Em eucariotos os íntrons de mRNAs codificantes de proteína são retirados e os éxons são mantidos junto ao transcrito primário pela maquinaria do spliceossomo. Este consiste em um grande complexo RNA-proteína que contém mais de 200 proteínas e cinco tipos de RNAs não codificantes, metabolicamente estáveis, conhecidos como snRNAs U, que incluem U1, U2, U4, U5 e U6. Os genes snRNA U estão presentes em múltiplas cópias dispersas no genoma de diversos eucariotos e parecem apresentar comportamento semelhante aqueles dos elementos móveis exibindo pouca conservação sintênica. No presente trabalho pretendia-se estudar a organização genômica e a localização cromossômica do gene snRNA U1 em espécies de peixes do gênero Leporinus, que é um grupo de peixes que se configura como um modelo interessante para estudo de DNAs repetitivos e evolução genômica em peixes. Porém, após diversas tentativas não foi possível amplificar este gene e então optou-se por estudar o gene snRNA U2. O DNA genômico de diferentes espécies de Leporinus e de Schizodon (grupo próximo evolutivamente) foi amplificado utilizando primers específicos para o gene, por meio da técnica de PCR e os produtos obtidos enviados para o sequenciamento. O tamanho encontrado para essa sequência correspondeu a aproximadamente 200 pb, valor esse já encontrado para outras espécies. As sequências foram analisadas e resultados não concisos das sequencias obtidas não permitiram análises subsequentes. A localização cromossômica do gene foi realizada por meio da técnica de hibridação in situ e as marcações foram evidenciadas em um par cromossômico submetacêntrico de tamanho médio em todas as espécies. A localização destas sequências não mostrou relação com cromossomos sexuais, presentes em algumas das espécies analisadas, mas demonstrou forte evidência de conservação do gene entre as diferentes espécies estudadas

The main objective of this study is to determine the activity of Po-210, Pb-210 and gross alpha/beta radioactivity in groundwater collected in Botucatu, municipality of the São Paulo state, Brazil. Samples were collected in wells to perform the radiochemical analysis. Most wells are registered in DAEE, Sabesp or CPRM. The activity of polonium was obtained by the method of alpha spectrometry, held in LABIDRO at UNESP, Rio Claro. All samples showed radioactivity values below the maximum allowed by WHO, that are: gross alpha radioactivity = 0,5 Bq/L, gross beta radioactivity = 1,0 Bq/L, Po210 = 0,2 Bq/L and Pb210 = 0,1 Bq/L. The results of hydrochemical analysis were compared with the 1469 Ordinance of the Health Ministry, and all values are bellow the maximum allowed. The data also allowed obtain the samples classification using the Piper diagram. The samples AAB, Quinta do Manacás, Faz. Quatro Irmãs, Sesi, Sitio São José and ITE are classified as calcic bicarbonate; the samples Staroup and Caio are potassic; the samples Banespa and BTC are bicarbonate and mixed interms of dissolved cations; Sitio das Palmeiras sample is sodium bicarbonate; the sample Faz. São Paulo is chlorinated calcic; the sample Sítio Btu-Pardinho chlorinated and mixed interms of dissolved cations

The aim of this work is to analyze the stability of the rotational motion’s artificial satellite using the Routh Hurwitz Algorithm (CRH) and the quaternions to describe the satellite’s attitude. This algorithm allows the investigation of the stability of the motion using the coefficients of the characteristic equation associated with the equation of the rotational motion in the linear form. The equations of the rotational motion are given by the four cinematic equations for the quaternion and the three equations of Euler for the spin velocity’s components. In the Euler equations are included the components of the gravity gradient torque (TGG) and the solar radiation torque (TRS). The TGG is generated by the difference of the Earth gravity force direction and intensity actuating on each satellite mass element and it depends on the mass distribution and the form of the satellite. The TRS is created by changing of the linear momentum, which happens due to the interactions of solar photons with the satellite surface. The equilibrium points are gotten by the equation of rotational motion and the CRH is applied in the linear form of these equations. Simulations are developed for small and medium satellites, but the gotten equilibrium points are not stable by CRH. However, when some of the eigenvalues of the characteristic equation are analyzed, it is found some equilibrium points which can be pointed out as stables for an interval of the time, due to small magnitude of the real part of these eigenvalue

O fator de início de tradução 5A (eukaryotic translation iniciation factor 5A, eIF5A) é altamente conservado entre arqueas a eucariotos, sendo que as proteínas eIF5A de Saccharomyces cerevisiae e de mamíferos são 63% idênticas. eIF5A sofre uma modificação pós-traducional única na célula, a hipusinação de um resíduo de lisina. Essa proteína já foi relacionada ao início da tradução, transporte nucleocitoplasmático, decaimento de mRNA e proliferação celular, mas a função crítica de eIF5A ainda não foi esclarecida. A depleção deste fator em S. cerevisiae leva a uma diminuição (30%) da taxa de síntese protéica, sugerindo que eIF5A seja um fator envolvido na tradução de um grupo específico de mensageiros. Dados do laboratório demonstram interação física entre eIF5A e proteínas ribossomais bem como com o fator de elongação 2 da tradução (eEF2). A interação com eEF2, sugere que eIF5A atua na etapa de elongação da tradução, ao invés do início da tradução, como proposto inicialmente. Com o objetivo de avaliar a relação de eIF5A com a etapa de elongação, foram realizadas análises de interações genéticas entre o gene codificador de eIF5A (TIF51A) e diversos genes codificadores de proteínas envolvidas na tradução. Através de análises de interações genéticas, foi observado que o mutante estável de eIF5A, tif51AK56A, apresenta um defeito de crescimento quando o mutante de eEF2, eft2H699K, está expresso em alto número de cópias, enquanto que o mutante tif51AQ22H/L93F não apresenta defeitos nesta condição. Foi observado também que o mutante tif51AQ22H/L93F apresenta um defeito de crescimento mais severo quando ocorre superexpressão de EFT2, gene codificador de eEF2. Foi observado ainda que não há complementação alélica entre os mutantes estáveis de eIF5A e que a reversibilidade do fenótipo de sensibilidade a temperatura... The Brazilian fauna has been constantly threatened by deforestation and forest fragmentation. As a result, many populations become isolated and small which negatively impacts their genetic diversity, putting them at a higher risk of extinction than large and stable populations. The aim of this work was to estimate the genetic diversity of white-lipped peccaries (Tayassu pecari) in the region of Taboco (Corguinho, MS), a fragmented area; and to compare these estimates with that obtained previously for two populations from Brazilian Pantanal, which is considered a relatively well-preserved biome and where the species is not threatened. A total of 18 blood and 72 hair samples of white-lipped peccaries had their DNA extracted and amplified for five polymorphic microsatellite loci. With the individuals identified, genetic diversity indicators (such as number of alleles, allelic richness, expected end and observed) and the inbreeding coefficient FIS were calculated. In addition, to verify if the population suffered a recent population bottleneck, we used the tests implemented in the program Bottleneck. The population of Taboco showed no evidence of recent population bottleneck (p > 0.05) or inbreeding (FIS = 0.008; p > 0.022). In addition, the levels of genetic diversity in this population (mean number of alleles = 2.60; mean allelic richness = 2.56 mean observed and expected heterozygosities = 0.45 and 0.47, respectively) were statistically similar to those found previously for the two populations from Pantanal (p > 0,05); although the region of Taboco is more impacted than the Pantanal. Even though we showed no evidence of loss of genetic diversity, it does not mean that the population is not suffering with fragmentation; but that there was not sufficient time to evidence the genetic changes. In recent years speleothem has been intensely studied due to its great potential of registering paleoclimate proxies but some considerably uncertaintiesregarding speleothem proxy interpretation still exist. In order to minimize these uncertainties, multi-proxy approach has been used. Here is presented the strontium isotope record from Bunker cave, northwest Germany. This cave was previously studied and has proved well record paleoclimate changes during Holocene for central Europe.87Sr/86Sr ratio is presented for rain water, A-horizon soil (water and leachate), C-horizon soil (water and leachate), host rock and host rock leachate, drip water and from a stalagmite (Bu4) previously dated covering the Holocene. Upper soil presented the higher values in contrast with host rock (lower values). Drip water and C-horizon presented intermediated ratios. Sr isotopesare used to track the source of 87Sr/86Sr in the Bunker system, resulting in a mixture between A-horizon soil, C-horizon soil and host rock. A decreasing trend in Bu4 indicates change in the Sr source in the system

Nuclear medicine is a medical specialty related to imagery that deals with imaging techniques, diagnosis and therapy, allowing observing the physiological state of tissues noninvasively by marking the molecules participating of these physiological processes with radioactive isotopes, thus creating the called radionuclides. The image of a radionuclide is one of the most important applications of radioactivity in nuclear medicine. The equipment’s of nuclear medicine imaging use the principle of radiation detection, turning it into an electrical signal which, through specific algorithms, allows forming tomographic images that provide information about the functional status of organs. New detection systems have been developed for tomographic acquisitions using solid state detectors. These devices use crystals of cadmium zinc telluride (CdZnTe). Some of the advantages of this detector are a significant improvement of signal to noise ratio, the increased spectral and spatial resolution, which in sum, result in greater clarity of the images obtained, opening new perspectives for imaging protocols previously unattainable. In contrast, all other gamma-cameras equipped with vacuum tubes have remained relatively unchanged for nearly fifty years. In these gamma-cameras, the images are obtained using two steps significantly less efficient: the gamma rays are converted to light through a first device, and then the light is converted into an electrical signal through a second device. One of functions the Medical Physicist is related to the quality control of equipment. This control ensures that the information and images provided are true and thus credible to be used in medical reports. The ceramic pigments are colored inorganic substances that during the process of dispersion in the ceramic glazes and subsequent calcination, are stable against physical and chemical attack. Therefore, they are crystalline compounds applied in the ceramic industry for coloring vitreous base. In this study, the Pechini method was used for obtaining the pigment CuCr2O4 with heat treatment at the temperatures of 800, 900 and 1000 ° C. The powder pigments were characterized on their structural, morphological and colorimetric aspects. The thermal analysis conducted on an amorphous precursor in a TG / DTA indicates the weight loss in the entire temperature range investigated, with characteristic exothermic peak of the elimination of the organic composition of the precursor around 300°C. The development of the crystalline phases were investigated by XRD, using a diffractometer with Cu Ka radiation and graphite monochromator, where it was observed the presence of crystalline phases corresponding to Cr2O3 and CuCr2O4.The measurements of the specific surface area of the powders pigments were carried out in an equipment Micromeritcs, model ASAP 2000, using N2 as gas of adsorption/desorption. The colorimetric measurements of the pigments were made in a colorimeter Gretac Macbeth Color-eye spectrophotometer 2180 / 2180UV in CIELAB standards. Based on the obtained results, it can be verified the thermal stability of the powder pigments of green coloration, which enables it as an alternative to the materials currently used in the manufacture of ceramic tiles.
