We analyse the dynamics of a relativistic particle moving in a uniform magnetic field and perturbed by a stationary electrostatic wave. We show that a pulsed wave produces an infinite number of perturbing terms with the same winding number. The perturbation coupling alters the number of island chains as a function of the parameters of the wave. We also observe that the number of chains in is always even if the number of islands in each chain is odd.

Compartmentalization of self-replicating molecules (templates) in protocells is a necessary step towards the evolution of modern cells. However, coexistence between distinct template types inside a protocell can be achieved only if there is a selective pressure favoring protocells with a mixed template composition. Here we study analytically a group selection model for the coexistence between two template types using the diffusion approximation of population genetics. The model combines competition at the template and protocell levels as well as genetic drift inside protocells. At the steady state, we find a continuous phase transition separating the coexistence and segregation regimes, with the order parameter vanishing linearly with the distance to the critical point. In addition, we derive explicit analytical expressions for the critical steadystate probability density of protocell compositions.

Brown rot caused by Monilinia laxa and Monilinia fructigena is considered one of the most important diseases affecting Prunus species. Although some losses can result from the rotten fruits in the orchard, most of the damage is caused to fruits during the post-harvest phase. Several studies reported that brown rot incidence during fruit development highly varies; it was found that at a period corresponding to the the pit hardening stage, fruit susceptibility drastically decreases, to be quickly restored afterwards. However the molecular basis of this phenomenon is still not well understood. Furthermore, no difference in the rot incidence was found between wound and un-wound fruits, suggesting that resistance associated more to a specifc biochemical response of the fruit, rather than to a higher mechanical resistance. So far, the interaction Monilinia-peach was analyzed through chemical approaches. In this study, a bio-molecular approach was undertaken in order to reveal alteration in gene expression associated to the variation of susceptibility. In this thesis three different methods for gene expression analysis were used to analyze the alterations in gene expression occurring in peach fruits during the pit hardening stage, in a period encompassing the temporary change in Monilinia susceptibility: real time PCR, microarray and cDNA AFLP techniques. In 2005, peach fruits (cv.K2) were weekly harvested during a 19-week long-period, starting from the fourth week after full bloom, until full maturity. At each sampling time, three replicates of 5 fruits each were dipped in the M.laxa conidial suspension or in distilled water, as negative control. The fruits were maintained at room temperature for 3 hours; afterwards, they were peeled with a scalpel; the peel was immediately frozen in liquid nitrogen and transferred to -80 °C until use. The degree of susceptibility of peach fruit to the pathogen was determined on 3 replicates of 20 fruits each, as percentage of infected fruits, after one week at 20 °C. Real time PCR analysis was performed to study the variation in expression of those genes encoding for the enzymes of the phenylpropanoid pathway (phenylalanine ammonia lyase (PAL), chalcone synthase (CHS), cinnamate 4-hydroxylase (C4H), leucoanthocyanidine reductase (LAR), hydroxycinnamoyl CoA quinate hydroxycinnamoyl transferase (HQT) and of the jasmonate pathway, such as lipoxygenase (LOX), both involved in the production of important defense compounds. Alteration in gene expression was monitored on fruit samples of a period encompassing the pit hardening stage and the corresponding temporary resistance to M.laxa infections, weekly, from the 6thto the 12th week after full bloom (AFB) inoculated with M. laxa or mock-inoculated. The data suggest a critical change in the expression level of the phenylpropanoid pathway from the 7th to the 8th week AFB; such change could be directly physiologically associated to the peach growth and it could indirectly determine the decrease of susceptibility of peach fruit to Monilinia rot during the subsequent weeks. To investigate on the transcriptome variation underneath the temporary loss of susceptibility of peach fruits to Monilinia rot, the microarray and the cDNA AFLP techniques were used. The samples harvested on the 8th week AFB (named S, for susceptible ones) and on the 12th week AFB (named R, for resistant ones) were compared, both inoculated or mock-inoculated. The microarray experiments were carried out at the University of Padua (Dept. of Environmental Agronomy and Crop Science), using the μPEACH1.0 microarray together with the suited protocols. The analysis showed that 30 genes (corresponding to the 0.6% of the total sequences (4806) contained in the μPeach1.0 microarray) were found up-regulated and 31 ( 0.6%) down regulated in RH vs. SH fruits. On the other hand, 20 genes (0.4%) were shown to be up-regulated and 13 (0.3%) down-regulated in the RI vs. SI fruit. No genes were found differentially expressed in the mock-inoculated resistant fruits (RH) vs. the inoculated resistant ones (RI). Among the up-regulated genes an ATP sulfurylase, an heat shock protein 70, the major allergen Pru P1, an harpin inducing protein and S-adenosylmethionine decarboxylase were found, conversely among the down-regulated ones, cinnamyl alcohol dehydrogenase, an histidine- containing phosphotransfer protein and the ferritin were found. The microarray experimental results and the data indirectly derived, were tested by Real Time PCR analysis. cDNA AFLP analysis was also performed on the same samples. 339 transcript derived fragments considered significant for Monilinia resistance, were selected, sequenced and classified. Genes potentially involved in cell rescue and defence were well represented (8%); several genes (12.1%) involved in the protein folding, post-transductional modification and genes (9.2%) involved in cellular transport were also found. A further 10.3% of genes were classified as involved in the metabolism of aminoacid, carbohydrate and fatty acid. On the other hand, genes involved in the protein synthesis (5.7%) and in signal transduction and communication (5.7%) were found. Among the most interesting genes found differentially expressed between susceptible and resistant fruits, genes encoding for pathogenesis related (PR) proteins were found. To investigate on the association of Monilinia resistance and PR biological function, the major allergen Pru P1 (GenBank accession AM493970) and its isoform (here named Pru P2), were expressed in heterologous system and in vitro assayed for their anti-microbial activity. The ribonuclease activity of the recombinant Pru P1 and Pru P2 proteins was assayed against peach total RNA. As the other PR10 proteins, they showed a ribonucleolytic activity, that could be important to contrast pathogen penetration. Moreover Pru P1 and Pru P2 recombinant proteins were checked for direct antimicrobial activity. No inhibitory effect of Pru P1 or Pru P2 was detected against the selected fungi.

Many potential diltiazem related L-VDCC blockers were developed using a multidisciplinary approach. This current study was to investigate and compare diltiazem with to the newly developed compounds by mouse Langendorff-perfused heart, Ca2+-transient and on recombinant L-VDCC. Twenty particular compounds were selected by the ligand-based virtual screening procedure (LBVS). From these compounds, five of them (5b, M2, M7, M8 and P1) showed a potent and selective inotropic activity on guinea-pig left atria driven 1 Hz. Further assays displayed an interesting negative inotropic effect of M2, M8, P1 and M7 on guinea pig isolated left papillary muscle driven at 1 Hz, a relevant vasorelaxant activity of 5b, M2, M7, M8 and P1 on K+-depolarized guinea-pig ileum longitudinal smooth muscle and a significant inhibition of contraction of 5b, M2, M8 and P1 on carbachol stimulated ileum longitudinal smooth muscle. Wild-type human heart and rabbit lung α1 subunits were expressed (combined with the regulatory α2δ and β3 subunits) in Xenopus Leavis oocytes using a two-electrode voltage clamp technique. Diltiazem is a benzothiazepine Ca2+ channel blocker used clinically for its antihypertensive and antiarrhythmic effects. Previous radioligand binding assays revealed a complex interaction with the benzothiazepine binding site for M2, M7 and M8. (Carosati E. et al. J. Med Chem. 2006, 49; 5206). In agreement with this findings, the relative order of increased rates of contraction and relaxation at lower concentrations s(≤10-6M) in unpaced hearts was M7>M2>M8>P1. Similar increases in Ca2+ transient were observed in cardiomyocytes. Diltiazem showed negative inotropic effects whereas 5b had no significant effect. Diltiazem blocks Ca2+current in a use-dependent manner and facilitates the channel by accelerating the inactivation and decelerating the recovery from inactivation. In contrast to diltiazem, the new analogs had no pronounced use-dependence. Application of 100 μM M8, M2 showed ~ 10% tonic block; in addition, M8, M2 and P1 shifted the steady state inactivation in hyperpolarized direction and the current inactivation time was significantly decreased compared with control (219.6 ± 11.5 ms, 226 ± 14.5 vs. 269 ± 12.9 vs. 199.28 ± 8.19 ms). Contrary to diltiazem, the recovery from the block by M8 and M2 was comparable to control. Only P1 showed a significantly decrease of the time for the recovery from inactivation. All of the compounds displayed the same sensitivity on the Ca2+ channel rabbit lung α1 except P1. Taken together, these findings suggest that M8, M2 and P1 might directly decrease the binding affinity or allow rapid dissociation from the benzothiazepine binding site.

In this thesis we will disclose the results obtained from the diastereoisomeric salt formation (n salt, p salt and p1,n1 salt) between non-racemic trans-chrysanthemic acid (trans-ChA) and pure enantiomers of threo-2-dimethylamino-1-phenyl-1,3-propanediol (DMPP). The occurrence of p1,n1 salt formation can have profound effects on enantiomer separation of scalemic (non-racemic) mixtures. This phenomenon when accompanied by substrate self-association impedes the complete recovery of the major enantiomer through formation of an inescapable racemate cage. A synthetic sequence for the asymmetric synthesis of bicyclo[3.2.0]heptanones and bicyclo[3.2.0]hept-3-en-6-ones through a cycloaddition strategy is reported. The fundamental step is a [2+2]-cycloaddition of an enantiopure amide derived from the reaction between a set of acids and an oxazolidinone as the chiral auxiliary. The inter- and intramolecular cycloaddition of in situ-generated keteniminium salts gives bicycles with a good enantioselection. A key intermediate of Iloprost, a chemically stable and biologically active mimic of prostacyclin PGI2 is synthesized following a ‘green approach’. An example of simple optical resolution of this racemic intermediate involving the diastereoisomeric salt formation is described.

Benessere delle popolazioni, gestione sostenibile delle risorse, povertà e degrado ambientale sono dei concetti fortemente connessi in un mondo in cui il 20% della popolazione mondiale consuma più del 75% delle risorse naturali. Sin dal 1992 al Summit della Terra a Rio de Janeiro si è affermato il forte legame tra tutela dell’ambiente e riduzione della povertà, ed è anche stata riconosciuta l’importanza di un ecosistema sano per condurre una vita dignitosa, specialmente nelle zone rurali povere dell’Africa, dell’Asia e dell’America Latina. La natura infatti, soprattutto per le popolazioni rurali, rappresenta un bene quotidiano e prezioso, una forma essenziale per la sussistenza ed una fonte primaria di reddito. Accanto a questa constatazione vi è anche la consapevolezza che negli ultimi decenni gli ecosistemi naturali si stanno degradando ad un ritmo impressionate, senza precedenti nella storia della specie umana: consumiamo le risorse più velocemente di quanto la Terra sia capace di rigenerarle e di “metabolizzare” i nostri scarti. Allo stesso modo aumenta la povertà: attualmente ci sono 1,2 miliardi di persone che vivono con meno di un dollaro al giorno, mentre circa metà della popolazione mondiale sopravvive con meno di due dollari al giorno (UN). La connessione tra povertà ed ambiente non dipende solamente dalla scarsità di risorse che rende più difficili le condizioni di vita, ma anche dalla gestione delle stesse risorse naturali. Infatti in molti paesi o luoghi dove le risorse non sono carenti la popolazione più povera non vi ha accesso per motivi politici, economici e sociali. Inoltre se si paragona l’impronta ecologica con una misura riconosciuta dello “sviluppo umano”, l’Indice dello Sviluppo Umano (HDI) delle Nazioni Unite (Cfr. Cap 2), il rapporto dimostra chiaramente che ciò che noi accettiamo generalmente come “alto sviluppo” è molto lontano dal concetto di sviluppo sostenibile accettato universalmente, in quanto i paesi cosiddetti “sviluppati” sono quelli con una maggior impronta ecologica. Se allora lo “sviluppo” mette sotto pressione gli ecosistemi, dal cui benessere dipende direttamente il benessere dell’uomo, allora vuol dire che il concetto di “sviluppo” deve essere rivisitato, perché ha come conseguenza non il benessere del pianeta e delle popolazioni, ma il degrado ambientale e l’accrescimento delle disuguaglianze sociali. Quindi da una parte vi è la “società occidentale”, che promuove l’avanzamento della tecnologia e dell’industrializzazione per la crescita economica, spremendo un ecosistema sempre più stanco ed esausto al fine di ottenere dei benefici solo per una ristretta fetta della popolazione mondiale che segue un modello di vita consumistico degradando l’ambiente e sommergendolo di rifiuti; dall’altra parte ci sono le famiglie di contadini rurali, i “moradores” delle favelas o delle periferie delle grandi metropoli del Sud del Mondo, i senza terra, gli immigrati delle baraccopoli, i “waste pickers” delle periferie di Bombay che sopravvivono raccattando rifiuti, i profughi di guerre fatte per il controllo delle risorse, gli sfollati ambientali, gli eco-rifugiati, che vivono sotto la soglia di povertà, senza accesso alle risorse primarie per la sopravvivenza. La gestione sostenibile dell’ambiente, il produrre reddito dalla valorizzazione diretta dell’ecosistema e l’accesso alle risorse naturali sono tra gli strumenti più efficaci per migliorare le condizioni di vita degli individui, strumenti che possono anche garantire la distribuzione della ricchezza costruendo una società più equa, in quanto le merci ed i servizi dell’ecosistema fungono da beni per le comunità. La corretta gestione dell’ambiente e delle risorse quindi è di estrema importanza per la lotta alla povertà ed in questo caso il ruolo e la responsabilità dei tecnici ambientali è cruciale. Il lavoro di ricerca qui presentato, partendo dall’analisi del problema della gestione delle risorse naturali e dal suo stretto legame con la povertà, rivisitando il concetto tradizionale di “sviluppo” secondo i nuovi filoni di pensiero, vuole suggerire soluzioni e tecnologie per la gestione sostenibile delle risorse naturali che abbiano come obiettivo il benessere delle popolazioni più povere e degli ecosistemi, proponendo inoltre un metodo valutativo per la scelta delle alternative, soluzioni o tecnologie più adeguate al contesto di intervento. Dopo l’analisi dello “stato del Pianeta” (Capitolo 1) e delle risorse, sia a livello globale che a livello regionale, il secondo Capitolo prende in esame il concetto di povertà, di Paese in Via di Sviluppo (PVS), il concetto di “sviluppo sostenibile” e i nuovi filoni di pensiero: dalla teoria della Decrescita, al concetto di Sviluppo Umano. Dalla presa di coscienza dei reali fabbisogni umani, dall’analisi dello stato dell’ambiente, della povertà e delle sue diverse facce nei vari paesi, e dalla presa di coscienza del fallimento dell’economia della crescita (oggi visibile più che mai) si può comprendere che la soluzione per sconfiggere la povertà, il degrado dell’ambiente, e raggiungere lo sviluppo umano, non è il consumismo, la produzione, e nemmeno il trasferimento della tecnologia e l’industrializzazione; ma il “piccolo e bello” (F. Schumacher, 1982), ovvero gli stili di vita semplici, la tutela degli ecosistemi, e a livello tecnologico le “tecnologie appropriate”. Ed è proprio alle Tecnologie Appropriate a cui sono dedicati i Capitoli successivi (Capitolo 4 e Capitolo 5). Queste sono tecnologie semplici, a basso impatto ambientale, a basso costo, facilmente gestibili dalle comunità, tecnologie che permettono alle popolazioni più povere di avere accesso alle risorse naturali. Sono le tecnologie che meglio permettono, grazie alle loro caratteristiche, la tutela dei beni comuni naturali, quindi delle risorse e dell’ambiente, favorendo ed incentivando la partecipazione delle comunità locali e valorizzando i saperi tradizionali, grazie al coinvolgimento di tutti gli attori, al basso costo, alla sostenibilità ambientale, contribuendo all’affermazione dei diritti umani e alla salvaguardia dell’ambiente. Le Tecnologie Appropriate prese in esame sono quelle relative all’approvvigionamento idrico e alla depurazione dell’acqua tra cui: - la raccolta della nebbia, - metodi semplici per la perforazione di pozzi, - pompe a pedali e pompe manuali per l’approvvigionamento idrico, - la raccolta dell’acqua piovana, - il recupero delle sorgenti, - semplici metodi per la depurazione dell’acqua al punto d’uso (filtro in ceramica, filtro a sabbia, filtro in tessuto, disinfezione e distillazione solare). Il quinto Capitolo espone invece le Tecnolocie Appropriate per la gestione dei rifiuti nei PVS, in cui sono descritte: - soluzioni per la raccolta dei rifiuti nei PVS, - soluzioni per lo smaltimento dei rifiuti nei PVS, - semplici tecnologie per il riciclaggio dei rifiuti solidi. Il sesto Capitolo tratta tematiche riguardanti la Cooperazione Internazionale, la Cooperazione Decentrata e i progetti di Sviluppo Umano. Per progetti di sviluppo si intende, nell’ambito della Cooperazione, quei progetti che hanno come obiettivi la lotta alla povertà e il miglioramento delle condizioni di vita delle comunità beneficiarie dei PVS coinvolte nel progetto. All’interno dei progetti di cooperazione e di sviluppo umano gli interventi di tipo ambientale giocano un ruolo importante, visto che, come già detto, la povertà e il benessere delle popolazioni dipende dal benessere degli ecosistemi in cui vivono: favorire la tutela dell’ambiente, garantire l’accesso all’acqua potabile, la corretta gestione dei rifiuti e dei reflui nonché l’approvvigionamento energetico pulito sono aspetti necessari per permettere ad ogni individuo, soprattutto se vive in condizioni di “sviluppo”, di condurre una vita sana e produttiva. È importante quindi, negli interventi di sviluppo umano di carattere tecnico ed ambientale, scegliere soluzioni decentrate che prevedano l’adozione di Tecnologie Appropriate per contribuire a valorizzare l’ambiente e a tutelare la salute della comunità. I Capitoli 7 ed 8 prendono in esame i metodi per la valutazione degli interventi di sviluppo umano. Un altro aspetto fondamentale che rientra nel ruolo dei tecnici infatti è l’utilizzo di un corretto metodo valutativo per la scelta dei progetti possibili che tenga presente tutti gli aspetti, ovvero gli impatti sociali, ambientali, economici e che si cali bene alle realtà svantaggiate come quelle prese in considerazione in questo lavoro; un metodo cioè che consenta una valutazione specifica per i progetti di sviluppo umano e che possa permettere l’individuazione del progetto/intervento tecnologico e ambientale più appropriato ad ogni contesto specifico. Dall’analisi dei vari strumenti valutativi si è scelto di sviluppare un modello per la valutazione degli interventi di carattere ambientale nei progetti di Cooperazione Decentrata basato sull’Analisi Multi Criteria e sulla Analisi Gerarchica. L’oggetto di questa ricerca è stato quindi lo sviluppo di una metodologia, che tramite il supporto matematico e metodologico dell’Analisi Multi Criteria, permetta di valutare l’appropriatezza, la sostenibilità degli interventi di Sviluppo Umano di carattere ambientale, sviluppati all’interno di progetti di Cooperazione Internazionale e di Cooperazione Decentrata attraverso l’utilizzo di Tecnologie Appropriate. Nel Capitolo 9 viene proposta la metodologia, il modello di calcolo e i criteri su cui si basa la valutazione. I successivi capitoli (Capitolo 10 e Capitolo 11) sono invece dedicati alla sperimentazione della metodologia ai diversi casi studio: - “Progetto ambientale sulla gestione dei rifiuti presso i campi Profughi Saharawi”, Algeria, - “Programa 1 milhão de Cisternas, P1MC” e - “Programa Uma Terra e Duas Águas, P1+2”, Semi Arido brasiliano.

Diese Arbeit beschreibt ein Experiment zur Photoproduktionneutraler Pionen am Proton im Schwellenbereich. DurchVerwendung linear polarisierter Photonen konnte neben dentotalen und differentiellen Wirkungsquerschnitten zum erstenMal die Photonasymmetrie nahe der Schwelle gemessen werden.Besonderes Interesse galt dem aus diesen physikalischenObservablen bestimmbaren s-Wellen-Multipol E0+ sowie der erstmaligen Bestimmung aller drei p-Wellen-KombinationenP1, P2 und P3 im Bereich der Schwelle.Das Experiment wurde 1995/1996 am ElektronenbeschleunigerMAMI (Mainzer Mikrotron) der Universität Mainz durchgeführt.Durch Verwendung eines Diamanten als Bremsstrahltarget fürdie Elektronen wurden über den Prozeß der kohärentenBremsstrahlung linear polarisierte Photonen erzeugt. DieEnergie der Photonen wurde über die Messung der Energie der gestreuten Elektronen in der MainzerPhotonenmarkierungsanlage bestimmt. Der Detektor TAPS, eineAnordnung aus 504 BaF2-Modulen, war um einFlüssigwasserstofftarget aufgebaut. In den Modulen wurdendie im Target produzierten neutralen Pionen über ihrenZerfall in zwei Photonen nachgewiesen.Die totalen und differentiellen Wirkungsquerschnitte wurdenim Energiebereich zwischen der Schwelle von 144.7 MeV und168 MeV gemessen. Die erstmals gemessene Photonasymmetriefür 159.5 MeV ist positiv und hat einen Wert von+0.217+/-0.046 für einen Polarwinkel von 90 Grad. Der Multipol E0+ und die drei p-Wellen-Kombinationen wurden andie physikalischen Observablen über zwei unterschiedlicheMethoden angepaßt, die übereinstimmende Ergebnisselieferten. Die Vorhersagen der Niederenergietheoreme derchiralen Störungstheorie für P1 und P2 stimmen beiEinbeziehung der statistischen und systematischen Fehler mitden experimentellen Werten überein.

Das Wolf-Hirschhorn-Syndrom (WHS) ist ein komplexes und variables Fehlbildungs- Retardierungssyndrom, das durch Deletion in der distalen Chromosomenregion 4p16.3 hervorgerufen wird und dessen Ätiologie und Pathogenese bisher weitgehend unverstanden sind. Die Zielsetzung in der vorliegenden Arbeit bestand in der Identifizierung und vorläufigen Charakterisierung neuer Gene, die an der Entstehung des Syndroms beteiligt sein könnten. Die Wolf-Hirschhorn-Syndrom-kritische Region (WHSCR) konnte zu Beginn der vorliegenden Arbeit auf einen ca. 2 Mb großen Bereich zwischen den Markern D4S43 und D4S142 eingegrenzt werden. Für die Identifizierung neuer Gene wurden zunächst drei größere genomische Cosmid-/PAC-Contigs (I-III) im Bereich der Marker D4S114 bis D4S142 erstellt und mittels Exonamplifikation auf transkribierte Bereiche (Exons) untersucht. Es konnten insgesamt 67 putative 'Exons' isoliert werden, von denen einige bereits bekannten Genen (ZNF141, PDEB, MYL5, GAK, DAGK4 und FGFR3) entsprechen. Zwei dieser Gene konnten im Rahmen dieser Arbeit erstmals (DAGK4) bzw. genauer (GAK) in die distale Region 4p16.3 kartiert werden. Die restlichen Exons können aufgrund von Homologievergleichen und/oder EST-cDNA-Homologien vermutlich neuen Genen oder auch Pseudogenen (z. B. YWEE1hu) zugeordnet werden. Durch die im Verlaufe der vorliegenden Arbeit publizierte weitere Eingrenzung der WHSCR auf einen 165 Kb-großen Bereich proximal des FGFR3-Gens konzentrierten sich weitere Untersuchungen auf die detaillierte Analyse der WHSCR zwischen dem Marker D4S43 und FGFR3. Mit Hilfe von Exonamplifikation bzw. computergestützter Auswertung vorliegender Sequenzdaten aus diesem Bereich ('GRAIL', 'GENSCAN' und Homologievergleiche in den EST-Datenbanken des NCBI) konnten mehrere neue Gene identifiziert werden. In distaler-proximaler Reihenfolge handelt es sich dabei um die Gene LETM1, 51, 43, 45, 57 und POL4P. LETM1 kodiert für ein putatives Transmembran-Protein mit einem Leucin-Zipper- und zwei EF-Hand-Motiven und könnte aufgrund seiner möglichen Beteiligung an der Ca2+-Homeostase und/oder der Signal-transduktion zu Merkmalen des WHS (Krampfanfällen, mentale Retardierung und muskuläre Hypotonie) beitragen. Das Gen 51 entspricht einem in etwa zeitgleich durch Stec et al. (1998) und Chesi et al. (1998) als WHSC1 bzw. MMSET bezeichnetem Gen und wurde daher nicht weiter charakterisiert. Es wird genauso wie das Gen 43, das zeitgleich von Wright et al. (1999b) als WHSC2 beschrieben werden konnte und eine mögliche Rolle bei der Transkriptionselongation spielt, ubiquitär exprimiert. Das in der vorliegenden Arbeit identifizierte Gen 45 zeigt demgegenüber ein ausgesprochen spezifisches Expressionsmuster (in Nervenzellen des Gehirns sowie in Spermatiden). Dies stellt zusammen mit der strukturellen Ähnlichkeit des putativen Genprodukts zu Signalmolekülen einen interessanten Zusammenhang zu Merkmalen des WHS (beispielsweise Kryptorchismus, Uterusfehlbildungen oder auch neurologische Defekte) her. Demgegenüber handelt es sich bei dem Gen 57 möglicherweise um ein trunkiertes Pseudogen des eRFS-Gens auf Chromosom 6q24 (Wallrapp et al., 1998). Das POL4P-Gen schließlich stellt allein aufgrund seiner genomischen Lokalisation sowie seiner möglichen Funktion (als DNA-Polymerase-ähnliches Gen) kein gutes Kandidatengen für spezifische Merkmale des Syndroms dar und wurde daher nicht im Detail charakterisiert. Um die Beteiligung der Gene an der Ätiologie und Pathogenese des Syndroms zu verstehen, ist die Entwicklung eines Mausmodells (über das Einfügen gezielter Deletionen in das Mausgenom) geplant. Um dies zu ermöglichen, wurde in der vorliegenden Arbeit die Charakterisierung der orthologen Region bei der Maus vorgenommen. Zunächst wurden die orthologen Gene der Maus (Letm1, Whsc1, Gen 43 (Whsc2h), Gen 45 und Pol4p) identifiziert. Durch die Erstellung sowie die genaue Kartierung eines murinen genomischen P1/PAC-Klon-Contigs konnte gezeigt werden, daß die murinen Gene Fgfr3, Letm1, Whsc1, Gen 43 (Whsc2h), Gen 45 und Pol4p sowie einige weitere der überprüften EST-cDNA-Klone der Maus in einem durchgehenden Syntänieblock zwischen Mensch (POL4P bis FGFR3) und Maus (Mmu 5.20) enthalten sind, der in seiner genomischen Ausdehnung in etwa den Verhältnissen beim Menschen (zwischen POL4P und FGFR3) entspricht.

Das Elektronentransportsystem von E. coli enthält zwei verschiedene NADH-Dehydrogenasen. Die NADH-DehydrogenaseI (nuoA-N) koppelt im Gegensatz zur NADH-DehydrogenaseII die Oxidation von NADH an eine Protonentranslokation und trägt zur Energiekonservierung bei. Die NADH-DehydrogenaseI wird über die Promotoren P1 und P2 exprimiert und besitzt mehrere Bindestellen für verschiedene Regulatoren.Die separate Klonierung der Promotoren, lacZ-Fusionen, Inaktivierung von Transkriptionsfaktoren, sowie die Nutzung mutierter Regulatorbindestellen in vivo zeigen, dass P1 im wesentlichen die Expressionshöhe bestimmt und ist unter aeroben und anaeroben Bedingungen aktiv. P2 trägt in wesentlich geringerem Maße als P1 zur Expression des Enzyms bei. Er ist stark abhängig von ArcA und IHF. Beide Promotoren wirken nicht additiv.Unter anaeroben Bedingungen wird die Transkription von nuo durch das Zweikomponenten-System ArcB/A reprimiert. ArcA bindet unabhängig und mit unterschiedlicher Affinität an die beiden Bindestellen arc1 und arc2. Von den 8 ArcA-Konsensussequenzen führen nur Mutationen der Konsensussequenzen arc1ab in vitro zu verminderter Bindungsaffinität von ArcA an die Bindestelle arc1. Dieselben führen in vivo unter anaeroben Bedingungen zur Derepression des Promotors P1 bzw. P1+P2. Unter aeroben Bedingungen zeigen nur Mutationen in arc2 eine Derepression, die nicht durch ArcA vermittelt wird. Der veröffentliche ArcA-Konsensus scheint deshalb hier in dieser einfachen Form nicht gültig zu sein.

Peptides presented by MHC class I molecules for CTL recognition are derived mainly from cytosolic proteins. For antigen presentation on the cell surface, epitopes require correct processing by cytosolic and ER proteases, efficient TAP transport and MHC class I binding affinity. The efficiency of epitope generation depends not only on the epitope itself, but also on its flanking regions. In this project, the influence of the C-terminal region of the model epitope SIINFEKL (S8L) from chicken ovalbumin (aa 257-264) on antigen processing has been investigated. S8L is a well characterized epitope presented on the murine MHC class I molecule, H-2Kb. The Flp-In 293Kb cell line was transfected with different constructs each enabling the expression of the S8L sequence with different defined C-terminal flanking regions. The constructs differed at the two first C-terminal positions after the S8L epitope, so called P1’ and P2’. At these sites, all 20 amino acids were exchanged consecutively and tested for their influence on H-2Kb/S8L presentation on the cell surface of the Flp-In 293Kb cells. The detection of this complex was performed by immunostaining and flow cytometry. The prevailing assumption is that proteasomal cleavages are exclusively responsible for the generation of the final C-termini of CTL epitopes. Nevertheless, recent publications showed that TPPII (tripeptidyl peptidase II) is required for the generation of the correct C-terminus of the HLA-A3-restricted HIV epitope Nef(73-82). With this background, the dependence of the S8L generation on proteasomal cleavage of the designed constructs was characterized using proteasomal inhibitors. The results obtained indicate that it is crucial for proteasomal cleavage, which amino acid is flanking the C-terminus of an epitope. Furthermore, partially proteasome independent S8L generation from specific S8L-precursor peptides was observed. Hence, the possibility of other existing endo- or carboxy-peptidases in the cytosol that could be involved in the correct trimming of the C-terminus of antigenic peptides for MHC class I presentation was investigated, performing specific knockdowns and using inhibitors against the target peptidases. In parallel, a purification strategy to identify the novel peptidase was established. The purified peaks showing an endopeptidase activity were further analyzed by mass spectrometry and some potential peptidases (like e.g. Lon) were identified, which have to be further characterized.

Die Metalloproteasen Meprin α und Meprin β sind an essentiellen (patho)physiologischen Prozessen beteiligt. Um die Funktion dieser Proteasen zu verstehen, ist es von Bedeutung, sie nicht isoliert, sondern im gesamten proteolytischen Netzwerk zu betrachten.rnDie Meprine werden in einer Vielzahl von Geweben, in Leukozyten, aber auch in Krebszellen exprimiert. In der Haut konnten die beiden Enzyme in unterschiedlichen dermalen Schichten detektiert werden, wo sie u.a. an der Kollagenassemblierung durch Abspaltung der Propeptide beteiligt sind. rnIm Zuge von Proteomics Analysen konnten mehr als 3000 proteolytische Schnittstellen von fünf Astacin-Metalloproteasen (Meprin α, Meprin β, Astacin, LAST und LAST_MAM) in Peptiden und nativen Substraten identifiziert werden und somit eine Aussage über die Spaltspezifität getroffen werden. In der vorliegenden Arbeit konnten diese Spaltspezifitäten mit Hilfe von fluorogenen Substraten in vitro verifiziert werden. Bemerkenswert hierbei ist die starke Präferenz der beiden Meprine und LAST_MAM für die Aminosäuren Aspartat und Glutamat in der P1‘ Position. rnMeprine werden als Zymogene exprimiert und müssen durch proteolytische Prozessierung einer tryptischen Protease aktiviert werden. Ein Schwerpunkt der vorliegenden Arbeit waren Aktivitätsbestimmungen beider Meprine unter Berücksichtigung potentieller Aktivatoren und Substrate. Es konnten die kallikrein-related peptidases (KLK) 4, 5 und 8 als spezifische Aktivatoren identifiziert werden, wobei nur KLK5 beide Proteasen aktiviert. Sowohl KLK4 als auch KLK8 sind lediglich in der Lage, das Propeptid von Meprin β abzuspalten. Außerdem konnte biochemisch und mittels Proteomics gezeigt werden, dass proKLK7 von Meprin β prozessiert wird. Durch N-terminale Sequenzierung wurde eine Schnittstelle zwei Aminosäuren N-terminal der eigentlichen Aktivierungsstelle identifiziert. Dieser Schritt beschleunigt die Aktivierung von KLK7, wenn durch Trypsin noch das verbliebene Dipeptid abgespalten wird. rnDa einige Vertreter der humanen kallikrein-related peptidases (KLK) als Meprin-Aktivatoren identifiziert werden konnten, sollten diese im Zuge dieser Arbeit im Modellorganismus Danio rerio untersucht werden. Durch in silico und RT-PCR Analysen konnte gezeigt werden, dass keine funktionellen KLK-Homologe im Zebrafisch codiert sind. Da somit andere tryptische Proteasen an der Aktivierung der Meprine beteiligt sein müssen, wurde die Transmembran-Serinprotease TMPRSS4 analysiert. In der Tat zeigte die Reduktion des Expressionslevels von TMPRSS4 durch Morpholino-Injektion drastische Störungen in der embryonalen Entwicklung von Zebrabärblingen. Mittels Licht- und Rasterelektronenmikroskopie ließ sich eine Fehlbildung der epidermalen Haut bis zu einem Ablösen der Keratinozyten von dem darunter liegenden Gewebe feststellen. rn

Die Zinkendopeptidasen Meprin α und β sind Schlüsselkomponenten in patho(physiologischen) Prozessen wie Entzündung, Kollagenassemblierung und Angiogenese. Nach ihrer Entdeckung in murinen Bürstensaummembranen und humanen Darmepithelien, wurden weitere Expressionsorte identifiziert, z.B. Leukozyten, Krebszellen und die humane Haut. Tiermodelle, Zellkulturen und biochemische Analysen weisen auf Funktionen der Meprine in der Epithelialdifferenzierung, Zellmigration, Matrixmodellierung, Angiogenese, Bindegewebsausbildung und immunologische Prozesse hin. Dennoch sind ihre physiologischen Substrate weitgehend noch unbekannt. Massenspektrometrisch basierte Proteomics-Analysen enthüllten eine einzigartige Spaltspezifität für saure Aminosäurereste in der P1´ Position und identifizierten neue biologische Substratkandidaten. Unter den 269 extrazellulären Proteinen, die in einem Substratscreen identifiziert wurden, stellten sich das amyloid precursor protein (APP) and ADAM10 (a disintegrin and metalloprotease 10) als sehr vielversprechende Kandidaten heraus. Mehrere Schnittstellen innerhalb des APP Proteins, hervorgerufen durch verschiedenen Proteasen, haben unterschiedlichen Auswirkungen zur Folge. Die β-Sekretase BACE (β-site APP cleaving enzyme) prozessiert APP an einer Schnittstelle, welche als initialer Schritt in der Entwicklung der Alzheimer Erkrankung gilt. Toxische Aβ (Amyloid β)-Peptide werden in den extrazellulären Raum freigesetzt und aggregieren dort zu senilen Plaques. Membran verankertes Meprin β hat eine β-Sekretase Aktivität, die in einem Zellkultur-basierten System bestätigt werden konnte. Die proteolytische Effizienz von Meprin β wurde in FRET (Fluorescence Resonance Energy Transfer)-Analysen bestimmt und war um den Faktor 104 höher als die von BACE1. Weiterhin konnte gezeigt werden, dass Meprin β die ersten zwei Aminosäuren prozessiert und somit aminoterminal einen Glutamatrest freisetzt, welcher nachfolgend durch die Glutaminylzyklase in ein Pyroglutamat zykliert werden kann. Trunkierte Aβ-Peptide werden nur in Alzheimer Patienten generiert. Aufgrund einer erhöhten Hydrophobie weisen diese Peptide eine höhere Tendenz zur Aggregation auf und somit eine erhöhte Toxizität. Bis heute wurde keine Protease identifiziert, welche diese Schnittstelle prozessiert. Die Bildung der Meprin vermittelten N-terminalen APP Fragmenten wurde in vitro und in vivo detektiert. Diese N-APP Peptide hatten keine cytotoxischen Auswirkungen auf murine und humane Gehirnzellen, obwohl zuvor N-APP als Ligand für den death receptor (DR) 6 identifiziert wurde, der für axonale Degenerationsprozesse verantwortlich ist. rnIm nicht-amyloidogenen Weg prozessiert ADAM10 APP und entlässt die Ektodomäne von der Zellmembran. Wir konnten das ADAM10 Propeptid als Substrat von Meprin β identifizieren und in FRET Analysen, in vitro und in vivo zeigen, dass die Meprin vermittelte Prozessierung zu einer erhöhten ADAM10 Aktivität führt. Darüber hinaus wurde ADAM10 als Sheddase für Meprin β identifiziert. Shedding konnte durch Phorbol 12-myristate 13-acetate (PMA) oder durch das Ionophor A23187 hervorgerufen werden, sowie durch ADAM10 Inhibitoren blockiert werden. rnDiese Arbeit konnte somit ein komplexes proteolytisches Netwerk innerhalb der Neurophysiologie aufdecken, welches für die Entwicklung der Alzheimer Demenz wichtig sein kann.rn

Long-term potentiation in the neonatal rat rnbarrel cortex in vivo rnLong-term potentiation (LTP) is important for the activity-dependent formation of early cortical circuits. In the neonatal rodent barrel cortex LTP has been so far only studied in vitro. I combined voltage-sensitive dye imaging with extracellular multi-electrode recordings to study whisker stimulation-induced LTP for both the slope of field potential and the number of multi-unit activity in the whisker-to-barrel cortex pathway of the neonatal rat barrel cortex in vivo. Single whisker stimulation at 2 Hz for 10 min induced an age-dependent expression of LTP in postnatal day (P) 0 to P14 rats with the strongest expression of LTP at P3-P5. The magnitude of LTP was largest in the stimulated barrel-related column, smaller in the surrounding septal region and no LTP could be observed in the neighboring barrel. Current source density analyses revealed an LTP-associated increase of synaptic current sinks in layer IV / lower layer II/III at P3-P5 and in the cortical plate / upper layer V at P0-P1. This study demonstrates for the first time an age-dependent and spatially confined LTP in the barrel cortex of the newborn rat in vivo. These activity-dependent modifications during the critical period may play an important role in the development and refinement of the topographic map in the barrel cortex. (An et al., 2012)rnEarly motor activity triggered by gamma and spindle bursts in neonatal rat motor cortexrnSelf-generated neuronal activity generated in subcortical regions drives early spontaneous motor activity, which is a hallmark of the developing sensorimotor system. However, the neuronal activity patterns and functions of neonatal primary motor cortex (M1) in the early movements are still unknown. I combined voltage-sensitive dye imaging with simultaneous extracellular multi-electrode recordings in the neonatal rat S1 and M1 in vivo. At P3-P5, gamma and spindle bursts observed in M1 could trigger early paw movements. Furthermore, the paw movements could be also elicited by the focal electrical stimulation of M1 at layer V. Local inactivation of M1 could significantly attenuate paw movements, suggesting that the neonatal M1 operates in motor mode. In contrast, the neonatal M1 can also operate in sensory mode. Early spontaneous movements and sensory stimulations of paw trigger gamma and spindle bursts in M1. Blockade of peripheral sensory input from the paw completely abolished sensory evoked gamma and spindle bursts. Moreover, both sensory evoked and spontaneously occurring gamma and spindle bursts mediated interactions between S1 and M1. Accordingly, local inactivation of the S1 profoundly reduced paw stimulation-induced and spontaneously occurring gamma and spindle bursts in M1, indicating that S1 plays a critical role in generation of the activity patterns in M1. This study proposes that both self-generated and sensory evoked gamma and spindle bursts in M1 may contribute to the refinement and maturation of corticospinal and sensorimotor networks required for sensorimotor coordination.rn

BACKGROUND: Porcine IGF2 and the H19 genes are imprinted. The IGF2 is paternally expressed, while the H19 gene is maternally expressed. Extensive studies in mice established a boundary model indicating that the H19 differentially methylated domain (DMD) controls, upon binding with the CTCF protein, reciprocal imprinting of the IGF2 and the H19 genes. IGF2 transcription is tissue and development specific involving the use of 4 promoters. In the liver of adult Large White boars IGF2 is expressed from both parental alleles, whereas in skeletal muscle and kidney tissues we observed variable relaxation of IGF2 imprinting. We hypothesized that IGF2 expression from both paternal alleles and relaxation of IGF2 imprinting is reflected in differences in DNA methylation patterns at the H19 DMD and IGF2 differentially methylated regions 1 and 2 (DMR1 and DMR2). RESULTS: Bisulfite sequencing analysis did not show any differences in DNA methylation at the three porcine CTCF binding sites in the H19 DMD between liver, muscle and kidney tissues of adult pigs. A DNA methylation analysis using methyl-sensitive restriction endonuclease SacII and 'hot-stop' PCR gave consistent results with those from the bisulfite sequencing analysis. We found that porcine H19 DMD is distinctly differentially methylated, at least for the region formally confirmed by two SNPs, in liver, skeletal muscle and kidney of foetal, newborn and adult pigs, independent of the combined imprinting status of all IGF2 expressed transcripts. DNA methylation at CpG sites in DMR1 of foetal liver was significantly lower than in the adult liver due to the presence of hypomethylated molecules. An allele specific analysis was performed for IGF2 DMR2 using a SNP in the IGF2 3'-UTR. The maternal IGF2 DMR2 of foetal and newborn liver revealed a higher DNA methylation content compared to the respective paternal allele. CONCLUSIONS: Our results indicate that the IGF2 imprinting status is transcript-specific. Biallelic IGF2 expression in adult porcine liver and relaxation of IGF2 imprinting in porcine muscle were a common feature. These results were consistent with the IGF2 promoter P1 usage in adult liver and IGF2 promoter P2, P3 and P4 usages in muscle. The results showed further that bialellic IGF2 expression in liver and relaxation of imprinting in muscle and kidney were not associated with DNA methylation variation at and around at least one CTCF binding site in H19 DMD. The imprinting status in adult liver, muscle and kidney tissues were also not reflected in the methylation patterns of IGF2 DMRs 1 and 2.

Calculation of electrolyte-free water clearance (EFWC) allows for quantification of renal losses of free water and was shown to be helpful in the differential diagnosis of dysnatremias and might help in the correction of the electrolyte disorders. A modified EFWC formula (MEFWC) was described to be more accurate than the conventional one; however, it has never been evaluated clinically.
