Este documento especifica o trabalho realizado no estágio, que decorreu na Colquímica, sediada em Valongo. Este trabalho teve como objetivo colaborar no desenvolvimento de uma cola com características biodegradáveis. O estágio foi orientado pela Engenheira Cristina Frutuoso, no que diz respeito à orientação por parte da Colquímica, e pelo Professor Dr. Gilberto Pinto e Alfredo Crispim, ISEP. O trabalho foi dividido em duas instâncias, sendo a primeira direcionada para a formulação de uma cola com características biodegradáveis e a segunda para a realização do teste de Sturm, um teste de biodegradabilidade capaz de quantificar a quantidade de CO2 produzido pelo processo de biodegradação. Inicialmente foram desenvolvidas e estudadas dez formulações através das variações de resinas e óleo plastificante e estudado o seu comportamento reológico relacionado à variação de viscosidade em função da temperatura. Com base nos resultados obtidos verificou-se que a melhor formulação seria a A8. Estudaram-se as propriedades mecânicas da formulação escolhida e decidiu-se que esta estaria apta a ser usada no fecho de caixas de cartão de produtos ultra congelados assim como no fecho de caixas de cartão de detergentes em pó com peso até 3 Kg. Depois de escolhida a formulação deu-se início ao teste de Sturm, teste meramente orientativo. Durante dezassete dias avaliou-se a quantidade de CO2 produzida durante o processo de biodegradação. Após avaliação dos resultados verificou-se que a taxa de biodegradação para a amostra foi de 0,055 g de CO2. Com a finalidade de confirmar os resultados obtidos no teste de Sturm foi efetuada uma réplica do ensaio, a qual se encontra a decorrer sendo os resultados facultados, posteriormente, à empresa.

A growing number of studies have identified cleaners as a group at risk for adverse health effects of the skin and the respiratory tract. Chemical substances present in cleaning products could be responsible for these effects. Currently, only limited information is available about irritant and health hazardous chemical substances found in cleaning products. We hypothesized that chemical substances present in cleaning products are known health hazardous substances that might be involved in adverse health effects of the skin and the respiratory tract. We performed a systematic review of cleaning products used in the Swiss cleaning sector. We surveyed Swiss professional cleaning companies (n = 1476) to identify the most used products (n = 105) for inclusion. Safety data sheets (SDSs) were reviewed and hazardous substances present in cleaning products were tabulated with current European and global harmonized system hazard labels. Professional cleaning products are mixtures of substances (arithmetic mean 3.5 +/- 2.8), and more than 132 different chemical substances were identified in 105 products. The main groups of chemicals were fragrances, glycol ethers, surfactants, solvents; and to a lesser extent, phosphates, salts, detergents, pH-stabilizers, acids, and bases. Up to 75% of products contained irritant (Xi), 64% harmful (Xn) and 28% corrosive (C) labeled substances. Hazards for eyes (59%) and skin (50%), and hazards by ingestion (60%) were the most reported. Cleaning products potentially give rise to simultaneous exposures to different chemical substances. As professional cleaners represent a large workforce, and cleaning products are widely used, it is a major public health issue to better understand these exposures. The list of substances provided in this study contains important information for future occupational exposure assessment studies.

Résumé La ribonucléase P (RNase P) est une ribonucléoprotéine omniprésente dans tous les règnes du vivant, elle est responsable de la maturation en 5’ des précurseurs des ARNs de transfert (ARNts) et quelques autres petits ARNs. L’enzyme est composée d'une sous unité catalytique d'ARN (ARN-P) et d'une ou de plusieurs protéines selon les espèces. Chez les eucaryotes, l’activité de la RNase P cytoplasmique est distincte de celles des organelles (mitochondrie et chloroplaste). Chez la plupart des espèces, les ARN-P sont constituées de plusieurs éléments structuraux secondaires critiques conservés au cours de l’évolution. En revanche, au niveau de la structure, une réduction forte été observé dans la plupart des mtARN-Ps. Le nombre de protéines composant la RNase P est extrêmement variable : une chez les bactéries, environ quatre chez les archéobactéries, et dix chez la forme cytoplasmique des eucaryotes. Cet aspect est peu connu pour les formes mitochondriales. Dans la plupart des cas, l’identification de la mtRNase P est le résultat de longues procédures de purification comprenant plusieurs étapes dans le but de réduire au minimum le nombre de protéines requises pour l’activité (exemple de la levure et A. nidulans). Cela mène régulièrement à la perte de l’activité et de l’intégrité des complexes ribonucléo-protéiques natifs. Dans ce travail, par l’utilisation de la technique de BN-PAGE, nous avons développé une procédure d’enrichissement de l’activité RNase P mitochondriale native, donnant un rendement raisonnable. Les fractions enrichies capables de cette activité enzymatique ont été analysées par LC/MS/MS et les résultats montrent que l’holoenzyme de la RNase P de chacune des fractions contient un nombre de protéines beaucoup plus grand que ce qui était connue. Nous suggérons une liste de protéines (principalement hypothétiques) qui accompagnent l’activité de la RNase P. IV De plus, la question de la localisation de la mtRNase P de A. nidulans a été étudiée, selon nos résultats, la majorité de la mtRNase P est attachée á la membrane interne de la mitochondrie. Sa solubilisation se fait par l’utilisation de différents types de détergent. Ces derniers permettent l’obtention d’un spectre de complexes de la RNase P de différentes tailles.

Les liposomes sont des structures sphériques formés par l'auto-assemblage de molécules amphiphiles sous forme d'une bicouche. Cette bicouche sépare le volume intérieur du liposome du milieu extérieur, de la même manière que les membranes cellulaires. Les liposomes sont donc des modèles de membranes cellulaires et sont formulés pour étudier les processus biologiques qui font intervenir la membrane (transport de molécules à travers la membrane, effets des charges en surface, interactions entre la matrice lipidique et d'autres molécules, etc.). Parce qu'ils peuvent encapsuler une solution aqueuse en leur volume intérieur, ils sont aussi utilisés aujourd'hui comme nanovecteurs de principes actifs. Nous avons formulé des liposomes non-phospholipidiques riches en stérol que nous avons appelés stérosomes. Ces stérosomes sont composés d'environ 30 % d'amphiphiles monoalkylés et d'environ 70 % de stérols (cholestérol, Chol, et/ou sulfate de cholestérol, Schol). Quand certaines conditions sont respectées, ces mélanges sont capables de former une phase liquide ordonnée (Lo) pour donner, par extrusion, des vésicules unilamellaires. Certaines de ces nouvelles formulations ont été fonctionnalisées de manière à libérer leur contenu en réponse à un stimulus externe. En incorporant des acides gras dérivés de l’acide palmitique possédant différents pKa, nous avons pu contrôler le pH auquel la libération débute. Un modèle mathématique a été proposé afin de cerner les paramètres régissant leur comportement de libération. En incorporant un amphiphile sensible à la lumière (un dérivé de l’azobenzène), les liposomes formés semblent répondre à une radiation lumineuse. Pour ce système, il serait probablement nécessaire de tracer le diagramme de phase du mélange afin de contrôler la photo-libération de l’agent encapsulé. Nous avons aussi formulé des liposomes contenant un amphiphile cationique (le chlorure de cétylpyridinium). En tant que nanovecteurs, ces stérosomes montrent un potentiel intéressant pour la libération passive ou contrôlée de principes actifs. Pour ces systèmes, nous avons développé un modèle pour déterminer l’orientation des différentes molécules dans la bicouche. La formation de ces nouveaux systèmes a aussi apporté de nouvelles connaissances dans le domaine des interactions détergents-lipides. Aux nombreux effets du cholestérol (Chol) sur les systèmes biologiques, il faut ajouter maintenant que les stérols sont aussi capables de forcer les amphiphiles monoalkylés à former des bicouches. Cette nouvelle propriété peut avoir des répercussions sur notre compréhension du fonctionnement des systèmes biologiques. Enfin, les amphiphiles monoalkylés peuvent interagir avec la membrane et avoir des répercussions importantes sur son fonctionnement. Par exemple, l'effet antibactérien de détergents est supposé être dû à leur insertion dans la membrane. Cette insertion est régie par l'affinité existant entre le détergent et cette dernière. Dans ce cadre, nous avons voulu développer une nouvelle méthode permettant d'étudier ces affinités. Nous avons choisi la spectroscopie Raman exaltée de surface (SERS) pour sa sensibilité. Les hypothèses permettant de déterminer cette constante d’affinité se basent sur l’incapacité du détergent à exalter le signal SERS lorsque le détergent est inséré dans la membrane. Les résultats ont été comparés à ceux obtenus par titration calorimétrique isotherme (ITC). Les résultats ont montré des différences. Ces différences ont été discutées.

The present work is a base line attempt to investigate and assess the toxicities of three surfactants viz. anionic sodium dodecyl sulfate (SDS), non ionic Triton X-1OO (TX-IOO) and cationic cetyl trimethyl ammonium bromide (CTAB). These compounds represent simple members of the often neglected group of aquatic pollutants i.e. the anionic alkyl sulfates, non ionics and the cationics. These compounds are widely used In plastic industry, pesticide/herbicide formulations, detergents, oil spill dispersants, molluscicides etc. The test organisms selected for the present study are the cyanobacterium Synechocystis salina Wislouch representing a primary producer in the marine environment and a fresh water adapted euryhaline teleost Oreochromis mossambicus (peters) at the consumer level of the ecological pyramid. The fish species, though not indigenous to our country, is now found ubiquitously in fresh water systems and estuaries. Also it is highly resistant to pollutants and has been suggested as an indicator of pollution in tropical region .

The thesis presents a detailed account of the alkaline protease produced by Vibrio sp.(V26) a mangrove isolate,and the application of this enzyme in different fields.The protease producer strain was identified on the basis of biochemical characteristice,putative virulence traits and 16S rRNA gene sequencing.The purification and characterization of the protease has been carried out. Along with this, an attempt has been made to identifiy the protease gene. The physical parameters as well as the media components influencing protease production were optimized using Response Surfce Methodology(RSM).The scale up of the application of the protease from Vibrio sp.(V26) in the dissociation of cells in animal cell culture,in the recovery of silver from used X-ray films as well as an ingredient in commercial detergents were investigated.

Bacillus smithii BTMS 11, isolated from marine sediment, produced alkaline and thermostable lipase. The enzyme was purified to homogeneity by ammonium sulfate precipitation and ion exchange chromatography which resulted in 0.51 % final yield and a 4.33 fold of purification. The purified enzyme was found to have a specific activity of 360 IU/mg protein. SDS-PAGE analyses, under non-reducing and reducing conditions, yielded a single band of 45 kDa indicating the single polypeptide nature of the enzyme and zymogram analysis using methylumbelliferyl butyrate as substrate confirmed the lipolytic activity of the protein band. The enzyme was found to have 50 C and pH 8.0 as optimum conditions for maximal activity. However, the enzyme was active over wide range of temperatures (30–80 C) and pH (7.0–10.0). Effect of a number of metal salts, solvents, surfactants, and other typical enzyme inhibitors on lipase activity was studied to determine the novel characteristics of the enzyme. More than 90 % of the enzyme activity was observed even after 3 h of incubation in the presence of commercial detergents Surf, Sunlight, Ariel, Henko, Tide and Ujala indicating the detergent compatibility of B. smithii lipase. The enzyme was also found to be efficient in stain removal from cotton cloths. Further it was observed that the enzyme could catalyse ester synthesis between fatty acids of varying carbon chain lengths and methanol with high preference for medium to long chain fatty acids showing 70 % of esterification. Results of the study indicated scope for application of this marine bacterial lipase in various industries

Aquesta activitat terciari d’ús comercial és classifica com un comerç al detall amb predomini de productes alimentaris, begudes i una altra àmplia gamma de mercaderies (com són els cosmètics, detergents, productes de papereria, etc.)

Jabones El Tigre y Roca S.A. es una compañía productora y comercializadora de jabones en barra y detergentes para el lavado de ropa y de jabones en pasta para la limpieza de la loza. Con el presente trabajo enmarcado dentro de las etapas descriptiva, de diagnóstico y prescriptiva, se pretende analizar y evaluar la situación actual de la empresa y a su vez dar alternativas de exportación para el producto detergente presentando recomendaciones que pueden orientar futuras decisiones.

Objectives: AcrA can function as the periplasmic adaptor protein (PAP) in several RND tripartite efflux pumps, of which AcrAB-TolC is considered the most important. This system confers innate multiple antibiotic resistance. Disruption of acrB or tolC impairs the ability of Salmonella Typhimurium to colonize and persist in the host. The aim of this study was to investigate the role of AcrA alone in multidrug resistance and pathogenicity. Methods: The acrA gene was inactivated in Salmonella Typhimurium SL1344 by insertion of the aph gene and this mutant complemented with pWKS30acrA. The antimicrobial susceptibility of the mutant to six antibiotics as well as various dyes and detergents was determined. In addition, efflux activity was quantified. The ability of the mutant to adhere to, and invade, tissue culture cells in vitro was measured. Results: Following disruption of acrA, RT-PCR and western blotting confirmed that acrB/AcrB was still expressed when acrA was disrupted. The acrA mutant was hypersusceptible to antibiotics, dyes and detergents. In some cases, lower MICs were seen than for the acrB or tolC mutants. Efflux of the fluorescent dye Hoechst H33342 was less than in wild-type following disruption of acrA. acrA was also required for adherence to, and invasion of, tissue culture cells. Conclusions: Inactivation of acrA conferred a phenotype distinct to that of acrB::aph and tolC::aph. These data indicate a role for AcrA distinct to that of other protein partners in both efflux of substrates and virulence.

Aims: In Escherichia coli, increased expression of efflux pumps and/or decreased expression of porins can confer multiple antibiotic resistance (MAR), causing resistance to at least three unrelated classes of antibiotics, detergents and dyes. It was hypothesized that in Campylobacter jejuni, the efflux systems CmeABC, CmeDEF and the major outer membrane porin protein, MOMP (encoded by porA) could confer MAR. Methods: The expression of cmeB, cmeF and porA in 32 MAR C. jejuni isolated from humans or poultry was determined by comparative (C)-reverse transcriptase (RT)-PCR and denaturing DHPLC. A further 13 ethidium bromide-resistant isolates and three control strains were also investigated. Accumulation of ciprofloxacin carbonyl cyanide-m-chlorophenyl hydrazone (CCCP) was also determined for all strains. Results: Although resistance to ethidium bromide has been associated with MAR, expression of all three genes was similar in the ethidium bromide-resistant isolates. These data indicate that CmeB, CmeF and MOMP play no role in resistance to this agent in C. jejuni. Six MAR isolates over-expressed cmeB, 3/32 over-expressed cmeB and cmeF. No isolates over-expressed cmeF alone. Expression of porA was similar in all isolates. All nine isolates that over-expressed cmeB contained a mutation in cmeR, substituting glycine 86 with alanine. All cmeB over-expressing isolates also accumulated low concentrations of ciprofloxacin, which were restored to wild-type levels in the presence of CCCP. Conclusions: These data indicate that over-expression of cmeB is associated with MAR in isolates of C. jejuni. However, as cmeB was over-expressed by only one-third (9/32) of MAR isolates, these data also indicate other mechanisms of MAR in C. jejuni.
The AcrAB-TolC efflux system of Salmonella enterica serovar Typhimurium plays a role in pathogenesis

The ability of an isogenic set of mutants of Salmonella enterica serovar Typhimurium L354 (SL1344) with defined deletions in genes encoding components of tripartite efflux pumps, including acrB, acrD, acrF and tolC, to colonize chickens was determined in competition with L354. In addition, the ability of L354 and each mutant to adhere to, and invade, human embryonic intestine cells and mouse monocyte macrophages was determined in vitro. The tolC and acrB knockout mutants were hyper-susceptible to a range of antibiotics, dyes and detergents; the tolC mutant was also more susceptible to acid pH and bile and grew more slowly than L354. Complementation of either gene ablated the phenotype. The tolC mutant poorly adhered to both cell types in vitro and was unable to invade macrophages. The acrB mutant adhered, but did not invade macrophages. In vivo, both the acrB mutant and the tolC mutant colonized poorly and did not persist in the avian gut, whereas the acrD and acrF mutant colonized and persisted as well as L354. These data indicate that the AcrAB-TolC system is important for the colonization of chickens by S. Typhimurium and that this system has a role in mediating adherence and uptake into target host cells.

The interaction between angiotensin II (AII, DRVYIHPF) and its analogs carrying 2,2,6,6-tetramethylpiperidine-1-oxyl-4-amino-4-carboxylic acid (TOAC) and detergents-negatively charged sodium dodecyl sulfate (SDS) and zwitterionic N-hexadecyl-N,N-dimethyl-3-ammonio-1-propanesulfonate (HPS)-was examined by means of EPR, CD, and fluorescence. EPR spectra of partially active TOAC(1)-AII and inactive TOAC(3)-AII in aqueous solution indicated fast tumbling, the freedom of motion being greater at the N-terminus. Line broadening occurred upon interaction with micelles. Below SDS critical micelle concentration, broader lines indicated complex formation with tighter molecular packing than in micelles. Small changes in hyperfine splittings evinced TOAC location at the micelle-water interface. The interaction with anionic micelles was more effective than with zwitterionic micelles. Peptide-micelle interaction caused fluorescence increase. The TOAC-promoted intramolecular fluorescence quenching was more, pronounced for TOAC(3)-AII because of the proximity between the nitroxide and Tyr(4). CD spectra showed that although both AII and TOAC(1)-AII presented flexible conformations in water, TOAC(3)-AII displayed conformational restriction because of the TOAC-imposed bend (Schreier et al., Biopolymers 2004, 74, 389). In HPS, conformational changes were observed for the labeled peptides at neutral and basic pH. In SDS, all peptides underwent pH-dependent conformational changes. Although the spectra suggested similar folds for All and TOAC(1)-AII, different conformations were acquired by TOAC(3)-AII. The membrane environment has been hypothesized to shift conformational equilibria so as to stabilize the receptor-bound conformation of ligands. The fact that TOAC(3)-AII is unable to acquire conformations similar to those of native AII and partially active TOAC(1)-AII is probably the explanation for its lack of biological activity.
