The Iberian viticultural regions are convened according to the Denomination of Origin (DO) and present different climates, soils, topography and management practices. All these elements influence the vegetative growth of different varieties throughout the peninsula, and are tied to grape quality and wine type. In the current study, an integrated analysis of climate, soil, topography and vegetative growth was performed for the Iberian DO regions, using state-of-the-art datasets. For climatic assessment, a categorized index, accounting for phenological/thermal development, water availability and grape ripening conditions was computed. Soil textural classes were established to distinguish soil types. Elevation and aspect (orientation) were also taken into account, as the leading topographic elements. A spectral vegetation index was used to assess grapevine vegetative growth and an integrated analysis of all variables was performed. The results showed that the integrated climate-soil-topography influence on vine performance is evident. Most Iberian vineyards are grown in temperate dry climates with loamy soils, presenting low vegetative growth. Vineyards in temperate humid conditions tend to show higher vegetative growth. Conversely, in cooler/warmer climates, lower vigour vineyards prevail and other factors, such as soil type and precipitation acquire more important roles in driving vigour. Vines in prevailing loamy soils are grown over a wide climatic diversity, suggesting that precipitation is the primary factor influencing vigour. The present assessment of terroir characteristics allows direct comparison among wine regions and may have great value to viticulturists, particularly under a changing climate.

A crescente necessidade de tornar a construção mais sustentável e o intuito de contribuir com soluções de reaproveitamento e de escoamento dos desperdícios produzidos na indústria da construção foram determinantes para o financiamento atribuído a um projeto de investigação, com o objetivo de avaliar a viabilidade da utilização de agregados reciclados de betão provenientes da indústria de pré-fabricação para a produção de betões autocompactáveis (BAC). Desta forma, não só se pode contribuir positivamente para a melhoria da qualidade final das peças produzidas pela indústria de pré-fabricação, dispensando a utilização de métodos de vibração falíveis, mas também se contribui para a sustentabilidade do meio ambiente, minimizando o consumo de recursos naturais e a deposição de aterros de resíduos. O projeto é subdividido em diversas fases de otimização e de análise. Numa fase inicial, serão produzidos e selecionados os betões de origem (BO), onde se incluem os materiais produzidos em laboratório e os materiais provenientes de estruturas pré-fabricadas, os quais darão origem aos agregados a analisar. Esses agregados reciclados (AR) sofrerão um processo de trituração orientado (trituração primária e trituração primária e secundária) que pode influenciar as características do próprio betão. Dos resultados obtidos, seleciona-se o método de trituração a adotar para as fases subsequentes. Estas consistirão na produção de betões convencionais e autocompactáveis, que reproduzam aproximadamente a mesma classe de resistência dos betões de origem, nomeadamente 20, 45, 65 e 80 MPa, variando a sua composição (rácio a/c, adições e superplastificantes) e as taxas de agregados reciclados finos (AFR) e grossos (AGR). As percentagens de substituição AFR/AGR a considerar serão: 0/25, 25/0, 25/25, 0/100, 100/0 e 100/100%. Posteriormente, será efetuada a otimização desses betões para classes de resistência mais elevadas, utilizando, para o efeito, agregados reciclados com resistências mecânicas correspondentes a uma classe inferior. Serão incluídos neste estudo os betões de elevado desempenho autocompactáveis, cuja resistência se pretende que seja igual ou superior a 80 MPa. Em todas as fases, serão executados ensaios de avaliação às características mecânicas e de durabilidade dos betões, incluindo a análise microestrutural para a observação da evolução da estrutura porosa da pasta e dos agregados reciclados e da interface pasta/agregados. Pretende-se obter como resultado final um betão com agregados reciclados com agregados reciclados, cujas propriedades sejam compatíveis ou superiores às de um betão auto-compactável com agregados naturais.

The aim of the present work is to provide insight into the mechanism of laccase reactions using syringyl-type mediators. We studied the pH dependence and the kinetics of oxidation of syringyl-type phenolics using the low CotA and the high redox potential TvL laccases. Additionally, the efficiency of these compounds as redox mediators for the oxidation of non-phenolic lignin units was tested at different pH values and increasing mediator/non-phenolic ratios. Finally, the intermediates and products of reactions were identified by LC-MS and H-1 NMR. These approaches allow concluding on the (1) mechanism involved in the oxidation of phenolics by bacterial laccases, (2) importance of the chemical nature and properties of phenolic mediators, (3) apparent independence of the enzyme's properties on the yields of non-phenolics conversion, (4) competitive routes involved in the catalytic cycle of the laccase-mediator system with several new C-O coupling type structures being proposed.

Background: Mushroom polysaccharides play an important role in functional foods because they exhibit biological modulator properties such as antitumour, antiviral and antibacterial activities. The present study involved the production, purification and characterisation of intracellular and extracellular free and protein-bound polysaccharides from Pleurotus ostreatus and the investigation of their growth-inhibitory effect on human carcinoma cell lines. Results: Several fermentation parameters were obtained: batch polysaccharide productivities of 0.013 +/- 8.12 x 10-5 and 0.037 +/- 0.0005 g L-1 day-1 for intracellular and extracellular polysaccharides respectively, a maximum biomass concentration of 9.35 +/- 0.18 g L-1, Pmax = 0.935 +/- 0.018 g L-1 day-1, µmax = 0.218 +/- 0.02 day-1, YEP/X = 0.040 +/- 0.0015 g g-1 and YIP/X = 0.014 +/- 0.0003 g g-1. Some polysaccharides exhibited superoxide dismutase (SOD)-like activity of 50-200 units. Fourier transform infrared analysis of the polysaccharides revealed absorption bands characteristic of such biological macromolecules. Cytotoxicity assays showed that both intracellular and extracellular polysaccharides exhibited antitumour activity towards several tested human carcinoma cell lines in a dose-dependent manner. Conclusion - The polysaccharides of P. ostreatus exhibited high SOD-like activity, which strongly supports their biological effect on tumour cell lines. The extracellular polysaccharides presented the highest antitumour activity towards the RL95 carcinoma cell line and should be further investigated as an antitumour agent.

Eucalyptus globulus sapwood and heartwood showed no differences in lignin content (23.0% vs. 23.7%) and composition: syringyl-lignin (17.9% vs. 18.0%) and guaiacyl-lignin (4.8% vs. 5.2%). Delignification kinetics of S- and G-units in heartwood and sapwood was investigated by Py-GC–MS/FID at 130, 150 and 170 °C and modeled as double first-order reactions. Reactivity differences between S and G-units were small during the main pulping phase and the higher reactivity of S over G units was better expressed in the later pulping stage. The residual lignin composition in pulps was different from wood or from samples in the initial delignification stages, with more G and H-units. S/G ratio ranged from 3 to 4.5 when pulp residual lignin was higher than 10%, decreasing rapidly to less than 1. The S/H was initially around 20 (until 15% residual lignin), decreasing to 4 when residual lignin was about 3%.

A study of chemical transformations of cork during heat treatments was made using colour variation and FTIR analysis. The cork enriched fractions from Quercus cerris bark were subjected to isothermal heating in the temperature range 150–400 ◦C and treatment time from 5 to 90 min. Mass loss ranged from 3% (90 min at 150 ◦C) to 71% (60 min at 350 ◦C). FTIR showed that hemicelluloses were thermally degraded first while suberin remained as the most heat resistant component. The change of CIE-Lab parameters was rapid for low intensity treatments where no significant mass loss occurred (at 150 ◦C L* decreased from the initial 51.5 to 37.3 after 20 min). The decrease in all colour parameters continued with temperature until they remained substantially constant with over 40% mass loss. Modelling of the thermally induced mass loss could be made using colour analysis. This is applicable to monitoring the production of heat expanded insulation agglomerates.
