Chemical admixtures, when properly selected and quantified, play an important role in obtaining adequate slurry systems for quality primary cementing operations. They assure the proper operation of a well and reduce costs attributed to corrective cementing jobs. Controlling the amount lost by filtering through the slurry to permeable areas is one of the most important requirements in an operation, commonly controlled by chemical admixtures, such as carboxymethylcellulose (CMC). However, problems related to temperature, salttolerance and the secundary retarding effect are commonly reported in the literature. According to the scenario described above, the use of an aqueous dispersion of non-ionic poliurethane was proposed to control the filter loss, given its low ionic interaction with the free ions present in the slurries in humid state. Therefore, this study aims at assessing the efficiency of poliurethane to reduce filter loss in different temperature and pressure conditions as well as the synergistic effect with other admixtures. The temperatures and pressures used in laboratory tests simulate the same conditions of oil wells with depths of 500 to 1200 m. The poliurethane showed resistance to thermal degradation and stability in the presence of salts. With the increase in the concentration of the polymer there was a considerable decrease in the volume lost by filtration, and this has been effective even with the increase in temperature

O processo de desintegração de tubérculos de yacon promove a incorporação de oxigênio que favorece a oxidação de compostos fenólicos e decorrente contaminação do extrato aquoso com cores e odores objetáveis que prejudicam a sua utilização. Este estudo buscou verificar uma metodologia simplificada de extração dos carboidratos presentes nos tubérculos de yacon e efetuar uma primeira remoção de resíduos e compostos originários das cores e odores indesejáveis, utilizando processos de coagulação e precipitação, aplicando tecnologias de baixo custo. Após a etapa de trituração e remoção dos restos celulares, o extrato teve seu pH aumentado para 9,5 e a temperatura para 90ºC, seguido de sedimentação. A remoção deste precipitado foi realizada por filtração em papel após a coagulação com diferentes concentrações de sulfato de alumínio comercial. Verificou-se que a melhor concentração de coagulante no extrato foi de 100ppm, removendo 90,6% dos compostos coloridos. O balanço de massa mostrou uma recuperação de 47,5% dos sólidos totais sendo que 35,6% estão na forma de carboidratos, verificado através da concentrção de carbono orgânico. O processo de pré-tratamento químico causou hidrólise nos oligofrutanos, detectado por um aumento na concentração de açúcares redutores totais, diretamente proporcional à concentração de sulfato de alumínio utilizado no tratamento de coagulação. Análises cromatográficas indicaram o perfil dos açúcares no hidrolisado e a extensão desta hidrólise que foi considerada pequena. A metodologia aplicada mostrou-se fácil, de baixo custo, eficiente e aplicável em agroindústrias nas zonas produtoras desta espécie de raiz tropical.

The extraction, chemical and structural characterization of a wide variety of compounds derived from plants has been a major source of bioactive molecules. Several proteases have been isolated in the plant kingdom, with numerous pharmacological and biotechnological applications. Among the proteases isolated from plants, are the fibrinogenolytic, with relevant application in the treatment of disorders in the coagulation cascade, in addition to potential use as a tool in clinical laboratories. In this study, in addition to evaluating the effects of the protein extract of Cnidoscolus urens (L.) Arthur (Euphorbiaceae) in the coagulation cascade also investigates the presence of antimicrobial activity and characterizes the proteolytic activity detected in this extract, aiming to determine their potential pharmacological and biotechnological application. In this way, crude protein extracts obtained from the leaves of C. urens in Tris-HCl 0.05M, NaCl 0.15M, pH 7.5, were precipitated in different concentrations of acetone, and assessed for the presence of proteolytic activity in azocaseína and fibrinogen. The most active fraction (F1.0) in these tests was chosen for assessment of biological activity and biochemical characterization. The Aα chain and Bβ of fibrinogen were completely cleaved at a concentration of 0.18 μg/μL of protein fraction in 4 minutes. Fibrinogenolytic activity presented total inhibition in the presence of E-64 and partial in the presence of EDTA. The fraction demonstrated coagulant activity in plasm and reduced the APTT, demonstrating acting on the factors coagulation of the intrinsic pathway and common, not exerting effects on the PT. Fibrinolytic activity on plasma clot was detected only in SDS-PAGE in high concentrations of fraction, and there were no defibrinating. Although several proteases isolated from plants and venomous animals are classically toxic, the fraction F1.0 of C. urens not expressed hemorrhagic nor hemolytic activities. Fraction F1.0 also showed no antimicrobial activity. In proteolytic activity on the azocasein, the optimal pH was 5.0 and optimum temperature of 60ºC. The enzyme activity has been shown to be sensitive to the presence of salts tested, with inhibition for all compounds. The surfactant triton did not influence the enzyme activity, but the tween-20 and SDS inhibited the activity. In the presence of reducing agents increase in enzyme activity occurred, a typical feature of enzymes belonging to the class of cysteine proteases. Several bands with proteolytic activity were detected in zymogram, in the region of high-molecular-weight, which were inhibited by E-64. In this study, we found that C. urens presents in its constitution cysteine proteases with fibrinogenolytic and procoagulant activity, which may be isolated, with potential application in treatment of bleeding disorders, thrombolytic and clinical laboratory

Seed germination and seedling establishment are critical processes for commercial plantation and depend directly on reserve mobilization as a source of cellular fuels and biosynthetic precursors. In this way, we investigated the coordination among reserve mobilization, metabolite partitioning, and mobilizing enzyme activities in Moringa oleifera Lam (moringa) an oil-seeded species employed in biofuel production. Seeds were germinated under controlled conditions and seedlings were grown hydroponically at a greenhouse. Samples were harvested at 0, 4, 8, 10, 12, 16, and 20 days after imbibition (DAI). The contents of dry mass (DM), neutral lipids (NL), soluble proteins (SP), starch, total soluble sugars (TSS), non-reducing sugars (NRS), and total free amino acids (TFAA) as the activity of isocitrate lyase (ICL), acid proteases, and amylases were determined. The mobilization of storage proteins was initiated during seed germination whereas the mobilization of storage lipids and starch was triggered throughout seedling establishment although all reserves have been depleted until 20 DAI. The partitioning of DM and metabolites to the roots and the shoots was uneven during seedling establishment. Low shoot/root ratio on the basis of DM could be related to the natural occurrence of moringa in drought climates. In the roots, TSS, NRS, and TFAA were accumulated from 12 to 16 DAI and then were consumed until the end of the experiment. In the shoots, TSS and TFAA were consumed in parallel with NRS accumulation from 12 to 20 DAI. The activity of ICL, acid proteases, and amylases was coordinated with the mobilization of lipids, proteins and starch respectively. Thus, we propose that the patterns of reserve mobilization and metabolite partitioning verified in moringa seem distinct from those found to other tree species and may be involved in metabolic strategies to enable environment colonization

Dormancy is an inherent property of the seeds that define the environmental conditions in which they are able to germinate and their presence is an adaptive trait common in species inhabiting semiarid regions. Moreover, the ability of seedling establishment in these environments has been related to the size, strength and chemical characteristics of the seeds. This study investigated patterns of dormancy and germination speed in tree species of the Caatinga, exploring how the seed size influence the processes of germination, seedling size and biomass allocation. In addition, we aim to investigate the chemical characteristics of the reserves, to verify a possible relationship between nutritional content and the process of seed germination. Therefore, seeds were collected from ten species of woody Caatinga for tests of breaking dormancy, germination and biochemical characterization. Overall, the results show that the scarification treatments mechanical and chemical, and thermal shock influenced the percentage and speed of germination in 50 % of the species, suggesting that they have some level of physical dormancy in the seeds. Biochemical characterization showed the existence of large amounts of carbohydrates in the seeds of all species, low proportion of protein and low amounts of neutral lipids. Using linear regression, we demonstrated the existence of a significant relationship between seed size and the ratio of root/shoot where the largest seeds invested a greater amount of resources for shoot growth. The relationship between germination speed and non-reducing sugar content was also significant, so these compounds is related to the maintenance of physiological seed quality. These results confirm some relationships discussed in the literature for cultivated species, but can be applied to the species native to the Caatinga

O principal objetivo deste trabalho foi avaliar os efeitos residuais, por quatro anos seguidos, de diferentes tipos de resíduos, aplicados de formas e em doses distintas, sobre a qualidade tecnológica da cana-de-açúcar de quinto corte (cultivar SP 81-3250). Os tratamentos testados são combinações de dois tipos de resíduos (lodo de esgoto; vinhaça; lodo de esgoto + vinhaça) com dois modos de aplicação (ao lado da linha de cultivo e em área total) e com duas doses (100 e 200%), e um tratamento-testemunha. O delineamento experimental foi em blocos casualizados, com três repetições, totalizando 39 parcelas. As variáveis analisadas foram: percentagem de sólidos solúveis no caldo extraído (Brix % C.E.), percentagem aparente de sacarose no caldo extraído (Pol % C.E.), percentagem da fibra na cana (Fibra % cana), pureza (%), percentagem aparente de sacarose na cana (Pol % cana), percentagem de açúcares redutores na cana (AR % cana), percentagem de açúcares redutores totais na cana (ART % cana) e açúcares totais recuperáveis (ATR). Os fatores de variação testados (tipo de resíduo, modo de aplicação e doses) não promoveram alterações na qualidade e não comprometeram a valorização da cana-de-açúcar.

O presente trabalho foi conduzido com o objetivo de estudar a liberação de amônia pela cama de frangos de corte, bem como o teor de umidade e de compostos glicídicos (carboidratos redutores) e nitrogenados (nitrogênio total, nitrogênio solúvel total e nitrato) presentes na mesma, em função da densidade populacional, tempo de confinamento e sexo das aves. Realizou-se o experimento em delineamento inteiramente casualizado. A cama de frango foi submetida às densidades de 10; 14; 18 e 22 aves/m² de piso, para ambos os sexos, e analisada em três épocas diferentes (24, 38 e 42 dias de idade das aves), utilizando-se de esquema fatorial 4x2x3 (densidades x sexos x épocas). de modo geral, observou-se que o sexo não influenciou o comportamento das variáveis estudadas. O aumento da densidade e o avanço da época de amostragem induziram a aumentos da quantidade de amônia liberada, do teor de umidade e dos teores de nitrogênio total e de nitrogênio solúvel, bem como diminuição do teor de carboidratos redutores. Estes resultados sugerem a necessidade de um controle rigoroso da amônia no ar dos galpões, principalmente em densidades elevadas e no período final de criação.

Among the sugar cane potential uses include those related to the extraction of products and byproducts from the stalk. The sugarcane flowering causes morphological and physiological changes in plant and in the presence of pith, promotes changes in the stalk, reducing the raw matter quality. Most of the areas planted with sugarcane in the South-Central region of Brazil are subject to flowering, however, this feature varies depending on the variety used. There are different pith diameters related to the flowered or induced stalks, and damage to the raw matter quality are still controversial. The objective of this experiment was to evaluate the response of cane sugar varieties in relation to the flowering, pith and chemical and technological parameters, and their relationship with the pith degree. The experimental design was a completely randomized with sis treatments (varieties: IAC91-1099-4004IAC94, IAC95-5000, SP81-3250, CTC and RB855536 15) and three replications. Were evaluated the flowering, based on morphological changes in the apical bud, the pith intensity by the percentage determination of internodes occupied by whitish and spongy tissues and and pith diameter, by measuring the diameters of the changes in the internodes, expressed in percentage, in addition to chemical (juice total acidity) and technological (fiber, reducing sugars and sugar cane Pol) parameters. The flowering occurrences were accompanied by higher pith intensity. The flowering induction contributed to the lower raw matter quality, although the pith volume was lower. The pith and flowering did not affected the sugarcane sucrose content. Increases in levels of fiber, sugars and acidity were observed only in cases where the diameter of the area spongeous exceeded 50% of stalk diameter. The variety that had the best and worse response was RB855536 and CTC 15, respectively.

O objetivo deste trabalho foi avaliar, física e quimicamente, produtos minimamente processados de abacaxi-'Pérola', rodelas e metades, armazenados sob diferentes temperaturas (3ºC, 6ºC e 9ºC). Frutos selecionados, quanto ao grau de maturação e ausência de danos, foram lavados, desinfeccionados com cloro (200 mg.L-1) e armazenados a 10ºC, por 12 horas, antes do processamento. O produto minimamente processado foi embalado em bandejas de isopor recobertas com filme de PVC esticável (metades) ou bandejas de tereftalato de polietileno (rodelas) e armazenado sob refrigeraç ;ão, com avaliação a cada 3 dias, quanto à textura, coloração, pH, e conteúdos de sólidos solúveis totais, acidez total titulável, ácido ascórbico e de açúcares, solúveis e redutores. Durante o armazenamento, o produto tornou-se menos firme, e sua polpa apresentou escurecimento. Os conteúdos de açúcares solúveis e redutores e de sólidos solúveis totais não foram afetados pelo tipo de preparo, temperatura ou embalagem. Os teores de acidez total titulável aumentaram e foram influenciados pela temperatura, sendo que os mantidos a 9ºC apresentaram os maiores teores, havendo, como conseqüência, decréscimo no pH. Os produtos armazenados a 9ºC também apresentaram evolução mais rápida no escurecimento, na redução do teor de ácido ascórbico e menor vida útil (6 dias), enquanto, para os armazenados a 3ºC e 6ºC, este período foi de 9 dias. Os resultados obtidos permitiram concluir que a temperatura de armazenamento foi o fator limitante para a vida útil destes produtos.
