[ 1] There has been a paucity of information on trends in daily climate and climate extremes, especially from developing countries. We report the results of the analysis of daily temperature ( maximum and minimum) and precipitation data from 14 south and west African countries over the period 1961 - 2000. Data were subject to quality control and processing into indices of climate extremes for release to the global community. Temperature extremes show patterns consistent with warming over most of the regions analyzed, with a large proportion of stations showing statistically significant trends for all temperature indices. Over 1961 to 2000, the regionally averaged occurrence of extreme cold ( fifth percentile) days and nights has decreased by - 3.7 and - 6.0 days/decade, respectively. Over the same period, the occurrence of extreme hot (95th percentile) days and nights has increased by 8.2 and 8.6 days/decade, respectively. The average duration of warm ( cold) has increased ( decreased) by 2.4 (0.5) days/decade and warm spells. Overall, it appears that the hot tails of the distributions of daily maximum temperature have changed more than the cold tails; for minimum temperatures, hot tails show greater changes in the NW of the region, while cold tails have changed more in the SE and east. The diurnal temperature range (DTR) does not exhibit a consistent trend across the region, with many neighboring stations showing opposite trends. However, the DTR shows consistent increases in a zone across Namibia, Botswana, Zambia, and Mozambique, coinciding with more rapid increases in maximum temperature than minimum temperature extremes. Most precipitation indices do not exhibit consistent or statistically significant trends across the region. Regionally averaged total precipitation has decreased but is not statistically significant. At the same time, there has been a statistically significant increase in regionally averaged daily rainfall intensity and dry spell duration. While the majority of stations also show increasing trends for these two indices, only a few of these are statistically significant. There are increasing trends in regionally averaged rainfall on extreme precipitation days and in maximum annual 5-day and 1-day rainfall, but only trends for the latter are statistically significant.

Observations at the Mauna Loa Observatory, Hawaii, established the systematic increase of anthropogenic CO2 in the atmosphere. For the same reasons that this site provides excellent globally averaged CO2 data, it may provide temperature data with global significance. Here, we examine hourly temperature records, averaged annually for 1977–2006, to determine linear trends as a function of time of day. For night-time data (22:00 to 06:00 LST (local standard time)) there is a near-uniform warming of 0.040 °C yr−1. During the day, the linear trend shows a slight cooling of −0.014 °C yr−1 at 12:00 LST (noon). Overall, at Mauna Loa Observatory, there is a mean warming trend of 0.021 °C yr−1. The dominance of night-time warming results in a relatively large annual decrease in the diurnal temperature range (DTR) of −0.050 °C yr−1 over the period 1977–2006. These trends are consistent with the observed increases in the concentrations of CO2 and its role as a greenhouse gas (demonstrated here by first-order radiative forcing calculations), and indicate the possible relevance of the Mauna Loa temperature measurements to global warming.

The urban heat island (UHI) phenomenon has been studied extensively, but there are relatively fewer reports on the so-called urban cool island (UCI) phenomenon. We reveal here that the UCI phenomenon exists in Hong Kong during the day, and is associated with the UHI at night under all wind and cloud conditions. The possible mechanisms for the UCI phenomenon in such a high-rise compact city have been discovered using a lumped urban air temperature model. A new concept of urban cool island degree hours (UCIdh) to measure the UCI intensity and duration is proposed. Our analyses reveal that when anthropogenic heat is small or absent, a high-rise and high-density city experiences a significant daytime UCI effect. This is explained by an intensified heat storage capacity and the reduced solar radiation gain of urban surfaces. However, if anthropogenic heat in the urban area increases further, the UCI phenomenon still exists, yet UCIdh decrease dramatically in a high-rise compact city. In a low-rise, low-density city, the UCI phenomenon also occurs when there is no anthropogenic heat, but easily disappears when there is little anthropogenic heat, and the UHI phenomenon dominates. This probably explains why the UHI phenomenon is often observed, but the UCI phenomenon is rarely observed. The co-existence of urban heat/cool island phenomena implies reduction of the daily temperature range (DTR) in such cities, and its dependence on urban morphology also implies that urban morphology can be used to control the urban thermal environment.

Analysis of observations indicates that there was a rapid increase in summer (June-August, JJA) mean surface air temperature (SAT) since the mid-1990s over Western Europe. Accompanying this rapid warming are significant increases in summer mean daily maximum temperature, daily minimum temperature, annual hottest day temperature and warmest night temperature, and an increase in frequency of summer days and tropical nights, while the change in the diurnal temperature range (DTR) is small. This study focuses on understanding causes of the rapid summer warming and associated temperature extreme changes. A set of experiments using the atmospheric component of the state-of-the-art HadGEM3 global climate model have been carried out to quantify relative roles of changes in sea surface temperature (SST)/sea ice extent (SIE), anthropogenic greenhouse gases (GHGs), and anthropogenic aerosols (AAer). Results indicate that the model forced by changes in all forcings reproduces many of the observed changes since the mid-1990s over Western Europe. Changes in SST/SIE explain 62.2% ± 13.0% of the area averaged seasonal mean warming signal over Western Europe, with the remaining 37.8% ± 13.6% of the warming explained by the direct impact of changes in GHGs and AAer. Results further indicate that the direct impact of the reduction of AAer precursor emissions over Europe, mainly through aerosol-radiation interaction with additional contributions from aerosol-cloud interaction and coupled atmosphere-land surface feedbacks, is a key factor for increases in annual hottest day temperature and in frequency of summer days. It explains 45.5% ± 17.6% and 40.9% ± 18.4% of area averaged signals for these temperature extremes. The direct impact of the reduction of AAer precursor emissions over Europe acts to increase DTR locally, but the change in DTR is countered by the direct impact of GHGs forcing. In the next few decades, greenhouse gas concentrations will continue to rise and AAer precursor emissions over Europe and North America will continue to decline. Our results suggest that the changes in summer seasonal mean SAT and temperature extremes over Western Europe since the mid-1990s are most likely to be sustained or amplified in the near term, unless other factors intervene.

Além de armazenar e distribuir o aço líquido para os moldes, uma das funções do distribuidor de lingotamento contínuo é a flotação e separação das inclusões para uma melhor qualidade do produto final. A eficiência e otimização deste processo requer o conhecimento das características do escoamento do aço líquido dentro do distribuidor. Esta dissertação trata do modelamento matemático do escoamento do aço num distribuidor DELTA-T, sem e com modificadores de fluxo e inibidores de turbulência, em condições isotérmicas e não isotérmicas em diferentes situações de vazão e condições térmicas. O software CFX-4 foi utilizado para resolver as equações de movimento, continuidade e transferência de calor utilizando modelo k-ε de turbulência para uma geometria tri-dimensional. A validação de modelo matemático com o modelo físico foi feita através da comparação de planos de laser entre os vetores de velocidade. Para a trajetória do escoamento foi utilizado no modelo físico a técnica do corante. Para a obtenção das curvas DTR – distribuição de tempos de residência no modelo físico foi usada a técnica de injeção de pulso, enquanto que no modelo numérico esta mesma técnica foi aplicada para um escoamento multifásico homogêneo. O perfil de temperatura calculado do aço na superfície foi comparado com medidas experimentais realizadas na usina O objetivo deste trabalho é caracterizar o escoamento do aço através dos perfis de velocidade, energia cinética de turbulência, perfis de temperatura e curvas de distribuição de tempos de residência, avaliando os tempos mínimo e médio de residência e as porções de volumes característicos. Os resultados obtidos indicam que o uso de modificadores de fluxo e inibidores de turbulência são eficientes no controle da emulsificação. É observado que o escoamento apresenta diferenças quando tratado isotermicamente e não isotermicamente.

Expanded Bed Adsorption (EBA) is an integrative process that combines concepts of chromatography and fluidization of solids. The many parameters involved and their synergistic effects complicate the optimization of the process. Fortunately, some mathematical tools have been developed in order to guide the investigation of the EBA system. In this work the application of experimental design, phenomenological modeling and artificial neural networks (ANN) in understanding chitosanases adsorption on ion exchange resin Streamline® DEAE have been investigated. The strain Paenibacillus ehimensis NRRL B-23118 was used for chitosanase production. EBA experiments were carried out using a column of 2.6 cm inner diameter with 30.0 cm in height that was coupled to a peristaltic pump. At the bottom of the column there was a distributor of glass beads having a height of 3.0 cm. Assays for residence time distribution (RTD) revelead a high degree of mixing, however, the Richardson-Zaki coefficients showed that the column was on the threshold of stability. Isotherm models fitted the adsorption equilibrium data in the presence of lyotropic salts. The results of experiment design indicated that the ionic strength and superficial velocity are important to the recovery and purity of chitosanases. The molecular mass of the two chitosanases were approximately 23 kDa and 52 kDa as estimated by SDS-PAGE. The phenomenological modeling was aimed to describe the operations in batch and column chromatography. The simulations were performed in Microsoft Visual Studio. The kinetic rate constant model set to kinetic curves efficiently under conditions of initial enzyme activity 0.232, 0.142 e 0.079 UA/mL. The simulated breakthrough curves showed some differences with experimental data, especially regarding the slope. Sensitivity tests of the model on the surface velocity, axial dispersion and initial concentration showed agreement with the literature. The neural network was constructed in MATLAB and Neural Network Toolbox. The cross-validation was used to improve the ability of generalization. The parameters of ANN were improved to obtain the settings 6-6 (enzyme activity) and 9-6 (total protein), as well as tansig transfer function and Levenberg-Marquardt training algorithm. The neural Carlos Eduardo de Araújo Padilha dezembro/2013 9 networks simulations, including all the steps of cycle, showed good agreement with experimental data, with a correlation coefficient of approximately 0.974. The effects of input variables on profiles of the stages of loading, washing and elution were consistent with the literature

Expanded Bed Adsorption plays an important role in the downstream processing mainly for reducing costs as well as steps besides could handling cells homogenates or fermentation broth. In this work Expanded Bed Adsorption was used to recover and purify whey proteins from coalho cheese manufacture using Streamline DEAE and Streamline SP both ionic resins as well as a hydrophobic resin Streamline Phenyl. A column of 2.6 cm inner diameter with 30 cm in height was coupled to a peristaltic pump. Hydrodynamics study was carried out with the three resins using Tris-HCl buffer in concentration of 30, 50 and 70 mM, with pH ranging from 7.0 to 8.0. In this case, assays of the expansion degree as well as Residence Time Distribution (RTD) were carried out. For the recovery and purification steps, a whey sample of 200 mL, was submitted to a column with 25mL of resin previously equilibrated with Tris/HCl (50 mM, pH 7.0) using a expanded bed. After washing, elution was carried out according the technique used. For ionic adsorption elution was carried out using 100 mL of Tris/HCl (50 mM, pH 7.0 in 1M NaCl). For Hydrophobyc interaction elution was carried out using Tris/HCl (50 mM, pH 7.0). Adsorption runs were carried out using the three resins as well as theirs combination. Results showed that for hydrodynamics studies a linear fit was observed for the three resins with a correlation coefficient (R2) about 0.9. In this case, Streamline Phenyl showed highest expansion degree reaching an expansion degree (H0/H) of 2.2. Bed porosity was of 0.7 when both resins Streamline DEAE and Streamline SP were used with StremLine Phenyl showing the highest bed porosity about 0.75. The number of theorical plates were 109, 41.5 and 17.8 and the axial dipersion coefficient (Daxial) were 0.5, 1.4 and 3.7 x 10-6 m2/s, for Streamline DEAE, Streamline SP and Streamline Phenyl, respectively. Whey proteins were adsorved fastly for the three resins with equilibrium reached in 10 minutes. Breakthrough curves showed that most of proteins stays in flowthrough as well as washing steps with 84, 77 and 96%, for Streamline DEAE, Streamline SP and Streamline Phenyl, respectively. It was observed protein peaks during elution for the three resins used. According to these peaks were identified 6 protein bands that could probably be albumin (69 KDa), lactoferrin (76 KDa), lactoperoxidase (89 KDa), β-lactoglobulin (18,3 KDa) e α-lactoalbumin (14 KDa), as well as the dimer of beta-lactoglobulin. The combined system compound for the elution of Streamline DEAE applied to the Streamline SP showed the best purification of whey proteins, mainly of the α-lactoalbumina

Die Lunge stellt einen Hauptort der CMV-Latenz dar. Die akute CMV-Infektion wird durch infiltrierende antivirale CD8 T-Zellen terminiert. Das virale Genom verbleibt jedoch im Lungengewebe in einem nicht replikativen Zustand, der Latenz, erhalten. Es konnte bereits gezeigt werden, dass während der Latenz die Major Immediate Early- (MIE) Gene ie1- und ie2 sporadisch transkribiert werden. Bisher konnte diese beginnende Reaktivierung latenter CMV-Genome nur in einer Momentaufnahme gezeigt werden (Kurz et al., 1999; Grzimek et al., 2001; Simon et al., 2005; zur Übersicht: Reddehase et al., 2008). Die sporadische Expression der MIE-Gene führt jedoch zur Präsentation eines antigenen IE1-Peptids und somit zur Stimulation antiviraler IE1-Peptid-spezifischer CD8 T-Zellen, die durch ihre Effektorfunktion die beginnende Reaktivierung wieder beenden. Dies führte uns zu der Hypothese, dass MIE-Genexpression über einen Zeitraum betrachtet (period prevalence) häufiger stattfindet als es in einer Momentaufnahme (point prevalence) beobachtet werden kann.rnrnUm die Häufigkeit der MIE-Genexpression in der Dynamik in einem definierten Zeitraum zu erfassen, sollte eine Methode entwickelt werden, welche es erstmals ermöglicht, selektiv und konditional transkriptionell aktive Zellen sowohl während der akuten Infektion als auch während der Latenz auszulöschen. Dazu wurde mit Hilfe der Zwei-Schritt BAC-Mutagenese ein rekombinantes death-tagged Virus hergestellt, welches das Gen für den Diphtherie Toxin Rezeptor (DTR) unter Kontrolle des ie2-Promotors (P2) enthält. Ist der P2 transkriptionell aktiv, wird der DTR an der Zelloberfläche präsentiert und die Zelle wird suszeptibel für den Liganden Diphtherie Toxin (DT). Durch Gabe von DT werden somit alle Zellen ausgelöscht, in denen virale Genome transkriptionell aktiv sind. Mit zunehmender Dauer der DT-Behandlung sollte also die Menge an latenten viralen Genomen abnehmen.rnrnIn Western Blot-Analysen konnte das DTR-Protein bereits 2h nach der Infektion nachgewiesen werden. Die Präsentation des DTR an der Zelloberfläche wurde indirekt durch dessen Funktionalität bewiesen. Das rekombinante Virus konnte in Fibroblasten in Gegenwart von DT nicht mehr replizieren. In akut infizierten Tieren konnte die virale DNA-Menge durch eine einmalige intravenöse (i.v.) DT-Gabe signifikant reduziert werden. Verstärkt wurde dieser Effekt durch eine repetitive i.v. DT-Gabe. Auch während der Latenz gelang es, die Zahl der latenten viralen Genome durch repetitive i.v. und anschließende intraperitoneale (i.p.) DT-Gabe zu reduzieren, wobei wir abhängig von der Dauer der DT-Gabe eine Reduktion um 60\% erreichen konnten. Korrespondierend zu der Reduktion der DNA-Menge sank auch die Reaktivierungshäufigkeit des rekombinanten Virus in Lungenexplantatkulturen. rnrnrnUm die Reaktivierungshäufigkeit während der Latenz berechnen zu können, wurde durch eine Grenzverdünnungsanalyse die Anzahl an latenten viralen Genomen pro Zelle bestimmt. Dabei ergab sich eine Kopienzahl von 9 (6 bis 13). Ausgehend von diesen Ergebnissen lässt sich berechnen, dass, bezogen auf die gesamte Lunge, in dem getesteten Zeitraum von 184h durch die DT-Behandlung 1.000 bis 2.500 Genome pro Stunde ausgelöscht wurden. Dies entspricht einer Auslöschung von 110 bis 280 MIE-Gen-exprimierenden Lungenzellen pro Stunde. Damit konnte in dieser Arbeit erstmals die Latenz-assoziierte Genexpression in ihrer Dynamik dargestellt werden.rn

Die primäre, produktive Cytomegalovirus (CMV)-Infektion wird im immunkompetenten Patienten effizient durch antivirale CD8+ T-Zellen kontrolliert. Das virale Genom besitzt jedoch die Fähigkeit, in einem nicht replikativen, Latenz genannten Zustand, in gewissen Zelltypen zu persistieren, ohne dass infektiöse Nachkommenviren produziert werden. Die molekularen Mechanismen, welche der Etablierung und Aufrechterhaltung der Latenz zugrundeliegen, sind noch weitestgehend unbekannt. Es gibt Hinweise darauf, dass zelluläre Verteidigungsmechanismen die Zirkularisierung und Chromatinisierung viraler Genome hervorrufen und dadurch die virale Genexpression größtenteils verhindert wird (Marks & Spector, 1984; Reeves et al., 2006).rnAllerdings liegen die Genome nicht in einem komplett inaktiven Zustand vor. Vielmehr konnte für das murine CMV (mCMV) bereits die sporadische Transkription der Gene ie1 und ie2 während der Latenz nachgewiesen werden (Kurz et al., 1999; Grzimek et al., 2001).rnIn der vorliegenden Arbeit wurde zum ersten Mal eine umfassende in vivo Latenz-Analyse zur Charakterisierung der viralen Transkription in einer Kinetik anhand der alle drei kinetischen Klassen repräsentierenden Transkripte IE1, IE3, E1, m164, M105 und M86 vorgenommen.rnNach Latenz-Etablierung, verifiziert durch Abwesenheit von infektiösem Virus, konnten alle getesteten Transkripte in der Lunge quantifiziert werden. Interessanterweise war die transkriptionelle Aktivität zu keinem Analyse-Zeitpunkt mit der klassischen IE-E-L-Kinetik der produktiven Infektion kompatibel. Stattdessen lag eine stochastische Transkript-Expression vor, deren Aktivität mit voranschreitender Zeit immer weiter abnahm.rnWährend der Latenz exprimierte Transkripte, die für antigene Peptide kodieren, können infizierte Zellen für das Immunsystem sichtbar machen, was zu einer fortwährenden Restimulation des memory T-Zell-pools führen würde. Durch zeitgleiche Analyse der Transkript-Expression, sowie der Frequenzen Epitop-spezifischer CD8+ T-Zellen während der Latenz (IE1, m164, M105), wurde eine möglicher Zusammenhang zwischen der transkriptionellen Aktivität und der Expansion des memory T-Zell-pools untersucht. Die weitere Charakterisierung von Subpopulationen der Epitop-spezifischen CD8+ T-Zellen identifizierte die SLECs (short-lived-effector cells; CD127low CD62Llow KLRG1high) als die dominante Population in Lunge und Milz während der mCMV-Latenz.rnIn einem weiteren Teil der Arbeit sollte untersucht werden, ob IE-Genexpression zur Etablierung von Latenz notwendig ist. Mit Hilfe der Rekombinanten mCMV-Δie2-DTR, die die Gensequenz des Diphtherietoxin-Rezeptors (DTR) anstelle des Gens ie2 trägt, konnten infizierte, DTR exprimierende Zellen durch eine DT-Applikation konditional depletiert werden.rnIm latent infizierbaren Zelltyp der Leber, den LSECs (liver sinusoidal endothelial cells) wurde die virale Load durch 90-stündige DT–Applikation nach mCMV-Δie2-DTR Infektion auf das Level latent infizierter LSECs reduziert. Diese Daten sprechen für die Hypothese eines von Beginn an inaktiven Genoms, das keine IE-Genexpression zur Latenz-Etablierung benötigt. Zusätzlich stellt dieser Ansatz ein neues Tier-Modell zur Latenz-Etablierung dar. Verringerte Wartezeiten bis zur vollständigen Latenz-Etablierung, im Vergleich zum bisherigen Knochenmarktransplantations-Modell, könnten anfallende Tierhaltungskosten erheblich reduzieren und das Voranschreiten der Forschung beschleunigen.

We analyze the impact of stratospheric volcanic aerosols on the diurnal temperature range (DTR) over Europe using long-term subdaily station records. We compare the results with a 28-member ensemble of European Centre/Hamburg version 5.4 (ECHAM5.4) general circulation model simulations. Eight stratospheric volcanic eruptions during the instrumental period are investigated. Seasonal all- and clear-sky DTR anomalies are compared with contemporary (approximately 20 year) reference periods. Clear sky is used to eliminate cloud effects and better estimate the signal from the direct radiative forcing of the volcanic aerosols. We do not find a consistent effect of stratospheric aerosols on all-sky DTR. For clear skies, we find average DTR anomalies of −0.08°C (−0.13°C) in the observations (in the model), with the largest effect in the second winter after the eruption. Although the clear-sky DTR anomalies from different stations, volcanic eruptions, and seasons show heterogeneous signals in terms of order of magnitude and sign, the significantly negative DTR anomalies (e.g., after the Tambora eruption) are qualitatively consistent with other studies. Referencing with clear-sky DTR anomalies to the radiative forcing from stratospheric volcanic eruptions, we find the resulting sensitivity to be of the same order of magnitude as previously published estimates for tropospheric aerosols during the so-called “global dimming” period (i.e., 1950s to 1980s). Analyzing cloud cover changes after volcanic eruptions reveals an increase in clear-sky days in both data sets. Quantifying the impact of stratospheric volcanic eruptions on clear-sky DTR over Europe provides valuable information for the study of the radiative effect of stratospheric aerosols and for geo-engineering purposes.

BACKGROUND Exposure to food allergens through a disrupted skin barrier has been recognized as a potential factor in the increasing prevalence of food allergy. OBJECTIVE We sought to test the immunologic mechanisms by which epicutaneous sensitization to food allergens predisposes to intestinal food allergy. METHODS Mice were epicutaneously sensitized with ovalbumin or peanut on an atopic dermatitis-like skin lesion, followed by intragastric antigen challenge. Antigen-specific serum IgE levels and T(H)2 cytokine responses were measured by ELISA. Expression of type 2 cytokines and mast cell proteases in the intestine were measured by using real-time PCR. Accumulation of basophils in the skin and mast cells in the intestine was examined by using flow cytometry. In vivo basophil depletion was achieved by using diphtheria toxin treatment of Baso-DTR mice. For cell-transfer studies, the basophil population was expanded in vivo by means of hydrodynamic tail vein injection of thymic stromal lymphopoietin (TSLP) cDNA plasmid. RESULTS Sensitization to food allergens through an atopic dermatitis-like skin lesion is associated with an expansion of TSLP-elicited basophils in the skin that promote antigen-specific T(H)2 cytokine responses, increased antigen-specific serum IgE levels, and accumulation of mast cells in the intestine, promoting the development of intestinal food allergy. Critically, disruption of TSLP responses or depletion of basophils reduced the susceptibility to intestinal food allergy, whereas transfer of TSLP-elicited basophils into intact skin promoted disease. CONCLUSION Epicutaneous sensitization on a disrupted skin barrier is associated with accumulation of TSLP-elicited basophils, which are necessary and sufficient to promote antigen-induced intestinal food allergy.

Three long-term temperature data series measured in Portugal were studied to detect and correct non-climatic homogeneity breaks and are now available for future studies of climate variability. Series of monthly minimum (Tmin) and maximum (Tmax) temperatures measured in the three Portuguese meteorological stations of Lisbon (from 1856 to 2008), Coimbra (from 1865 to 2005) and Porto (from 1888 to 2001) were studied to detect and correct non-climatic homogeneity breaks. These series together with monthly series of average temperature (Taver) and temperature range (DTR) derived from them were tested in order to detect homogeneity breaks, using, firstly, metadata, secondly, a visual analysis and, thirdly, four widely used homogeneity tests: von Neumann ratio test, Buishand test, standard normal homogeneity test and Pettitt test. The homogeneity tests were used in absolute (using temperature series themselves) and relative (using sea-surface temperature anomalies series obtained from HadISST2 close to the Portuguese coast or already corrected temperature series as reference series) modes. We considered the Tmin, Tmax and DTR series as most informative for the detection of homogeneity breaks due to the fact that Tmin and Tmax could respond differently to changes in position of a thermometer or other changes in the instrument's environment; Taver series have been used, mainly, as control. The homogeneity tests show strong inhomogeneity of the original data series, which could have both internal climatic and non-climatic origins. Homogeneity breaks which have been identified by the last three mentioned homogeneity tests were compared with available metadata containing data, such as instrument changes, changes in station location and environment, observing procedures, etc. Significant homogeneity breaks (significance 95% or more) that coincide with known dates of instrumental changes have been corrected using standard procedures. It was also noted that some significant homogeneity breaks, which could not be connected to the known dates of any changes in the park of instruments or stations location and environment, could be caused by large volcanic eruptions. The corrected series were again tested for homogeneity: the corrected series were considered free of non-climatic breaks when the tests of most of monthly series showed no significant (significance 95% or more) homogeneity breaks that coincide with dates of known instrument changes. Corrected series are now available in the frame of ERA-CLIM FP7 project for future studies of climate variability.

Physiognomic traits of plant leaves such as size, shape or margin are decisively affected by the prevailing environmental conditions of the plant habitat. On the other hand, if a relationship between environment and leaf physiognomy can be shown to exist, vegetation represents a proxy for environmental conditions. This study investigates the relationship between physiognomic traits of leaves from European hardwood vegetation and environmental parameters in order to create a calibration dataset based on high resolution grid cell data. The leaf data are obtained from synthetic chorologic floras, the environmental data comprise climatic and ecologic data. The high resolution of the data allows for a detailed analysis of the spatial dependencies between the investigated parameters. The comparison of environmental parameters and leaf physiognomic characters reveals a clear correlation between temperature related parameters (e.g. mean annual temperature or ground frost frequency) and the expression of leaf characters (e.g. the type of leaf margin or the base of the lamina). Precipitation related parameters (e.g. mean annual precipitation), however, show no correlation with the leaf physiognomic composition of the vegetation. On the basis of these results, transfer functions for several environmental parameters are calculated from the leaf physiognomic composition of the extant vegetation. In a next step, a cluster analysis is applied to the dataset in order to identify "leaf physiognomic communities". Several of these are distinguished, characterised and subsequently used for vegetation classification. Concerning the leaf physiognomic diversity there are precise differences between each of these "leaf physiognomic classes". There is a clear increase of leaf physiognomic diversity with increasing variability of the environmental parameters: Northern vegetation types are characterised by a more or less homogeneous leaf physiognomic composition whereas southern vegetation types like the Mediterranean vegetation show a considerable higher leaf physiognomic diversity. Finally, the transfer functions are used to estimate palaeo-environmental parameters of three fossil European leaf assemblages from Late Oligocene and Middle Miocene. The results are compared with results obtained from other palaeo-environmental reconstructing methods. The estimates based on a direct linear ordination seem to be the most realistic ones, as they are highly consistent with the Coexistence Approach.
