Objectives: To reevaluate the longevity and intraocular safety of recombinant adenovirus (rAd)-mediated gene delivery after subretinal injection, and to prolong transgene expression through the combination of 2 synergistic immunosuppressants. Methods: An rAd vector carrying green fluorescent protein (GFP) gene was delivered subretinally in the rat eye. The GFP expression was monitored in real time by fundus fluorescent photography. Intraocular safety was examined by observation of changes of retinal pigmentation, cell infiltration in virus-contacted area, immunophenotyping for CD4(+) and CD8(+) cytotoxic T lymphocytes, and CD68(+) macrophages, histologic findings, and dark-adapted electroretinography. Two synergistic immunosuppressants, cyclosporine and sirolimus, were used alone or in combination to prolong transgene expression by temporary immunosuppression. Results: The GFP expression peaked on day 4, dramatically decreased on day 10, and was not detectable on day 14. The decreased GFP expression was coincident with cell infiltration in virus-contacted area. Immunostaining showed that the infiltrating cells were CD4(+) and CD8(+) cytotoxic T lymphocytes and CD68(+) macrophages. Clumped retinal pigmentation and decreased b wave of dark-adapted electroretinogram were observed at 3 to 4 weeks after injection. Histologic examination confirmed rAd-induced retinal degeneration. Transient immunosuppression by cyclosporine and sirolimus, either alone or in combination, improved transgene expression, with the combination being the most efficient. The combined immunosuppression attenuated but did not retard the rAd-induced retinal damage. Conclusions: Transgene expression mediated by rAd after subretinal delivery is short-term and toxic to the retina. Combination of cyclosporine and sirolimus may act as an immunosuppressive adjunct to prolong rAd-mediated gene transfer. Clinical Relevance: The intraocular safety of rAd should be carefully considered before clinical trials are performed.

We compared changes in muscle fibre composition and muscle strength indices following a 10 week isokinetic resistance training programme consisting of fast (3.14 rad(.)s(-1)) or slow (0.52 rad(.)s(-1)) velocity eccentric muscle contractions. A group of 20 non-resistance trained subjects were assigned to a FAST (n = 7), SLOW (n = 6) or non-training CONTROL (n = 7) group. A unilateral training protocol targeted the elbow flexor muscle group and consisted of 24 maximal eccentric isokinetic contractions (four sets of six repetitions) performed three times a week for 10 weeks. Muscle biopsy samples were obtained from the belly of the biceps brachii. Isometric torque and concentric and eccentric torque at 0.52 and 3.14 rad(.)s(-1) were examined at 0, 5 and 10 weeks. After 10 weeks, the FAST group demonstrated significant [mean (SEM)] increases in eccentric [29.6 (6.4)%] and concentric torque [27.4 (7.3) %] at 3.14 rad(.)s(-1), isometric torque [21.3 (4.3)%] and eccentric torque [25.2 (7.2) %] at 0.52 rad(.)s(-1). The percentage of type I fibres in the FAST group decreased from [53.8 (6.6)% to 39.1 (4.4)%] while type lib fibre percentage increased from [5.8 (1.9)% to 12.9 (3.3)%; P < 0.05]. In contrast. the SLOW group did not experience significant changes in muscle fibre type or muscle torque. We conclude that neuromuscular adaptations to eccentric training stimuli may be influenced by differences in the ability to cope with chronic exposure to relatively fast and slow eccentric contraction velocities. Possible mechanisms include greater cumulative damage to contractile tissues or stress induced by slow eccentric muscle contractions.

The aim of this experiment was to establish a mouse model of irradiation-induced oral candidiasis and to explore the cellular populations and mechanisms by which the infection is cleared from the oral mucosa. BALB/c mice received irradiation to the head and neck equivalent to 800 Rad using a Cobalt 60 gamma source. Both irradiated and non-irradiated mice were infected orally with 1 X 10(8) Candida albicans yeasts. Compared with untreated controls, irradiated animals developed a more severe infection of longer duration, with hyphae penetrating the oral mucosa. Monoclonal antibody depletion of CD4(+) but not CD8(+) T cells from the systemic circulation prolonged the infection in irradiated mice, but not in controls. Supernatants of submandibular and superficial cervical lymph node cultures from irradiated animals demonstrated significantly higher titers of interleukin-12, but similar levels of interferon-gamma compared with controls. Screening for cytokine production by an RNase protection assay detected only macrophage migration inhibition factor in irradiated and non-irradiated oral tissues from day 8 onwards. The results of this study demonstrate a requirement for CD4(+) T cells in the recovery from oral candidiasis induced by head and neck irradiation in mice, and are consistent with a role for Th-1-type cytokines in host resistance.

The aim of this study was to determine the effects of 7 weeks of high- and low-velocity resistance training on strength and sprint running performance in nine male elite junior sprint runners (age 19.0 +/- 1.4 years, best 100 m times 10.89 +/- 0.21 s; mean +/- s). The athletes continued their sprint training throughout the study, but their resistance training programme was replaced by one in which the movement velocities of hip extension and flexion, knee extension and flexion and squat exercises varied according to the loads lifted (i.e. 30-50% and 70-90% of 1-RM in the high- and low-velocity training groups, respectively). There were no between-group differences in hip flexion or extension torque produced at 1.05, 4.74 or 8.42 rad . s(-1), 20 m acceleration or 20 m 'flying' running times, or 1-RM squat lift strength either before or after training. This was despite significant improvements in 20 m acceleration time (P < 0.01), squat strength (P< 0.05), isokinetic hip flexion torque at 4.74 rad . s(-1) and hip extension torque at 1.05 and 4.74 rad . s(-1) for the athletes as a whole over the training period. Although velocity-specific strength adaptations have been shown to occur rapidly in untrained and non-concurrently training individuals, the present results suggest a lack of velocity-specific performance changes in elite concurrently training sprint runners performing a combination of traditional and semi-specific resistance training exercises.

A interação entre o diâmetro da mediana volumétrico (DMV), a densidade (DEN) e a cobertura (COB) proporcionada pelas gotas produzidas na pulverização, é pouco estudada e pode ter influência nas aplicações de herbicidas. Objetivou-se estimar parâmetros técnicos da tecnologia de aplicação do glyphosate no controle de Brachiaria decumbens analisando os fatores que constituem a pulverização. Os tratamentos constaram da aplicação de glyphosate em diferentes modos de operação de um pulverizador centrífugo, visando a obtenção de parâmetros da aplicação: cinco rotações do dispositivo pulverizador (105, 157, 210, 262 e 315 rad s-1) e seis velocidades de deslocamento (0,5; 1,0; 1,5; 3,0; 4,5; e 6,0 km h-1), obtendo-se trinta parâmentros. O herbicida foi aplicado aos 20 dias após a emergência (DAE). O pulverizador foi adaptado em barra equipada com motor elétrico e comando para controle da velocidade de deslocamento. Foram realizadas avaliações visuais e calculada a porcentagem de acúmulo de massa de matéria seca das plantas em relação às testemunhas, que não receberam herbicida. O ensaio foi conduzido em delineamento inteiramente casualizado, com cinco repetições, e os dados analisados por meio de regressão múltipla. Foram obtidos controles superiores a 91%, quando o pulverizador foi utilizado de modo a proporcionar DMV de 327 mm e COB 2,51%, desde que a DEN fosse de 30 gotas cm-2. Os fatores que mais afetaram o controle de B. decumbens foram a cobertura e a densidade de gotas. A pulverização do glyphosate com menores DMV e COB e maiores DEN, proporcionou melhor controle das plantas.

We examine the instability behavior of nanocrystalline silicon (nc-Si) thin-film transistors (TFTs) in the presence of electrical and optical stress. The change in threshold voltage and sub-threshold slope is more significant under combined bias-and-light stress when compared to bias stress alone. The threshold voltage shift (Delta V-T) after 6 h of bias stress is about 7 times larger in the case with illumination than in the dark. Under bias stress alone, the primary instability mechanism is charge trapping at the semiconductor/insulator interface. In contrast, under combined bias-and-light stress, the prevailing mechanism appears to be the creation of defect states in the channel, and believed to take place in the amorphous phase, where the increase in the electron density induced by electrical bias enhances the non-radiative recombination of photo-excited electron-hole pairs. The results reported here are consistent with observations of photo-induced efficiency degradation in solar cells.

Ao longo deste trabalho será apresentada uma solução para sistemas de informação empresarial no domínio da Qualidade. Desenvolvida num ambiente real com recurso a plataforma .NET, a sua eficácia foi posta à prova em várias implementações. Várias organizações necessitam de implementar processos baseados num documento único, que deve conter o registo de toda a informação, mas que exige a intervenção de diferentes colaboradores. Esta necessidade varia muito entre organizações. Para dar resposta a este problema foi desenvolvido um sistema, denominado de Documentos Inteligentes, que, através de várias ferramentas, permite configurar processos, sem recorrer a codificação, baseados no registo de informação, workflow, controlo de acessos e alertas. É justamente a descrição dos vários componentes tecnológicos utilizados para dar resposta aos problemas, que se irá efectuar neste trabalho. Em termos de desenvolvimento de software foram abordados conceitos práticos de engenharia de domínio, RAD, arquitecturas por camadas, abstracção e workflow que possibilitaram uma maior flexibilidade do sistema desenvolvido e que conduz, simultaneamente, a uma rápida implementação de soluções de software baseadas nesta área.

To evaluate whether the intensity of the hepatic granulomatous response induced by S. mansoni eggs plays a role in drug metabolism, mice were infected with 40 cercariae and tested to assess the sodic pentobarbital induced sleeping-time. To decrease the inflammatory reaction the animals were irradiated with 400 Rad or received azathioprine, 20mg/kg, 3 times a week, for 4 weeks, respectively in or beginning in the 33th post-infection day. In infected animals receiving azathioprine the area of the hepatic granulomas was smaller and the sleeping-time was similar to that of non-infected ones (controls). In mice infected and irradiated the granuloma dimensions were similar to those of animals only infected, in these two latter groups of animals, the sleeping-time was more prolonged than that of the control animals. These results show that: 1) mice with unaltered hepatic granulomatous reaction show reduction in metabolism of sodic pentobarbital; 2) granulomatous response diminished by azathioprine does not interfere with the capacity of metabolism of the anesthetic drug.

Dipteryx odorata (Aubl.) Willd. (Fabaceae) é popularmente conhecida como cumaru. Suas sementes são utilizadas pela população, por seus efeitos terapêuticos, dados pela cumarina, hoje comercializada para distúrbios vasculares e linfáticos e pela produção de óleo. Realizou-se um estudo morfo-anatômico de sementes de seis indivíduos desta espécie através da caracterização morfológica, determinação do peso da matéria fresca e das dimensões de 100 sementes, além de cortes histológicos transversais e longitudinais, considerando-se o tegumento e o embrião. A semente é oblonga, levemente comprimida na região do hilo. O tegumento seminal apresenta cutícula delgada e lisa, macroesclereídeos, osteoesclereídeos, mesofilo interno e membrana basal. O embrião constitui-se de dois cotilédones e o eixo embrionário retilíneo, formado por plúmula, epicótilo e radícula.
