PURPOSE. To examine the effects of transcorneal electrical stimulation (TES) on retinal degeneration of light-exposed rats. METHODS. Thirty-three Sprague Dawley albino rats were divided into three groups: STIM (n = 15) received 60 minutes of TES, whereas SHAM (n = 15) received identical sham stimulation 2 hours before exposure to bright light with 16,000 lux; healthy animals (n = 3) served as controls for histology. At baseline and weekly for 3 consecutive weeks, dark-and light-adapted electroretinography was used to assess retinal function. Analysis of the response versus luminance function retrieved the parameters Vmax (saturation amplitude) and k (luminance to reach 1/2Vmax). Retinal morphology was assessed by histology (hematoxylin-eosin [HE] staining; TUNEL assay) and immunohistochemistry (rhodopsin staining). RESULTS. Vmax was higher in the STIM group compared with SHAM 1 week after light damage (mean intra-individual difference between groups 116.06 mu V; P = 0.046). The b-wave implicit time for the rod response (0.01 cd.s/m(2)) was lower in the STIM group compared with the SHAM group 2 weeks after light damage (mean intra-individual difference between groups 5.78 ms; P = 0.023); no other significant differences were found. Histological analyses showed photoreceptor cell death (TUNEL and HE) in SHAM, most pronounced in the superior hemiretina. STIM showed complete outer nuclear layer thickness preservation, reduced photoreceptor cell death, and preserved outer segment length compared with SHAM (HE and rhodopsin). CONCLUSIONS. This sham-controlled study shows that TES can protect retinal cells against mild light-induced degeneration in Sprague Dawley rats. The ALICE experiment at the LHC has studied J/psi production at mid-rapidity in pp collisions at root s = 7 TeV through its electron pair decay on a data sample corresponding to an integrated luminosity L-int = 5.6 nb(-1). The fraction of J/psi from the decay of long-lived beauty hadrons was determined for J/psi candidates with transverse momentum p(t) > 1,3 GeV/c and rapidity vertical bar y vertical bar < 0.9. The cross section for prompt J/psi mesons, i.e. directly produced J/psi and prompt decays of heavier charmonium states such as the psi(2S) and chi(c) resonances, is sigma(prompt J/psi) (p(t) > 1.3 GeV/c, vertical bar y vertical bar < 0.9) = 8.3 +/- 0.8(stat.) +/- 1.1 (syst.)(-1.4)(+1.5) (syst. pol.) mu b. The cross section for the production of b-hadrons decaying to J/psi with p(t) > 1.3 GeV/c and vertical bar y vertical bar < 0.9 is a sigma(J/psi <- hB) (p(t) > 1.3 GeV/c, vertical bar y vertical bar < 0.9) = 1.46 +/- 0.38 (stat.)(-0.32)(+0.26) (syst.) mu b. The results are compared to QCD model predictions. The shape of the p(t) and y distributions of b-quarks predicted by perturbative QCD model calculations are used to extrapolate the measured cross section to derive the b (b) over bar pair total cross section and d sigma/dy at mid-rapidity.

The aim of the study was to screen 11 selected traditional medicinal plants from West Africa for their in vitro antiplasmodial activity in order to determine the activity of single and of combination of plant extracts and to examine the activity of isolated pure compounds. Ethanolic and aqueous extracts of the 11 selected plants and pure compounds from Phyllanthus muellerianus and Anogeissus leiocarpus were tested in vitro against Plasmodium falciparum 3D7. Proliferation inhibitory effects were monitored after 48 h. Among the plants and pure compounds investigated in this study, geraniin from P. muellerianus, ellagic, gentisic, and gallic acids from A. leiocarpus, and extracts from A. leiocarpus, P. muellerianus and combination of A. leiocarpus with P. muellerianus affected the proliferation of P. falciparum most potently. Significant inhibitory activity was observed in combination of A. leiocarpus with P. muellerianus (IC50 = 10.8 mu g/ml), in combination of A. leiocarpus with Khaya senegalensis (IC50 = 12.5 mu g/ml), ellagic acid (IC50 = 2.88 mu M), and geraniin (IC50 = 11.74 mu M). In general growth inhibition was concentration-dependent revealing IC50 values ranging between 10.8 and -40.1 mu g/ml and 2.88 and 11.74 mu M for plant extracts and pure substances respectively. Comparison with literature sources of in vivo and in vitro toxicity data revealed that thresholds are up to two times higher than the determined IC50 values. Thus, the present study suggests that geraniin from P. muellerianus; ellagic acid, gallic acid, and gentisic acid from A. leiocarpus; and combination of extracts from A. leiocarpus with either P. muellerianus or K. senegalensis could be a potential option for malaria treatment.

Bacterial capsular polysaccharides (PS) which naturally contain zwitterionic charge motifs (ZPS) possess specific immunostimulatory activity, leading to direct activation of antigen-presenting cells (APCs) through Toll-like receptor 2 (TLR2) and of T cells in co-culture systems. When administered intraperitoneally, ZPS and bacteria expressing them are involved in the induction or regulation of T-cell dependent inflammatory processes such as intra-abdominal abscess formation. Moreover it has been published that ZPSs are processed to low molecular weight carbohydrates and presented to T cells through a pathway similar to that used for protein antigens. These findings were in contrast with the paradigm according to which polysaccharides are T-independent antigens unable to be presented in association with MHC class II molecules and unable to induce a protective immune response. For this reason in glycoconjugate vaccines polysaccharides often need to be conjugated to a carrier protein to induce protection. The aim of our work was to generate vaccine candidates with antigen and adjuvant properties in one molecule by the chemical introduction of a positive charge into naturally anionic PS from group B streptococcus (GBS). The resulting zwitterionic PS (ZPS) has the ability to activate human and mouse APCs, and in mixed co-cultures of monocytes and T cells, ZPS induce MHC II-dependent T-cell proliferation and up-regulation of activation markers. TLR2 transfectants show reporter gene transcription upon incubation with ZPS and these stimulatory qualities can be blocked by anti-TLR2 mAbs or by the destruction of the zwitterionic motif. However, in vivo, ZPS used alone as vaccine antigen failed to induce protection against GBS challenge, a result which does not confirm the above mentioned postulate that ZPS are T-cell dependent Ags by virtue of their charge motif. Thus to make ZPS visible to the immune system we have conjugated ZPS with a carrier protein. ZPS-glycoconjugates induce higher T cell and Ab responses to carrier and PS, respectively, compared to control PS-glycoconjugates made with the native polysaccharide form. Moreover, protection of mothers or neonate offspring from lethal GBS challenge is better when mothers are immunized with ZPS-conjugates compared to immunization with PS-conjugates. In TLR2 knockout mice, ZPS-conjugates lose both their increased immunogenicity and protective effect after vaccination. When ZPS are co-administered as adjuvants with unconjugated tetanus toxoid (TT), they have the ability to increase the TT-specific antibody titer. In conclusion, glycoconjugates containing ZPS are potent vaccines. They target Ag to TLR2-expressing APCs and activate these APCs, leading to better T cell priming and ultimately to higher protective Ab titers. Thus, rational chemical design can generate potent novel PS-adjuvants with wide application, including glycoconjugates and co-administration with unrelated protein Ags.

I poriferi rappresentano un importante campo di ricerca anche in ambito applicativo in quanto potenzialmente utili come fonte di metaboliti secondari da impiegarsi in ambito clinico (antitumorali, antibiotici, antivirali, ecc.) e industriale (antifouling). I processi di biosilicificazione interessano invece per aspetti legati alle biotecnologie marine. Questo Phylum ha un importante ruolo strutturale e funzionale nell’ecologia dei popolamenti bentonici, in quanto può essere dominante in numerosi habitat e svolgere un ruolo ecologico fondamentale nelle dinamiche degli ecosistemi marini. Per questo, la variazione spaziale e temporale della loro abbondanza può avere effetti considerevoli su altri membri della comunità. Lo studio delle dinamiche di popolazione e del ciclo riproduttivo dei poriferi potrebbe permettere di valutare come i cambiamenti climatici ne influenzino la crescita e la riproduzione e potrebbe quindi fornire una base per lo sviluppo di corrette tecniche di gestione ambientale. La spugna Axinella polypoides è inserita all’interno delle liste di protezione della Convenzione di Berna e di Barcellona, dove sono elencate le specie da proteggere perché minacciate o in pericolo di estinzione. Questa specie, avendo una morfologia eretta, è fortemente minacciata soprattutto da attività antropiche quali pesca e ancoraggi, ma nonostante questo la letteratura relativa ad essa è scarsa, La sua importanza è legata soprattutto al recente utilizzo come modello per numerosi esperimenti. A. polypoides rappresenta, infatti, il più basso livello nella scala evolutiva in cui sono stati rinvenuti meccanismi biochimici cellulari di reazione all’aumento di temperatura (incremento dell’attività ADP-ribosil ciclasica, sintesi di ossido nitrico) tipici degli organismi superiori. Lo scopo di questa tesi è di aumentare le conoscenze sull’ecologia e sulla biologia di questo porifero, al fine di consentire una migliore predisposizione di eventuali piani di tutela. Dallo studio delle colonie effettuato presso l’Isola Gallinara (SV), emerge una dinamica di crescita lenta ed un ciclo riproduttivo estivo, coerentemente con quanto osservato per altre specie mediterranee del genere Axinella. Le analisi istologiche effettuate hanno mostrato variabilità temporale nella densità e nella dimensione di particolari cellule sferulose, che si ipotizza siano collegate a fenomeni di proliferazione cellulare e rigenerazione in seguito a danni. È stata individuata inoltre la presenza di una particolare tipologia cellulare dendritica la cui funzione si ritiene abbia affinità con le funzioni sensoriali di Phyla superiori. Queste osservazioni, e l’evidente vulnerabilità della specie agli impatti antropici, hanno evidenziato la necessità di sviluppare adeguati piani di monitoraggio e di conservazione.

Diese Arbeit befasst sich mit Eduard Study (1862-1930), einem der deutschen Geometer um die Jahrhundertwende, der seine Zeit zum Einen durch seine Kontakte zu Klein, Hilbert, Engel, Lie, Gordan, Halphen, Zeuthen, Einstein, Hausdorff und Weyl geprägt hat, zum Anderen in ihr aber auch für seine beißenden und stilistisch ausgefeilten Kritiken ebenso berühmt wie berüchtigt war. Da sich Study mit einer Vielzahl mathematischer Themen beschäftigt hat, führen wir zunächst in die von ihm bearbeiteten Gebiete der Geometrie des 19. Jahrhunderts ein (analytische und synthetische Geometrie im Sinne von Monge, Poncelet, Plücker und Reye, Invariantentheorie Clebsch-Gordan'scher Prägung, abzählende Geometrie von Chasles und Halphen, die Werke Lie's und Grassmann’s, Liniengeometrie sowie Axiomatik und Grundlagenkrise). In seiner darauf folgenden Biographie finden sich als zentrale Stellen seine Habilitation bei Klein über die Chasles’sche Vermutung, sein Streit mit Zeuthen darüber als eine der Debatten der Mathematischen Annalen (aus der er historisch zwar nicht, mathematisch aber tatsächlich als Gewinner hätte herausgehen müssen, wie wir an der Lösung des Problems durch van der Waerden sehen werden) und seine Auseinandersetzungen als etablierter Bonner Professor mit Engel über Lie, Weyl über Invariantentheorie, zahlreichen philosophischen Richtungen über das Raumproblem, Pasch’s Axiomatik, Hilbert’s Formalismus sowie Brouwer’s und Weyl’s Intuitionismus.

In dieser Arbeit stelle ich Aspekte zu QCD Berechnungen vor, welche eng verknüpft sind mit der numerischen Auswertung von NLO QCD Amplituden, speziell der entsprechenden Einschleifenbeiträge, und der effizienten Berechnung von damit verbundenen Beschleunigerobservablen. Zwei Themen haben sich in der vorliegenden Arbeit dabei herauskristallisiert, welche den Hauptteil der Arbeit konstituieren. Ein großer Teil konzentriert sich dabei auf das gruppentheoretische Verhalten von Einschleifenamplituden in QCD, um einen Weg zu finden die assoziierten Farbfreiheitsgrade korrekt und effizient zu behandeln. Zu diesem Zweck wird eine neue Herangehensweise eingeführt welche benutzt werden kann, um farbgeordnete Einschleifenpartialamplituden mit mehreren Quark-Antiquark Paaren durch Shufflesummation über zyklisch geordnete primitive Einschleifenamplituden auszudrücken. Ein zweiter großer Teil konzentriert sich auf die lokale Subtraktion von zu Divergenzen führenden Poltermen in primitiven Einschleifenamplituden. Hierbei wurde im Speziellen eine Methode entwickelt, um die primitiven Einchleifenamplituden lokal zu renormieren, welche lokale UV Counterterme und effiziente rekursive Routinen benutzt. Zusammen mit geeigneten lokalen soften und kollinearen Subtraktionstermen wird die Subtraktionsmethode dadurch auf den virtuellen Teil in der Berechnung von NLO Observablen erweitert, was die voll numerische Auswertung der Einschleifenintegrale in den virtuellen Beiträgen der NLO Observablen ermöglicht. Die Methode wurde schließlich erfolgreich auf die Berechnung von NLO Jetraten in Elektron-Positron Annihilation im farbführenden Limes angewandt.

Die großen christlichen Kirchen in Deutschland befinden sich in einem strukturellen Umbruch, der sich zunehmend auf die Pastoral der örtlichen Pfarreien und Kirchengemeinden auswirkt. Durch die Einführung von Verfahren aus dem New Public Management erhoffen sich Kirchenleitungen beider Konfessionen eine innerkirchliche Personalentwicklung, die vor allem das hauptberufliche Seelsorgepersonal neu motivieren soll. Insbesondere in der katholischen Kirche soll so - in Verbindung mit einem möglichst missionarisch wirksamen Neuaufbruch - die Transformation der ehemals volkskirchlich geprägten Pfarrgemeinden in eine sich neu abzeichnende Sozialgestalt von Kirche als Netzwerk größerer pastoraler Einheiten unterstützt werden. Die vorliegende Arbeit untersucht an der interdisziplinären Schnittstelle von Organisationforschung und praktischer Theologie, inwieweit die Einführung von Zielvereinbarungsgesprächen (ZVG) führungsverantwortliche Priester in der Wahrnehmung ihrer Leitungsaufgabe stärkt. Über 300 leitende Priester aus den Erzbistümern Freiburg und Paderborn hatten an der empirischen Studie mit zwei Messzeitpunkten teilgenommen. 73 Priester waren an beiden Messzeitpunkten A (2007) und B (2008) beteiligt. Unmittelbar nach dem Zeitpunkt A besuchten die befragten Priester der Erzdiözese Freiburg eine ZVG-Einführungsschulung, der sich eine einjährige Gesprächspraxis anschloss. Die in der Erzdiözese Paderborn befragten Priester bildeten die Vergleichsgruppe (ohne entsprechendes Treatment).rn rnWesentliches Ergebnis der Studie sind empirische Hinweise auf signifikante Zusammenhänge zwischen dem priesterlichem Leitungsselbstverständnis, der ZVG-Umsetzung und der Einschätzung der eigenen Führungsqualität.rnrnIn den Selbsteinschätzungen der leitenden Priester (z.B. im Hinblick auf Leitungszufriedenheit, Bedeutung bzw. Gelingen von einzelnen Leitungs- und Gesprächsaspekten, etc.) zeigten sich zum Zeitpunkt A im Vergleich zwischen beiden Diözesen nur geringe Unterschiede. Dies gilt auch für die in A erfolgte Auswertung offener Fragen zum priesterlichen Selbstverständnis, welches als leitungs-amtliche Handlungs- und Objektorientierung in beiden Diözesen am wenigsten stark ausgeprägt war.Zum Zeitpunkt B verdeutlichte die Untersuchung des Tätigkeitsfelds Gemeindeleitung, dass dessen Bedeutung in Freiburg größer war als in Paderborn. Der hierfür erbrachte Kraft- bzw. Zeitaufwand war in Freiburg jedoch niedriger als in Paderborn, was als eine Frucht der verbindlichen ZVG-Einführung in Freiburg interpretiert werden kann. Deutlich wird auch, dass Priester, die der ZVG-Einführung eine hohe Bedeutung beimessen, mit ihren Mitarbeitern viele Gespräche planen. Weil Folgegespräche zum Zeitpunkt B im gleichen Umfang wie schon zum Zeitpunkt A geplant wurden, müssen die konkreten Gesprächserfahrungen in der A-Phase hinreichend positiv gewesen sein. Die Umsetzung zum Zeitpunkt B war jedoch nicht in dem zum Zeitpunkt A geplanten Maß erfolgt, was sich mit Prioritätsverschiebungen erklären lässt. Interessanterweise korreliert die Anzahl der geführten Zielvereinbarungsgespräche mit dem Dienst- und Lebensalter der Priester. Erfahrene Priester, die sich auf eine Mitarbeiterführung durch Zielvereinbarung einlassen, machen demnach gute Erfahrungen mit diesem Personalentwicklungsinstrument. rnrnInsgesamt können die Ergebnisse der Studie zu einer weiteren Kultivierung der Zielvereinbarungsgespräche im kirchlichen Dienst ermutigen. Bistümern, die noch keine Zielvereinbarungsgespräche eingeführt haben, kann eine Implementierung angeraten werden. rn

In clinical medicine, plane radiography is used for detecting the remains of medications in the stomach in oral medication intoxication cases. Since postmortem computed tomography (CT), performed prior to autopsy, is currently intensively entering the forensic routine, the technique was applied to three fatal cases of oral medication intoxication. Here we report CT and autopsy findings for these cases. In all three cases, hyperdense areas within the stomach content were documented. The measurement of Hounsfield Units (HU) beyond 74HU showed mean values of 338, 88 and 98HU. Postmortem CT also showed brain edema and pulmonary aspiration in one case. At autopsy, tablet remains in the stomach were detected microscopically in all three cases. The ex vivo CT scans of the ingested medicaments showed similar HU values. Despite the fact that further case studies are necessary beyond this one, and in spite of its limitations, postmortem CT was found to be a useful screening and documentation method for stomach contents in oral medication intoxication.

Tumor necrosis factor related apoptosis-inducing ligand (TRAIL) and agonistic anti-DR4/TRAIL-R1 and anti-DR5/TRAIL-R2 antibodies are currently under clinical investigation for treatment of different malignancies. TRAIL activates DR4 and DR5 and thereby triggers apoptotic and non-apoptotic signaling pathways, but possible different roles of DR4 or DR5 in these responses has poorly been addressed so far. In the present work, we analyzed cell viability, DISC formation as well as IL-8 and NF-kappaB activation side by side in responses to TRAIL and agonistic antibodies against DR4 (mapatumumab) and against DR5 (lexatumumab) in pancreatic ductal adenocarcinoma cells. We found that all three reagents are able to activate cell death and pro-inflammatory signaling. Death-inducing signaling complex (DISC) analysis revealed that mapatumumab and lexatumumab induce formation of homocomplexes of either DR4 or DR5, whereas TRAIL additionally stimulated the formation of heterocomplexes of both receptors. Notably, blocking of receptors using DR4- and DR5-specific Fab fragments indicated that TRAIL exerted its function predominantly via DR4. Interestingly, inhibition of PKC by Goe6983 enabled DR5 to trigger apoptotic signaling in response to TRAIL and also strongly enhanced lexatumumab-mediated cell death. Our results suggest the existence of mechanisms that silence DR5 for TRAIL- but not for agonistic-antibody treatment.

Oncocytomas are defined as tumors containing in excess of 50% large mitochondrion-rich cells, irrespective of histogenesis and dignity. Along the central neuraxis, oncocytomas are distinctly uncommon but relevant to the differential diagnosis of neoplasia marked by prominent cytoplasmic granularity. We describe an anaplastic ependymoma (WHO grade III) with a prevailing oncocytic component that was surgically resected from the right fronto-insular region of a 43-year-old female. Preoperative imaging showed a fairly circumscribed, partly cystic, contrast-enhancing mass of 2 cm × 2 cm × 1.7 cm. Histology revealed a biphasic neoplasm wherein conventional ependymal features coexisted with plump epithelioid cells replete with brightly eosinophilic granules. Whereas both components displayed an overtly ependymal immunophenotype, including positivity for S100 protein and GFAP, as well as "dot-like" staining for EMA, the oncocytic population also tended to intensely react with the antimitochondrial antibody 113-1. Conversely, failure to bind CD68 indicated absence of significant lysosomal storage. Negative reactions for both pan-cytokeratin (MNF 116) and low molecular weight cytokeratin (CAM 5.2), as well as synaptophysin and thyroglobulin, further assisted in ruling out metastatic carcinoma. In addition to confirming the presence of "zipper-like" intercellular junctions and microvillus-bearing cytoplasmic microlumina, electron microscopy allowed for the pervasive accumulation of mitochondria in tumor cells to be directly visualized. A previously not documented variant, oncocytic ependymoma, is felt to add a reasonably relevant novel item to the differential diagnosis of granule-bearing central nervous system neoplasia, in particular oncocytic meningioma, granular cell astrocytoma, as well as metastatic deposits by oncocytic malignancies from extracranial sites.
