Background: Recent data have suggested that a population of CD4+ CD25high T cells, phenotypically characterized by the expression of CD45RO and CD127, is significantly expanded in stable liver and kidney transplant recipients and represents alloreactive T cells. We analyzed this putative new alloreactive cellular marker in various groups of kidney transplant recipients. Patients and methods: Flow cytometry was used to analyze the expression of CD25, CD45RO and CD127 on peripheral CD4+ T cells. Of 73 kidney recipients, 59 had a stable graft function under standard immunosuppressive therapy (IS), 5 had biopsy-proven chronic humoral rejection (CHR), 8 were stable under minimal IS and one was an operationally "tolerant" patient who had discontinued IS for more than 3 years. Sixty-six healthy subjects (HS) were studied as controls. Results: Overall, the alloreactive T cell population was found to be significantly increased in the 73 kidney recipients (mean ± SE: 15.03 ± 1.04% of CD4+ CD25high T cells) compared to HS (5.93 ± 0.39%) (p <0.001). In the 5 patients with CHR, this population was highly expanded (31.33 ± 4.16%), whereas it was comparable to HS in the 8 stable recipients receiving minimal IS (6.12 ± 0.86%), in 4 patients who had been switched to sirolimus (4.21 ± 0.53%) as well as in the unique "tolerant" recipient (4.69%). Intermediate levels (15.84 ± 0.93%) were found in the 55 recipients with stable graft function on standard CNI-based IS. Regulatory T cells, defined as CD4+ CD25high FoxP3+ CD127low, were found to be significantly reduced in all recipients except in those with minimal or no IS, and this reduction was particularly striking in recipients with CHR. Conclusion: After kidney transplantation, an alloreactive T cell population was found to be significantly expanded and it correlates with the clinical status of the recipients. Interestingly, in stable patients with minimal (or no) IS as well as in patients on sirolimus, alloreactive T cells were comparable the healthy controls. Measuring circulating CD4+ CD25high CD45RO+ CD127high T cells may become a useful monitoring tool after transplantation.

BACKGROUND: Up to 10% of the patients in whom suspected betalactam hypersensitivity (HS) has been excluded by skin and challenge tests report suspected allergic reactions during subsequent treatments with the same or very similar betalactams. It has been suggested that the reactions may result from a resensitization induced by the challenge performed at the time of the allergological work-up. However, most patients did not undergo a second allergological work-up, to determine if the reactions resulted from betalactam HS or not. OBJECTIVES: We aimed to determine if children diagnosed nonallergic to betalactams have tolerated subsequent treatments with the initially suspected and/or other betalactams, and, in case of a reaction, if the reaction resulted from betalactam HS. Methods: We sent a questionnaire concerning the clinical history of their children to the parents of 256 children previously diagnosed nonallergic to betalactams. A second allergological work-up was performed in the children reporting suspected allergic reactions during subsequent treatments with the same and/or other betalactams. Skin tests were performed with the soluble form of the suspected (or very similar) betalactams and other betalactams from the same and other classes. Skin test responses were assessed at 15-20 min (immediate), 6-8 h (semi-late) and 48-72 h (late). Oral challenge (OC) was performed in children with negative skin tests, either at the hospital (immediate and accelerated reactions), or at home (delayed reactions). RESULTS: A response was obtained from 141 children (55.3%). Forty-eight (34%) of those children had not been treated with the betalactams for whom a diagnosis of allergy had been ruled out previously. Seven (7.5%) of the 93 children who had been treated again reported suspected allergic reactions. Skin tests and OC were performed in six of those children, and gave negative results in five children. In one child previously diagnosed nonallergic to amoxicillin associated with clavulanic acid, we diagnosed a delayed HS to clavulanic acid and a serum sickness-like disease to cefaclor. Thus, the frequency of reactions resulting from betalactam HS in children with negative skin and challenge tests is very low, and does not exceed 2.1% (2/93) if we consider that the child which refused a second allergological work-up is really allergic to betalactams. CONCLUSION: Our results in a very large number of children show that reactions presumed to result from betalactam HS are rare in children in whom the diagnosis of betalactam allergy has been ruled out previously. Moreover, they suggest that, as shown for the initial reactions, most of the reactions during subsequent treatments are rather a consequence of the infectious diseases for whom betalactams have been prescribed than a result of betalactam HS. Finally, they suggest that the risk of resensitization by OC is very low, and do not support the notion that skin testing should be repeated in children diagnosed nonallergic to betalactams.

The coreid Leptoglossus zonatus (Dallas, 1852) is commonly found in corn (Zea mays L.) fields in Brazil, and it has been observed flying and landing on objects or persons near these fields. During January, 1995, this behavior was studied in corn plantations. Results indicated that the bugs concentrated on objects (plastic cylinders traps) introduced into their habitat and that their number increased during the first 24 hs. However, as time passed (8 days), this possible territorial or recognition behavior gradually decreased, and tended to disappear.

O cetoprofeno (ácido 2-(3-benzoilfenil) propiónico) é um anti-inflamatório não esteroidal (AINE) utilizado no tratamento de uma grande variedade de doenças inflamatórias agudas e crónicas incluindo a artrite reumatoide, osteoartrite e espondilite anquilosante. A sua administração oral prolongada está associada a diversas reações gastrointestinais, tais como irritações e ulcerações. Neste contexto, é importante desenvolver sistemas alternati-vos, nomeadamente sistemas de libertação controlada para administração oral, transdér-mica ou intradérmica. Este trabalho tem como objetivo testar a possibilidade de utilização de dispersões aquosas de poliuretano (PUDs) como material de suporte para a produção de sistemas de liberta-ção controlada de cetoprofeno. Numa primeira etapa, foram sintetizadas PUDs de base poliéster (policaprolactona, PCL) e poliéter (polipropileno-glicol, PPG) utilizando o méto-do de pré-polímero modificado. As dispersões obtidas foram caracterizadas em termos de pH, viscosidade, teor de sólidos e tamanho de partícula. Numa segunda etapa, foi testada a incorporação do cetoprofeno nas PDUs produzidas utilizando duas estratégias para incre-mentar a sua solubilidade em água: (i) utilização de um co-solvente (acetona, DMSO e HYD) e (ii) utilização de um surfactante não iónico (Tween 80). A incorporação foi testada para teores de 5% e 10% (razão fármaco/polímero, m/m). Os filmes produzidos pelo méto-do da evaporação do solvente foram avaliados quanto à sua homogeneidade e caracteriza-dos por FTIR e DSC. Numa terceira fase realizaram-se estudos de libertação em tampão de fosfato salino (PBS) de pH 7.5 tendo como objetivo avaliar a viabilidade de desenvolvimen-to de diferentes tipologias de dispositivos dependendo de um compromisso entre as pro-priedades dos filmes e o comportamento de libertação. Os resultados obtidos podem ajudar na seleção do material de base mais adequado para um determinado fim. Adicionalmente, e mais importante, comprovou-se a viabilidade de utilizar PUDs como material base para o desenvolvimento de sistemas de libertação con-trolada, utilizando como exemplo o cetoprofeno. A avaliação da toxicidade e da atividade anti-inflamatória dos filmes produzidos foi considerada estando em curso neste momento no grupo do Professor Armando Cunha Júnior.

Esta pesquisa relaciona-se com a temática Higiene e Segurança no Trabalho (HST). Por meio de questionário com os funcionários da sociedade Cabo-verdiana de Cerveja e Refrigerantes (CERIS) e entrevista com o Técnico de Recursos Humanos (TRH) da referida empresa, procuramos saber em que medida a existência das condições HST poderá afectar a motivação do trabalhador e consequentemente conduzir a uma maior ou menor produtividade e desempenho no trabalho da CERIS. Para análise dos dados utilizamos tanto a metodologia quantitativa como qualitativa, pois recorremos ao inquérito por meio de questionário e entrevista. Como resultado do estudo de caso, chegamos a conclusão de que as condições de HST oferecido pela empresa não é um factor que exerce muita influência e/ou motivação para a execução do trabalho, ou seja, as condições físicas do trabalho (iluminação, ruído, vibrações, entre outros), o reconhecimento profissional, o ambiente de trabalho, são consideradas pelos inquiridos como sendo factores que não contribuem para a motivação na execução das tarefas.

A dessalinização da água do mar é um método usado para obter água potável. Na dessalinização da água também é necessária a desinfecção de água com o cloro. Esta desinfecção garante à água condições seguras em termos microbiológicos mas promove a formação de novas substâncias consideradas tóxicas, resultantes da reacção do cloro com a matéria orgânica presente na água. Essas substâncias são designados por subprodutos de desinfecção de água com o cloro (DBP) dos quais destacam-se os trihalometanos (THM), os ácidos haloacéticos (HAA), os haloacetonitrilos (HAN), as haloacetonas (HK). A concentração dessas substâncias em águas para consumo humano é muito baixa e constitui sempre um desafio a sua detecção e quantificação. Neste trabalho desenvolveu-se e optimizou-se métodos de análises de DBP em água para consumo humano. Os DBP voláteis foram analisados por headspace e cromatografia gasosa com detecção por captura electrónica (HS-GC-ECD) e por microextracção em fase sólida no headspace e cromatografia gasosa com detecção por captura electrónica (SPME-HS-GC-ECD). Para os DBP não voláteis (HAA) aplicou-se a extracção em fase sólida e cromatografia líquida com detector de fila de diodos (SPE-RPHPLC- DAD). Foram analisados os DBP na água dessalinizada por osmose inversa proveniente de Cabo Verde e na água da cidade de Porto, de forma a verificar as diferenças em termos qualitativos e quantitativos. Usou-se um planeamento factorial de Box-Behnken para a optimização de métodos analíticos. Este modelo foi igualmente aplicado na simulação laboratorial de dessalinização da água do mar por osmose inversa e desinfecção com o cloro, tendo como objectivo identificar os factores mais significativos na formação de DBP em água dessalinizada (pH, temperatura e tempo de desinfecção).

Glioblastomas (GBMs) are the most frequent and malignant brain tumors in adults. Glucocorticoids (GCs) are routinely used in the treatment of GBMs for their capacity to reduce the tumor-associated edema. Few in vitro studies have suggested that GCs inhibit the migration and invasion of GBM cells through the induction of MAPK phosphatase 1 (MKP-1). Macrophage migration inhibitory factor (MIF), an endogenous GC antagonist is up-regulated in GBMs. Recently, MIF has been involved in tumor growth and migration/invasion and specific MIF inhibitors have been developed on their capacity to block its enzymatic tautomerase activity site. In this study, we characterized several glioma cell lines for their MIF production. U373 MG cells were selected for their very low endogenous levels of MIF. We showed that dexamethasone inhibits the migration and invasion of U373 MG cells, through a glucocorticoid receptor (GR)- dependent inhibition of the ERK1/2 MAPK pathway. Oppositely, we found that exogenous MIF increases U373 MG migration and invasion through the stimulation of the ERK1/2 MAP kinase pathway and that this activation is CD74 independent. Finally, we used the Hs 683 glioma cells that are resistant to GCs and produce high levels of endogenous MIF, and showed that the specific MIF inhibitor ISO-1 could restore dexamethasone sensitivity in these cells. Collectively, our results indicate an intricate pathway between MIF expression and GC resistance. They suggest that MIF inhibitors could increase the response of GBMs to corticotherapy.

Over 94% of Iowa ’ s land is held in private ownership, and the programs of the Iowa Department of Agriculture and Land Stewardship Division of Soil Conservation (IDALS-DSC) have been established to work with these landowners. Over 90% of the landscape is used for agricultural production so much of our focus is in rural communities, but we haven’t overlooked the importance of land management in urban areas. It is crucial to understanding of both conservation and hydrology issues, that what happens on the landscape has immense consequences to the environmental health of our state and the quality of life we enjoy. IDALS-DSC is striving to integrate our programs with other agencies and local concerns to improve water and local concerns to improve water and soil quality throughout the state and nation.

A compactação, uma das principais causas de degradação dos solos agrícolas, tem sido avaliada por meio de diversos indicadores: (a) de qualidade estrutural do solo para o crescimento das plantas, como o intervalo hídrico ótimo (IHO), e (b) de capacidade de suporte do solo, como a pressão de preconsolidação (σp). Este trabalho foi realizado com o objetivo não só de relacionar o IHO e a σp, mas também de determinar valores de pressões críticas (Pcr) que possam ser aplicados ao solo sem restringir o crescimento das plantas ou provocar a compactação adicional do solo. O estudo foi realizado com amostras de Podzólico Vermelho-Amarelo, cultivado com cana-de-açúcar, no município de Piracicaba (SP). Na camada superficial do solo, foram coletadas trinta e seis amostras com estrutura indeformada para a quantificação do IHO e da σp. A partir do IHO, foi obtido o valor de densidade do solo crítica (Dsc) para o crescimento das plantas. A σp foi linearmente relacionada com a densidade e umidade do solo, permitindo a incorporação dos valores de Dsc e do IHO para a quantificação de Pcr. Os resultados indicaram que a Pcr diminuiu linearmente com o incremento da umidade, variando de 360 kPa a 500 kPa para a faixa de umidade de θ = 0,18 m³ m-3 a θ = 0,12 m³ m-3. O parâmetro proposto permitiu definir pressões máximas que podem ser aplicadas ao solo, para diferentes umidades, sem promover a degradação da qualidade estrutural do solo para o crescimento das plantas.

BACKGROUND: The relation of serum uric acid (SUA) with systemic inflammation has been little explored in humans and results have been inconsistent. We analyzed the association between SUA and circulating levels of interleukin-6 (IL-6), interleukin-1beta (IL-1beta), tumor necrosis factor- alpha (TNF-alpha) and C-reactive protein (CRP). METHODS AND FINDINGS: This cross-sectional population-based study conducted in Lausanne, Switzerland, included 6085 participants aged 35 to 75 years. SUA was measured using uricase-PAP method. Plasma TNF-alpha, IL-1beta and IL-6 were measured by a multiplexed particle-based flow cytometric assay and hs-CRP by an immunometric assay. The median levels of SUA, IL-6, TNF-alpha, CRP and IL-1beta were 355 micromol/L, 1.46 pg/mL, 3.04 pg/mL, 1.2 mg/L and 0.34 pg/mL in men and 262 micromol/L, 1.21 pg/mL, 2.74 pg/mL, 1.3 mg/L and 0.45 pg/mL in women, respectively. SUA correlated positively with IL-6, TNF-alpha and CRP and negatively with IL-1beta (Spearman r: 0.04, 0.07, 0.20 and 0.05 in men, and 0.09, 0.13, 0.30 and 0.07 in women, respectively, P<0.05). In multivariable analyses, SUA was associated positively with CRP (beta coefficient +/- SE = 0.35+/-0.02, P<0.001), TNF-alpha (0.08+/-0.02, P<0.001) and IL-6 (0.10+/-0.03, P<0.001), and negatively with IL-1beta (-0.07+/-0.03, P = 0.027). Upon further adjustment for body mass index, these associations were substantially attenuated. CONCLUSIONS: SUA was associated positively with IL-6, CRP and TNF-alpha and negatively with IL-1beta, particularly in women. These results suggest that uric acid contributes to systemic inflammation in humans and are in line with experimental data showing that uric acid triggers sterile inflammation.

Soil organic matter from the surface horizon of two Brazilian soils (a Latosol and a Chernosol), in bulk samples (in situ SOM) and in HF-treated samples (SOM), was characterized by elemental analyses, diffuse reflectance (DRIFT) and transmission Fourier transform infrared spectroscopy (T-FTIR). Humic acids (HA), fulvic acids (FA) and humin (HU) isolated from the SOM were characterized additionally by ultraviolet-visible spectroscopy (UV-VIS). After sample oxidation and alkaline treatment, the DRIFT technique proved to be more informative for the detection of "in situ SOM" and of residual organic matter than T-FTIR. The higher hydrophobicity index (HI) and H/C ratio obtained in the Chernosol samples indicate a stronger aliphatic character of the organic matter in this soil than the Latosol. In the latter, a pronounced HI decrease was observed after the removal of humic substances (HS). The weaker aliphatic character, the higher O/C ratio, and the T-FTIR spectrum obtained for the HU fraction in the Latosol suggest the occurrence of surface coordination of carboxylate ions. The Chernosol HU fraction was also oxygenated to a relatively high extent, but presented a stronger hydrophobic character in comparison with the Latosol HU. These differences in the chemical and functional group composition suggest a higher organic matter protection in the Latosol. After the HF treatment, decreases in the FA proportion and the A350/A550 ratio were observed. A possible loss of FA and condensation of organic molecules due to the highly acid medium should not be neglected.

Fully biodegradable composite materials were obtained through reinforcement of a commercially available thermoplastic starch (TPS) matrix with rapeseed fibers (RSF). The influence of reinforcement content on the water sorption capacity, as well as thermal and thermo-mechanical properties of composites were evaluated. Even though the hydrophilic character of natural fibers tends to favor the absorption of water, results demonstrated that the incorporation of RSF did not have a significant effect on the water uptake of the composites. DSC experiments showed that fibers restricted the mobility of the starch macromolecules from the TPS matrix, hence reducing their capacity to crystallize. The viscoelastic behaviour of TPS was also affected, and reinforced materials presented lower viscous deformation and recovery capacity. In addition, the elasticity of materials was considerably diminished when increasing fiber content, as evidenced in the TMA and DMTA measurements

Genótipos de milho com variabilidade genética contrastante apresentam potenciais produtivos diferentes. Isso pode ser causado, pelo menos em parte, por diferenças morfológicas no sistema radicular e nos parâmetros cinéticos de absorção de nutrientes. Este trabalho objetivou quantificar esses parâmetros em três cultivares de milho. Foram comparados um híbrido simples (HS), um híbrido duplo (HD) e uma variedade de polinização aberta (VPA). Determinaram-se os parâmetros de absorção (influxo máximo, Imax, constante de Michalies-Menten, Km, e concentração na solução onde a absorção cessa, Cmin) para N, P, K, Ca e Mg, além de atributos morfológicos radiculares, em experimentos efetuados em câmara de crescimento, com solução nutritiva. A morfologia das raízes variou pouco entre os genótipos, provavelmente por causa do cultivo das plantas em meio líquido. As diferenças entre genótipos quanto aos parâmetros cinéticos de absorção dependeram do nutriente. O Imax diferiu entre os cultivares para P; o Km, para N e P, e o Cmin, para N e K. A VPA, por apresentar maior variabilidade genética, deveria apresentar menores valores para Km e Cmin do que os híbridos. Contudo, isso só aconteceu para P em relação ao Km. O HS, por apresentar maior potencial produtivo, deveria expressar os maiores valores para Imax, mas isso não ocorreu com nenhum dos macronutrientes avaliados. Portanto, a absorção de nutrientes não parece ser um fator determinante nas diferenças de rendimento de grãos entre genótipos de milho com bases genéticas contrastantes.
