Although somatic cell nuclear transfer (SCNT) is a promising tool, its potential use is hampered by the high mortality rates during the development to term of cloned offspring. Abnormal epigenetic reprogramming of donor nuclei after SCNT is thought to be the main cause of this low efficiency. We hypothesized that chromatin-modifying agents (CMAs) targeting chromatin acetylation and DNA methylation could alter the chromatin configuration and turn them more amenable to reprogramming. Thus, bovine fibroblasts were treated with 5-aza-2'-deoxycytidine (AZA) plus trichostatin (TSA) or hydralazine (HH) plus valproic acid (VPA) whereas, in another trial, cloned bovine zygotes were treated with TSA. The treatment of fibroblasts with either AZA + TSA or HH + VPA increased histone acetylation, but did not affect the level of DNA methylation. However, treatment with HH + VPA decreased cellular viability and proliferation. The use of these cells as nuclear donors showed no positive effect on pre- and postimplantation development. Regarding the treatment of cloned zygotes with TSA, treated one-cell embryos showed an increase in the acetylation patterns, but not in the level of DNA methylation. Moreover, this treatment revealed no positive effect on pre- and postimplantation development. This work provides evidence the treatment of either nuclear donor cells or cloned zygotes with CMAs has no positive effect on pre- and postimplantation development of cloned cattle.

Schizophrenia has been defined as a neurodevelopmental disease that causes changes in the process of thoughts, perceptions. and emotions, usually leading to a mental deterioration and affective blunting. Studies have shown altered cell respiration and oxidative stress response in schizophrenia; however, most of the knowledge has been acquired from postmortem brain analyses or from nonneural cells. Here we describe that neural cells, derived from induced pluripotent stem cells generated from skin fibroblasts of a schizophrenic patient, presented a twofold increase in extramitochondrial oxygen consumption as well as elevated levels of reactive oxygen species (ROS), when compared to controls. This difference in ROS levels was reverted by the mood stabilizer valproic acid. Our model shows evidence that metabolic changes occurring during neurogenesis are associated with schizophrenia, contributing to a better understanding of the development of the disease and highlighting potential targets for treatment and drug screening.

Das metastasierende maligne Melanom ist durch eine geringe p53-Mutations-Rate und eine hohe Resistenz gegenüber Chemotherapie mit alkylierenden Agenzien wie Fotemustin (FM) und Temozolomid (TMZ) gekennzeichnet. In der vorliegenden Arbeit wurde die Rolle von p53 in der Resistenz von malignen Melanomzellen gegenüber FM untersucht und Möglichkeiten zur Sensitivierung von Melanomzellen gegenüber TMZ und FM aufgezeigt.rnAusgangspunkt war die Beobachtung, dass p53 Wildtyp (p53wt) Melanomzellen resistenter gegenüber FM sind als p53 mutierte (p53mt) Zellen. In der vorliegenden Arbeit wurde gezeigt, dass eine FM-Behandlung in p53wt Zellen eine Stabilisierung von p53 und eine Induktion des p53-Zielproteins p21 bewirkte. Mithilfe einer p53wt Zelllinie, welche einen p53 Knockdown trägt, konnte gezeigt werden, dass p53 für die geringe Apoptose-Rate nach FM-Behandlung verantwortlich ist. Eine Untersuchung der Interstrang-Crosslink (ICL)-Reparaturkapazität zeigte, dass p53mt Zellen im Gegensatz zu p53wt Zellen nicht in der Lage sind, FM-induzierte ICL zu reparieren. Dies ging mit einer im Vergleich zu p53wt Zellen starken DNA-Schadensantwort einher. Die Gene für die Proteine DDB2 und XPC wurden als durch FM regulierte DNA-Reparatur-Gene identifiziert, deren Induktion p53-abhängig und lang anhaltend (bis zu 144 h) erfolgt. Da XPC Knockdown-Zellen sensitiver als ihre Kontrollzellen gegenüber FM reagierten, konnte die biologische Relevanz von XPC bei der ICL-Reparatur bestätigt werden. Anhand von Xenograft-Tumoren wurde gezeigt, dass FM auch in situ eine Induktion von DDB2 und XPC auslöst. Die Beobachtung, dass DNA-Reparatur-Gene nach FM-Behandlung hochreguliert werden, liefert eine Erklärung für das schlechte Ansprechen von Melanomen auf eine Therapie mit ICL-induzierenden Chemotherapeutika.rnDes Weiteren befasste sich die vorliegende Arbeit mit Möglichkeiten zur Sensitivierung von Melanomzellen gegenüber den Chemotherapeutika TMZ und FM. In diesem Zusammenhang wurde Valproinsäure (VPA), ein in der Epilepsie-Therapie verwendetes Medikament und Histondesacetylase (HDAC)-Hemmer, bezüglich der chemosensitivierenden Wirkung untersucht. Zunächst konnte der in der Literatur häufig beschriebene stabilisierende Effekt von VPA auf „wildtypisches“ p53-Protein und destabilisierende Effekt auf mutiertes p53-Protein bestätigt werden. Zwei der vier untersuchten Zelllinien konnten mithilfe von VPA gegenüber TMZ sensitiviert werden, während nur eine der vier untersuchten Zelllinien gegenüber FM sensitiviert werden konnte. VPA begünstigt die Induktion von Apoptose, während der Effekt auf die Induktion von Nekrose nur gering ausfiel. Eine Wirkung von VPA auf die Aktivität des Resistenz-vermittelnden Enzyms O6-Methylguanin-DNA-Methyltransferase (MGMT) wurde nicht beobachtet. Zudem wurde ausgeschlossen, dass die Sensitivierung gegenüber TMZ und FM, welche S-Phase abhängige Gentoxine sind, auf einer VPA-induzierten Erhöhung der Proliferation beruht. Mithilfe einer Zelllinie, welche stabil dominant-negatives FADD (Fas-associated death domain) exprimiert, konnten keine Hinweise auf eine Beteiligung des extrinsischen Apoptose-Signalwegs an der VPA-vermittelten Sensitivierung gewonnen werden. Gleichzeitig wurde gezeigt, dass VPA keine Induktion der niedrig exprimierten Procaspase-8 verursachte. Mithilfe eines PCR-Arrays wurden transaktivierende und –reprimierende Effekte von VPA auf die Genexpression gezeigt, wobei das proapoptotische Protein BAX (Breakpoint cluster-2-associated x protein) als ein in der Sensitivierung involviertes Kandidatengen identifiziert wurde. Obwohl eine vollständige Aufklärung der dem Sensitivierungseffekt von VPA zu Grunde liegenden Mechanismen nicht erbracht werden konnte, zeigen die in dieser Arbeit erlangten Beobachtungen einen vielversprechenden Weg zur Überwindung der Resistenz von Melanomzellen gegenüber DNA-alkylierenden Zytostatika auf.rn

Background DNA polymerase γ (POLG) is the only known mitochondrial DNA (mtDNA) polymerase. It mediates mtDNA replication and base excision repair. Mutations in the POLG gene lead to reduction of functional mtDNA (mtDNA depletion and/or deletions) and are therefore predicted to result in defective oxidative phosphorylation (OXPHOS). Many mutations map to the polymerase and exonuclease domains of the enzyme and produce a broad clinical spectrum. The most frequent mutation p.A467T is localised in the linker region between these domains. In compound heterozygote patients the p.A467T mutation has been described to be associated amongst others with fatal childhood encephalopathy. These patients have a poorer survival rate compared to homozygotes. Methods mtDNA content in various tissues (fibroblasts, muscle and liver) was quantified using quantitative PCR (qPCR). OXPHOS activities in the same tissues were assessed using spectrophotometric methods and catalytic stain of BN-PAGE. Results We characterise a novel splice site mutation in POLG found in trans with the p.A467T mutation in a 3.5 years old boy with valproic acid induced acute liver failure (Alpers-Huttenlocher syndrome). These mutations result in a tissue specific depletion of the mtDNA which correlates with the OXPHOS-activities. Conclusions mtDNA depletion can be expressed in a high tissue-specific manner and confirms the need to analyse primary tissue. Furthermore, POLG analysis optimises clinical management in the early stages of disease and reinforces the need for its evaluation before starting valproic acid treatment.

PURPOSE Women with epilepsy apparently have a higher incidence of polycystic ovary syndrome (PCOS) than do women without epilepsy. Whether the underlying disease or the antiepileptic drug (AED) treatment is responsible for this increased risk is unknown, although clinical reports implicate valproic acid (VPA) as a potential cause. The steroidogenic enzymes 3beta HSDII (3beta-hydroxysteroid dehydrogenase) and P450c17 (17alpha-hydroxylase/17,20 lyase) are essential for C19 steroid biosynthesis, which is enhanced during adrenarche and in PCOS. METHODS To determine whether the AEDs VPA, carbamazepine (CBZ), topiramate (TPM), or lamotrigine (LYG) directly affect the activities of human 3beta HSDII and P450c17, we added them to yeast expressing human P450c17 or 3beta HSDII and assayed enzymatic activities in the microsomal fraction. RESULTS Concentrations of VPA < or = 10 mM had no effect on activities of P450c17; however, VPA inhibited 3beta HSDII activity starting at 0.3 mM (reference serum unbound concentration, 0.035-0.1 mM) with an IC50 of 10.1 mM. CBZ, TPM, and LTG did not influence 3beta HSDII or P450c17 activities at typical reference serum unbound concentrations, but did inhibit 3beta HSDII and P450c17 at concentrations >10-fold higher. CONCLUSIONS None of the tested AEDs influenced 3beta HSDII or P450c17 activities at concentrations normally used in AED therapy. However, VPA started to inhibit 3beta HSDII activity at concentrations 3 times above the typical reference serum unbound concentration. Because inhibition of 3beta HSDII activity will shift steroidogenesis toward C19 steroid production when P450c17 activities are unchanged, very high doses of VPA may promote C19 steroid biosynthesis, thus resembling PCOS. CBZ, TPM, and LTG influenced 3beta HSDII and P450c17 only at toxic concentrations.

Free drug measurement and pharmacodymanic markers provide the opportunity for a better understanding of drug efficacy and toxicity. High-performance liquid chromatography (HPLC)-mass spectrometry (MS) is a powerful analytical technique that could facilitate the measurement of free drug and these markers. Currently, there are very few published methods for the determination of free drug concentrations by HPLC-MS. The development of atmospheric pressure ionisation sources, together with on-line microdialysis or on-line equilibrium dialysis and column switching techniques have reduced sample run times and increased assay efficiency. The availability of such methods will aid in drug development and the clinical use of certain drugs, including anti-convulsants, anti-arrhythmics, immunosuppressants, local anaesthetics, anti-fungals and protease inhibitors. The history of free drug measurement and an overview of the current HPLC-MS applications for these drugs are discussed. Immunosuppressant drugs are used as an example for the application of HPLC-MS in the measurement of drug pharmacodynamics. Potential biomarkers of immunosuppression that could be measured by HPLC-MS include purine nucleoside/nucleotides, drug-protein complexes and phosphorylated peptides. At the proteomic level, two-dimensional gel electrophoresis combined with matrix-assisted laser desorption/ionisation time-of-flight (TOF) MS is a powerful tool for identifying proteins involved in the response to inflammatory mediators. Valproic acid (VPA) is a major therapeutic agent in the treatment of epilepsy and other neurological disorders. It is metabolized in humans and rats primarily along two pathways: direct glucuronidation to yield the acyl glucuronide (VPA-G) and beta-oxidation. We have shown much earlier in the Sprague-Dawley rat that i.v. administration of sodium valproate (NaVPA) caused a marked choleresis ( mean of 3.3 times basal bile flow after doses of 150 mg/kg), ascribed to the passive osmotic flow of bile water following excretion of VPA-G across the canalicular membrane. Active biliary pumping of anionic drug conjugates across the canalicular membrane is now believed to be attributable to transporter proteins, in particular Mrp2, which is deficient in the TR- ( a mutant Wistar) rat. In the present study, normal Wistar and Mrp2-deficient TR- rats were dosed i.v. with NaVPA at 150 mg/kg. In the Wistar rats, there was a peak choleretic effect of about 3.2 times basal bile flow, occurring at about 30 to 45 min postdose ( as seen previously with Sprague-Dawley rats). In TR- rats given the same i.v. dose, there was no evidence of postdose choleresis. The choleresis was correlated with the excretion of VPA-G into bile. In Wistar rats, 62.8 +/- 7.7% of the NaVPA dose was excreted in bile as VPA-G, whereas in TR- rats, only 2.0 +/- 0.6% of the same dose was excreted as VPA-G in bile ( with partial compensatory excretion of VPA-G in urine). This study underlines the functional ( bile flow) consequences of biliary transport of xenobiotic conjugated metabolites.

Developmental neurotoxicity is a major issue in human health and may have lasting neurological implications. In this preliminary study we exposed differentiating Ntera2/clone D1 (NT2/D1) cell neurospheres to known human teratogens classed as non-embryotoxic (acrylamide), weakly embryotoxic (lithium, valproic acid) and strongly embryotoxic (hydroxyurea) as listed by European Centre for the Validation of Alternative Methods (ECVAM) and examined endpoints of cell viability and neuronal protein marker expression specific to the central nervous system, to identify developmental neurotoxins. Following induction of neuronal differentiation, valproic acid had the most significant effect on neurogenesis, in terms of reduced viability and decreased neuronal markers. Lithium had least effect on viability and did not significantly alter the expression of neuronal markers. Hydroxyurea significantly reduced cell viability but did not affect neuronal protein marker expression. Acrylamide reduced neurosphere viability but did not affect neuronal protein marker expression. Overall, this NT2/D1 -based neurosphere model of neurogenesis, may provide the basis for a model of developmental neurotoxicity in vitro.

Introdução: existem múltiplos estudos acerca da interacção entre o ácido valpróico (AVP) e o meropenem, mas não estão bem definidas as recomendações para o seu uso concomitante, visto haver uns que indicam que os dois fármacos não devem ser administrados juntos e outros que apenas recomendam que os níveis de valproatémia sejam monitorizados. Também não existe informação consistente sobre possíveis repercussões clínicas desta interacção. O objectivo deste estudo foi analisar a interacção entre o AVP e o meropenem e as suas repercussões clínicas, em doentes hospitalizados e cronicamente medicados com o primeiro. Material e Métodos: trata-se de um estudo retrospectivo de uma amostra de doentes internados num hospital distrital no período de um ano. Os critérios de inclusão foram a existência de pelo menos uma determinação do nível sérico de AVP na semana anterior à prescrição de meropenem e outra durante o tratamento concomitante com os dois fármacos. Foi efectuada a análise estatística de 18 episódios. Resultados: Verificou-se um declínio médio estatisticamente significativo (p<0,001) de cerca de 83,4% dos níveis séricos de AVP após o início da terapêutica com meropenem e foram objectivadas crises convulsivas em 50% dos doentes. Discussão: a interacção entre os dois fármacos em questão é potencialmente grave, sendo que o número de doentes com manifestações clínicas desta interacção é maior que em estudos semelhantes. Conclusão: concluiu-se que a utilização concomitante dos dois fármacos deve ser evitada.

Les Troubles du Spectre Autistique (TSA) sont caractérisés par deux principaux symptômes : des difficultés de communication sociale et des comportements stéréotypés et intérêts restreints. Les TSA touchent 5 fois plus les garçons que les filles et une augmentation de la prévalence exponentielle et continue a été observée aux États-Unis ces dernières décennies. Cette augmentation ne peut s’expliquer par les facteurs génétiques à eux seuls qui ne représentent que 5 à 15% des cas de TSA. Il est donc indispensable d’identifier de potentiels facteurs de risque environnementaux des TSA. Le but de ce travail est d’étudier différents facteurs environnementaux potentiellement modifiables dans le développement de phénotypes autistiques dans différents modèles précliniques des TSA. Les objectifs spécifiques sont : (i) caractériser les effets neurocomportementaux provoqués par une exposition périnatale simultanée à 5 perturbateurs endocriniens parmi les plus prévalent dans notre environnement quotidien (DEHP, DBP, DiNP, BDE-47, BDE-99) à de faibles doses pertinentes pour l’exposition humaine, (ii) identifier les effets neurocomportementaux associés à une altération périconceptionnelle du microbiote maternelle (iii) déterminer les effets neurocomportementaux associés à une altération périconceptionnelle du métabolisme monocarboné. Les résultats présentés dans cette thèse démontrent le potentiel de chacun de ces facteurs environnementaux d’altérer le développement cérébral fœtal. Chaque condition expérimentale a provoqué l’apparition de traits autistiques chez les rats, avec des spécificités comportementales pour chaque exposition développementale. Des déficits d’interactions sociales ont été observés dans chaque situation expérimentale, associés soit à de l’anxiété, de l’hyperactivité, des altérations d’intégration sensorimotrice, et/ou des stéréotypies. Cela nous force à considérer les TSA comme une pathologie aux multiples facettes où l’hétérogénéité des tableaux cliniques est représentative de l’hétérogénéité des causes possibles. La multitude des interactions environnementales courantes possibles avec l’épigénome pourrait être à la base de la grande diversité observée dans la sévérité des symptômes et / ou des comorbidités des TSA. Ce travail ouvre des perspectives futures de prévention ciblée des TSA fondées sur de potentielles modifications de l’environnement comme la réduction de l’exposition aux perturbateurs endocriniens, ou des supplémentations en donneurs monocarbonés (e.g. acide folique) et/ou probiotiques.

Background: Over the last few years, microdeletions of the 22q11.2 region responsible for DiGeorge syndrome, or velocardiofacial syndrome, have been increasingly related to neuropsychiatric disorders including schizophrenia and bipolar disorders. These signs seem to be related to certain genes located in the hemideleted region as the proline dehydrogenase (PRODH) and the catecholo-methyltransferase (COMT) genes. The PRODH or proline oxidase deficiency is responsible for hyperprolinemia type 1 (HPI) also causing psychiatric manifestations. Case Report: We describe a 17 year old boy with previous mild psychomotor and speech delay, mild cognitive impairment, and obsessive behaviours who started his adolescent psychiatric care presenting irritablemood and aggressive behaviour with schizophrenia symptoms that scored a “severely ill” level PANSS assessment. Symptoms got worse when he was treated with valproic acid and plasma aminoacids showing increase in alanine and proline, suggested a mitochondrial involvement of the proline metabolic pathway. Results: Mild dysmorphia suggested a possible 22q11.2 deletion genetically confirmed involving both the PRODH and COMT regions. HPI that can present with psychiatric features is however a recessive disorder and therefore the symptoms could not be solely explained by this genetic deletion. Additional investigations also showed disclosed a p.L289m (c.1865 T > A) mutation in the PRODH gene. Discussion: We believe that the association of this mutation together with the 22q11.2 deletion would lead to a decrease of functional protein. Although it may be difficult to diagnosis chromosomal abnormalities in patients with no clear malformations and mild dysmorphic features as in this patient we emphasize need to investigate the aetiology in patients with psychiatric symptoms, especially if they have other systemic manifestations such as developmental delay or psychotic symptoms, as it may be important in the management of the patients.

Introduction: Abdominal pain, in etiology sometimes difficult to be defined, is a frequent complaint in childhood. Abdominal epilepsy is a rare cause of abdominal pain. Objectives: In this article, we report on 5 year old girl patient with abdominal epilepsy. Methods: Some investigations (stool investigation, routine blood tests, ultrasonography (USG), electrocardiogram (ECHO) and electrocardiograpy (ECG), holter for 24hr.) were done to understand the origin of these complaints; but no abnormalities were found. Finally an EEG was done during an episode of abdominal pain and it was shown that there were generalized spikes especially precipitated by hyperventilation. The patient did well on valproic acid therapy and EEG was normal 1 month after beginning of the treatment. Discussion: The cause of chronic recurrent paroxymal abdominal pain is difficult for the clinicians to diagnose in childhood. A lot of disease may lead to paroxysmal gastrointestinal symptoms like familial mediterranean fever and porfiria. Abdominal epilepsy is one of the rare but easily treatable cause of abdominal pain. Conclusion: In conclusion, abdominal epilepsy should be suspected in children with recurrent abdominal pain.

L’acétylation est une modification post-traductionnelle des protéines essentielles. Elle est impliquée dans bon nombre de processus cellulaires importants comme la régulation de la structure de la chromatine et le recrutement de protéines. Deux groupes d’enzymes, soient les lysines acétyltransférases et les lysines désacétylases, régulent cette modification, autant sur les histones que sur les autres protéines. Au cours des dernières années, de petites molécules inhibitrices des désacétylases ont été découvertes. Certaines d’entre elles semblent prometteuses contre diverses maladies telles le cancer. L’acide valproïque, un inhibiteur de deux des trois classes des désacétylases, a un effet antiprolifératif chez plusieurs organismes modèles. Toutefois, les mécanismes cellulaires sous-jacents à cet effet restent encore méconnus. Ce mémoire met en lumière l’effet pH dépendant de l’acide valproïque sur différentes voies cellulaires importantes chez la levure Saccharomyces cerevisiae. Il démontre que ce composé a la capacité d’inhiber la transition entre les phases G1 et S par son action sur l’expression des cyclines de la phase G1. De plus, il inhibe l’activation de la kinase principale de la voie activée suite à un stress à la paroi cellulaire. L’acide valproïque occasionne également un arrêt dans la réplication de l’ADN sans y causer de dommage. Il s’agit là d’un effet unique qui, à notre connaissance, n’est pas observable avec d’autres agents qui inhibent la progression en phase S.

Viscosupplements, used for treating joint and cartilage diseases, restore the rheological properties of synovial fluid, regulate joint homeostasis and act as scaffolds for cell growth and tissue regeneration. Most viscosupplements are hydrogels composed of hyaluronic acid (HA) microparticles suspended in fluid HA. These microparticles are crosslinked with chemicals to assure their stability against enzyme degradation and to prolong the action of the viscosupplement. However, the crosslinking also modifies the mechanical, swelling and rheological properties of the HA microparticle hydrogels, with consequences on the effectiveness of the application. The aim of this study is to correlate the crosslinking degree (CD) with these properties to achieve modulation of HA/DVS microparticles through CD control. Because divinyl sulfone (DVS) is the usual crosslinker of HA in viscosupplements, we examined the effects of CD by preparing HA microparticles at 1:1, 2:1, 3:1, and 5:1 HA/DVS mass ratios. The CD was calculated from inductively coupled plasma spectrometry data. HA microparticles were previously sized to a mean diameter of 87.5 µm. Higher CD increased the viscoelasticity and the extrusion force and reduced the swelling of the HA microparticle hydrogels, which also showed Newtonian pseudoplastic behavior and were classified as covalent weak. The hydrogels were not cytotoxic to fibroblasts according to an MTT (3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide) assay. © 2014 Wiley Periodicals, Inc.
