The traditional analysis of tourism, having in mind only its economic impacts has been shown to be reductive and insufficient to explain the numerous and versatile modifications these can and will stimulate in a society at many levels, e.g. socially, culturally, politically and in the environment. The complexity of touristic activities and the insufficient measuring instruments that can provide exact data about these, gives terrain to the emergence of myths and value judgments around the effects in countries where tourism is a reality. This study aims at understanding how the impacts of tourism are grasped by the local community in Sal island – Cape Verde – convinced as we are that a quality and sustainable touristic offer can only be done by trialing the population, and involving them in the planning, managing and monitoring processes. The analysis of the perception of the impacts of touristic activities by the population tells us a lot about the levels of satisfaction of such communities towards the way in which the touristic development has been carried out in their surroundings. This study has been made through the inquiry of 231 locals, by means of a questionnaire, that showed that the population in this island has a very clear conscience of the impacts of tourism in their day-to-day lives. Conclusions drawn are that the negative economic and social impacts are greater than the positive; the cultural and environmental impacts are not so significant, and that the people feel that their voice has not been heard in what planning touristic activities is concerned. Nevertheless, they have high expectations regarding tourism as a way of ameliorating their life conditions. The inexistence of a linear behavior of impacts of touristic activities in the receptive countries and a perfect and adjustable model for tourism development make these countries delineate new politics aiming at the sustainability and the creation of conditions that help them monitor and mitigate its negative impacts.

Models are presented for the optimal location of hubs in airline networks, that take into consideration the congestion effects. Hubs, which are the most congested airports, are modeled as M/D/c queuing systems, that is, Poisson arrivals, deterministic service time, and {\em c} servers. A formula is derived for the probability of a number of customers in the system, which is later used to propose a probabilistic constraint. This constraint limits the probability of {\em b} airplanes in queue, to be lesser than a value $\alpha$. Due to the computational complexity of the formulation. The model is solved using a meta-heuristic based on tabu search. Computational experience is presented.

Psychological control refers to parental behaviors that intrude on the psychological and emotional development of the child. In 2010, Soenens and colleagues proposed a distinction between two domain-specific expressions of psychological control, that is, Dependency-oriented Psychological Control (DPC) and Achievement-oriented Psychological Control (APC). The aim of this study was to evaluate the factor structure, reliability, and convergent validity of the French form of the Dependency-oriented and Achievement-oriented Psychological Control Scale (DAPCS; Soenens, Vansteenkiste, and Luyten, 2010) in a sample of late adolescents (N = 291, mean age = 21.65). Confirmatory factor analyses confirmed the hypothesized two-factor solution of the DAPCS for paternal as well as for maternal ratings. Moreover, high indices of internal consistency indicated that both subscales produced reliable scores. Further, convergent validity was confirmed by theoretically consistent associations between the DAPCS' subscales and well-established assessments of general parenting style dimensions. Finally, results evidenced gender specific patterns supporting the relevance of domain differentiation in the assessment of psychological control. Overall, the results of this study indicated that the French form of the DAPCS might be a useful instrument to assess two domainspecific types of parental psychological control among French-speaking adolescents.

Akt/protein kinase B (PKB) plays a critical role in the regulation of metabolism, transcription, cell migration, cell cycle progression, and cell survival. The existence of viable knockout mice for each of the three isoforms suggests functional redundancy. We generated mice with combined mutant alleles of Akt1 and Akt3 to study their effects on mouse development. Here we show that Akt1-/- Akt3+/- mice display multiple defects in the thymus, heart, and skin and die within several days after birth, while Akt1+/- Akt3-/- mice survive normally. Double knockout (Akt1-/-) Akt3-/-) causes embryonic lethality at around embryonic days 11 and 12, with more severe developmental defects in the cardiovascular and nervous systems. Increased apoptosis was found in the developing brain of double mutant embryos. These data indicate that the Akt1 gene is more essential than Akt3 for embryonic development and survival but that both are required for embryo development. Our results indicate isoform-specific and dosage-dependent effects of Akt on animal survival and development.

The respective production of specific immunoglobulin (Ig)G2a or IgG1 within 5 d of primary immunization with Swiss type mouse mammary tumor virus [MMTV(SW)] or haptenated protein provides a model for the development of T helper 1 (Th1) and Th2 responses. The antibody-producing cells arise from cognate T cell B cell interaction, revealed by the respective induction of Cgamma2a and Cgamma1 switch transcript production, on the third day after immunization. T cell proliferation and upregulation of mRNA for interferon gamma in response to MMTV(SW) and interleukin 4 in response to haptenated protein also starts during this day. It follows that there is minimal delay in these responses between T cell priming and the onset of cognate interaction between T and B cells leading to class switching and exponential growth. The Th1 or Th2 profile is at least partially established at the time of the first cognate T cell interaction with B cells in the T zone. The addition of killed Bordetella pertussis to the hapten-protein induces nonhapten-specific IgG2a and IgG1 plasma cells, whereas the anti-hapten response continues to be IgG1 dominated. This indicates that a Th2 response to hapten-protein can proceed in a node where there is substantial Th1 activity.

A three-dimensional cell culture system was used as a model to study the influence of low levels of mercury in the developing brain. Aggregating cell cultures of fetal rat telencephalon were treated for 10 days either during an early developmental period (i.e., between days 5 and 15 in vitro) or during a phase of advanced maturation (i.e., between days 25 and 35) with mercury. An inorganic (HgCl2) and an organic mercury compound (monomethylmercury chloride, MeHgCl) were examined. By monitoring changes in cell type-specific enzymes activities, the concentration-dependent toxicity of the compounds was determined. In immature cultures, a general cytotoxicity was observed at 10(-6) M for both mercury compounds. In these cultures, HgCl2 appeared somewhat more toxic than MeHgCl. However, no appreciable demethylation of MeHgCl could be detected, indicating similar toxic potencies for both mercury compounds. In highly differentiated cultures, by contrast, MeHgCl exhibited a higher toxic potency than HgCl2. In addition, at 10(-6) M, MeHgCl showed pronounced neuron-specific toxicity. Below the cytotoxic concentrations, distinct glia-specific reactions could be observed with both mercury compounds. An increase in the immunoreactivity for glial fibrillary acidic protein, typical for gliosis, could be observed at concentrations between 10(-9) M and 10(-7) M in immature cultures, and between 10(-8) M and 3 x 10(-5) M in highly differentiated cultures. A conspicuous increase in the number and clustering of GSI-B4 lectin-binding cells, indicating a microglial response, was found at concentrations between 10(-10) M and 10(-7) M. These development-dependent and cell type-specific effects may reflect the pathogenic potential of long-term exposure to subclinical doses of mercury.

Työssä käsitellään Helsingin Energian sähköasemaprojektoinnin kehittämistä. Helsingin kaupunkiolosuhteissa valmiiden ratkaisujen hyödyntäminen ei ole usein mahdollista, vaan asemat on suunniteltava itsenäisinä, erilaisina projekteina. Rakennettavat sähköasemat ovat suuria ja eri osien kilpailuttamisella saavutetaan taloudellista hyötyä. HelenEngineeringillä on mahdollisuus toimia haastavissa projekteissa konsulttimaisena projektinhoitajana. Kun sähköasemaprojektien tarve on jatkuva, Helsingin Energialla on syytä säilyttää oma sähköasemaprojektointiosaaminen. Työtä varten tehtiin haastattelututkimus sähköasemaprojekteja toteuttaville yrityksille ja perehdyttiin kahteen viimeisimpään Helsingin Energialla toteutettuun sähköasemaprojektiin, jotka olivat Viikinmäen 20 kV ja Suvilahden110 kV sähköasemien uusimiset. Näiden perusteella muodostettiin malli toteuttaasähköasemaprojekteja Helsingin Energialla. Malli sisältää muun muassa rungon projektivaiheistukselle, ositukselle, organisaatiokaaviolle, aikataululle ja näkökulmia sopimusten laadintaan. Mallin täysimittainen soveltaminen vaatii kuitenkinjatkuvaa, suunniteltua kehitystyötä. Myös mallia on kehitettävä jatkuvasti.

Työssä tutkittiin kunnossapidon mahdollisuuksia vaikuttaa laitoksen elinkaareen ja elinkaaren aikaisiin kustannuksiin. Lisäksi tutkittiin teknisen suunnittelun vaikutusta kunnossapidettävyyteen ja laitoksen elinkaarikustannuksiin. Työn kirjallisuusosassa on tarkasteltu nykyaikaisia kunnossapidon menetelmiä.Erityisesti on tutkittu luotettavuuskeskeistä kunnossapitoa ja kunnossapidon tehtäviä laitoksen elinkaaren aikana. Investoinnin laitevalinnoilla ja niiden suunnittelulla on myös merkittävä vaikutus käyttöön ja erityisesti kunnossapitoon. Työn empiirisessä osassa selvitettiin Botnia Mill Servicen jaMetsä - Botnian toimintamalleja Joutsenon sellutehtaalla. Erityisesti keskityttiin suuriin korjauksiin ja niiden kustannusten hallintaan. Myös kunnossapidon toimintamallia ja sen kehittämistä on tutkittu. Kokeellisessa osassa selvitettiin kunnossapidolle tyypillinen toimintamalli laitoksen elinkaaren hallintaa varten. Tehdyn selvityksen pohjalta voidaan todeta, että pitkän tähtäimen suunnittelu luo pohjaa kustannusten hallinnalle ja laitteiden kunnon ylläpidolle. Suunnittelun aikajänne tulisi olla laitteiden kunnostusten osalta vähintään kolme vuotta. Koko laitoksen kattava elinkaarisuunnitelma on hyvä laatia joheti käyttöönottovaiheen jälkeen. Pitkäjänteisellä suunnittelulla ja sen toteutuksella pystytään parhaalla mahdollisella tavalla ylläpitämään käyttöomaisuuden arvo. Kunnossapidolla on mahdollisuus vaikuttaa laitoksen käyttöomaisuuden arvonylläpitoon hallitsemalla elinkaarikustannuksia. Kunnossapidon onnistuneen toteutuksen takaa oikeanlainen johtaminen ja jatkuva osaamisen ylläpito. Myös kunnossapidossa sovitut toimintamallit ja järjestelmien jatkuva ylläpito ja kehittäminen takaavat onnistuneen toiminnan.

Tämän pro gradu -tutkielman aiheena on yritysverkostojen kehittäminen sekä siihen liittyvät käsitteet ja menetelmät. Tutkielman tavoitteena on selkiyttää verkostoitumiseen, verkostosuhteen kehittymiseen ja yritysverkoston kehittämiseen liittyviä käsitteitä. Tutkielmassa verkostoitumista tarkastellaan transaktiokustannusteorian, resurssiperusteisen näkemyksen ja IMP-tutkimusryhmän vuorovaikutusnäkökulman mukaisesti. Verkostoituminen nähdään osana organisaation uudistumisprosessia ja sitä mallinnetaan verkostosuhteen kehittymisprosessin avulla. Verkoston kehittäminen on jatke organisaation uudistumisprosessille ja osa verkostosuhteen kehittymisprosessia. Verkoston kehittämiseen liittyen tutkielmassa tarkastellaan, miten verkoston kehittäminen eroaa yrityksen sisäisestä kehittämisestä, ja millaisia haasteita ja menestystekijöitä yhteisestä kehittämistoiminnasta on löydettävissä. Tutkielmassa luokitellaan myös kehittämiskohteita sekä tarkastellaan, millaisella kehittämistyön organisoitumismallilla sekä työkaluilla ja menetelmillä yritysverkoston kehittämistä voidaan tukea.

Finland has large forest fuel resources. However, the use of forest fuels for energy production has been low, except for small-scale use in heating. According to national action plans and programs related to wood energy promotion, the utilization of such resources will be multiplied over the next few years. The most significant part of this growth will be based on the utilization of forest fuels, produced from logging residues of regeneration fellings, in industrial and municipal power and heating plants. Availability of logging residues was analyzed by means of resource and demand approaches in order to identify the most suitable regions with focus on increasing the forest fuel usage. The analysis included availability and supply cost comparisons between power plant sites and resource allocation in a least cost manner, and between a predefined power plant structure under demand and supply constraints. Spatial analysis of worksite factors and regional geographies were carried out using the GIS-model environment via geoprocessing and cartographic modeling tools. According to the results of analyses, the cost competitiveness of forest fuel supply should be improved in order to achieve the designed objectives in the near future. Availability and supply costs of forest fuels varied spatially and were very sensitive to worksite factors and transport distances. According to the site-specific analysis the supply potential between differentlocations can be multifold. However, due to technical and economical reasons ofthe fuel supply and dense power plant infrastructure, the supply potential is limited at plant level. Therefore, the potential and supply cost calculations aredepending on site-specific matters, where regional characteristics of resourcesand infrastructure should be taken into consideration, for example by using a GIS-modeling approach constructed in this study.

Reusability has become more popular factor in modern software engineering. This is mainly because object-orientation has brought methods that allow reusing more easily. Today more and more application developer thinks how they can reuse already existing applications in their work. If the developer wants to use existing components outside the current project, he can use design patterns, class libraries or frameworks. These provide solution for specific or general problems that has been already encountered. Application frameworks are collection of classes that provides base for the developer. Application frameworks are mostly implementation phase tools, but can also be used in application design. The main purpose of the frameworks is separate domain specific functionalities from the application specific. Usually the frameworks are divided into two categories: black and white box. Difference between those categories is the way the reuse is done. The application frameworks provide properties that can be examined and compared between different frameworks. These properties are: extensibility, reusability, modularity and scalability. These examine how framework will handle different platforms, changes in framework, increasing demand for resources, etc. Generally application frameworks do have these properties in good level. When comparing general purpose framework and more specific purpose framework, the main difference can be located in reusability of frameworks. It is mainly because the framework designed to specific domain can have constraints from external systems and resources. With general purpose framework these are set by the application developed based on the framework.

Diplomityön tavoitteena on suunnitella ja toteuttaa tuotannon budjettimalli elektroniikka-alalla toimivalle yritykselle. Työn onnistumisen kannalta keskeistä on löytää vanhan budjettimallin puutteet sekä uudet tietotarpeet, joita tuotannon johto tarvitsee pyrittäessä kustannustehokkaaseen ja kannattavaan tuotantotoimintaan. Näiden tietojen pohjalta suunnitellaan ja toteutetaan uusi budjettimalli sekä kuvataan sen toimintaperiaatteet. Työn teoreettisessa osuudessa esitellään budjetoinnin tavoitteet, hyödyt, kritiikin kohteet sekä tyypillisen teollisuusyrityksen osabudjetit. Myös perinteiseen budjetointiin läheisesti liittyvät muodot ja sovellukset, kuten toimintopohjainen budjetointi, vastuualuelaskenta ja nollapohjabudjetointi esitellään. Budjettiseurannan ja eroanalyysin tärkeä merkitys tuodaan esiin. Ennen uuden mallin esittämistä tutkitaan vanhan budjettimallin toimintaperiaatteet ja heikkoudet. Empiirisessä osassa keskitytään tuotannon budjetin toimintaperiaatteisiin sen sisältämien osabudjettien kautta. Työn loppupuolella esitellään uuden budjettimallin antamat tulokset ja poikkeamat sekä arvioidaan budjettimallin toimivuutta. Budjettimallin tuottaman informaation pohjalta tuotannonjohtoa ja suunnittelua ryhdyttiin välittömästi tehostamaan välillisten kustannusten alentamiseksi. Tehostustoimet ilmenivät konkreettisesti positiivisena erona poikkeama-analyysissä suhteessa budjettiin. Rakennetun budjettimallin mallinnustarkkuus osoittautui hyväksi ja uudet esiin tuodut kustannuserät hyödylliseksi informaatioksi.

Neurofilament proteins (NFs) are the major components of the intermediate filaments of the neuronal cytoskeleton. The three different NF proteins; the low (NF-L), medium (NF-M),and dendrites.NF proteins play an important role in neuronal development, and plasticity,and seem to contribute to the pathophysiology of several diseases. However, the detailed expression patterns of NF proteins in the course of postnatal aturation, and in response to seizures in the rat have remained unknown. In this work, I have studied the developmental expression and cellular distribution of the three NF proteins in the rat hippocampus during the postnatal development. The reactivity of NF proteins in response to kainic acid (KA)-induced status epilepticus (SE)was studied in the hippocampus of 9-day-old rats, and using in vitro organotypic hippocampal slices cultures prepared from P6-7 rats. The results showed that NF-L and NF-M proteins are expressed already at the postnatal day 1, while the expression of NF-H mainly occurred during the second postnatal week. The immunoreactivity of NF proteins varied depending on the cell type and sub-cellular location in the hippocampus. In adult rats, KA-induced SE typically results in severe and permanent NF degradation. However, in our P9 rats KA-induced SE resulted in a transient increase in the expression of NF proteins during the first few hours but not degradation. No neuronal death or mossy fiber sprouting was observed at any time after SE. The in vitro studies with OHCs, which mimick the in vivo developing models where a local injection of KA is applied(e.g. intrahippocampal), indicated that NF proteins were rapidly degraded in response to KA treatment, this effect being effectively inhibited by the treatment with the AMPA receptor antagonist CNQX, and calpain inhibitor MDL-28170. These compounds also significantly ameliorated the KA-induced region-specific neuronal damage. The NMDA receptor antagonist and the L-type Ca2+ channel blocker did not have any significant effect. In conclusion, the results indicate that the developmental expression of NF in the rat hippocampus is differentially regulated and targeted in the different hippocampal cell types during the postnatal development. Furthermore, despite SE, the mechanisms leading to NF degradation and neuronal death are not activated in P9 rats unlike in adults. The reason for this remains unknown. The results in organotypic hippocampal cultures confirm the validity of this in vitro model to study development processes, and to perform pharmacological studies. The results also suggest that calpain proteases as interesting pharmacological targets to reduce neuronal damage after acute excitotoxic insults.

A major problem in developmental neurotoxicity (DNT) risk assessment is the lack of toxicological hazard information for most compounds. Therefore, new approaches are being considered to provide adequate experimental data that allow regulatory decisions. This process requires a matching of regulatory needs on the one hand and the opportunities provided by new test systems and methods on the other hand. Alignment of academically and industrially driven assay development with regulatory needs in the field of DNT is a core mission of the International STakeholder NETwork (ISTNET) in DNT testing. The first meeting of ISTNET was held in Zurich on 23-24 January 2014 in order to explore the concept of adverse outcome pathway (AOP) to practical DNT testing. AOPs were considered promising tools to promote test systems development according to regulatory needs. Moreover, the AOP concept was identified as an important guiding principle to assemble predictive integrated testing strategies (ITSs) for DNT. The recommendations on a road map towards AOP-based DNT testing is considered a stepwise approach, operating initially with incomplete AOPs for compound grouping, and focussing on key events of neurodevelopment. Next steps to be considered in follow-up activities are the use of case studies to further apply the AOP concept in regulatory DNT testing, making use of AOP intersections (common key events) for economic development of screening assays, and addressing the transition from qualitative descriptions to quantitative network modelling.

Current-day web search engines (e.g., Google) do not crawl and index a significant portion of theWeb and, hence, web users relying on search engines only are unable to discover and access a large amount of information from the non-indexable part of the Web. Specifically, dynamic pages generated based on parameters provided by a user via web search forms (or search interfaces) are not indexed by search engines and cannot be found in searchers’ results. Such search interfaces provide web users with an online access to myriads of databases on the Web. In order to obtain some information from a web database of interest, a user issues his/her query by specifying query terms in a search form and receives the query results, a set of dynamic pages that embed required information from a database. At the same time, issuing a query via an arbitrary search interface is an extremely complex task for any kind of automatic agents including web crawlers, which, at least up to the present day, do not even attempt to pass through web forms on a large scale. In this thesis, our primary and key object of study is a huge portion of the Web (hereafter referred as the deep Web) hidden behind web search interfaces. We concentrate on three classes of problems around the deep Web: characterization of deep Web, finding and classifying deep web resources, and querying web databases. Characterizing deep Web: Though the term deep Web was coined in 2000, which is sufficiently long ago for any web-related concept/technology, we still do not know many important characteristics of the deep Web. Another matter of concern is that surveys of the deep Web existing so far are predominantly based on study of deep web sites in English. One can then expect that findings from these surveys may be biased, especially owing to a steady increase in non-English web content. In this way, surveying of national segments of the deep Web is of interest not only to national communities but to the whole web community as well. In this thesis, we propose two new methods for estimating the main parameters of deep Web. We use the suggested methods to estimate the scale of one specific national segment of the Web and report our findings. We also build and make publicly available a dataset describing more than 200 web databases from the national segment of the Web. Finding deep web resources: The deep Web has been growing at a very fast pace. It has been estimated that there are hundred thousands of deep web sites. Due to the huge volume of information in the deep Web, there has been a significant interest to approaches that allow users and computer applications to leverage this information. Most approaches assumed that search interfaces to web databases of interest are already discovered and known to query systems. However, such assumptions do not hold true mostly because of the large scale of the deep Web – indeed, for any given domain of interest there are too many web databases with relevant content. Thus, the ability to locate search interfaces to web databases becomes a key requirement for any application accessing the deep Web. In this thesis, we describe the architecture of the I-Crawler, a system for finding and classifying search interfaces. Specifically, the I-Crawler is intentionally designed to be used in deepWeb characterization studies and for constructing directories of deep web resources. Unlike almost all other approaches to the deep Web existing so far, the I-Crawler is able to recognize and analyze JavaScript-rich and non-HTML searchable forms. Querying web databases: Retrieving information by filling out web search forms is a typical task for a web user. This is all the more so as interfaces of conventional search engines are also web forms. At present, a user needs to manually provide input values to search interfaces and then extract required data from the pages with results. The manual filling out forms is not feasible and cumbersome in cases of complex queries but such kind of queries are essential for many web searches especially in the area of e-commerce. In this way, the automation of querying and retrieving data behind search interfaces is desirable and essential for such tasks as building domain-independent deep web crawlers and automated web agents, searching for domain-specific information (vertical search engines), and for extraction and integration of information from various deep web resources. We present a data model for representing search interfaces and discuss techniques for extracting field labels, client-side scripts and structured data from HTML pages. We also describe a representation of result pages and discuss how to extract and store results of form queries. Besides, we present a user-friendly and expressive form query language that allows one to retrieve information behind search interfaces and extract useful data from the result pages based on specified conditions. We implement a prototype system for querying web databases and describe its architecture and components design.
