The porcine reproductive and respiratory syndrome virus (PRRSV) is an economically important pathogen of swine and is known to cause abortion and infertility in pregnant sows and respiratory distress in piglets. PRRSV contains a major glycoprotein (GP5) and three minor glycoproteins (GP2a, GP3, and GP4) on the virion envelope, all of which are required for infectious virus production. To study their interactions amongst each other and with a cellular receptor for PRRSV, CD163, I cloned each of the viral glycoproteins and CD163 in various expression vectors. My studies have shown that while the GP2a, GP3, and GP4 are co-translationally glycosylated, the GP5 is post-translationally glycosylated. By using co-immunoprecipitation (co-IP) assays, strong interaction was demonstrated between GP4 and GP5 proteins, although weak interactions among the other envelope glycoproteins were also detected. Further, GP4 was found to mediate interactions leading to formation of multiprotein glycoprotein complex. My results also show that GP2a and GP4 proteins are the only two GPs that specifically interact with the CD163 molecule and that glycosylation of these GPs is required for efficient interaction. Based on these studies, I have developed an interactome map of the viral GPs and CD163 and have proposed a model of the viral glycoprotein complex and its interaction with CD163. Studies reported here also show that glycan addition at residue 184 (N184) of GP2a, and residues N42, N50, and N131 of GP3 is essential for recovery of infectious virus. Although single site glycosylation mutants of GP4 had no effect on infectious virus production, introduction of double mutations was lethal. The loss of glycan moieties of GP2a, GP3, and GP4 proteins had no effect on host neutralizing antibody production. Overall, I conclude that the PRRSV glycoproteins are co-translationally and post-translationally glycosylated, the GP4 protein is central to mediating interglycoprotein interactions, and along with GP2a, serves as the viral attachment protein that is responsible for interactions with the viral receptor, CD163. Further, glycosylation of GP2a, GP3, and GP4 proteins is required for infectious virus production, efficient interaction with CD163, but does not play any role in neutralizing antibody response in infected animals.

Die supramolekularen Organisationen von 5-n-Alkoxyisophthaläuren und 2,5-Di-n-alkoxyterephthalsäuren bilden durch sekundäre Wechselwirkungen in Form van der Waals-Kräften und Wasserstoffbrückenbindungen hochgeordnete lamellare Strukturen, die sowohl mittels Röntgen-kristallgraphie als auch durch Rastertunnelmikroskopie (STM) visualisierbar sind. Diese Art der Aggregationsmuster ermöglicht den potentiell reaktiven Gruppen (Diacetylene, Zimtsäuren) in der Alkoxykette, einen optimalen Abstand und geometrische Orientierungen im Kristall einzunehmen, um lichtinduzierte Reaktionen im Festkörper durchzuführen. Einführung einer Amidgruppe an die Alkoxykette erweitert dieses Konzept, durch Erhalt einer zusätzlichen Wasserstoffbrückenbindung im hydrophoben Bereich der Organisation.Die Stabilität solche supramolekularer Aggregate kann durch Einführung semifluorierter Alkoxyketten stark beeinflußt werden, da die Inkompatibilität der pefluorierten Gruppen durch viele organische Verbindungen das Gleichgewicht nicht-kovalenter Wechselwirkung drastisch verändert. Diese Eigenschaft der semifluorierten 5-n-Alkoxyisopthalsäuren und 2,5-Di-n-alkoxyterephthalsäuren zeigt sich erstaunlicherweise jedoch nur bei einem großen Anteil des perfluorierten Alkylteils als der des nicht-perfluorierten. Da sich dann die perfluorierten Alkylteile untereinander organisieren können, erscheint die Änderung der Assoziationsstruktur von interdigitiert-lamellar zu nicht-interdigitiert-lamellar plausibel.Das gezielte Design eines neuen Organisationsmusters der 5-n-Alkoxyisophthalsäuren gelang durch Brechung der Symmetrie dieses Bausteins. Die Einführung zweier Alkoxyketten ließ die zweidimensionale Anordnung der wasserstoffbrückengebundenen Aromate unverändert, resultierte aber mit Ausbruch einer Alkoxykette aus dieser Ebene.

Ziel der hier vorliegenden Dissertation ist es, Übergangsmetallpivalate durch gezielte Substitution monodentater Donorliganden in apikalen Positionen, unter Erhalt ihrer Grundstruktur, zu höherdimensionalen Verbindungen zu verknüpfen. Als Ausgangs-verbindungen dienen dabei [Fe3O(O2C-tBu)6(OH2)3]O2C-tBu und [Ni2(OH2)(O2C-tBu)4(HO2C-tBu)4].rnrnIm ersten Teil dieser Arbeit konnten, in Abhängigkeit der in den Reaktionen eingesetzten Liganden mit [Fe3O(O2C-tBu)6(OH2)3]O2C-tBu, symmetrisch oder asymmetrisch substituierte dreikernige Verbindungen erhalten werden. Deren strukturellen und magnetischen Eigenschaften konnten untersucht werden und die daraus resultierenden magnetostrukturellen Korrelationen auf die folgenden vorgestellten mehrkernigen bzw. höherdimensionalen Verbindungen übertragen werden, die erheblich an Komplexität zugenommen haben.rnDie 0-dimensionalen dreikernigen Einheiten zeigen, abhängig von ihren Fe-O-Bindungslängen in den µ3-Oxo verbrückten Einheiten, unterschiedlich starke antiferro-magnetische Austauschwechselwirkungen. Wenn in den Verbindungen eine längere Fe-O-Bindung und zwei kürzere Fe-O-Bindungen existieren, können diese Typ 2:a zugeordnet werden. Daraus folgt, dass die Daten der magnetischen Suszeptibilität mit zwei unterschiedlich starken Austauschwechselwirkungen (J-Kopplungen) zu simulieren sind. Es liegen eine stärkere J-Kopplung über die kurzen Fe-O-Bindungen und zwei schwächere über die lange Fe-O-Bindung vor (J1 > J2). Existieren hingegen eine kürzere Fe-O-Bindung und zwei längere Fe-O-Bindungen (Typ 2:b) sind nun die magnetischen Suszeptibilitätsdaten nur mit zwei stärkeren und einer schwächeren Kopplung zu simulieren (J1 < J2). Die vorgestellten Verbindungen zeigen alle einen Spingrundzustand S≠0, der durch konkurrierende Wechselwirkungen der Spinzentren in Dreieckssituationen begründet ist. rnDer zweite Teil der Arbeit beschäftigte sich mit dem gezielten Aufbau mehrkerniger Verbindungen, in denen die dreikernige Einheit als Grundmotiv erhalten bleiben konnte. Die Austauschwechselwirkungen der fünf- und sechskernigen Verbindungen konnten in Abhängigkeit der Bindungslängen und basierend auf den Ergebnissen der dreikernigen Einheiten aus dem ersten Teil, bestimmt werden. rnDie Synthesen der 4-Hydroxybenzaldehyd verbrückten Kettenverbindung sowie des über 3,5,3’,5’-Tetramethyl-1H,1’H-[4,4’]bipyrazolyl verknüpften 3-dimensionalen Nickelnetzwerks zeigten die erfolgreiche Umsetzung des „Bottom Up“ Ansatzes. Durch Erhaltung des jeweiligen Grundmotivs der verwendeten Ausgangsverbindung konnten die magnetischen Austauschwechselwirkungen unter Einbeziehung schwacher Wechselwirkungen durch den Raum, mit Hilfe der Theta-Weiss Temperatur, in den Simulationen bestimmt werden.rnrnDamit stellt der „Bottom Up“ Ansatz eine hervorragende Syntesestrategie für den Aufbau höherdimensionaler Verbindungen, ausgehend von zwei- bzw. dreikernigen Übergangs-metallkomplexen, dar.rn

We used single molecule dynamic force spectroscopy to unfold individual serine/threonine antiporters SteT from Bacillus subtilis. The unfolding force patterns revealed interactions and energy barriers that stabilized structural segments of SteT. Substrate binding did not establish strong localized interactions but appeared to be facilitated by the formation of weak interactions with several structural segments. Upon substrate binding, all energy barriers of the antiporter changed thereby describing the transition from brittle mechanical properties of SteT in the unbound state to structurally flexible conformations in the substrate-bound state. The lifetime of the unbound state was much shorter than that of the substrate-bound state. This leads to the conclusion that the unbound state of SteT shows a reduced conformational flexibility to facilitate specific substrate binding and a reduced kinetic stability to enable rapid switching to the bound state. In contrast, the bound state of SteT showed an increased conformational flexibility and kinetic stability such as required to enable transport of substrate across the cell membrane. This result supports the working model of antiporters in which alternate substrate access from one to the other membrane surface occurs in the substrate-bound state.

An increasing number of neuroimaging studies are concerned with the identification of interactions or statistical dependencies between brain areas. Dependencies between the activities of different brain regions can be quantified with functional connectivity measures such as the cross-correlation coefficient. An important factor limiting the accuracy of such measures is the amount of empirical data available. For event-related protocols, the amount of data also affects the temporal resolution of the analysis. We use analytical expressions to calculate the amount of empirical data needed to establish whether a certain level of dependency is significant when the time series are autocorrelated, as is the case for biological signals. These analytical results are then contrasted with estimates from simulations based on real data recorded with magnetoencephalography during a resting-state paradigm and during the presentation of visual stimuli. Results indicate that, for broadband signals, 50–100 s of data is required to detect a true underlying cross-correlations coefficient of 0.05. This corresponds to a resolution of a few hundred milliseconds for typical event-related recordings. The required time window increases for narrow band signals as frequency decreases. For instance, approximately 3 times as much data is necessary for signals in the alpha band. Important implications can be derived for the design and interpretation of experiments to characterize weak interactions, which are potentially important for brain processing.

Efforts to increase the potency of transcriptional activators are generally unsuccessful because poor expression of activators in mammalian cells limits their delivery to target promoters. Here we report that the effectiveness of chimeric activators can be dramatically improved by expressing them as noncovalent tetrameric bundles. Bundled activation domains are much more effective at activating a reporter gene than simple monomeric activators, presumably because, at similar expression levels, up to 4 times as many the activation domains are delivered to the target promoter. These bundled activation domains are also more effective than proteins in which activation domains are tandemly reiterated in the same polypeptide chain, because such proteins are very poorly expressed and therefore not delivered effectively. These observations suggest that there is a threshold number of activation domains that must be bound to a promoter for activation, above which promoter activity is simply a function of the number of activators bound. We show that bundling can be exploited practically to enhance the sensitivity of mammalian two-hybrid assays, enabling detection of weak interactions or those between poorly expressed proteins. Bundling also dramatically improves the performance of a small-molecule-regulated gene expression system when the expression level of regulatory protein is limiting, a situation that may be encountered in gene therapy applications.

Recruitment of intracellular proteins to the plasma membrane is a commonly found requirement for the initiation of signal transduction events. The recently discovered pleckstrin homology (PH) domain, a structurally conserved element found in ∼100 signaling proteins, has been implicated in this function, because some PH domains have been described to be involved in plasma membrane association. Furthermore, several PH domains bind to the phosphoinositides phosphatidylinositol-(4,5)-bisphosphate and phosphatidylinositol-(3,4,5)-trisphosphate in vitro, however, mostly with low affinity. It is unclear how such weak interactions can be responsible for observed membrane binding in vivo as well as the resulting biological phenomena. Here, we investigate the structural and functional requirements for membrane association of cytohesin-1, a recently discovered regulatory protein of T cell adhesion. We demonstrate that both the PH domain and the adjacent carboxyl-terminal polybasic sequence of cytohesin-1 (c domain) are necessary for plasma membrane association and biological function, namely interference with Jurkat cell adhesion to intercellular adhesion molecule 1. Biosensor measurements revealed that phosphatidylinositol-(3,4,5)-trisphosphate binds to the PH domain and c domain together with high affinity (100 nM), whereas the isolated PH domain has a substantially lower affinity (2–3 μM). The cooperativity of both elements appears specific, because a chimeric protein, consisting of the c domain of cytohesin-1 and the PH domain of the β-adrenergic receptor kinase does not associate with membranes, nor does it inhibit adhesion. Moreover, replacement of the c domain of cytohesin-1 with a palmitoylation–isoprenylation motif partially restored the biological function, but the specific targeting to the plasma membrane was not retained. Thus we conclude that two elements of cytohesin-1, the PH domain and the c domain, are required and sufficient for membrane association. This appears to be a common mechanism for plasma membrane targeting of PH domains, because we observed a similar functional cooperativity of the PH domain of Bruton’s tyrosine kinase with the adjacent Bruton’s tyrosine kinase motif, a novel zinc-containing fold.

The crystal structure of the pheromone Er-1 from the unicellular eukaryotic organism Euplotes raikovi was determined at 1.6 A resolution and refined to a crystallographic R factor of 19.9%. In the tightly packed crystal, two extensive intermolecular helix-helix interactions arrange the Er-1 molecules into layers. Since the putative receptor of the pheromone is a membrane-bound protein, whose extracellular C-terminal domain is identical in amino acid sequence to the soluble pheromone, the interactions found in the crystal may mimic the pheromone-receptor interactions as they occur on a cell surface. Based on this, we propose a model for the interaction between soluble pheromone molecules and their receptors. In this model, strong pheromone-receptor binding emerges as a consequence of the cooperative utilization of several weak interactions. The model offers an explanation for the results of binding studies and may also explain the adhesion between cells that occurs during mating.

Compósitos de polímeros de polietileno linear de baixa densidade (LLDPE) possuem baixo desempenho mecânico devido principalmente à sua fraca interação, intermolecular, entre a cadeia polimérica e a carga. Uma maneira de minimizar esse baixo desempenho mecânico se faz com a mudança da estrutura química da poliolefina com a inserção de um grupo polar a sua cadeia, ou seja, faz-se a funcionalização das poliolefinas. O sistema de funcionalização adotado foi o processamento reativo, no qual foi utilizado para este sistema de processamento o misturador de dupla rosca acoplado a um reâmetro de torque. Neste trabalho, os grupos polares inseridos à cadeia dos polímeros de LLDPE\'s de copolímeros 1-buteno e 1-octeno (LLDPE-but e LLDPE-oct) foram o anidrido maléico (AM) e o anidrido tetrahidroftálico (ATF). Para a confecção dos compósitos foram utilizadas as cargas de microesferas de sílica modificada, no qual foi inserido compostos silanados em sua superfície (3-aminopropilsilano - APS - e trimetoxiclorosilano TMCISi) para estudo de interação com as poliolefinas funcionalizadas. Neste trabalho foram realizados ensaios de caracterização térmica, vibracional além de análises de torque do polímero fundido, análises do grau de reticulação e ensaios mecânicos de tração por elongação. Na caracterização térmica foram utilizadas as técnicas: termogravimetria (TG) e calorimetria exploratória diferencial (DSC). Na caracterização vibracional utilizou-se a espectroscopia fotoacústica no infravermelho (PAS-IR) e a espectroscopia de espalhamento Raman. Pela técnica PAS-IR foi possível comprovar a inserção dos anidridos à cadeia das poliolefinas assim como foi possível verificar a interação entre o polímero funcionalizado e a carga. Pelas técnicas térmicas de DSC e TG foi possível verificar mudanças das propriedades do compósito frente aos polímeros originais ou funcionalizados. Os ensaios mecânicos comprovaram que os compósitos de polímeros funcionalizados possuem maior elongação e tensão à ruptura comparada aos compósitos dos LLDPE\'s não funcionalizados

Data provided by 7380 middle managers from 60 nations are used to determine whether demographic variables are correlated with managers’ reliance on vertical sources of guidance in different nations and whether these correlations differ depending on national culture characteristics. Significant effects of Hofstede’s national culture scores, age, gender, organization ownership and department function are found. After these main effects have been discounted, significant although weak interactions are found, indicating that demographic effects are stronger in individualist, low power distance nations than elsewhere. Significant non-predicted interaction effects of uncertainty avoidance and masculinity-femininity are also obtained. The implications for theory and practice of the use of demographic attributes in understanding effective management procedures in various parts of the world are discussed.

We study the properties of radiation generated in ultralong fiber lasers and find an interesting link between these optical systems and the theory of weak wave turbulence. Experimental observations strongly suggest that turbulentlike weak interactions between the multitude of laser cavity modes are responsible for practical characteristics of ultralong fiber lasers such as spectra of the output radiation.

We analyse industry–academic links in the context of a dual economy (or disarticulated industrial structure) in Ireland, as an example of a peripheral territory in the EU. The duality found in the Irish industrial structure is the result of a FDI-led industrialisation strategy which has resulted in two distinct economic sectors – foreign and indigenous, respectively – with weak interactions between the two. Through increased public funding of academic research, the Irish government aimed to attract and embed new waves of higher-value foreign direct investment and increase the dynamism of its indigenous enterprise base. Based on a combination of quantitative and qualitative data, the paper analyses a crucial aspect of Ireland’s recent emphasis on STI policy – industry-academic linkages – and finds that the measures introduced reproduce in the public research system the uneven development found in Ireland’s productive system between indigenous industry and the foreign-owned industrial base.

Depuis que la haute énantiopureté est nécessaire dans l’industrie pharmaceutique, les études visant à découvrir les mécanismes pour l’hydrogénation énantiosélective de cétones ou céto-esters sur les surfaces, et à rechercher de nouveaux et plus performants catalyseurs asymétriques, sont d’une grande importance. La microscopie à effet tunnel (STM), la spectroscopie infrarouge de réflexion-absorption, la spectroscopie de désorption à température programmée et la spectrométrie de photoélectrons induits par rayons X sont des méthodes performantes facilitant la compréhension des mécanismes de réaction. En plus de nous permettre de comprendre les mécanismes réactionnels, les études peuvent fournir des informations sur la dynamique des réactions en catalyse hétérogène ainsi que sur le développement de la théorie de la fonctionnelle de la densité (DFT) afin de calculer des interactions faibles dans les processus de surface. D’autres parts, les calculs DFT fournissent une aide essentielle à l’interprétation des données de STM et spectroscopie de surface. Dans cette thèse, certains cétones et céto-esters sur la surface de platine sont étudiées par les techniques sophistiquées mentionnées ci-dessus. Mes études démontrent que la combinaison de l’utilisation de la spectroscopie de routine, des nanotechnologies et de nombreux calculs élaborés, est une méthode efficace pour étudier les réactions à la surface car ces techniques explorent les différents aspects de la surface ainsi que s’entraident mutuellement lors de certaines interprétations.
