This paper attempts to demonstrate the advantages of adaptive boundary element techniques and their implementation in microcomputers. It is shown how it is possible to treat elastostatics problems using adaptive BEM techniques. The paper comments on the solutions proposed for the problems encountered when tryng to implement adaptive BEM in minicomputers.

A poluição relacionada a metais pesados tem recebido uma atenção especial devido a sua alta toxicidade, não biodegradabilidade e tendência de acumular-se na cadeia alimentar. Apesar disso, metais pesados também são considerados recursos valiosos, portanto a sua remoção em conjunto com a sua recuperação torna-se ainda mais importante. Este caso aplica-se aos rejeitos de mineração de cobre, os quais oferecem a possibilidade de recuperação do metal e de sua contenção de maneira segura do meio ambiente. Tais rejeitos se caracterizam por ocuparem enormes áreas inundadas e abrigarem soluções diluídas de cobre (II), porém, muitas vezes, acima dos limites seguros. Diversos processos tradicionais de tratamento mostram-se disponíveis para remover o cobre de tais soluções, no entanto, em certas aplicações eles podem ser ineficientes ou muito onerosos. Nesse contexto, a biossorção é uma alternativa interessante. Nesse processo, certos microrganismos, como fungos, bactérias e algas, ligam-se passivamente ao cobre na forma íons ou outras moléculas em soluções. No presente trabalho foi avaliado o potencial de biossorção de íons cobre (II) pela biomassa do fungo Rhizopus microsporus, coletado e isolado da área de rejeitos da Mina do Sossego, na região norte do Brasil. Isotermas de biossorção foram determinadas experimentalmente em bateladas sob temperatura de 25°C, agitação de 150 rpm, concentração de biomassa de 2,0 a 2,5 g/L e tempo de contato mínimo de 4 horas. O pH mostrou ser um fator importante no equilíbrio da biossorção, sendo o valor máximo da capacidade de biossorção de 33,12 mg de cobre / g biomassa encontrado em pH 6. Valores sucessivamente menores são encontrados pela acidificação da solução, sendo o pH 1 considerado adequado para o processo de dessorção, correspondendo a uma capacidade de biossorção de 1,95 mg/g. Modelos de adsorção de Langmuir e de Freundlich ajustaram-se adequadamente às isotermas tanto com pH controlado quanto não controlado. Foi constatado que a troca iônica é um dos mecanismos envolvidos na biossorção do cobre com Rhizopus microsporus. Tanto o modelo de pseudo-primeira ordem quanto o de pseudo-segunda ordem ajustaram-se aos dados cinéticos da biossorção, sendo que o equilíbrio ocorre em aproximadamente 4 horas. A biomassa conservou a capacidade de biossorção ao operar repetidamente em três ciclos de sorção-dessorção. A biomassa viável e a morta não apresentaram diferença estatisticamente significativa na capacidade de biossorção.

Rhizopus arrhizus, strain DAR 36017, produced L(+)-lactic acid in a simultaneous saccharification and fermentation process using starch waste effluents. Lactic acid at 19.5 - 44.3 g l(-1) with a yield of 0.85 - 0.96 g g(-1) was produced in 40 h using 20 - 60 g starch l(-1). Supplementation of nitrogen source may be unnecessary if potato or corn starch waste effluent was used as a production medium.

This paper describes a feasibility study of a for lactic acid production integrated with are treatment of wastewater from an industrial starch plant. Rhizopus oryzae two strains, Rhizopus arrhizus and Rhizopus oligosporus were tested with respect to their capability to carry out simultaneous saccharification and fermentation to lactic acid using potato wastewater. Rhizopus arrhizus DAR 36017 was identified as a suitable strain that demonstrated a high capacity for starch saccharification and lactic acid synthesis. The optimal conditions, in terms of pH, temperature and starch concentration, for lactic acid production were determined. The selected fungal strain grew well in a pH range from 3.0 to 7.0. The addition of CaCO(3)10 g dm(-3) maintained the pH at 5.0-6.0 and significantly enhanced lactic acid production. Kinetic study revealed that almost complete starch saccharification and a lactic acid yield of 450g kg(-1) could be achieved in 20 h and 28 h cultivation, respectively. The maximum lactic acid production 21 g dm(-3) and mycelial biomass (1.7 g dm(-3)) were obtained at 30degreesC. Besides the multiple bioproducts, total removal of suspended solids and 90% reduction of COD were achieved in a single no-aseptic operation. (C) 2003 Society of Chemical Industry.

The fungal species of Rhizopus oryzae 2062 has the capacity to carry out a single stage fermentation process for lactic acid production from potato starch wastewater. Starch hydrolysis, reducing sugar accumulation, biomass formation, and lactic acid production were affected with variations in pH, temperature, and starch source and concentration. A growth condition with starch concentration approximately 20 g/L at pH 6.0 and 30degreesC was favourable for starch fermentation, resulting in a lactic acid yield of 78.3%similar to85.5% associated with 1.5similar to2.0 g/L fungal biomass produced in 36 h of fermentation.

The biochemical kinetic of direct fermentation for lactic acid production by fungal species of Rhizopus arrhizus 3,6017 and Rhizopus oryzae 2,062 was studied with respect to growth pH, temperature and substrate. The direct fermentation was characterized by starch hydrolysis, accumulation of reducing sugar, and production of lactic acid and fungal biomass. Starch hydrolysis, reducing sugar accumulation, biomass formation and lactic acid production were affected with the variations in pH, temperature, and starch source and concentration. A growth condition with starch concentration approximately 20 g/l at pH 6.0 and 30 degrees C was favourable for both starch saccharification and lactic acid fermentation, resulting in lactic acid yield of 0.87-0.97 g/g starch associated with 1.5-2.0 g/l fungal biomass produced in 36 h fermentation. R. arrhizus 3,6017 had a higher capacity to produce lactic acid, while R. oryzae 2,062 produced more fungal biomass under similar conditions.

Both R. arrhizus 36017 and R. oryzae 2062 had a capacity to carry out a single stage SSF process for lactic acid production from potato starch wastewater. The kinetic characteristics, termed as starch hydrolysis, accumulation of reducing sugars, lactic acid production and fungal biomass formation, were affected with variations in pH, temperature, and starch source and concentration. A growth condition with starch concentration approximately 20 g/l at pH 6.0 and 30 degrees C was favourable for both starch saccharification and lactic acid fermentation, resulting in lactic acid yield of 0.85-0.92 g/g associated with 1.5-3.5 g/l fungal biomass produced in 36-48 h fermentation. This thesis explores the interaction between Micros (<10 employees) from non-creative sectors and website designers ("Creatives") that occurred when creating a website of a higher order than a basic template site. The research used Straussian Grounded Theory Method with a longitudinal design, in order to identify what knowledge transferred to the Micros during the collaboration, how it transferred, what factors affected the transfer and outcomes of the transfer including behavioural additionality. To identify whether the research could be extended beyond this, five other design areas were also examined, as well as five Small to Medium Enterprises (SMEs) engaged in website and branding projects. The findings were that, at the start of the design process, many Micros could not articulate their customer knowledge, and had poor marketing and visual language skills, knowledge core to web design, enabling targeted communication to customers through images. Despite these gaps, most Micros still tried to lead the process. To overcome this disjoint, the majority of the designers used a knowledge transfer strategy termed in this thesis as ‘Bi-Modal Knowledge Transfer’, where the Creative was aware of the transfer but the Micro was unaware, both for drawing out customer knowledge from the Micro and for transferring visual language skills to the Micro. Two models were developed to represent this process. Two models were also created to map changes in the knowledge landscapes of customer knowledge and visual language – the Knowledge Placement Model and the Visual Language Scale. The Knowledge Placement model was used to map the placement of customer knowledge within the consciousness, extending the known Automatic-Unconscious -Conscious model, adding two more locations – Peripheral Consciousness and Occasional Consciousness. Peripheral Consciousness is where potential knowledge is held, but not used. Occasional Consciousness is where potential knowledge is held but used only for specific tasks. The Visual Language Scale was created to measure visual language ability from visually responsive, where the participant only responds personally to visual symbols, to visually multi-lingual, where the participant can use visual symbols to communicate with multiple thought-worlds. With successful Bi-Modal Knowledge Transfer, the outcome included not only an effective website but also changes in the knowledge landscape for the Micros and ongoing behavioural changes, especially in marketing. These effects were not seen in the other design projects, and only in two of the SME projects. The key factors for this difference between SMEs and Micros appeared to be an expectation of knowledge by the Creatives and failure by the SMEs to transfer knowledge within the company.

A contaminação aleatória de alimentos por micotoxinas afeta as condições de sanidade das dietas de humanos e animais. Dentre as toxinas fúngicas, deoxinivalenol (DON) se destaca pela freqüente contaminação de produtos agrícolas e alimentos e pela sua resistência a degradação pelo emprego de métodos tradicionais de processamento, o que motiva políticas de controle e a busca por técnicas de descontaminação. A descontaminação biológica empregando processsos fermentativos tem sido apontada como uma alternativa promissora, pois permite degradar micotoxinas através do sistema enzimático microbiano e melhorar características funcionais e sensoriais de matérias-primas e insumos alimentícios. Este trabalho teve por objetivo estudar condições e mecanismos de biodegradação de deoxinivalenol empregando Aspergillus oryzae e Rhizopus sp. em sistemas fermentativos submersos. Para tanto, foi necessário adequar metodologia para reação de derivação na determinação cromatográfica de DON; estudar o potencial e condições de degradação via fermentação submersa por Aspergillus oryzae e Rhizopus sp.; e avaliar a atividade de oxidoredutases e a citotoxicidade dos extratos fementados. A otimização da metodologia estabeleceu a melhor condição para a reação de derivação com 200 µL de anidrido trifluoroacético e 18 mg de bicarbonato de sódio, durante 6 minutos a 74 °C na faixa entre 7 e 21 µg de DON. A quantificação de DON residual no meio fermentado mostrou que as espécies fúngicas Rhizopus sp. e Aspergillus oryzae possuem a capacidade de degradar DON demonstrando índices médios de 87,4 e 62,4% respectivamente, principalmente quando o meio submerso foi água estéril e fermentação realizada durante 48 horas. A velocidade máxima de degradação neste intervalo foi de 10,8 e 12,4 ppb/h, observando também um aumento na atividade específica da enzima peroxidase. Os extratos dos fermentados com A. oryzae e Rhizopus sp. apresentaram efeito de inibição de proliferação celular (IC50) quando concentrados 10 vezes em 48 e 72 horas respectivamente. Os meios fermentados com Rhizopus sp. apresentaram menor efeito (1,5 vezes) quando comparado com Aspergillus oryzae.

A capacidade destes fungos degradarem compostos xenobióticos e recalcitrantes é uma característica biotecnologicamente importante tornando-os potencialmente úteis para processos de biorremediação. Gongronella sp. e Rhizopus sp. foram isolados do solo de vinhas da região do Alentejo, Portugal. Estes isolados mostraram elevada capacidade de degradarem o fungicida acilalanina metalaxil. No presente estudo, para a identificação polifásica de Gongronella sp. e Rhizopus sp., presuntivamente identificado como R. stolonifer, várias linhagens de referência da ordem Mucorales (Absidia, Circinella, Gongronella e Rhizopus) foram incluídas. A abordagem polifásica combinou a análise das macro- e micromorfologias, a análise da sequência inteira da região ITS ribossomal (i.e., ITS1/5.8S rDNA/ITS2) e ainda o uso da espectrometria de massas por Matrix Assisted Laser Desorption Ionization Time-of-Flight (MALDI-TOF).

Six pimarane-type diterpenes isolated from Viguiera arenaria Baker and two semi-synthetic derivatives were evaluated in vitro against a panel of representative microorganisms responsible for dental root canal infections. The microdilution method was used for the determination of the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) against Porphyromonas gingivalis, Prevotella nigrescens, Prevotella intermedia, Prevotella buccae, Fusobacterium nucleatum, Bacteroides fragilis, Actinomyces naeslundii, Actinomyces viscosus, Peptostreptococcus micros, Enterococcus faecalis and Aggregatibacter actinomycetemcomitans. The compounds ent-pimara-8(14), 15-dien-19-oic acid, its sodium salt and ent-8(14), 15-pimaradien-3 beta-ol were the most active, displaying MIC values ranging from 1 to 10 mu g mL(-1). The results also allow us to conclude that minor structural differences among these diterpenes significantly influence their antimicrobial activity, bringing new perspectives to the discovery of new chemicals for use as a complement to instrumental endodontic procedures.

Whole cells of hydrocarbon-degrading bacteria, isolated from polluted sediments in the Santos Estuary (Baixada Santista, Sao Paulo, Brazil), were able to catalyse oxidoreduction reactions with various substituted phenylethanols and acetophenones as substrates. A number of substituted phenylethanols were formed with high (>99 %) enantiomeric excess. The results of microbial oxidation of phenylethanols 2, 3, 5-7 by Acinetobacter sp. 6.4T and the reduction of acetophenones 1a-6a by Serratia marcescens 5.4T showed that the bacteria used as biocatalysts in this study present significant potential for exploitation in biotechnological processes. The reduction of prochiral acetophenones by Serratia marcescens 3.5T yielded optically active alcohols with 90-99 % enantiomeric excess, and Acinetobacter sp. 6.4T is a potential biocatalyst for the oxidation of alcohols.

Seven food grade commercially available lipases were immobilized by covalent binding on polysiloxane-polyvinyl alcohol (POS-PVA) hybrid composite and screened to mediate reactions of industrial interest. The synthesis of butyl butyrate and the interesterification of tripalmitin with triolein were chosen as model reactions. The highest esterification activity (240.63 mu M/g min) was achieved by Candida rugosa lipase, while the highest interesterification yield (31%, in 72 h) was achieved by lipase from Rhizopus oryzae, with the production of about 15 mM of the triglycerides C(50) and C(52). This lipase also showed a good performance in butyl butyrate synthesis, with an esterification activity of 171.14 mu M/g min. The results demonstrated the feasibility of using lipases from C. rugosa for esterification and R. oryzae lipase for both esterification and interesterification reactions.
