In variant Creutzfeldt-Jakob disease (vCJD), a disease linked to bovine spongiform encephalopathy (BSE), florid-type prion protein (PrP(sc)) deposits are aggregated around the larger diameter (> 10 µm) cerebral microvessels. Clustering of PrP(sc) deposits around blood vessels may result from blood-borne prions or be a consequence of the cerebral vasculature influencing the development of the florid deposits. To clarify the factors involved, the dispersion of the florid PrP(sc) deposits was studied around the larger diameter microvessels in the neocortex, hippocampus, and cerebellum of ten cases of vCJD. In the majority of brain regions, florid deposits were clustered around the larger diameter vessels with a mean cluster size of between 50 µm and 628 µm. With the exception of the molecular layer of the dentate gyrus, the density of the florid deposits declined as a negative exponential function of distance from a blood vessel profile suggesting that diffusion of molecules from blood vessels is a factor in the formation of the florid deposits. Diffusion of PrP(sc) directly into the brain via the microvasculature has been demonstrated in vCJD in a small number of cases. However, the distribution of the prion deposits in vCJD is more likely to reflect molecular 'chaperones' diffusing from vessels and promoting the aggregation of pre-existing PrP(sc) in the vicinity of the vessels to form florid deposits.

The objective of this article was to determine whether the pathological changes of Creutzfeldt-Jacob disease (CJD) were related to the brain microcirculation. Hence, the spatial correlations between the vacuolation, prion protein (PrP) deposits, and the blood vessel profiles were studied in immunolabelled sections of the cerebral cortex, hippocampus, and cerebellum in two subtypes of CJD, viz., sporadic CJD (sCJD) and variant CJD (vCJD). In sCJD, both the vacuolation and the ‘synaptic-type’ PrP deposits were spatially correlated with the microvessels; the PrP deposits being more strongly correlated than the vacuoles. In vCJD, there were no significant spatial correlations between either the vacuolation or the diffuse-type of PrP deposit and the microvessels. By contrast, a consistent pattern of spatial correlation was observed in gyri of the cerebral cortex between the florid PrP deposits and microvessels. In both sCJD and vCJD, the frequency of positive spatial correlations was similar in the different gyri of the cerebral cortex and in the upper compared with the lower laminae. In conclusion, the microcirculation may be more significantly involved in determining the pathological changes in sCJD than in vCJD. The spatial correlations of the florid PrP deposits in vCJD and the synaptic deposits in sCJD and the blood vessels may be attributable to factors associated with the microcirculation which enhance the aggregation of PrP molecules rather than representing a possible haematogenous spread of the disease.

During the second half of the nineteenth century, a German business community of about one hundred merchants and commercial clerks developed in Glasgow. Their trade networks extended not only to Germany but also to other world markets. The main arguments and findings of the microhistorical analysis include: numbers were significantly higher than previously assumed; endogenous recruitment based on ethnic and family ties was prevalent; migrants benefited from their migration-induced social capital (training, languages, intercultural competence) to fill a skills-gap in Britain; labour market competition at the junior career level was less pronounced than contemporaneous assessments suggested; naturalisation was taken out for purely pragmatic reasons; there was a sense of community at intra-ethnic level, but also with the local business elite. The case study is embedded into the larger context of Anglo-German economic relations and globalisation. A purely local perspective does not suffice to do justice to the wider significance of expatriate business communities in an age of economic globalisation.

Maintaining the structural health of prestressed concrete nuclear containments is a key element in ensuring nuclear reactors are capable of meeting their safety requirements. This paper discusses the attachment, fabrication and characterisation of optical fibre strain sensors suitable for the prestress monitoring of irradiated steel prestressing tendons. The all-metal fabrication and welding process allowed the instrumented strand to simultaneously monitor and apply stresses up to 1300 MPa (80% of steel's ultimate tensile strength). There were no adverse effects to the strand's mechanical properties or integrity. After sensor relaxation through cyclic stress treatment, strain transfer between the optical fibre sensors and the strand remained at 69%. The fibre strain sensors could also withstand the non-axial forces induced as the strand was deflected around a 4.5 m bend radius. Further development of this technology has the potential to augment current prestress monitoring practices, allowing distributed measurements of short- and long-term prestress losses in nuclear prestressed-concrete vessels. © 2014 Elsevier B.V.

The main driver for the investigation of fast pyrolysis oil marine fuel blends is EU directive 2012/33/EU which aims to cut the sulphur content of marine fuel and thereby reduce air pollution caused by marine vessels. The aim of this study was to investigate the miscibility of 3- and 4- component blends containing pyrolysis oil, 1-butanol, biodiesel (RME) and/or marine gas oil (MGO). The ideal blend would be a stable homogenous product with a minimum amount of butanol, whilst maximising the amount of pyrolysis oil. A successful blend would have properties suitable for use in marine engines. In order to successfully utilise a marine fuel blend in commercial vessels it should meet minimum specification requirements such as a flash point ≥60°C. Blends of pyrolysis oil, RME, MGO and 1-butanol were evaluated and characterised. The mixed blends were inspected after 48 hours for homogeneity and the results plotted on a tri-plot phase diagram. Homogenous samples were tested for water content, pH, acid number, viscosity and flash point as these give indicate a blend’s suitability for engine testing. The work forms part of the ReShip Project which is funded by Norwegian industry partners and the Research Council of Norway (The ENERGIX programme).

Tissue-engineered blood vessels (TEBV) can serve as vascular grafts and may also play an important role in the development of organs-on-a-chip. Most TEBV construction involves scaffolding with biomaterials such as collagen gel or electrospun fibrous mesh. Hypothesizing that a scaffold-free TEBV may be advantageous, we constructed a tubular structure (1 mm i.d.) from aligned human mesenchymal cell sheets (hMSC) as the wall and human endothelial progenitor cell (hEPC) coating as the lumen. The burst pressure of the scaffold-free TEBV was above 200 mmHg after three weeks of sequential culture in a rotating wall bioreactor and perfusion at 6.8 dynes/cm(2). The interwoven organization of the cell layers and extensive extracellular matrix (ECM) formation of the hMSC-based TEBV resembled that of native blood vessels. The TEBV exhibited flow-mediated vasodilation, vasoconstriction after exposure to 1 μM phenylephrine and released nitric oxide in a manner similar to that of porcine femoral vein. HL-60 cells attached to the TEBV lumen after TNF-α activation to suggest a functional endothelium. This study demonstrates the potential of a hEPC endothelialized hMSC-based TEBV for drug screening.

The fishing sector has been suffering a strong setback, with reduction in fishing stocks and more recently with the reduction of the fishing fleet. One of the most important factors for this decrease, is related to the continuous difficulty to find fish with quality and quantity, allowing the sector work constantly all year long. However other factors are affecting negatively the fishing sector, in particular the huge maintenance costs of the ships and the high diary costs that are necessary for daily work of each vessel. One of the main costs associated with daily work, is the fuel consumption. As an example, one boat with 30 meters working around 17 hours every day, consumes 2500 liters of fuel/day. This value is very high taking into account the productivity of the sector. Supporting this premise was developed a project with the aim of reducing fuel consumption in fishing vessels. The project calls “ShipTrack” and aims the use of forecasts of ocean currents in the routes of the ships. The objective involves the use of ocean currents in favor, and avoiding ocean currents against, taking into account the course of the ship, in order to reduce fuel consumption and increase the ship speed. The methodology used underwent the creation of specific Software, in order to optimize routes, taking into account the forecasts of the ocean currents. These forecasts are performed using numerical modelling, methodology that become more and more important in all communities, because through the modeling, it can be analyzed, verified and predicted important phenomena to all the terrestrial ecosystem. The objective was the creation of Software, however its development was not completed, so it was necessary a new approach in order to verify the influence of the ocean currents in the navigation of the fishing ship "Cruz de Malta". In this new approach, and during the various ship routes it was gathering a constant information about the instant speed, instantaneous fuel consumption, the state of the ocean currents along the course of the ship, among other factors. After 4 sea travels and many routes analyzed, it was possible to verify the influence of the ocean currents in the Ship speed and in fuel consumption. For example, in many stages of the sea travels it was possible to verify an increase in speed in zones where the ocean currents are in favor to the ships movements. This incorporation of new data inside the fishing industry, was seen positively by his players, which encourages new developments in this industry.

Tract consultants are a landscape architecture practice, founded in 1973 as an offshoot to the highly innovative, interdisciplinary design and build company Merchant Builders, and was perhaps the first truly corporate practice of this type in Australia. Founding directors Rodney Wulff and Steve Calhoun were both instrumental in establishing the undergraduate landscape architecture course at RMIT University, and bringing our Jim Sinatra, who had taught Calhoun at the University of Iowa. Wulff remained for many years the holder of the only doctorate in landscape architecture in the country. This combination of an academic, design and professional agenda was a rich one for Tract in their early days. This founding generosity and interest in the intellectual aspects of landscape architecture continues in relation to the university in a number of ways, including information ones, such as the regular employment of applicants who fail to get into the course at RMIT. In preparing them for re-applying, he has given a number of individuals a way into the profession that the university could not allow.

Purpose The aim of this work is to develop a more complete understanding of the in vivo histology of the human palpebral conjunctiva and tarsal plate. Methods. The upper eyelids of 11 healthy human volunteer subjects were everted, and laser scanning confocal microscopy was used to examine the various tissue layers of the palpebral conjunctiva and tarsal plate. Results The superficial and basal epithelial layers are composed of cells with gray cytoplasm and thick, light gray borders.Nuclei can not be seen. The stroma has a varied appearance; fibrous tissue is sometimes observed, interspersed with dark,amorphous lacunae, and crevases. Numerous single white or gray cells populate this tissue, and fine blood vessels are seen traversing the field. Occasional conjunctival microcysts and Langerhans cells are observed. The tarsal plate is dark and amorphous, and meibomian gland acini with convoluted borders are clearly observed. Acini are composed of an outer lining of large cuboidal cells, and differentiated secretory cells can be seen within the acini lumen. Conclusions Laser scanning confocal microscopy is capable of studying the human palpebral conjunctiva, tarsal plate, and acini of meibomian glands in vivo. The observations presented here may provide useful supplementary anatomical information relating to the morphology of this tissue.
