A commercial casein hydrolysate was microencapsulated in liposomes produced with non-purified soy lecithin, cryoprotected with two different disaccharides and lyophilized. The encapsulation efficiency of casein hydrolysate ranged from 30 to 40%. The powders were analyzed by differential scanning calorimetry (DSC), scanning electron micrography (SEM), infrared spectroscopy (FTIR) and wide angle X-ray diffraction (WAXD). DSC data revealed the presence of an exothermal transition in empty lyophilized liposomes, which was ascribed to the presence of a quasicrystalline lamellar phase (intermediary characteristics between the L-beta and L-c phases). The addition of peptides to the liposomal system caused the disappearance of this exothermic phenomenon, as they were located in the polar headgroup portion of the bilayer, causing disorder and preventing the formation of the quasicrystalline phase. Infrared data indicated the presence of the peptides in the lyophilized formulations and showed that the cryoprotectants interacted effectively with the polar heads of phospholipids in the bilayer.

Neben Tomatensaft wurde eine Vielzahl von Säften und Blattextrakten als Medienzusätze auf Wachstumsförderung bei 30 verschiedenen Oenococcus oeni-Stämmen getestet. Es zeigte sich eine breite Wachstumsförderung bei allen Zusätzen mit Ausnahme von Zitronensaft, sodass die Wachstumsfaktoren keine tomatenspezifischen Inhaltsstoffe sein können und eher ubiquitär in der Pflanzenwelt vorkommen. Das Ausmaß der Wachstumsförderung war stammabhängig sehr unterschiedlich und Tomatensaft stellte keineswegs für alle Stämme den optimalen Medienzusatz dar. Durch Schälen der Früchte war eine für die Analytik hilfreiche Abtrennung schalenspezifischer Inhaltsstoffe möglich, wobei auch die Schalenextrakte großes Potential für die Suche nach Wachstumsfaktoren offenbarten und die Wichtigkeit einer Auftrennung der Frucht in die verschiedenen Fruchtbereiche betonte. Aus Tomatensaft konnte analytisch der anorganische Wachstumsfaktor Mangan identifiziert werden. Die größten Zelldichten der Oenokokken-Stämme wurden hierbei bei 67 µM und 34 mM Manganzusatz erreicht. Bei 13 von 20 getesteten Oenokokkenstämmen konnte bei Zusatz von 34 mM Mangan der Tomatensaft ersetzt werden, bei 4 Stämmen (z. B. Stamm B2) fehlten jedoch noch weitere Wachstumsfaktoren und bei 3 Stämmen (z. B. Stamm B120) kam es zu einem verfrühten Absterben. Da weitere Mineralstoffe sowie veraschte Säfte und Blattextrakte keinen positiven Einfluß auf die Oenokokken-Zelldichte hatten, wurde mittels semipräparativer HPLC nach zusätzlichen organischen Wachstumsfaktoren für den Stamm B2 gesucht. Hierzu wurde der nachfolgende Wachstums-Assay miniaturisiert und erfolgreich auf Microtiterplatten etabliert. Es gelang die Isolierung und Identifizierung eines wachstumsfördernden Trisaccharides aus Mangoschalen-Extrakt, das aus den Zuckern Glucose, Rhamnose und Arabinose bestand. Von den monomeren Zuckern erhöhte lediglich die Arabinose die Zelldichte, das Optimum lag bei 1,5 g/l. Auch aus Zitronenmesokarp-Extrakt war die Isolierung eines wachstumsfördernden arabinosehaltigen Disaccharides möglich, die Menge reichte jedoch noch nicht für eine genaue Identifizierung aus. Desweiteren erwies sich 1,5 g/l Cystein als wachstumsstimulierend. Ein Zusatz aller gefundenen Wachstumsfaktoren (34 mM Mangan, 1,5 g/l Arabinose und 1,5 g/l Cystein) ersetzte den Tomatensaft bei weiteren Oenokokken-Stämmen (z.B. Stamm B120) komplett, wobei bei allen Stämmen sogar eine schnellere Anzucht erfolgte. Neben dem Tomatensaft war auch der Zusatz von Hefeextrakt zum Grundmedium nicht mehr nötig, sodass ein neues vereinfachtes Medium für die Anzucht von Oenokokken mit komplexen Nährstoffansprüchen vorgeschlagen werden konnte. Lediglich beim Stamm B2 zeigte sich noch ein OD-Unterschied von 0,2 in der stationären Phase, der nach Adsorptionsversuchen an Polyvinylpolypyrrolidon auf noch unidentifizierte Polyphenole im Tomatensaft zurückzuführen ist. Aus grünem Tee erwies sich das Polyphenol Epigallocatechingallat (EGCG) konzentrationsabhängig sowohl als Hemmstoff (>550 mg/l EGCG) als auch Wachstumsfaktor (400-500 mg/l EGCG) für den Oenokokken-Stamm B2. Der hemmende als auch der fördernde Einfluss auf das Wachstum wurde mittels Sytox/DAPI-Färbung bestätigt. Der sogenannte „Tomatensaft-Faktor“ ist also nicht eine spezielle Substanz, sondern das synergistische Zusammenwirken mehrerer einfacher Substanzen wie Mineralstoffe, Aminosäuren, Kohlenhydrate und Polyphenole. Auch sind die Oenokokken-Stämme bezüglich ihres Nährstoffbedarfes sehr unterschiedlich, sodass für jeden Stamm einzeln das optimale Substratspektrum ermittelt werden muss.

Krebserkrankungen gehen oft mit der Überexpression von mucinartigen Glycoproteinen auf der Zelloberfläche einher. In vielen Krebserkrankungen wird aufgrund der fehlerhaften Expression verschiedener Glycosyltransferasen das transmembranständige Glycoprotein MUC1, mit verkürzten Glycanstrukturen, überexprimiert. Das Auftreten der verschiedenen tumor-assoziierten Antigene (TACA) korreliert meist mit dem Fortschreiten des Krebs und der Metastasierung. Daher stellen TACAs interessante Zielmoleküle für die Entwicklung einer aktiven Tumorimmuntherapie zur spezifischen Behandlung von Adenokarzinomen dar. In dieser Arbeit galt das Interesse dem epithelialen Mucin MUC1, auf Basis dessen ein synthetischer Zugang zu einheitlichen Antitumorvakzinen, welche aus mucinanalogen Glyco-peptid¬konjugaten des MUC1 und Carrierproteinen bestehen, hergestellt werden sollten.rnUm eine tumorspezifische Immunantwort zu erhalten, müssen die selbst schwach immunogenen MUC1-Antigene über einen nicht-immunogenen Spacer mit einem geeigneten Trägerprotein, wie Tetanus Toxoid oder Rinderserumalbumin (BSA), verbunden werden. rnDa ein Einsatz von Glycokonjugaten in Impfstoffen durch die metabolische Labilität der O-glycosidischen Bindungen eingeschränkt ist, wurden hierzu erstmals fluorierte Vetreter von MUC1-analogen Glycopeptiden verwendet, in denen das Kohlenhydrat-Epitop durch den strategischen Einbau von Fluor¬atomen gegenüber einem raschen Abbau durch Glycosidasen geschützt werden soll. Dazu wurden auf Basis des literaturbekannten Thomsen-Friedenreich-Antigens Synthesestrategien zur Herstellung eines 2’F- und eines 2’,6’-bisfluorierten-Analogons erarbeitet. rnSchlüsselschritte in der Synthese stellten neben der elektrophilen Fluorierung eines Galactalvorläufers auch die -selektive 3-Galactosylierung des TN-Antigen-Bausteins zum 2’F- und 2’,6’-bisfluorierten-Analogons des TF-Disaccharids dar. Durch entsprechende Schutzgruppentransformationen wurden die beiden Derivate in entsprechende Glycosyl¬amino-säure-Bausteine für die Festphasensynthese überführt.rnNeben den beiden Analoga des TF-Antigens wurde auch erstmals ein 2F-Analogon des 2,6-Sialyl-T-Antigens hergestellt. Dazu wurde der entsprechende 2’F-TF-Baustein mit Sialinsäure-xanthogenat nach bereits bekannten Syntheseprotokollen umgesetzt. Aufgrund von Substanzmangel konnte die Verbindung nicht zur Synthese eines MUC1-Glycopeptid-Analogons herangezogen werden.rnDer Einbau der hergestellten Glycosylaminosäure-Bausteine erfolgte in die aus 20 Amino-säuren bestehende vollständige Wiederholungseinheit aus der tandem repeat-Sequenz des MUC1, wobei die entsprechenden Glycanseitenketten stets in Position 6 eingeführt wurden. Um die erhaltenen Glycopeptide für immunologische Studien an Carrier-Proteine anbinden zu können und so ggf. zu funktionsfähigen Impfstoff-Konjugaten zu gelangen, wurden diese stets N-terminal mit einem nicht-immunogenen Triethylenglycol-Spacer verknüpft. Die anschließende Funktionalisierung mit Quadratsäurediethylester erlaubte die spätere chemoselektive Konjugation an Trägerproteine, wie Tetanus Toxoid oder BSA.rnIn ersten immunologischen Bindungsstudien wurden die synthetisierten BSA-Glycopeptid-Konjugate mit Serum-Antikörpern aus Vakzinierungsstudien von MUC1-Tetanus Toxoid-Konjugaten, die (i) eine natürliche TF-Antigenstruktur und (ii) ein entsprechendes TF-Antigenderivat mit Fluorsubstituenten an C-6 des Galactosamin-Bausteins und C-6’ des Galactoserests tragen, untersucht.rn

The goals of this article are to (1) provide further validation of the Glycam06 force field, specifically for its use in implicit solvent molecular dynamic (MD) simulations, and (2) to present the extension of G.N. Ramachandran's idea of plotting amino acid phi and psi angles to the glycosidic phi, psi, and omega angles formed between carbohydrates. As in traditional Ramachandran plots, these carbohydrate Ramachandran-type (carb-Rama) plots reveal the coupling between the glycosidic angles by displaying the allowed and disallowed conformational space. Considering two-bond glycosidic linkages, there are 18 possible conformational regions that can be defined by (α, ϕ, ψ) and (β, ϕ, ψ), whereas for three-bond linkages, there are 54 possible regions that can be defined by (α, ϕ, ψ, ω) and (β, ϕ, ψ, ω). Illustrating these ideas are molecular dynamic simulations on an implicitly hydrated oligosaccharide (700 ns) and its eight constituent disaccharides (50 ns/disaccharide). For each linkage, we compare and contrast the oligosaccharide and respective disaccharide carb-Rama plots, validate the simulations and the Glycam06 force field through comparison to experimental data, and discuss the general trends observed in the plots.

Disaccharide intolerance I or congenital sucrase-isomaltase deficiency (CSID) is a disorder leading to maldigestion of disaccharides, which is autosomal recessively inherited. Here we analyzed the sucrase-isomaltase (SI) gene from 11 patients of Hungarian origin with congenital sucrase-isomaltase deficiency. Variants in the SI gene had previously been described in CSID patients, which cause amino acid exchanges that affect the transport, the processing, or the function of the SI protein. None of our patients had known mutations for CSID. Our analyses revealed 43 SI variants in total, 15 within exons and one at a splice site. Eight of the exonic mutations lead to amino acid exchanges, causing hypomorph or null alleles. One new variation affects a splice site, which is also predicted to result in a null allele. All potential pathological alterations were present on one allele only. In six out of the 11 patients the phenotype of CSID could be explained by compound heterozygosity.

BACKGROUND: Contagious bovine pleuropneumonia (CBPP) caused by Mycoplasma mycoides subsp. mycoides small-colony type (SC) is among the most serious threats for livestock producers in Africa. Glycerol metabolism-associated H2O2 production seems to play a crucial role in virulence of this mycoplasma. A wide number of attenuated strains of M. mycoides subsp. mycoides SC are currently used in Africa as live vaccines. Glycerol metabolism is not affected in these vaccine strains and therefore it does not seem to be the determinant of their attenuation. A non-synonymous single nucleotide polymorphism (SNP) in the bgl gene coding for the 6-phospho-beta-glucosidase (Bgl) has been described recently. The SNP differentiates virulent African strains isolated from outbreaks with severe CBPP, which express the Bgl isoform Val204, from strains to be considered less virulent isolated from CBPP outbreaks with low mortality and vaccine strains, which express the Bgl isoform Ala204. RESULTS: Strains of M. mycoides subsp. mycoides SC considered virulent and possessing the Bgl isoform Val204, but not strains with the Bgl isoform Ala204, do trigger elevated levels of damage to embryonic bovine lung (EBL) cells upon incubation with the disaccharides (i.e., beta-D-glucosides) sucrose and lactose. However, strains expressing the Bgl isoform Val204 show a lower hydrolysing activity on the chromogenic substrate p-nitrophenyl-beta-D-glucopyranoside (pNPbG) when compared to strains that possess the Bgl isoform Ala204. Defective activity of Bgl in M. mycoides subsp. mycoides SC does not lead to H2O2 production. Rather, the viability during addition of beta-D-glucosides in medium-free buffers is higher for strains harbouring the Bgl isoform Val204 than for those with the isoform Ala204. CONCLUSION: Our results indicate that the studied SNP in the bgl gene is one possible cause of the difference in bacterial virulence among strains of M. mycoides subsp. mycoides SC. Bgl does not act as a direct virulence factor, but strains possessing the Bgl isoform Val204 with low hydrolysing activity are more prone to survive in environments that contain high levels of beta-D-glucosides, thus contributing in some extent to mycoplasmaemia.

I solved equations that describe coupled hydrolysis in and absorption from a continuously stirred tank reactor (CSTR), a plug flow reactor (PFR), and a batch reactor (BR) for the rate of ingestion and/or the throughput time that maximizes the rate of absorption (=gross rate of gain from digestion). Predictions are that foods requiring a single hydrolytic step (e.g., disaccharides) yield ingestion rates that vary inversely with the concentration of food substrate ingested, whereas foods that require multiple hydrolytic and absorptive reactions proceeding in parallel (e.g., proteins) yield maximal ingestion rates at intermediate substrate concentrations. Counterintuitively, then, animals acting to maximize their absorption rates should show compensatory ingestion (more rapid feeding on food of lower concentration), except for the lower range of diet quality fur complex diets and except for animals that show purely linear (passive) uptake. At their respective maxima in absorption rates, the PFR and BR yield only modestly higher rates of gain than the CSTR but do so at substantially lower rates of ingestion. All three ideal reactors show milder than linear reduction in rate of absorption when throughput or holding time in the gut is increased (e.g., by scarcity or predation hazard); higher efficiency of hydrolysis and extraction offset lower intake. Hence adding feeding costs and hazards of predation is likely to slow ingestion rates and raise absorption efficiencies substantially over the cost-free optima found here.

Difficulties in determining composition and sequence of glycosaminoglycans, such as those related to heparin, have limited the investigation of these biologically important molecules. Here, we report methodology, based on matrix-assisted laser desorption ionization MS and capillary electrophoresis, to follow the time course of the enzymatic degradation of heparin-like glycosaminoglycans through the intermediate stages to the end products. MS allows the determination of the molecular weights of the sulfated carbohydrate intermediates and their approximate relative abundances at different time points of the experiment. Capillary electrophoresis subsequently is used to follow more accurately the abundance of the components and also to measure sulfated disaccharides for which MS is not well applicable. For those substrates that produce identical or isomeric intermediates, the reducing end of the carbohydrate chain was converted to the semicarbazone. This conversion increases the molecular weight of all products retaining the reducing terminus by the “mass tag” (in this case 56 Da) and thus distinguishes them from other products. A few picomoles of heparin-derived, sulfated hexa- to decasaccharides of known structure were subjected to heparinase I digestion and analyzed. The results indicate that the enzyme acts primarily exolytically and in a processive mode. The methodology described should be equally useful for other enzymes, including those modified by site-directed mutagenesis, and may lead to the development of an approach to the sequencing of complex glycosaminoglycans.

Inhibitors of glycosylation provide a tool for studying the biology of glycoconjugates. One class of inhibitors consists of glycosides that block glycoconjugate synthesis by acting as primers of free oligosaccharide chains. A typical primer contains one sugar linked to a hydrophobic aglycone. In this report, we describe a way to use disaccharides as primers. Chinese hamster ovary cells readily take up glycosides containing a pentose linked to naphthol, but they take up hexosides less efficiently and disaccharides not at all. Linking phenanthrol to a hexose improves its uptake dramatically but has no effect on disaccharides. To circumvent this problem, analogs of Xyl beta 1-->6Gal beta-O-2-naphthol were tested as primers of glycosaminoglycan chains. The unmodified disaccharide did not prime, but methylated derivatives had activity in the order Xyl beta 1-->6Gal(Me)3-beta-O-2-naphthol > Xyl beta 1-->6Gal (Me)2 beta-O-2-naphthol >> Xyl beta 1-->6Gal(Me)beta-O-2-naphthol. Acetylated Xyl beta 1-->6Gal beta-O-2-naphthol also primed glycosaminoglycans efficiently, suggesting that the terminal xylose residue was exposed by removing the acetyl groups. The general utility of using acetyl groups to create disaccharide primers was shown by the priming of oligosaccharides on peracetylated Gal beta 1-->4GlcNAc beta-O-naphthalenemethanol. This disaccharide inhibited sialyl Lewis X expression on HL-60 cells.

Mannose-binding type 1 pili are important virulence factors for the establishment of Escherichia coli urinary tract infections (UTIs). These infections are initiated by adhesion of uropathogenic E. coli to uroplakin receptors in the uroepithelium via the FimH adhesin located at the tips of type 1 pili. Blocking of bacterial adhesion is able to prevent infection. Here, we provide for the first time binding data of the molecular events underlying type 1 fimbrial adherence, by crystallographic analyses of the FimH receptor binding domains from a uropathogenic and a K-12 strain, and affinity measurements with mannose, common mono- and disaccharides, and a series of alkyl and aryl mannosides. Our results illustrate that the lectin domain of the FimH adhesin is a stable and functional entity and that an exogenous butyl alpha- D-mannoside, bound in the crystal structures, exhibits a significantly better affinity for FimH (K-d = 0.15 muM) than mannose (K-d = 2.3 muM). Exploration of the binding affinities of alpha-D-mannosides with longer alkyl tails revealed affinities up to 5 nM. Aryl mannosides and fructose can also bind with high affinities to the FimH lectin domain, with a 100-fold improvement and 15-fold reduction in affinity, respectively, compared with mannose. Taken together, these relative FimH affinities correlate exceptionally well with the relative concentrations of the same glycans needed for the inhibition of adherence of type 1 piliated E. coli. We foresee that our findings will spark new ideas and initiatives for the development of UTI vaccines and anti-adhesive drugs to prevent anticipated and recurrent UTIs.

Oligosaccharide synthesis using aminosugars requires the presence of a suitable amino protecting group. A number of protecting groups are currently used, and while many display favorable properties, most agents available still suffer from certain disadvantages. This report details the use of a hydrazine labile aminosugar protecting group, N -[1-(4,4-dimethyl-2,6-dioxocyclohex-1-ylidene)ethyl] (Dde), which can be introduced and removed in a facile and cost-effective manner.

Carbohydrates are a major source of energy in the diet. Classified according to their chemistry, carbohydrates can be divided into sugars (monosaccharides and disaccharides), polyols, oligosaccharides (malto-oligosaccharides and non-digestible oligosaccharides) and polysaccharides (starch and non-starch polysaccharides). However, this classification does not allow a simple translation into nutritional effects since each class of carbohydrates has overlapping physiological properties and effects on health. Carbohydrates can also be classified according to their digestion and absorption in the human small intestine. Digestible carbohydrates are absorbed and digested in the small intestine; non-digestible carbohydrates are resistant to hydrolysis in the small intestine and reach the large intestine where they are at least partially fermented by the commensal bacteria present in the colon. There is no universal definition of the term ‘dietary fibre’; broadly speaking, it refers to some or all of the constituents of non-digestible carbohydrates and may also include other quantitatively minor components (e.g.lignin) that are associated with non-digestible carbohydrates in plant cell walls.
