La Enfermedad de Chagas es una enfermedad causada por un parásito llamado Trypanosoma cruzi, que puede vivir en la sangre y en los tejidos de personas y animales y en el tubo digestivo de unos insectos conocidos como triatóminos o chinches. El OBJETIVO de esta investigación fue detectar casos de la Enfermedad de Chagas en la población de 5 a 15 años de edad en el caserío Los Orellana y determinar la presencia de Triatoma dimidiata en el ámbito familiar de la población en estudio. METODOLOGÍA: El estudio fue prospectivo, transversal, descriptivo, de campo y de laboratorio. La población estuvo constituida por 66 menores en edades de 5 a 15 años que cumplieron con los criterios de inclusión. Así como también la búsqueda de Triatoma dimidiata que se llevó a cabo en las viviendas donde se buscó el vector. Se llenó una cédula de entrevista con respuestas de los representantes de cada hogar inspeccionado la cual constaba de 8 preguntas referentes a la investigación. A los vectores capturados se les realizó la técnica de compresión abdominal en busca del parásito Trypanosoma cruzi en la deyecciones de estos. Posteriormente se procedió a tomar una muestra de sangre a los menores que participaron en el estudio para realizar las pruebas parasitológicas que fueron el frotis de sangre periférica, gota gruesa y concentrado de Strout en busca del parásito en la sangre y las pruebas serológicas de ELISA Tercera y Cuarta generación e Inmunofluorescencia Indirecta para confirmar los casos que resultaron reactivos. RESULTADOS OBTENIDOS: El porcentaje de casas infestadas con Triatoma dimidiata fue de 9 (21.43 %) habiendo capturado 28 triatóminos los cuales 3 presentaron el parásito que corresponde al 10.71 %. En los métodos parasitológicos directos (Frotis de sangre periférica, Gota gruesa y Concentrado de Strout) no se observó la presencia del parásito; El método serológico indirecto ELISA de Tercera generación dio como resultado 3 casos reactivos (4.5%) procediéndose a realizar su respectiva confirmación a través de ELISA de Cuarta generación e Inmunofluorescencia Indirecta, detectándose anticuerpos anti - Trypanosoma cruzi tipo IgG en las tres muestras. CONCLUSIÓN: Se encontró en un 21.43 % (9) de las casas la presencia del vector transmisor de la Enfermedad de Chagas los cuales se analizaron encontrándose en el 10.71 % (3) la presencia del parásito. Finalmente se encontró el 4.5 % (3) de los menores con la enfermedad en su fase crónica.

Reprogramming of somatic cells to pluripotency promises to boost cellular therapy. Most instances of direct reprogramming have been achieved by forced expression of defined exogenous factors using multiple viral vectors. The most used 4 transcription factors, octamer-binding transcription factor 4 (OCT4), (sex determining region Y)-box 2 (SOX2), Kruppel-like factor 4 (KLF4), and v-myc myelocytomatosis viral oncogene homolog (C-MYC), can induce pluripotency in mouse and human fibroblasts. Here, we report that forced expression of a new combination of transcription factors (T-cell leukemia/lymphoma protein 1A [TCL-1A], C-MYC, and SOX2) is sufficient to promote the reprogramming of human fibroblasts into pluripotent cells. These 3-factor pluripotent cells are similar to human embryonic stem cells in morphology, in the ability to differentiate into cells of the 3 embryonic layers, and at the level of global gene expression. Induced pluripotent human cells generated by a combination of other factors will be of great help for the understanding of reprogramming pathways. This, in turn, will allow us to better control cell-fate and apply this knowledge to cell therapy.

As previously shown, higher levels of NOTCH1 and increased NF-kappa B signaling is a distinctive feature of the more primitive umbilical cord blood (UCB) CD34+ hematopoietic stem cells (HSCs), as compared to bone marrow ( BM). Differences between BM and UCB cell composition also account for this finding. The CD133 marker defines a more primitive cell subset among CD34+ HSC with a proposed hemangioblast potential. To further evaluate the molecular basis related to the more primitive characteristics of UCB and CD133+ HSC, immunomagnetically purified human CD34+ and CD133+ cells from BM and UCB were used on gene expression microarrays studies. UCB CD34+ cells contained a significantly higher proportion of CD133+ cells than BM (70% and 40%, respectively). Cluster analysis showed that BM CD133+ cells grouped with the UCB cells ( CD133+ and CD34+) rather than to BM CD34+ cells. Compared with CD34+ cells, CD133+ had a higher expression of many transcription factors (TFs). Promoter analysis on all these TF genes revealed a significantly higher frequency ( than expected by chance) of NF-kappa B-binding sites (BS), including potentially novel NF-kappa B targets such as RUNX1, GATA3, and USF1. Selected transcripts of TF related to primitive hematopoiesis and self-renewal, such as RUNX1, GATA3, USF1, TAL1, HOXA9, HOXB4, NOTCH1, RELB, and NFKB2 were evaluated by real-time PCR and were all significantly positively correlated. Taken together, our data indicate the existence of an interconnected transcriptional network characterized by higher levels of NOTCH1, NF-kappa B, and other important TFs on more primitive HSC sets.

A dynamic systems simulation model of water resources was developed as a tool to help analyze alternatives to water resources management for the Piracicaba, Capivari and Jundiai River Water Basins (RB-PCJ), and used to run six 50-year simulations from 2004 to 2054. The model estimates water supply and demand, as well as contamination load by several consumers. Six runs were performed using a constant mean precipitation value, changing water supply and demand and different volumes diverted from RB-PCJ to RB-Alto Tiet. For the Business as Usual scenario, the Sustainability Index went from 0.44 in 2004 to 0.20 by 2054. The Water Sustainability Index changed from 74% in 2004 to 131% by 2054. The Falkenmark Index changed from 1,403 m(3) person (-aEuro parts per thousand 1) year (-aEuro parts per thousand 1) in 2004 to 734 m(3) person (-aEuro parts per thousand 1) year (-aEuro parts per thousand 1) by 2054. We concluded that sanitation is one of the major problems for the PCJ River Basins.

The Piracicaba, Capivari, and Jundiai River Basins (RB-PCJ) are mainly located in the State of So Paulo, Brazil. Using a dynamics systems simulation model (WRM-PCJ) to assess water resources sustainability, five 50-year simulations were run. WRM-PCJ was developed as a tool to aid decision and policy makers on the RB-PCJ Watershed Committee. The model has 254 variables. The model was calibrated and validated using available information from the 80s. Falkenmark Water Stress Index went from 1,403 m(3) person (-aEuro parts per thousand 1) year (-aEuro parts per thousand 1) in 2004 to 734 m(3) P (-aEuro parts per thousand 1) year (-aEuro parts per thousand 1) in 2054, and Xu Sustainability Index from 0.44 to 0.20. In 2004, the Keller River Basin Development Phase was Conservation, and by 2054 was Augmentation. The three criteria used to evaluate water resources showed that the watershed is at crucial water resources management turning point. The WRM-PCJ performed well, and it proved to be an excellent tool for decision and policy makers at RB-PCJ.

Using a dynamic systems model specifically developed for Piracicaba, Capivari and Jundia River Water Basins (BH-PCJ) as a tool to help to analyze water resources management alternatives for policy makers and decision takers, five simulations for 50 years timeframe were performed. The model estimates water supply and demand, as well as wastewater generation from the consumers at BH-PCJ. A run was performed using mean precipitation value constant, and keeping the actual water supply and demand rates, the business as usual scenario. Under these considerations, it is expected an increment of about similar to 76% on water demand, that similar to 39% of available water volume will come from wastewater reuse, and that waste load increases to similar to 91%. Falkenmark Index will change from 1,403 m(3) person(-1) year(-1) in 2004, to 734 m(3) P(-1) year(-1) by 2054, and the Sustainability Index from 0.44 to 0.20. Another four simulations were performed by affecting the annual precipitation by 90 and 110%; considering an ecological flow equal to 30% of the mean daily flow; and keeping the same rates for all other factors except for ecological flow and household water consumption. All of them showed a tendency to a water crisis in the near future at BH-PCJ.

Objective. The relationship of multipotent mesenchymal stromal cells (MSC) with pericytes and fibroblasts has not been established thus far, although they share many markers of primitive marrow stromal cells and the osteogenic, adipogenic, and chondrogenic differentiation potentials. Materials and Methods. We compared MSCs from adult or fetal tissues, MSC differentiated in vitro, fibroblasts and cultures of retinal pericytes obtained either by separation with anti-CD146 or adhesion. The characterizations included morphological, immunophenotypic, gene-expression profile, and differentiation potential. Results. Osteogenic, adipocytic, and chondrocytic differentiation was demonstrated for MSC, retinal perivascular cells, and fibroblasts. Cell morphology and the phenotypes defined by 22 markers were very similar. Analysis of the global gene expression obtained by serial analysis of gene expression for 17 libraries and by reverse transcription polymerase chain reaction of 39 selected genes from 31 different cell cultures, revealed similarities among MSC, retinal perivascular cells, and hepatic stellate cells. Despite this overall similarity, there was a heterogeneous expression of genes related to angiogenesis, in MSC derived from veins, artery, perivascular cells, and fibroblasts. Evaluation of typical pericyte and MSC transcripts, such as NG2, CD146, CD271, and CD140B on CD146 selected perivascular cells and MSC by real-time polymerase chain reaction confirm the relationship between these two cell types. Furthermore, the inverse correlation between fibroblast-specific protein-1 and CD146 transcripts observed on pericytes, MSC, and fibroblasts highlight their potential use as markers of this differentiation pathway. Conclusion. Our results indicate that human MSC and pericytes are similar cells located in the wall of the vasculature, where they function as cell sources for repair and tissue maintenance, whereas fibroblasts are more differentiated cells with more restricted differentiation potential. Background/Aims: The expression of cancer/testis antigens (CTAs) on additional normal tissues or stem cells may restrict their use as cancer targets. The objective of the present study was to evaluate the mRNA levels of some CTAs in a variety of tissues. Materials and Methods: mRNA of pericytes, fibroblasts and mesenchymal stem cells (MSCs) derived from adult and fetal tissues, human umbilical vein endothelial cells, MSC-derived adipocytes, selected normal tissues and control cancer cell lines (CLs) were extracted and quantitative polymerase chain reaction was performed for MAGED1, PRAME, CTAG1B, MAGEA3 and MAGEA4. Results: MAGED1 was expressed in all normal tissues and cells evaluated. CTAG1B was expressed at levels comparable to control CLs on MSCs derived from arterial, fetal skin, adipose tissue and saphenous vein, heart, brain and skin tissues. MAGEA4 was detected only in fibroblasts and differentiated adipocytes from MSCs, at levels comparable to the control CLs. Conclusion: The potential use of CTAs in immunotherapy should take into account the potential off-target effects on MSCs.

Este trabajo es un análisis crítico de la gestión organizacional que estudia la participación de la mujer en formatos productivos locales en dos regiones: la provincia de Santa Fe en Argentina y en las regiones rurales de la Zona Central de Chile. Ambos análisis exponen una visión acerca del riesgo del retorno de la inversión del capital socio-económico en formatos productivos de desarrollo local que son afectados por intervenciones de la administración de políticas públicas en el ámbito de la mujer. Este trabajo constituye un análisis cualitativo descriptivo y de proceso heurístico sobre la visión que se tiene del género, y particularmente de la mujer, para entender qué tipos iniciativas estimulan la participación hacia esa forma de organización de trabajo de coproducción y bajo qué condiciones estas ocurren.

O retrovírus linfotrópico de células T humanas tipo 1 é o agente etiológico da leucemia das células T do adulto e da paraparesia espástica tropical/mielopatia associada ao HTLV-1. O genoma proviral é composto por 9.032 pares de bases, contendo genes estruturais e regulatórios. A glicoproteína transmembrana gp 21 é codificada pelo gene estrutural env. O desenvolvimento de metodologias para a expressão heteróloga de proteínas, assim como a obtenção de uma linhagem celular que expresse a gp 21 recombinante constitutivamente são os principais objetivos do trabalho. O fragmento codificante da gp 21 foi amplificado por Nested-PCR e clonado no vetor pCR2.1-TOPO. Posteriormente, foi realizada a subclonagem no vetor de expressão pcDNA3.1+. A transfecção da linhagem celular de mamíferos HEK 293 foi realizada de maneira transitória e permanente. A produção da gp 21 recombinante foi confirmada por citometria de fluxo e a linhagem celular produtora será utilizada em ensaios de imunogenicidade.

Aparentemente os primeiros dados documentados sobre a história dos muiraquitãs, ainda que inferidos, surgem com Orellana, tido como o primeiro explorador a navegar o rio Amazonas rio baixo, ainda em 1542, quando teria combatido com índias guerreiras valentes, sem maridos, as quais denominou de Amazonas. Posteriormente De la Condamine em sua viagem ao longo deste rio, em 1735, descreve amuletos batraquianos em pedras verdes semelhantes a jade. Spix e Martius escrevem sobre pierres divines como pingente batraquiano em madrepérola e com nome de muraquêitã. Antigos escritos de Maurício Heriarte de 1662 descrevem amuleto com termos similares: baraquitãs, buraquitãs, puúraquitan, uuraquitan e mueraquitan. Finalmente Barboza Rodrigues em 1875 emprega o termo muyrakytã modificado por Barata em 1954 para muiraquitã, que entende como artefatos em forma de sapo confeccionados em pedra verde, com dois furos laterais. Teriam sido usados pelos povos Tapajó/Santarem e Conduri, que habitavam o Baixo Amazonas até a chegada do colonizador europeu, que os dizimou. Para elaborar os muiraquitãs acredita-se que esses povos utilizavam instrumentos engenhosos, mas principalmente muita paciência e habilidade. Os estudos mais recentes mostram que o maior centro produtor estava na região, da mesma forma como a fonte da matéria-prima e não na Asia como suposto pelos historiadores. Eram utilizados como amuletos, símbolos de poder, e ainda como material para compra e troca de objetos valiosos. Sobre eles há muitas lendas e mitos sempre envolvidos com as índias Amazonas, extintas ou lendárias.

OBJETIVO: Analisar as taxas e algumas características da mortalidade por suicídio entre indígenas e não indígenas no Amazonas. MÉTODO: Estudo de coorte retrospectiva, em que os dados de óbito foram obtidos no Sistema de Informações sobre Mortalidade e os populacionais no Instituto Brasileiro de Geografia e Estatística. Foram utilizados todos os registros de suicídio do período de 2006-2010 e a interpolação geométrica anual para a estimação das subpopulações. RESULTADOS: Ocorreram 688 suicídios no Amazonas, dos quais 19,0% em indígenas. A taxa ajustada de mortalidade por suicídio (TAMS) nos indígenas, de 18,4/100 mil, foi 4,4 vezes superior a dos não indígenas. A TAMS em indígenas aumentou 1,6 vez em 2010 em relação a 2006. Nos municípios de Tabatinga e São Gabriel da Cachoeira, as TAMS foram muito altas, 75,8 e 41,9/100 mil, respectivamente. CONCLUSÕES: Evidenciou-se o comportamento desigual das taxas de mortalidade por suicídio entre indígenas e não indígenas, expondo não só sua importância local, como também sua invisibilidade como problema de saúde pública, principalmente entre jovens 15 e 24 anos.

Anomalías conotruncales – Hibridación in situ Fluorescente La microdeleción cromosómica 22q11.2, es la más frecuente en humanos. Se estima una prevalencia de 1 cada 4000 recién nacidos, presentando una variabilidad clínica que abarca: Síndrome de DiGeorge, Velo-Cardio-Facial, anomalías cardíacas conotruncales aisladas, entre otras, hasta la descripción de pacientes con microdeleción subclínica. Actualmente todos estos pacientes son denominados como Síndromes de Microdeleción 22q. Hipótesis de trabajo: existe en nuestro medio un subdiagnóstico atribuible al desconocimiento de la gran variabilidad fenotípica, sospechándose sólo los casos clásicos. Objetivos: -Estimar la prevalencia relativa de microdeleción 22q11.2 en una muestra de pacientes asistidos en el Hospital de Niños de la Santísima Trinidad de Córdoba. -Correlacionar los datos clínicos con la citogenética clásica y molecular. -Diagnosticar las formas heredables para realizar asesoramiento familiar.
