In recent decades, metabolic syndrome has become a public health problem throughout the world. Longitudinal studies in humans have several limitations due to the invasive nature of certain analyses and the size and randomness of the study populations. Thus, animal models that are able to mimic human physiological responses could aid in investigating metabolic disease. Thus, the present study was designed to analyze metabolic syndrome markers in albino Wistar rats (Rattus norvegicus) of different ages. The following parameters were assessed at two (young), four (adult), six (adult), and twelve (mature) months of age: glucose tolerance (glucose tolerance test); insulin sensitivity (insulin tolerance test); fasting serum glucose, triglycerides, total cholesterol, HDL cholestero, and LDL cholesterol concentrations; glucose uptake in isolated soleus muscle; and total lipid concentration in subcutaneous, mesenteric, and retroperitoneal adipose tissue. We found that aging triggered signs of metabolic syndrome in Wistar rats. For example, mature rats showed a significant increase in body weight that was associated. In addition, mature rats showed an increase in the serum concentration of triglycerides, total cholesterol, and LDL cholesterol, which is characteristic of dyslipidemia. There was also an increase in serum glucose compared with the younger groups of animals. Therefore, aging Wistar rats appear to be an interesting model to study the changes related to metabolic syndrome.

OBJECTIVE: To assess biochemical, anthropometric, and dietary variables considered risk factors for coronary artery disease. METHODS: Using anthropometrics, dietary allowance, and blood biochemistry, we assessed 84 patients [54 males (mean age of 55± 8 years) and 30 females (mean age of 57±7 years)], who had severe ( > or =70% coronary artery obstruction) and nonsevere forms of coronary artery disease determined by cardiac catheterization. The severe form of the disease prevailed in 70% of the males and 64% of the females, and a high frequency of familial antecedents (92% ' 88%) and history of acute myocardial infarction (80% ' 70%) were observed. Smoking predominated among males (65%) and diabetes mellitus among females (43%). RESULTS: Males and females had body mass index and body fat above the normal values. Females with nonsevere lesions had HDL > 35 mg/dL, and this constituted a discriminating intergroup indicator. Regardless of the severity of the disease, hyperglycemia and hypertriglyceridemia were found among females, and cholesterolemia > 200 mg/dL in both sexes, but only males had LDL fraction > 160 mg/dL and homocysteine > 11.7 mmol/L. The male dietary allowance was inadequate in nutrients for homocysteine metabolism and in nutrients with an antioxidant action, such as the vitamins B6, C, and folate. Individuals of both sexes had a higher lipid and cholesterol intake and an inadequate consumption of fiber. The diet was classified as high-protein, high-fat, and low-carbohydrate. CONCLUSION: The alterations found had no association with the severity of lesions, indicating the need for more effective nutritional intervention.

INTRODUÇÃO: A prática de atividade física é reconhecida como fator importante para a preservação, recuperação e manutenção da saúde. O estímulo à prática de exercícios é crescente, mas quando relacionado à gravidez, dúvidas surgem sobre os efeitos deletérios ou salutares na mãe e no feto. OBJETIVO: O objetivo deste trabalho foi avaliar os efeitos do exercício físico intervalado e contínuo no perfil bioquímico de ratas Wistar prenhes e avaliar o efeito destes exercícios no peso da placenta e dos filhotes. MÉTODOS: Utilizou-se 45 ratas Wistar divididas em grupos de 15 animais segundo o tipo de exercício: controle (GC), exercício contínuo (GCO) e exercício intermitente (GIN). Os exercícios constituíram-se de natação forçada, cinco dias por semana, em piscinas individuais: exercício contínuo (duração de 45 minutos diários com sobrecarga de 5% do peso corporal) e intermitente (45 minutos com estímulos de 15 segundos de exercício e 15 de repouso com sobrecarga de 15% do peso corporal). O exercício foi praticado do primeiro ao 20º dia de prenhez. Após este período avaliou-se o peso e os níveis de glicemia, colesterol total, LDL-C, HDL-C e triglicérides das ratas, assim como o peso da placenta e dos filhotes. RESULTADOS: Não se observou modificação no peso das mães. Houve redução significativa nos níveis de LDL-C. O peso das placentas não variou, mas os pesos dos filhotes variaram estatisticamente entre os três grupos (4,153 ± 0,649; 3,682 ± 0,070 e 3,453 ± 0,052, respectivamente, para os filhos de mães do GC, GIN e GCO). CONCLUSÕES: Conclui-se que a prática do exercício físico contínuo e intermitente por ratas prenhes, neste modelo experimental, não interferiu no peso corpóreo das mesmas, mas interferiu no peso dos filhotes ao nascer

Os objetivos deste estudo foram comparar medidas de circunferências da cintura e abdominal e analisar suas relações com fatores de risco cardiometabólico em servidores de uma universidade da Bahia. Para tanto, cinquenta e cinco homens e setenta e uma mulheres (36,4 ± 11,2 anos) foram submetidos à avaliação antropométrica bem como medidas das pressões arteriais sistólica e diastólica, glicemia, triglicerídeos, colesterol total e frações HDL e LDL. Apesar das fortes correlações (P < 0.01) entre as diferentes medidas (r > 0,93), a circunferência da cintura foi significativamente menor que a circunferência abdominal em ambos os sexos, sendo a diferença média entre locais maior em mulheres (8,6 ± 4,1 vs 3,8 ± 4,2 cm; P < 0,01). A circunferência da cintura foi significativamente relacionada a dois e quatro fatores de risco em homens e mulheres, respectivamente. Por outro lado, a circunferência abdominal foi significativamente relacionada a um fator em homens e cinco em mulheres. Não foram observadas diferenças significativas (P > 0,05) entre coeficientes de correlação nos casos em que ambas as circunferências se relacionaram significativamente a um fator de risco. Esses resultados sugerem que o local de mensuração tem influência substancial sobre a circunferência tomada na região inferior do tronco, particularmente, em mulheres, porém, não evidenciam clara superioridade de uma das medidas quanto às relações com fatores de risco cardiometabólico tradicionais em amostra brasileira. Estudos devem ser conduzidos, buscando comparar a capacidade preditiva de diferentes medidas de circunferência para o desenvolvimento de fatores de risco e doenças cardiovasculares em diferentes populações.

OBJETIVO: avaliar a prevalência do hipotiroidismo subclínico e suas repercussões sobre o perfil lipídico e a densidade mineral óssea (DMO) em mulheres na pós-menopausa. Métodos: trata-se de estudo transversal com recuperação de dados de prontuários de pacientes acompanhadas em ambulatório de climatério. Critérios de inclusão: mulheres na pós-menopausa com dosagem do hormônio estimulador da tiróide (TSH) e de tiroxina livre (T4-L). Critérios de exclusão: hipertiroidismo e carcinoma de tiróide. Considerou-se hipotiroidismo subclínico valores de TSH superiores a 5,0 mUI/mL e T4-L normal. Foram selecionadas 320 pacientes (idade 55,2±6,4 anos) divididas em 3 grupos: função tiroideana normal (n=208), hipotiroidismo subclínico (n=53) e hipotiroidismo clínico sob tratamento (n=59). Foram analisados dados clínicos, uso de terapia hormonal, índice de massa corpórea (IMC=kg/m²), perfil lipídico (colesterol total, HDL, LDL, triglicerídeos) e DMO da coluna lombar e fêmur. Na análise estatística, as diferenças entre as médias dos grupos foram comparadas utilizando-se a análise de variância (ANOVA). Para múltipla comparação, assumindo que a variância era diferente entre os grupos, utilizou-se o método de Tukey. RESULTADOS: o hipotiroidismo subclínico foi diagnosticado em 16,1% dos casos. Os grupos foram homogêneos quanto às características clínicas, IMC e perfil lipídico e uso de terapêutica hormonal. Nas pacientes com hipotiroidismo subclínico ou clínico encontrou-se menor freqüência de osteopenia na coluna lombar e fêmur quando comparadas às eutiroidianas (p<0,001). Houve correlação negativa entre os valores de TSH e DMO da coluna lombar e fêmur (p<0,001). Não se constatou correlação entre os valores de TSH e idade, tempo de menopausa, IMC e perfil lipídico. O total de usuárias de terapia hormonal foi de 65,1%, duração média de 3,43±2,42 anos, não diferindo entre os grupos. CONCLUSÃO: o hipotiroidismo subclínico com prevalência de 16,1% na pós-menopausa associou-se à baixa DMO, mas sem repercussões sobre o perfil lipídico.

OBJETIVO: O excesso de peso é um grande problema de saúde pública em todo o mundo, atingindo crianças e adolescentes. O presente estudo teve por objetivo verificar se a condição de portador de sobrepeso se associa à presença de fatores de risco à saúde. MÉTODOS: Foram estudados 84 sujeitos entre seis e 17 anos, constituindo-se um grupo de 34 pacientes portadores de sobrepeso e outro de 50 obesos, atendidos em ambulatório de Nutrologia Pediátrica da Universidade de Ribeirão Preto (Unaerp), durante o período de outubro de 2004 a outubro de 2005. Foram obtidas as variáveis: pressão arterial, circunferência abdominal, glicemia de jejum, glicemia de segunda hora, colesterol total, HDL-c, LDL-c e triglicérides. Comparam-se os dois grupos por meio de duas abordagens estatísticas: comparação entre as distribuições dos valores absolutos (teste de Mann-Whitney) e entre as frequências de valores considerados alterados (teste exato de Fisher). RESULTADOS: Quanto à primeira abordagem, verificou-se diferença estatística apenas para o HDL-c, com maiores valores para o subgrupo de portadores de sobrepeso de seis a dez anos. Quanto à segunda comparação, verificou-se diferença estatística para a circunferência abdominal (segundo os critérios de Freedman), HDL-c e glicemia de jejum, com maiores frequências de valores alterados entre os obesos. CONCLUSÕES: Os portadores de sobrepeso apresentaram alterações indicativas de prejuízos à sua saúde, semelhantemente aos obesos, sendo necessário também incluí-los nos programas de prevenção e tratamento.

The mechanism by which adiposity can raise blood pressure is not fully understood. Leptin has been suggested as a possible cause of the arterial hypertension in obese subjects because leptin induces an increase in sympathetic activity. The aim of the present study is to evaluate serum leptin level, blood pressure, lipid profile, blood glucose, and insulin in obese women. Leptin, total cholesterol, low-density lipoprotein (LDL) cholesterol, and triglycerides were measured. Serum leptin was markedly higher in hypertensive obese women (92.3 +/- 22 ng/mL, n = 7) as compared with normotensive obese women (37.7 +/- 11 ng/mL, n = 7). Similarly, total cholesterol and LDL cholesterol were significantly elevated in the hypertensive group. No changes were observed in triglycerides and high-density lipoprotein cholesterol concentration between the 2 groups. There were no significant differences in plasma insulin concentration or blood glucose in both groups. In conclusion, our findings suggest a link among dyslipidemia, leptin level, and hypertension that might be relevant to the development of cardiovascular disease in obese subjects.

The aim of this study was to investigate the effects of training programs on serum lipid profile and myocardial oxidative stress. Male Wistar rats (2 mo-old) were divided into three groups (n=8): sedentary (S), loadless trained (T) and trained-overload 2% body weight (TL). T and TL were trained through swimming for 9 weeks. T and TL rats had increased myocardial lipoperoxide (TBA) and lipid hydroperoxide (HP), whereas HP was higher in TL than in T animals. Superoxide dismutase (SOD) activities were lowest in TL. Myocardial glutathione peroxidase (GSH-Px) was lower in TL than in T and S rats. TL decreased HDL-cholesterol and increased LDL-cholesterol. The serum lactate dehydrogenase and TBA were increased, while SOD and GSH-Px activities were decreased in TL rats. Loadless training was able to improve HDL-cholesterol and to reduce LDL-cholesterol. In conclusion, the loadless training program induced beneficial effects on lipid profile, while overload training induced dyslipidemic profile that was associated with serum oxidative stress. The overload training program was deleterious relative to loadless training program, increasing myocardial oxidative stress.

The present study was undertaken to determine anthropometrical parameters in male adult Wistar rats. We tested the hypothesis that the anthropometrical index may identify obesity and may predict its adverse effects on lipid profile and oxidative stress in rats. Two experimental protocols were performed. in the first experiment, 50 male Wistar rats, 21 days old and fed a control chow were studied up to 150 days of age. in the second experiment, male Wistar rats, 60 days old, were divided into three groups (n = 8): control (C) given free access to a control chow; (S) receiving the control chow and drinking 30% sucrose ad libitum and (HQ fed a high-carbohydrate diet ad libitum. The first experiment showed that food consumption, energy intake and body weight increased with increasing age, while specific rate of body mass gain was significantly decreased. There were no significant differences in body length and thoracic circumference of rats from 60 days of age. The abdominal circumference (AC) and body mass index (BMI) significantly increased with enhancing age in rats up to 90 days of age and remained constant thereafter. In the second experiment, after 30 days of dietary treatment, the final body weight, body mass gain, carcass fat and BMI were higher in S and HC rats than in C. There were no significant alterations in body length and carcass protein among the groups. Triacylglycerol (TG), total cholesterol (CT), low-density lipoprotein cholesterol (LDL-C) and lipid hydroperoxide (LH) were higher in S and HC rats than in C. High-density lipoprotein cholesterol (HDL-C) decreased in HC rats and total antioxidant substances (TAS) decreased in S and HC rats. There were positive correlations between BMI with carcass fat, BMI with LH and BMI and serum TG concentration. In conclusion, the BMI for male adult Wistar rats ranged between 0.45 and 0.68 g/cm(2). Obesity may be easily estimated from the BMI in rats. Alterations in BMI were associated with dyslipidemic profile and oxidative stress in serum of rats and BMI may predict these adverse consequences of the obesity in rats.

Objective: To assess the effect of soy protein and progressive resistance training on body composition and lipids in postmenopausal women.Design: In a controlled trial, 46 postmenopausal women were randomized to one of four groups: 25 g of soy protein (SP, n = 10), 25 g of soy protein plus resistance exercise (SPE, n = 14), 25 g of maltodextrine (placebo) (PL, n = 11), or placebo plus resistance exercise (PLE, n = 11). Progressive resistance training was held three times a week for 16 weeks and included 8 exercises (3 series of 8-12 repetitions). At baseline and after 16 weeks, body mass index, waist circumference (WC), body fat, muscle mass and serum lipid levels were measured. To confirm isoflavone absorption, urinary concentrations were determined. The t-test of Student and ANOVA were used in the statistical analysis.Results: Subjects were classified as overweight and showed android fat distribution: Urinary isoflavone excretion indicated compliance to soy protein treatment. After 16 weeks of intervention, both SPE and PLE groups showed a significant increase of 1.3 kg in muscle mass and reduction in WC of -1.4 and -2.1 cm, respectively (p < 0.05). Significant decreases in the mean values of total cholesterol and LDL (-29.0 and -24.0 mg/dL, p < 0.001 and p < 0.006, respectively) were observed in the users of soy protein alone (SP).Conclusions: Soy protein supplementation did not influence the indicators of body composition. However, it exerted possible favorable effects on lipid profile in postmenopausal women. The increase in muscle mass and reduction in abdominal fat were correlated with resistance training. Objective: To evaluate the effects of isoflavones on vasomotor symptoms and blood lipids in postmenopausal women with contraindication for conventional hormone replacement therapy (HRT). Methods: This prospective, double-blind and placebo-controlled study included 50 postmenopausal women randomly divided into two groups: 25 women on soy germ isoflavones (60 mg per day, capsules) and 25 women on placebo. Inclusion criteria included: non-vegetarian, non-asian women whose last menstruation dated at least 12 months prior to the beginning of the study, With FSH > 40 mIU/ml, hot flushes and contraindication for HRT, not using tamoxifen or antibiotic and no disease of the gastrointestinal tract. For 6 months, the Kupperman menopausal index (KMI), the vaginal cytological maturation value (MV) and both hormonal and lipid profiles were assessed. The t-test and analysis of variance (ANOVA) were employed to compare the two groups. Results: In both groups, a decreased KI rate was observed. However, isoflavone was significantly superior to placebo in reducing hot flushes (44% versus 10%, respectively) (P < 0.05). After 6 months, the isoflavone group showed increased estradiol levels with unchanged FSH, LH, and vaginal cytology, and a reduction of 11.8% in LDL and an increase of 27.3% in HDL (P < 0.05). In the placebo group, just a reduction in MV was observed after 6 months (P < 0.05). Conclusions: Soy germ isoflavone exerted favorable effects on vasomotor symptoms and lipid profile, showing itself to be an interesting alternative therapy for the postmenopausal women with contraindication for conventional HRT. In previous studies, it was shown that lipid microemulsions resembling LDL (LDE) but not containing protein, acquire apolipoprotein E when injected into the bloodstream and bind to LDL receptors (LDLR) using this protein as ligand. Aiming to evaluate the effects of apolipoprotein (apo) B-100 on the catabolism of these microemulsions, LDE with incorporated apo B-100 (LDE-apoB) and native LDL, all labeled with radioactive lipids were studied after intraarterial injection into Wistar rats. Plasma decay curves of the labels were determined in samples collected over 10 h and tissue uptake was assayed from organs excised from the animals sacrificed 24 h after injection. LDE-apo B had a fractional clearance rate (FCR) similar to native LDL (0.40 and 0.33, respectively) but both had FCR pronouncedly smaller than LDE (0.56, P<0.01). Liver was the main uptake site for LDE, LDE-apoB, and native LDL, but LDE-apoB and native LDL had lower hepatic uptake rates than LDE. Pre-treatment of the rats with 17 alpha-ethinylestradiol, known to upregulate LDLR, accelerated the removal from plasma of both LDE and LDE-apoB, but the effect was greater upon LDE than LDE-apoB. These differences in metabolic behavior documented in vivo can be interpreted by the lower affinity of LDLR for apo B-100 than for apo E, demonstrated in in vitro studies. Therefore, our study shows in vivo that, in comparison with apo E, apo B is a less efficient ligand to remove lipid particles such as microemulsions or lipoproteins from the intravascular compartment.
