[ES] IPOL es una revista científica de procesamiento digital de imágenes y diversos métodos de análisis de imágenes. En cada publicación se incorpora una demo donde cualquier persona puede probar, vía web, el funcionamiento del método descrito en dicha publicación. De esta forma, se puede usar el método sin tener conocimiento de programación ni tener que instalarlo en su ordenador. En este proyecto fin de carrera se quiere desarrollar una aplicación que permita la ejecución de las demos desde un dispositivo móvil. Con ello, se pretende hacer más accesible la ejecución de algoritmo de procesamiento de imágenes y aumentar su divulgación científica.

ABSTRACT Il presente lavoro vuole introdurre la problematica del rigonfiamento del terreno a seguito di grandi scavi in argilla. Il sollevamento del terreno dopo lo scavo può passare inosservato ma sono numerosi i casi in cui il rigonfiamento dura per molti anni e addirittura decenni, Shell Centre, London, Lion Yard, Cambridge, Bell Common, London, ecc. Questo rigonfiamento il più delle volte è impedito dalla presenza di fondazioni, si genera quindi una pressione distribuita che se non considerata in fase di progetto può portare alla fessurazione della fondazione stessa. L’anima del progetto è la modellazione e l’analisi del rigonfiamento di grandi scavi in argilla, confrontando poi i risultati con i dati reali disponibili in letteratura. L’idea del progetto nasce dalla difficoltà di ottenere stime e previsioni attendibili del rigonfiamento a seguito di grandi scavi in argilla sovraconsolidata. Inizialmente ho esaminato la teoria e i fattori che influenzano il grado e la velocità del rigonfiamento, quali la rigidezza, permeabilità, fessurazione, struttura del suolo, etc. In seguito ho affrontato lo studio del comportamento rigonfiante di argille sovraconsolidate a seguito di scarico tensionale (scavi), si è evidenziata l’importanza di differenziare il rigonfiamento primario e il rigonfiamento secondario dovuto al fenomeno del creep. Il tema centrale del progetto è l’analisi numerica tramite Flac di due grandi scavi in argilla, Lion Yard, Cambridge, e, Bell Common, London. Attraverso una dettagliata analisi parametrica sono riuscito a trovare i migliori parametri che modellano il comportamento reale nei due casi in esame, in questo modo è possibile arrivare a stime e previsioni attendibili del fenomeno rigonfiante del terreno a seguito di grandi scavi. Gli scavi modellati Lion Yard e Bell Common sono rispettivamente in Gault Clay e London Clay, grazie a famosi recenti articoli scientifici sono riuscito a evidenziare la principali propietà che diversificano i due terreni in esame, tali propietà sono estremamente differenti dalle normali caratteristiche considerate per la progettazione in presenza di terreno argilloso; sono così riuscito a implementare i migliori parametri per descrivere il comportamento dei due terreni nei diversi modelli. Ho inoltre studiato l’interazione terreno-struttura, la pressione esercitata dal rigonfiamento del terreno è strettamente funzione delle caratteristiche di connesione tra fondazione superficiale e muro di sostegno, tale pressione non deve essere ignorata in fase progettuale poichè può raggiungere importanti valori. Nello scavo di Lion Yard, considerando la presenza delle fondazioni profonde ho evidenziato il fatto che il rigonfiamento crea una forza distribuita di taglio tra i pali di fondazione ed il terreno, anche tale sollecitazione dovrebbe essere considerata ai fini della progettazione. La problematica non si ferma solo sull’interazione terreno-fondazioni, infatti durante gli scavi di importanti fondazioni londinesi lo scarico tensionale ha creato uno spostamento significativo positivo verso la superfice di tratti di tunnel della metropolita, questo fenomeno può creare seri problemi di sicurezza nella rete dei trasporti pubblici. Infine sono stati messi a confronto i risultati del programma Flac con quelli di metodi semplificati, ho trovato che utilizzando il metodo iterativo di O’Brien i risultati sono simili alla realtà e il tempo di calcolo è molto inferiore di quello richiesto utilizzando Flac, 2-3 giorni. In conclusione posso affermare che grazie ad una dettagliata analisi parametrica è stato possibile stimare il rigonfiamento del terreno, argilla sovraconsolidata, nei due casi analizzati.

Background. One of the phenomena observed in human aging is the progressive increase of a systemic inflammatory state, a condition referred to as “inflammaging”, negatively correlated with longevity. A prominent mediator of inflammation is the transcription factor NF-kB, that acts as key transcriptional regulator of many genes coding for pro-inflammatory cytokines. Many different signaling pathways activated by very diverse stimuli converge on NF-kB, resulting in a regulatory network characterized by high complexity. NF-kB signaling has been proposed to be responsible of inflammaging. Scope of this analysis is to provide a wider, systemic picture of such intricate signaling and interaction network: the NF-kB pathway interactome. Methods. The study has been carried out following a workflow for gathering information from literature as well as from several pathway and protein interactions databases, and for integrating and analyzing existing data and the relative reconstructed representations by using the available computational tools. Strong manual intervention has been necessarily used to integrate data from multiple sources into mathematically analyzable networks. The reconstruction of the NF-kB interactome pursued with this approach provides a starting point for a general view of the architecture and for a deeper analysis and understanding of this complex regulatory system. Results. A “core” and a “wider” NF-kB pathway interactome, consisting of 140 and 3146 proteins respectively, were reconstructed and analyzed through a mathematical, graph-theoretical approach. Among other interesting features, the topological characterization of the interactomes shows that a relevant number of interacting proteins are in turn products of genes that are controlled and regulated in their expression exactly by NF-kB transcription factors. These “feedback loops”, not always well-known, deserve deeper investigation since they may have a role in tuning the response and the output consequent to NF-kB pathway initiation, in regulating the intensity of the response, or its homeostasis and balance in order to make the functioning of such critical system more robust and reliable. This integrated view allows to shed light on the functional structure and on some of the crucial nodes of thet NF-kB transcription factors interactome. Conclusion. Framing structure and dynamics of the NF-kB interactome into a wider, systemic picture would be a significant step toward a better understanding of how NF-kB globally regulates diverse gene programs and phenotypes. This study represents a step towards a more complete and integrated view of the NF-kB signaling system.

The comparative genomic sequence analysis of a region in human chromosome 11p15.3 and its homologous segment in mouse chromosome 7 between ST5 and LMO1 genes has been performed. 158,201 bases were sequenced in the mouse and compared with the syntenic region in human, partially available in the public databases. The analysed region exhibits the typical eukaryotic genomic structure and compared with the close neighbouring regions, strikingly reflexes the mosaic pattern distribution of (G+C) and repeats content despites its relative short size. Within this region the novel gene STK33 was discovered (Stk33 in the mouse), that codes for a serine/threonine kinase. The finding of this gene constitutes an excellent example of the strength of the comparative sequencing approach. Poor gene-predictions in the mouse genomic sequence were corrected and improved by the comparison with the unordered data from the human genomic sequence publicly available. Phylogenetical analysis suggests that STK33 belongs to the calcium/calmodulin-dependent protein kinases group and seems to be a novelty in the chordate lineage. The gene, as a whole, seems to evolve under purifying selection whereas some regions appear to be under strong positive selection. Both human and mouse versions of serine/threonine kinase 33, consists of seventeen exons highly conserved in the coding regions, particularly in those coding for the core protein kinase domain. Also the exon/intron structure in the coding regions of the gene is conserved between human and mouse. The existence and functionality of the gene is supported by the presence of entries in the EST databases and was in vivo fully confirmed by isolating specific transcripts from human uterus total RNA and from several mouse tissues. Strong evidence for alternative splicing was found, which may result in tissue-specific starting points of transcription and in some extent, different protein N-termini. RT-PCR and hybridisation experiments suggest that STK33/Stk33 is differentially expressed in a few tissues and in relative low levels. STK33 has been shown to be reproducibly down-regulated in tumor tissues, particularly in ovarian tumors. RNA in-situ hybridisation experiments using mouse Stk33-specific probes showed expression in dividing cells from lung and germinal epithelium and possibly also in macrophages from kidney and lungs. Preliminary experimentation with antibodies designed in this work, performed in parallel to the preparation of this manuscript, seems to confirm this expression pattern. The fact that the chromosomal region 11p15 in which STK33 is located may be associated with several human diseases including tumor development, suggest further investigation is necessary to establish the role of STK33 in human health.

The aim of the work is to conduct a finite element model analysis on a small – size concrete beam and on a full size concrete beam internally reinforced with BFRP exposed at elevated temperatures. Experimental tests performed at Kingston University have been used to compare the results from the numerical analysis for the small – size concrete beam. Once the behavior of the small – size beam at room temperature is investigated and switching to the heating phase reinforced beams are tested at 100°C, 200°C and 300°C in loaded condition. The aim of the finite element analysis is to reflect the three – point bending test adopted into the oven during the exposure of the beam at room temperature and at elevated temperatures. Performance and deformability of reinforced beams are straightly correlated to the material properties and a wide analysis on elastic modulus and coefficient of thermal expansion is given in this work. Develop a good correlation between the numerical model and the experimental test is the main objective of the analysis on the small – size concrete beam, for both modelling the aim is also to estimate which is the deterioration of the material properties due to the heating process and the influence of different parameters on the final result. The focus of the full – size modelling which involved the last part of this work is to evaluate the effect of elevated temperatures, the material deterioration and the deflection trend on a reinforced beam characterized by a different size. A comparison between the results from different modelling has been developed.

Articular cartilage lesions, with their inherent limited healing potential, are hard to treat and remain a challenging problem for orthopedic surgeons. Despite the development of several treatment strategies, the real potential of each procedure in terms of clinical benefit and effects on the joint degeneration processes is not clear. Aim of this PhD project was to evaluate the results, both in terms of clinical and imaging improvement, of new promising procedures developed to address the challenging cartilage pathology. Several studies have been followed in parallel and completed over the 3-year PhD, and are reported in detail in the following pages. In particular, the studies have been focused on the evaluation of the treatment indications of a scaffold based autologous chondrocyte implantation procedure, documenting its results for the classic indication of focal traumatic lesions, as well as its use for the treatment of more challenging patients, older, with degenerative lesions, or even as salvage procedure for more advanced stages of articular degeneration. The second field of study involved the analysis of the results obtained treating lesions of the articular surface with a new biomimetic osteochondral scaffold, which showed promise for the treatment of defects where the entire osteochondral unit is involved. Finally, a new minimally invasive procedure based on the use of growth factors derived from autologous platelets has been explored, showing results and underlining indicatios for the treatment of cartilage lesions and different stages of joint degeneration. These studies shed some light on the potential of the evaluated procedures, underlining good results as well as limits, they give some indications on the most appropriate candidates for their application, and document the current knowledge on cartilage treatment procedures suggesting the limitations that need to be addressed by future studies to improve the management of cartilage lesions.

Laterally loaded piles are a typical situation for a large number of cases in which deep foundations are used. Dissertation herein reported, is a focus upon the numerical simulation of laterally loaded piles. In the first chapter the best model settings are largely discussed, so a clear idea about the effects of interface adoption, model dimension, refinement cluster and mesh coarseness is reached. At a second stage, there are three distinct parametric analyses, in which the model response sensibility is studied for variation of interface reduction factor, Eps50 and tensile cut-off. In addition, the adoption of an advanced soil model is analysed (NGI-ADP). This was done in order to use the complex behaviour (different undrained shear strengths are involved) that governs the resisting process of clay under short time static loads. Once set a definitive model, a series of analyses has been carried out with the objective of defining the resistance-deflection (P-y) curves for Plaxis3D (2013) data. Major results of a large number of comparisons made with curves from API (America Petroleum Institute) recommendation are that the empirical curves have almost the same ultimate resistance but a bigger initial stiffness. In the second part of the thesis a simplified structural preliminary design of a jacket structure has been carried out to evaluate the environmental forces that act on it and on its piles foundation. Finally, pile lateral response is studied using the empirical curves.

In this work, the Generalized Beam Theory (GBT) is used as the main tool to analyze the mechanics of thin-walled beams. After an introduction to the subject and a quick review of some of the most well-known approaches to describe the behaviour of thin-walled beams, a novel formulation of the GBT is presented. This formulation contains the classic shear-deformable GBT available in the literature and contributes an additional description of cross-section warping that is variable along the wall thickness besides along the wall midline. Shear deformation is introduced in such a way that the classical shear strain components of the Timoshenko beam theory are recovered exactly. According to the new kinematics proposed, a reviewed form of the cross-section analysis procedure is devised, based on a unique modal decomposition. Later, a procedure for a posteriori reconstruction of all the three-dimensional stress components in the finite element analysis of thin-walled beams using the GBT is presented. The reconstruction is simple and based on the use of three-dimensional equilibrium equations and of the RCP procedure. Finally, once the stress reconstruction procedure is presented, a study of several existing issues on the constitutive relations in the GBT is carried out. Specifically, a constitutive law based on mirroring the kinematic constraints of the GBT model into a specific stress field assumption is proposed. It is shown that this method is equally valid for isotropic and orthotropic beams and coincides with the conventional GBT approach available in the literature. Later on, an analogous procedure is presented for the case of laminated beams. Lastly, as a way to improve an inherently poor description of shear deformability in the GBT, the introduction of shear correction factors is proposed. Throughout this work, numerous examples are provided to determine the validity of all the proposed contributions to the field.

This thesis is focused on Smart Grid applications in medium voltage distribution networks. For the development of new applications it appears useful the availability of simulation tools able to model dynamic behavior of both the power system and the communication network. Such a co-simulation environment would allow the assessment of the feasibility of using a given network technology to support communication-based Smart Grid control schemes on an existing segment of the electrical grid and to determine the range of control schemes that different communications technologies can support. For this reason, is presented a co-simulation platform that has been built by linking the Electromagnetic Transients Program Simulator (EMTP v3.0) with a Telecommunication Network Simulator (OPNET-Riverbed v18.0). The simulator is used to design and analyze a coordinate use of Distributed Energy Resources (DERs) for the voltage/var control (VVC) in distribution network. This thesis is focused control structure based on the use of phase measurement units (PMUs). In order to limit the required reinforcements of the communication infrastructures currently adopted by Distribution Network Operators (DNOs), the study is focused on leader-less MAS schemes that do not assign special coordinating rules to specific agents. Leader-less MAS are expected to produce more uniform communication traffic than centralized approaches that include a moderator agent. Moreover, leader-less MAS are expected to be less affected by limitations and constraint of some communication links. The developed co-simulator has allowed the definition of specific countermeasures against the limitations of the communication network, with particular reference to the latency and loss and information, for both the case of wired and wireless communication networks. Moreover, the co-simulation platform has bee also coupled with a mobility simulator in order to study specific countermeasures against the negative effects on the medium voltage/current distribution network caused by the concurrent connection of electric vehicles.

The association of several favorable factors has resulted in the development of a wide barchan dune field that stands out as a fundamental element in the coastal landscape of southern Santa Catarina state in Brazil. This original ecosystem is being destroyed and highly modified, due to urbanization. This work identifies and discusses its basic characteristics and analyzes the favorable factors for its preservation, in the foreseen of both a sustainable future and potential incomes from ecotourism. The knowledge of the geologic evolution allows to associate this transgressive Holocene dunes formation to more dissipative beach conditions. Spatial differences on morphodynamics are related to local and regional contrasts in the sediment budget, with an influence on gradients of wave attenuation in the inner shelf and consequently with influence in the level of coastal erosion. The link between relative sea level changes and coastal eolian sedimentation can be used to integrate coastal eolian systems to the sequence stratigraphy model. The main accumulation phase of eolian sediments would occur during the final transgressive and highstand systems tracts. Considering the global character of Quaternary relative sea level changes, the Laguna transgressive dune field should be correlated with similar eolian deposits developed along other parts of the Brazilian coast compatibles with the model of dunefield initiation during rising and highstand sea level phases.

Il presente lavoro di tesi è stato svolto presso il servizio di Fisica Sanitaria del Policlinico Sant'Orsola-Malpighi di Bologna. Lo studio si è concentrato sul confronto tra le tecniche di ricostruzione standard (Filtered Back Projection, FBP) e quelle iterative in Tomografia Computerizzata. Il lavoro è stato diviso in due parti: nella prima è stata analizzata la qualità delle immagini acquisite con una CT multislice (iCT 128, sistema Philips) utilizzando sia l'algoritmo FBP sia quello iterativo (nel nostro caso iDose4). Per valutare la qualità delle immagini sono stati analizzati i seguenti parametri: il Noise Power Spectrum (NPS), la Modulation Transfer Function (MTF) e il rapporto contrasto-rumore (CNR). Le prime due grandezze sono state studiate effettuando misure su un fantoccio fornito dalla ditta costruttrice, che simulava la parte body e la parte head, con due cilindri di 32 e 20 cm rispettivamente. Le misure confermano la riduzione del rumore ma in maniera differente per i diversi filtri di convoluzione utilizzati. Lo studio dell'MTF invece ha rivelato che l'utilizzo delle tecniche standard e iterative non cambia la risoluzione spaziale; infatti gli andamenti ottenuti sono perfettamente identici (a parte le differenze intrinseche nei filtri di convoluzione), a differenza di quanto dichiarato dalla ditta. Per l'analisi del CNR sono stati utilizzati due fantocci; il primo, chiamato Catphan 600 è il fantoccio utilizzato per caratterizzare i sistemi CT. Il secondo, chiamato Cirs 061 ha al suo interno degli inserti che simulano la presenza di lesioni con densità tipiche del distretto addominale. Lo studio effettuato ha evidenziato che, per entrambi i fantocci, il rapporto contrasto-rumore aumenta se si utilizza la tecnica di ricostruzione iterativa. La seconda parte del lavoro di tesi è stata quella di effettuare una valutazione della riduzione della dose prendendo in considerazione diversi protocolli utilizzati nella pratica clinica, si sono analizzati un alto numero di esami e si sono calcolati i valori medi di CTDI e DLP su un campione di esame con FBP e con iDose4. I risultati mostrano che i valori ricavati con l'utilizzo dell'algoritmo iterativo sono al di sotto dei valori DLR nazionali di riferimento e di quelli che non usano i sistemi iterativi.
