In order to select superior hybrids for the concentration of favorable alleles for resistance to papaya black spot, powdery mildew and phoma spot, 67 hybrids were evaluated in two seasons, in 2007, in a randomized block design with two replications. Genetic gains were estimated from the selection indices of Smith & Hazel, Pesek & Baker, Williams, Mulamba & Mock, with selection intensity of 22.39%, corresponding to 15 hybrids. The index of Mulamba & Mock showed gains more suitable for the five traits assessed when it was used the criterion of economic weight tentatively assigned. Together, severity of black spot on leaves and on fruits, characteristics considered most relevant to the selection of resistant materials, expressed percentage gain of -44.15%. In addition, there were gains for other characteristics, with negative predicted selective percentage gain. The results showed that the index of Mulamba & Mock is the most efficient procedure for simultaneous selection of papaya hybrid resistant to black spot, powdery mildew and phoma spot.

Grapevine is one of the most cultivated fruit crop worldwide with Vitis vinifera being the species with the highest economical importance due to the high quality standards of its berries. Nonetheless, it is also the most susceptible Vitis species to fungal pathogens. Among others, relevant fungal diseases currently threatening grapevine cultures are powdery mildew, caused by Erysiphe necator, and esca, a disease complex comprised of several fungi in which Phaeomoniella chlamydospora and Phaeoacremonium aleophilum participate.(...)

Nutzen und Umweltrisiken von gentechnisch verändertem (GV) Sommerweizen mit verbesserter Mehltauresistenz wurde von einem Verbund Schweizer Forschungsgruppen von 2008 bis 2010 an zwei Standorten im Freiland untersucht. GV Weizenlinien mit spezifischer Resistenz gegen Mehltau und allgemeiner Pilzresistenz wurde mit Kontrolllinien, kommerziellen Weizensorten sowie Gerste und Triticale verglichen. Neben der Mehltauresistenz wurden die Auswirkungen auf Insekten und Bodenlebewesen (Bakterien, Mykorrhizapilze, Bodenfauna) sowie die Auskreuzung auf Weizen untersucht. Mehrere GV Weizenlinien waren deutlich resistenter gegen Mehltau als ihre Kontrollen. Es wurden keine relevanten Auswirkungen auf Nichtzielorganismen, deren Biodiversität und ausgewählte Ökosystemleistungen gefunden. Insgesamt waren die Unterschiede zwischen GV und Kontrolllinien geringer als zwischen kommerziellen Weizensorten oder anderen Getreidearten. Auskreuzungsereignisse waren selten und wurden nur in kurzer Distanz zu GV Versuchspflanzen gefunden.

This study aimed to evaluate the impact of genetically modified (GM) wheat with introduced pm3b mildew resistance transgene, on two types of root-colonizing microorganisms, namely pseudomonads and arbuscular mycorrhizal fungi (AMF). Our investigations were carried out in field trials over three field seasons and at two locations. Serial dilution in selective King's B medium and microscopy were used to assess the abundance of cultivable pseudomonads and AMF, respectively. We developed a denaturing gradient gel electrophoresis (DGGE) method to characterize the diversity of the pqqC gene, which is involved in Pseudomonas phosphate solubilization. A major result was that in the first field season Pseudomonas abundances and diversity on roots of GM pm3b lines, but also on non-GM sister lines were different from those of the parental lines and conventional wheat cultivars. This indicates a strong effect of the procedures by which these plants were created, as GM and sister lines were generated via tissue cultures and propagated in the greenhouse. Moreover, Pseudomonas population sizes and DGGE profiles varied considerably between individual GM lines with different genomic locations of the pm3b transgene. At individual time points, differences in Pseudomonas and AMF accumulation between GM and control lines were detected, but they were not consistent and much less pronounced than differences detected between young and old plants, different conventional wheat cultivars or at different locations and field seasons. Thus, we conclude that impacts of GM wheat on plant-beneficial root-colonizing microorganisms are minor and not of ecological importance. The cultivation-independent pqqC-DGGE approach proved to be a useful tool for monitoring the dynamics of Pseudomonas populations in a wheat field and even sensitive enough for detecting population responses to altered plant physiology.

Les bénéfices et les risques environnementaux du blé de printemps génétiquement modifié (GM), plus résistant à l'oïdium, ont été étudiés sur deux sites en plein champ de 2008 à 2010, dans le cadre d'un réseau de groupes de recherche suisses. Les lignées de blé GM, possédant une résistance spécifique à l'oïdium et aux maladies fongiques en général, ont été comparées à des variétés de blé commercialisées, à des lignées témoins ainsi qu'à de l'orge et du triticale. Outre la résistance à l'oïdium, l'étude a également porté sur les répercussions sur les insectes et les organismes du sol (bactéries, champignons mycorhiziens, faune du sol) ainsi que sur les croisements fortuits du blé. Plusieurs lignées de blé GM étaient nettement plus résistantes à l'oïdium que les témoins. Aucun impact significatif sur les organismes non-cibles n'a été constaté, sur leur biodiversité ou sur certaines fonctions sélectionnées des écosystèmes. Dans l'ensemble, les différences entre le blé GM et les lignées témoins étaient moindres qu'entre les variétés de blé commerciales ou les autres espèces de céréales. Les cas de croisement fortuits étaient rares et n'ont été trouvés qu'à faible distance des plantes GM de l'essai.

Mungbean cultivar MGS Esmeralda was developed by Asian Vegetable Research and Development Center (Shanhua, Taiwan), as a result of crossing between the lines VC 1973A and VC 2768A. In ten trials conducted in the State of Minas Gerais, Brazil, it produced 13.5% more grains than 'Ouro Verde MG-2' (control cultivar), and its highest yield was 2,550 kg ha-1. The cultivar MGS Esmeralda is more susceptible to lodging, and its pods mature more uniformly than Ouro Verde MG-2 pods. One hundred-seed mass of 'MGS Esmeralda' ranged between 5.5 and 6.8 g. Both cultivars are susceptible to powdery mildew and cercospora leaf spot.

A powdery mildew disease was observed on leaves of Solanum gilo, S. melongena, S. tuberosum S. chacoense, Nicotiana rustica and N. tabacum in Brasília (Federal District), Brazil. Symptoms were mainly characterized by adaxial yellow areas in the leaves corresponding to white fungal colonies on the abaxial surface. Profuse sporulation was often observed. Light microscopy of the fungal colonies revealed the presence of conidiophores emerging through stomata with some having two or three branches. Ellipsoidal, subhyaline conidia were predominantly born singly and terminally on the conidiophore. All morphometrical characteristics agreed with those of Oidiopsis haplophylli (Syn. O. sicula). The teleomorph (Leveillula taurica) was not observed. Inoculation tests indicated that O. haplophylli isolates obtained from S. gilo, S. melongena, S. tuberosum, S. chacoense, Nicotiana rustica and N. tabacum were also pathogenic to sweet pepper (Capsicum annuum) and tomato (Lycopersicon esculentum). This is apparently the first report of these Solanaceae species as hosts of O. haplophylli in Brazil. This disease may become important in these crops, especially in greenhouses, and in hot and dry areas where drip irrigation is employed.

The main objective of this work was to identify sources of resistance in sorghum (Sorghum bicolor) to Peronosclerospora sorghi, the causal agent of downy mildew, through the evaluation of 42 sorghum genotypes under natural infection in the field. Genotypes were planted in single row plots between two rows of the susceptible line SC283, planted 30 days before, to act as spreader rows, in two separate nurseries. The experimental design was a completely randomized block design with three replications. Sorghum genotypes CMSXS156, CMSXS157, CMSXS243, TxARG-1, 8902, 9902054, 9910032, 9910296, Tx430, QL-3, SC170-6-17, CMSXS762 and BR304 were classified as highly resistant in both nurseries. Among these, SC170-6-17 and 9910296 showed 0% systemic infection. Results indicated the possible occurrence of different pathotypes of P. sorghi in the two nurseries.

Gerbera (Gerbera jamesonii) is one of the most popular ornamental plants in the world, used as cut or vase flower. The experiment aimed to evaluate the effect of milk in natura to control powdery mildew (Erysiphe cichoracearum) on gerbera. The treatments tested were: two concentrations of cow's milk in natura (8 and 16%), with and without adjuvant (vegetal oil), three fungicides (fenarimol -6 g a.i.100 L-1 H2O, chlorothalonil -150 g a.i.100 L-1 H2O and azoxystrobin + mineral oil - 10 g a.i.100 L-1 H2O) and control treatment. The area under the disease progress curve (AUDPC) was calculated for each plot based on disease severity. Milk in natura applied with adjuvant controlled the disease and did not allow the infection and development of pathogen. The addition of adjuvant improves the efficiency of milk in natura, but it requires subsequent washing of plants with H2O + detergent. The fungicide fenarimol promoted the best control and also the best gerbera visual aspect. The fungicide azoxystrobin + mineral oil and the chlorothalonil were phytotoxic to gerbera plants, affecting the aspect of leaves and flowers.

Experiments were carried out in a growth chamber with controlled temperature and photoperiod to test two populations of Blumeria graminis f. sp. hordei from Guarapuava, Paraná State, and Passo Fundo, Rio Grande do Sul State, Brazil. Treatments consisted in application of the fungicide triadimenol (Baytan 150 SC®) at three rates of its commercial formulation: 150, 250, 350 mL/100 Kg barley seeds. The experiments were conducted separately in a growth chamber for each population, adopting the same temperature and photoperiod. For inoculation, pots containing barley seedlings colonized by the fungus were placed among the plots. After emergence of the first symptoms, the disease severity was assessed at two-day intervals. The experiments were repeated twice for each fungus population. Data were expressed as area under the disease progress curve and as powdery mildew control by comparing the severity after the fungicide treatments to that of control. Data were subjected to analysis of variance and regression analysis; the area under the disease progress curve was also calculated. Comparing the data obtained in the present study with those reported in the literature and the control, the maximum value of 26.1% is considered insufficient to prevent the damages caused by the disease. The control response to the fungicide rate was significant. We can conclude that there was a reduction in the sensitivity of both B. graminis f.sp. hordei populations to the fungicide triadimenol, which explains the control failure observed in barley farms.

A model was devised to describe simultaneously the grain masses of water and dry matter against thermal time during grain filling and maturation of winter wheat. The model accounted for a linear increase in water mass of duration anthesis-m(1) (end of rapid water assimilation phase) and rate a, followed by a more stable water mass until in,, after which water mass declined rapidly at rate e. Grain dry matter was described as a linear increase of rate bgf until a maximum size (maxgf) was attained at m(2).The model was fitted to plot data from weekly samples of grains taken from replicated field experiments investigating effects of grain position (apical or medial), fungicide (five contrasting treatments), sowing date (early or late), cultivar (Malacca or Shamrock) and season (2001/2002 and 2002/2003) on grain filling. The model accounted for between 83 and 99% of the variation ( 2) when fitted to data from individual plots, and between 97 and 99% when fitted to treatment means. Endosperm cell number of grains from early-sown plots in the first season were also counted. Differences in maxgf between grain positions and also between cultivars were mostly the result of effects on bgf and were empirically associated with water mass at nil. Fungicide application controlled S. tritici and powdery mildew infection, delayed flag leaf senescence, increased water mass at m(1) (wm(1)), and also increased m(2), bgf and maxgf. Fungicide effects on water mass were detected before fungicide effects on dry matter, but comparison of the effects of individual fungicide treatments showed no evidence that effects on wm(1), nor on endosperm cell numbers at about m(1), were required for fungicide effects on maxgf, (c) 2005 Elsevier B.V. To further our understanding of powdery mildew biology during infection, we undertook a systematic shotgun proteomics analysis of the obligate biotroph Blumeria graminis f. sp. hordei at different stages of development in the host. Moreover we used a proteogenomics approach to feed information into the annotation of the newly sequenced genome. We analyzed and compared the proteomes from three stages of development representing different functions during the plant-dependent vegetative life cycle of this fungus. We identified 441 proteins in ungerminated spores, 775 proteins in epiphytic sporulating hyphae, and 47 proteins from haustoria inside barley leaf epidermal cells and used the data to aid annotation of the B. graminis f. sp. hordei genome. We also compared the differences in the protein complement of these key stages. Although confirming some of the previously reported findings and models derived from the analysis of transcriptome dynamics, our results also suggest that the intracellular haustoria are subject to stress possibly as a result of the plant defense strategy, including the production of reactive oxygen species. In addition, a number of small haustorial proteins with a predicted N-terminal signal peptide for secretion were identified in infected tissues: these represent candidate effector proteins that may play a role in controlling host metabolism and immunity.
