic categories, building support for each theme with the accumulated data. Coding themes were modified to accommodate new data when it challenged existing themes. ^ Findings suggest that adults with mental retardation do have the requisite knowledge, interest, and ability to participate in decisions about their end-of-life experience and handling of affairs. Siblings are overwhelmingly the chosen future surrogates and they (or their children) will likely be the end-of-life advocates for their brothers and sisters affected by mental retardation. Findings further point to a need for increased awareness, accurate information, and improved communication about end-of-life issues, both in general and particular to adults affected by mental retardation. Also suggested by the findings is a need to focus on creating accommodations and adaptations that can best uncover a person's authentic views on life and death and related end-of-life preferences. Practical implications and suggestions for further research are also discussed. ^

Sucinta semblanza y temprana valoración de la vida y la obra de Gregorio Weinberg sobre su quehacer ligado a un vasto proyecto cultural para sentar las bases de un nuevo humanismo incluyente, que tiene en el libro su símbolo más alto. En una primera aproximación se lo descubre siempre preocupado por repensar y corregir la tentación esencialista, a histórica y excluyente en que puede incurrir el quehacer filosófico. Su labor consistió en una vida al servicio de la reflexión crítica, la educación y la edición. Vio en libros, editoriales y bibliotecas, auténticas empresas culturales de largo alcance y radical incidencia en el avance de una sociedad.

En este ensayo se procura conceptualizar acerca de cómo los niños, niñas y docentes construyen interpretaciones compartidas en el aula. Desde el análisis dialógico (Bajtin), entendemos el aprendizaje de los alumnos desde sus “respuestas" en relación con las voces de los maestros, textos y pares. Distinguimos el tratamiento unívoco del lenguaje, propio del recitado; de su tratamiento como instrumento de pensamiento, en el intercambio dialógico y en el discurso interactivo. Se analiza como se guía a un grupo de escolares en una institución pública de educación especial; centrando la mirada en las interacciones conversacionales del debate y preguntándonos qué patrones contribuyen más eficazmente a sus aprendizajes. El diseño supone la búsqueda de lo cualitativo, lo irrepetible y lo singular; incorporando el paradigma “indiciario" (Guinzburg). En la sistematización registramos, que las preguntas auténticas indican que el maestro otorga prioridad al razonamiento por sobre la memorización. Se caracteriza la interacción cercana al debate, como apuntes colaborativos. Esta construcción dialógica de sentido promueve el aprendizaje, porque genera respuestas con argumentación y facilita el diálogo entre lo nuevo y lo ya conocido. Así, el modo en que los alumnos piensan y lo que pueden aprender está relacionado a la modalidad que los docentes desarrollan para responder a sus preguntas.

Sin embargo, la distinción hecha por el filósofo entre escepticismo pirrónico y académico no es correcta en absoluto. El objetivo de este trabajo es mostrar que quizá la filosofía escéptico-naturalista de Hume no se halla tan distanciada del pirronismo auténtico como lo afirma su propio autor. En tal sentido, intentaremos desentrañar si el 'mitigado' escepticismo del escocés no esconde cierto pirronismo inconsciente, señalando que la clave puede hallarse en el desenlace del escepticismo humeano: la 'cura de la naturaleza'

La magia ha regresado, y los narradores, aunque sus relatos sean inconclusos, indiferentes e incluso frívolos, interpelan al lector como los grandes realistas (Lukács 1971) porque el efecto de los textos es indicial. Señalan -con brutalidad o con delicia- el desfasaje entre lo que se dice y lo que se hace o lo que se deja de hacer. Nombran así, con palabras comunes y sin ninguna distancia, deseos singulares: los suyos y los nuestros (Aira 1988: 59)

Dentro del expandido mundo doméstico que el poema presenta no se halla una respuesta clara para la presencia de los pretendientes, como tampoco para el rol social desempeñado por algunos servidores cuya característica definitoria es haber sido nobles y expresar en la actualidad una prueba fehaciente de las variaciones de la fortuna. Tal es el caso de Euriclea y Eumeo. El trabajo propone la discusión de la elaboración de los límites entre una clase social y otra, así como la constitución de las fronteras entre ellas, para interpretar la forma en que se articulan en el poema la situación social de personajes menores con la adhesión al héroe protagonista.
