Expression of mitogenic basic fibroblast growth factor (bFGF) in the central nervous system is inhibited by direct cell contact and is implicated in reactive and neoplastic transformation of astrocytes. The molecular mechanisms controlling expression of bFGF were examined in cultures of human astrocytes. Cell-density-dependent depletion of bFGF mRNA levels parallels changes in bFGF gene protein. Regulation of transcription of a bFGF luciferase reporter gene containing an upstream region (bp -1800 to +314) of the bFGF gene promoter mimicks the density-dependent regulation of the endogenous bFGF gene in transfected astrocytes. Deletion analysis has identified a fragment (bp -650 to -513) and sequences further downstream (bp -274 to +314) as the regions required for the regulation of bFGF gene activity by cell density. Unlike in astrocytes, changing the cell density of glioma cell cultures does not affect the levels of bFGF protein and mRNA. bFGF luciferase constructs were expressed at the same level in high- or low-density cultures of glioma cells, indicating altered regulation of the bFGF gene promoter. Electrophoretic mobility shift assays showed binding of nuclear proteins to a fragment of bFGF gene promoter from bp -650 to -453. This binding was abolished by a deletion of the upstream cell-density-responsive region (bp -650 to -512). Binding was observed with nuclear extracts from subconfluent astrocytes but was reduced in extracts from confluent astrocytes. Our results indicate that induction of bFGF in astrocytes upon reduction of cell density is mediated transcriptionally by positive trans-acting factors interacting with bFGF promoter. In contrast, nuclear proteins from glioma cells bind to the promoter region from bp -650 to -453 independent of cell density. Thus, the constitutive binding of trans-acting factor(s) to the region of the bFGF promoter from bp -650 to -453 may be responsible for the continuous expression of bFGF that leads to the uncontrolled growth of glioma cells.

Nonlinear analyses of infant heart rhythms reveal a marked rise in the complexity of the electrocardiogram with maturation. We find that normal mature infants (gestation greater than or equal to 35 weeks) have complex and distinctly nonlinear heart rhythms (consistent with recent reports for healthy adults) but that such nonlinearity is lacking in preterm infants (gestation > or = to 27 weeks) where parasympathetic-sympathetic interaction and function are presumed to be less well developed. Our study further shows that infants with clinical brain death and those treated with atropine exhibit a similar lack of nonlinear feedback control. These three lines of evidence support the hypothesis championed by Goldberger et al. [Goldberger, A.L., Rigney, D.R. & West, B.J. (1990) Sci. Am. 262, 43-49] that autonomic nervous system control underlies the nonlinearity and possible chaos of normal heart rhythms. This report demonstrates the acquisition of nonlinear heart rate dynamics and possible chaos in developing human infants and its loss in brain death and with the administration of atropine. It parallels earlier work documenting changes in the variability of heart rhythms in each of these cases and suggests that nonlinearity may provide additional power in characterizing physiological states.

beta-Amyloid deposition and neurofibrillary tangle formation are two histopathological features of Alzheimer disease. We have previously reported that beta-amyloid immunoreactive deposits form in the brains of transgenic mice programmed for neuronal expression of the 751-amino acid isoform of human beta-amyloid precursor protein (beta-APP751) and now describe that these animals also display Alz50 intraneuronal immunoreactivity similar to that seen in early Alzheimer disease. This suggests that abnormal beta-APP expression and/or beta-amyloid deposition promotes pathogenic alterations in tau protein. The frequency of both beta-amyloid deposition and Alz50-positive neurons was twice as prevalent in brains from old (22 months) as compared to young (2-3 months) beta-APP751 transgenic mice. This increase in histopathology with age in beta-APP751 transgenic mice parallels the time-dependent progression seen in the human disease.

A method for simultaneous iontophoretic injections of the anterograde tracer Phaseolus vulgaris leukoagglutinin and the retrograde tracer fluorogold was used to characterize in the rat a hypothalamothalamocortical pathway ending in a region thought to regulate attentional mechanisms by way of eye and head movements. The relevant medial hypothalamic nuclei receive pheromonal information from the amygdala and project to specific parts of the thalamic nucleus reuniens and anteromedial nucleus, which then project to a specific lateral part of the retrosplenial area (or medial visual cortex). This cortical area receives a convergent input from the lateral posterior thalamic nucleus and projects to the superior colliculus. Bidirectional connections with the hippocampal formation suggest that activity in this circuit is modified by previous experience. Striking parallels with basal ganglia circuitry are noted.

Experimental information on the structure and dynamics of molten globules gives estimates for the energy landscape's characteristics for folding highly helical proteins, when supplemented by a theory of the helix-coil transition in collapsed heteropolymers. A law of corresponding states relating simulations on small lattice models to real proteins possessing many more degrees of freedom results. This correspondence reveals parallels between "minimalist" lattice results and recent experimental results for the degree of native character of the folding transition state and molten globule and also pinpoints the needs of further experiments.

This study examines some aspects of Caldwell's (200 I) hypotheses about the adaptations associated with elevations on the clinical scales of the Minnesota Multiphasic Personality Assessment-2 (MMPI-2). The argument is made that the adaptation to early rejection and neglect, which he attributes to Scale 4, parallels the adaptation represented by a dismissing attachment. Archival data of 72 individuals who completed assessment batteries that included both the MMPI-2 and that Adult Attachment Projective (AAP) were analyzed to determine if there was a relationship between elevations on Scale 4 and classification as dismissing attachment. No significant relationship was found for the primary hypothesis; however, some small effects were noted in related exploratory analyses. Possible reasons for the lack of a significant result are discussed a well as directions for future research.

Abstract This paper investigates themes and parallels related to the traumatic experiences women face within the correctional setting and how these experiences influence women's behavior choices that increase their risk of recidivism. Intersubjective Systems Theory is used to conceptualize the distinct dynamics and impact of trauma with this particular population. Intersubjectivity also informs the changes needed to create an environment that would help women in correctional settings to heal, avoid recidivism, and foster successful community reintegration. Principles from Intersubjective Systems Theory are reviewed in this paper to demonstrate: (a) how developmental trauma impacts the lives of incarcerated women, (b) how these women's attempts at self-healing are often maladaptive and lead to arrests, (c) how the current climate in corrections leads to retraumatization and promotes later recidivism, and (d) what changes in the corrections system would promote optimal healing and better outcomes for incarcerated women. Improved outcomes are defined as healthy boundaries, empowerment in choice of relationships, improvement of social support and occupational skills, and reduction of relapse and recidivism.

We introduce the notion of Lipschitz compact (weakly compact, finite-rank, approximable) operators from a pointed metric space X into a Banach space E. We prove that every strongly Lipschitz p-nuclear operator is Lipschitz compact and every strongly Lipschitz p-integral operator is Lipschitz weakly compact. A theory of Lipschitz compact (weakly compact, finite-rank) operators which closely parallels the theory for linear operators is developed. In terms of the Lipschitz transpose map of a Lipschitz operator, we state Lipschitz versions of Schauder type theorems on the (weak) compactness of the adjoint of a (weakly) compact linear operator.

In the wake of the disclosures surrounding PRISM and other US surveillance programmes, this paper assesses the large-scale surveillance practices by a selection of EU member states: the UK, Sweden, France, Germany and the Netherlands. Given the large-scale nature of these practices, which represent a reconfiguration of traditional intelligence gathering, the paper contends that an analysis of European surveillance programmes cannot be reduced to a question of the balance between data protection versus national security, but has to be framed in terms of collective freedoms and democracy. It finds that four of the five EU member states selected for in-depth examination are engaging in some form of large-scale interception and surveillance of communication data, and identifies parallels and discrepancies between these programmes and the NSA-run operations. The paper argues that these programmes do not stand outside the realm of EU intervention but can be analysed from an EU law perspective via i) an understanding of national security in a democratic rule of law framework where fundamental human rights and judicial oversight constitute key norms; ii) the risks posed to the internal security of the Union as a whole as well as the privacy of EU citizens as data owners and iii) the potential spillover into the activities and responsibilities of EU agencies. The paper then presents a set of policy recommendations to the European Parliament.

A new Stata command called -mgof- is introduced. The command is used to compute distributional tests for discrete (categorical, multinomial) variables. Apart from classic large sample $\chi^2$-approximation tests based on Pearson's $X^2$, the likelihood ratio, or any other statistic from the power-divergence family (Cressie and Read 1984), large sample tests for complex survey designs and exact tests for small samples are supported. The complex survey correction is based on the approach by Rao and Scott (1981) and parallels the survey design correction used for independence tests in -svy:tabulate-. The exact tests are computed using Monte Carlo methods or exhaustive enumeration. An exact Kolmogorov-Smirnov test for discrete data is also provided.

A detailed d18O and d13C stratigraphy has been generated from analysis of well-preserved Albian - Early Maastrichtian foraminifera from Deep Sea Drilling Project (DSDP) Sites 511 and 327 (Falkland Plateau; ~58°S - 62°S paleolatitude) in the southern South Atlantic, and Cenomanian and Coniacian - Santonian foraminifera from DSDP Site 258 (Naturaliste Plateau; ~58°S paleolatitude) in the southern Indian Ocean. These results, when combined with previously published Maastrichtian stable isotope data from Ocean Drilling Program (ODP) Site 690 (Weddell Sea, ~65°S paleolatitude), provide new insight into the climatic and oceanographic history of the southern high latitudes during Middle-Late Cretaceous time. The planktonic foraminifer d18O curves reveal a gradual warming of surface waters from the Albian through the Cenomanian followed by extremely warm surface waters from the Turonian through the early Campanian. Long-term cooling of surface waters began in the late early Campanian and continued through the end of the Maastrichtian. The benthic foraminifer d18O record generally parallels changes in the oxygen isotopic curves defined by shallow-dwelling planktonic foraminifera. The vertical oxygen and carbon isotopic gradients were relatively low during the Albian - Cenomanian, high from the Turonian - Early Campanian, and then low during the late Campanian and Maastrichtian.

The principal gaseous carbon-containing components identified in the first 400 m of sediment at Deep Sea Drilling Project Site 533, Leg 76, are methane (CH4) and carbon dioxide (CO2). Below a sub-bottom depth of about 25 m, sediment cores commonly contained pockets caused by the expansion of gas upon core recovery. The carbon isotopic composition (d13C per mil relative to PDB standard) of CH4 and CO2 in these gas pockets has been measured, resulting in the following observations: (1) d13C-CH4 values increase with depth from approximately -94 per mil in the uppermost sediment to about -66 per mil in the deepest sediment, reflecting a systematic but nonlinear depletion of 12C with depth. (2) d13C-CO2 values also increase with depth of sediment from about -25 per mil to about -4 per mil, snowing a depletion of 12C that closely parallels the trend of the isotopic composition of CH4. The magnitude and parallel distribution of d13C values for both CH4 and CO2 are consistent with the concept that the formation of the CH4 resulted from the microbiological reduction of CO2 from organic substances. These results imply that CH4 and CO2 incorporated in gas hydrates at this site are biogenic.

The Early Toarcian was characterized by two large perturbations to the carbon cycle: a positive trend associated with increased organic matter burial and ocean anoxia, and a pronounced negative carbon isotope excursion (CIE). We contrast the geochemical evolution in the carbonate phases of two successions: one from the Paris Basin (Sancerre core, comprising black shales), the other from the Lusitanian Basin (Peniche section with very minor lithological expression of bottom water anoxia). Our aim was to identify whether these carbon cycle perturbations were related, and differentiate between the common (global) versus regional expressions of the biogeochemical response and ocean chemistry. Our results highlight contrasts in timing of different phases of anoxia in both locations through the widely documented negative CIE. Widespread anoxic conditions were not a necessary prerequisite for generating a pronounced CIE, as required by the recycling (so-called "Küspert") model. The production of carbonate simultaneously dropped during the d13C negative shift in both locations, likely in response to lowered seawater saturation rate induced by substantial absorption of CO2 from the atmosphere. The recovery interval was accompanied by a rapid reestablishment of seawater alkalinity, and primary and carbonate productivity in epicontinental seas, as evidenced by high d13C and Sr/Ca, in contrast with the more open ocean regime in the Lusitanian Basin. Our results confirm that parallels can be draw between the ocean productivity response and feedback during the Toarcian CIE and the PETM. Both events are characterized by ocean acidification and reduced pelagic calcification followed by a peak in nearshore coccolith productivity, which could have helped the recovery from the perturbation.
