In this study we have investigated the role of the N-terminal region of thyroid hormone receptors (TRs) in thyroid hormone (TH)-dependent transactivation of a thymidine kinase promoter containing TH response elements composed either of a direct repeat or an inverted palindrome. Comparison of rat TR beta 1 with TR beta 2 provides an excellent model since they share identical sequences except for their N termini. Our results show that TR beta 2 is an inefficient TH-dependent transcriptional activator. The degree of transactivation corresponds to that observed for the mutant TR delta N beta 1/2, which contains only those sequences common to TR beta 1 and TR beta 2. Thus, TH-dependent activation appears to be associated with two separate domains. The more important region, however, is embedded in the N-terminal domain. Furthermore, the transactivating property of TR alpha 1 was also localized to the N-terminal domain between amino acids 19 and 30. Using a coimmunoprecipitation assay, we show that the differential interaction of the N terminus of TR beta 1 and TR beta 2 with transcription factor IIB correlates with the TR beta 1 activation function. Hence, our results underscore the importance of the N-terminal region of TRs in TH-dependent transactivation and suggest that a transactivating signal is transmitted to the general transcriptional machinery via a direct interaction of the receptor N-terminal region with transcription factor IIB.

Vascular cell adhesion molecule 1 (VCAM-1) represents a structurally and functionally distinct class of immunoglobulin superfamily molecules that bind leukocyte integrins and are involved in inflammatory and immune functions. X-ray crystallography defines the three-dimensional structure of the N-terminal two-domain fragment that participates in ligand binding. Residues in domain 1 important for ligand binding reside in the C-D loop, which projects markedly from one face of the molecule near the contact between domains 1 and 2. A cyclic peptide that mimics this loop inhibits binding of alpha 4 beta 1 integrin-bearing cells to VCAM-1. These data demonstrate how crystallographic structural information can be used to design a small molecule inhibitor of biological function.

The murine p53 protein contains two nucleic acid-binding sites, a sequence-specific DNA-binding region localized between amino acid residues 102-290 and a nucleic acid-binding site without sequence specificity that has been localized to residues 364-390. Alternative splicing of mRNA generates two forms of this p53 protein. The normal, or majority, splice form (NSp53) retains its carboxyl-terminal sequence-nonspecific nucleic acid-binding site, which can negatively regulate the sequence-specific DNA-binding site. The alternative splice form of p53 (ASp53) replaces amino acid residues 364-390 with 17 different amino acids. This protein fails to bind nucleic acids nonspecifically and is constitutive for sequence-specific DNA binding. Thus, the binding of nucleic acids at the carboxyl terminus regulates sequence-specific DNA binding by p53. The implications of these findings for the activation of p53 transcriptional activity following DNA damage are discussed.

A global cellular reorganization occurs during the reticulocyte stage of erythroid differentiation. This reorganization is accomplished partly through programmed protein degradation. The selection of proteins for degradation can be mediated by covalent attachment of ubiquitin. We have cloned cDNAs encoding two ubiquitin-conjugating (E2) enzymes, E2-20K and E2-230K, and found their genes to be strongly induced during the differentiation of erythroblasts into reticulocytes. Induction of the E2-20K and E2-230K genes is specific, as transcript levels for at least two other ubiquitinating enzymes fall during erythroblast differentiation. In contrast to most proteins induced in reticulocytes, E2-20K and E2-230K enzymes are present at strongly reduced levels in erythrocytes and thus decline in abundance as reticulocyte maturation is completed. This result suggests that both enzymes function during the reticulocyte stage, when enhanced protein degradation has been observed. These data implicate regulated components of the ubiquitin conjugation machinery in erythroid differentiation.

The effects of NusA on the RNA polymerase contacts made by nucleotides at internal positions in the nascent RNA in Escherichia coli transcription complexes were analyzed by using the photocrosslinking nucleotide analog 5-[(4-azidophenacyl) thio]-UMP. It was placed at nucleotides between +6 and +15 in RNA transcribed from the phage lambda PR' promoter. Crosslinks of analog in these positions in RNAs which contained either 15, 28, 29, or 49 nt were examined. Contacts between the nascent RNA and proteins in the transcription complex were analyzed as the RNA was elongated, by placing the crosslinker nearest the 5' end of the RNA 10, 23, 24, or 44 nt away from the 3' end. The beta or beta' subunit of polymerase, and NusA when added, were contacted by RNA from 15 to 49 nt long. When the upstream crosslinker was 24 nt from the 3" end of the RNA (29-nt RNA), alpha was also contacted in the absence of NusA. The addition of NusA prevented RNA crosslinking to alpha. When the crosslinker was 44 nt from the 3' end (49-nt RNA), alpha crosslinks were still observed, but crosslinks to beta or beta' and NusA were greatly diminished. RNA crosslinking to alpha, and loss of this crosslink when NusA was added, was observed in the presence of NusB, NusE, and NusG and when transcription was carried out in the presence of an E. coli S100 cell extract. Peptide mapping localized the RNA interactions to the C-terminal domain of alpha.

Hexamethylenebisacetamide-induced terminal differentiation of Friend virus-transformed murine erythroleukemia (MEL) cells can be inhibited by okadaic acid, an inhibitor of type 1 and type 2A protein phosphatases. The inhibition is shown to be correlated with prevention of dephosphorylation of retinoblastoma protein (pRB) in cells and bypass of G1 prolongation in the cell cycle. These results suggest that pRB-mediated G1 prolongation is necessary for MEL cells to commit to terminal differentiation. However, further experiments demonstrate that the simple cell cycle exit is not sufficient for commitment to terminal differentiation. Induction of dephosphorylation of pRB and subsequent G1 prolongation by forskolin does not lead MEL cells to differentiate. Additional pRB has been expressed in MEL cells by transfection with a neo-resistant plasmid containing RB cDNA under the control of a cytomegalovirus promoter. Exogenously expressed pRB is hyperphosphorylated in logarithmically growing MEL cells without any noticeable change in growth rate between the transfected cell line and the parental cell line. This result suggests that pRB in MEL cells is regulated by protein kinases and protein phosphatases and not by transcription.

Ribotoxins are cytotoxic members of the family of fungal extracellular ribonucleases best represented by RNase T1. They share a high degree of sequence identity and a common structural fold, including the geometric arrangement of their active sites. However, ribotoxins are larger,with a well-defined N-terminal β-hairpin, and display longer and positively charged unstructured loops. These structural differences account for their cytotoxic properties.Unexpectedly, the discovery of hirsutellin A (HtA), a ribotoxin produced by the invertebrate pathogen Hirsutella thompsonii, showed how it was possible to accommodate these features into a shorter amino acid sequence. Examination of HtA N-terminal β-hairpin reveals differences in terms of length, charge, and spatial distribution. Consequently,four different HtA mutants were prepared and characterized. One of them was the result of deleting this hairpin [Δ(8-15)] while the other three affected single Lys residues in its close spatial proximity (K115E, K118E, and K123E). The results obtained support the general conclusion that HtA active site would show a high degree of plasticity,being able to accommodate electrostatic and structural changes not suitable for the other previously known larger ribotoxins, as the variants described here only presented small differences in terms of ribonucleolytic activity and cytotoxicity against cultured insect cells.

La comunicación entre las neuronas ocurre en regiones anatómicamente identificables denominadas sinapsis. Existen dos tipos de transmisión sináptica, las sinapsis químicas y las eléctricas, aunque predominan las sinapsis químicas. En este tipo de sinapsis, la comunicación neuronal ocurre en zonas especializadas de los axones, denominados terminales sinápticos, los cuales almacenan en su interior pequeñas vesículas que contienen un neurotransmisor. Ante la llegada de un potencial de acción al terminal presináptico, el flujo de calcio, generado a través de la apertura de canales de calcio voltaje-dependientes, provoca la fusión de las vesículas con la membrana del terminal presináptico, y la liberación del neurotransmisor a la hendidura sináptica. La fusión vesicular tiene lugar en regiones de membrana del terminal presináptico, molecularmente especializadas para dicho evento exocitótico, denominadas Zonas Activas. Este neurotransmisor liberado difunde por la hendidura sináptica y se une a receptores específicos ubicados en la membrana de la neurona postsináptica, propagándose así el impulso nervioso. Tras este evento exocitótico, que implica la fusión de multitud de vesículas, es necesario un proceso de endocitosis, que ocurre en las zonas perisinápticas, y que está encargado de recuperar las fracciones de membrana que formaban las vesículas sinápticas, con dos objetivos: 1. Impedir el aumento de la superficie de la zona activa, lo cual llevaría a su desestructuración, y 2. La formación de nuevas vesículas que se rellenen de neurotransmisor y puedan prepararse para una nueva ronda de exocitosis. La endocitosis que sigue a un estímulo moderado, está mediada por clatrina y recicla vesículas independientes preparadas para el rellenado con neurotransmisor. Tras estímulos intensos que provocan exocitosis de múltiples vesículas, la retirada de membrana ocurre a través de otro mecanismo más lento y menos eficaz, denominado endocitosis en masa, el cual recicla grandes fragmentos de membrana y acumula estructuras endosomales en el interior del terminal, los cuales no siempre rinden vesículas funcionales inmediatamente...

La infección por el virus de la hepatitis C (VHC) tiene un gran impacto a nivel mundial, considerándose una de las principales causas de hepatitis crónica, enfermedad hepática terminal y hepatocarcinoma. Hasta la incorporación de los nuevos agentes antivirales, el genotipo 1b era el que tenía tasas más bajas de respuesta viral sostenida (RVS); y a su vez, el de mayor prevalencia en nuestro medio. Lo que ha llevado a la búsqueda de marcadores predictores de respuesta, con el fin de identificar a aquellos pacientes que pudieran beneficiarse del tratamiento. Estudios previos han mostrado que la variabilidad genómica en la región codificante de la proteína C del Core y en la región comprendida entre los aminoácidos 2209 y 2248 (región determinante de la sensibilidad al IFN: ISDR) de la región que codifica la proteína no-­‐estructural 5A (NS5A) están relacionados con la respuesta al tratamiento con biterapia. Es por ello, que el objetivo de nuestro estudio es caracterizar a nivel genómico ambas regiones y analizar su impacto en pacientes con infección crónica por el genotipo 1b. Los dispositivos de asistencia mecánica circulatoria (AMC) son una opción terapéutica prometedora para los pacientes con insuficiencia cardíaca avanzada. Pueden actuar como puente al trasplante, como terapia de destino para los pacientes con contraindicaciones para el trasplante, o como un puente hacia un futuro de recuperación. En las últimas décadas, las AMC se han utilizado cada vez más en los pacientes con insuficiencia cardíaca terminal, ya que el trasplante cardiaco está limitado por una falta de donantes. El principal objetivo de una AMC es mantener la perfusión de los órganos vitales. Para mejorar la salida clínica de la AMC, es necesario optimizar las condiciones perioperatorias (AMC de flujo continuo, monitores hemodinámicos, y los fármacos anestésicos) en la implantación de estos dispositivos. Aunque varios estudios muestran los efectos de la AMC en el flujo de los órganos (corazón, cerebro, hígado y riñón), el efecto de los anestésicos en el flujo de los órganos en pacientes con una AMC no ha sido analizado hasta la fecha...

Hemos estudiado una muestra (n=IOO) de profesionales de enfermería de las provincias de Alicante y Valencia, analizando las actitudes, según el modelo de Ajzen y Fishbein, con respecto a dos grupos de enfermos: crónicos y terminales. En general las actitudes de estos profesionales son bastante positivas en ambos casos y en todos su s componentes; aunque encontramos como hecho más destacado una tendencia actitudinal más baja significativamente hacia el enfermo terminal en sus componentes afectivo y comportamental.

A highly regio- and stereoselective oxime palladacycle/imidazolinium-catalyzed head to head dimerization of terminal aryl alkynes in water is presented. The reaction, which is carried out at 130 °C under microwave irradiation in the presence of 1,3-bis-(2,6-diisopropylphenyl)imidazolinium chloride as ligand, triethylamine as base, and TBAB as surfactant, allows the synthesis of (E)-1,4-enynes as single stereoisomers in good isolated yields.

4-1BB (CD137) est un membre de la superfamille TNFR qui est impliqué dans la transmission des signaux de survie aux lymphocytes. TRAF1 est une protéine adaptatrice qui est recrutée par 4-1BB et autres TNFRs et est caractérisée par une expression très restreinte aux lymphocytes, cellules dendritiques et certaines cellules épithéliales. TRAF1 est nécessaire pour l’expansion et la survie des cellules T mémoire en présence d'agonistes anti-4-1BB in vivo. De plus, TRAF1 est requise en aval de 4-1BB pour activer (phosphoryler) la MAP kinase Erk impliquée dans la régulation de la molécule pro-apoptotique Bim. Suite à l’activation du récepteur 4-1BB, TRAF1 et ERK sont impliqués dans la phosphorylation de Bim et la modulation de son expression. L’activation et la régulation de TRAF1 et Bim ont un rôle important dans la survie des cellules T CD8 mémoires. Dans cette étude, nous avons utilisé une approche protéomique afin de pouvoir identifier de nouveaux partenaires de liaison de TRAF1. Utilisant cette stratégie, nous avons identifié que LSP1 (Leukocyte Specific Protein 1) est recruté dans le complexe de signalisation 4-1BB de manière TRAF1 dépendante. Une caractérisation plus poussée de l’interaction entre TRAF1 et LSP1 a montré que LSP1 lie la région unique N-terminal de TRAF1 de façon indépendante de la région conservée C-terminal. À l’instar des cellules T déficientes en TRAF1, les cellules T déficientes en LSP1 ne sont pas capables d’activer ERK en aval de 4-1BB et par conséquent ne peuvent pas réguler Bim. Ainsi, TRAF1 et LSP1 coopèrent en aval de 4-1BB dans le but d’activer ERK et réguler en aval les niveaux de Bim dans les cellules T CD8. Selon la littérature, le récepteur 4-1BB n’est pas exprimé à la surface des cellules B murines, mais le récepteur 4-1BB favorise la prolifération et la survie des cellules B humaines. Cependant, il est important d'étudier l'expression du récepteur 4-1BB dans les cellules B murines afin de disposer d'un modèle murin et de prédire la réponse clinique à la manipulation de 4-1BB. En utilisant différentes stimulations de cellules B murines primaires, nous avons identifié que le récepteur 4-1BB est exprimé à la surface des cellules B de souris suite à une stimulation avec le LPS (Lipopolysaccharides). Une caractérisation plus poussée a montré que le récepteur 4-1BB est induit dans les cellules B murines d'une manière dépendante de TLR4 (Toll Like Receptor 4). Collectivement, notre travail a démontré que la stimulation avec le LPS induit l’expression du récepteur 4-1BB à la surface des cellules B murines, menant ainsi à l'induction de TRAF1. De plus, TRAF1 et LSP1 coopèrent en aval de 4-1BB pour activer la signalisation de la Map kinase ERK dans les cellules B murines de manière similaire aux cellules T. Les cellules B déficientes en TRAF1 et les cellules B déficientes en LSP1 ne sont pas en mesure d'activer la voie ERK en aval de 4-1BB et montrent un niveau d’expression du récepteur significativement diminué comparé aux cellules B d’une souris WT. Ainsi, TRAF1 et LSP1 sont nécessaires pour une expression maximale du récepteur 4-1BB à la surface cellulaire de cellules B murines et coopèrent en aval de 4-1BB afin d'activer la cascade ERK dans les cellules B murines.

The morphological variability (coiling properties, size and shape) of the planktic foraminifer Contusortuncana contusa (Cushman) in the terminal Cretaceous ocean was examined at eight deep-sea sites and two continental sections from low (16°) to middle (42°) paleolatitudes in both hemispheres. The material used in this study includes samples from the South Atlantic (DSDP Sites 356, 527 and 525A), North Atlantic (Sites 384 and 548A), Indian and Pacific Oceans (DSDP Site 465A and ODP Sites 761C and 762C) and Tethyan Ocean (outcrop sections from El-Kef and Caravaca). On average 45 specimens from two samples per location were analysed, from an interval corresponding approximately to the last 60 kyr of the Cretaceous. No differences in coiling direction (dextral proportions were > 90% in all samples), percentage of kummerform specimens (usually > 50%) and number of chambers in the last whorl (4-5) were observed between the sites. Both test size (expressed as spiral outline area and test volume) and total number of chambers increase significantly towards lower latitudes. Similarly, test conicity, examined by shape coordinate and eigenshape methods, and angularity of the spiral outline show a rather continuous, slight increase towards lower latitudes. Kummerform specimens of C. contusa were slightly larger and more conical than normalforms and possessed substantially more chambers (both totally and in the last whorl). A principal components analysis of the sample means of five variables describing size and shape clearly distinguished high-latitude sites (525A, 527, 548A, 761C and 762C) from low-latitude sites (384, 465A, Caravaca and El-Kef). Specimens from Site 356 are transitional with respect to those two groups. The results indicate: (1) considerable morphological variation in C. contusa during the terminal Cretaceous comparable to that known in many Recent planktic foraminiferal species and (2) a geographical distribution of this variation corresponding to previously suggested biogeographic schemes based on quantitative analysis of planktic foraminiferal assemblages. Despite the differences in sample means, the overall morphology of C. contusa overlaps among the sites studied, supporting the classification of all C. contusa morphotypes as a single species. Similarly, no discrete morphologic groups could be distinguished within any of the samples.

The problem of preparation of a program to perform it on multiprocessor system of a cluster type is considered. When developing programs for a cluster computer the technology based on use of the remote terminal is applied. The situation when such remote terminal is the computer with operational system Windows is considered. The set of the tool means, allowing carrying out of editing program texts, compiling and starting programs on a cluster computer, is suggested. Advantage of an offered way of preparation of programs to execution is that it allows as much as possible to use practical experience of programmers used to working in OS Windows environment.
