INTRODUCTION: Emerging evidence indicates that nitric oxide (NO), which is increased in osteoarthritic (OA) cartilage, plays a role in 4-hydroxynonenal (HNE) generation through peroxynitrite formation. HNE is considered as the most reactive product of lipid peroxidation (LPO). We have previously reported that HNE levels in synovial fluids are more elevated in knees of OA patients compared to healthy individuals. We also demonstrated that HNE induces a panoply of inflammatory and catabolic mediators known for their implication in OA cartilage degradation. The aim of the present study was to investigate the ability of inducible NO synthase (iNOS) inhibitor, L-NIL (L-N6-(L-Iminoethyl)Lysine), to prevent HNE generation through NO inhibition in human OA chondrocytes. METHOD: Cells and cartilage explants were treated with or without either an NO generator (SIN or interleukin 1beta (IL-1β)) or HNE in absence or presence of L-NIL. Protein expression of both iNOS and free-radical-generating NOX subunit p47 (phox) were investigated by western blot. iNOS mRNA detection was measured by real-time RT-PCR. HNE production was analysed by ELISA, Western blot and immunohistochemistry. S-nitrosylated proteins were evaluated by Western Blot. Prostaglandin E2 (PGE2) and metalloproteinase 13 (MMP-13) levels as well as glutathione S-transferase (GST) activity were each assessed with commercial kits. NO release was determined using improved Griess method. Reactive oxygen species (ROS) generation was revealed using fluorescent microscopy with the use of commercial kits. RESULTS: L-NIL prevented IL-1β-induced NO release, iNOS expression at protein and mRNA levels, S-nitrosylated proteins and HNE in a dose dependent manner after 24h of incubation. Interestingly, we revealed that L-NIL abolished IL-1β-induced NOX component p47phox as well as ROS release. The HNE-induced PGE2 release and both cyclooxygenase-2 (COX-2) and MMP-13 expression were significantly reduced by L-NIL addition. Furthermore, L-NIL blocked the IL-1β induced inactivation of GST, an HNE-metabolizing enzyme. Also, L-NIL prevented HNE induced cell death at cytotoxic levels. CONCLUSION: Altogether, our findings support a beneficial effect of L-NIL in OA by preventing LPO process in NO-dependent and/or independent mechanisms.

Les champignons mycorhizien à arbuscules (CMA) sont des organismes pouvant établir des symbioses avec 80% des plantes terrestres. Les avantages d'une telle symbiose sont de plus en plus caractérisés et exploités en agriculture. Par contre, jusqu'à maintenant, il n'existe aucun outil permettant à la fois l'identification et la quantification de ces champignons dans le sol de façon fiable et rapide. Un tel outil permettrait, entre autres, de mieux comprendre les dynamiques des populations des endomycorhizes dans le sol. Pour les producteurs d'inoculum mycorhiziens, cela permettrait également d'établir un suivi de leurs produits en champs et d'avoir un contrôle de qualité de plus sur leurs inoculants. C'est ce que nous avons tenté de développer au sein du laboratoire du Dr. Hijri. Depuis environ une trentaine d'années, des outils d'identification et/ou de quantification ont été développés en utilisant les profiles d'acides gras, les isozymes, les anticorps et finalement l'ADN nucléaire. À ce jour, ces méthodes d’identification et de quantification sont soit coûteuses, soit imprécises. Qui plus est, aucune méthode ne permet à la fois la quantification et l’identification de souches particulières de CMA. L’ADN mitochondrial ne présente pas le même polymorphisme de séquence que celui qui rend l’ADN nucléaire impropre à la quantification. C'est pourquoi nous avons analysé les séquences d’ADN mitochondrial et sélectionné les régions caractéristiques de deux espèces de champignons mycorhiziens arbusculaires (CMA). C’est à partir de ces régions que nous avons développé des marqueurs moléculaires sous forme de sondes et d’amorces TaqMan permettant de quantifier le nombre de mitochondries de chacune de ces espèces dans un échantillon d’ADN. Nous avons ensuite tenté de déterminer une unité de quantification des CMA, soit un nombre de mitochondries par spore. C’est alors que nous avons réalisé que la méthode de préparation des échantillons de spores ainsi que la méthode d’extraction d’ADN avaient des effets significatifs sur l’unité de quantification de base. Nous avons donc optimisé ces protocoles, avant d’en e tester l’application sur des échantillons de sol et de racines ayant été inoculés avec chacune des deux espèces cibles. À ce stade, cet outil est toujours semi-quantificatif, mais il permet 9 l’identification précise de deux espèces de CMA compétentes dans des milieux saturés en phosphore inorganique. Ces résultats , en plus d’être prometteurs, ont permis d’augmenter les connaissances méthodologiques reliées à la quantification des CMA dans le sol, et suggèrent qu’à cause de leurs morphologies différentes, l’élaboration d’un protocole de quantification standardisé pour toutes les espèces de CMA demeure un objectif complexe, qui demande de nouvelles études in vivo.

An efficient one-pot synthesis of two new heterocyclic perimidines 4-(2,3-dihydro-1H-perimidin-2-yl)-2-methoxyphenol and 2-(quinoxalin-2-yl)-2,3-dihydro-1H-perimidine in good yields is presented. This methodology provides a simple, straightforward synthetic route to these interesting classes of heterocycles. Crystal structure, solvatochromism and antibacterial activity of these organic compounds are discussed.

A novel bisazomethine Schiff base was synthesised by the condensation of 3-hydroxyquinoxaline-2- carboxaldehyde and 2,3-diaminomaleonitrile. 1H NMR, 13C NMR, HPLC and FT-IR studies revealed that the compound exists in two major tautomeric forms. The Schiff base exhibits positive absorption and fluorescent solvatochromism and displays dual fluorescence with large stoke shifts. Cyclic voltammetric analysis of the compound in 1:1 methanol–THF was influenced by scan rate. Thermal analysis of the compound was undertaken using TG–DTA and DSC

The third-order non-linear absorption coefficient, b, was 1.48 x 10-6 cm W-1 and the imaginary part of the third-order non-linear optical susceptibility, Im c(3), was 3.36 x10-10 esu. The optical limiting threshold for the compound was found to be 340 MW cm-2.

Pulsed photoacoustic technique which is found to be a very convenient and accurate method, is used for the determination of absolute fluorescence quantum yield of laser dye rhodamine B. Concentration and power dependence of quantum yield of rhodamine B in methanol for excitation at 532 nm is reported here. Results show that a rapid decrease in quantum yield as the concentration is increased and finally it reaches the limit corresponding to fluorescence quenching.

Multimodal imaging agents that combine magnetic and fluorescent imaging capabilities are desirable for the high spatial and temporal resolution. In the present work, we report the synthesis of multifunctional fluorescent ferrofluids using iron oxide as the magnetic core and rhodamine B as fluorochrome shell. The core–shell structure was designed in such a way that fluorescence quenching due to the inner magnetic core was minimized by an intermediate layer of silica. The intermediate passive layer of silica was realized by a novel method which involves the esterification reaction between the epoxy group of prehydrolysed 3-Glyidoxypropyltrimethoxysilane and the surfactant over iron oxide. The as-synthesized ferrofluids have a high saturation magnetization in the range of 62–65 emu/g and were found to emit light of wavelength 640 nm ( excitation = 446 nm). Time resolved life time decay analysis showed a bi-exponential decay pattern with an increase in the decay life time in the presence of intermediate silica layer. Cytotoxicity studies confirmed the cell viability of these materials. The in vitro MRI imaging illustrated a high contrast when these multimodal nano probes were employed and the R2 relaxivity of these ∗Author to whom correspondence should be addressed. This thesis entitled “Studies on Nitrifying Microorganisms in Cochin Estuary and Adjacent Coastal Waters” reports for the first time the spatial andtemporal variations in the abundance and activity of nitrifiers (Ammonia oxidizingbacteria-AOB; Nitrite oxidizing bacteria- NOB and Ammonia oxidizing archaea-AOA) from the Cochin Estuary (CE), a monsoon driven, nutrient rich tropicalestuary along the southwest coast of India. To fulfil the above objectives, field observations were carried out for aperiod of one year (2011) in the CE. Surface (1 m below surface) and near-bottomwater samples were collected from four locations (stations 1 to 3 in estuary and 4 in coastal region), covering pre-monsoon, monsoon and post-monsoon seasons. Station 1 is a low saline station (salinity range 0-10) with high freshwater influx While stations 2 and 3 are intermediately saline stations (salinity ranges 10-25). Station 4 is located ~20 km away from station 3 with least influence of fresh water and is considered as high saline (salinity range 25- 35) station. Ambient physicochemical parameters like temperature, pH, salinity, dissolved oxygen (DO), Ammonium, nitrite, nitrate, phosphate and silicate of surface and bottom waters were measured using standard techniques. Abundance of Eubacteria, total Archaea and ammonia and nitrite oxidizing bacteria (AOB and NOB) were quantified using Fluorescent in situ Hybridization (FISH) with oligonucleotide probes labeled withCy3. Community structure of AOB and AOA was studied using PCR Denaturing Gradient Gel Electrophoresis (DGGE) technique. PCR products were cloned and sequenced to determine approximate phylogenetic affiliations. Nitrification rate in the water samples were analyzed using chemical NaClO3 (inhibitor of nitrite oxidation), and ATU (inhibitor of ammonium oxidation). Contribution of AOA and AOB in ammonia oxidation process was measured based on the recovered ammonia oxidation rate. The contribution of AOB and AOA were analyzed after inhibiting the activities of AOB and AOA separately using specific protein inhibitors. To understand the factors influencing or controlling nitrification, various statistical tools were used viz. Karl Pearson’s correlation (to find out the relationship between environmental parameters, bacterial abundance and activity), three-way ANOVA (to find out the significant variation between observations), Canonical Discriminant Analysis (CDA) (for the discrimination of stations based on observations), Multivariate statistics, Principal components analysis (PCA) and Step up multiple regression model (SMRM) (First order interaction effects were applied to determine the significantly contributing biological and environmental parameters to the numerical abundance of nitrifiers). In the CE, nitrification is modulated by the complex interplay between different nitrifiers and environmental variables which in turn is dictated by various hydrodynamic characteristics like fresh water discharge and seawater influx brought in by river water discharge and flushing. AOB in the CE are more adapted to varying environmental conditions compared to AOA though the diversity of AOA is higher than AOB. The abundance and seasonality of AOB and NOB is influenced by the concentration of ammonia in the water column. AOB are the major players in modulating ammonia oxidation process in the water column of CE. The distribution pattern and seasonality of AOB and NOB in the CE suggest that these organisms coexist, and are responsible for modulating the entire nitrification process in the estuary. This process is fuelled by the cross feeding among different nitrifiers, which in turn is dictated by nutrient levels especially ammonia. Though nitrification modulates the increasing anthropogenic ammonia concentration the anthropogenic inputs have to be controlled to prevent eutrophication and associated environmental changes.

Chemical sensors have growing interest in the determination of food additives, which are creating toxicity and may cause serious health concern, drugs and metal ions. A chemical sensor can be defined as a device that transforms chemical information, ranging from the concentration of a specific sample component to total composition analysis, into an analytically useful signal. The chemical information may be generated from a chemical reaction of the analyte or from a physical property of the system investigated. Two main steps involved in the functioning of a chemical sensor are recognition and transduction. Chemical sensors employ specific transduction techniques to yield analyte information. The most widely used techniques employed in chemical sensors are optical absorption, luminescence, redox potential etc. According to the operating principle of the transducer, chemical sensors may be classified as electrochemical sensors, optical sensors, mass sensitive sensors, heat sensitive sensors etc. Electrochemical sensors are devices that transform the effect of the electrochemical interaction between analyte and electrode into a useful signal. They are very widespread as they use simple instrumentation, very good sensitivity with wide linear concentration ranges, rapid analysis time and simultaneous determination of several analytes. These include voltammetric, potentiometric and amperometric sensors. Fluorescence sensing of chemical and biochemical analytes is an active area of research. Any phenomenon that results in a change of fluorescence intensity, anisotropy or lifetime can be used for sensing. The fluorophores are mixed with the analyte solution and excited at its corresponding wavelength. The change in fluorescence intensity (enhancement or quenching) is directly related to the concentration of the analyte. Fluorescence quenching refers to any process that decreases the fluorescence intensity of a sample. A variety of molecular rearrangements, energy transfer, ground-state complex formation and collisional quenching. Generally, fluorescence quenching can occur by two different mechanisms, dynamic quenching and static quenching. The thesis presents the development of voltammetric and fluorescent sensors for the analysis of pharmaceuticals, food additives metal ions. The developed sensors were successfully applied for the determination of analytes in real samples. Chemical sensors have multidisciplinary applications. The development and application of voltammetric and optical sensors continue to be an exciting and expanding area of research in analytical chemistry. The synthesis of biocompatible fluorophores and their use in clinical analysis, and the development of disposable sensors for clinical analysis is still a challenging task. The ability to make sensitive and selective measurements and the requirement of less expensive equipment make electrochemical and fluorescence based sensors attractive.

Der Wechsel von Tag und Nacht erzeugt einen regelmäßigen Rhythmus von verschiedenen Umweltreizen, allen voran Licht und Temperatur. Fast jedes bis zum heutigen Tage untersuchte Lebewesen besitzt einen endogenen Mechanismus zur Zeitwahrnehmung, und diese "innere Uhr" befähigt Lebewesen dazu, sich vorausschauend an rhythmische Umwelt-Änderungen anzupassen. Circadiane Rhythmen bestehen auch ohne jegliche äußere Reize und basieren auf einem molekularen Rückkopplungs-Mechanismus, der Rhythmen in Genexpression und Proteinkonzentration von etwa 24 Stunden erzeugt. Obwohl sich die grundsätzlichen Mechanismen und Komponenten dieses molekularen Uhrwerks in allen Insekten ähneln, zeigte sich jedoch immer mehr, dass es im Detail doch wesentliche Unterschiede zwischen verschiedenen Insektengruppen gibt. Während das molekulare Uhrwerk der Fruchtfliege Drosophila melanogaster inzwischen sehr gut untersucht ist, fehlen bei den meisten Insektengruppen immernoch eingehende Untersuchungen. Fast nichts ist über die molekulare Basis von circadianen Rhythmen bei der Schabe Rhyparobia maderae bekannt, obwohl diese Art bereits seit Langem als Modellorganismus in der Chronobiologie dient. Um mit der Forschung am molekularen, circadianen System von R. maderae zu beginnen, wurde die Struktur und das Expressionsprofil der core feedback loop Gene per, tim1 und cry2 analysiert. Mittels degenerierten Primern und RACE konnte das vollständige offene Leseraster (OLR) von rmPer und rmCry2, und ein Teil des rmTim1 OLR kloniert werden. Eine phylogenetische Analyse gruppierte rmPER und rmCRY2 gemeinsam mit den Orthologa hemimetaboler Insekten. Viele bei D. melanogaster funktionell charakterisierte Domänen sind bei diesen Proteinen konserviert, was auf eine ähnliche Funktion in der inneren Uhr von R. maderae hinweist. Mittels quantitativer PCR konnte gezeigt werden, dass die mRNA von rmPer, rmTim1 und rmCry2 in verschiedenen Lichtregimen in der gleichen Phasenlage Tageszeit-abhängig schwankt. Die Phasenlage stellte sich bei unterschiedlichen Photoperioden jeweils relativ zum Beginn der Skotophase ein, mit Maxima in der ersten Hälfte der Nacht. Auch im Dauerdunkel zeigen sich Rhythmen in der rmTim1 und rmCry2 Expression. Die Amplitude der rmPer Expressionsrhythmen war jedoch so gering, dass keine signifikanten Unterschiede zwischen den einzelnen Zeitgeberzeiten (ZT) festgestellt werden konnten. Mittels Laufrad-Assays wurde untersucht wie Kurz- und Langtag Lichtregime die Verhaltensrhythmen beeinflussen. Es konnten nur Unterschiede in der Periodenlänge unter freilaufenden Bedingungen festgestellt werden, wenn höhere Lichtintensitäten (1000lx) zur Synchronisation (entrainment) genutzt wurden. Die Periode des freilaufenden Rhythmus war bei Tieren aus dem Kurztag länger. Die photoperiodische Plastizität zeigte sich also auch auf Verhaltensebene, obwohl höhere Lichtintensitäten notwendig waren um einen Effekt zu beobachten. Basierend auf den Sequenzen der zuvor klonierten OLR wurden gegen rmPER, rmTIM1 und rmCRY2 gerichtete Antikörper hergestellt. Die Antikörper gegen rmPER und rmTIM1 erkannten in western blots sehr wahrscheinlich spezifisch das jeweilige Protein. Zeitreihen von Gehirngewebe-Homogenisaten zeigten keinen offensichtlichen circadianen Rhythmus in der Proteinkonzentration, wahrscheinlich auf Grund einer Oszillation mit niedriger Amplitude. In Immunhistochemischen Färbungen konnte nur mit dem gegen rmPER gerichteten Antikörper aus Kaninchen ein Signal beobachtet werden. Beinahe jede Zelle des Zentralnervensystems war rmPER-immunreaktiv im Zellkern. Es konnten keine Unterschiede zwischen den untersuchten ZTs festgestellt werden, ähnlich wie bei den western blot Zeitreihen. In dieser Studie konnten erstmals molekulare Daten der circadianen Uhr von R. maderae erfasst und dargestellt werden. Die Uhrgene per, tim1 und cry2 werden in dieser Schabenart exprimiert und ihre Domänenstruktur sowie das circadiane Expressionsmuster ähneln dem hypothetischen ursprünglichen Insektenuhrwerk, welches der circadianen Uhr von Vertebraten nahesteht. Das molekulare Uhrwerk von R. maderae kann sich an unterschiedliche Photoperioden anpassen, und diese Anpassungen manifestieren sich im Expressionsprofil der untersuchten Uhrgene ebenso wie im Verhalten.

INTRODUCCION: El dolor torácico es una de las principales causas de consulta en los servicios de urgencias y cardiología, se convierte en un reto clasificar a los pacientes empleando una herramienta diagnóstica lo suficientemente sensible y especifica para establecer riesgo y pronóstico, la estrecha relación existente entre enfermedad aterosclerótica e inflamación ha dirigido su atención al papel de marcadores plasmáticos de inflamación como predictores de riesgo de eventos cardiovasculares. La Proteína C reactiva (PCR) ha sido ampliamente estudiada en pacientes con factores de riesgo cardiovascular y Eventos coronarios Agudos, pero se desconoce el comportamiento en pacientes con dolor torácico de probabilidad intermedia. OBJETIVOS: Determinar la utilidad y comportamiento de la Proteína C reactiva en pacientes con dolor torácico de probabilidad Intermedia para síndrome coronario. MATERIALES Y METODOS: Este estudio fue realizado entre junio 2008 y febrero de 2009 en una institución de referencia en cardiológica ( Fundación Cardio Infantil, Bogotá-Colombia), Se Estudiaron pacientes con EKG normal o no diagnostico y marcadores de injuria miocardica negativos. Los pacientes continuaron su estudio según las recomendaciones y guías internacionales para dolor torácico. Nosotros realizamos dos tomas de PCR, Una PCR antes de 12 horas de iniciado el dolor torácico y otra PCR después de las 18 Hrs de iniciado el dolor torácico, se realizo la deferencia entre estas dos PCR (PCR 18 hrs vs PCR basal) Con estos 3 resultados se hizo el análisis estadístico para hallar sensibilidad, especificidad, valor predictivo positivo, valor predictivo negativo, comparándolo contra las pruebas de provocación de isquemia y cateterismo. RESULTADOS: Un total de 203 pacientes fueron analizaron. Con un promedio de edad fue de 60.8 ± 11 años, Los dos géneros tuvieron una distribución sin diferencia significativas. Los factores de riesgo asociados fueron: Hipertensión arterial 76%(n=155), Dislipidemia 68.1%(n=139), Diabetes Mellitus 20.6%(n=42), Obesidad 7.4%(n=15) y tabaquismo 9.3%(n=19). El total de cateterismos realizados fueron 66 pruebas: Normal el 27%(n=18), lesiones no significativas el 25.8%(n=17) y lesiones Obstructivas 47%(n=31). La PCR tuvo una utilidad diagnostica baja, la PCR a las 18 horas es la mejor prueba diagnóstica , con un mejor comportamiento del área de la curva ROC 0.74 (IC , 0.64-0.83), con sensibilidad del 16.13% (IC 95%, 1.57-30.69), especificidad del 98.26%( IC 955, 96.01-100), un valor predictivo negativo de 86.67%(IC 95%, 81.64-91.69). En el seguimiento a los 30 días no encontró nuevas hospitalizaciones de causa cardiovascular. CONCLUSIONES: Nuestro estudio muestra una utilidad diagnostico baja de la PCR en el dolor torácico de probabilidad intermedia para enfermedad coronaria, el mejor comportamiento diagnostico se encontró en la PCR a las 18 hrs con una alta especificidad y un alto Valor predictivo negativo para un valor de PCR > de 3mg/dl, siendo menor la utilidad de la PCR basal y diferencia de la PCR. diferencia de la PCR. Estos hallazgos no se correlacionaron con estudios previos. No se pudo establecer un punto de Corte de la PCR diferente a los ya existentes debido a la variabilidad de la PCR entre la población de estudio. Las limitaciones encontradas en nuestro estudio hacen necesaria la realización de un estudio multicéntrico.

La PCR es el marcador de inflamación vascular más estudiado y validado hasta la fecha. Los niveles de PCR ultrasensible pueden predecir el riesgo de enfermedad cardiovascular. En el programa “salud integral para la mujer” de la fundación cardio-infantil se realizan estratificaciones de riesgo cardiovascular a mujeres adultas. Se midieron los niveles de PCR en este grupo de pacientes entre Octubre del 2007 y Mayo del 2009 y se evaluó la correlación entre la estratificación del riesgo cardiovascular por escala de Framingham y niveles de PCR en esta población. Objetivo: establecer el grado de correlación entre los niveles de PCR ultrasensible y el grado de riesgo cardiovascular y otros factores de riesgo cardiovascular. Resultados: Edad promedio 48 años (18 a 98 años). 62 pacientes hipertensas (40,7%). 19 pacientes con cifras alteradas de glicemia en ayunas (12%). Hay un 83% de la población estudiada con dislipidemia (127 pacientes). 78 pacientes con sobrepeso u obesidad. El 87% de la población tiene al menos un factor de riesgo presente. Discusión y resultados: En la población estudiada existe una alta prevalencia de factores de riesgo cardiovasculares, dentro de los cuales predomina los trastornos del metabolismo de los lípidos. Sin embargo no hay correlación entre los niveles de PCR ultrasensible y el riesgo cardiovascular según la escala de Framingham. En la literatura médica publicada sobre PCR ultrasensible y su relación con el riesgo cardiovascular los resultados son muy divergentes con los encontrados en este estudio.

La proteína C reactiva ultrasensible es un biomarcador de inflamación vascular más estudiado y validado hasta la fecha. En el presente estudio se determino la correlación entre la PCR y el riesgo cardiovascular estratificado mediante la escala europea y el método de farmingham, en pacientes incluidas desde septiembre de 2007 hasta septiembre de 2011. Objetivo: establecer el grado de correlación entre los niveles de PCR ultrasensible y el grado de riesgo cardiovascular. Resultados: se incluyeron un total (N=299) mujeres mayores de 18 años, con promedio de edad de 48 años; según los niveles de PCR (89%) N= 269 tenían riesgo cardiovascular bajo. Con la estratificación de riesgo según la escala europea solo un (0.66%) N=2 se catalogaban como alto riesgo cardiovascular, implementando la escala de framingham un (74%) N=222 estaban en bajo riesgo. Predominando el riesgo cardiovascular bajo y datos de PCR en riesgo bajo. Encontramos muy débil correlación positiva entre los niveles de PCR y el riesgo cardiovascular calculado por el método de framingham para las pacientes (correlación de Pearson 0.323 con significancia estadística de 0,01) Finalmente encontramos una correlación inversa entre los niveles de PCR y HDL en la muestra estudiada estadísticamente significativa. Discusión: En la población estudiada existe una alta prevalencia de factores de riesgo cardiovasculares, destacamos la utilidad del método de framingham para discriminar riesgo cardiovascular en comparación con la escala europea. Además en el presente estudio, se destacan las correlaciones de la PCR con el perfil lipídico. Finalmente encontramos un correlación inversa entre los niveles de PCR y HDL en la muestra estudiada estadísticamente significativa, con lo cual podemos concluir posible utilidad tanto para el diagnostico y alternativa de estratificación en riesgo cardiovascular en nuestra población.
