We investigated the role of cAMP/cGMP, protein kinases and intracellular calcium ( [Ca2+](i)) in pentoxifylline-stimulated hamster sperm capacitation and the acrosome reaction (AR) in vitro. Treatment with pentoxifylline [0.45 mM) initially increased sperm cAMP values 2.8-fold, compared with untreated controls (396 +/- 9.2 versus 141 +/- 6.0 fmoles/10(6) spermatozoa; mean +/- SEM, n = 6) after 15 min, although by 3 h, cAMP values were similar (503-531 fmoles/10(6) spermatozoal, cGMP values (similar to 27 fmoles/10(6) spermatozoa) were the same in treated and control spermatozoa. Both sperm capacitation and the AR, determined from the absence of an acrosomal cap, were stimulated by pentoxifylline; these were almost completely inhibited by a Cl-/HCO(3)(-)antiporter inhibitor (4,4-diisothiocyanato-stilbene-2,2 disulphonic acid; 1 mM) defined from the degree of sperm motility and by a protein kinase A inhibitor (H89; 10 mu M) A protein kinase G inhibitor (staurosporine, 1 nM) did not affect pentoxifylline-stimulated capacitation but inhibited the AR by 50%. A protein tyrosine kinase inhibitor (tyrphostin A-47, 0.1 mM) had no effect on either pentoxifylline-stimulated capacitation or AR, A phospholipase A(2) inhibitor (aristolochic acid, 0.4 mM) markedly inhibited the pentoxifylline-stimulated AR but not capacitation. When intracellular sperm calcium [Ca2+](i) was measured using fura-2-AM, there was an early rise 271 nM at 0.5 hi in pentoxifylline(-treated spermatozoa; this appeared to be due to intracellular mobilization rather than to uptake. In the absence of extracellular Ca2+, sperm motility was maintained in the presence of pentoxifylline, but capacitation did not occur; spermatozoa exhibited a low level of hyperactivated motility and had a poor rate of AR(20.5 +/- 2.3%). These results suggest that: (i) the pentoxifylline-stimulated early onset of sperm capacitation may be mediated by an early rise in cAMP and [Ca2+/-](i) and involves protein kinase A activity; and (ii) pentoxifylline-stimulated AR may require phospholipase A;A(2) and protein kinase C activity.

The role of FIC (Filamentation induced by cAMP)(2) domain containing proteins in the regulation of many vital pathways, mostly through the transfer of NMPs from NTPs to specific target proteins (NMPylation), in microorganisms, higher eukaryotes, and plants is emerging. The identity and function of FIC domain containing protein of the human pathogen, Mycobacterium tuberculosis, remains unknown. In this regard, M. tuberculosis fic gene (Mtfic) was cloned, overexpressed, and purified to homogeneity for its biochemical characterisation. It has the characteristic FIC motif, HPFREGNGRSTR (HPFxxGNGRxxR), spanning 144th to 155th residue. Neither the His-tagged nor the GST-tagged MtFic protein, overexpressed in Escherichia coil, nor expression of Mtfic in Mycobacterium smegmatis, yielded the protein in the soluble fraction. However, the maltose binding protein (MBP) tagged MtFic (MBP-MtFic) could be obtained partly in the soluble fraction. The cloned, overexpressed, and purified recombinant MBP-MtFic showed conversion of ATP, GTP, CTP, and UTP into AMP. GMP, CMP, and UMP, respectively. Sequence alignment with several FIC motif containing proteins, complemented with homology modeling on the FIC motif containing protein, VbhT of Bartonella schoenbuchensis as the template, showed conservation and interaction of residues constituting the FIC domain. Site-specific mutagenesis of the His144, or Glu148, or Asn150 of the FIC motif, or of Arg87 residue that constitutes the FIC domain, or complete deletion of the FIC motif, abolished the NTP to NMP conversion activity. The design of NMP formation assay using the recombinant, soluble MtFic would enable identification of its target substrate for NMPylation. (C) 2012 Elsevier Inc. All rights reserved.

The highly modular nature of protein kinases generates diverse functional roles mediated by evolutionary events such as domain recombination, insertion and deletion of domains. Usually domain architecture of a kinase is related to the subfamily to which the kinase catalytic domain belongs. However outlier kinases with unusual domain architectures serve in the expansion of the functional space of the protein kinase family. For example, Src kinases are made-up of SH2 and SH3 domains in addition to the kinase catalytic domain. A kinase which lacks these two domains but retains sequence characteristics within the kinase catalytic domain is an outlier that is likely to have modes of regulation different from classical src kinases. This study defines two types of outlier kinases: hybrids and rogues depending on the nature of domain recombination. Hybrid kinases are those where the catalytic kinase domain belongs to a kinase subfamily but the domain architecture is typical of another kinase subfamily. Rogue kinases are those with kinase catalytic domain characteristic of a kinase subfamily but the domain architecture is typical of neither that subfamily nor any other kinase subfamily. This report provides a consolidated set of such hybrid and rogue kinases gleaned from six eukaryotic genomes-S. cerevisiae, D. melanogaster, C. elegans, M. musculus, T. rubripes and H. sapiens-and discusses their functions. The presence of such kinases necessitates a revisiting of the classification scheme of the protein kinase family using full length sequences apart from classical classification using solely the sequences of kinase catalytic domains. The study of these kinases provides a good insight in engineering signalling pathways for a desired output. Lastly, identification of hybrids and rogues in pathogenic protozoa such as P. falciparum sheds light on possible strategies in host-pathogen interactions.

Vários estudos sugerem que a desnutrição materna no período pós-natal poderia causar alterações na homeostase glicêmica da prole na vida adulta. Neste trabalho objetivamos investigar a interferência da programação metabólica induzida pela desnutrição protéica materna durante o início da lactação sobre a homeostase glicêmica e a sinalização da insulina nos tecidos muscular e adiposo. Animais desnutridos (D-dieta da mãe contendo 0% de proteína nos primeiros 10 dias de lactação) ou controle (C-dieta da mãe contendo 22% de proteína) foram estudados do nascimento até a vida adulta. Em resumo, observamos uma diminuição na insulina plasmática acompanhada de normoglicemia nos animais adultos desnutridos. A ativação do receptor de insulina (IR), após a estimulação com o hormônio apresentou-se diminuída durante o período de restrição protéica em músculo isolado destes animais experimentais. Durante o período da lactação, observamos uma diminuição na captação de glicose, na fosforilação do substrato para o receptor de insulina (IRS 1) e na translocação do GLUT 4 no tecido muscular. Na idade adulta, entretanto, houve aumento significativo na captação de glicose e translocação do GLUT 4 no músculo, associado com o aumento na expressão da PI3 quinase associada ao IRS 1. No tecido adiposo de ratos desnutridos adultos observamos menor fosforilação em tirosina tanto do IR quanto do IRS 1, que foi compensada pela maior ativação do IRS 2 e da PI3 quinase. Os níveis basais de pAkt e de GLUT 4 na membrana estavam aumentados, culminando em um aumento na captação de glicose. Observamos também uma redistribuição do citoesqueleto de actina e maior resistência aos efeitos da Ltrunculina B nos adipócitos dos ratos desnutridos. Em conclusão, este estudo demonstrou que a desnutrição materna no início da lactação é capaz de causar alterações na prole na vida adulta, o que parece estar relacionado com a expressão e ativação de proteínas chave na cascata da sinalização da insulina nos tecidos periféricos, importantes na regulação do metabolismo da glicose.

To understand the molecular events of ovarian development in penaeid shrimp, RNA arbitrarily primed polymerase chain reaction (RAP-PCR) was used to identify differentially expressed genes during ovarian maturation in Metapenaeus ensis. From a screening of 700 clones in a cDNA library of the shrimp ovary by the products of RAP-PCR of different maturation stages, 91 fragments with differentially expressed pattern as revealed by dot-blot hybridization were isolated and sequenced. Forty-two of these fragments show significant sequence similarity to known gene products and the differentially expressed pattern of 10 putative genes were further characterized via Northern hybridization. Putative glyceraldehyde-3-phosphate dehydrogenase and arginine kinase are related to provision of energy for active cellular function in oocyte development. Translationally controlled tumor protein, actin, and keratin are related to the organization of cytoskeleton to accomplish growth and development of oocytes. High mobility group protein DSP1, heat shock protein 70, and nucleoside diphosphate kinase may act as repressors before the onset of ovarian maturation. Peptidyl-prolyl cis-trans isomerase and glutathione peroxidase are related to the stabilization of proteins and oocytes. This study provides new insights on the molecular events in the ovarian development in the shrimp.

We used microarray technology to study differentially expressed genes in white spot syndrome virus (WSSV)-infected shrimp. A total of 3136 cDNA targets, including 1578 unique genes from a cephalothorax cDNA library and 1536 cDNA clones from reverse and forward suppression subtractive hybridization (SSH) libraries of Fenneropenaeus chinensis, plus 14 negative and 8 blank control clones, were spotted onto a 18 x 18 mm area of NH2-modified glass slides. Gene expression patterns in the cephalothorax of shrimp at 6 h after WSSV injection and moribund shrimp naturally infected by WSSV were analyzed. A total of 105 elements on the arrays showed a similar regulation pattern in artificially infected shrimp and naturally infected moribund shrimp; parts of the results were confirmed by semiquantitative reverse transcriptase-polymerase chain reaction (RT-PCR). The up-regulated expression of immune-related genes, including heat shock proteins (HSP70 and HSP90), trehalose-phosphate synthase (TPS), ubiquitin C, and so forth, were observed when shrimp were challenged with WSSV. Genes including myosin LC2, ATP synthase A chain, and arginine kinase were found to be down-regulated after WSSV infection. The expression of housekeeping genes such as actin, elongation factor, and tubulin is not stable, and so these genes are not suitable as internal standards for semiquantitative RT-PCR when shrimp are challenged by WSSV. As a substitute, we found that triosephosphate isomerase (TPI) was an ideal candidate of interstandards in this situation.

Anthropogenic pollutant chemicals pose a major threat to aquatic organisms. There is a need for more research on emerging categories of environmental chemicals such as nanomaterials, endocrine disruptors and pharmaceuticals. Proteomics offers options and advantages for early warning of alterations in environmental quality by detecting sub-lethal changes in sentinel species such as the mussel, Mytilus edulis. This thesis aimed to compare the potential of traditional biomarkers (such as enzyme activity measurement) and newer redox proteomic approaches. Environmental proteomics, especially a redox proteomics toolbox, may be a novel way to study pollutant effects on organisms which can also yield information on risks to human health. In particular, it can probe subtle biochemical changes at sub-lethal concentrations and thus offer novel insights to toxicity mechanisms. In the first instance, the present research involved a field-study in three stations in Cork Harbour, Ireland (Haulbowline, Ringaskiddy and Douglas) compared to an outharbour control site in Bantry Bay, Ireland. Then, further research was carried out to detect effects of anthropogenic pollution on selected chemicals. Diclofenac is an example of veterinary and human pharmaceuticals, an emerging category of chemical pollutants, with potential to cause serious toxicity to non-target organisms. A second chemical used for this study was copper which is a key source of contamination in marine ecosystems. Thirdly, bisphenol A is a major anthropogenic chemical mainly used in polycarbonate plastics manufacturing that is widespread in the environment. It is also suspected to be an endocrine disruptor. Effects on the gill, the principal feeding organ of mussels, were investigated in particular. Effects on digestive gland were also investigated to compare different outcomes from each tissue. Across the three anthropogenic chemicals studied (diclofenac, copper and bisphenol A), only diclofenac exposure did not show any significant difference towards glutathione transferase (GST) responses. Meanwhile, copper and bisphenol A significantly increased GST in gill. Glutathione reductase (GR) enzyme analysis revealed that all three chemicals have significant responses in gill. Catalase activity showed significant differences in digestive gland exposed to diclofenac and gills exposed to bisphenol A. This study focused then on application of redox proteomics; the study of the oxidative modification of proteins, to M. edulis. Thiol proteins were labelled with 5-iodoacetamidofluorescein prior to one-dimensional and two-dimensional electrophoresis. This clearly revealed some similarities on a portion of the redox proteome across chemical exposures indicating where toxicity mechanism may be common and where effects are unique to a single treatment. This thesis documents that proteomics is a robust tool to provide valuable insights into possible mechanisms of toxicity of anthropogenic contaminants in M. edulis. It is concluded that future research should focus on gill tissue, on protein thiols and on key individual proteins discovered in this study such as calreticulin and arginine kinase which have not previously been considered as biomarkers in aquatic toxicology prior to this study.

Induced by a literature review, this paper presents a framework of dimensions and indicators highlighting the underpinning aspects and values of social learning within teacher groups. Notions of social networks, communities of practice and learning teams were taken as the main perspectives to influence this social learning framework. The review exercise resulted in four dimensions: (1) practice, (2) domain and value creation, (3) collective identity and (4) organization. The indicators corresponding to these dimensions serve as the foundation for understanding social learning in practice. The framework of dimensions and indicators can be of assistance for researchers as well as teacher groups that aim to assess their views on social learning and analyse whether these views fit the learning goals of the group, or that adjustments are required. In this way, learning processes within groups of teachers can be improved.

Signal initiation by engagement of the TCR triggers actin rearrangements, receptor clustering, and dynamic organization of signaling complexes to elicit and sustain downstream signaling. Nef, a pathogenicity factor of HIV, disrupts early TCR signaling in target T cells. To define the mechanism underlying this Nef-mediated signal disruption, we employed quantitative single-cell microscopy following surface-mediated TCR stimulation that allows for dynamic visualization of distinct signaling complexes as microclusters (MCs). Despite marked inhibition of actin remodeling and cell spreading, the induction of MCs containing TCR-CD3 or ZAP70 was not affected significantly by Nef. However, Nef potently inhibited the subsequent formation of MCs positive for the signaling adaptor Src homology-2 domain-containing leukocyte protein of 76 kDa (SLP-76) to reduce MC density in Nef-expressing and HIV-1-infected T cells. Further analyses suggested that Nef prevents formation of SLP-76 MCs at the level of the upstream adaptor protein, linker of activated T cells (LAT), that couples ZAP70 to SLP-76. Nef did not disrupt pre-existing MCs positive for LAT. However, the presence of the viral protein prevented de novo recruitment of active LAT into MCs due to retargeting of LAT to an intracellular compartment. These modulations in MC formation and composition depended on Nef's ability to simultaneously disrupt both actin remodeling and subcellular localization of TCR-proximal machinery. Nef thus employs a dual mechanism to disturb early TCR signaling by limiting the communication between LAT and SLP-76 and preventing the dynamic formation of SLP-76-signaling MCs.

Complex animals use a wide variety of adaptor proteins to produce specialized sites of interaction between actin and membranes. Plants do not have these protein families, yet actin-membrane interactions within plant cells are critical for the positioning of subcellular compartments, for coordinating intercellular communication, and for membrane deformation [1]. Novel factors are therefore likely to provide interfaces at actin-membrane contacts in plants, but their identity has remained obscure. Here we identify the plantspecific Networked (NET) superfamily of actin-binding proteins, members of which localize to the actin cytoskeleton and specify different membrane compartments. The founding member of the NET superfamily, NET1A, is anchored at the plasma membrane and predominates at cell junctions, the plasmodesmata. NET1A binds directly to actin filaments via a novel actin-binding domain that defines a superfamily of thirteen Arabidopsis proteins divided into four distinct phylogenetic clades. Members of other clades identify interactions at the tonoplast, nuclear membrane, and pollen tube plasma membrane, emphasizing the role of this superfamily in mediating actin-membrane interactions.

A novel reusable molecularly imprinted polymer (MIP) assembled on a polymeric layer of carboxylated poly(vinyl chloride) (PVCsingle bondCOOH) for myoglobin (Myo) detection was developed. This polymer was casted on the gold working area of a screen printed electrode (Au-SPE), creating a novel disposable device relying on plastic antibodies. Electrochemical impedance spectroscopy (EIS), cyclic voltammetry (CV) and Fourier transform infrared spectroscopy (FTIR) studies confirmed the surface modification. The MIP/Au-SPE devices displayed a linear behaviour in EIS from 0.852 to 4.26 μg mL−1, of positive slope 6.50 ± 1.48 (kΩ mL μg−1). The limit of detection was 2.25 μg mL−1. Square wave voltammetric (SWV) assays were made in parallel and showed linear responses between 1.1 and 2.98 μg mL−1. A current decrease was observed against Myo concentration, producing average slopes of −0.28 ± 0.038 μA mL μg−1. MIP/Au-SPE also showed good results in terms of selectivity. The error% found for each interfering species were 7% for troponin T (TnT), 11% for bovine serum albumin (BSA) and 2% for creatine kinase MB (CKMB), respectively. Overall, the technical modification over the Au-SPE was found a suitable approach for screening Myo in biological fluids.

Fibroblast growth factor receptors (FGFRs) are involved in proliferative and differentiation physiological responses. Deregulation of FGFR-mediated signaling involving the Ras/PI3K/Akt and the Ras/Raf/ERK MAPK pathways is causally involved in the development of several cancers. The caspase-3/p120 RasGAP module is a stress sensor switch. Under mild stress conditions, RasGAP is cleaved by caspase-3 at position 455. The resulting N-terminal fragment, called fragment N, stimulates anti-death signaling. When caspase-3 activity further increases, fragment N is cleaved at position 157. This generates a fragment, called N2, that no longer protects cells. Here, we investigated in Xenopus oocytes the impact of RasGAP and its fragments on FGF1-mediated signaling during G2/M cell cycle transition. RasGAP used its N-terminal Src homology 2 domain to bind FGFR once stimulated by FGF1, and this was necessary for the recruitment of Akt to the FGFR complex. Fragment N, which did not associate with the FGFR complex, favored FGF1-induced ERK stimulation, leading to accelerated G2/M transition. In contrast, fragment N2 bound the FGFR, and this inhibited mTORC2-dependent Akt Ser-473 phosphorylation and ERK2 phosphorylation but not phosphorylation of Akt on Thr-308. This also blocked cell cycle progression. Inhibition of Akt Ser-473 phosphorylation and entry into G2/M was relieved by PHLPP phosphatase inhibition. Hence, full-length RasGAP favors Akt activity by shielding it from deactivating phosphatases. This shielding was abrogated by fragment N2. These results highlight the role played by RasGAP in FGFR signaling and how graded stress intensities, by generating different RasGAP fragments, can positively or negatively impact this signaling.

Nous avons récemment démontré que les espèces réactives oxygénées induisent une augmentation de l’expression des protéines Giα dans les cellules du muscle lisse vasculaire (CMLV) provenant d’aortes de rats spontanément hypertendus (SHR, de l’anglais spontaneously hypertensive rats). La présente étude a pour but d’étudier les effets du peroxyde d’hydrogène (H2O2), un oxydant qui induit le stress oxydatif, sur l’expression de Giα et sur l’activité de l’adénylate cyclase, et d’explorer les voies de signalisation sous-jacentes responsables de cette réponse. Nos résultats montrent que H2O2 induit une augmentation de l’expression des protéines Giα-2 et Giα-3 de manière dose- et temps-dépendante avec une augmentation maximale de 40-50% à 100 µM après 1 heure, sans affecter l’expression de Gsα. L’expression des protéines Giα a été maintenue au niveau normal en presence de AG 1478, AG1295, PD98059 et la wortmannine, des inhibiteurs d’EGF-R (de l’anglais epidermal growth factor receptor), PDGFR-β (de l’anglais platelet-derived growth factor receptor β), de la voie de signalisation ras-ERK1/2 (de l’anglais extracellular regulated kinase1/2), et de la voie de la PI3Kinase-AKT (de l’anglais phosphatidyl inositol-3 kinase), respectivement. En outre, le traitement des CMLV avec H2O2 a induit une augmentation du degré de phosphorylation d’EGF-R, PDGF-R, ERK1/2 et AKT; et cette expression a été maintenue au niveau témoin par leurs inhibiteurs respectifs. Les inhibiteurs d’EGF-R et PDGF-R ont aussi induit une diminution du degré de phosphorylation de ERK1/2, et AKT/PKB. En outre, la transfection des cellules avec le siRNA (de l’anglais, small interfering ribonucleic acid) de EGF-R et PDGFR-β a atténué la surexpression des protéines Giα-2 et Giα-3 induite par le traitement au H2O2. La surexpression des protéines Giα induite par H2O2 a été corrélée avec une augmentation de la fonction de la protéine Giα. L’inhibition de l’activité de l’adénylate cyclase par de faibles concentrations de GTPγS après stimulation par la forskoline a augmenté de 20% dans les cellules traitées au H2O2. En outre, le traitement des CMLV au H2O2 a aussi accru l’inhibition de l’activité de l’adénylate cyclase par les hormones inhibitrices telles que l’angiotensine II, oxotrémorine et C-ANP4-23. D’autre part, la stimulation de l’adénylate cyclase induite par GTPγS, glucagon, isoprotérénol, forskoline, et le fluorure de sodium (NaF) a été atténuée de façon significative dans les cellules traitées au H2O2. Au cours de la cytokinèse, le génome dédoublé est compartimentalisé en deux cellules filles. L’anneau contractile, une structure dynamique, est constitué d’actine, myosine (NMY-II) et d’autres protéines accessoires. NMY-2 est le seul moteur protéique impliqué dans la contraction de l’anneau durant la cytokinèse. Depuis longtemps, il a été considéré que celle-ci glissait le long des filaments d’actine grâce à sa capacité de traction. Récemment, plusieurs études ont découvert que son activité réticulante joue un rôle en cytokinèse et il est connu que la NMY-2 peut s’assembler en filaments bipolaires à partir de dimères. Ainsi, nous postulons que leur dimension (nombre de moteurs ATPasiques) pourrait dicter leur contribution en activité motrice et réticulante. Afin de déterminer la composition des filaments corticaux de NMY-2, nous avons utilisé une technique d'imagerie de molécules individuelles à l’aide de la microscopie TIRF. J’ai trouvé à travers l’analyse statistique de la distribution des NMY-2 mesurés que les filaments sont assemblés à deux dimensions constantes: Des filaments composés de 20 dimères et 30 dimères. La kinase Rho est une activatrice de NMY-2 nécessaire pour les niveaux physiologiques de NMY-2 sur l’anneau contractile, pour des cinétiques et fermeture concentrique de l’anneau. La déplétion de RhoK augmente l’abondance relative des filaments de 20 dimères. Ainsi, RhoK pourrait réguler le recrutement de la NMY et aussi l’assemblage des filaments corticaux de NMY-2. De plus, à l’aide de la microscopie confocale à temps réel, j’ai trouvé que lors de la déplétion de RhoK, il se produit une réduction du recrutement et du délai d’initiation du sillon, une fermeture lente et une augmentation significative de la concentricité de l’anneau. De plus, j’ai mesuré des défauts dans l’organisation corticale de l’anneau contractile en patch. La déplétion de MRCK-1 n’affecte pas l’initiation du sillon, les cinétiques de fermeture, ou la fermeture concentrique de l’anneau. Paradoxalement, la déplétion de MRCK-1 augmente le recrutement cortical de NMY-2, mais quand depleté simultanément avec Rho-K il diminue NMY-2 à l’équateur comparé à la déplétion seule de Rho-K. De plus, la double déplétion, conduit à un phénotype de concentricité de l’anneau, suivie d’un recentrage.

Le système rénine-angiotensine-aldostérone (SRAA) régule l’homéostasie de la contraction des artères. Or, suivant la liaison de l’angiotensine II (Ang II) à son récepteur AT1, le SRAA est également impliqué dans l’activation de voies de signalisation à l’origine de l’inflammation et de l’hypertrophie des cellules musculaires lisses vasculaires (CMLV), soit deux processus participant au remodelage vasculaire caractéristique de diverses maladies cardiovasculaires, telles l’hypertension et l’athérosclérose. Ces pathologies sont les premières causes de mortalité naturelle en Amérique et les traitements les ciblant ne sont pas optimaux puisqu’ils visent seulement quelques facteurs de risque qui leur sont associés. Ainsi, la détermination des effecteurs intracellulaires régulant ces voies délétères est nécessaire à l'identification de nouvelles cibles thérapeutiques. L’inflammation Ang II-dépendante dans les CMLV est attribuée au facteur de transcription nuclear factor-kappa B (NF-κB). Cependant, les processus moléculaires couplant le récepteur AT1 à son activation sont peu caractérisés. L’étude abordant cette question démontre in vitro que NF-κB est activé par la protéine IκB kinase β (IKKβ) dans les CMLV exposées à l’Ang II et que cette kinase est régulée par deux voies de signalisation indépendantes, mais complémentaires afin d’assurer son activation rigoureuse et soutenue. L’une des voies est précoce et dépend des seconds messagers ainsi que de deux nouveaux effecteurs sous-jacents au récepteur AT1, soit la E3 ligase TNF receptor-associated factor 6 (TRAF6) et la IKK kinase transforming growth factor-beta-activated kinase 1 (TAK1) tandis que la seconde est tardive et résulte de la signalisation mitogen-activated protein kinase kinase 1/2 (MEK1/2) - extracellular signal-regulated kinase 1/2 (ERK1/2) - ribosomal S6 kinase (RSK). L’inhibition conjointe de ces voies abroge complètement la réponse inflammatoire, ce qui indique qu’elles en sont la seule source. Ainsi, l’inhibition d’IKKβ pourrait suffire à contrer l’inflammation impliquée dans le remodelage vasculaire associé à une suractivation du SRAA. Une découverte des plus novatrices découle de cette étude, qui veut que la E3 ligase TRAF6 est un nouvel effecteur des récepteurs couplés aux protéines G et est à l’origine de la formation d’un nouveau type de second messager, soit des chaînes libres de poly-ubiquitines. Les mécanismes moléculaires à la base de l’hypertrophie Ang II-dépendante dans les CMLV sont également peu définis. Or, suivant la parution d’un article démontrant qu’IKKβ dans les cellules cancéreuses participe aux mécanismes d’initiation de la traduction en réponse au facteur de nécrose tumorale α (TNFα) via la phosphorylation de la protéine Tuberous sclerosis 1 (TSC1) et donc l’activation du complexe mammalian target of rapamycin (mTORC1), une hypothèse a été émise selon laquelle cette kinase serait impliquée dans la synthèse protéique Ang II-dépendante dans les CMLV. Les expériences effectuées in vitro dans des CMLV exposées à l’Ang II démontrent qu’IKKβ induit la phosphorylation de TSC1 ainsi que l’activation de mTORC1 et de ses substrats S6 kinase 1 (S6K1) et translational regulators eukaryotic translation initiation factor 4E-binding protein (4E-BP1), deux protéines impliquées directement dans l’hypertrophie. Par ailleurs, la synthèse protéique au niveau des CMLV exposées à l’Ang II est réduite de 75% suivant la diminution de l’expression d’IKKβ et suivant la surexpression d’un mutant de TSC1 dont le site consensus d’IKKβ a été modifié, faisant de cette kinase un médiateur majeur au niveau de ce processus. Ainsi, in vitro IKKβ en réponse à l’Ang II est en amont de deux processus impliqués dans un remodelage vasculaire à l’origine de maladies cardiovasculaires. De plus, plusieurs facteurs de risque de ces pathologies convergent à l’activation d’IKKβ, ce qui en fait une cible thérapeutique particulièrement attrayante. Qui plus est, l’administration d’un inhibiteur d’IKKβ à des rats diminue non seulement la synthèse protéique dépendante de l’Ang II au niveau de l’aorte et des artères mésentériques, mais également la synthèse de la protéine pro-inflammatoire VCAM-1 par les cellules composant l’aorte, ce qui confirme son envergure en tant que cible.
