A escola portuguesa do século XXI tem sido alvo de reformas sucessivas procedentes de imposições económicas, políticas e sociais, sem o devido acompanhamento de inovação bottom up. A mudança decorrente tem-se processado sobretudo formalmente (inovação top down), menosprezando, ainda em larga escala, as pessoas e o seu capital (intelectual, emocional, social e psicológico), num momento em que a globalização impulsiona a transição do paradigma funcional para o paradigma das competências. Sendo os indivíduos parte integrante das soluções e êxitos organizacionais, bem como cada vez mais manifesta a preocupação dos líderes com a eficácia, eficiência e qualidade do serviço prestado ao nível das escolas de interesse público, direcionamos a análise para a gestão dos recursos humanos, mais particularmente ao nível da dualidade: instabilidade na carreira docente/estabilidade do diretor de turma. O interesse supremo da temática reside nas potencialidades deste cargo de gestão intermédia na concretização da missão da escola, a partir do papel que o seu detentor assume na teia de relações em que se insere por imposição das suas funções, e que pode fomentar quando dotado de determinadas características. Este estudo de caso, de natureza essencialmente quantitativa, traduz a análise crítica acerca do impacto da continuidade do desempenho do cargo ao longo do 2.º ciclo do ensino básico no sucesso educativo dos alunos, partindo da análise dos resultados de medidas adotadas no ano escolar 2010/2011. Para o efeito recorreu-se à análise do aproveitamento, da atuação do conselho de turma e da comunicação escola-família em seis turmas do sexto ano de escolaridade, três com o mesmo diretor de turma, de uma escola pública do concelho de Câmara de Lobos. Concluímos que a manutenção do diretor de turma nas turmas visadas teve repercussões na vertente relacional, principalmente, não tendo sido confirmada a conexão entre a continuidade do profissional que assume o cargo ao longo do ciclo e os resultados académicos dos discentes, pese embora a opinião contrária de 87,9% dos docentes inquiridos, e de a maioria quer dos estudantes, quer dos seus encarregados de educação, ser favorável à sua prossecução.

Notre système visuel extrait d'ordinaire l'information en basses fréquences spatiales (FS) avant celles en hautes FS. L'information globale extraite tôt peut ainsi activer des hypothèses sur l'identité de l'objet et guider l'extraction d'information plus fine spécifique par la suite. Dans les troubles du spectre autistique (TSA), toutefois, la perception des FS est atypique. De plus, la perception des individus atteints de TSA semble être moins influencée par leurs a priori et connaissances antérieures. Dans l'étude décrite dans le corps de ce mémoire, nous avions pour but de vérifier si l'a priori de traiter l'information des basses aux hautes FS était présent chez les individus atteints de TSA. Nous avons comparé le décours temporel de l'utilisation des FS chez des sujets neurotypiques et atteints de TSA en échantillonnant aléatoirement et exhaustivement l'espace temps x FS. Les sujets neurotypiques extrayaient les basses FS avant les plus hautes: nous avons ainsi pu répliquer le résultat de plusieurs études antérieures, tout en le caractérisant avec plus de précision que jamais auparavant. Les sujets atteints de TSA, quant à eux, extrayaient toutes les FS utiles, basses et hautes, dès le début, indiquant qu'ils ne possédaient pas l'a priori présent chez les neurotypiques. Il semblerait ainsi que les individus atteints de TSA extraient les FS de manière purement ascendante, l'extraction n'étant pas guidée par l'activation d'hypothèses.

This paper combines the idea of a hierarchical distributed genetic algorithm with different inter-agent partnering strategies. Cascading clusters of sub-populations are built from bottom up, with higher-level sub-populations optimising larger parts of the problem. Hence higher-level sub-populations search a larger search space with a lower resolution whilst lower-level sub-populations search a smaller search space with a higher resolution. The effects of different partner selection schemes for (sub-)fitness evaluation purposes are examined for two multiple-choice optimisation problems. It is shown that random partnering strategies perform best by providing better sampling and more diversity.

The article includes the development of the thesis that the analysis carried out in the framework of comparative education, as a discipline dealing with the phenomenon of education in the broad context, showing the "entanglement education" 1, often they provide students an important source of reflection and a kind of "discovery" of what they experience, but can not be called. Knowledge and understanding of the impact of a number of conditions for the development of education, in particular the political and economic although it is only a precondition, may encourage the development of a sense of agency and taking intentional actions aimed positive bottom-up change in the environment of human life.

Re-creating and understanding the origin of life represents one of the major challenges facing the scientific community. We will never know exactly how life started on planet Earth, however, we can reconstruct the most likely chemical pathways that could have contributed to the formation of the first living systems. Traditionally, prebiotic chemistry has investigated the formation of modern life’s precursors and their self-organisation under very specific conditions thought to be ‘plausible’. So far, this approach has failed to produce a living system from the bottom-up. In the work presented herein, two different approaches are employed to explore the transition from inanimate to living matter. The development of microfluidic technology during the last decades has changed the way traditional chemical and biological experiments are performed. Microfluidics allows the handling of low volumes of reagents with very precise control. The use of micro-droplets generated within microfluidic devices is of particular interest to the field of Origins of Life and Artificial Life. Whilst many efforts have been made aiming to construct cell-like compartments from modern biological constituents, these are usually very difficult to handle. However, microdroplets can be easily generated and manipulated at kHz rates, making it suitable for high-throughput experimentation and analysis of compartmentalised chemical reactions. Therefore, we decided to develop a microfluidic device capable of manipulating microdroplets in such a way that they could be efficiently mixed, split and sorted within iterative cycles. Since no microfluidic technology had been developed before in the Cronin Group, the first chapter of this thesis describes the soft lithographic methods and techniques developed to fabricate microfluidic devices. Also, special attention is placed on the generation of water-in-oil microdroplets, and the subsequent modules required for the manipulation of the droplets such as: droplet fusers, splitters, sorters and single/multi-layer micromechanical valves. Whilst the first part of this thesis describes the development of a microfluidic platform to assist chemical evolution, finding a compatible set of chemical building blocks capable of reacting to form complex molecules with endowed replicating or catalytic activity was challenging. Abstract 10 Hence, the second part of this thesis focuses on potential chemistry that will ultimately possess the properties mentioned above. A special focus is placed on the formation of peptide bonds from unactivated amino acids, despite being one of the greatest challenges in prebiotic chemistry. As opposed to classic prebiotic experiments, in which a specific set of conditions is studied to fit a particular hypothesis, we took a different approach: we explored the effects of several parameters at once on a model polymerisation reaction, without constraints on hypotheses on the nature of optimum conditions or plausibility. This was facilitated by development of a new high-throughput automated platform, allowing the exploration of a much larger number of parameters. This led us to discover that peptide bond formation is less challenging than previously imagined. Having established the right set of conditions under which peptide bond formation was enhanced, we then explored the co-oligomerisation between different amino acids, aiming for the formation of heteropeptides with different structure or function. Finally, we studied the effect of various environmental conditions (rate of evaporation, presence of salts or minerals) in the final product distribution of our oligomeric products.

En esta tesis se realiza un estudio contrastivo de los textos en español y alemán para el contrato de arrendamiento por temporada, centrándose en el caso de alojamiento no compartido en una casa rural, donde se muestran las influencias que existen desde el punto de vista cultural, legal y social en los textos tanto a nivel de macroestructural como a nivel de microfunciones del lenguaje. En el trabajo se exponen las dificultades para realizar un estudio a nivel microfuncional completo, por lo que se centra, una vez establecido un contrato tipo a nivel macroestructural, en los aspectos más importantes dentro del estudio realizado: las microfunciones temporales y condicionales. El trabajo se ha dividido en dos partes principales para las que se justifica un estudio bottom-up. Para el análisis de los textos se ha utilizado la herramienta Atlas.ti incluyéndose en los anexos toda la documentación generada con dicho software. Las conclusiones principales que se desprenden de este trabajo son: El contrato de alojamiento turístico extrahotelero en español está en desuso, por lo que es necesario usar el contrato de arrendamiento por temporada. En el caso alemán, se usa el «Mietvertrag für ein Ferienhaus oder eine Ferienwohnung» (arrendamiento de un chalé vacacional o de un apartamento turístico) que es un subgénero de «Beherbergungsvertrag» (contrato de alojamiento u hospedaje). Ambos contratos se someten a la legislación arrendaticia. Es necesario centrarse en ramas muy limitadas de texto para poder elaborar un corpus comparable, por lo que se debe usar un planteamiento empírico con el género textual de contrato en el primer nivel y el análisis de las microfunciones en segundo nivel para poder confeccionar una tipología contrastiva. Es imprescindible incluir los aspectos culturales de las lenguas a comparar, ya que el texto no se debe estudiar solamente bajo aspectos lingüísticos. El hecho de que existan términos en un idioma que no hay en otro a nivel léxico se reconfirma a nivel textual, mejor dicho a nivel de género o a nivel de subgénero de texto. Los contratos, al estar dentro de un ordenamiento jurídico no son textos estables, los cambios en legislación influyen en los textos o incluso en su desaparición en un determinado contexto comunicativo. Un mismo género de texto no supone coincidencia a nivel de macroestructura, existen bloques comunes pero también diferencias importantes por legislación, convenciones culturales, normativas europeas, etc. La determinación de una microfunción depende del género textual analizado. En el caso de las microfunciones temporales, para el género textual analizado, se definen siete criterios temporales. Frente a la clasificación jerarquizada en tres niveles (funciones generales, subfunciones y microfunciones), se plantea establecer las microfunciones dominantes del texto para confeccionar más fácilmente una tipología textual.

Humans use their grammatical knowledge in more than one way. On one hand, they use it to understand what others say. On the other hand, they use it to say what they want to convey to others (or to themselves). In either case, they need to assemble the structure of sentences in a systematic fashion, in accordance with the grammar of their language. Despite the fact that the structures that comprehenders and speakers assemble are systematic in an identical fashion (i.e., obey the same grammatical constraints), the two ‘modes’ of assembling sentence structures might or might not be performed by the same cognitive mechanisms. Currently, the field of psycholinguistics implicitly adopts the position that they are supported by different cognitive mechanisms, as evident from the fact that most psycholinguistic models seek to explain either comprehension or production phenomena. The potential existence of two independent cognitive systems underlying linguistic performance doubles the problem of linking the theory of linguistic knowledge and the theory of linguistic performance, making the integration of linguistics and psycholinguistic harder. This thesis thus aims to unify the structure building system in comprehension, i.e., parser, and the structure building system in production, i.e., generator, into one, so that the linking theory between knowledge and performance can also be unified into one. I will discuss and unify both existing and new data pertaining to how structures are assembled in understanding and speaking, and attempt to show that the unification between parsing and generation is at least a plausible research enterprise. In Chapter 1, I will discuss the previous and current views on how parsing and generation are related to each other. I will outline the challenges for the current view that the parser and the generator are the same cognitive mechanism. This single system view is discussed and evaluated in the rest of the chapters. In Chapter 2, I will present new experimental evidence suggesting that the grain size of the pre-compiled structural units (henceforth simply structural units) is rather small, contrary to some models of sentence production. In particular, I will show that the internal structure of the verb phrase in a ditransitive sentence (e.g., The chef is donating the book to the monk) is not specified at the onset of speech, but is specified before the first internal argument (the book) needs to be uttered. I will also show that this timing of structural processes with respect to the verb phrase structure is earlier than the lexical processes of verb internal arguments. These two results in concert show that the size of structure building units in sentence production is rather small, contrary to some models of sentence production, yet structural processes still precede lexical processes. I argue that this view of generation resembles the widely accepted model of parsing that utilizes both top-down and bottom-up structure building procedures. In Chapter 3, I will present new experimental evidence suggesting that the structural representation strongly constrains the subsequent lexical processes. In particular, I will show that conceptually similar lexical items interfere with each other only when they share the same syntactic category in sentence production. The mechanism that I call syntactic gating, will be proposed, and this mechanism characterizes how the structural and lexical processes interact in generation. I will present two Event Related Potential (ERP) experiments that show that the lexical retrieval in (predictive) comprehension is also constrained by syntactic categories. I will argue that the syntactic gating mechanism is operative both in parsing and generation, and that the interaction between structural and lexical processes in both parsing and generation can be characterized in the same fashion. In Chapter 4, I will present a series of experiments examining the timing at which verbs’ lexical representations are planned in sentence production. It will be shown that verbs are planned before the articulation of their internal arguments, regardless of the target language (Japanese or English) and regardless of the sentence type (active object-initial sentence in Japanese, passive sentences in English, and unaccusative sentences in English). I will discuss how this result sheds light on the notion of incrementality in generation. In Chapter 5, I will synthesize the experimental findings presented in this thesis and in previous research to address the challenges to the single system view I outlined in Chapter 1. I will then conclude by presenting a preliminary single system model that can potentially capture both the key sentence comprehension and sentence production data without assuming distinct mechanisms for each.

This thesis pursuits to contextualize the theoretical debate between the implementation of public education policy of the Federal Government focused in a distance learning and legal foundations for its enforcement, in order to raise questions and comments on the topic in question. Its importance is back to provide scientific input and can offer to the academy, particularly in the UFRN, and elements of society to question and rethink the complex relationship between the socio-economic and geographic access to higher education. It consists of a descriptive study on the institutionalization of distance education in UFRN as a mechanism for expanding access to higher education, for both, the research seeks to understand if the distance undergraduate courses offered by the UAB system and implemented at UFRN, promote expanding access to higher education, as it is during implementation that the rules, routines and social processes are converted from intentions to action. The discussion of this study lasted between two opposing views of Implementation models: Top-down and Bottom-up. It is worth noting that the documents PNE, PDE and programs and UAB MEETING reflect positively in improving the educational level of the population of the country It is a qualitative study, using the means Bibliographic, Document and Field Study, where they were performed 04 (four) in 2010 interviews with the management framework SEDIS / UAB in UFRN. The data were analyzed and addressed through techniques: Document Analysis and Content Analysis. The results show that the process of implementation of distance education at UFRN is in progress. According to our results, the research objective is achieved, but there was a need to rethink the conditions of the infrastructure of poles, the structure of the academic calendar, the management of the SEDIS UFRN, regarding the expansion of existing vacancies and the supply of new courses by the need for a redesign as the Secretariat's ability to hold the offerings of undergraduate courses offered by the Federal Government to be implemented in the institution. It was also found that levels of evasion still presents a challenge to the teaching model. Given the context, we concluded that the greatest contribution of UAB and consequently UFRN by distance learning for undergraduate courses (Bachelor in Mathematics, Physics, Chemistry, Geography and Biological Sciences, beyond the bachelor's degrees in Business and Public Administration ) is related to increasing the number of vacancies and accessibility of a population that was previously deprived of access to university

Los factores que inciden en el proceso de desarrollo de los territorios rurales y que explican el éxito o el fracaso de las estrategias impulsadas desde abajo (bottom-up) o inducidas desde arriba (top-down), han preocupado desde hace varias décadas a los analistas, que observan las limitaciones de los enfoques del “desarrollo territorial rural” para aprehender la complejidad de dicho proceso. Habiéndose centrado, sobre todo, en el ámbito de las políticas públicas y sus efectos en el desarrollo de los territorios rurales, los analistas del desarrollo han visto la necesidad de apoyarse en otras perspectivas que capten las dinámicas que acontecen en el ámbito de la sociedad civil local, tanto en lo que se refiere a las relaciones entre los diversos actores socioeconómicos e institucionales presentes en el territorio, como a su interacción con los organismos públicos encargados de implementar dichas políticas. El objetivo general de esta tesis doctoral ha sido analizar las dinámicas sociales que surgen en espacios naturales sometidos a políticas de gestión y regulación, mostrando el grado de influencia que tienen en la aplicación de esas políticas las diversas redes en que se organizan las poblaciones locales. De la investigación empírica realizada y de su integración en el marco teórico utilizado, hemos podido extraer resultados referidos a la realidad concreta y localizada de la REBISE que muestran cómo es que conciliar los objetivos de la “conservación” y el “desarrollo” en territorios poblados por comunidades locales estrechamente vinculadas a espacios naturales, exige abordar de forma integral los problemas ambientales, sociales y económicos. Tratar de alcanzar esos objetivos con políticas sectoriales conduce al fracaso de los programas de protección, ya que sólo se logran objetivos parciales y limitados. Por muy elevado que sea el valor ecológico de este tipo de espacios naturales y por muy alta que sea la protección que reciban por parte de los organismos internacionales (como ocurre con las “reservas de la biosfera” del programa MaB de la UNESCO), “conservar” estas áreas naturales no puede lograrse sin contar con la colaboración de las poblaciones locales. Esto exige combinar estrategias top-down y bottom-up buscando establecer sinergias entre los responsables públicos y los grupos sociales presentes en el territorio. De nuestra investigación se deduce la necesidad de empoderar a las comunidades locales para inducir en ellos un capital social tipo bridging dirigido a la construcción de un gran pacto territorial que trascienda los intereses particulares de cada grupo y que persiga el interés general del territorio en pro de la conservación de los recursos naturales y de la mejora del bienestar y calidad de vida de las familias que residen allí. Si no se hace así, continuarán promoviéndose proyectos “balsámicos” que paliarán a corto plazo algunos de los problemas de las poblaciones locales, pero que las mantendrán en el estancamiento y la pobreza.

Exosomes released by myeloid-derived suppressor cells (MDSC) are 30 nm in diameter extracellular vesicles that have been shown to carry biologically active proteins as well as ubiquitin molecules. Ubiquitin is known to have many functions, including involvement in the formation of exosomes, although the exact role is highly contested. In the study reported here, the proteome and ubiquitome of MDSC exosomes has been investigated by bottom-up proteomics techniques. This report identifies more than 1000 proteins contained in the MDSC exosome cargo and 489 sites of ubiquitination in more than 300 ubiquitinated proteins based on recognition of glycinylglycine tagged peptides without antibody enrichment. This has allowed extensive chemical and biological characterization of the ubiquitinated cohort compared to that of the entire protein cargo to support hypotheses on the role of ubiquitin in exosomes.
