Tässä työssä suunniteltiin lappeenrantalaisen Astex Oy:n tilauksesta liikkuvan työkoneen runkorakenne. Työ tehtiin Lappeenrannan teknillisen yliopiston teräsrakenteiden laboratoriossa. Suunniteltava rakenne kuului linkkuohjattuun, noin 5000 kg painoiseen trukkitraktoriin. Lähtökohtana rakenteen suunnittelulle olivat tilaajan asettamat rakenteen geometriaan ja suorituskykyyn liittyvät reunaehdot ja rajoitteet. Uuden rakenteen suunnittelussa hyödynnettiin myös tilaajatahon kehittelemää vastaavan tyyppistä prototyyppirakennetta. Rakenteen suunnitteluprosessi koostui neljästä eri vaiheesta. Suunniteltavan rakenteen lähtökohtana olleelle prototyyppirakenteelle suoritettiin koneen käytönaikaisia venymäliuskamittauksia rakenteen kuormitushistorian selvittämiseksi. Mittauksista saatujen tulosten perusteella määritettiin kuormitukset uudelle suunniteltavalle rakenteelle. Määritettyjä kuormituksia hyödyntäen ideoitiin, suunniteltiin ja mallinnettiin uusi tilaajan vaatimuksia vastaava rakenne. Uudelle rakenteelle suoritettiin lujuusanalyysit FE-analyysiä hyödyntäen. Uuden rakenteen suunnittelussa kiinnitettiin huomiota rakenteen hyvään valmistettavuuteen ja suunniteltiin rakenneratkaisut tilaajataholle mahdollisimman optimaalisiksi. Suunnittelu- ja mallinnustyö tehtiin Solidworks 2014 ohjelmistolla. Rakenteen lujuustekniset tarkastelut sisälsivät rakennedetaljien analyyttistä mitoitusta ja laskentaa. FE-laskennalla selvitettiin rakenteen ääri- ja väsymiskestävyys. Laskenta sisälsi koko rakenteen globaaleja tarkasteluja, sekä eri kriittisten rakennedetaljien yksityiskohtaisempia analyysejä. FE-laskennan pääpaino oli rakenteen väsymisanalyyseissä, jotka toteutettiin Hot-Spot- ja särönkasvumenetelmillä. Rakenteen FE-analyysien suorittamisessa käytettiin Femap, NxNastran ja Abaqus-ohjelmistoja.

Tässä työssä tutkittiin kuinka eri tavoin rakenteen mallintaminen vaikuttaa laskettuun väsy-misikään. Työssä tutkittavana rakenteena toimi Sandvik Mining and Construction OY:n las-tauskoneen nostovarsi. Nostovarrelle väsytyskokeet suoritettiin Lappeenrannan teknillisen yliopiston teräsrakenteiden laboratoriossa. Työn tavoitteena oli tutkia miten eri digitaalinen valmistuksen tasot vaikuttavat hitsatulle rakenteelle saatuun kestoikään. Työssä tutkittiin myös miten todellista geometriaa voidaan hyödyntää rakenteen kestoiän arvioinnissa. Väsytyskoejärjestely mallinnettiin FE-menetelmällä, ja järjestelystä tehtiin useita malleja käyttäen solidi- ja laattaelementtejä. Malleista laskettiin väsymisiät hot spot- ja tehollisen lovijännityksen menetelmällä, ja saatuja tuloksia vertailtiin toisiinsa ja väsytyskokeen tulok-siin. Väsytyskokeessa vaurioituneista kohdista tarkemman tutkimuksen kohteena oli nosto-varren palstalevyn kärki. Hot spot-menetelmällä saadut kestoiät vaihtelivat paikoin melko paljon eri mallien välillä. Tehollisen lovijännityksen menetelmällä saaduissa tuloksissa erot olivat pienempiä mallien välillä. FE-mallin ja venymäliuskojen jännitykset poikkesivat toisistaan paikoin melko pal-jon. Todellisen hitsatun rakenteen kestoikään vaikuttaa moni asia, ja täten FE-menetelmällä las-kettu kestoikä voi poiketa huomattavasti todellisesta kestoiästä. Varsinkin hot spot-menetel-mällä tulokset voivat poiketa hyvinkin paljon todellisuudesta, mikäli jännitystila tutkitta-vassa kohdassa on moniaksiaalinen. Todellisen geometrian mallintaminen vaatii tarkkuutta, ja alkuperäisdatan tulee olla mahdollisimman tarkkaa ja riittävän suurelta alueelta, jotta malli vastaa tarpeeksi todellista.

El interés de esta monografía es analizar la incidencia de la estrategia de seguridad ciudadana de los Hot Spots en los niveles de delincuencia de Bogotá durante el periodo de la Alcaldía de Samuel Moreno 2009-2011, tomando como casos de estudio la Avenida 19 y la Zona Rosa (Chicó El Lago). Esta investigación se realiza desde los conceptos teórico-prácticos que ha arrojado el modelo de Hot Spot tras su aplicación en 18 ciudades del mundo, y que sirven como herramienta para analizar las transformaciones del delito en las zonas de estudio. De esta manera se avanza hacia el resultado que permite identificar los factores que impidieron que el modelo de Hot Spots se formulara y ejecutara efectivamente y por lo tanto no disminuyera los niveles de delincuencia en Bogotá.

Arousal sometimes enhances and sometimes impairs perception and memory. In our Glutamate Amplifies Noradrenergic Effects (GANE) model, glutamate at active synapses interacts with norepinephrine released by the locus coeruleus to create local ‘hot spots’ of activity that enable the selective effects of arousal. This hot spot mechanism allows local cortical regions to self-regulate norepinephrine release based on current activation levels. In turn, hot spots bias global energetic delivery and functional network connectivity to enhance processing of high priority representations and impair processing of lower priority representations.

The Atlantic Rain Forest, an important biodiversity hot spot, has faced severe habitat loss since the last century which has resulted in a highly fragmented landscape with a large number of small forest patches (<100 ha). For conservation planning it is essential to understand how current and future forest regeneration depends on ecological processes, fragment size and the connection to the regional seed pool. We have investigated the following questions by applying the forest growth simulation model FORMIND to the situation of the Atlantic Forest in the state of Sao Paulo, SE Brazil: (1) which set of parameters describing the local regeneration and level of density regulation can reproduce the biomass distribution and stem density of an old growth forest in a reserve? (2) Which additional processes apart from those describing the dynamics of an old growth forest, drive forest succession of small isolated fragments? (3) Which role does external seed input play during succession? Therefore, more than 300 tree species have been classified into nine plant functional types (PFTs), which are characterized by maximum potential height and shade tolerance. We differentiate between two seed dispersal modes: (i) local dispersal, i.e. all seedlings originated from fertile trees within the simulated area and (ii) external seed rain. Local seed dispersal has been parameterized following the pattern oriented approach, using biomass estimates of old growth forest. We have found that moderate density regulation is essential to achieve coexistence for a broad range of regeneration parameters. Considering the expected uncertainty and variability in the regeneration processes it is important that the forest dynamics are robust to variations in the regeneration parameters. Furthermore, edge effects such as increased mortality at the border and external seed rain have been necessary to reproduce the patterns for small isolated fragments. Overall, simulated biomass is much lower in the fragments compared to the continuous forest, whereas shade tolerant species are affected most strongly by fragmentation. Our simulations can supplement empirical studies by extrapolating local knowledge on edge effects of fragments to larger temporal and spatial scales. In particular our results show the importance of external seed rain and therefore highlight the importance of structural connectivity between regenerating fragments and mature forest stands. Soil CO2 efflux is the primary source of CO2 emissions from terrestrial ecosystems to the atmosphere. The rates of this flux vary in time and space producing hot moments (sudden temporal high fluxes) and hot spots (spatially defined high fluxes), but these high reaction rates are rarely studied in conjunction with each other. We studied temporal and spatial variation of soil CO2 efflux in a water-limited Mediterranean ecosystem in Baja California, Mexico. Soil CO2 efflux increased 522% during a hot moment after rewetting of soils following dry summer months. Monthly precipitation was the primary driver of the seasonal trend of soil CO2 efflux (including the hot moment) and through changes in soil volumetric water content (VWC) it influenced the relationship between CO2 efflux and soil temperature. Geostatistical analyses showed that the spatial dependence of soil CO2 efflux changed between two contrasting seasons (dry and wet). During the dry season high soil VWC was associated with high soil CO2 efflux, and during the wet season the emergence of a hot spot of soil CO2 efflux was associated with higher root biomass and leaf area index. These results suggest that sampling designs should accommodate for changes in spatial dependence of measured variables. The spatio-temporal relationships identified in this study are arguably different from temperate ecosystems where the majority of soil CO2 efflux research has been done. This study provides evidence of the complexity of the mechanisms controlling the spatio-temporal variability of soil CO2 efflux in water-limited ecosystems. (C) 2014 Elsevier Ltd. Trypanosoma cruzi is an organism highly resistant to ionizing radiation. Following a dose of 500 Gy of gamma radiation, the fragmented genomic DNA is gradually reconstructed and the pattern of chromosomal bands is restored in less than 48 hours. Cell growth arrests after irradiation but, while DNA is completely fragmented, RNA maintains its integrity. In this work we compared the transcriptional profiles of irradiated and non-irradiated epimastigotes at different time points after irradiation using microarray. In total, 273 genes were differentially expressed; from these, 160 were up-regulated and 113 down-regulated. We found that genes with predicted functions are the most prevalent in the down-regulated gene category. Translation and protein metabolic processes, as well as generation of precursor of metabolites and energy pathways were affected. In contrast, the up-regulated category was mainly composed of obsolete sequences (which included some genes of the kinetoplast DNA), genes coding for hypothetical proteins, and Retrotransposon Hot Spot genes. Finally, the tyrosyl-DNA phosphodiesterase 1, a gene involved in double-strand DNA break repair process, was up-regulated. Our study demonstrated the peculiar response to ionizing radiation, raising questions about how this organism changes its gene expression to manage such a harmful stress.

Although it is well known that the thyroid hormone (T3) is an important positive regulator of cardiac function over a short term and that it also promotes deleterious effects over a long term, the molecular mechanisms for such effects are not yet well understood. Because most alterations in cardiac function are associated with changes in sarcomeric machinery, the present work was undertaken to find novel sarcomeric hot spots driven by T3 in the heart. A microarray analysis indicated that the M-band is a major hot spot, and the structural sarcomeric gene coding for the M-protein is severely down-regulated by T3. Real-time quantitative PCR-based measurements confirmed that T3 (1, 5, 50, and 100 physiological doses for 2 days) sharply decreased the M-protein gene and protein expression in vivo in a dose-dependent manner. Furthermore, the M-protein gene expression was elevated 3.4-fold in hypothyroid rats. Accordingly, T3 was able to rapidly and strongly reduce the M-protein gene expression in neonatal cardiomyocytes. Deletions at the M-protein promoter and bioinformatics approach suggested an area responsive to T3, which was confirmed by chromatin immunoprecipitation assay. Functional assays in cultured neonatal cardiomyocytes revealed that depletion of M-protein (by small interfering RNA) drives a severe decrease in speed of contraction. Interestingly, mRNA and protein levels of other M-band components, myomesin and embryonic-heart myomesin, were not altered by T3. We concluded that the M-protein expression is strongly and rapidly repressed by T3 in cardiomyocytes, which represents an important aspect for the basis of T3-dependent sarcomeric deleterious effects in the heart.

In many animals, males congregate in leks that females visit for the sole purpose of mating. We observed male and female behavior on 3 different-sized leks of the bower-building cichlid fish Nyassachromis cf. microcephalus to test predictions of 3 prominent lek models: the "hotshot," "hot spot," and "female preference" models. In this system, we were able to refine these predictions by distinguishing between indirect mate choice, by which females restrict their set of potential mates in the absence of individual male assessment, and direct mate choice, by which females assess males and their territories through dyadic behavioral interactions. On no lek were males holding central territories favored by indirect or direct mate choice, contrary to the prediction of the hotshot model that leks form because inferior males establish territories surrounding hotshot males preferred by females. Average female encounter rate of males increased with lek size, a pattern typically interpreted as evidence that leks form through female preference for lekking males, rather than because males congregate in hot spots of high female density. Female propensity to engage in premating behavior once courted did not increase with lek size, suggesting female preference for males on larger leks operated through indirect choice rather than direct choice based on individual assessment. The frequency of male-male competitive interactions increased with lek size, whereas their foraging rate decreased, implying a cost to males maintaining territories on larger leks. Together these data most strongly support the female preference model, where females may benefit through indirect mate choice for males able to meet the competitive cost of occupying larger leks.

Tumor necrosis factor (TNF)-Receptor Associated Factors (TRAFs) are a family of signal transducer proteins. TRAF6 is a unique member of this family in that it is involved in not only the TNF superfamily, but the toll-like receptor (TLR)/IL-1R (TIR) superfamily. The formation of the complex consisting of Receptor Activator of Nuclear Factor κ B (RANK), with its ligand (RANKL) results in the recruitment of TRAF6, which activates NF-κB, JNK and MAP kinase pathways. TRAF6 is critical in signaling with leading to release of various growth factors in bone, and promotes osteoclastogenesis. TRAF6 has also been implicated as an oncogene in lung cancer and as a target in multiple myeloma. In the hopes of developing small molecule inhibitors of the TRAF6-RANK interaction, multiple steps were carried out. Computational prediction of hot spot residues on the protein-protein interaction of TRAF6 and RANK were examined. Three methods were used: Robetta, KFC2, and HotPoint, each of which uses a different methodology to determine if a residue is a hot spot. These hot spot predictions were considered the basis for resolving the binding site for in silico high-throughput screening using GOLD and the MyriaScreen database of drug/lead-like compounds. Computationally intensive molecular dynamics simulations highlighted the binding mechanism and TRAF6 structural changes upon hit binding. Compounds identified as hits were verified using a GST-pull down assay, comparing inhibition to a RANK decoy peptide. Since many drugs fail due to lack of efficacy and toxicity, predictive models for the evaluation of the LD50 and bioavailability of our TRAF6 hits, and these models can be used towards other drugs and small molecule therapeutics as well. Datasets of compounds and their corresponding bioavailability and LD50 values were curated based, and QSAR models were built using molecular descriptors of these compounds using the k-nearest neighbor (k-NN) method, and quality of these models were cross-validated.

Selected basalts from a suite of dredged and drilled samples (IPOD sites 525, 527, 528 and 530) from the Walvis Ridge have been analysed to determine their rare earth element (REE) contents in order to investigate the origin and evolution of this major structural feature in the South Atlantic Ocean. All of the samples show a high degree of light rare earth element (LREE) enrichment, quite unlike the flat or depleted patterns normally observed for normal mid-ocean ridge basalts (MORBs). Basalts from Sites 527, 528 and 530 show REE patterns characterised by an arcuate shape and relatively low (Ce/Yb)N ratios (1.46-5.22), and the ratios show a positive linear relationship to Nb content. A different trend is exhibited by the dredged basalts and the basalts from Site 525, and their REE patterns have a fairly constant slope, and higher (Ce/Yb)N ratios (4.31-8.50). These differences are further reflected in the ratios of incompatible trace elements, which also indicate considerable variations within the groups. Mixing hyperbolae for these ratios suggest that simple magma mixing between a 'hot spot' type of magma, similar to present-day volcanics of Tristan da Cunha, and a depleted source, possibly similar to that for magmas being erupted at the Mid-Atlantic Ridge, was an important process in the origin of parts of the Walvis Ridge, as exemplified by Sites 527, 528 and 530. Site 525 and dredged basalts cannot be explained by this mixing process, and their incompatible element ratios suggest either a mantle source of a different composition or some complexity to the mixing process. In addition, the occurrence of different types of basalt at the same location suggests there is vertical zonation within the volcanic pile, with the later erupted basalts becoming more alkaline arid more enriched in incompatible elements. The model proposed for the origin and evolution of the Walvis Ridge involves an initial stage of eruption in which the magma was essentially a mixture of enriched and depleted end-member sources, with the N-MORB component being small. The dredged basalts and Site 525, which represent either later-stage eruptives or those close to the hot spot plume, probably result from mixing of the enriched mantle source with variable amounts and variable low degrees of partial melting of the depleted mantle source. As the volcano leaves the hot spot, these late-stage eruptives continue for some time. The change from tholeiitic to alkalic volcanism is probably related either to evolution in the plumbing system and magma chamber of the individual volcano, or to changes in the depth of origin of the enriched mantle source melt, similar to processes in Hawaiian volcanoes.

The hot-spot phenomenon is a relatively frequent problem in current photovoltaic generators. It entails both a risk for the photovoltaic module's lifetime and a decrease in its operational efficiency. Nevertheless, there is still a lack of widely accepted procedures for dealing with them in practice. This paper presents the IES UPM observations on 200 affected modules. Visual and infrared inspection, electroluminescence, peak power and operating voltage tests have been accomplished. Hot-spot observation procedures and well defined acceptance and rejection criteria are proposed, addressing both the lifetime and the operational efficiency of the modules. The operating voltage has come out as the best parameter to control effective efficiency losses for the affected modules. This procedure is oriented to its possible application in contractual frameworks.

Seamounts, submarine banks, volcanoes and undercurrent channels are prominent geomorphic features that have become an important target for minerals research and exploration with the goal of future exploitation. Polymetallic ferromanganese deposits are common types of mineralization on these settings. Co-rich ferromanganese crusts are important as potential resources of Mn and Co, but also Ti, Ni, Tl, REEs, PGEs, and other metals. Many seamounts and channels along the Atlantic Spanish continental margin are known to hold mineral deposits but are poorly studied. This work presents and briefly describes the most recent activities of the Spanish Geological Survey (IGME) on exploration and investigation of ferromanganese deposits along the Atlantic Spanish continental margin. Different submarine areas from the northwestern margin of the Iberian Peninsula to the west off Canary Islands have been surveyed by geophysical, sampling and underwater observations from 89 to 4000 m water depth. The mineral deposits cover a large diversity of submarine geological and geomorphical features: mud volcanoes and diapirs related to hydrocarbon seeps, seamounts associated with hot spot volcanism, hydrothermal vents in active magmatic volcanoes, structural basement highs and banks or contourite channels. Considering the collected dataset, we present the preliminary results of the study of these mineral deposits, including ferromanganese nodules and crusts and phosphate pavements and nodules, which can be considered as potential sources of raw materials.

The hot-spot phenomenon is a relatively frequent problem occurring in current photovoltaic generators. It entails both a risk for the photovoltaic module’s lifetime and a decrease in its operational efficiency. Nevertheless, there is still a lack of widely accepted procedures for dealing with them in practice. This paper presents the IES–UPM observations on 200 affected photovoltaic modules. Visual and infrared inspection, as well as electroluminescence, peak power rating and operating voltage tests have been carried out. Thermography under steady state conditions and photovoltaic module operating voltage, both at normal photovoltaic system operating conditions, are the selected methods to deal in practice with hot-spots. The temperature difference between the hot-spot and its surroundings, and the operating voltage differences between affected and non-affected photovoltaic modules are the base for establishing defective criteria, at the lights of both lifetime and operating efficiency considerations. Hot-spots temperature gradients larger than 20 °C, in any case, and larger than 10 °C when, at the same time, voltage operating losses are larger than the allowable power losses fixed at the photovoltaic module warranties, are proposed as rejecting conditions for routine inspections under contractual frameworks. The upper threshold of 20 °C is deduced for temperate climates from the basic criterion of keeping absolute hot-spot temperatures below 20 °C.

Organization of transgenes in rice transformed through direct DNA transfer strongly suggests a two-phase integration mechanism. In the “preintegration” phase, transforming plasmid molecules (either intact or partial) are spliced together. This gives rise to rearranged transgenic sequences, which upon integration do not contain any interspersed plant genomic sequences. Subsequently, integration of transgenic DNA into the host genome is initiated. Our experiments suggest that the original site of integration acts as a hot spot, facilitating subsequent integration of successive transgenic molecules at the same locus. The resulting transgenic locus may have plant DNA separating the transgenic sequences. Our data indicate that transformation through direct DNA transfer, specifically particle bombardment, generally results in a single transgenic locus as a result of this two-phase integration mechanism. Transgenic plants generated through such processes may, therefore, be more amenable to breeding programs as the single transgenic locus will be easier to characterize genetically. Results from direct DNA transfer experiments suggest that in the absence of protein factors involved in exogenous DNA transfer through Agrobacterium, the qualitative and/or quantitative efficiency of transformation events is not compromised. Our results cast doubt on the role of Agrobacterium vir genes in the integration process.
