Nas últimas décadas, as empresas têm sido sujeitas a uma maior concorrência, ao desenvolvimento tecnológico mais acelerado e a novos desafios no que respeita à qualidade. A Metrologia deverá assim acompanhar estas tendências do mercado. O Departamento de Metrologia (DMET) do Instituto Português da Qualidade, I.P. (IPQ) tem vindo a desenvolver esforços significativos que visam a melhoria contínua das técnicas e dos processos de medição e de calibração realizados nos laboratórios. A presente dissertação de mestrado foi realizada no Laboratório de Volume e Caudal (LVC) do DMET. Os objetivos iniciais do trabalho centraram-se na caraterização metrológica de contadores de fluidos, tendo como base a medição do caudal, utilizando como método de referência o método gravimétrico. De forma a validar o sistema de medição foram realizados ensaios entre 1 mL/h a 100 mL/h, em condições de reprodutibilidade, com dois sistemas padrão distintos de acordo com o caudal a ensaiar. Em todos os ensaios efetuados foi necessário monitorizar as condições ambientais, i.e. temperatura, pressão atmosférica, humidade relativa e a temperatura do líquido padrão, para a aplicação das correções necessárias. Outro feito importante foi a caraterização das fontes de incerteza, que permitiram caraterizar o resultado das medições. A metodologia utilizada para a avaliação e para a estimativa da incerteza de medição encontra-se descrita no Guide to the Expression of Uncertainty in Measurement (GUM). Neste projeto foram contempladas fontes de incerteza como as associadas à evaporação, à impulsão do tubo, à resolução da balança, entre outras. Com os resultados obtidos foi possível elaborar um procedimento técnico para a Calibração de Caudalímetros de Líquidos. Os resultados obtidos permitiram garantir a rastreabilidade das medições de caudal de líquidos ao SI, essencial para a calibração de equipamentos no Laboratório Nacional de Metrologia (LNM) do IPQ, nomeadamente para micro caudalímetros no intervalo de medição entre 0,12 mL/h e 600 mL/h.

Até ao final dos anos 40, o DNA não era reconhecido como portador da informação genética. Era uma molécula demasiado simples, difícil de isolar e incompatível com os métodos de análise da química orgânica e da biologia. Quando alguns cientistas começam a acreditar na importância do DNA, percebem que são incapazes, tecnicamente, de determinar a sua estrutura. É nesse espírito que James Watson vai para a Europa e, na primavera de 1951, ao assistir à conferência de Maurice Wilkins, da King’s College, onde vê uma fotografia do padrão de difração de raios X, percebe que será esta a técnica chave para a determinação da estrutura do DNA e, subsequentemente, dos segredos da vida. É este o início de uma das possíveis narrativas sobre uma das principais descobertas científicas do séc. XX que muitas vezes se reduz a: “A dupla hélice do DNA foi descoberta em 1953 por Watson e Crick”. Esta dissertação propõe-se a demonstrar que, apesar de em termos estritos, se tratar de uma afirmação verdadeira, não é suficiente para garantir uma experiência pedagógica significativa, nem fazer jus ao que é o funcionamento da ciência, com todas as implicações humanas, contextuais, éticas, consequências e impacto.

Taking into account the fact that the sun’s radiation is estimated to be enough to cover 10.000 times the world’s total energy needs (BRAKMANN & ARINGHOFF, 2003), it is difficult to understand how solar photovoltaic systems (PV) are still such a small part of the energy source matrix across the globe. Though there is an ongoing debate as to whether energy consumption leads to economic growth or whether it is the other way around, the two variables appear correlated and it is clear that ensuring the availability of energy to match a country’s growth targets is one of the prime concerns for any government. The topic of centralized vs distributed electricity generation is also approached, especially in what regards the latter fit to developing countries needs, namely the lack of investment capabilities and infrastructure, scattered population, and other factors. Finally, Brazil’s case is reviewed, showing that the current cost of electricity from the grid versus the cost from PV solutions still places an investment of this nature with 9 to 16 years to reach breakeven (from a 25 year panel lifespan), which is too high compared to the required 4 years for most Brazilians. Still, recently passed legislation opened the door, even if unknowingly, to the development of co-owned solar farms, which could reduce the implementation costs by as much as 20% and hence reduce the number of years to breakeven by 3 years.

This paper presents the development of the power electronics needed for the interaction between the electrical generator of a wind turbine and an isolated ac micro grid. In this system there are basically two types of receptors for the energy produced by the wind turbine, which are the loads connected to the isolated micro grid and the batteries used to store energy. There are basically two states in which the system will work. One of the states is when there is enough wind power to supply the loads and the extra energy is used to charge the batteries. The other state is when there is low wind power and the batteries have to compensate the lack of power, so that the isolated micro grid has enough power to supply at least the priority loads. In this paper are presented the hardware and the control algorithm for the developed system. The topology was previously tested in computer simulations, using the software PSIM 9.0, and then validated with the implementation of a laboratory prototype.

Considerando a ampla variabilidade genética de cubiu (Solanum sessiliflorum Dunal), quantificaram-se os macro e micro-nutrientes, objetivando a ampliação da tabela de composição química de alimentos típicos da região amazônica. Os frutos provenientes da Estação Experimental de Hortaliças Alejo von der Pahlen (EEH) do Instituto Nacional de Pesquisas da Amazônia (INPA), localizados no km 14 da Rodovia AM 010 em Manaus, AM, foram processados no Laboratório de Alimentos e Nutrição do INPA. Avaliaram-se oito etnovariedades de cubiu identificados como: 2 I, 3 I, 6, 7, 12, 14, 17, 29 I e III em estádio de maturação comercial. Os teores de elementos minerais foram quantificados pela técnica de Ativação por Nêutrons Instrumental e a fibra alimentar pelo método enzímico-gravimétrico. Os resultados demonstram ser o cubiu um fruto com baixo conteúdo energético (média de 33 kcal), com conteúdo de fibra alimentar total na ordem de 1,6%. Em relação aos macros elementos minerais, a etnovariedade 6, apresentou a maior concentração em potássio (513,5±3,1mg), cálcio (18,9±0,6mg) e a etnovariedade 2 I em Fe (564,4±58,1µg) e Cr (99,3±8,3µg). A menor concentração foi constatada na etnovariedade 12 para os elementos K (229,0±4,5mg), Na (53,7±5,5µg) e Zn (89,3±4,7µg). Apesar das variações em relação as diferentes etno variedades e conseqüentemente concentrações em elementos minerais, o cubiu, pode estar contribuindo para atingir as recomendações desses nutrientes.

DNA microarrays are one of the most used technologies for gene expression measurement. However, there are several distinct microarray platforms, from different manufacturers, each with its own measurement protocol, resulting in data that can hardly be compared or directly integrated. Data integration from multiple sources aims to improve the assertiveness of statistical tests, reducing the data dimensionality problem. The integration of heterogeneous DNA microarray platforms comprehends a set of tasks that range from the re-annotation of the features used on gene expression, to data normalization and batch effect elimination. In this work, a complete methodology for gene expression data integration and application is proposed, which comprehends a transcript-based re-annotation process and several methods for batch effect attenuation. The integrated data will be used to select the best feature set and learning algorithm for a brain tumor classification case study. The integration will consider data from heterogeneous Agilent and Affymetrix platforms, collected from public gene expression databases, such as The Cancer Genome Atlas and Gene Expression Omnibus.

Neste trabalho, a espécie Piper hispidinervum (pimenta longa), endêmica do estado do Acre, foi cultivada na região do Vale do Itajaí - SC, e sua adaptação foi avaliada em relação à composição química do óleo essencial obtido pelos processos de hidrodestilação convencional e por micro-ondas. O safrol foi identificado como o constituinte majoritário do óleo essencial desta espécie, o qual foi empregado como parâmetro de avaliação da capacidade de adaptação desta planta à região sul do Brasil, já que a proposta abrange avaliar a utilização desta espécie como fonte alternativa de safrol, em substituição a Canela Sassafrás (Ocotea odorifera), espécie muito explorada até a década de 90 nesta região. As amostras da planta foram obtidas de diferentes regiões do Acre e foram cultivadas na estação experimental da EPAGRI - Itajaí-SC. O óleo essencial das folhas forneceu um teor médio de safrol entre 76,6% e 89,9%. A análise por CG-DIC e CG-EM do óleo com maior concentração de safrol, apresentou os seguintes constituintes: safrol (89,93%), α-terpineno (0,35%), (E)-β-ocimeno (0,54%), terpinoleno (3,10%), valenceno (0,21%), (Z)-β-bisaboleno (1,70%) e guaiol (0,29%).

High-risk human papillomavirus (hrHPV) is an essential cause of cervical carcinoma and is also strongly related to anal cancer development. The hrHPV E6 oncoprotein plays a major role in carcinogenesis. We aimed to evaluate the frequency of hrHPV DNA and E6 oncoprotein in the anuses of women with cervical carcinoma. We analyzed 117 women with cervical cancer and 103 controls for hrHPV and the E6 oncogene. Positive test results for a cervical carcinoma included 66.7 % with hrHPV-16 and 7.7 % with hrHPV-18. One case tested positive for both HPV variants (0.9 %). The samples from the anal canal were positive for HPV-16 in 59.8 % of the cases. Simultaneous presence of HPV in the cervix and anal canal was found in 53.8 % of the cases. Regarding expression of E6 RNA, positivity for HPV-16 in the anal canal was found in 21.2 % of the cases, positivity for HPV-16 in the cervix was found in 75.0 %, and positivity for HPV-18 in the cervix was found in 1.9 %. E6 expression in both the cervix and anal canal was found in 19.2 % of the cases. In the controls, 1 % tested positive for HPV-16 and 0 % for HPV-18. Anal samples from the controls showed a hrHPV frequency of 4.9 % (only HPV16). The presence of hrHPV in the anal canal of women with cervical cancer was detected at a high frequency. We also detected E6 RNA expression in the anal canal of women with cervical cancer, suggesting that these women are at risk for anal hrHPV infection.

We assessed aquatic hyphomycete diversity in autumn and spring on oak leaves decomposing in five streams along a gradient of eutrophication in the Northwest of Portugal. Diversity was assessed through microscopy-based (identification by spore morphology) and DNA-based techniques (Denaturing Gradient Gel Electrophoresis and 454 pyrosequencing). Pyrosequencing revealed five times greater diversity than DGGE. About 21% of all aquatic hyphomycete species were exclusively detected by pyrosequencing and 26% exclusively by spore identification. In some streams, more than half of the recorded species would have remained undetected if we had relied only on spore identification. Nevertheless, in spring aquatic hyphomycete diversity was higher based on spore identification, probably because many species occurring in this season are not yet connected to ITS barcodes in genetic databases. Pyrosequencing was a powerful tool for revealing aquatic hyphomycete diversity on decomposing plant litter in streams and we strongly encourage researchers to continue the effort in barcoding fungal species.

Nanocomposite materials with an organic-inorganic urea-silicate (di-ureasil) based matrix containing gold nanoparticles (NPs) were synthesized and characterized by optical (UV/Vis) spectroscopy and indentation measurement. The urea silicate gels were obtained by reaction between silicon alkoxyde modified by isocyanate group and polyethylene glycol oligomer with amine terminal groups in presence of catalyst. The latter ensures the successful incorporation of citrate-stabilized gold NPs in the matrix. It is shown that using a convenient destabilizing agent (AgNO3) and governing the preparative conditions, the aggregation degree of gold NPs can be controlled. The developed synthesis procedure significantly simplifies the preparative procedure of gold/urea silicate nanocomposites, compared to the procedure using gold NPs, preliminary covered with silica shells. Mechanical properties of the prepared sample were characterised using depth sensing indentation methods (DSI) and an idea about the type of aggregation structures was suggested.

Combining ionic liquids (ILs) with polymers offers the prospect of new applications, where they surpass the performance of conventional media, such as organic solvents, giving advantages in terms of improved safety and a higher operating temperature range. In this work we have investigated the morphology, thermal and electrochemical properties of polymer electrolytes prepared through the addition of con- trolled quantities of the cholinium based IL N,N,N-trimethyl-N-(2-hydroxyethyl)ammonium bis(trifluo- romethylsulfonyl)imide ([N1 1 1 2(OH)] [NTf2]) to a deoxyribonucleic acid (DNA) host network. These novel IL-based electrolytes have been analyzed aiming at applications in electrochemical devices. An optimized sample showed good thermal stability up to 155 °C and a wide electrochemical window of ~3.5 V. The highest conductivity was registered for the DNA[N1 1 1 2(OH)][NTf2] (1:1) (2.82 × 10-5 and 1.09 × 10-3 S cm-1 at 30 and 100 °C, respectively).

BACKGROUND: Knowledge of cervical human papillomavirus (HPV) status might influence a cytotechnician's assessment of cellular abnormalities. The authors compared original cytotechnicians' Papanicolaou (Pap) readings for which HPV status was concealed with Pap rereads for which HPV status was revealed separately for 3 screening populations. METHODS: Previously collected cervical Pap smears and clinical data were obtained from the Canadian Cervical Cancer Screening Trial (study A), the Democratic Republic of Congo Community-Based Screening Study (study B), and the Brazilian Investigation into Nutrition and Cervical Cancer Prevention (study C). Smears were reread with knowledge of HPV status for all HPV-positive women as well as a sample of HPV-negative women. Diagnostic performance of Pap cytology was compared between original readings and rereads. RESULTS: A total of 1767 Pap tests were reread. Among 915 rereads for HPV-positive women, the contrast between "revealed" and "concealed" Pap readings demonstrated revisions from negative to positive results for 109 women (cutoff was atypical squamous cells of undetermined significance or worse) and 124 women (cutoff was low-grade squamous intraepithelial lesions [LSIL] or worse). For a disease threshold of cervical intraepithelial neoplasia of grade 2 or worse, specificity significantly declined at the atypical squamous cells of undetermined significance cutoff for studies A (86.6% to 75.3%) and C (42.5% to 15.5%), and at the LSIL cutoff for study C (61.9% to 37.6%). Sensitivity remained nearly unchanged between readings, except in study C, in which reread performance was superior (91.3% vs 71.9% for the LSIL cutoff). CONCLUSIONS: A reduction in the diagnostic accuracy of Pap cytology was observed when revealing patients' cervical HPV status, possibly due to a heightened awareness of potential abnormalities, which led to more false-positive results.
