Selenium binding proteins (SeBP) represent a family of proteins that are believed to be involved in controlling the oxidation/reduction in many physiological processes. The cDNA of Zhikong Scallop Chlamys farreri selenium binding protein (zSeBP) was cloned by expressed sequence tag (EST) and RACE techniques. The high similarity of zSeBP deduced amino acid sequence with the SeBP in other organisms, such as bird, fish, frog, mosquito, fruit fly, mammalian, and even nematode and microorganism indicated that zSeBP should be a member of SeBP family. The temporal expression of zSeBP in the hemocytes was measured by semi-quantitative RT-PCR after scallops were stimulated by either oxidative stress or microbial challenge. The expression of zSeBP was up-regulated progressively after stimulation, and then dropped gradually to the original level. Meanwhile, malondialdehyde (MDA) measured by the colorimetric method in the microbial challenged scallops increased immediately after scallops was challenged by microbes, and was significantly higher than that in the control scallops. Results indicated that the microbial infection could incense the disorder of oxidation/reduction and may result in high MDA production. The negative correlation between the expression level of zSeBP and the MDA content suggested that zSeBP could play an important role in mediating the anti-oxidation mechanisms and immune response in marine invertebrates. Heat shock protein 70 (HSP70) is an important member of the heat shock protein superfamily, and it plays a key role in the process of protecting cells, facilitating the folding of nascent peptides and responding to stress. The cDNA of bay scallop Argopecten irradians HSP70 (designated AIHSP70) was cloned by the techniques of homological cloning and rapid amplification of cDNA end (RACE). The full length of AIHSP70 cDNA was 2651 bp in length, having a 5' untranslated region (UTR) of 96 bp, a 3' UTR of 575 bp, and an open reading frame (ORF) of 1980 bp encoding a polypeptide of 659 amino acids with an estimated molecular mass of 71.80 kDa and an estimated isoelectric point of 5.26. BLAST analysis revealed that the AIHSP70 gene shared high identity with other known HSP70 genes. Three classical HSP signature motifs were detected in AIHSP70 by InterPro, analysis. 3-D structural prediction of AIHSP70 showed that its N terminal ATPase activity domain and,C terminal substrate-binding domain shared high similarity with that in human heat shock protein 70. The results indicated that the AIHSP70 was a member of the heat shock protein 70 family. A semi-quantitive RT-PCR method was used to analyse the expression of AIHSP70 gene after the treatment of naphthalin which is one kind of polycyclic aromatic hydrocarbon (PAH) and the challenge of bacteria. mRNA expression of AIHSP70 in scallop was up-regulated significantly after the stimulation of naphthalin and increased with increasing naphthalin concentration. A clearly time-dependent expression pattern of AIHSP70 was observed after the scallops were infected by Vibrio anguillarum, and the mRNA expression reached a maximum level at 8 h and lasted to 16 h, and then dropped progressively. Myf-5, a member of the myogenic regulatory factors (MRF), has been shown to be expressed in muscle precursors in early stage zebrafish embryos. The MRFs, including MyoD, Myf-5, Myogenin and MR-F4, belong to the basic Helix-Loop-Helix transcription factors that contain a conserved basic Helix-Loop-Helix (bHLH) domain. To better understand the role of Myf-5 in the development of fish muscles, we have isolated the Myf-5 genomic sequence and cDNA from Flounder (Paralichthys olivaceus), and analyzed its structures and patterns of expression. Promoter analysis identified several putative transcription factor binding sites such as an E-box, NF-Y sites that might confer muscle-specific expression. Myf-5 transcripts were first detected in the paraxial mesoderm that gives rise to slow muscles. During somitogenesis, Myf-5 expression was found in developing somites. Myf-5 expression decreased gradually in somites in the anterior region, but remained strong in the newly formed somites. In the hatching stage, the expression was also detected in other muscle cells such as head muscle and fin muscle. In the growing fish, RT-PCR results showed that Myf-5 was expressed in the skeletal muscle and intestine. (c) 2006 Elsevier Inc. All rights reserved.

Translationally controlled tumor protein (TCTP) is one of the abundant and ubiquitously expressed proteins in metazoans In the present study, the first molluscan TCTP (denoted as VpTCTP) was identified from Venerupis philippinarum haemocytes by EST and RACE approaches The full-length cDNA of VpTCTP consisted of 1148 nucleotides with an open-reading frame of 555 bp encoding 184 amino acids The deduced amino acid sequence of VpTCTP shared high similarity with TCTPs from other species, indicating that VpTCTP should be a new member of TCTP family Several highly conserved motifs, including 5'terminal ologopyrimidine (5'TOP) starting sequence and rich AU and AUUT elements in 3'UTR, were also identified in VpTCTP The tissue and temporal expression of VpTCTP after Vi boo anguillarum challenge was recorded by quantitative real-time RT-PCR. VpTCTP transcript could be detected in all examined tissues with the highest expression level in haemocytes and the lowest in hepatopancreas Concerning the time-course expression in haemocytes, the relative expression of VpTCTP mRNA was down-regulated sharply from 6 h to 12 h post-infection. Then, the expression level was obviously up-regulated and reached 3.4-fold to that in the control group at 48 h post challenge As time progressed, the expression of VpTCTP recovered to the original level at 96 h. We used microarray technology to study differentially expressed genes in white spot syndrome virus (WSSV)-infected shrimp. A total of 3136 cDNA targets, including 1578 unique genes from a cephalothorax cDNA library and 1536 cDNA clones from reverse and forward suppression subtractive hybridization (SSH) libraries of Fenneropenaeus chinensis, plus 14 negative and 8 blank control clones, were spotted onto a 18 x 18 mm area of NH2-modified glass slides. Gene expression patterns in the cephalothorax of shrimp at 6 h after WSSV injection and moribund shrimp naturally infected by WSSV were analyzed. A total of 105 elements on the arrays showed a similar regulation pattern in artificially infected shrimp and naturally infected moribund shrimp; parts of the results were confirmed by semiquantitative reverse transcriptase-polymerase chain reaction (RT-PCR). The up-regulated expression of immune-related genes, including heat shock proteins (HSP70 and HSP90), trehalose-phosphate synthase (TPS), ubiquitin C, and so forth, were observed when shrimp were challenged with WSSV. Genes including myosin LC2, ATP synthase A chain, and arginine kinase were found to be down-regulated after WSSV infection. The expression of housekeeping genes such as actin, elongation factor, and tubulin is not stable, and so these genes are not suitable as internal standards for semiquantitative RT-PCR when shrimp are challenged by WSSV. As a substitute, we found that triosephosphate isomerase (TPI) was an ideal candidate of interstandards in this situation.

The ovary of triploid shrimp Fenneropenaeus chinensis was apparently impaired compared to that of the diploid shrimp at the same age. Therefore triploid shrimp ovary is possible to be taken as a model to understand the mechanism of ovary development of shrimp compared to that of the ovary of diploid shrimp at the same age. In the present study, a suppression subtractive hybridization (SSH) technique was applied to identify differentially expressed genes in the ovary between diploid and triploid shrimp. For the forward library (RNA from the ovary of triploid shrimp as the tester), 54 genes were identified. For the reverse library (RNA from the ovary of diploid shrimp as the tester), 16 genes were identified. The identified genes encoded proteins with multiple functions, including extracellular matrix components, cytoskeleton, cell growth and death, metabolism, genetic information processing, signal transduction/transport or immunity related proteins. Eleven differentially expressed genes were selected to be confirmed in the ovaries of triploid and diploid shrimp by semi-quantitative RT-PCR. Genes encoding spermatogonial stem-cell renewal factor, cytochrome c oxidase subunits I and II, clottable protein, antimicrobial peptide and transposase showed up-regulated expressions in the ovary of triploid shrimp. Genes encoding tubulin, cellular apoptosis susceptibility protein, farnesoic acid O-methyltransferase, thrombospondin and heat shock protein 90 genes showed higher expressions in the ovary of diploid shrimp. The differential expressions of the above genes are suggested to be related to the ovary development of shrimp. It will provide a new clue to uncover the molecular mechanisms underlying the ovarian development in penaeid shrimp. (C) 2010 Elsevier Inc. All rights reserved.

Ferritin, the iron storage protein, plays a key role in iron metabolism. A cDNA encoding ferritin (FcFer) was cloned from hepatopancreas of Chinese shrimp, Fenneropenaeus chinensis. The predicted protein contains 170 amino acid residues with a predicted molecular weight (MW) about 19, 422.89 Da and theoretical isoelectric point (PI) of 4.73. Amino acid alignment of FcFer revealed 97% homology with Litopenaeus vannamei ferritin. Results of the RT-PCR showed that the expression of FcFer mRNA was up-regulated after shrimp was challenged with either white spot syndrome virus (WSSV) or heavy metal ions (Zn2+ and Cu2+) in the laboratory. A fusion protein containing FcFer was produced and the purified recombinant protein exhibited similar function of iron uptake in vitro. The result of in-gel digestion and identification using LC-ESI-MS showed that two peptide fragments (-DDVALPGFAK- and -LLEDEYLEEQVDS1KK-) of the recombinant protein were identical to the corresponding sequence of L. vannamei ferritin. The recombinant FcFer protein will be proved useful for study on the structure and function of ferritin in F chinensis. (c) 2006 Elsevier B.V. All rights reserved.

Serine protease inhibitors, critical regulators of endogenous proteases, are found in all multicellular organisms and play crucial roles in host physiological and immunological effector mechanisms. The first mollusk serine proteinase inhibitor (designated AISPI) cDNA was obtained from the bay scallop Argopecten irradians by randomly sequencing a whole tissue cDNA library and rapid amplification of cDNA ends (RACE). The full-length cDNA of the scallop serine protease inhibitor was 1020 bp, consisting of a 5'-terminal untranslated region (UTR) of 39 bp, a 3'-terminal UTR of 147 bp with a canonical polyadenylation signal sequence AATAAA and a poly(A) tail, and an open reading frame of 834 bp. The AISPI cDNA encoded a polypeptide of 278 amino acids with a putative signal peptide of 22 amino acids and a mature protein of 256 amino acids. The deduced amino-acid sequence of AISPI contained six tandem and homologous domains similar to that of Kazal-type serine protease inhibitors, including the conserved sequence C-X(7)-C-X(6)-Y-X(3)-C-X(2,3)-C and six cysteine residues responsible for the formation of disulfide bridges, indicating that the AISPI protein from bay scallop should be a member of the Kazal-type serine protease inhibitor family. The temporal expression of AISPI was measured by semi-quantitative RT-PCR after injury or bacterial challenge. After the adductor muscle was wounded or injected with Vibrio anguillarum, the expression of AISPI mRNA in hemolymph was up-regulated and reached the maximum level at 8 and 16 h, respectively, and then progressively dropped back to the original level. The results indicated that AISPI could play an important role in injury healing and immune response in mollusks as it could be induced by injury and bacterial challenge. (c) 2005 Elsevier Ltd. A new member of antimicrobial protein genes of the Crustin family was cloned from haemocytes of the Chinese shrimp Fenneropenaeus chinensis by 3' and 5' RACE. The full-length cDNA of Crustin-like gene contains a 390 bp open reading frame, encoding 130 amino acids. The deduced peptide contains a putative signal peptide of 17 amino acids and mature peptide of 113 amino acids. The molecular mass of the deduced mature peptide is 12.3 ku. It is highly cationic with a theoretical isoelectric point of 8.5. The deduced amino acids sequence of this Crustin showed high homology with those of Penaeus (Litopenaeas) setferus. Northern blotting showed that the cloned Crustin gene was mainly expressed in haemocytes, gill, intestine, and RNA in situ hybridization indicated that the Crustin gene was constitutively expressed exclusively in haemocytes of these tissues. Capillary electrophoresis RT-PCR analysis showed that Crustin was up-regulated dramatically from 12 to 48 h after a brief decrease of mRNA during first 6 h in response to microbe infection. The level of Crustin mRNA began to restore at 72 h post-challenge. This indicated that Crustin gene might play an important role when shrimps are infected by bacterial pathogen.

Thymidylate synthase (TS), an essential enzyme for DNA de novo synthesis, is a critical therapeutic target in cancer therapy. Previous study has shown that TS was able to bind to its own mRNA in human and E.coli, resulting in translational repression. Zebrafish is the best animal model for vertebrate study. In order to study the regulatory mechanism of zebrafish TS, the enzyme were expressed in E. coli BL21 (DE3) and it was purified to homogeneity. Electrophoretic mobility shift assay (EMSA) was used to detect the interaction of zebrafish TS protein and its own TS transcript in vitro and the results showed that zebrafish TS could bound with its own mRNA specifically. Further study revealed that zebrafish TS was able to interact with its own mRNA in vivo using immunoprecipitation : RT-PCR technique. The results provide evidence that zebrafish may be developed as an useful model for studying the anti-metabolism agents.

SNARE蛋白家族是所有真核细胞胞吐及分泌作用中的关键因子,由其介导的运输囊泡膜与靶膜的锚靠、融合为胞内蛋白的运出提供了一条重要途径。体外试验表明,Syntaxin6-Syntaxin7-Vti1b,SNAP-23-Syntaxin4等SNARE核心蛋白之间精确的相互作用是哺乳动物巨噬细胞肿瘤坏死因子α (TNF-α)运输和分泌的必备条件,在机体非特异性免疫应答反应过程中起重要作用。 本研究受上述启示,旨在揭示SNARE蛋白在海洋鱼类免疫细胞内重要细胞因子白细胞介素1β (IL-1β)的分泌过程中的作用。参照Percoll密度梯度离心技术,从鲈鱼头肾组织分离纯化巨噬细胞进行稳定培养;利用RT-PCR方法克隆出鲈鱼t-SNARE蛋白SNAP-23和Syntaxin3的部分cDNA序列,再结合先前克隆的VAMP2和已知的鲈鱼IL-1β,TNF-α和IL-8的基因序列,设计特异性引物。利用Real-time PCR技术在mRNA水平上精确测定鲈鱼巨噬细胞中上述6种基因在革兰氏阴性菌脂多糖(LPS)分子刺激下的表达变化,发现SNAP-23基因与三种细胞因子基因的表达正相关;通过免疫印迹检测SNAP-23蛋白表达变化,利用酶联免疫吸附试验(ELISA)检测IL-1β的分泌水平,在蛋白水平上验证了SNAP-23表达与IL-1β分泌的正相关性;利用5`RACE和3`RACE技术克隆出鲈鱼SNAP-23全长基因,结合定点突变策略和靶向PCR克隆手段,构建鲈鱼SNAP-23野生型融合质粒pEGFP-SNAP-23wt,Cys缺失突变融合质粒pEGFP-SNAP-23ΔCys和模拟E型肉毒神经毒素(BoNT/E)切割突变融合质粒pEGFP-SNAP-23ΔBoNT/E,以及鲈鱼IL-1β野生型融合表达质粒IL-1β-pEGFP和IL-1β-pEYFP。所有融合蛋白均在鲈鱼巨噬细胞内成功表达,结合ELISA实验结果发现,SNAP-23野生型的表达对IL-1β的分泌有促进作用,而Cys缺失突变体的表达则抑制IL-1β向胞外分泌。首次证实了鱼类巨噬细胞内SNAP-23蛋白在IL-1β分泌过程中的重要作用。此外通过与GFP共表达,定位了IL-1β分子在巨噬细胞内的分布,发现新合成的IL-1β分子很可能像TNFα一样经“内质网-胞质-伪足-胞外” 的分泌路径运出胞外。

本文分别提取了中国对虾、中华绒螯蟹、鹰爪虾和蓝对虾的眼柄总RNA。用无DNA污染的眼柄总RNA为模板,根据日本对虾的MIH设计兼并引物P1和P2,进行RT-PCR扩增,在适宜的反应条件下,均分别得到一特异性的产物。以蓝对虾基因组DNA为模板的PCR扩增也能得到一与特异性cDNA片段大小相似的特异性的DNA片段。分别将这些片段亚克隆到载体中进行测序,测得中国对虾特异性cDNA片段由203个碱基组成,中华绒螯蟹、鹰爪虾、蓝对虾的特异性cDNA片段及蓝对虾的特异性DNA片段由215个碱基组成,其中蓝对虾特异性cDNA与由蓝对虾的基因组DNA扩增得到的特异性DNA片段的碱基序列几乎完全相同。利用互联网上的在线工具Fasta3查找这些cDNA推定的氨基酸序列的相似序列,可以发现大量的甲壳动物的CHH家族的神经肽与它们相似。在所有的相似序列中,分别由中国对虾、中华绒螯蟹、鹰爪虾和蓝对虾的特异性cDNA片段推定的氨基酸序列与蜕皮抑制激素(MIH)的相似度最高,这一结果提示分别由中国对虾、中华绒螯蟹、鹰爪虾和蓝对虾的眼柄特异性cDNA片段推定的氨基酸序列可能是这四种甲壳动物的MIH片段。根据核苷酸数据和氨基酸数据,比较这四种甲壳动物的MIH以及已发表的六种MIH之间的相似度,同时分析这十种MIH的系统进化关系,结果是蓝对虾、鹰爪虾和中华绒螯蟹的MIH相似性较高,亲缘关系较近,这与物种的系统演化不相符。

对虾病害在世界范围内的广泛传播,给水产养殖和沿海农村经济造成了重大损失。深入开展对虾免疫机制研究并在此基础上寻找对虾疾病防治的有效方法已成为当务之急。研究表明,当对虾等甲壳动物受到外界病原刺激时,其体内的吞噬细胞在吞噬活动中会激活磷酸己糖支路的代谢,引起呼吸爆发,产生多种活性氧分子。另外,受到病原侵染的对虾还会产生其他多种免疫反应,这些免疫反应将消耗大量的能量(ATP),产能的呼吸链会加速运转,由此也会引发大量活性氧的产生。这些活性氧分子可以杀灭入侵的病原微生物,但同时由于活性氧分子反应的非特异性,它们也会对宿主的细胞、组织和器官造成严重伤害,进而导致对虾生理机能的损伤和免疫系统的破坏。所以,消除对虾体内因过度免疫反应产生的过量氧自由基将能够增强其抵御病原侵染的能力,提高免疫力。本论文从中国明对虾体内克隆了线粒体型超氧化物歧化酶(mMnSOD)、胞质型超氧化物歧化酶(cMnSOD)、过氧化氢酶(Catalase)和过氧化物还原酶(Peroxiredoxin)等四种与免疫系统相关的抗氧化酶基因,分析了它们的分子结构特征,组织分布及应答不同病原刺激的表达变化模式,并对其中的mMnSOD基因和Peroxiredoxin基因进行了体外重组表达、分离纯化和酶活性分析。 采用RACE技术从中国明对虾血细胞中克隆了两个超氧化物歧化酶(SOD)基因,通过序列比对分析发现,其中一个为mMnSOD基因,另一个为cMnSOD基因。mMnSOD基因的cDNA全长为1185个碱基,其中开放阅读框为660个碱基,编码220个氨基酸,其中推测的信号肽为20个氨基酸。多序列比对结果显示中国明对虾mMnSOD基因的推导氨基酸序列与罗氏沼虾、蓝蟹的推导氨基酸序列同源性分别为88%和82%。Northern blot结果表明,该基因在对虾的肝胰脏、血细胞、淋巴器官、肠、卵巢、肌肉和鳃等组织中均有表达。半定量RT-PCR结果显示,对虾感染病毒3 h时,该基因在血细胞和肝胰脏中的转录水平显著升高。此外,通过构建原核表达载体,本研究对该基因进行了体外重组表达,并对纯化的重组蛋白进行了质谱鉴定和酶活分析。cMnSOD基因的cDNA全长为1284个碱基,其中开放阅读框为861个碱基,编码287个氨基酸。多序列比对结果显示中国明对虾cMnSOD基因的推导氨基酸序列与斑节对虾和凡纳滨对虾的同源性高达98%和94%。组织半定量结果显示,cMnSOD基因在对虾被检测的各个组织中均有表达。 另外,半定量RT-PCR结果表明,对虾感染病毒23h时,该基因在肝胰脏中的转录上升到正常水平的3.5倍;而感染后59 h时,该基因在血细胞中的转录上升到正常水平的2.5倍。 利用根据其他生物过氧化氢酶保守氨基酸序列设计的简并引物,结合RACE技术,从中国明对虾肝胰脏中克隆到了过氧化氢酶基因的部分片段,片段长1725个碱基。多序列比对结果发现目前所得中国明对虾Catalase基因部分片段的推导氨基酸序列与罗氏沼虾和皱纹盘鲍Catalase氨基酸序列的同源性分别达到95%和73%。通过实时荧光定量PCR技术对中国明对虾Catalase基因在各个组织中的分布情况及病毒感染后该基因在血细胞和肝胰脏中的转录变化进行了研究。结果发现,该基因在肝胰脏、鳃、肠和血细胞中表达水平较高,在卵巢、淋巴器官和肌肉中的表达水平相对较弱;感染病毒23 h和37 h时,对虾血细胞和肝胰脏中该基因mRNA的表达量分别出现显著性上升。 依据中国明对虾头胸部cDNA文库提供的部分片段信息,结合SMART-RACE技术,从中国明对虾肝胰脏中克隆到了过氧化物还原酶基因(Peroxiredoxin), 该基因的cDNA全长为942个碱基,其中开放阅读框为594个碱基,编码198个氨基酸。中国明对虾Peroxiredoxin基因的推断氨基酸序列与伊蚊、文昌鱼和果蝇等Peroxiredoxin基因的推断氨基酸序列同源性分别为77%、76%和73%。其蛋白理论分子量为22041.17 Da,pI为5.17。Northern blot结果表明,Peroxiredoxin基因在对虾的肝胰脏、血细胞、淋巴器官、肠、卵巢、肌肉和鳃等组织中均有表达。实时荧光定量PCR结果显示,弧菌感染后,该基因在对虾血细胞和肝胰脏中的转录水平都有明显变化并且表达模式不同。另外,对该基因进行了体外重组表达,并对纯化的重组蛋白进行了质谱鉴定和酶活性分析。酶活性分析表明,复性后的重组蛋白能在DTT存在的条件下还原H2O2。

热休克蛋白70是热休克蛋白家族中最重要的一类家族成员,它具有“分子伴侣”、细胞保护、抗凋亡以及肿瘤免疫治疗等独特复杂的生物学功能,它的研究已成为生命科学领域研究的热点之一。采用RACE方法从皱纹盘鲍中克隆到一种HSP70基因的全长cDNA序列,该序列与其它物种的HSP70基因高度同源。克隆得到的皱纹盘鲍HSP70基因的全长cDNA序列为2631bp,开放阅读框为1968bp,另外其5'-和3'-非翻译区长度分别是90和573bp,其中3'-非翻译区具有AATAAA mRNA加尾信号及PolyA寡聚腺苷酸。开放阅读框编码655个氨基酸,包含N端包含的382个氨基酸的ATP酶结合域(约45kDa)、由161个氨基酸编码的底物肽结合结构域(18kDa)及112个氨基酸编码的C端结构域(10kDa)。构建了含有皱纹盘鲍HSP70的表达载体,以期得到体外表达的重组热激蛋白70,为制备抗体,通过Western blot杂交或酶联免疫等方法检测HSPs蛋白表达情况作准备。 为了研究皱纹盘鲍HSP70的转录表达,采用半定量RT-PCR方法,对HSP70不同组织及热休克、鳗弧菌感染下的转录表达情况分析。试验结果表明,采用热应激及鳗弧菌处理皱纹盘鲍,均能导致皱纹盘鲍肌肉中HSP70 mRNA转录水平明显增加,并在处理后96 h恢复至正常水平;与肌肉组织不同的是,鳃组织在温度及鳗弧菌处理后12 h后达到最高值,并一直维持相对较高的表达水平。在温度和鳗弧菌处理下,皱纹盘鲍HSP70上调表达表明HSP70的响应没有胁迫特异性,可能参与了皱纹盘鲍的免疫反应。 以皱纹盘鲍近交和杂交群体为材料,研究了不同群体的生长速率和HSP70的表达量的差异。试验结果表明,在较低的处理温度下,皱纹盘鲍近交群体HSP70的表达量高于杂交群体,随着处理温度的升高,在接近致死温度时,杂交群体的HSP70表达量要高于近交群体;不同群体达到最大HSP70表达量时的温度(Tpeak)也不同,近交群体的Tpeak为26C,低于杂交群体(28C);近交群体的最大HSP70表达量高于杂交群体。以上试验结果表明杂交群体的HSP70的表达变化幅度较近交群体的变化幅度小,具有更宽的温度适应性。可能提供了关于杂交能够缓解皱纹盘鲍内在遗传压力的证据。 用荧光定量PCR方法分析不同壳色皱纹盘鲍在热激胁迫下不同恢复时间和南方与北方越冬的皱纹盘鲍在不同程度温度胁迫下的HSP70变化。在南方与北方越冬的皱纹盘鲍由于其越冬环境的温度明显不同,表现出明显的热响应差异。具体表现如下几个方面:首先,两个群体在非胁迫的正常条件下,其HSP70的初始表达量明显不同。南方越冬群体在其正常生长温度下(16C)的表达量高于北方群体的相应的表达量(12C);其次,随着热激温度的增高,两个群体HSP70的表达量升高的幅度也不同。第三,两个群体HSP70表达量达到最高值后,随着温度的升高,表达量开始下降,但两个群体的表达量下降幅度也有所不同。结果表明在南方的适用后,体内可能聚集了较多的热休克蛋白,可能更有利于渡过北方的高温季节。

类胡萝卜素在生物体内具有重要的生理功能,其中虾青素的抗氧化活性、提高动物的免疫能力,预防癌症等生理功能更为显著。类胡萝卜素的代谢工程在大肠杆菌、酵母和高等植物中已取得了较大的进展。本文对真核微藻类胡萝卜素代谢工程进行了初步的探索。 1.克隆雨生红球藻β-胡萝卜素羟化酶基因crtR-B,基因枪法转入衣藻叶绿体,经强光处理转化子,HPLC分析表明细胞的叶黄素库(V+A+Z)量较野生型增加,表明是外源的β-胡萝卜素羟化酶将β-胡萝卜素生成玉米黄素。 2.克隆雨生红球藻β-胡萝卜素酮化酶基因bkt,基因枪法转入衣藻叶绿体,RT-PCR及RT-PCR Southern blot分析表明外源基因具有转录活性,但未检测到转化子中积累虾青素。 3.根据盐生杜氏藻大量积累β-胡萝卜素的特点,对其遗传转化体系进行了研究。发现20 µg/ml草丁膦Basta,能够完全抑制1.0×106个/ml细胞生长;通过GUS报告基因在细胞内的瞬时表达,确立了轰击压力450 psi、距离6 cm为基因枪法转化盐生杜氏藻的最佳条件;将bar基因转入细胞,得到稳定的转化子,初步建立了盐生杜氏藻稳定遗传转化体系。克隆了盐生杜氏藻八氢番茄红素合成酶基因psy的cDNA序列、基因组序列及psy基因上游459 bp片段。 本文首次开展了衣藻类胡萝卜素代谢工程研究,发现在强光处理下,衣藻crtR-B转化子外源的羟化酶作用使叶黄素库量增加,实现了对衣藻类胡萝卜素代谢途径的修饰。确立了bar基因为选择标记、Basta为筛选试剂的基因枪转化盐生杜氏藻的遗传转化体系,并克隆了其内源的psy基因5’上游序列,为通过基因工程手段在盐生杜氏藻细胞内积累虾青素奠定了基础。
