Intense exercise results in muscular inflammation. Molecular techniques were used to identify novel inflammatory proteins in human muscle. Males and females displayed different levels of exercise-induced inflammatory proteins. Interestingly, dairy protein supplements reduced these inflammatory proteins post-exercise. Increased dietary red meat consumption, with training, had no impact on muscle inflammation, although strength gain was improved.

This thesis found that light exercise between repeated sprints improved performance in a subsequent bout. This was attributed to a reduction in potentially fatiguing by-products within the muscle and an increased aerobic metabolism in the second sprint.

The results of this thesis indicate that vegetarianism and prior exercise can increase skeletal muscle creatine loading. The mechanisms by which these phenomena occur remain unknown but may be related to changes in the gene expression of a cellular pump within the muscle known as the "creatine transporter".

Demonstrates that anaerobic capacity, using the accumulated oxygen deficit technique, can be measured rapidly and with a high degree of precision, however significant difficulties remain to be addressed. Significant further insights into some of the underlying cellular mechanisms associated with increases in anaerobic capacity were also obtained.

The effects of exercise on novel signalling enzymes in skeletal muscle of humans was investigated. It was shown that exercise increased the activity of a calcium and calmodulin activated kinase. High-intensity, but submaximal, exercise increased the activity of some but not all isoforms of protein kinase C, a lipid-activated kinase family. These findings suggest that these enzymes may be part of the signalling process leading to beneficial adaptation to repeated exercise as well as the control of function within skeletal muscle during exercise.

The findings from this 18-month, community-based study revealed that an exercise program involving strength training and jumping activities was feasible and effective for improving bone density, muscle mass and strength in older men. There were no additional skeletal benefits derived from consuming a high calcium-vitamin D milk drink.

Ý-lactoglobulin enriched whey protein isolate, but not carbohydrate, increased growth signalling in human skeletal muscle when consumed in conjunction with resistance exercise. Ageing did not impair the anabolic signalling response; however this response was attenuated after training. These findings help identify strategies to prevent, or delay the onset of sarcopenia.

This qualitative study explores reasons why women who participate in exercise to music continue to do so long-term. Thirteen women were interviewed. Generational differences of body perceptions were easily identifiable.

Female athletes may demonstrate a poor ability to quantify portion sizes and educational sessions using food models do not always improve accuracy. Female cyclists with the highest body fat do not report the highest energy intakes during 70 days of training and racing. Training loads can influence reported daily energy intake.

In activities such as walking individuals can select an optimum speed that minimises energy expenditure. When learning to row, individuals initially selected fast inefficient stroke rates but learned to become more efficient by taking longer, slower strokes. The research showed, therefore, that optimum pacing depends on extensive practice and sensitivity to energy cost helps us to change movements in order to become more efficient.

Understanding muscle adaptation and repair is vital for preserving muscle loss with aging. Analysis of the inflammatory-responsive signalling pathway, JAK/STAT was performed. After intense exercise, the STAT3 pathway is highly activated, potentially by the pro-inflammatory regulator IL-6. This pathway is suppressed in older individuals, possible leading to altered inflammatory regulation.

The collective findings of this dissertation demonstrated little effect of exercise on the absolute or relative expression of glycogen regulatory proteins associated with a glycogen enriched fraction in human skeletal muscle. However the findings of this thesis help inform methodological approaches to future investigations into glycogen-protein associations.

This thesis investigated the physical activity behaviours of employees who commonly sit for work tasks. Specifically, this PhD explored the key influences on light-intensity physical activity during work hours and issues associated with subjectively assessing this behaviour. Overall, this PhD provided insights into a novel approach to address the important public health issue of physical inactivity.
