The relationships among avian uncoupling protein (avUCP) mRNA expression, heat production, and thyroid hormone metabolism were investigated in 7-14-day-old broiler chicks (Gallus gallus) exposed to a low temperature (cold-exposed chicks, CE) or a thermoneutral temperature (TN). After 7 days of exposure, CE chicks exhibited higher heat production (+83%, P < 0.01), avUCP mRNA expression (+20%, P < 0.01), and circulating triiodothyronine (T-3) levels (+104%, P = 0.07) for non-statistically different body weights and feed intake between 3 and 7 days of exposure as compared to TN chicks. Plasma thyroxine (T-4) concentration was clearly decreased in CE chicks (-33%, P = 0.06). The lower hepatic inner-ring deiodination activity (-47%) and the higher renal outer-ring deiodination activity (+75%) measured in CE compared to TN chicks could partly account for their higher plasma T3 concentrations. This study describes for the first time the induction of avUCP mRNA expression by low temperature in chickens, as it has been previously shown in ducklings, and supports the possible involvement of avUCP in avian thermogenesis. Two experiments were carried out to evaluate the effect of supplementation with different nitrogenous compounds on the activities of carboxymethil cellulase (CMCase) and glutamate dehydrogenase (GDH). In the first experiment, four treatments were evaluated in vitro: cellulose, cellulose with casein, cellulose with urea, and cellulose with casamino acids. After 6, 12 and 24 hours of incubation, CMCase and GDH activity, pH, and concentrations of ammonia nitrogen (AN) and microbial protein were measured. In the three incubation periods, the concentration of AN was higher when urea was used as a supplemental source of nitrogen. The activity of CMCase was higher with the addition of urea and casamino acids when compared with the control and the casein treatment. Supplementation with casamino acids provided higher GDH activity when compared with the control at 6 hours of incubation. At 12 hours of incubation, the GHD activity was also stimulated by casein. At 24 hours, there was no difference in GHD activity among treatments. In the second experiment, three rumen-fistulated bulls were used for in situ evaluation. Animals were fed Tifton hay (Cynodon sp.) ad libitum. The treatments consisted of control (no supplementation), supplementation with non-protein nitrogenous compounds (urea and ammonium sulphate, 9:1) and supplementation with protein (albumin). In treatments with nitrogenous compound supplementation, 1 g of crude protein/kg of body weight was supplied. The experiment was conducted in a 3 × 3 Latin square design. The measurements were performed at 6, 12 and 24 hours after supplementation. No difference in GDH activity was observed among treatments. The control treatment showed higher CMCase activity when compared with the treatments containing supplemental sources of nitrogen. However, urea supplementation provided higher CMCase activity compared to albumin.

O objetivo neste trabalho foi avaliar o consumo, a digestibilidade total e parcial da matéria seca (MS) e dos nutrientes e os parâmetros ruminais de novilhos alimentados com rações à base de resíduo do processamento do caroço de algodão (RPCA) suplementado com levedura seca de cana-de-açúcar. Foram utilizados seis novilhos Nelore canulados no rúmen e duodeno, alimentados com rações compostas de 80% RPCA e 20% concentrado, composto de milho, ureia e levedura nos níveis de 0, 20 ou 40%. O delineamento experimental adotado para análise do consumo e da digestibilidade foi em blocos casualisados e a análise dos parâmetros ruminais foi em parcela subdividida. Os consumos e a digestibilidade de nutrientes não foram influenciados pela suplementação com levedura, fato que pode estar relacionado ao alto valor da fração C do resíduo do processamento do RPCA (44,51% MS) e à baixa fração proteica de lenta degradação da levedura, que impediu a sincronização energia/nitrogênio para maior degradação do resíduo. O pH do líquido ruminal decresceu linearmente nos animais mantidos com as dietas suplementadas com levedura. em todas as dietas, o pH e a concentração de N-NH3 foram adequados para o crescimento microbiano. As concentrações ruminais de acetato, propionato, butirato e ácidos graxos voláteis totais e a relação acetato:propionato não foram influenciadas pela suplementação com levedura. O resíduo do processamento do caroço de algodão é uma opção a ser utilizada no arraçoamento de bovinos.

Neste trabalho, avaliou-se a fermentação da cana-de-açúcar queimada, ensilada com ou sem uso de aditivo seco. Os tratamentos (seis no total) consistiram da silagem de cana crua ou queimada, adicionada de 0, 50 ou 100 g/kg de milho desintegrado com palha e sabugo (MDPS), com base no peso verde da forragem. Foram determinados os teores de MS, PB, nitrogênio insolúvel em detergente ácido (NIDA), FDN, FDA, celulose, hemicelulose e lignina. Na avaliação das características fermentativas, foram determinados os valores de carboidratos solúveis, o poder tamponante, o pH e as concentrações de nitrogênio amoniacal e etanol. Como características microbiológicas, avaliou-se o desenvolvimento de leveduras. A inclusão de MDPS elevou os teores de MS e reduziu discretamente os teores de N-NH3 e etanol das silagens, não ocasionando efeito nos valores de pH e na população de leveduras. A presença do fogo reduziu a concentração de MS das silagens, elevou os teores de etanol e leveduras e diminuiu os teores de N-NH3. A fermentação etanólica durante a ensilagem não foi controlada com a inclusão de aditivo seco ou com o uso do fogo.

The effect of four extracts from neem seeds (Azadirachta indica) containing 2000, 5000, 9000 and 10,000 ppm of azadirachtin A (AZA), quantified by high-performance liquid chromatography (HPLC) and diluted to 1.25%; 2.5%; 5.0%; 10.0% and 12.8% was verified by in vitro tests with engorged females and larvae of the cattle tick Rhipicephalus micro plus. The results from the bioassays with the engorged females showed that the main toxic effect of the extracts was reduction of the reproductive parameters, with a sharp drop in the number of eggs laid and the hatching rate, mainly when the extracts were diluted to 10.0% and 12.8%. The product effectiveness (PE) calculations for all the solutions tested showed that the AZA solution at 10,000 ppm (N10) was the most effective. However, statistical analysis of the PE data obtained for the proportional AZA concentrations in the different diluted extracts showed significance (P<0.05) of the effects included in the model (extract dilution, principle effect (classificatory) of the assay (extract) and the interaction between the two), indicating significant variations due to the dilution, the test and the interaction between the two factors in the tests with engorged females. For solutions N2, N5, and N9, it was not possible to estimate LC(90) values in the dilution range tested. The lowest LC(50) was observed for extract N5, and although extract N10 was the only extract for which the LC(90) could be estimated within the range tested, the LC(50) was higher than for N5 and N9. These results suggest that substances other than AZA present in the extracts influenced the efficacy, especially up to a certain LC range. In the tests with larvae, no mortality was observed, indicating zero effectiveness of all the extracts tested. The results of the tests with engorged females showed that the neem extracts had acaricide activity, inhibiting egg laying and the larval hatching rate. Complementary studies are necessary to develop new methods to isolate and/or identify other substances besides AZA contained in this plant, to enable using products made from it as acaricides. In Natal still dominates the use of individual disposal systems for domestic sewage, once only 29% of the city has a sewarage system. Wastes that are accumulated in these individual treatment systems should be exhausted periodically, service provided by collector entrepreneurs. Some of these companies causing major damage to the environment. In Natal, only two companies have their own septage (RESTI) treatment system, which were designed with parameters from domestic sewage generating strain and inefficient systems. Therefore, the characterization becomes essential as a source of parameters for their design. Thus, this work presents the physical-chemical and microbiological characterization of waste pumped from individual sewage treatment systems. Samples collections were made weekly from 5 different trucks at the reception point on the treatment plant on the point of the preliminary treatment. From each truck it was taken 5 samples during the discharge in order to make a composite sample. Afterwards, samples were carried out to laboratory and analyses for determination of temperature, pH, conductivity, BOD, COD, nitrogen (ammonia e organic), alkalinity, oils, phosphorus, solids, faecal coliforms and helminth egg. The results were treated as a single database, and ranked according to its generating source (multi and single house, lodging, health, services and / or food), area of origin (metropolitan, south and north) and type of system (cesspits, septic tank and / or sink). Through these data it was possible to verify that the type of system adopted by most in Natal and the metropolitan region is cesspit, besides to confirm the difference between the septage of areas with a population have different social and economical characteristics. It was found that the septage have higher concentrations than domestic sewage, except for thermotolerant coliforms that showed concentrations of 1,38E+07. Among the parameters studied, is the median values identified for COD (3,549 mg / L), BOD (973mg / L) and total solids (3.557mg / L). The volatile fraction constitutes about 70% of the total solids of the septage. For helminths has been a median of 7 eggs/L. In general, the characteristics of the waste followed the variability found in the literature reviewed for all variables, showing high amplitudes

The urban drainage is one of the powers of environmental sanitation and its scope is the quantitative and qualitative aspects. In decision making of managers and the engineering aspects of design are almost always taken into account only the quantitative aspects. However, the waters of the runoff have the highest concentrations of pollutants at the beginning of precipitation. Thus, if the plot pollution removed, the remaining portion can be used for other purposes. This work has aimed to present the variation of water quality of two drainage basins in the city of Natal / RN-Brazil to support the implementation of drainage to consider the qualitative aspect, and identify potential for the use of water. The basins (M and C) are analyzed closed-type, are in the urban area, are predominantly residential occupation and its waters are used for detention ponds and infiltration. The samples were divided into three phases, the first two direct to final points in a basin and the third in traps distributed over the surface drainage. The parameters had been analyzed were pH, conductivity, dissolved oxygen, Color, Turbidity, COD, Ammonia, nitrite, nitrate, total phosphorus, orthophosphate, Sediments solids, total solids, chloride, sulfate, alkalinity, calcium, magnesium, sodium, potassium, Heavy Metals (Chromium, Cadmium, Lead, Zinc and Copper), Eschichia coli and total coliforms. The parameters studied showed high initial pollution load, events and located in different proportions, except nitrite, heavy metals and biological indicators. The size of the surface drainage and topographic its features influence the quality of water. However, the form of sampling is crucial in the qualitative study in the basin. The samplers developed at work, were generated economic and representative results. The urban rainwater presents organic faecal indicators. The runoff of water from both basins shows no risk of salinity and sodicity for use in irrigation, should be noted the content of chloride in the choice of method of irrigation

Only 32% of the population of Natal is attended by sewage, while the remaining population use pits and septic tanks. The characterization of the contents of septic tanks and pits contributes to the performance of such system and may guide the decision on treatment of these contents. The main of this research is to characterize the contents of interior residential pits and septic tanks in the greater Natal, with the following specific goals: to develop and manufacture a sampler capable of collecting a representative sample of the entire column (the surface scum, the clarified liquid and sludge bottom); to compare the contents of the tanks with the pits; to compare the contents of the septage from vacuum trucks; to relate the composition content with socioeconomic characteristics of households; to compare the content in both chambers of the septic tanks in series; to assess the situation of the content before and six months after the cleanness; and ultimately propose a pilot scale plant for treatment of septage. Once the sampler was developed, samples were collected within 14 septic tanks and 10 pits in many districts of Natal. Medians of the 24 systems were obtained: temperature, pH, conductivity, oil and grease, total solids, total suspended solids and sediments of 28.0 °C, 6.95; 882 mS/cm, 75.2 mg/L; 10,169 mg/L, 6,509 mg/L and 175 mL/L respectively; 111.0 mgN/L for ammonia, 130.5 mgN/ L for organic nitrogen, 0.2 mgN/L for nitrite, 0.4 mg/L for nitrate; 8935 mgO2/L for COD, 29.2 mgP/L for total phosphorus, thermotolerant coliforms from 9.95 E +06 CFU/100mL helminth eggs and 9.2 eggs/L with a maximum concentration of 688 eggs/L and minimum of 0 eggs/L. Medians of organic nitrogen and TKN were significantly different between groups of tanks and pits. The systems with cleanness gap from 11 and 20 years presented the higher concentrations for most variables. The effluent from the toilets and bathrooms participate more effectively in contributing fractions of solids, alkalinity, nitrogen, COD, total phosphorus, thermotolerant coliforms and helminth eggs. The systems used by socioeconomics class with income from R$ 3,700.00 to R$ 7,600.00, presented higher concentrations for COD, nitrogen, solids and helminth eggs. The first of the two chambers had always presented higher concentrations over the second compartment. The analysis of variance for most variables, showed that the values of septic tanks, pits and septage from vacuum trucks belong to the same group. In the samples taken after cleanness, the median of pH and temperature increased, while alkalinity, COD, organic nitrogen, total phosphorus, ammonia and helminth eggs decreased. The oils and greases and thermotolerant coliforms had slightly varied due to the continuous release of sewage into the systems that maintained their steady state concentrations.

Although the good performance in organic matter and suspended solids removal, the anaerobic reactors are unable to remove ammonia nitrogen from sewage, which makes indispensable to include a step of post-treatment for removal of ammonia or nitrate as necessary. This paper presents the performance of a new variant technology, where the nitrification unit, preceded by anaerobic units, is a submerged aerated biological filter, without continuous sludge discharge in their daily operation. The oxygenation system is very simple and inexpensive, consisting of perforated hoses and compressors. The anaerobic reactors are a septic tank with two chambers followed (8.82 m³) and two parallel anaerobic filters (36 m³ each) filled with ceramic bricks and conics plastic parts. Both followed aerated filters were filled with cut corrugated conduit. The study evaluated the behavior of the system with constant domestic sewage flow (10 m³/d) and different aeration conditions, are these: stage 01, when applied air flow of 0.01 m³ air/min in both aerated filter; stage 02, remained in the initial air flow rate in the second aerated filter and increased at the first to 0.05 m³ air/min; at last, at last, in stage 03, the air flow rate of first aerated filter was 0.10 m³ air/min and on the second remained at 0.01 m³ air/min. The filter FA1 received load of 0.41 kg COD/m³.d, 0.37 kg COD/m³.d and 0.26 kg COD/m³.d on phases 01, 02 and 03, respectively. The FA2 received loads of 0.25 kg COD/m³.d, 0.18 kg COD/m³.d and 0.14 kg COD/m³.d on phases 01, 02 and 03, respectively. During stage 01, were found the following results: 98% removals of BODtotal and 92% of CODtotal, with effluent presenting 9 mg/L of BODtotal final average and 53 mg/L of CODtotal average; suspended solids removals of 93%, with a mean concentration of 10 mg/L in the final effluent; 47% reduction of ammonia of FA2 to FAN 's, presenting average of 28 mg NNH3/ L of ammonia in the effluent with; the dissolved oxygen levels always remained around 2.0 mg/L. During stage 02, were found removals of 97% and 95% to BODtotal and suspended solids, respectively, with average final concentrations of 8 and 7 mg/L, respectively; was removed 60% of ammonia, whose final concentration was 16.3 mg NNH3/ L, and nitrate was increased to a final average concentration of 16.55 mg N-NO3/L. Finally, the stage 03 provided 6 mg/L of DBOtotal (98% removal) and 23 mg/L of CODtotal (95% removal) of final effluent concentrations average. At this stage was identified the higher ammonia oxidation (86%), with final effluent showing average concentration of 6.1 mg N-NH3/L, reaching a minimum of 1.70 mg N-NH3/L. In some moments, during stage 03, there was a moderate denitrification process in the last aerated filter. The average turbidity in the effluent showed around 1.5 NTU, proving the good biomass physical stability. Therefore, the results demonstrate the submerged biological filters potential, filled with high void ratio material (98%), and aerated with hoses and compressor adoption, in the carbonaceous and nitrogenous matter oxidation, also generating an effluent with low concentration of solids

Waste stabilization ponds are the main technology in use for domestic sewage treatment in Rio Grande do Norte State (RN), northeast Brazil. The are around 80 systems, constructed mainly by municipal city halls, being series comprised by a primary facultative pond followed by two maturation ponds the most used configuration. Due to problems related with the production and destination of sludge and generation of bad odors, the designers have avoided the use of anaerobic lagoons. The majority of systems are rarely monitored to verify their efficiencies and to get new project parameters for future designing. This work has as purpose to make a diagnosis of efficiency of three series of waste stabilization pond series (WSPS) of Jardim Lola 1, Jardim Lola 2 and Beira Rio, located in the North Zone of the city of the Natal/RN, treating domestic raw sewage, on the removal of organic matter and thermotolerant coliform, comparing the operational conditions of the systems this inside of the bands foreseen in the project, through parameters BOD5, QOD, thermotolerant coliforms, dissolved oxygen, pH, temperature, ammoniac nitrogen, total and suspended solids. The work was carried through in the WSPS, all constituted by a primary facultative pond followed by two maturation ponds. Socioeconomic characteristics of population are predominantly low and all the plants are very near of the contributing basins. The series were monitored from of May the November of 2002, totalizing 20 collections of grab samples of raw sewage and ponds effluents between 8:00 and 9:50 h. The main aspect to be detached by the results was the great concentration of organic matter (BOD and COD) and microorganisms the raw sewage which were around two times more concentrated than those values foreseen one in project. Considering all series the highest removals of organic matter were observed in system Beira Rio (84 and 78% of BOD and COD, respectively), which presented high hydraulic detention time (TDH = 89 days). On the other hand, Jardim Lola 1 and Jardim Lola 2 presented a much lower values of HDT (36 days and 18 days respectively) and their removals of BOD and COD were the same (76% and 60%, respectively). The Beira Rio WSPS, was the most efficient verified in relation to solids and ammonia, proving the great influence of the operational variables such as HDT and applied surface organic loadings on the performance of pond series. Although the treatment plants have reached efficiencies of thermotolerant coliforms around 99,999%, the concentrations in the final effluent can be considered very high for launching in aquatic bodies, particularly those produced by Jardim Lola 1 and Jardim Lola 2 series

The release of reactive oxygen specie (ROS) by activated neutrophil is involved in both the antimicrobial and deleterious effects in chronic inflammation. The objective of the present investigation was to determine the effect of therapeutic plasma concentrations of non-steroidal anti-inflammatory drugs (NSAIDs) on the production of ROS by stimulated rat neutrophils. Diclofenac (3.6 µM), indomethacin (12 µM), naproxen (160 µM), piroxicam (13 µM), and tenoxicam (30 µM) were incubated at 37ºC in PBS (10 mM), pH 7.4, for 30 min with rat neutrophils (1 x 10(6) cells/ml) stimulated by phorbol-12-myristate-13-acetate (100 nM). The ROS production was measured by luminol and lucigenin-dependent chemiluminescence. Except for naproxen, NSAIDs reduced ROS production: 58 ± 2% diclofenac, 90 ± 2% indomethacin, 33 ± 3% piroxicam, and 45 ± 6% tenoxicam (N = 6). For the lucigenin assay, naproxen, piroxicam and tenoxicam were ineffective. For indomethacin the inhibition was 52 ± 5% and diclofenac showed amplification in the light emission of 181 ± 60% (N = 6). Using the myeloperoxidase (MPO)/H2O2/luminol system, the effects of NSAIDs on MPO activity were also screened. We found that NSAIDs inhibited both the peroxidation and chlorinating activity of MPO as follows: diclofenac (36 ± 10, 45 ± 3%), indomethacin (97 ± 2, 100 ± 1%), naproxen (56 ± 8, 76 ± 3%), piroxicam (77 ± 5, 99 ± 1%), and tenoxicam (90 ± 2, 100 ± 1%), respectively (N = 3). These results show that therapeutic levels of NSAIDs are able to suppress the oxygen-dependent antimicrobial or oxidative functions of neutrophils by inhibiting the generation of hypochlorous acid.

Special diets are used to mitigate many human diseases. When these diets require changes in carbohydrate content, then sweetness becomes an important characteristic. The range of low-calorie sweeteners available to the food industry is expanding. It is essential to have an exact knowledge of the relative sweetness of various sweeteners in relation to different sucrose concentrations. The objective of this study was to determine the variation on the relative sweetness of aspartame (APM), stevia [Stevia rebaudiana (Bert.) Bertoni] leaf extract (SrB) and the mixture cyclamate/saccharin - two parts of cyclamate and one part of saccharin - (C/S) with the increase in their concentrations, and in neutral and acid pH in equi-sweet concentration to 10% sucrose, using magnitude estimation. Sweetness equivalence of SrB in relation to sucrose concentrations of 20% or higher and of APM and C/S to sucrose concentrations of 40% or higher could not be determined, because a bitter taste predominated. The potency of all sweeteners decreased as the level of sweetner increased. In equi-sweet concentration of sucrose at 10%, with pH 7.0 and pH 3.0, the potency was practically the same for all sweeteners evaluated.
