International Perspective The development of GM technology continues to expand into increasing numbers of crops and conferred traits. Inevitably, the focus remains on the major field crops of soybean, maize, cotton, oilseed rape and potato with introduced genes conferring herbicide tolerance and/or pest resistance. Although there are comparatively few GM crops that have been commercialised to date, GM versions of 172 plant species have been grown in field trials in 31 countries. European Crops with Containment Issues Of the 20 main crops in the EU there are four for which GM varieties are commercially available (cotton, maize for animal feed and forage, and oilseed rape). Fourteen have GM varieties in field trials (bread wheat, barley, durum wheat, sunflower, oats, potatoes, sugar beet, grapes, alfalfa, olives, field peas, clover, apples, rice) and two have GM varieties still in development (rye, triticale). Many of these crops have hybridisation potential with wild and weedy relatives in the European flora (bread wheat, barley, oilseed rape, durum wheat, oats, sugar beet and grapes), with escapes (sunflower); and all have potential to cross-pollinate fields non-GM crops. Several fodder crops, forestry trees, grasses and ornamentals have varieties in field trials and these too may hybridise with wild relatives in the European flora (alfalfa, clover, lupin, silver birch, sweet chestnut, Norway spruce, Scots pine, poplar, elm, Agrostis canina, A. stolonifera, Festuca arundinacea, Lolium perenne, L. multiflorum, statice and rose). All these crops will require containment strategies to be in place if it is deemed necessary to prevent transgene movement to wild relatives and non-GM crops. Current Containment Strategies A wide variety of GM containment strategies are currently under development, with a particular focus on crops expressing pharmaceutical products. Physical containment in greenhouses and growth rooms is suitable for some crops (tomatoes, lettuce) and for research purposes. Aquatic bioreactors of some non-crop species (algae, moss, and duckweed) expressing pharmaceutical products have been adopted by some biotechnology companies. There are obvious limitations of the scale of physical containment strategies, addressed in part by the development of large underground facilities in the US and Canada. The additional resources required to grow plants underground incurs high costs that in the long term may negate any advantage of GM for commercial productioNatural genetic containment has been adopted by some companies through the selection of either non-food/feed crops (algae, moss, duckweed) as bio-pharming platforms or organisms with no wild relatives present in the local flora (safflower in the Americas). The expression of pharmaceutical products in leafy crops (tobacco, alfalfa, lettuce, spinach) enables growth and harvesting prior to and in the absence of flowering. Transgenically controlled containment strategies range in their approach and degree of development. Plastid transformation is relatively well developed but is not suited to all traits or crops and does not offer complete containment. Male sterility is well developed across a range of plants but has limitations in its application for fruit/seed bearing crops. It has been adopted in some commercial lines of oilseed rape despite not preventing escape via seed. Conditional lethality can be used to prevent flowering or seed development following the application of a chemical inducer, but requires 100% induction of the trait and sufficient application of the inducer to all plants. Equally, inducible expression of the GM trait requires equally stringent application conditions. Such a method will contain the trait but will allow the escape of a non-functioning transgene. Seed lethality (‘terminator’ technology) is the only strategy at present that prevents transgene movement via seed, but due to public opinion against the concept it has never been trialled in the field and is no longer under commercial development. Methods to control flowering and fruit development such as apomixis and cleistogamy will prevent crop-to-wild and wild-to-crop pollination, but in nature both of these strategies are complex and leaky. None of the genes controlling these traits have as yet been identified or characterised and therefore have not been transgenically introduced into crop species. Neither of these strategies will prevent transgene escape via seed and any feral apomicts that form are arguably more likely to become invasives. Transgene mitigation reduces the fitness of initial hybrids and so prevents stable introgression of transgenes into wild populations. However, it does not prevent initial formation of hybrids or spread to non-GM crops. Such strategies could be detrimental to wild populations and have not yet been demonstrated in the field. Similarly, auxotrophy prevents persistence of escapes and hybrids containing the transgene in an uncontrolled environment, but does not prevent transgene movement from the crop. Recoverable block of function, intein trans-splicing and transgene excision all use recombinases to modify the transgene in planta either to induce expression or to prevent it. All require optimal conditions and 100% accuracy to function and none have been tested under field conditions as yet. All will contain the GM trait but all will allow some non-native DNA to escape to wild populations or to non-GM crops. There are particular issues with GM trees and grasses as both are largely undomesticated, wind pollinated and perennial, thus providing many opportunities for hybridisation. Some species of both trees and grass are also capable of vegetative propagation without sexual reproduction. There are additional concerns regarding the weedy nature of many grass species and the long-term stability of GM traits across the life span of trees. Transgene stability and conferred sterility are difficult to trial in trees as most field trials are only conducted during the juvenile phase of tree growth. Bio-pharming of pharmaceutical and industrial compounds in plants Bio-pharming of pharmaceutical and industrial compounds in plants offers an attractive alternative to mammalian-based pharmaceutical and vaccine production. Several plantbased products are already on the market (Prodigene’s avidin, β-glucuronidase, trypsin generated in GM maize; Ventria’s lactoferrin generated in GM rice). Numerous products are in clinical trials (collagen, antibodies against tooth decay and non-Hodgkin’s lymphoma from tobacco; human gastric lipase, therapeutic enzymes, dietary supplements from maize; Hepatitis B and Norwalk virus vaccines from potato; rabies vaccines from spinach; dietary supplements from Arabidopsis). The initial production platforms for plant-based pharmaceuticals were selected from conventional crops, largely because an established knowledge base already existed. Tobacco and other leafy crops such as alfalfa, lettuce and spinach are widely used as leaves can be harvested and no flowering is required. Many of these crops can be grown in contained greenhouses. Potato is also widely used and can also be grown in contained conditions. The introduction of morphological markers may aid in the recognition and traceability of crops expressing pharmaceutical products. Plant cells or plant parts may be transformed and maintained in culture to produce recombinant products in a contained environment. Plant cells in suspension or in vitro, roots, root cells and guttation fluid from leaves may be engineered to secrete proteins that may be harvested in a continuous, non-destructive manner. Most strategies in this category remain developmental and have not been commercially adopted at present. Transient expression produces GM compounds from non-GM plants via the utilisation of bacterial or viral vectors. These vectors introduce the trait into specific tissues of whole plants or plant parts, but do not insert them into the heritable genome. There are some limitations of scale and the field release of such crops will require the regulation of the vector. However, several companies have several transiently expressed products in clinical and pre-clinical trials from crops raised in physical containment.

O trabalho em turnos faz com que os ritmos humanos sejam alterados, com repercussões sobre os sistemas orgânicos e também sobre o ciclo vigília-sono, resultando em prejuízos, uma vez que o sono diurno não tem as mesmas características que o sono noturno. A equipe de enfermagem, que trabalha em turnos noturnos, merece atenção, pois estão submetidos a essa inadequação. Objetivou-se com este estudo conhecer a qualidade do sono de trabalhadores da equipe de enfermagem que trabalham nos plantões noturnos. Verificou-se que o trabalhador noturno sofre alterações significativas orgânicas, emocionais e sociais. Dessa forma, é de grande importância que haja um ajuste do sono desse profissional por meio de recomendações, que favoreçam o funcionamento adequado do organismo, como a adequação da vida social, afetiva e comportamental desse profissional.

The “Albergue Noturno de Bauru” (Night Shelter of Bauru) is a social assistance entity mantained by the “Centro Espírita Amor e Caridade (CEAC)” in partnership with the “Secretaria do Bem Estar Social (SEBES)”, that aims to temporarily shelter migrants in town and people in homeless situation. Every day, about 50 people look for the Shelter, because of several reasons. Opened in 1951, the entity – the only public service that offers overnight stay in town - surpassed social transformations over the years and never stopped its work. With the opening of its new headquarters in July 2011, it also began to develop a systematic monitoring of some sheltered people, looking for the reintegration of them into the social context. All this work has made the Night Shelter accumulate many stories, especially from those people who have gone through or are still in it. Thus, this paper aims to present, through a non-fiction novel, this scenario, exposing the history and functioning of the Bauru Night Shelter, while exposing the lives of different people who are part of this universe. Through the monitoring of several night shifts, narrated in diary format, it was possible to understand how the service started, the dilemmas involved in their development and how sheltered and volunteers build this reality.

A presença cada vez mais disseminada de organismos exóticos (muitos dos quais se tornam invasores) nas diferentes regiões do planeta levou ao surgimento de uma linha de pesquisa na ecologia voltada às invasões biológicas. E para permitir a comunicação entre autores também foi desenvolvido um arcabouço terminológico. Mas, apesar disso, a terminologia relativa às bioinvasões tem sido ignorada por boa parte dos botânicos no Brasil. Há uma boa dose de confusão entre botânicos sobre o que seja uma espécie exótica, naturalizada, invasora, daninha e ruderal, levando ao uso inconsistente da terminologia. Além disso, diferentes autores têm adotado posturas praticamente opostas ao lidar com espécies exóticas em suas áreas de estudo, seja na preparação de tratamentos taxonômicos, seja na publicação de levantamentos florísticos e fitossociológicos. Enquanto alguns pesquisadores incluem em floras mesmo espécies cultivadas que não se reproduzem, outros excluem plantas invasoras comuns e conspícuas. Nós apresentamos aqui, em português, os principais conceitos relativos ao tema da bioinvasão e chamamos a atenção dos autores brasileiros para a necessidade de utilizar de modo consistente o arcabouço terminológico já existente na literatura. Também propomos a adoção de rótulos claros para informar quais espécies são exóticas na área estudada, diferenciando-as das nativas, e sugerimos critérios para ajudar botânicos a decidirem quando uma planta exótica deve ou não ser incluída em tratamentos taxonômicos ou levantamentos de florística.

While many developed countries have invested heavily in research on plant invasions over the last 50 years, the immense region of Latin America has made little progress. Recognising this, a group of scientists working on plant invasions in Latin America met in Chile in late 2010 to develop a research agenda for the region based on lessons learned elsewhere. Our three main findings are as follows. (1) Globalisation is inevitable, but the resultant plant introductions can be slowed or prevented by effective quarantine and early intervention. Development of spatially explicit inventories, research on the invasion process and weed risk assessments can help prioritise and streamline action. (2) Eradication has limited application for plants and control is expensive and requires strict prioritisation and careful planning and evaluation. (3) Accepting the concept of novel ecosystems, new combinations of native and introduced species that no longer depend on human intervention, may help optimise invasive species management. Our vision of novel ecosystem management is through actions that: (a) maintain as much native biodiversity and ecosystem functionality as possible, (b) minimise management intervention to invasives with known impact, and (c) maximise the area of intervention. We propose the creation of a Latin American Invasive Plants Network to help focus the new research agenda for member countries. The network would coordinate research and training and establish funding priorities, develop and strengthen tools to share knowledge, and raise awareness at the community, governmental and intergovernmental levels about the social, economic and environmental costs of plant invasions.

Noninvasive molecular-imaging technologies are playing a keyrole in drug discovery, development and delivery. Positron Emission Tomography (PET) is such a molecular imaging technology and a powerful tool for the observation of various diseases. However, it is limited by the availability of agents with high selectivity to the target and a physical half-life of the used positron emitting nuclide which matches the biological half-life of the observed process. For the long lasting enrichment of antibodies in tumor tissue few suitable isotopes for PET imaging are currently available. The element arsenic provides a range of isotopes, which could be used for diagnosis and also for endoradiotherapy. This work describes the development of radiochemical separation procedures to separate arsenic isotopes in no-carrier-added (nca) purity from reactor or cyclotron irradiated targets, the development and evaluation of a labeling chemistry to attach these separated arsenic isotopes to monoclonal antibodies, the in vitro and in vivo evaluation of antibodies labeled with radioactive arsenic isotopes and the molecular imaging using small animal PET.

Die Positronen-Emissions-Tomographie (PET) ist ein leistungsstarkes, nicht-invasives, bildgebendes Verfahren in der Nuklearmedizin und hat darüber hinaus zunehmende Bedeutung in der Arzneistoffentwicklung. Zur Verbesserung des therapeutischen Index von niedermolekularen Pharmaka werden vermehrt Wirkstofftransportsysteme eingesetzt. Eine Klasse dieser Wirkstofftransportsysteme sind Liposomen. Die Weiterentwicklung der klassischen Liposomen sind sogenannte „Stealth“-Liposomen, die eine Polyethylenglykol (PEG)-Korona zur Herabsetzung der Erkennung und Ausscheidung tragen. Zur (Weiter-)Entwicklung und deren in vivo-Evaluierung bietet die PET die Möglichkeit, die Auswirkungen von strukturellen Anpassungen auf die pharmakokinetischen Eigenschaften solcher Transportsysteme zu untersuchen. Zur Evaluierung neuartiger, cholesterolverankerter, linear-hyperverzweigter Polyglycerole (Ch-PEG-hbPG) als sterisch stabilisierende Polymere in Liposomen wurden diese im Rahmen dieser Arbeit mit der prosthetischen Gruppe 18F-TEG-N3 über kupferkatalysierte Alkin-Azid Cycloaddition (CuAAC) in sehr hohen Ausbeuten radiomarkiert. Zum systematischen Vergleich des in vivo-Verhaltens wurde ebenfalls ein cholesterolbasiertes lineares PEG (Ch-PEG) mit CuAAC nahezu quantitativ radiomarkiert. Als drittes Element wurde die Direktmarkierung von Cholesterol mit [18F]F- entwickelt. Diese drei Verbindungen wurden zuerst separat als Einzelkomponenten und anschließend, in Liposomen formuliert, in Tierstudien an Mäusen hinsichtlich ihrer initialen Pharmakokinetik und Biodistribution untersucht. Dabei zeigte sich ein ähnliches Verhalten der neuartigen Ch-PEG-hbPG-Derivate zu den bekannten Ch-PEG, mit dem Vorteil der Multifunktionalität an den hyperverzweigten Strukturen. Die liposomalen Strukturen mit der neuartigen sterischen Stabilisierung wiesen eine erhöhte Blutzirkulationszeit und vorteilhafte Blut-zu-Leber- und Blut-zu-Lunge-Verhältnisse im Vergleich zu den linear stabilisierten Analoga auf.rnEine weitere Klasse von Wirkstofftransportsystemen sind polymere Trägersysteme wie pHPMA. Alkinfunktionalisierte Polymere konnten in zwei verschiedenen Größen (~12 und 60 kDa) mittels CuAAC in sehr hohen Ausbeuten mit der prosthetischen Gruppe 18F-TEG-N3 radiomarkiert werden. Bicyclononinderivate der gleichen Größen konnten ohne Kupferkatalyse über ringspannungsvermittelte Alkin-Azid-Cycloaddition (SPAAC) mikrowellengestützt markiert werden und stehen somit zur in vivo-Untersuchung hinsichtlich des Einflusses der Markierungsart zur Verfügung.

In traditional medicine of Cameroon, Nymphaea lotus Linn. (Nympheaceae) is used for treatment of male sexual disorders. The aim of this study was to evaluate the effects of the N. lotus flowers aqueous extracts on general mating behavior, fertility and some androgenic parameters of normoglycemic and diabetic adult male rats. Mating behavior assessment was carried out with primiparous and with oestrus female rats. Male rats were divided into 5 groups and subjected once in a day to the following treatment: distilled water (10 mL/kg), Sildenafil citrate (5mg/kg), Testosterone enanthate (5mg/kg) and plant extract (75 mg/kg and 150 mg/kg). Treatment lasted for 8 and 55 days before sacrifice. Organ weight, epididymal sperm counts, motility, viability, histomorphometric analysis, muscular strength, seminal fructose, cholesterol level, epididymal and serum proteins testicular were determined. Results showed a dose- dependant influence of the treatment in sperm count and motility which significantly increases compared to the distilled water-administered control. An increase in some sexual performance parameters (mount and intromission frequencies) was observed in both diabetic and normal rats compared to respective controls. The latencies of mount and intromission were significantly reduced. Computed frequencies of penile licking, index of libido and sexual motivation were higher in normal and diabetic extract-treated animals; which suggest an enhancement of motivational parameters. The treatment also caused significant increases in the weight of the testis, prostate, epididymis, in serum cholesterol and epididymal protein level in normal rats compared to the distilled water-administered control. These results indicate an androgenic pro-sexual potential of N. lotus in male rats and justify the empirical use of N. lotus in the management of erectile dysfunction and fertility disorders in males. Key-words: Nymphaea lotus, prosexual, androgenic, fertility, diabetic, male rat. //Selon les tradipraticiens de l’Ouest-Cameroun, Nymphaea lotus Linn. (N. lotus), de la famille des Nymphéacées, est utilisé pour le traitement des dysfonctionnements sexuels chez les hommes. Dans cette étude, l’effet de l’extrait aqueux des fleurs de N. lotus sur la fonction de reproduction des rats mâles adultes normaux et diabétiques a été évalué. Dans le but d’évaluer les effets de l’extrait aqueux des fleurs de N. lotus sur les paramètres généraux de copulation, d’androgénicité et de fertilité, les rats normoglycémiques et diabétiques ont été divisé en 3 groupes contrôle [Groupe I- recevant de l’eau distillée, Groupe II et III recevant respectivement du citrate de sildénafil (5 mg/kg) et l’énanthate de testostérone (5 mg/kg)] et deux groupes expérimentaux traités à l’extrait aux doses 75 mg/kg (Groupe IV) et 150 mg/kg (Groupe V). L’eau distillée, l’extrait et la substance de référence était administré per os une fois par jour. Pour analyser le comportement sexuel et la fertilité, des femelles primipares et en oestrus étaient utilisées. Le traitement a duré 8 et 55 jours avant le sacrifice. Le poids relatif des organes, le nombre de spermatozoïde, la motilité, les analyses morphométriques, la force musculaire, le taux de cholestérol, le taux de protéines sériques et épididymaires étaient déterminé. Le temps de latence de monte et d’intromission a diminué significativement alors que la fréquence d’éjaculation a augmenté. L’index de libido, la fréquence de monte, d’intromission, d’éjaculation, d’orientation des mâles vis-à-vis d’eux même et l’index de motivation sexuelle a augmenté chez les animaux traités l’extrait comparés à ceux ayant reçu de l’eau distillée aussi bien chez les normaux que chez le diabétiques qui n’enregistrent d’ailleurs aucune éjaculation. Le traitement a augmenté significativement (P < 0,01) le poids des testicules, de la prostate et de l’épididyme. Il est observé une augmentation dose-dépendante du nombre et de la motilité des spermatozoïdes (P < 0,05), ainsi qu’une augmentation significative (P < 0,001) temps-dépendant du taux de cholestérol sérique et de protéines épididymaires. Ces résultats indiquent un potentiel androgénique pro-érectile de N. lotus chez les rats mâles et justifient l’utilisation empirique des fleurs de N. lotus dans le traitement des dysfonctions érectiles et des problèmes de fertilité chez les hommes. A 20-year old Swiss Warmblood mare was referred to the Swiss Institute of Equine Medicine with a history of poor performance, coughing and ataxia and hindlimb weakness which progressed to recumbency. Lung auscultation revealed pronounced wheezing, blood work showed signs of chronic inflammation and increased bone turnover and thoracic ultrasound indicated patchy pulmonary consolidation. Cerebrospinal fluid revealed only mild, unspecific changes allowing exclusion of meningoencephalomyelitis and clinically relevant bleeding. Despite medical treatment and support in a sling the mare did not improve and was euthanized. Necropsy revealed a poorly demarcated, non-encapsulated and invasively growing mass dorsally in the musculature at the level of the forth cervical vertebra (C4) infiltrating the vertebral body and the spinal canal at the level of C1–C2. Multiple nodular, firm masses were present in all lobes of the lung and appeared to be mainly located in vessels. Histologically the masses were composed of spindle cells with marked anisocytosis, anisocaryosis, a high mitotic activity and showed invasive growth. These neoplastic cells stained positive for CD31, an endothelial cell marker, which confirmed diagnosis of a hemangiosarcoma. Definite ante mortem diagnosis of hemangiosarcoma, which is rare in horses, is challenging. Besides the vertebral localization, disseminated, locally invasive and cutaneous forms of hemangiosarcoma exist and can be either acquired or congenital. Prognosis for equine hemangiosarcoma and response to treatment are usually poor and progression of clinical signs is rapid. Vertebral hemangiosarcoma is an uncommon cause of spinal ataxia in horses.
