Adipose tissue secretes a variety of adipokines, including leptin and adiponectin, which are involved in endocrine processes regulating glucose and fatty metabolism, energy expenditure, inflammatory response, immunity, cardiovascular function, and reproduction. The present article describes the fluctuations in circulating leptin and adiponectin as well as their patterns of secretion in women from birth to menopause. During pregnancy, leptin and adiponectin seem to act in an autocrine/paracrine fashion in the placenta and adipose tissue, playing a role in the maternal-fetal interface and contributing to glucose metabolism and fetal development. In newborns, adiponectin levels are two to three times higher than in adults. Full-term newborns have significantly higher leptin and adiponectin levels than preterms, whereas small-for-gestational-age infants have lower levels of these adipokines than adequate-for-gestational-age newborns. However, with weight gain, leptin concentrations increase significantly. Children between 5 and 8 years of age experience an increase in leptin and a decrease in adiponectin regardless of body mass index, with a reversal of the newborn pattern for adiponectin: plasma adiponectin levels at age five are inversely correlated with percentage of body fat. In puberty, leptin plays a role in the regulation of menstrual cycles. In adults, it has been suggested that obese individuals exhibit both leptin resistance and decreased serum adiponectin levels. In conclusion, a progressive increase in adiposity throughout life seems to influence the relationship between leptin and adiponectin in women.

Antiviral nucleosides are compounds that are used against viruses, such as human immunodeficiency virus (HIV) and hepatitis C virus (HCV). To act as therapeutic agent, the antiviral nucleoside needs to be phosphorylated to nucleotide in the body in three consecutive phosphorylation steps by cellular or viral enzymes. The first phosphorylation to the nucleoside monophosphate is often inefficient and leads to poor antiviral activity. The antiviral efficacy can be improved by applying a prodrug strategy and delivering the antiviral nucleoside directly as its monophosphate. In prodrug strategies of antiviral nucleotides, the negative charges on the phosphate moiety are temporarily masked with protecting groups. Once inside the cell, the protecting groups are removed by enzymatic or chemical processes. Many prodrug strategies apply biodegradable protecting groups, the removal of which is triggered by esterase enzymes. Several studies have, however, demonstrated that the removal rate of the second and subsequent esterase labile protecting groups significantly slows down after the first protecting group is removed due to the negative charge on the phosphodiester intermediate, which disturbs the catalytic site of the enzyme. In this thesis, esterase labile protecting group strategies where the issue of retardation could be avoided were studied. Prodrug candidates of antiviral nucleotides were synthesized and kinetic studies on the chemical and enzymatic stability were carried out. In the synthesized compounds, the second protecting group is cleaved from the monophosphate some other mechanism than esterase triggered activation or the structure of prodrug requires only one protecting group. In addition, esterase labile protecting group which is additionally thermally removable was studied. This protecting group was cleaved from oligomeric phosphodiesters both enzymatically and thermally and seems most attractive of the studied phosphate protecting groups. However, the rate of the thermal removal still is too slow to allow efficient protection of longer oligonucleotides and needs optimization. This study discusses the formation phase of Chinese-Finnish joint ventures in China. The purpose of this thesis is to create best practices for Finnish software companies in forming a joint venture with a local Chinese company in China. Therefore, the main research question, in what are the best practices for forming Sino-Finnish joint ventures in China for Finnish software firms, is examined through four different themes within the joint venture formation phase; the motives, the partner se- lection, the choice of a joint venture type and joint venture negotiations. The theoretical background of the study consists of literature relating to the establishment process of Sino-Western joint ventures in China. The empirical research conducted for this study is based on the expert interviews. The empirical data was gathered via nine semi-structured interviews with both Chinese and Finnish experts in software and technology industry, who have experience or knowledge in establishing Sino-Finnish joint ventures in China. Thematic analysis was used to cat- egorize and interpret the interview data. In addition, a thematic network was built to act as a basis of the analysis. According to the main findings, the main motives for Finnish software companies to establish a joint venture in China are lack of skills or experience, little resources to enter on their own, and China’s large market. The main motives for Chinese companies are to gain new technology or man- agerial skills, and expand internationally. The intellectual property rights (IPR) have recently im- proved a lot in China, but the Finnish companies’ knowledge on IPR is inadequate. The Finnish software companies should conduct a market and industry research in order to understand their po- sition in the market and to find a suitable location and potential joint venture partners. It is essential to define partner selection criteria and partner attributes. In addition, it is important to build the joint venture around complementary motives and a win-win situation between the joint venture partners. The Finnish companies should be prepared that the joint venture negotiations will be challenging and they will take a long time. The challenges can be overcome by gaining understanding about the Chinese culture and business environment. The findings of this study enhance understanding of the joint venture formation phase in China. This study provides guidelines for Finnish software companies to establish a joint venture in China. In addition, this study brings new insights to the Sino-Western joint venture literature with its soft- ware industry context. Future research is, however, necessary in order to gain an understanding of the advantages and disadvantages of a joint venture as an entry mode into China for Finnish soft- ware companies

Digitalization has been predicted to change the future as a growing range of non-routine tasks will be automated, offering new kinds of business models for enterprises. Serviceoriented architecture (SOA) provides a basis for designing and implementing welldefined problems as reusable services, allowing computers to execute them. Serviceoriented design has potential to act as a mediator between IT and human resources, but enterprises struggle with their SOA adoption and lack a linkage between the benefits and costs of services. This thesis studies the phenomenon of service reuse in enterprises, proposing an ontology to link different kinds of services with their role conceptually as a part of the business model. The proposed ontology has been created on the basis of qualitative research conducted in three large enterprises. Service reuse has two roles in enterprises: it enables automated data sharing among human and IT resources, and it may provide cost savings in service development and operations. From a technical viewpoint, the ability to define a business problem as a service is one of the key enablers for achieving service reuse. The research proposes two service identification methods, first to identify prospective services in the existing documentation of the enterprise and secondly to model the services from a functional viewpoint, supporting service identification sessions with business stakeholders.

Yacon is considered a functional food due to its the fructooligosaccharide (FOS) content, however its perishability and low production volume is a problem. The aim of this study was to analyze the changes in aging during one year of storage and conduct sensory analysis of fermented of yacon. For one year total acidity, volatile acidity, free and total sulfur dioxide, reducing sugars, sucrose, phenols and FOS and its antioxidant power were studied. At the end of aging a sensory profile and acceptance panel was performed. The total and volatile acidity increased significantly (p < 0.05). A decrease in fructooligosaccharide was also observed, indicating that yeasts are probably capable of hydrolyzing the latter. The total sulfur dioxide decreased significantly, demonstrating its ability to act well against oxidation products. This product showed good antioxidant capacity and sensory profiles of considerable acceptance. Therefore it can be affirmed that the alcoholic fermentation of yacon can be a good alternative for the industrial sector and farmers in the region could be encouraged to use large-scale production.

Tämän tutkimuksen tavoitteena oli selvittää miten omistajuus vaikuttaa yrityksen toimintaan ja tulokseen ja syventää näkökulmaa omistajan vaikutuksesta yrityksen menestymiseen. Tutkimus tehtiin kvalitatiivisen ja kvantitatiivisen tutkimusmenetelmän yhdistelmällä, joka mahdollistaa laajemman näkökulman tutkittavaan asiaan ja tuo tutkimukseen luotettavuutta. Omistajan kompetenssilla toimia aktiivisena omistajana sekä hallita erilaisia liiketoimintariske-jä on suuri merkitys. Kestävä menestys, uudistuminen ja yrityksen arvon luominen vaativat niin asiantuntemusta, näkemyksiä, sitoutunutta riskirahoitusta kuin vuorovaikutustaitoja. Omis-tajalla on vaikutusvalta osaavan johdon ja henkilöstön motivaation ylläpysymiseen ja tahtoti-laan toimia yrityksen etujen mukaisesti ja yritykseen sitoutuneena. Ymmärtämällä omistamansa yrityksen kilpailuedut ja hallitsemalla organisatoriset riskit, omis-taja voi vaikuttaa valitsemansa roolin kautta yrityksen menestykseen ja tuottoarvon kasvuun. Tutkimuksen tulokset viittaavat myös siihen, että henkilöstön kokemat positiiviset psykologi-sen ja sosiaalisen omistajuuden tunteet selittävät onnistumisia.

As human action, drinking alcohol or smoking cigarettes, for instance, are phenomena that are familiar to most people. We recognise when people use drugs or gamble whether that happens in the television or around us. Sometimes we call these actions addictions. Addictive actions are actions that puzzle us. This puzzlement has raised different kinds of views on addiction that describe the phenomenon in different ways. The proponents of the views pick features that they consider to be sufficient in capturing the phenomenon. The disease view emphasises that addicted individuals are not in control over their own actions, whereas the choice view highlights addicted individuals’ capacity to act according to their own preferences. Some see addiction as a defect of will and addictive action is a manifestation of that. Sometimes they all insist on referring to the same group of people and describing the same actions with seemingly contradictory terms. What happens when an addicted individual acts in accordance with his addiction? This thesis also tries to answer this question and make this kind of action and agency understandable. By showing that these three common views fall short of capturing the phenomenon, I will provide characteristics that jointly suffice for something to be labelled addiction, but which are not, however, individually necessary (or sufficient) for addiction. Those characteristics are strong desire, myopia, biased decision-making, and weakness of will. Furthermore, they should be understood in the framework of diachronic, active agency. They contribute to a view on addictive action that consists of different kinds of actions. Acknowledging the variety of nuanced human action, the understanding of addictive action is increased and this may also be utilised in addiction-related policies and treatment. The emphasis of my study lies specifically on understanding addiction as action by the means of analytical philosophy.

Strateginen osaaminen on yrityksen strategian toteutumisen kannalta olennaista osaamista. Sen tunnistamista voidaan pitää koko strategisen osaamisen johtami-sen tärkeimpänä – ja vaikeimpana - vaiheena. Vasta sen jälkeen, kun strategiset osaamisalueet on tunnistettu, voidaan niitä menestyksekkäästi johtaa. Tämän tutkimuksen tavoitteena oli selvittää, mitkä tekijät vaikuttavat strategisen osaamisen tunnistamiseen ja johtamiseen. Tutkimus suoritettiin laadullisena tapaustutkimuksena energia-alan yrityksessä ja empiirinen aineisto kerättiin haastattelemalla konsernin johtoryhmän jäseniä. Tutkimustulos osoittaa, että strategisen osaamisen tunnistamisen ja johtamisen kannalta ratkaisevaa on yrityksen strategisten linjausten vieminen riittävän konk-reettiselle tasolle. Mitä turbulenssisempi toimintaympäristö on, sitä tiheämmin strategisen osaamisen kehittämisen suuntaa tulee tarkastella.

Cancer affects more than 20 million people each year and this rate is increasing globally. The Ras/MAPK-pathway is one of the best-studied cancer signaling pathways. Ras proteins are mutated in almost 20% of all human cancers and despite numerous efforts, no effective therapy that specifically targets Ras is available to date. It is now well established that Ras proteins laterally segregate on the plasma membrane into transient nanoscale signaling complexes called nanoclusters. These Ras nanoclusters are essential for the high-fidelity signal transmission. Disruption of nanoclustering leads to reduction in Ras activity and signaling, therefore targeting nanoclusters opens up important new therapeutic possibilities in cancer. This work describes three different studies exploring the idea of membrane protein nanoclusters as novel anti-cancer drug targets. It is focused on the design and implementation of a simple, cell-based Förster Resonance Energy Transfer (FRET)-biosensor screening platform to identify compounds that affect Ras membrane organization and nanoclustering. Chemical libraries from different sources were tested and a number of potential hit molecules were validated on full-length oncogenic proteins using a combination of imaging, biochemical and transformation assays. In the first study, a small chemical library was screened using H-ras derived FRET-biosensors. Surprisingly from this screen, commonly used protein synthesis inhibitors (PSIs) were found to specifically increase H-ras nanoclustering and downstream signalling in a H-ras dependent manner. Using a representative PSI, increase in H-ras activity was shown to induce cancer stem cell (CSC)-enriched mammosphere formation and tumor growth of breast cancer cells. Moreover, PSIs do not increase K-ras nanoclustering, making this screening approach suitable for identifying Ras isoform-specific inhibitors. In the second study, a nanoncluster-directed screen using both H- and K-ras derived FRET biosensors identified CSC inhibitor salinomycin to specifically inhibit K-ras nanocluster organization and downstream signaling. A K-ras nanoclusteringassociated gene signature was established that predicts the drug sensitivity of cancer cells to CSC inhibitors. Interestingly, almost 8% of patient tumor samples in the The Cancer Genome Atlas (TCGA) database had the above gene signature and were associated with a significantly higher mortality. From this mechanistic insight, an additional microbial metabolite screen on H- and K-ras biosensors identified ophiobolin A and conglobatin A to specifically affect K-ras nanoclustering and to act as potential breast CSC inhibitors. In the third study, the Ras FRET-biosensor principle was used to investigate membrane anchorage and nanoclustering of myristoylated proteins such as heterotrimeric G-proteins, Yes- and Src-kinases. Furthermore, Yes-biosensor was validated to be a suitable platform for performing chemical and genetic screens to identify myristoylation inhibitors. The results of this thesis demonstrate the potential of the Ras-derived FRETbiosensor platform to differentiate and identify Ras-isoform specfic inhibitors. The results also highlight that most of the inhibitors identified predominantly perturb Ras subcellular distribution and membrane organization through some novel and yet unknown mechanisms. The results give new insights into the role of Ras nanoclusters as promising new molecular targets in cancer and in stem cells.

As information technology evolves, organizations have begun to adopt big data and apply it as a tool throughout their decision-making processes. Research on big data has grown in the past years however mainly from a technical stance and there is a void in business related cases. This thesis fills the gap in the research by addressing big data challenges and failure cases. The Technology-Organization-Environment framework was applied to carry out a literature review on trends in Business Intelligence and Knowledge management information system failures. A review of extant literature was carried out using a collection of leading information system journals. Academic papers and articles on big data, Business Intelligence, Decision Support Systems, and Knowledge Management systems were studied from both failure and success aspects in order to build a model for big data failure. I continue and delineate the contribution of the Information System failure literature as it is the principal dynamics behind technology-organization-environment framework. The gathered literature was then categorised and a failure model was developed from the identified critical failure points. The failure constructs were further categorized, defined, and tabulated into a contextual diagram. The developed model and table were designed to act as comprehensive starting point and as general guidance for academics, CIOs or other system stakeholders to facilitate decision-making in big data adoption process by measuring the effect of technological, organizational, and environmental variables with perceived benefits, dissatisfaction and discontinued use.

Turun 1600-luvun johtavan porvariston naiset toimivat aktiivisesti ja päättäväisesti omiin ja perheensä etuihin liittyvissä asioissa. He jatkoivat jo keskiajalla alkanutta traditiota, jossa myös porvarisvaimojen taloudellisen toiminnan merkitys korostui. He eivät kuitenkaan olleet perheestään, suvustaan tai Turun paikallisyhteisöstä erillään olevia yksilöitä, vaan tärkeä osa niitä. Siksi heidän toimijuuttaan ei voi eikä tule erottaa aikakautensa ja yhteisönsä kontekstista. Johtavan porvariston naiset ajoivat asioitaan Turun kaupungin oikeusasteissa kaikissa kolmessa siviilisäädyssä, naimattomina tyttärinä, aviovaimoina ja leskinä, aktiivisimmat heistä elämänsä aikana yli sata kertaa. Verrattuna muihin saman ajanjakson kaupunkeihin naimattomaksi jääneet turkulaiset porvaristyttäret esiintyivät korostuneen aktiivisesti valvomassa taloudellisia etujaan. Heidän toimintansa hyväksyttiin silloin, kun se ei ylittänyt porvariyhteisön sovinnaisuuden rajoja, joita he pyrkivät koettelemaan parantaakseen talouttaan. Leskinaisten oikeustoimikelpoisuus kertoo siitä, että naisia ei pidetty vaimoinakaan lähtökohtaisesti kyvyttöminä hoitamaan perheen asioita, vaan ainoastaan velvollisina alistumaan miehensä edusmiehisyyteen ja määräysvaltaan tämän eläessä. Talouden pää oli yleensä mies ja hänen velvollisuutenaan oli ajaa perheensä asioita. Siksi aviovaimot esiintyvät oikeudessa siviilisäädyistä harvimmin. Leskillä oli puolestaan oikeus hoitaa itse omaisuuttaan. Tämä tarkoitti myös velvollisuutta valvoa lasten perintöä. Turun johtava porvaristo koostui joistakin kymmenistä perheistä, joista suurin osa oli 1600-loppupuolelle tultaessa muodostanut avioliittojen kautta laajan sukulaisverkoston. Tätä verkostoa ei olisi voinut syntyä ilman porvarisnaisia, joiden avioliitot mahdollistivat omaisuuden siirtämisen uusille yrittäjille. Vaikka sukulaisuussuhteet olivat tärkeitä kaupankäynnissä, kokivat naiset ne myös uhkaksi itselleen. Sukulaismiehet pyrkivät usein saamaan naisten omaisuutta käyttöönsä vetoamalla näiden heikkoon kykyyn hallita omaisuutta. Porvarisnaiset kuitenkin puolustautuivat tällaisissa tilanteissa voimakkaasti ja hakivat turvaa porvariyhteisöä edustaneelta raadilta. He osasivat vedota asemaansa yhteisön jäseninä ja saivat yleensä raadin ja porvariyhteisön tuen. Aviomiehen kuoltua porvarisnaisen oli päätettävä, jatkoiko hän miehensä liiketoimintaa vai hankkiko hän elantonsa jollain muulla tavalla. Monet lesket järjestelivät taloudellisia asioitaan heti miehensä kuoltua ja jäivät sitten elämään esimerkiksi kaupunkitalonsa, maatilojensa ja pääomansa turvin. Osa naisista jatkoi yrittäjyyttä pienimuotoisesti esimerkiksi lainaamalla rahaa korkoa vastaan tai panemalla olutta. Muutamat johtavan porvariston naiset jatkoivat kuitenkin laajaa taloudellista toimintaa vuosikausia. Heidän menestyksensä osoittaa, että he olivat saaneet jo kotonaan perusteellisen koulutuksen porvarisammattiin. He olivat myös toimineet miehensä kumppaneina yrityksen toiminnassa, joten heillä oli hyvät edellytykset itsenäiseen asioiden hoitamiseen. Saamansa koulutuksen turvin porvarisnaiset siirsivät osaamisensa myös lapsilleen.

Tutkimuksen tavoitteena on kuvata ja ymmärtää osuustoiminnallisen pankin yhteiskuntavastuun toteuttamista. Yhteiskuntavastuussa on kysymys vastuusta toiminnan aiheuttamista vaikutuksista sekä teoista, joilla vastataan yhteiskunnan ja sidosryhmien odotuksiin. Tutkimuksessa pyritään selvittämään, mitä odotuksia jäsenistöllä on osuustoiminnallisen pankin yhteiskuntavastuusta. Tutkimus on tapaustutkimus ja kohdeyhteisönä on itsenäisesti toimiva osuuspankki. Tutkimus on luonteeltaan laadullinen ja aineiston keräämisessä käytetään triangulaatiota. Empiirinen aineisto koostuu valmiista aineistosta, kohdeyhteisön kahden henkilöstön edustajan teemahaastatteluista sekä kyselytutkimuksesta, joka osoitettiin kohdeyhteisön jäsenistöä edustavalle edustajistolle. Jäsenistö odottaa osuustoiminnallisen pankin toimivan vastuullisesti. Osuuspankin yhteiskuntavastuussa korostuu paikallisuus sekä jäsenistön hyvinvointi. Yhteiskuntavastuullisuuden edellytyksenä pidetään toiminnan kannattavuutta, pankkipalveluiden saatavuutta sekä lakien ja säännösten noudattamista.

In Finland, entrepreneurship policy priorities are encouragement and support for business innovation, growth and internationalization. SMEs are having economically significant employment and wealth creation potential. During the following years, there will be plenty of businesses for sale on the Finnish market. The challenge is to find and identify growth-oriented, talented, innovative, capable to rapid experimentation new entrepreneurs, in whose hands the company's growth targets are going to be realized. In these research is examined frameworks of the company's growth and the factors influencing growth on the basis of previous studies and literature. In addition, the aim is to create a suitable framework to identify the early stage of entrepreneurs, who are growth-oriented. The other aim is create the supposed directions of influences for growth-orientation. The objectives of the work of the results is to act as an umbrella topic, contextual analysis and forming a new applied framework for further studies. The results of these research are the background variables, the desire for growth are composed of motives, attitudes and desires; the skills for growth are composed of capabilities, experiences, self-efficacy and the willingness for risk-taking; the opportunities for growth are composed of IPR, financing, megatrends and other observed chances.
