A implementação convencional do método de migração por diferenças finitas 3D, usa a técnica de splitting inline e crossline para melhorar a eficiência computacional deste algoritmo. Esta abordagem torna o algoritmo eficiente computacionalmente, porém cria anisotropia numérica. Esta anisotropia numérica por sua vez, pode levar a falsos posicionamentos de refletores inclinados, especialmente refletores com grandes ângulos de mergulho. Neste trabalho, como objetivo de evitar o surgimento da anisotropia numérica, implementamos o operador de extrapolação do campo de onda para baixo sem usar a técnica splitting inline e crossline no domínio frequência-espaço via método de diferenças finitas implícito, usando a aproximação de Padé complexa. Comparamos a performance do algoritmo iterativo Bi-gradiente conjugado estabilizado (Bi-CGSTAB) com o multifrontal massively parallel solver (MUMPS) para resolver o sistema linear oriundo do método de migração por diferenças finitas. Verifica-se que usando a expansão de Padé complexa ao invés da expansão de Padé real, o algoritmo iterativo Bi-CGSTAB fica mais eficientes computacionalmente, ou seja, a expansão de Padé complexa atua como um precondicionador para este algoritmo iterativo. Como consequência, o algoritmo iterativo Bi-CGSTAB é bem mais eficiente computacionalmente que o MUMPS para resolver o sistema linear quando usado apenas um termo da expansão de Padé complexa. Para aproximações de grandes ângulos, métodos diretos são necessários. Para validar e avaliar as propriedades desses algoritmos de migração, usamos o modelo de sal SEG/EAGE para calcular a sua resposta ao impulso.

Implementações dos métodos de migração diferença finita e Fourier (FFD) usam fatoração direcional para acelerar a performance e economizar custo computacional. Entretanto essa técnica introduz anisotropia numérica que podem erroneamente posicionar os refletores em mergulho ao longo das direções em que o não foi aplicado a fatoração no operador de migração. Implementamos a migração FFD 3D, sem usar a técnica do fatoração direcional, no domínio da frequência usando aproximação de Padé complexa. Essa aproximação elimina a anisotropia numérica ao preço de maior custo computacional buscando a solução do campo de onda para um sistema linear de banda larga. Experimentos numéricos, tanto no modelo homogêneo e heterogêneo, mostram que a técnica da fatoração direcional produz notáveis erros de posicionamento dos refletores em meios com forte variação lateral de velocidade. Comparamos a performance de resolução do algoritmo de FFD usando o método iterativo gradiente biconjugado estabilizado (BICGSTAB) e o multifrontal massively parallel direct solver (MUMPS). Mostrando que a aproximação de Padé complexa é um eficiente precondicionador para o BICGSTAB, reduzindo o número de iterações em relação a aproximação de Padé real. O método iterativo BICGSTAB é mais eficiente que o método direto MUMPS, quando usamos apenas um termo da expansão de Padé complexa. Para maior ângulo de abertura do operador, mais termos da série são requeridos no operador de migração, e neste caso, a performance do método direto é mais eficiente. A validação do algoritmo e as propriedades da evolução computacional foram avaliadas para a resposta ao impulso do modelo de sal SEG/EAGE.

O trabalho visa estudar a ocorrência e os aspectos epidemiológicos do parasitismo intestinal na aldeia Paranatinga da tribo indígena Parakanã, Amazônia Oriental Brasileira. Foram realizados dois inquéritos coproparasitológicos, em abril de 1992 e em fevereiro de 1995. Os métodos utilizados na identificação dos agentes parasitários foram os de Hoffman e exame direto, dois métodos simples, facilmente exeqüíveis em aldeias indígenas. Da amostra de 126 índios em abril de 1992 (população de 215 índios), 101 (80,2%) encontravam-se parasitados por pelo menos um enteroparasita. Ancilostomídeos foram encontrados em 33,3%, A. lumbricoides, em 42,8%, T. trichiura, em 0,8%, e S. stercoralis, em 5,6%. Em relação aos protozoários, E. histolytica foi encontrada em 65,0% e G. lamblia, em 46,8%. No inquérito de fevereiro de 1995, apesar do aumento da prevalência total em comparação com o de abril de 1992 (p = 0,04), houve diminuição das prevalências de ancilostomídeos, E. histolytica, G. lamblia, e ausência de S. stercoralis (p<0,05). A despeito das ações de saúde e saneamento empreendidas nas aldeias Parakanã, a prevalência de enteroparasitas ainda se encontra elevada na aldeia Paranatinga, sugerindo que as medidas de atenção primária e secundária devem ser imediatamente incrementadas.

The dental professionals are exposed to many risks of development of occupational diseases and the musculoskeletal disorders in the upper limbs are the most common disease. This problem may result in lower productivity and / or early retirement. Therefore, early diagnosis should be done through a reliable and valid system of risk assessment. Because of that, this study set out to conduct a review of the literature on methods of risk assessment of musculoskeletal disorders in the upper limbs in dental professionals. Conclusion: It was concluded that the available methods for assessing the risk of musculoskeletal disorders in dentistry are self-reports, observational and direct. Among these methods, the self-reports were frequently used to detect the risk of musculoskeletal disorders among dentists, dental students and professionals from the dental team. Further studies should be performed in dentistry using observational and direct methods.

The contamination of the physical environment occurs as a result of the lack of planning and inadequate management of hazardous products. Due to the difficulty of the characterization and study of soil and groundwater, the contamination has been long neglected. The characterization of these areas by research techniques through direct methods such as physical and chemical analysis, are widely used in the diagnosis and monitoring. However, despite presenting quantitative results, this method is not representative in spatial terms. The purpose of this work is to bring geophysical complementation with the diagnosis, bringing the spatial element for the detection of the contamination plume. In the study area, there was a railway accident in 1999 in which three wagons fell and spilled 240,000 L of diesel onto the soil. The main objective is to assess the presence of contaminants in the residual phase and dissolved in the subsurface, using the technique of Electrical Tomography (ETR), understand the effects of natural attenuation process in the degradation of hydrocarbons and changes in electrical properties in soil and groundwater. The results allowed the definition of three distinct patterns of resistivity region with presence of dissolved phase, set in a historical chemical analyzes: low values (> 60Ω.m) associated with the presence of hydrocarbons in an advanced stage of degradation, intermediate values (260Ω.me 511Ω .m) indicative of coexistence of residual hydrocarbons and dissolved phase, and high values (> 1000Ω.m), which reveal the prevalence of residual phase with incipient degradation, possibly in high toxicity concentrations for effective action of biodegradation processes. The diagnostic of areas with potential accumulation of residual phase can subsidize the planning of remediation techniques and promote the complete decontamination of the area

This thesis is concerned with in-situ time-, temperature- and pressure-resolved synchrotron X-ray powder diffraction investigations of a variety of inorganic compounds with twodimensional layer structures and three-dimensional framework structures. In particular, phase stability, reaction kinetics, thermal expansion and compressibility at non-ambient conditions has been studied for 1) Phosphates with composition MIV(HPO4)2·nH2O (MIV = Ti, Zr); 2) Pyrophosphates and pyrovanadates with composition MIVX2O7 (MIV = Ti, Zr and X = P, V); 3) Molybdates with composition ZrMo2O8. The results are compiled in seven published papers and two manuscripts. Reaction kinetics for the hydrothermal synthesis of α-Ti(HPO4)2·H2O and intercalation of alkane diamines in α-Zr(HPO4)2·H2O was studied using time-resolved experiments. In the high-temperature transformation of γ-Ti(PO4)(H2PO4)·2H2O to TiP2O7 three intermediate phases, γ'-Ti(PO4)(H2PO4)·(2-x)H2O, β-Ti(PO4)(H2PO4) and Ti(PO4)(H2P2O7)0.5 were found to crystallise at 323, 373 and 748 K, respectively. A new tetragonal three-dimensional phosphate phase called τ-Zr(HPO4)2 was prepared, and subsequently its structure was determined and refined using the Rietveld method. In the high-temperature transformation from τ-Zr(HPO4)2 to cubic α-ZrP2O7 two new orthorhombic intermediate phases were found. The first intermediate phase, ρ-Zr(HPO4)2, forms at 598 K, and the second phase, β-ZrP2O7, at 688 K. Their respective structures were solved using direct methods and refined using the Rietveld method. In-situ high-pressure studies of τ-Zr(HPO4)2 revealed two new phases, tetragonal ν-Zr(HPO4)2 and orthorhombic ω-Zr(HPO4)2 that crystallise at 1.1 and 8.2 GPa. The structure of ν-Zr(HPO4)2 was solved and refined using the Rietveld method. The high-pressure properties of the pyrophosphates ZrP2O7 and TiP2O7, and the pyrovanadate ZrV2O7 were studied up to 40 GPa. Both pyrophosphates display smooth compression up to the highest pressures, while ZrV2O7 has a phase transformation at 1.38 GPa from cubic to pseudo-tetragonal β-ZrV2O7 and becomes X-ray amorphous at pressures above 4 GPa. In-situ high-pressure studies of trigonal α-ZrMo2O8 revealed the existence of two new phases, monoclinic δ-ZrMo2O8 and triclinic ε-ZrMo2O8 that crystallises at 1.1 and 2.5 GPa, respectively. The structure of δ-ZrMo2O8 was solved by direct methods and refined using the Rietveld method.

L’acquifero freatico costiero ravennate è intensamente salinizzato fino a diversi km nell’entroterra. Il corpo dell’acquifero è formato da sabbie che poggiano su un substrato argilloso ad una profondità media di 25 m, i depositi affioranti sono sabbie e argille. Il lavoro svolto consiste in una caratterizzazione dello stato di salinizzazione con metodologie indirette (geoelettrica) e metodologie dirette (letture dei parametri fisici delle acque in pozzo). I sondaggi elettrici verticali (V.E.S.) mostrano stagionalità dovuta alle differenti quantità di pioggia e quindi di ricarica, le aree con depositi superficiali ad alta conducibilità idraulica (sabbie) hanno una lente d’acqua dolce compresa tra 0,1 e 2,25 m di spessore, al di sotto della quale troviamo una zona di mescolamento con spessori che vanno da 1,00 a 12,00 m, mentre quando in superficie abbiamo depositi a bassa conducibilità idraulica (limi sabbiosi e argille sabbiose) la lente d’acqua dolce scompare e la zona di mescolamento è sottile. Le misure dirette in pozzo mostrano una profondità della tavola d’acqua quasi ovunque sotto il livello del mare in entrambi i mesi monitorati, Giugno e Dicembre 2010, presentando una profondità leggermente maggiore nel mese di Dicembre. Dalla ricostruzione litologica risulta un acquifero composto da 4×109 m3 di sabbia, per cui ipotizzando una porosità media del 30% sono presenti 1,2×109 m3 di acqua. Dalla modellazione numerica (Modflow-SEAWAT 2000) risulta che l’origine dell’acqua salata che si trova in falda trova più facilmente spiegazione ipotizzando la sua presenza fin dalla formazione dell’acquifero, residuo delle acque marine che regredivano. Un’altra problematica analizzata è valutare l’applicazione della metodologia a minifiltri in uno studio sulla salinizzazione delle acque di falda. É stata implementata la costruzione di un transetto sperimentale, che ha permesso la mappatura dell’interfaccia acqua dolce/salmastra/salata con una precisione finora non raggiungibile.

Many biological processes depend on the sequential assembly of protein complexes. However, studying the kinetics of such processes by direct methods is often not feasible. As an important class of such protein complexes, pore-forming toxins start their journey as soluble monomeric proteins, and oligomerize into transmembrane complexes to eventually form pores in the target cell membrane. Here, we monitored pore formation kinetics for the well-characterized bacterial pore-forming toxin aerolysin in single cells in real time to determine the lag times leading to the formation of the first functional pores per cell. Probabilistic modeling of these lag times revealed that one slow and seven equally fast rate-limiting reactions best explain the overall pore formation kinetics. The model predicted that monomer activation is the rate-limiting step for the entire pore formation process. We hypothesized that this could be through release of a propeptide and indeed found that peptide removal abolished these steps. This study illustrates how stochasticity in the kinetics of a complex process can be exploited to identify rate-limiting mechanisms underlying multistep biomolecular assembly pathways.

The study of glacier fronts combines different geomatics measurement techniques as the classic survey using total station or theodolite, technical GNSS (Global Navigation Satellite System), using laser-scanner or using photogrammetry (air or ground). The measure by direct methods (classical surveying and GNSS) is useful and fast when accessibility to the glaciers fronts is easy, while it is practically impossible to realize, in the case of glacier fronts that end up in the sea (tide water glaciers). In this paper, a methodology that combines photogrammetric methods and other techniques for lifting the front of the glacier Johnsons, inaccessible is studied. The images obtained from the front, come from a non-metric digital camera; its georeferencing to a global coordinate system is performed by measuring points GNSS support in accessible areas of the glacier front side and applying methods of direct intersection in inaccessible points of the front, taking measurements with theodolite. The result of observations obtained were applied to study the temporal evolution (1957-2014) of the position of the Johnsons glacier front and the position of the Argentina, Las Palmas and Sally Rocks lobes front (Hurd glacier).

In this paper, we present a real-time tracking strategy based on direct methods for tracking tasks on-board UAVs, that is able to overcome problems posed by the challenging conditions of the task: e.g. constant vibrations, fast 3D changes, and limited capacity on-board. The vast majority of approaches make use of feature-based methods to track objects. Nonetheless, in this paper we show that although some of these feature-based solutions are faster, direct methods can be more robust under fast 3D motions (fast changes in position), some changes in appearance, constant vibrations (without requiring any specific hardware or software for video stabilization), and situations where part of the object to track is out the field of view of the camera. The performance of the proposed strategy is evaluated with images from real-flight tests using different evaluation mechanisms (e.g. accurate position estimation using a Vicon sytem). Results show that our tracking strategy performs better than well known feature-based algorithms and well known configurations of direct methods, and that the recovered data is robust enough for vision-in-the-loop tasks.

Autonomous aerial refueling is a key enabling technology for both manned and unmanned aircraft where extended flight duration or range are required. The results presented within this paper offer one potential vision-based sensing solution, together with a unique test environment. A hierarchical visual tracking algorithm based on direct methods is proposed and developed for the purposes of tracking a drogue during the capture stage of autonomous aerial refueling, and of estimating its 3D position. Intended to be applied in real time to a video stream from a single monocular camera mounted on the receiver aircraft, the algorithm is shown to be highly robust, and capable of tracking large, rapid drogue motions within the frame of reference. The proposed strategy has been tested using a complex robotic testbed and with actual flight hardware consisting of a full size probe and drogue. Results show that the vision tracking algorithm can detect and track the drogue at real-time frame rates of more than thirty frames per second, obtaining a robust position estimation even with strong motions and multiple occlusions of the drogue.

The performance of the proposed tracking strategy on-board UAVs is evaluated with images from realflight tests using manually-generated ground truth information, accurate position estimation using a Vicon system, and also with simulated data from a simulation environment. Results show that the hierarchical tracking strategy performs better than wellknown feature-based algorithms and well-known configurations of direct methods, and that its performance is robust enough for vision-in-the-loop tasks, e.g. for vision-based landing tasks.

Human hepatitis B virus genome encodes a protein, termed HBx, that is widely recognized as a transcriptional transactivator. While HBx does not directly bind cis-acting transcriptional control elements, it has been shown to associate with cellular proteins that bind DNA. Because HBx transactivated a large number of viral/cellular transcriptional control elements, we looked for its targets within the components of the basal transcriptional machinery. This search led to the identification of its interactions with TFIIH. Here, we show that HBx interacts with yeast and mammalian TFIIH complexes both in vitro and in vivo. These interactions between HBx and the components of TFIIH are supported by several lines of evidence including results from immunoprocedures and direct methods of measuring interactions. We have identified ERCC3 and ERCC2 DNA helicase subunits of holoenzyme TFIIH as targets of HBx interactions. Furthermore, the DNA helicase activity of purified TFIIH from rat liver and, individually, the ERCC2 component of TFIIH is stimulated in the presence of HBx. These observations suggest a role for HBx in transcription and DNA repair.
