Currently, mammalian cells are the most utilized hosts for biopharmaceutical production. The culture media for these cell lines include commonly in their composition a pH indicator. Spectroscopic techniques are used for biopharmaceutical process monitoring, among them, UV–Vis spectroscopy has found scarce applications. This work aimed to define artificial neural networks architecture and fit its parameters to predict some nutrients and metabolites, as well as viable cell concentration based on UV–Vis spectral data of mammalian cell bioprocess using phenol red in culture medium. The BHK-21 cell line was used as a mammalian cell model. Off-line spectra of supernatant samples taken from batches performed at different dissolved oxygen concentrations in two bioreactor configurations and with two pH control strategies were used to define two artificial neural networks. According to absolute errors, glutamine (0.13 ± 0.14 mM), glutamate (0.02 ± 0.02 mM), glucose (1.11 ± 1.70 mM), lactate (0.84 ± 0.68 mM) and viable cell concentrations (1.89 105 ± 1.90 105 cell/mL) were suitably predicted. The prediction error averages for monitored variables were lower than those previously reported using different spectroscopic techniques in combination with partial least squares or artificial neural network. The present work allows for UV–VIS sensor development, and decreases cost related to nutrients and metabolite quantifications.

Multivariate analyses of UV-Vis spectral data from cachaca wood extracts provide a simple and robust model to classify aged Brazilian cachacas according to the wood species used in the maturation barrels. The model is based on inspection of 93 extracts of oak and different Brazilian wood species by a non-aged cachaca used as an extraction solvent. Application of PCA (Principal Components Analysis) and HCA (Hierarchical Cluster Analysis) leads to identification of 6 clusters of cachaca wood extracts (amburana, amendoim, balsamo, castanheira, jatoba, and oak). LDA (Linear Discriminant Analysis) affords classification of 10 different wood species used in the cachaca extracts (amburana, amendoim, balsamo, cabreuva-parda, canela-sassafras, castanheira, jatoba, jequitiba-rosa, louro-canela, and oak) with an accuracy ranging from 80% (amendoim and castanheira) to 100% (balsamo and jequitiba-rosa). The methodology provides a low-cost alternative to methods based on liquid chromatography and mass spectrometry to classify cachacas aged in barrels that are composed of different wood species.

Ar matrix photolysis of 1- and 2-naphthyl azides 3 and 4 at 313 nm initially affords the singlet naphthyl nitrenes, (1)1 and (1)2. Relaxation to the corresponding lower energy, persistent triplet nitrenes (3)1 and (3)2 competes with cyclization to the azirines 15 and 18, which can also be formed photochemically from the triplet nitrenes. On prolonged irradiation, the azirines can be converted to the seven-membered cyclic ketenimines 10 and 13, respectively, as described earlier by Dunkin and Thomson. However, instead of the o-quinoid ketenimines 16 and 19, which are the expected primary ring-opening products of azirines 15 and 18, respectively, we observed their novel bond-shift isomers 17 and 20, which may be formally regarded as cyclic nitrile ylides. The existence of such ylidic heterocumulenes has been predicted previously, but this work provides the first experimental observation of such species. The factors which are responsible for the special stability of the ylidic species 17 and 20 are discussed.

El aguacate (Persea americana Miller) es una conocida fruta arb´orea con un alto contenido nutricional que crece en varias partes del mundo. El presente estudio compara los espectros del UV-Vis y del espectr´ometro infrarrojo con transformada de Fourier (FTIR) de la fruta y de la hoja de aguacate (c´ascara, pulpa y aceite) cultivado en Ecuador y posteriormente eval´ua su actividad antioxidante empleando el 1,1-difenil-2-picrilhidrazil (DPPH•). El estudio de los espctros UV-Vis y FTIR revel´o que el aguacate tiene predominantemente flavonoides. Entre la hoja y el fruto del aguacate, se comprob´o mediante el ensayo DPPH• (captura de radicales libres), que la hoja tuvo una mayor actividad antioxidante, la que oscila entre 84,46% y 80,12%, con valores de 32.60-32.73 μg equivalentes de ´acido g´alico por mL. Se demostr´o que el orden de la actividad antioxidante de los extractos es: hoja de aguacate > c´ascara > aceite > pulpa. La actividad antioxidante tuvo una correlaci´on positiva con el contenido total de flavonoides y estos extractos de plantas (especialmente de las hojas del aguacate) son ´utiles para el desarrollo de futuros productos antioxidantes.

UV-vis photodissociation action spectroscopy is becoming increasingly prevalent because of advances in, and commercial availability of, ion trapping technologies and tunable laser sources. This study outlines in detail an instrumental arrangement, combining a commercial ion-trap mass spectrometer and tunable nanosecond pulsed laser source, for performing fully automated photodissociation action spectroscopy on gas-phase ions. The components of the instrumentation are outlined, including the optical and electronic interfacing, in addition to the control software for automating the experiment and performing online analysis of the spectra. To demonstrate the utility of this ensemble, the photodissociation action spectra of 4-chloroanilinium, 4-bromoanilinium, and 4-iodoanilinium cations are presented and discussed. Multiple photoproducts are detected in each case and the photoproduct yields are followed as a function of laser wavelength. It is shown that the wavelength-dependent partitioning of the halide loss, H loss, and NH3 loss channels can be broadly rationalized in terms of the relative carbon-halide bond dissociation energies and processes of energy redistribution. The photodissociation action spectrum of (phenyl)Ag-2 (+) is compared with a literature spectrum as a further benchmark.

Recent investigations into plant tissues have indicated that the free form of the natural polyphenolic antioxidant, ellagic acid (EA), is much more plentiful than first envisaged; consequently a re-assessment of solvent systems for the extraction of this water-insoluble form is needed. As EA solubility and its UV-Vis spectrum, commonly used for detection and quantification, are both governed by pH, an understanding of this dependence is vital if accurate EA measurements are to be achieved. After evaluating the pH effects on the solubility and UV-Vis spectra of commercial EA, an extraction protocol was devised that promoted similar pH conditions for both standard solutions and plant tissue extracts. The extraction so devised followed by HPLC with photodiode-array detection (DAD) provided a simple, sensitive and validated methodology that determined free EA in a variety of plant extracts. The use of 100 % methanol or a triethanolamine-based mixture as the standard dissolving solvents were the best choices, while these higher pH-generating solvents were more efficient in extracting EA from the plants tested with the final choice allied to the plants’ natural acidity. Two of the native Australian plants anise myrtle (Syzygium anisatum) and Kakadu plum (Terminalia ferdinandiana) exhibited high concentrations of free EA. Furthermore, the dual approach to measuring EA UV-Vis spectra made possible an assessment of the effect of acidified eluent on EA spectra when the DAD was employed.

As indústrias consomem volumes elevados de água e outras substâncias químicas na síntese dos seus produtos e geram grande quantidade de rejeitos. Entre os mais importantes poluentes encontrados nos efluentes dessas indústrias estão os corantes sintéticos que representam um problema, pois não são facilmente destruídos por tratamentos convencionais. A fotocatálise heterogênea tem sido considerada como uma alternativa efetiva no tratamento de efluentes contendo esses corantes. Neste trabalho, estudou-se a cinética de descoloração e o grau de mineralização dos corantes sintéticos reativos Yellow 145, Black 5, Red 4 e Blue 21 através da fotocatálise utilizando ZnO puro e impregnado com íons Fe2+ e Co2+. Testes preliminares foram realizados para otimizar a concentração dos corantes e a massa mínima de catalisador a ser utilizado nos experimentos fotocatalíticos. Além da fotocatálise, experimentos individuais de fotólise e adsorção também foram realizados, porém se mostraram poucos eficientes. Através da espectrofotometria UV-Vis, verificou-se o total descoramento individual dos corantes em aproximadamente 30 minutos de irradiação com ZnO. O grau de mineralização de cada corante foi determinado através de análise de carbono orgânico total (COT), atingindo-se cerca de 70 a 80% de mineralização após 240 minutos de tratamento fotocatalítico. Foram comparadas, ainda, as eficiências de cada fotocatalisador ZnO, Fe/ZnO e Co/ZnO na mineralização de uma solução contendo a mistura dos quatro corantes já mencionados após 240 minutos de reação. A eficiência na mineralização da mistura dos corantes seguiu a seguinte ordem: Co/ZnO (32%), ZnO (78%) e Fe/ZnO (87%). A reação de degradação fotocatalítica do corante Black 5 seguiu uma cinética de primeira ordem, enquanto que os corantes Yellow 145, Red 4 e Blue 21 seguiram uma cinética de ordem zero.

Electrochemically induced three conformational transitions of calf thymus DNA from B-10.4 to Z(10.2)-DNA and from B-10.2 to B-10.4 and to C-DNA in 10 mM phosphate buffer solution (pH 7.21) at glassy carbon electrode are found and studied by in situ circular dichroism (CD) thin layer spectroelectrochemistry with singular value decomposition least square (SVDLS) analysis. It indicates that the so-called B-10.2 form and the C-form of DNA may be composed of B-10.4 and left-A DNA and of B-10.4 and right-A DNA, respectively. The irreversible electrochemical reduction of adenine and cytosine groups in the DNA molecule is studied by UV-Vis spectroelectrochemistry. Some electrochemical parameters alphan = 0.17, E-0' = -0.70 V (vs. Ag/AgCl), and the standard heterogeneous electron transfer rate constant, k(0) = 1.8 x 10(-5) cm s(-1) are obtained by double logarithmic analysis and non-linear regression. Aniline pentamer and hexamer in the leucoemeraldine oxidation state were synthesized through a novel method. The method was accomplished by the reaction of parent aniline tetramer in the pernigraniline oxidation state with diphenylamine and N-phenyl-1,4-phenylenediamine in the leucoemeraldine oxidation state respectively. The oligomers in the leucoemeraldine oxidation state were characterized by IR, NMR, elemental analysis and MALDI-MS. Aniline pentamer and hexamer in the emeraldine oxidation state were synthesized by the oxidation of Ag2O in DMF. It was found that some fragmentation occurred when the pentamer and hexamer were oxidized by (NH4)(2)S2O8 and FeCl3. 6H(2)O. The pentamer and hexamer in the emeraldine oxidation state was studied by UV/Vis spectra. The relative intensity of exciton peak for pentaaniline showed a little increase compared with that of hexaaniline.

Silicalite-I, ZSM-5, and Fe-ZSM-5 zeolites prepared from two different silicon sources are characterized by UV resonance Raman (UVRR) spectroscopy, X-ray diffraction (XRD), electron spin resonance (ESR), and UV/visible diffuse reflectance spectroscopy (UV/Vis DRS). A new technique for investigating zeolitic structure, UV resonance Raman spectroscopy selectively enhances the Raman bands associated with framework iron atoms incorporated into MFI-type zeolites, and it is very sensitive in identifying the iron atoms in the framework of zeolites, while other techniques such as XRD, ESR, and UV/Vis DRS have failed in uncovering trace amounts of iron atoms in the framework of zeolites.
